Abstract:Due to the persistence of ionic liquids (ILs) in aquatic environments, it is necessary to reveal their ecological risk to aquatic organisms. Herein, the biotoxicity of two alkyl-methylimidazolium nitrate ILs ([C10mim]NO3 and [C12mim]NO3) against Scenedesmus obliquus were studied. Results showed that the growth inhibition of S. obliquus increased with increasing concentrations of ILs, maximum values of 94.61% at 4 mg/L of [C10mim]NO3 and 97.34% at 0.8 mg/L of [C12min]NO3 were observed. The fluorescence parameters of photosystem II, such as light quantum yield and electron transfer rate, showed a negative relationship with the exposure dose. [C12mim]NO3 had a more significant effect than [C10mim]NO3. Moreover, the redox homeostasis of algae was disrupted; the accumulation of reactive oxygen species, leading to obvious inhibition of superoxide dismutase and catalase activities was observed. A metabolomic analysis indicated that the contents of most metabolites were reduced significantly, and fructose and galactose decreased significantly by 42.3% and 88.6%, respectively, in the [C10mim]NO3 treatment compared to those in the control. The inhibition of amino acid biosynthesis and glyoxylate and dicarboxylate metabolism explained the more serious biotoxicity of [C12mim]NO3 than that of [C10mim]NO3. This study facilitates a better understanding of the environmental safety and ecological risks of ILs.
Imidazolium-based ionic liquids disrupt saccharomyces cerevisiae cell membrane integrity

Abstract:Ionic liquids (ILs) are interesting chemical compounds that have a wide range of industrial and scientific applications. They have extraordinary properties, such as the tunability of many of their physical properties and, accordingly, their activities; and the ease of synthesis methods. Hence, they became important building blocks in catalysis, extraction, electrochemistry, analytics, biotechnology, etc. This study determined antifungal activities of various imidazolium-based ionic liquids against yeast Saccharomyces cerevisiae via minimum inhibitory concentration (MIC) estimation method. Increasing the length of the alkyl group attached to the imidazolium cation, enhanced the antifungal activity of the ILs, as well as their ability of the disruption of the cell membrane integrity. FTIR studies performed on the S. cerevisiae cells treated with the ILs revealed alterations in the biochemical composition of these cells. Interestingly, the alterations in fatty acid content occurred in parallel with the increase in the activity of the molecules upon the increase in the length of the attached alkyl group. This trend was confirmed by statistical analysis and machine learning methodology. The classification of antifungal activities based on FTIR spectra of S. cerevisiae cells yielded a prediction accuracy of 83%, indicating the pharmacy and medicine industries could benefit from machine learning methodology. Furthermore, synthesized ionic compounds exhibit significant potential for pharmaceutical and medical applications.

Abstract:Progress in the development of biodegradable or biobased ionic liquids (ILs) has led to the design of green compounds for several applications. Herein, four biocompatible dicationic ionic liquids (DILs) with ammonium-phosphonium cations and amino acid anions were synthesized and investigated their environmental impact. The structures of the DILs were confirmed by spectral analyses (1H, 13C and 31P NMR). Furthermore, physicochemical properties such as density, viscosity and refractive index were determined. Water content, bromide content and solubility were thereafter determined as the parameters needed for further studies. Subsequently, their antifeedant activity towards economically important pests of grain in storage warehouses: the granary weevil, the confused flour beetle, and the khapra beetle was examined, showing the dependence on structure. Moreover, selected DILs were investigated for toxicity towards white mustard, Daphnia magna, and Artemia franciscana to specify the environmental impact. These studies were complemented by understand the biodegradation of DILs by bacterial communities derived from soil at the agricultural land. The result was DILs with limited environmental footprints that have great potential for further application studies.

