Abstract:Nicotinamide adenine dinucleotide (NADH) and nicotinamide adenine dinucleotide phosphate (NADPH), as two kinds of well-known cofactor, are widely used in the most of enzymatic redox reactions, playing an important role in industrial catalysis. In general, supply of NAD(P)H is a major challenged factor in redox fermentation systems due to its high cost and low stability, which have stimulated the development of NADH regeneration systems in recent years. Until now, a series of NAD(P)H regeneration systems have been developed. This review focuses primarily on new approaches of NAD(P)H cofactor regeneration in the biosynthesis systems, such as single cell in vivo NADH regeneration system, double cell coupling NADH regeneration system, in vitro enzyme-coupled NADH regeneration system, microbial cell surface display NADH regeneration system. Finally, the prospect and tendency of NADH regeneration are discussed.

Abstract:Recently, photobiocatalysis with oxidoreductases, inspired by natural photosynthesis, has drawn increasing attention due to its high catalytic efficiency and selectivity. However, although an important cofactor for the activation of redox enzymes, nicotinamide adenine dinucleotide (NADH) suffers from the drawbacks of cost and limited stability. Moreover, the design of an effective NADH regeneration system remains a huge challenge. Here, we report a visible light-driven conjugated microporous polymer DBTS-CMP1 for the heterogeneous photocatalytic regeneration of NADH. Thanks to various improved properties, such as extended visible light absorption, adequate fluorescence lifetime, enhanced wettability, and rapid charge separation and transfer, an NADH regeneration efficiency of 84% in 45 min was achieved with DBTS-CMP1. In addition, the small molecule model compound DBTS-Ph2, which shares similar structural motifs, was also designed to further study the photoinduced electron transfer process. The strong coordination interaction between dibenzo-[b,d]thiophene sulfone and the Rh metal center, as reflected in fluorescence quenching, cyclic voltammetry, and Fourier-transform infrared spectroscopy measurements, plays an essential role in electron transfer from the photocatalyst to the Rh complex, thus endowing DBTS-CMP1 with a high reaction conversion and selectivity (100%) for 1,4-NADH regeneration. Finally, a photobiocatalytic system was constructed by incorporating NADH-dependent alcohol dehydrogenase for the reduction of formaldehyde into methanol. Abstract:Photocatalytic regeneration of valuable cofactors by using sunlight has emerged as a promising strategy for biosynthesis and pharmaceutical manufacturing. Graphitic carbon nitride (g-C3N4) is very suitable for photocatalytic nicotinamide cofactor regeneration since it is metal-free, visible-light responsive and has strong binding with nicotinamide cofactor. However, its great potential is hindered by some intrinsic drawbacks such as low visible absorption, fast electron/hole recombination, and limited active sites. Here, we demonstrate dual-defect g-C3N4 (DDCN) with controllable defects of nitrogen vacancies and cyano groups for efficient photocatalytic cofactor regeneration via a KOH-assisted thermal polymerization by using urea as a precursor. Although DDCN is widely used for other photocatalytic applications such as organic degradation and hydrogen peroxide production, this work is original in its application to photocatalytic cofactor regeneration. Material characterizations confirm the successful introduction of nitrogen vacancies and cyano groups. Measurements of nicotinamide-cofactor generation show that the DDCN samples assisted with 0.1 g and 0.01 g KOH are 3.0 and 2.5 times that of pristine g-C3N4 in terms of nicotinamide cofactor yield, respectively. The high yields are attributed to the synergetic effect of both enhanced light absorption and improved charge separation, achieved through the introduction of energy levels and trap states via dual defects. This work provides a green, energy-saving, and promising strategy for nicotinamide cofactor regeneration and would promote its application in biosynthesis and drug manufacturing.

