Abstract:The synthetic method of polyester monomer 5-hydroxylevulinic acid(5-HLA) was improved and simplified.Using levulinic acid(LA) derived from biomass as raw material,5-HLA was synthesized via modified bromination and hydrolysis processes.In the bromization reaction of LA,the reaction conditions such as reaction temperature,drop time of Br_2 and reaction medium were optimized.The side product was reused via rearrangement of methyl 3-bromolevulinate to methyl 5-bromolevulinate(5-MBL) and the yield of 5-MBL was thus improved to 45% from an earlier reported value of 30%.Then pure 5-HLA in 30% yield was obtained via a simplified one-step hydrolysis of 5-MBL followed by concentration of the hydrolysate,extraction of 5-HLA in a continuous manner with stirring,and recrystallization in chloroform.Compared with the two-step method,the processing time was shortened and the yield of 5-HLA was improved.

Abstract:5-Hydroxylevulinic acid (5-HLA) was synthesized via modified bromination and hydrolysis using levulinic acid (LA) as starting material. Based on the hydroxyl and the carboxyl groups of the 5-HLA, poly(5-hydroxylevulinic acid), a novel aliphatic polyester, was synthesized by direct polycondensation using SnCl_2·2H_2O as catalyst. In addition, 1,6,9,13-tetraoxadispiro [4.2.4.2]tetradecane-2,10-dione (or altaicadispirolactone), a dimer of the 5-HLA, was synthesized by cyclolactonization catalyzed by BF_3·OEt_2. Altaicadispirolactone is one of the chemical components of Anemone altaica C.A.May, a traditional Chinese medicinal material. Structures of the dimer and the polymer were characterized with GC-MS, 1H NMR, FT-IR, element analysis and GPC, respectively. Two plausible mechanisms for the cyclolactonization reaction were presented.

Abstract:The product of methylaluminoxane（MAO） is available commercially, however, central challenges over poor process control, low product yield, and high safety risks, still remain in its synthesis using large-scale reactors.This review presents several conventional hydrolytic routes for MAO synthesis with different techniques to supply water in the reaction system, and primarily focuses upon the development of continuous flow platforms to synthesize MAO, isobutylaluminoxane（IBAO）, and modified methylaluminoxane（MMAO） using 3D printed functional devices.In particular, those flow platforms introduce monodisperse microdroplets of water to improve the heat and mass transfer, achieving inherently safe productions of alkylaluminoxanes with high yield and superior co-catalytic activity. The presented flow strategy has a wide array of applications in chemical synthesis, especially when handling fast, highly exothermic, multiphase reactions.

Abstract:5-Aminolevulinic acid (5-ALA) is a non-proteinogenic amino acid essential for synthesizing tetrapyrrole compounds, including heme, chlorophyll, cytochrome, and vitamin B12. As a plant growth regulator, 5-ALA is extensively used in agriculture to enhance crop yield and quality. The complexity and low yield of chemical synthesis methods have led to significant interest in the microbial synthesis of 5-ALA. Advanced strategies, including the: enhancement of precursor and cofactor supply, compartmentalization of key enzymes, product transporters engineering, by-product formation reduction, and biosensor-based dynamic regulation, have been implemented in bacteria for 5-ALA production, significantly advancing its industrialization. This article offers a comprehensive review of recent developments in 5-ALA production using engineered bacteria and presents new insights to propel the field forward.

Abstract:Abstract 5-Aminolevulinic acid (5-ALA), a non-proteinogenic five-carbon amino acid, has received intensive attentions in medicine due to its approval by the US Food and Drug Administration (FDA) for cancer diagnosis and treatment as photodynamic therapy. As chemical synthesis of 5-ALA performed low yield, complicated processes, and high cost, biosynthesis of 5-ALA via C4 (also called Shemin pathway) and C5 pathway related to heme biosynthesis in microorganism equipped more advantages. In C4 pathway, 5-ALA is derived from condensation of succinyl-CoA and glycine by 5-aminolevulic acid synthase (ALAS) with pyridoxal phosphate (PLP) as co-factor in one-step biotransformation. The C5 pathway involves three enzymes comprising glutamyl-tRNA synthetase (GltX), glutamyl-tRNA reductase (HemA), and glutamate-1-semialdehyde aminotransferase (HemL) from α-ketoglutarate in TCA cycle to 5-ALA and heme. In this review, we describe the recent results of 5-ALA production from different genes and microorganisms via genetic and metabolic engineering approaches. The regulation of different chassis is fine-tuned by applying synthetic biology and boosts 5-ALA production eventually. The purification process, challenges, and opportunities of 5-ALA for industrial applications are also summarized.

