Abstract:Background & objectives: The main aim of this research is to provide literature on the Ocimum plant, and to know the significance of the Ocimum species carried out by pharmacognostic study and experimental design for GC-MS. Ocimum genus are very important for their therapeutic potential among the most important aromatic herbs. Methods: Extreme attention has been put on literature reports in which the utilization of tulsi and their pharmacognostic study has been done by performing morphological and microscopic leaf experimental design and by using essential oil through the GC-MS instrumentation method. Results: The utilization of these characteristics would be important for the drug discovery scientist to develop a specific formulation of the crude drug, which will be a magical therapeutic agent in the future, with many advantages. GC-MS chromatogram of Ocimum sanctum, Ocimum canum, and Ocimum gratissimum oil showed major peaks and has been identified after comparison of the mass spectra with the NIST library, indicating the presence of three phytocomponents. From the results, the GC-MS study suggested that anethole which is well reported antimicrobial compound is more in O. canum (2.66%) in comparison to O. sanctum (1,28%) but absent in O. gratissimum. The results indicated that the antimicrobial activity is more in O. canum due to the presence ofa high amount of anethole in comparison to O. gratissimum and O. sanctum. Interpretation & conclusion: The result revealed that O. canum has a microscopic character that can be identified by the characteristic GC MS analysis of extracts to distinguish between different species of the ocimum plant.

Abstract:In recent years, the growing demand for herbal-based formulations, including functional foods, has acquired significant attention. This study highlights historical, botanical, ecological, and phytochemical descriptions and different extraction mechanisms of Ocimum sanctum utilized in its processing. Besides this, it explores the utilization of Ocimum sanctum as a functional food ingredient in various food products such as bakery products (biscuits, bread), dairy products (herbal milk, cheese), and beverages (tea, juice, wine) while focusing on their evaluation parameters, preparation techniques, and pharmacological activities. In terms of other pharmacological properties, Ocimum sanctum-infused functional foods exhibited cognitiveenhancing properties, adaptogenic qualities, anti-obesity effects, gastroprotective, antiinflammatory, hypoglycemic, and immuno-modulatory effects. Thus, the diverse properties of Ocimum sanctum offer exciting opportunities for the development of functional foods that can promote specific health issues, so future research should focus on developing and analyzing novel Ocimum sanctum-based functional foods to meet the growing demand of the functional food industry.

Abstract:Since ancient times, various scientists and doctors have utilized different herbs to heal diseases. Due to the rise in drug resistance and the negative effects of chemosynthetic drugs, researchers and the general public around the world have become more interested in medicinal herbs and plant metabolites/extracts. This is due to its non-toxicity and its several health benefits when used to treat diseases in clinical and medical settings. Ocimum basilicum is one such plant, possessing a wide range of bioactive phytochemicals including alkaloids, phenolics, flavonoids, tannins, saponins, reducing sugars, cardiac glycosides, steroids and glycosides, as well as complex pharmacological activities, including anti-inflammatory, antifungal, antibacterial, antioxidant, wound healing and antiviral properties. The results of many studies on Ocimum basilicum plant extracts are collected and presented in this review. The plant extracts have excellent potential to be used as medicinal raw materials, and exhibit an extensive variety of therapeutic capacities, including antibacterial, antioxidant, wound healing, anti-inflammatory, antifungal, and antiviral properties.