Abstract:A nicotinamide adenine dinucleotide phosphate (NADPH)-mediated photoenzyme-coupled catalytic system is an attractive and green strategy for chemical conversion. However, artificial photocatalytic NADPH regeneration with low activity and selectivity has posed a challenge when it is coupled with enzyme catalysis. Therefore, we developed a strategy to prepare an "all-in-one" photocatalyst (Rh-sp(2)c-COF) by immobilizing an electron mediator ([Cp*Rh(bpy)(H2O)](2+)) in a covalent organic framework (COF) with a donor-pi-acceptor structure for efficient photocatalytic NADPH regeneration. The immobilized [Cp*Rh(bpy)(H2O)](2+) not only achieves specific regeneration of 1,4-NADPH but also improves the efficiency of electron transfer and utilization by constructing intramolecular electron transfer channels and electron buffer tanks. In addition, the donor-pi-acceptor structure expands the range of light absorption and promotes effective carrier separation and directional transfer. The turnover frequency of NADPH regeneration by Rh-sp(2)c-COF reaches 2.17 mmol center dot g(COF)(-1)center dot h(-1), which is 3.28, 1.99, and 4.5 times higher than those of sp(2)c-COF, sp(2)c-COF + free [Cp*Rh(bpy)(H2O)](2+), and imine-COF, respectively. Finally, Rh-sp(2)c-COF coupled with ene reductase achieves efficient asymmetric hydrogenation of C=C. This work provides an approach for accurate predesign of photocatalysts to achieve efficient conversion and utilization of solar energy.

Abstract:Nicotinamide adenine dinucleotide (NAD+) is an essential metabolite that plays a crucial role in diverse biological processes, including energy metabolism, gene expression, DNA repair, and mitochondrial function. An aberrant NAD+ level mediates the development of cardiovascular dysfunction and diseases. Both in vivo and in vitro studies have demonstrated that nicotinamide mononucleotide (NMN), as a NAD+ precursor, alleviates the development of cardiovascular diseases such as heart failure, atherosclerosis, and myocardial ischemia/reperfusion injury. Importantly, NMN has suggested pharmacological activities mostly through its involvement in NAD+ biosynthesis. Several clinical studies have been conducted to investigate the efficacy and safety of NMN supplementation, indicating its potential role in cardiovascular protection without significant adverse effects. In this review, we systematically summarize the impact of NMN as a nutraceutical and potential therapeutic drug on cardiovascular diseases and emphasize the correlation between NMN supplementation and cardiovascular protection.

Abstract:For industrial enzymatic catalysis, efficient regeneration of nicotinamide adenine dinucleotide (NADH) is necessary. Herein, hollow resorcinol-formaldehyde resin nanospheres loaded with Au nanoparticles (HRF-Au) with shape-controllable nanostructures for photocatalysis were successfully prepared via the hard template method and photodeposition. As a novel photocatalyst, HRF-Au efficiently achieved a high NADH yield of 80.39% with a total reaction rate of 0.6 mmol·gcatalyst –1·h–1 under visible light illumination, which was four times higher than pure resorcinol–formaldehyde resin nanospheres (RF). Also, the photocatalytic system achieved high operational stability and faster rates of NADH production to the partial traditional enzymatic regeneration systems. This exceedingly good activity is partly due to the improvement of photoelectric properties of HRF-Au, which was confirmed by a series of characterization. More importantly, by finite element simulations and energy band structure determination, the hollow sphere structure and localized surface plasmon resonance effect induces the bending of the RF energy band to form a Schottky junction. These were conducive to the generation, separation, and migration of carriers, so as to improve the performance of photocatalytic regeneration of NADH. Our study will contribute to the development of materials with efficient photocatalytic NADH regeneration performance. Meanwhile, the photocatalytic NADH regeneration system could operate continuously and stably, and the significant reduction in byproducts and costs make this sustainable green technology have great potential in the future under both ecological and economical concerns.

