Preparation of Essential Oil Rich in Β-Asarone from Acorus Tatarinowii Schott Rhizome by Enzymatic Pretreatment and Solvent-Free Microwave-Assisted Method and Evaluation of Acetylcholinesterase Inhibitory Activity in Vitro

Abstract:The essential oil from fresh rhizomes of Acorus tatarinowii Schott was extracted by enzymatic pretreatment and solvent-free microwave-assisted method (EP-SFM) and characterized. Based on single-factor and response surface optimization experiments, the optimum conditions of essential oil extraction were obtained as follows: the amount of enzyme added was 1.5%, the enzymatic hydrolysis temperature was 49.43 oC, the enzymatic hydrolysis time was 2 h, the microwave power was 586.17 W and the microwave time was 47.33 min, respectively. The highest essential oil (1.48%) was obtained at the optimum conditions, which was characterized by scanning electron microscopy (SEM), thermogravimetric analysis (TGA), fourier transform infrared (FTIR), GC-MS and acetylcholinesterase inhibition activity analysis in vitro assay. The results indicated EP-SFM method effectively improved the extraction rate of essential oil and shortened the extraction time. More importantly, this method greatly increased the active component content of β-asarone (65.87%), in which the content of β-asarone was increased by 18.06% compared with the conventional steam distillation method (SD). In addition, the acetylcholinesterase inhibitory activity of essential oil from EP-SFM was analyzed, which had the best acetylcholinesterase inhibitory activity and were closest to the positive control group. The results will provide data to support the extraction of essential oil from the rhizomes of Acorus tatarinowii Schott by the EP-SFM method and contribute to its industrial development.