Abstract:BackgroundOcimum sanctum L. (O. tenuiflorum) family-Lamiaceae is an important component of Indian tradition of medicine as well as culture around the world, and hence is known as “Holy basil” in India. This plant is mentioned in the ancient texts of Ayurveda as an “elixir of life” (life saving) herb and worshipped for over 3000 years due to its healing properties. Although used in various ailments, validation of molecules for differential activities is yet to be fully analyzed, as about 80 % of the patents on this plant are on extracts or the plant parts, and mainly focussed on essential oil components. With a view to understand the full metabolic potential of this plant whole nuclear and chloroplast genomes were sequenced for the first time combining the sequence data from 4 libraries and three NGS platforms.ResultsThe saturated draft assembly of the genome was about 386 Mb, along with the plastid genome of 142,245 bp, turning out to be the smallest in Lamiaceae. In addition to SSR markers, 136 proteins were identified as homologous to five important plant genomes. Pathway analysis indicated an abundance of phenylpropanoids in O. sanctum. Phylogenetic analysis for chloroplast proteome placed Salvia miltiorrhiza as the nearest neighbor. Comparison of the chemical compounds and genes availability in O. sanctum and S. miltiorrhiza indicated the potential for the discovery of new active molecules.ConclusionThe genome sequence and annotation of O. sanctum provides new insights into the function of genes and the medicinal nature of the metabolites synthesized in this plant. This information is highly beneficial for mining biosynthetic pathways for important metabolites in related species.

The multifaceted antibacterial properties of tulsi including activity against a wide variety of human and animal pathogens claims that it has several applications, including water purification, mouthwash, sanitizer, and usage in animal husbandry, wound healing, traveler's health, and food preservation. It can also be used to raise animals, treat wounds preserve food and herbal raw materials, and raise animals. Tulsi plant cultivation ties the grower to the generative forces of nature on a spiritual and practical level, and organic agriculture provides answers for issues like food security, hunger, environmental, rural poverty, climate change, and degradation, The usage of tulsi in every day routines is a tribute to Ayurvedic expertise and serves as an illustration of how old wisdom may provide answers to contemporary issues. Abstract:The aim of the present study is to determine the minerals, chemical compounds and antibacterial activity of stem and seed extracts of Ocimum Kilimandcharicum plant using X-ray diffraction, Gas Chromatography—Mass Spectroscopy and UV–Visible spectroscopic studies. The potential antimicrobial activity is studied using Bacillus subtilis, Staphylococcus aureus and Pseudomonas aeruginosa . The results of X-ray diffraction showed that the stems and seeds are a rich source of minerals such as oxides of Potassium, Magnesium, Calcium and Chlorine. The GC–MS analysis indicated that the compounds Camphor, Thujopsene-[12] and Eugenol are present in higher amounts and UV–Visible studies showed the existence of Phytochemicals such as chromophores, carotenoids, xanthophylls, aromatic compounds, amines and chlorophyll. The ethanol extract of stems and seeds showed significant anti microbial activity surpassing the positive control streptomycin. The presence of potential bioactive compounds, minerals and its significant antimicrobial activity justifies the use of stems and seeds of the plant for curing infectious diseases and can be used as a mineral supplement.

Fungal Endophyte Diversity and Bioactivity in the Indian Medicinal Plant Ocimum sanctum Linn

Abstract:Endophytic mycopopulation isolated from India's Queen of herbs Tulsi (Ocimum sanctum) were explored and investigated for their diversity and antiphytopathogenic activity against widespread plant pathogens Botrytis cinerea, Sclerotinia sclerotiorum, Rhizoctonia solani and Fusarium oxysporum. 90 fungal isolates, representing 17 genera were recovered from 313 disease-free and surface sterilised plant segments (leaf and stem tissues) from three different geographic locations (Delhi, Hyderabad and Mukteshwar) during distinct sampling times in consequent years 2010 and 2011 in India. Fungal endophytes were subjected to molecular identification based on rDNA ITS sequence analysis. Plant pathogens such as F. verticillioides, B. maydis, C. coarctatum, R. bataticola, Hypoxylon sp., Diaporthe phaseolorum, Alternaria tenuissima and A. alternata have occurred as endophyte only during second sampling (second sampling in 2011) in the present study. Bi-plot generated by principal component analysis suggested tissue specificity of certain fungal endophytes. Dendrogram revealed species abundance as a function of mean temperature of the location at the time of sampling. Shannon diversity in the first collection is highest in Hyderabad leaf tissues (H' = 1.907) whereas in second collection it was highest from leaf tissues of Delhi (H' = 1.846). Mukteshwar (altitude: 7500 feet) reported least isolation rate in second collection. Nearly 23% of the total fungal isolates were considered as potent biocontrol agent. Hexane extract of M. phaseolina recovered from Hyderabad in first collection demonstrated highest activity against S. sclerotiorum with IC50 value of 0.38 mg/ml. Additionally, its components 2H-pyran-2-one, 5,6-dihydro-6-pentyl and palmitic acid, methyl ester as reported by GC-MS Chromatogram upon evaluation for their antiphytopathogenic activity exhibited IC50 value of 1.002 and 0.662 against respectively S. sclerotiorum indicating their significant role in antiphytopathogenic activity of hexane extract. The production of 2H-pyran-2-one, 5,6-dihydro-6-pentyl from M. phaseolina, an endophytic fungus is being reported for the first time.