Abstract:Ionic liquids (ILs) are solvents with salt structures. Typically, they contain organic cations (ammonium, imidazolium, pyridinium, piperidinium or pyrrolidinium), and halogen, fluorinated or organic anions. While ILs are considered to be environmentally-friendly compounds, only a few reasons support this claim. This is because of high thermal stability, and negligible pressure at room temperature which makes them non-volatile, therefore preventing the release of ILs into the atmosphere. The expansion of the range of applications of ILs in many chemical industry fields has led to a growing threat of contamination of the aquatic and terrestrial environments by these compounds. As the possibility of the release of ILs into the environment s grow systematically, there is an increasing and urgent obligation to determine their toxic and antimicrobial influence on the environment. Many bioassays were carried out to evaluate the (eco)toxicity and biodegradability of ILs. Most of them have questioned their “green” features as ILs turned out to be toxic towards organisms from varied trophic levels. Therefore, there is a need for a new biodegradable, less toxic “greener” ILs. This review presents the potential risks to the environment linked to the application of ILs. These are the following: cytotoxicity evaluated by the use of human cells, toxicity manifesting in aqueous and terrestrial environments. The studies proving the relation between structures versus toxicity for ILs with special emphasis on directions suitable for designing safer ILs synthesized from renewable sources are also presented. The representants of a new generation of easily biodegradable ILs derivatives of amino acids, sugars, choline, and bicyclic monoterpene moiety are collected. Some benefits of using ILs in medicine, agriculture, and the bio-processing industry are also presented.

Abstract:New ionic liquids (ILs) are continuously introduced involving an increasing number of organic and inorganic ions. Amino acid based ILs (AAILs) represent a specific interest due to their natural origin and, allegedly, low cost. We apply our recently developed force field for imidazolium-based AAILs to investigate structure properties in their aqueous solutions via molecular dynamics (MD) simulations. By reporting cluster analysis, radial distribution functions and spatial distribution functions, we argue that AAIL ions are well dispersed in the aqueous media, irrespective of the AAIL content. Aqueous solutions of AAILs exhibit desirable properties as solvents for chemical engineering. The AAILs in relatively dilute aqueous solutions (10 mol% AAIL) exist as ion pairs, while more concentrated solutions feature certain amount of larger ionic aggregates.

Abstract:The shortage of new antimicrobial drugs and increasing resistance of microbe to antimicrobial agents have been of some concern. The formulation studies of new antibacterial and antifungal agents have been an active research field. Ionic liquids are known as designed liquids with controllable physical/chemical/biological properties and specific functions, which have been attracting considerable interest over recent years. However, no attention has been made towards the preparation of ionic liquids with antimicrobial activities. In this paper, a new class of ionic liquids (ILs) with fumarate anion was synthesized by neutralization of aqueous 1-butyl-3-methylimidazolium hydroxide with equimolar monoester fumarate and characterized using NMR and thermal gravimetric analysis. The ILs are soluble in water and polar organic solvents and also soluble in the common ILs. The antimicrobial activities of the ILs are more active than commercially available potassium sorbate and are greatly affected by the alkyl chain length. The significant antimicrobial properties observed in this research suggest that the ILs may have potential applications in the modern biotechnology.

Abstract:Antimicrobial resistance has become a serious threat to global health. New antimicrobials are thus urgently needed. Ionic liquids (ILs), salts consisting of organic cations and anions with melting points less than 100°C, have been recently found to be promising in antimicrobial field as they may disrupt the bacterial wall and membrane and consequently lead to cell leakage and death. Different types of antimicrobial ILs are introduced in the review, including cationic, polymeric, and anionic ILs. Being the main type of the antimicrobial ILs, the review focuses on the structure and the antimicrobial mechanisms of cationic ILs. The quantitative structure-activity relationship (QSAR) models of the cationic ILs are also included. Increase in alkyl chain length and lipophilicity is beneficial to increase the antimicrobial effects of cationic ILs. Polymeric ILs are homopolymers of monomer ILs or copolymers of ILs and other monomers. They have great potential in the field of antibiotics as they provide stronger antimicrobial effects than the sum of the monomer ILs. Anionic ILs are composed of existing anionic antibiotics and organic cations, being capable to enhance the solubility and bioavailability of the original form. Nonetheless, the medical application of antimicrobial ILs is limited by the toxicity. The structural optimization aided by QSAR model and combination with existing antibiotics may provide a solution to this problem and expand the application range of ILs in antimicrobial field.Graphical Abstract

