The Extracts of Artemisia absinthium L. Suppress the Growth of Hepatocellular Carcinoma Cells through Induction of Apoptosis via Endoplasmic Reticulum Stress and Mitochondrial-Dependent Pathway

Abstract:Artemisia absinthium L. has pharmaceutical and medicinal effects such as antimicrobial, antiparasitic, hepatoprotective, and antioxidant activities. Here, we prepared A. absinthium ethanol extract (AAEE) and its subfractions including petroleum ether (AAEE-Pe) and ethyl acetate (AAEE-Ea) and investigated their antitumor effect on human hepatoma BEL-7404 cells and mouse hepatoma H22 cells. The cell viability of hepatoma cells was measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The apoptosis, cell cycle, mitochondrial membrane potential (Δψm), and reactive oxygen species (ROS) were analyzed by flow cytometry. The levels of proteins in the cell cycle and apoptotic pathways were detected by Western blot. AAEE, AAEE-Pe, and AAEE-Ea exhibited potent cytotoxicity for both BEL-7404 cells and H22 cells through the induction of cell apoptosis and cell cycle arrest. Moreover, AAEE, AAEE-Pe, and AAEE-Ea significantly reduced Δψm, increased the release of cytochrome c, and promoted the cleavage of caspase-3, caspase-9, and poly(ADP-ribose) polymerase (PARP) in BEL-7404 and H22 cells. AAEE, AAEE-Pe, and AAEE-Ea significantly upregulated the levels of ROS and C/EBP-homologous protein (CHOP). Further, AAEE, AAEE-Pe, and AAEE-Ea significantly inhibited tumor growth in the H22 tumor mouse model and improved the survival of tumor mice without side effects. These results suggest that AAEE, AAEE-Pe, and AAEE-Ea inhibited the growth of hepatoma cells through induction of apoptosis, which might be mediated by the endoplasmic reticulum stress and mitochondrial-dependent pathway.

This paper aims to solve the problems in the treatment of hepatocellular carcinoma (HCC). Specifically, the research attempts to provide a scientific basis for the development of new, safe and effective anti - tumor drugs by exploring the inhibitory effects and mechanisms of different extracts of Artemisia absinthium L. on liver cancer cells. ### Research Background and Problems Hepatocellular carcinoma is one of the common malignant tumors worldwide. In 2018, it was listed as the sixth most common cancer and the fourth leading cause of cancer death globally. Current treatment methods include surgical resection, liver transplantation, intra - arterial treatment and immunotherapy, etc., but the prognosis of most patients is still poor. Commonly used drugs such as oxaliplatin and sorafenib are not ideal in efficacy due to side effects and multi - drug resistance. Therefore, there is an urgent need to develop new effective drugs for HCC treatment. ### Research Objectives The main objective of this study is to evaluate the anti - tumor effects of Artemisia absinthium L. ethanol extract (AAEE) and its sub - components, petroleum ether extract (AAEE - Pe) and ethyl acetate extract (AAEE - Ea), on human liver cancer BEL - 7404 cells and mouse liver cancer H22 cells. Specifically, the study focuses on the following aspects: 1. **Inhibition of cell proliferation**: Detect the inhibitory effects of AAEE, AAEE - Pe and AAEE - Ea on the proliferation of liver cancer cells. 2. **Induction of cell apoptosis**: Analyze whether these extracts can induce apoptosis in liver cancer cells and explore their possible molecular mechanisms. 3. **Cell cycle arrest**: Investigate whether these extracts can cause cell cycle arrest. 4. **Changes in mitochondrial membrane potential**: Detect the changes in mitochondrial membrane potential (\(\Delta \psi_m\)). 5. **Generation of reactive oxygen species (ROS) and endoplasmic reticulum stress (ER Stress)**: Evaluate the effects of these extracts on ROS generation and ER stress. 6. **In vivo experiments**: Verify the anti - tumor effects and safety of AAEE, AAEE - Pe and AAEE - Ea in the H22 tumor - bearing mouse model. ### Main Findings The research shows that AAEE, AAEE - Pe and AAEE - Ea can significantly inhibit the growth of BEL - 7404 and H22 cells, mainly through the following mechanisms: - **Induction of cell apoptosis**: Through endoplasmic reticulum stress and mitochondria - dependent pathways. - **Cell cycle arrest**: Mainly in the G2/M phase. - **Reduction of mitochondrial membrane potential**: Leading to the release of cytochrome c and activation of caspase - 3, caspase - 9 and PARP cleavage. - **Increase in ROS levels**: Promoting cell apoptosis. - **Up - regulation of CHOP expression**: Indicating that endoplasmic reticulum stress is involved in the induction of cell apoptosis. In addition, the results of in vivo experiments show that AAEE, AAEE - Pe and AAEE - Ea can inhibit the growth of H22 tumors and improve the survival rate of tumor - bearing mice without obvious side effects. In conclusion, this study reveals the potential of Artemisia absinthium L. extracts in anti - liver cancer, providing a theoretical basis for the further development of safe and effective anti - tumor drugs.