Abstract:This article reported the highly selective light‐harvesting metal‐free photocatalyst formed by functionalizing native graphitic carbon nitride with BF3.OEt2, which enables the doping of boron and fluorine in the native structure. The photocatalytic performance increases by increasing the concentration of dopants. The formation of the BFGCN‐x photocatalyst was confirmed via various characterizations like UV‐DRS, FT‐IR, X‐ray diffraction spectroscopy (XRD), CV, electrochemical studies, SEM, and EDX. The synthesized BFGCN‐x photocatalyst was capable of regenerating NAD(P)H from NAD(P)+ cofactor and also has capability in forming acetal compounds, confirmed by gas chromatography and nuclear magnetic resonance spectroscopy. Nicotinamide Adenine Dinucleotide Phosphate (NAD(P)H) plays an important role in numerous biologically significant redox reactions. The photochemical restoration of its oxidized form (NAD(P)+) under physiological conditions is intriguing in the context of integrated photo and catalysis. Herein, we report the functionalized graphitic carbon‐based solar light active photocatalyst by doping boron and fluorine in the native graphitic carbon nitride (GCN) (nonfunctionalized) for the regeneration of enzymatically visible light active coenzyme and in photo‐acetalization reactions. The metal‐free functionalized photocatalyst systems such as BFGCN‐x leads to higher yield NADH and NADPH regeneration. They are also capable of catalyzing acetal reactions in the absence of any Lewis and Bronsted acids. The current research endeavor provides the advancement and the application of functionalized GCN‐based photocatalysts for NADH (61.89%), NADPH (59.84%) regeneration, and photo‐acetalization reactions.

Abstract:The integration of titanium dioxide with gold nanoparticles led to a 12‐fold enhancement in the plasmonically driven regeneration of nicotinamide cofactor, by following a less‐explored light‐independent pathway. Photocatalysis with plasmonic nanoparticles (NPs) is emerging as an attractive strategy to make and break chemical bonds. However, the fast relaxation dynamics of the photoexcited charge carriers in plasmonic NPs often result in poor yields. The separation and extraction of photoexcited hot‐charge carriers should be faster than the thermalization process to overcome the limitation of poor yield. This demands the integration of rationally chosen materials to construct hybrid plasmonic photocatalysts. In this work, the enhanced photocatalytic activity of gold nanoparticle‐titanium dioxide metal‐semiconductor heterostructure (Au‐TiO2) is used for the efficient regeneration of nicotinamide (NADH) cofactors. The modification of plasmonic AuNPs with n‐type TiO2 semiconductor enhanced the charge separation process, because of the Schottky barrier formed at the Au–TiO2 heterojunction. This led to a 12‐fold increment in the photocatalytic activity of plasmonic AuNP in regenerating NADH cofactor. Detailed mechanistic studies revealed that Au‐TiO2 hybrid photocatalyst followed a less‐explored light‐independent pathway, in comparison to the conventional light‐dependent path followed by sole AuNP photocatalyst. NADH regeneration yield reached ~70% in the light‐independent pathway, under optimized conditions. Thus, our study emphasizes the rational choice of components in hybrid nanostructures in dictating the photocatalytic activity and the underlying reaction mechanism in plasmon‐powered chemical transformations.

Abstract:Nickel hexacyanoferrate films have been deposited on the surface of a microband gold electrode by cyclic voltammetry. Cyclic voltammograms of the resulting nickel hexacyanoferrate films indicate the presence of a couple of redox peaks which correspond to the hexacyanoferrate(II/III) redox couple. The nickel hexacyanoferrate modified microband gold electrode shows an excellent electrocatalytic activity for oxidation of reduced nicotinamide adenine dinucleotide (NADH) in phosphate buffer solution (pH 7.0), with an overpotential ca. 340 mV lower than that of the bare electrode. The steady-state current increases linearly with NADH concentration in the range of 0.5–8.0 mmol/l. The catalytic rate constant of the modified electrode for the oxidation of NADH is determined using a rotating disk electrode. The further experimental results show that the rate constant decreases with increasing NADH concentration. The mechanism of the oxidation of NADH catalyzed by the nickel hexacyanoferrate modified gold electrode is discussed.
One-Pot Highly Efficient Synthesis of N-Enrich Graphene Quantum Dots as a Photocatalytic Platform for NAD+/NADP+ Reduction