Abstract:A family of novel ionic liquids with amino acids and their derivatives as cations and environmentally benign materials as anions have been synthesized using easy preparation techniques. The ionic liquids obtained have the same characteristics as conventional imidazolium ionic liquids and the same chiralities as natural amino acids. Thermal stabilities, phase behaviour, viscosities and miscibilities of the representative family members have been investigated, generally showing no difference from conventional ionic liquids. These amino acid ionic liquids may be used as catalysts and "fully green'' solvents in the cycloaddition of cyclopentadiene to methyl acrylate, which is a typical Diels-Alder reaction. This approach to treating amino acids and their derivatives can serve as an alternative to traditional ionic liquids having synthetic chemical components.
Comparative antioxidant status in freshwater fish Carassius auratus exposed to eight imidazolium bromide ionic liquids: a combined experimental and theoretical study

Abstract:Imidazolium bromide ionic liquids such as 1-alkyl-3-methylimidazolium bromides ([AMIm]Br) and 1-alkyl-2,3-dimethylimidazolium bromides ([AMMIm]Br) are common-use organic salts. However, data on comparative toxicological effects of these ILs are lacking for fish. In this study, a combined experimental and theoretical approach was applied to compare and analyze the effects of these ILs on biochemical biomarkers in liver of Carassius auratus treated with different concentrations (2 and 20mg/L) for 3 and 16d. Changes in the activities of superoxide dismutase, catalase, glutathione peroxidase, and in the levels of reduced glutathione and malondialdehyde were detected, indicating that these ILs exhibit potential biotoxicity. The integrated biomarker response (IBR) index suggested that 1-hexyl-3-methylimidazolium bromide ([HMIm]Br), 1-octyl-3-methylimidazolium bromide ([OMIm]Br), 1-hexyl-2,3-dimethylimidazolium bromide ([HMMIm]Br), and 1-octyl-2,3-dimethylimidazolium bromide ([OMMIm]Br) showed the highest biotoxicity under different concentrations or exposure time, while 1-ethyl-3-methylimidazolium bromide ([EMIm]Br) always showed the least stressful power towards the test organism. Quantum chemical calculations (electronic parameters, frontier molecular orbitals, and Wiberg bond order) were also conducted to interpret the experimental results. Notably, some descriptors were correlated with the toxicity order. In addition, theoretical calculations provided some valuable information on metabolic pathways of these ILs, which may help to get better understanding on their environmental behavior and fate. In general, the toxicological determination and analysis of these ILs were performed with a combined experimental and theoretical method, which may contribute to the future ecotoxicological studies.

Abstract:Ionic liquids (ILs) are new solvents widely used in many technologies due to their unique and advantageous physicochemical properties. In biotechnological applications, ILs can be used along with microorganisms such as Saccharomyces cerevisiae. Due to the enormous number of ILs that can be synthesized through the combination of different anions and cations, it is necessary to have an easy and quick tool for the preliminary screening of their biocompatibility for being used in biotechnological applications. In this work, the agar well diffusion test was successfully applied as a rapid method to identify toxic/nontoxic ILs toward S. cerevisiae. Sixty-three ILs containing a diverse set of cations and anions were used. Through this methodology, nine fully biocompatible ILs toward S. cerevisiae were identified, including: [Bmim+] [NO3−], [HOPmim+] [NO3−], [Bmim+] [NTf2−], [N8,8,8,1+] [NTf2−], [S2,2,2+] [NTf2−], [EMPyr+] [NTf2−], [BMPi+] [NTf2−], [Moxa+] [MeSO4−] and [Chol+] [H2PO4−]. The analysis of the results also provides preliminary rules to enable the design of biocompatible ILs with S. cerevisiae. In this context, the toxicity was mainly determined by the cation nature although some anions can also display a strong influence on the IL biocompatibility as the bistriflimide anion. Besides, it was observed that an increase in the alkyl chain length of cations, such as imidazolium or pyridinium, involves an increase in the IL toxicity.