Abstract:The family Lamiaceae encompasses a diverse array of therapeutic plants, with the genus Ocimum being particularly esteemed. The plant known as Kapur Tulsi in India, Ocimum kilimandscharicum Guerke is a perennial woody shrub characterized by its essential oil, which predominantly contains camphor, along with linalool, limonene, eugenol, 1,8-cineole, and camphene. This review aims to provide current information on the taxonomy, botany, distribution, ethnomedicinal uses, phytochemistry, pharmacology, and toxicological profile of O. kilimandscharicum. The information presented was critically analyzed to understand the current research on this species and to explore potential future opportunities for this plant in pharmaceutical research. A comprehensive literature search was conducted and relevant information was collected through an extensive exploration of bibliographic databases, including Google Scholar, ScienceDirect, PubMed, Web of Science, Semantic Scholar, Europe PMC, and Scopus. The literature suggests that the plant contains a range of compounds that underpin its broad spectrum of pharmacological effects. These phytoconstituents have been widely employed in the treatment and management of numerous conditions. This review consolidates the latest information on its phytochemical and pharmacological activities, traditional applications, and scientific research. The review highlights the significant phytoconstituents of Ocimum kilimandscharicum Guerke and summarizes the evidence supporting its pharmacological potential. The findings suggest that the plant's diverse compounds contribute to its extensive use in traditional medicine and underscore its potential for therapeutic applications in various diseases.

Abstract:In ayurveda, plants have been used for the treatment of so many diseases. Herbal drugs are easily available and have fewer side effects. So, many people are attracted towards the herbal drugs. Platycladus orientalis is one of the useful plant in Indian & Chinese medicine. It is used in treatment of so many diseases like diuretic, anticancer, anticonvulsant, stomachic, antipyretic, analgesic and anthelmintic etc. In Zimbabwe, Mozambique and South Africa Platycladus orientalis is grown as an ornamental. The essential oils extracted from leaves, cones and wood are important in body care products used in the Western world. Platycladus orientalis (Commonly- Morpankhi, Family- Cupressaceae) is an evergreen, monoecious trees or shrubs used in various forms of traditional medicines and homeopathy in various ways. In traditional practices Thuja is used for treatment of bronchial catarrh, enuresis, cystitis, psoriasis, uterine carcinomas, amenorrhea and rheumatism. Recent re-searches in different parts of the world have shown that p. orientalis and its active component thujone have the great potential against a various health problems The medicinal plants are widely used by the traditional medical practitioners for curing various diseases in their day to day practice. In traditional systems of medicine, different parts (leaves, stem, flower, root, seeds and even whole plant) of Ocimum canum Linn (known as kalaTulsi in Hindi), a small herb seen throughout India, have been recommended for the treatment of bronchitis, bronchial asthma, malaria, diarrhea, dysentery, skin diseases, arthritis, painful eye diseases, chronic fever, insect bite etc. The Ocimum canum L. has also been suggested to possess antifertility, anticancer, antidiabetic, antifungal, antimicrobial, hepatoprotective, cardioprotective, antiemetic, antispasmodic, analgesic,adaptogenic and diaphoretic actions.