Abstract:In the present study, an economic and environmentally friendly bi-functional catalyst, Zr-SBA-15, has been prepared to develop a single-step process for synthesis of methyl levulinate (MLA) directly from furfural (FAL) in near-critical methanol without added H2. The presence of Zr in SBA-15, the structure of the Zr-SBA-15 catalyst, and its physical properties have been determined by X-ray diffraction (XRD), N2 adsorption and desorption (N2-BET) and transmission electron microscopy (TEM). The Zr-SBA-15 catalyst has high activity for the production of MLA and good reusability. A possible reaction pathway containing two possible reaction pathways during this single-step synthesis of MLA has been proposed: the FAL has been transformed to β-Methoxy-2-furanethanol (MFE) and 2-Furfuryl methyl ether first, which then have been converted to MLA, and the conversion of FAL to MFE has been much easier and faster than that to 2-FME, which may be the major reaction pathway. The SBA-15 has given high activity for the conversion of FAL to MFE, and the incorporation of Zr has converted the MFE to MLA efficiently. The reaction of FAL to MFE to MLA has been found to be the dominant reaction. Finally, the catalyst has shown good reusability for at least five times without any reduction in the catalytic activity.

Abstract:5-Aminolevulinic acid (5-ALA) is the key precursor for the biosynthesis of tetrapyrrole compounds, with wide applications in medicine, agriculture and other burgeoning fields. Because of its potential applications and disadvantages of chemical synthesis, alternative biotechnological methods have drawn increasing attention. In this review, the recent progress in biosynthetic pathways and regulatory mechanisms of 5-ALA synthesis in biological hosts are summarized. The research progress on 5-ALA biosynthesis via the C4/C5 pathway in microbial cells is emphasized, and the corresponding biotechnological design strategies are highlighted and discussed in detail. In addition, the detection methods and applications of 5-ALA are also reviewed. Finally, perspectives on potential strategies for improving the biosynthesis of 5-ALA and understanding the related mechanisms to further promote its industrial application are conceived and proposed.

Abstract:5-Aminolevulinic acid (ALA), an important cell metabolic intermediate useful for cancer treatments or plant growth regulator, was produced by recombinant Escherichia coli expressing the codon optimized mitochondrial 5-aminolevulinic acid synthase (EC: 2.3.1.37, hem1) from Saccharomyces cerevisiae controlled via the plasmid encoding T7 expression system with a T7 RNA polymerase. When a more efficient autoinduced expression approach free of IPTG was applied, the recombinant containing antibiotic-free stabilized plasmid was able to produce 3.6 g/L extracellular ALA in shake flask studies under optimized temperature. A recombinant E. coli expressing synthesis pathways of poly-3-hydroxybutyrate (PHB) and ALA resulted in coproduction of 43% PHB in the cell dry weights and 1.6 g/L extracellular ALA, leading to further reduction on ALA cost as two products were harvested both intracellularly and extracellularly. This was the first study on coproduction of extracellular ALA and intracellular PHB for improving bioprocessing efficiency. The cost of ALA production could be further reduced by employing a Halomonas spp. TD01 able to grow and produce ALA and PHB under continuous and unsterile conditions even though ALA had the highest titer of only 0.7 g/L at the present time.
Efficient 5-aminolevulinic acid production through reconstructing the metabolic pathway in SDH-deficient Yarrowia lipolytica