Abstract:Ionic liquids (ILs), entirely composed of cations and anions, are liquid solvents at room temperature. They are interesting due to their low vapor pressure, high polarity and thermostability, and also for the possibility to fine-tune their physicochemical properties through modification of the chemical structures of their cations or anions. In recent years, ILs have been widely used in biotechnological fields involving whole-cell biotransformations of biodiesel or biomass, and organic compound synthesis with cells. Research studies in these fields have increased from the past decades and compared to the typical solvents, ILs are the most promising alternative solvents for cell biotransformations. However, there are increasing limitations and new challenges in whole-cell biotransformations with ILs. There is little understanding of the mechanisms of ILs' interactions with cells, and much remains to be clarified. Further investigations are required to overcome the drawbacks of their applications and to broaden their application spectrum. This work mainly reviews the applications of ILs in whole-cell biotransformations, and the possible mechanisms of ILs in microbial cell biotransformation are proposed and discussed.

Abstract:Ionic liquids (ILs) are widely used as reaction and separation media in many technologies due to their unique and advantageous physicochemical properties. Thus, further studies approaching the study of the toxicity of these materials are required. Moreover, they are utilized in devices in which microorganisms such as Shewanella sp . act as biocatalysts. Thus, in this work, the toxicity of 69 ILs on the marine bacterium Shewanella sp . was tested. Specifically, the ILs analyzed were based on the cations imidazolium, pyridinium, pyrrolidinium, piperidinium, morpholinium, oxazolinium, phosphonium, ammonium, and sulfonium, in combination with different anions. The toxicities of this wide group of ILs on Shewanella sp. were determined using two methods: 1) the agar disk-diffusion test and 2) the growth inhibition test in liquid media. The relationship between toxicity and IL chemical structure was elucidated. A decrease in the hydrophobicity of the alkyl chain length was found to be a key factor to reduce IL toxicity. On the other hand, phosphonium-based ILs containing long alkyl chains were shown to be largely incompatible with Shewanella sp .

Rheological properties of water-based amino acid ionic liquids

Abstract:The rheological properties and zero-shear viscosity of water-based lubricants (WBLs) containing amino acid ionic liquids (AAILs) were investigated using both equilibrium (EMD) and non-equilibrium molecular dynamics (NEMD) simulations. We also performed experimental measurements to validate the simulations. The simulations demonstrated that adding AAIL additives to water increased both the shear viscosity and zero-shear viscosity. We added tetrabutylphosphonium (P4444) as a cation and three different amino acids, serine (Ser), lysine (Lys), or phenylalanine (Phe), as anions. We varied the AAIL concentration from 5 to 10 wt. % for tetrabutylphosphonium-serine (P4444-Ser) ionic liquid additives, showing that AAILs increased water viscosity by 68%–125%, depending on concentration. The P4444-Ser WBLs also exhibited a significantly higher first normal stress difference than water, meaning they could support more load in lubrication. The improved rheology persisted over a wide range of shear rates up to ∼1011 s−1. We have extracted full rheological parameters by fitting data into Cross, Carreau–Yasuda, and Eyring models, including zero-shear viscosity and critical shear rates of onset shear thinning. The experimental values of zero-shear viscosity were close to zero-shear viscosity results obtained from fitting NEMD results to these models, demonstrating the high fidelity of the molecular model. We explored the formation of hydration shells around amino acid anions as a marker for low friction behavior. These findings suggest that AAIL WBLs can be potentially low-friction and biodegradable lubricants in tribological applications.