Abstract:An efficient photocatalytic regeneration of nicotinamide adenine dinucleotide (NADH) and nicotinamide adenine dinucleotide phosphate (NADPH) has been carried out by two-electron reduction and protonation of NAD+ /NADP+ , induced by photons in the visible light region. This functional artificial photosynthetic counterpart of the complete energy-trapping occurring in natural photosystem I (PS I) is achieved with nitrogen-enrich graphene quantum dot (N-EGQD) as the light-harvesting photocatalyst. In buffer aqueous solution, this compound photo catalytically recycles a rhodium hydride complex of the type [Cp*Rh(bpy)H]+ (Cp* = pentamethylcyclopentadienyl, bpy = 2,2'-bipyridine) which can mediate hydride transfer processes leading to nucleotide co-factor reduction. Very promising yields of 73.31%/78.45% of NADH/NADPH with the excellent thermal stability of N-EGQD photocatalyst is observed. Thus, in this work, an efficient light-harvesting photocatalyst is synthesized for the regeneration of nicotinamide cofactor that has pharmaceuticals application.
A Prebiotic Pathway to Nicotinamide Adenine Dinucleotide

Abstract:Enzymes play a fundamental role in cellular metabolism. A wide range of enzymes require the presence of complementary coenzymes and cofactors to function properly. While coenzymes are believed to have been part of the last universal ancestor (LUCA) or have been present even earlier, the syntheses of crucial coenzymes like the redox‐active coenzymes flavin adenine dinucleotide (FAD) or nicotinamide adenine dinucleotide (NAD+) remain challenging. Here, we present a pathway to NAD+ under prebiotic conditions starting with ammonia, cyanoacetaldehyde, prop‐2‐ynal and sugar‐forming precursors, yielding in situ the nicotinamide riboside. Regioselective phosphorylation and water stable light activated adenosine monophosphate derivatives allow for topographically and irradiation‐controlled formation of NAD+. Our findings indicate that NAD+, a coenzyme vital to life, can be formed non‐enzymatically from simple organic feedstock molecules via photocatalytic activation under prebiotically plausible early Earth conditions in a continuous process under aqueous conditions.

Photoenzymatic Catalytic Cascade System of a Pyromellitic Diimide/g-C 3 N 4 Heterojunction to Efficiently Regenerate NADH for Highly Selective CO 2 Reduction toward Formic Acid

Abstract:Photocatalytic reduction of carbon dioxide (CO2) holds great promise for both clean energy and environment protection. However, the low activity and poor selectivity of photocatalysts are the main bottlenecks. Herein, inspired by artificial photosynthesis and taking advantages of high efficiency and specificity of bioenzymes, we marry photo with enzyme to synergistically solve the above problems. A metal-free heterojunction of pyromellitic diimide/g-C3N4 (PDI/CN) with an excellent visible light response (λ < 660 nm) is fabricated for achieving a photoenzymatic catalytic cascade system, which efficiently regenerates nicotinamide adenine dinucleotide (NADH) and selectively reduces CO2 to formic acid (HCOOH). The highest NADH yield of the PDI/CN hybrid achieved is 75%, and the HCOOH generation rate achieved is 1.269 mmol g–1 h–1 with nearly 100% selectivity, which is much higher than those of the reported materials. The excellent photocatalytic performance is attributed to the unique photoenzymatic catalytic cascade system, heterointerface effect, good conductivity, and a wide sunlight response range of the PDI/CN heterojunction. Abstract:In this work, catechol violet modified graphite electrode shows significantly electrocatalytic activity to the oxidation of reduced nicotinamide adenine dinucleotide (NADH), making oxidation overpotential decrease by 250 mV compared with the direct oxidation of NADH at bare graphite electrode, In the medium of 0.1 mol/L phosphate buffer (pH= 7.0) the surface formal potential E(0') and apparent electron-transfer rate of the adsorbed mediator catechol violet are 0.10 V and 5.5 s(-1), respectively.
Elucidating the Intimate Mechanism of NAD+ hydrogenation with Phosphonic Acid Catalysed by Cp*Ir(Pyridine-2-sulfonamidate) Complexes