Abstract:5-Aminolevulinic acid (5-ALA) is a precursor for the biosynthesis of tetrapyrrole compounds such as chlorophyll a, heme, and vitamin B12, which has been widely used in the fields of agriculture and medicine. 5-ALA can be biosynthesized through C4 pathway that use succinyl-CoA and glycine as precursor, or through C5 pathway that use glutamate as precursor. Yarrowia lipolytica is an important unconventional yeast and has wide applications in metabolic engineering and industrial biotechnology. It possesses strong metabolic flux through tricarboxylic acid cycle, and prominent acid tolerance, which make it an attractive chassis cell for 5-ALA fermentation. However, there are no reports on the 5-ALA production in Y. lipolytica so far. In this study, a series of 5-ALA producing engineered strains were constructed by using succinate dehydrogenase (SDH)-deficient Y. lipolytica. The related genes of C4 and C5 pathway were co-expressed, and the best engineered strain PGC62-IAL (MatA, xpr2-322, axp-2, leu2-270, ura3-302, ΔSdh5::loxP, ΔAch1::loxP, ScPck, ScHemⅠ, StHemA, EcHemL) for efficient ALA production was obtained. Through optimizing culture conditions, the titer of 5-ALA was 1050 mg/L in shake flask fermentation with glycerol as carbon source. Finally, the 5-ALA titer reached 2216.8 mg/L in the fed-batch fermentation, which is the highest 5-ALA titer achieved using yeast globally. Due to accumulation of the by-products including succinic acid and porphyrin compounds, the 5-ALA yield only reached 0.024 g/g glucose. Nevertheless, Y. lipolytica was engineered to produce 5-ALA for the first time, and indicated that SDH-deficient strains can be used as platform for efficient 5-ALA production.

Abstract:5-aminolevulinic acid (ALA) is a non-protein amino acid that has been widely used in the fields of medicine and agriculture. This study aims to engineer the C5 pathway of the ALA biosynthesis in Escherichia coli BL21 to enhance ALA production. The ALA synthase genes gltX , hemA, and hemL were overexpressed in E. coli BL21 to lead to the increase in the production of ALA. The sRNA RyhB was also overexpressed to downregulate the expression of ALA dehydratase to reduce the downstream bioconversion of ALA to porphobilinogen. Next, the gene arcA was knocked out by CRISPR-Cas9 technology to open the TCA cycle to promote the respiratory metabolism of the strain to reduce the feedback inhibition of heme to ALA. The fermentation conditions of the engineered strain were optimized by response surface experiments. The time-course analysis of the ALA production was carried out in a 1 L shake flask. Through these efforts, the production of ALA in engineered strain reached 2953 mg/L in a 1 L shake flask. This study contributes to the industrial production of ALA by the engineered E. coli in the future.

Systematic development of a highly efficient cell factory for 5-aminolevulinic acid production

Abstract:The versatile applications of 5-aminolevulinic acid (5-ALA) across the fields of agriculture, livestock, and medicine necessitate a cost-efficient biomanufacturing process. In this study, we achieved the economic viability of biomanufacturing this compound through a systematic engineering framework. First, we obtained a 5-ALA synthase (ALAS) with superior performance by exploring its natural diversity with divergent evolution. Subsequently, using a genome-scale model, we identified and modified four key targets from distinct pathways in Escherichia coli, resulting in a final enhancement of 5-ALA titers up to 21.82 g/l in a 5-l bioreactor. Furthermore, recognizing that an imbalance of redox equivalents hindered further titer improvement, we developed a dynamic control system that effectively balances redox status and carbon flux. Ultimately, we collaboratively optimized the artificial redox homeostasis system at the transcription level with other cofactors at the feeding level, demonstrating the highest recorded performance to date with a titer of 63.39 g/l for the biomanufacturing of 5-ALA.

Efficient biosynthesis of 5-aminolevulinic acid from glutamate via whole-cell biocatalyst in immobilized engineered Escherichia coli

Abstract:5-Aminolevulinic acid (ALA), a natural plant growth regulator, herbicide, insecticide, and photodynamic therapy drug, has various applications in agriculture and medicine. Currently, ALA mainly relies on chemical synthesis; however, this method is unsustainable and cannot meet the increasing demand because of its low yields. In this study, through phylogenetic analysis and the statistic of gene numbers downstream to hemA , we selected 4 candidate hemA genes with high potential for ALA production from genome sequences of 562 bacterial strains and found that hemA from Burkholderia caryophylli had the best effect on ALA production. Next, we found the match of HemA and homologous glutamyl-tRNA Glu , and further properly overexpressing gltX and tRNA Glu increased glutamyl-tRNA Glu recognition by HemA. Moreover, through the rational combination of gene elements from B. caryophylli ( hemA , tRNA Glu ) and Salmonella typhimurium ( gltX , hemL ), optimizing the gene expression model, as well as E. coli hosts, the efficiency of ALA synthesis through the C5 pathway was significantly enhanced in engineered E. coli BurAt-SalSL1 (T7). Finally, we developed a whole-cell bioconversion system to produce ALA for the first time and 20.89 g ALA was obtained from 42.42 g glutamate with a conversion of 63.51% after six catalytic recycles. Overall, our study provides an alternative route for the green production of ALA.