Abstract:Ocimum tenuiflorum Linn. is an Ayurvedic herb commonly regarded as Tusli or Holy Basil. The plant has a long traditional medicinal history. In India Tulsi is worshipped by Hindus and is abundantly found in all regions. Each part of a plant like stems, roots, seeds, leaves, flowers, and fruits have their own curative properties and functions reported by many researchers. The varieties of active chemical constituents; terpenoids, phenols, flavonoids, phenylpropanoids, fatty acid derivatives, essential oil, fixed oil, and steroids are extracted from the Ocimum tenuiflorum plant and are determined of great therapeutic activities. The therapeutic actions of Tulsi compounds recreate their remedial attention and in the standardization of their medicinal results. Eugenol, Oleanolic Acid, Linalool, Ursolic Acid, Beta-Caryophyllene, Rosemarinic Acid, Estragole, Carvacrol, Methyl Cinnamate are the major compounds which are leads to various actions against diseases. This review article summarizes researchers validated various pharmacological actions of these active chemical constituents that have great potential against, antidiabetic, antioxidant, anti- inflammatory, antimicrobial, antiviral (especially against Covid-19/Coronavirus, helpful in the current scenario of “pandemic”), cardioprotective, antihypertensive, and other activities as well. This will be useful for developing new active principles and therapeutics or new life-saving drugs by researchers, practitioners, and students who were implicated in the field of ethnopharmacology.

Abstract:This study presents an innovative X-ray diffraction (XRD) application to identify minerals in the medicinal plant Ocimum kilimandscharicum . Additionally, this study includes the analysis of chemical compounds and screening antibacterial activity in the leaf extract, using Gas Chromatography-Mass Spectrometry (GC-MS) and UV-visible spectroscopy. The results of X-ray diffraction showed that the leaves are a rich source of minerals such as oxides of Potassium, Magnesium, and Calcium. The GC-MS analysis revealed that the leaf extract of Ocimum kilimandscharicum possesses Eugenol (27.53%), Camphor (16.91%), Thujopsene (13.53%) as major components and UV-visible studies showed the existence of Phytochemicals such as chromophores, carotenoids, and chlorophyll. The ethanol extract of leaves showed antimicrobial activity against four microorganisms. It significantly inhibited Bacillus subtilis , E.Coli, and Staphylococcus aureus, surpassing the positive control streptomycin. These findings suggest the potential of using leaves of Ocimum kilimandscharicum for curing infectious diseases and can be used as a mineral supplement.

Abstract:Ocimum sanctum, also known as Holy Basil, is an aromatic herb with a long history of use in traditional medicine. It is native to tropical and subtropical regions of Asia and is now cultivated worldwide. Plants have erect, much-branched sub-shrub with simple opposite green or purple leaves that are strongly scented and hairy stems and leaves has a strong, characteristic aroma that occurs mainly because of the volatile oils present. In this experiment, 30 genotypes of Ocimum sanctum were evaluated for morphological, yield and quality traits. At nursery stage, germination percentage, seedling shoot length and seedling root lengths were recorded. Among the accessions. OS-22 exhibited the highest germination percentages (82%), Seedling shoot and root length were highest in Os-18 and OS 21 (35 cm and 11 cm) respectively. In the main field, the accessions were characterized for qualitative traits viz growth habit, leaf colour and stem colour. The accessions were grouped into two, based on growth habit. Based on stem and leaf colour the accessions were grouped into five and four groups respectively. In case of quantitative traits, plant height achieved maximum in the accession OS 22 (88cm) followed by OS 17(75cm). The accessions OS-15 and OS-22 exhibited more extensive plants spread, while OS-21 and OS-13 had higher number of primary branches. Accessions OS-20 and O S-23 recorded more number of leaves (409 and 422) and the accessions O S-22 and OS-11 recorded highest fresh leaf weight (370.2 g & 301.67 g). The essential oil recovery and oil yield was highest in the accession OS 20 (green type) followed by OS 22 (purple type). Characterization of oil in GCMS revealed that the key component was eugenol, which showed highest content in accession Os-20 (46 %). This study can be used for further crop improvement studies and breeding programs.