Abstract:The reaction mechanism of nicotine amide dinucleotide hydrogenation (NAD+ to NADH) catalysed by Cp*Ir(2-pyridinesulfonamidate) complexes in the presence of phosphonic acid has been elucidated. Multivariate kinetic experiments and NMR spectroscopy revealed that the enhanced performance of this class of catalysts arise from the hemilability of the pyridine ligand, which is displaced during P-H bond activation and facilitates the generation of hydride intermediates. Experimental results are backed by DFT calculations showcasing the importance of hydrogen bonding interactions in the activation of phosphite anions. The direct comparison between the prototypical unsubstituted catalyst and the 6-aminopyridinesufonamidate derivative allowed to trace the molecular origin of the superior performance of the latter, paving the way for the intelligent design of better performing catalysts for NADH regeneration.

Abstract:Nicotinamide adenine dinucleotide (NAD+) is a redox active molecule that is universally found in biology. Despite the importance and simplicity of this molecule, few reports exist that investigate which molecular features are important for the activity of this ribodinucleotide. By exploiting the nonenzymatic reduction and oxidation of NAD+ by pyruvate and methylene blue, respectively, we were able to identify key molecular features necessary for the intrinsic activity of NAD+ through kinetic analysis. Such features may explain how NAD+ could have been selected early during the emergence of life. Simpler molecules, such as nicotinamide, that lack an anomeric carbon are incapable of accepting electrons from pyruvate. The phosphate moiety inhibits activity in the absence of metal ions but facilitates activity at physiological pH and model prebiotic conditions by recruiting catalytic Mg2+. Reduction proceeds through consecutive single electron transfer events. Of the derivatives tested, including nicotinamide mononucleotide, nicotinamide riboside, 3‐(aminocarbonyl)‐1‐(2,3‐dihydroxypropyl)pyridinium, 1‐methylnicotinamide, and nicotinamide, only NAD+ and nicotinamide mononucleotide would be capable of efficiently accepting and donating electrons from and to pyruvate within a nonenzymatic electron transport chain. The data are consistent with early metabolic chemistry exploiting NAD+ or nicotinamide mononucleotide and not simpler molecules.

Abstract:Nicotinamide, the amide form of vitamin B(3) (niacin), is changed to its mononucleotide compound with the enzyme nicotinic acide/nicotinamide adenylyltransferase, and participates in the cellular energy metabolism that directly impacts normal physiology. However, nicotinamide also influences oxidative stress and modulates multiple pathways tied to both cellular survival and death. During disorders that include immune system dysfunction, diabetes, and aging-related diseases, nicotinamide is a robust cytoprotectant that blocks cellular inflammatory cell activation, early apoptotic phosphatidylserine exposure, and late nuclear DNA degradation. Nicotinamide relies upon unique cellular pathways that involve forkhead transcription factors, sirtuins, protein kinase B (Akt), Bad, caspases, and poly (ADP-ribose) polymerase that may offer a fine line with determining cellular longevity, cell survival, and unwanted cancer progression. If one is cognizant of the these considerations, it becomes evident that nicotinamide holds great potential for multiple disease entities, but the development of new therapeutic strategies rests heavily upon the elucidation of the novel cellular pathways that nicotinamide closely governs.

Abstract:Here we report a new robust nicotinamide dinucleotide phosphate cofactor analogue (carba‐NADP+) and its acceptance by many enzymes in the class of oxidoreductases. Replacing one ribose oxygen with a methylene group of the natural NADP+ was found to enhance stability dramatically. Decomposition experiments at moderate and high temperatures with the cofactors showed a drastic increase in half‐life time at elevated temperatures, since it significantly disfavors hydrolysis of the pyridinium‐N‐glycoside bond. Overall, more than 27 different oxidoreductases were successfully tested, and a thorough analytical characterization and comparison is given. The cofactor carba‐NADP+ opens up the field of redox‐biocatalysis under harsh conditions.
