Genome-Wide Analysis of LAZ1 Gene Family from Maize

Abstract:Lazarus 1 (LAZ1) is a six transmembrane protein with DUF300 domain, and functions as organic solute transporter in vertebrates. However, there was no any available literature on genome-wide identification of the LAZ1 gene family in plant. In this study, nine members of the ZmLAZ1 genes were identified from the maize genome, and characterized for their chromosomal locations, gene duplication, gene structure, cis-acting elements, and expression pattern in different organs and developmental stages. Except motif 1, which forms the DUF300 domain and functions as transmembrane organic solute transporter, was highly conserved among all the ZmLAZ1 members, other six motifs were shared by the ZmLAZ1 members within the same phylogenetic clade. Except ZmLAZ1-6, other eight open reading frames (ORF) of ZmLAZ1s were experimentally cloned and sequenced. The results of bioinformatics prediction and laboratorial transient expression showed that the ZmLAZ1 is localized in the nucleus, cytoplasm, cytoplasm plasmalemma, as well as different organelles. The expression of the ZmLAZ1 genes in root and leaf was detected by real-time quantitative PCR. Their relative expression levels showed abundant diversity among different organs and development stages, and in response to abscisic acid, simulative drought and NaCl treatments. This might be partially explained by the abundant diversity of cis-acting elements in the promoter of the eight ZmLAZ1 s. The above results demonstrated that the ZmLZA1 protein function as transmembrane organic solute transporters in various pathways and organs, tissues or organelles at different development stages.
