The role and mechanism of abscisic acid in mitigating the adverse impacts of high temperature in Gracilariopsis lemaneiformis

Abstract:The commercial red seaweed Gracilariopsis lemaneiformis is extensively cultivated in China, with an annual production of 348,085 t dry weight in 2019, although high temperatures in the summer limit its production. The phytohormone abscisic acid (ABA) plays a major role in growth, development, and stress resistance in higher plants. However, the role and mechanism of ABA in algae remain obscure. Here we investigated the physio-chemical responses of high-temperature-stressed G. lemaneiformis after the exogenous application of ABA. The results showed that the growth rate and other parameters were decreased under high-temperature stress and were increased after ABA treatment. In particular, 50 μmol L−1 ABA increased the growth rate by 75% relative to the control, and the contents of proline, trehalose, mannitol, and superoxide dismutase activity were increased to some degree, whereas the malondialdehyde content was slightly decreased. By using the transcriptome sequencing technique, 1624 differentially expressed genes (DEGs) were screened after ABA treatment. Among these DEGs, the most significantly enriched pathway was oxidative phosphorylation, followed by the ribosome pathway; large numbers of DEGs were also annotated to the carbon fixation in photosynthetic organism, tricarboxylic acid (TCA) cycle, and pentose phosphate pathways. The aforementioned results indicated that exogenous ABA could mitigate the adverse impacts of high temperature by increasing the antioxidant capacity, eliminating reactive oxygen species, and activating energy metabolism and carbohydrate metabolism. This study provides a theoretical reference for algal cultivation in summer.