Abstract:Due to the numerous health benefits and therapeutic properties, herbs and plant metabolites/extracts are gaining popularity. This is particularly evident in the current era of drug resistance and the adverse effects of chemical drugs. Ocimum basilicum , also known as basil, has been extensively studied for its pharmacological benefits, including antimicrobial, antifungal, antioxidant, anti-inflammatory, antiviral, and wound healing properties. As a result, this plant has the potential to treat a wide range of diseases in both humans and animals. Ocimum basilicum contains various bioactive chemical compounds, such as neryl acetate, 1,8-cineole, p-allylanisole, geraniol, methyl eugenol, methyl chavicol, and trans-α-bergamotene. The latest advancements in technology can be utilised to enhance the beneficial properties of raw Ocimum basilicum extract. This review compiles and presents the profile of phytocomponents and pharmacological properties of Ocimum basilicum . The findings presented here will contribute to further research on this remarkable herb, aiming to develop effective pharmaceutical solutions for various health issues in humans and animals.

Abstract:Due to adverse effects of synthetic compounds, there is a growing interest in utilization of plant-derived natural products in the pharmaceutical and food industries. Iranian endemic medicinal plants widely used in traditional practice have attracted much attention as antibacterial and antioxidant agents.This review attempts to compile the accessible scientific research pertained to phytochemical compounds, antibacterial and antioxidant effects of essential oils obtained from some of the most widely used and distributed medicinal plants in Iran.This review has been compiled using references via reliable databases (Google Scholar, SID and Science Direct) from 2010 to 2020. This literature review was limited to references published in English and Persian languages.Based on studies heretofore carried out, essential oils isolated from mentioned medicinal plants exhibited strong antioxidant activity which is attributed to their main phytochemical compounds; thymol, carvacrol, p-cymene and γ-terpinene. In addition, the antibacterial activities of essential oils of most plant species from Apiaceae and Asteraceae families were more susceptible against Gram-positive bacteria; Staphylococcus aureus and Bacillus cereus than Gram-negative bacteria; however, essential oils of other studied plant species manifested similar behaviours against both Gram-positive and -negative bacteria.As there is rich ethnobotanical knowledge behind Iranian endemic medicinal plants, further scientific research is required to prove their safety and efficacy. This review revealed that there are numerous valuable medicinal plants adoptable in food and pharmaceutical industries in the near future.

In this context, the aim of this article is to highlight the results of a phytochemical analysis (HPLC), with particular emphasis on the antioxidant (DPPH, TAC, and FRAP), analgesic, anti-inflammatory, haemagglutinin-test-related, and hemolytic activities of the total extracts of these plants. Phytochemical analysis via HPLC revealed that licoflavone C (30%) is the main compound in Origanum grosii, while hesperidin (43%) is found in T. pallidus. Evaluation of the antioxidant capacity of Origanum grosii and Thymus pallidus using the DPPH, TAC, and FRAP methods revealed an IC50 of the order of 0.085 mg/mL and 0.146 mg/mL, an EC50 of the order of 0.167 mg/mL and 0.185 mg/mL, and a total antioxidant capacity of between 750 mg EQ/g and 900 mg EQ/g, respectively. Analgesic evaluations revealed writhes inhibition of the order of 97.83% for O. grosii and 90% for T. pallidus. In addition, both plant extracts showed limited hemolytic activity, not exceeding 30% at a concentration of 100 mg/mL. Evaluation of the anti-inflammatory potential showed edema inhibition of the order of 94% (800 mg/kg) for O. grosii and 86% (800 mg/kg) for T. pallidus. These results highlight the potential applications of these extracts in pharmacological research.