Abstract:5-Aminolevulinic acid (5-ALA) is a non-proteinogenic amino acid which has involved in heme metabolism of organisms, and has been widely applied in agriculture, and medical fields nowadays. 5-ALA is used in the elimination of pathogens or cancer cells by photodynamic therapy (PDT) owing to the photosensitizer reaction which releases the reactive oxygen species (ROS). Currently, biofabrication of 5-ALA is regarded as the most efficient and eco-friendly approach, but the complicated ingredient of medium causes the nuisance process of purification, resulting in low recovery and high producing cost. In this study, hydrogen chloride, sodium acetate, and ammonia were examined to maximize the recovery of 5-ALA from ion-exchange chromatography (IEC), thus a 92% recovery in 1 M ammonia at pH 9.5 was obtained. Afterward, the activated carbon was used for decolorization to further remove the pigments from the eluent. Four organic solvents, i.e., diethyl ether, methanol, ethanol, and acetone were compared to extract and form 5-ALA precipitation. The purified 5-ALA was verified to eliminate 74% of A549 human lung cancer and 83% of A375 melanoma skin cancer cell. Moreover, Proteus hauseri, Aeromonas hydrophila, Bacillus cereus, and Staphylococcus aureus were killed via anti-microbial PDT with 1% 5-ALA and reached 100% killing rate at optimal condition. With the addition of 0.05% 5-ALA during the culture, the growth of microalgae Chlorella sorokiniana was improved to against a common aquatic pathogen, A. hydrophila. The broad application of 5-ALA was demonstrated in this study for the first time.

Abstract:A gentle and effective method for the photocatalytic dual functionalization of allenes with high regio- and stereoselectivity using a nonmetallic catalyst is described. Inexpensive and easily available sulfinates and TMSN3 were employed as sulfone and azido sources, respectively. The method is characterized by satisfactory substrate compatibility and tolerance toward functional groups. The straightforward initial mechanistic experiments suggested that the reaction could follow a radical pathway. The synthesis of vinylsulfone azide derivatives presented here offers a promising scaffold for the future development of vinyl sulfone-based drugs and functional bioorthogonal reagents.

Abstract:5-Aminolevulinic acid (ALA) was produced aerobically in the dark during growth on glucose by a newly isolated Chlorella sp. When levulinic acid (20 mM), a competitive inhibitor of ALA dehydratase, was added repeatedly to the medium, about 1.5 mM of ALA was produced extracellularly. Glutamate (30 mM) added with levulinic acid (20 mM, given repeatedly) enhanced ALA production up to 1.9 mM, indicating that ALA might be synthesized via the C-5 pathway.

Abstract:In this study, 7 stains of Rhodopseudomonas sp. were selected from 36 photosynthetic bacteria stains storied in our laboratory. Rhodopseudomonas sp. strain 99-28 has the highest 5-aminolevulinic acid(ALA) production ability in these 7 strains. Rhodopseudomonas sp. 99-28 strain was mutated using ultraviolet radiation and a mutant strain L-1, which ALA production is higher than wild strain 99-28 about one times, was obtained. The elements affecting ALA formation of strain 99-28 and L-1 were studied. Under the optimal condition( pH 7.5, supplement of ALA dehydratase(ALAD) inhibitor, levulinic acid(LA) and precursors of ALA synthesis, glycine and succinat, 3000 Ix of light density), ALA formation of mutant L-1 was up to 22.15 mg/L. Strain L-1 was used to treat wastewater to remove COD(Cr) and produce ALA. ALA production was 2.819 mg/L, 1.531 mg/L, 2.166 mg/L, and 2.424 mg/L in monosodium glutamate wastewater(MGW), succotash wastewater(SW), brewage wastewater(BW), and citric acid wastewater(CAW) respectively. More than 90% of COD(Cr) was removed in four kinds of wastewater. When LA, glycin and succinate were supplied, ALA production was dramatically increased, however, COD(Cr) could hardly be removed.