Biodegradability of imidazolium and pyridinium ionic liquids by an activated sludge microbial community

Abstract:Ionic liquids (ILs) are novel organic salts that have enormous potential for industrial use as green replacements for harmful volatile organic solvents. Varying the cationic components can alter the chemical and physical properties of ILs, including solubility, to suit a variety of industrial processes. However, to complement designer engineering, it is crucial to proactively characterize the biological impacts of new chemicals, in order to fully define them as environmentally friendly. Before introduction of ILs into the environment, we performed an analysis of the biodegradability of six ILs by activated sludge microorganisms collected from the South Bend, Indiana wastewater treatment plant. We examined biodegradability of 1-butyl, 1-hexyl and 1-octyl derivatives of 3-methyl-imidazolium and 3-methyl-pyridinium bromide compounds using the standard Organisation for Economic Cooperation and Development dissolved organic carbon Die-Away Test, changes in total dissolved nitrogen concentrations, and 1H-nuclear magnetic resonance analysis of initial and final chemical structures. Further, we examined microbial community profiles throughout the incubation period using denaturing gradient gel electrophoresis (DNA-PCR-DGGE). Our results suggest that hexyl and octyl substituted pyridinium-based ILs can be fully mineralized, but that imidazolium-based ILs are only partially mineralized. Butyl substituted ILs with either cation, were not biodegradable. Biodegradation rates also increase with longer alkyl chain length, which may be related to enhanced selection of a microbial community. Finally, DGGE analysis suggests that certain microorganisms are enriched by ILs used as a carbon source. Based on these results, we suggest that further IL design and synthesis include pyridinium cations and longer alkyl substitutions for rapid biodegradability.
Cytotoxicity and cell membrane interactions of choline-based ionic liquids: comparing amino acids, acetate, and geranate anions

Abstract:Ionic liquids (ILs) have found diverse applications in research and industry. Biocompatible ILs, a subset considered less toxic than traditional ILs, have expanded their applications into biomedical fields. However, there is limited understanding of the toxicity profiles, safe concentrations, and underlying factors driving their toxicity. In this study, we investigated the cytotoxicity of 13 choline-based ILs using four different cell lines: Human dermal fibroblasts (HDF), epidermoid carcinoma cells (A431), cervical cancer cells (HeLa), and gastric cancer cells (AGS). Additionally, we explored the haemolytic activity of these ILs. Our findings showed that the cytotoxic and haemolytic activities of ILs can be attributed to the hydrophobicity of the anions and the pH of the IL solutions. Furthermore, utilising quartz crystal microbalance with dissipation (QCM-D) technique, we delved into the interaction of selected ILs, including choline acetate [Cho][Ac] and choline geranate [Cho][Ge], with model cell membranes composed of 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC). The QCM-D data showed that ILs with higher toxicities exhibited more pronounced interactions with membranes. Increased variations in frequency and dissipation reflected substantial changes in membrane fluidity and mass following the addition of the more toxic ILs. Furthermore, total internal reflection fluorescence microscopy study revealed that [Cho][Ac] could cause lipid rearrangements and pore formation in the membrane, while [Cho][Ge] disrupted the bilayer packing. This study advances our understanding of the cellular toxicities associated with choline-based ILs and provides valuable insights into their mechanisms of action concerning IL-membrane interactions. These findings have significant implications for the safe and informed utilisation of biocompatible ILs in the realm of drug delivery and biotechnology.

That is why aggregation of ILs in solutions has been in focus of many studies. Amino acid ionic liquids (AAIL) are considered a new, more eco-friendly type of IL. The aggregation behavior of the newly synthesized 1-decyl-3-methylimidazolium-based AAILs [C 10 mim] [AA] (AA = L-serine, L-threonine, L-lysine) in aqueous solutions was studied by means of densitometry at different temperatures (288.15–323.15) K and by conductometry at 298.15 K. Moreover, the viscosity of aqueous solutions of AAILs at 1.18 mol·kg −1 was determined in the temperature range (293.15–323.15) K. For comparison of the estimated characteristics, 1-decyl-3-methylimidazolium ILs with halogen anions, [C 10 mim]Br and [C 10 mim]Cl, were studied. The aggregation behavior of AAILs in aqueous solutions was discussed based on the data on critical micelle concentration ( cmc ) and degree of counter-ion binding; the values of standard Gibbs energy of micellization, the apparent molar volume at the cmc , apparent molar volume in the micellar region, and the change in the apparent molar volume upon micellization were derived. The obtained results were further analyzed to understand the specificity of the self-aggregation behavior of AAILs and to discuss this effect in terms of solute–solute and solute–solvent interactions. These results can open up opportunities for potential design of new surfactants.