Abstract:Origanum species are mostly distributed around the Mediterranean, Euro‐Siberian, and Iran‐Siberian regions. Since time immemorial, the genus has popularly been used in Southern Europe, as well as on the American continent as a spice now known all over the world under the name "oregano" or "pizza‐spice." Origanum plants are also employed to prepare bitter tinctures, wines, vermouths, beer, and kvass. The major components of Origanum essential oil are various terpenes, phenols, phenolic acids, and flavonoids with predominant occurrence of carvacrol and thymol (with reasonable amounts of p‐cymen and ‐terpinene) or of terpinene‐4‐ol, linalool, and sabinene hydrate. Many species of Origanum genus are used to treat kidney, digestive, nervous, and respiratory disorders, spasms, sore throat, diabetes, lean menstruation, hypertension, cold, insomnia, toothache, headache, epilepsy, urinary tract infections, etc. Origanum essential oil showed potent bioactivities owing to its major constituents' carvacrol, thymol, and monoterpenes. Several preclinical studies evidenced its pharmacological potential as antiproliferative or anticancer, antidiabetic, antihyperlipidemic, anti‐obesity, renoprotective, antiinflammatory, vasoprotective, cardioprotective, antinociceptive, insecticidal, and hepatoprotective properties. Its nanotechnological applications as a promising pharmaceutical in order to enhance the solubility, physicochemical stability, and the accumulation rate of its essential oils have been investigated. However, Origanum has been reported causing angioedema, perioral dermatitis, allergic reaction, inhibition of platelet aggregation, hypoglycemia, and abortion. Conclusive evidences are still required for its clinical applications against human medical conditions. Toxicity analyses and risk assessment will aid to its safe and efficacious application. In addition, elaborate structure–activity studies are needed to explore the potential use of Origanum‐derived phytochemicals as promising drug candidates.

Exploring of Chemical Profile and Biological Activities of Three Ocimum Species From Comoros Islands: A Combination of In Vitro and In Silico Insights

Abstract:Ocimum species have a great interest in different traditional medicinal systems. This study examined the chemical composition, antioxidant properties, enzyme inhibitory effects, and antibacterial and antifungal activities of the aerial parts of Ocimum gratissimum, Ocimum americanum, and Ocimum basilicum from the Comoros Islands. The extracts were analyzed using high-performance liquid chromatography-mass spectrometry (HPLC-MS) to determine their chemical composition. Antioxidant activity was assessed using 2,2-Diphenyl-1-picrylhydrazyl (DPPH), 2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS), cupric reducing antioxidant capacity (CUPRAC), ferric reducing antioxidant power (FRAP), chelating ability, and phosphomolybdenum radical scavenging assays. Enzyme inhibitory activities against acetylcholinesterase (AChE), butrylcholinesterase (BChE), tyrosinase, amylase, and glucosidase were evaluated using spectrophotometric methods. Antibacterial and antifungal activities were tested using the broth microdilution method against selected pathogenic microorganisms. The selected enzymes and proteins were evaluated using in silico methods with biomolecules from these plants. In addition, 111 different metabolites were identified in the tested extracts using advanced HPLC/MS techniques. The most significant number of detected compounds were derivatives of hydroxycinnamic acids, followed by flavonoid glycosides and aglycones and derivatives of hydroxybenzoic acids. All three Ocimum species exhibited significant antioxidant activities, O. gratissimum exhibited the best-reducing abilities in CUPRAC and FRAP assays. In addition, enzyme inhibitory assays revealed that O. americanum had the most potent inhibitory effect on tyrosinase (48.01 ± 3.89 mg kojic acid equivalent [KAE]/g), and amylase (1.08 ± 0.02 mmol acarbose equivalent [ACAE]/g). Antibacterial and antifungal tests demonstrated that the extracts possess broad-spectrum activity. Molecular docking results showed that compounds exhibited remarkable binding energies with target enzymes and proteins. The molecular dynamics simulations identified chicoric acid with MurE of Staphylococcus aureus complex as the most promising drug candidate. These findings support their traditional medical and nutraceutical uses and suggest possibilities for natural functional applications.