Abstract:Ionic liquids (ILs) are salt-like organic compounds that are composed of organic cations and organic/inorganic anions. The constituting ions play a crucial role in dictating their physicochemical properties and biological activities. ILs since their discovery have been explored for a variety of applications and they prove to be an excellent category of molecules which can be used in almost all sectors of research and applications. However, from the last few decades, the toxicity of ILs has also become a hot topic as they have been found to induce harmful impacts on all forms of life including marine life and the life on earth. There has been an ongoing effort to understand how the ILs and the structural features of ions induce toxicity and which structural parameters should be introduced or changed in order to design environment benign and greener ILs. The interaction of ILs with cell membranes is the most accepted mechanism which leads to the toxic character of the ILs. This review comprises the journey of ILs from their discovery to the times when they were considered a boon and now when their toxic impacts are well known. An overview of the studies is provided which have been carried on to study the toxic behavior of ILs towards a variety of living beings including all variety of plants, living organisms, and cell lines. The structural features of ions are discussed which impact these adverse impacts shown by the ILs. Moreover, this review focuses on the discussion of various techniques and studies which have been used to study and understand the interaction of ILs with lipid membranes. The studies have shown that the toxicity of ILs can be estimated from the interaction of ILs with membranes. ILs turn out to be more toxic when the hydrophobicity of the ions is increased. This review will be helpful to the readers to get a better insight into the toxicity and ILs-membrane interactions along with a scope to understand how the structural features of ions can be varied to design greener ILs.

Abstract:Evaluation of toxicity of ionic liquids has been less prioritized, compared to the development of new ionic liquids and their applications. Focus on the toxicity of ionic liquids should be enhanced, since ionic liquids have been intensively industrialized for decades. Their toxicity is related to not only biological applications but also others, since accidents, such as leakage, can occur at any time. The current review aimed to systematically summarize the general trend between chemical structure and toxicity of ionic liquids across biological species. It also provides a perspective on the current strategies for the development of low-toxicity ionic liquids and their applications.

Abstract:The non-volatility character of the ionic liquids makes them environmentally attractive to new separation processes development, but their water solubility emphasizes the importance of the study of their impact on the aquatic environment. The present work reports the toxicity of 24 imidazolium- and pyridinium-based ionic liquids through Vibrio fischeri inhibition basic test. The toxicity-structure relationship of the ionic liquids has been studied through the anion and cation core, the cation alkyl chain length, and the presence of functional groups on the cation alkyl chain. Ionic liquids whose chemical structure includes pyridinium cations, long cation alkyl chains, and bis(trifluoromethylsulfonyl) imide ([Tf2N]) anions present the highest toxicity. Finally, considering ionic liquids as potential solvents for the aromatic/aliphatic separation process, their toxicity has been compared to that of sulfolane, which is the current organic solvent used in this kind of process. Most of the studied ionic liquids can be classified as harmless or practically harmless; meanwhile, only four of them present a slightly or moderately toxicity, the same than that of sulfolane. The present study suggests that not only the atmospheric pollution would be decreased, but the aquatic impact as well by using ionic liquids as alternative solvents.Graphic abstractIonic liquids (ILs) stand as more environmentally friendly solvents in the petrochemical industry. Their toxicity towards Vibrio fischeri bacteria is lower than most of the “green” alternative deep eutectic solvents (DESs) and much lower than the current solvent, sulfolane.