Abstract:Ocimum plants are commonly used culinary herbs and fragrant ornamental plants. Beyond this feature, the present review firstly describes scientific studies on the antimicrobial and antioxidant activities of several Ocimum species. Secondly, the use of Ocimum plants as ingredients for food preservation, such as meat, fish and dairy products was covered. Thirdly, the phytochemical composition has been reviewed to reinforce further standardization to be Ocimum plants used as preservatives. Finally, other pharmacological properties (cardioprotective activity, anti-diabetic activity, hepato-renal protective activity, anticancer activity) are also reviewed. Overall, these plants can serve to promote food preservatives and functional ingredients, but there are still some challenges to be overcome as this review points out.

Abstract:Ocimum species represent commercially important medicinal and aromatic plants. The essential oil biosynthesized by Ocimum species is enriched with specialized metabolites specifically, terpenoids and phenylpropanoids. Interestingly, various Ocimum species are known to exhibit diverse chemical profiles, and this chemical diversity has been at the center of many studies to identify commercially important chemotypes. Here, we present various chemotypes from the Ocimum species and emphasize trends, implications, and strategies for the quality and yield improvement of essential oil. Globally, many Ocimum species have been analyzed for their essential oil composition in over 50 countries. Asia represents the highest number of chemotypes, followed by Africa, South America, and Europe. Ocimum basilicum L. has been the most widespread and well-studied species, followed by O. gratissimum L., O. tenuiflorum L., O. canum Sims, O. americanum and O. kilimandscharicum Gürke. Moreover, various molecular reasons, benefits, adverse health effects and mechanisms behind this vast chemodiversity have been discussed. Different strategies of plant breeding, metabolic engineering, transgenic, and tissue-culture, along with anatomical modifications, are surveyed to enhance specific chemotypic profiles and essential oil yield in numerous Ocimum species. Consequently, chemical characterization of the essential oil obtained from Ocimum species has become indispensable for its proper utilization The present chemodiversity knowledge from Ocimum species will help to exploit various applications in the industrial, agriculture, biopharmaceutical, and food sectors.

Identification of compounds from Origanum compactum and Origanum elongatum using HPLC/UV-ESI-MS and comparative analysis of their antioxidant, antimicrobial, anticoagulant, and antidiabetic properties

Abstract:The aim was to assess the phytochemical composition, phenolic component levels, and biological properties of the flowering tops of Origanum compactum and Origanum elongatum. The study employed phytochemical assays, spectrophotometric techniques for quantitative analysis of polyphenols, flavonoids, and tannins, and compound identification using HPLC/UV-ESI-MS. The antimicrobial, antioxidant, anticoagulant, and antidiabetic properties were examined both in vitro and in vivo. The results showed that the O. compactum extract had significantly high levels of total polyphenols, measuring 47.368 mg gallic acid equivalents per gram, and flavonoids, measuring 14.839 mg quercetin equivalents per gram. The phytochemical examination of O. compactum revealed that lithospermic acid accounted for 36.82 % of the chemicals detected, followed by salvianolic acid C at 12.57 % and ros-marinic acid at 6.01 %. The main constituents of O. elongatum are salvianolic acid C (14.46 %), luteolin-3-O-glucuronide (13.51 %), salvianolic acid B (12.24 %), rosmarinic acid (7.83 %), and rutin (6.18 %). The results demonstrated different levels of effectiveness against the investigated microorganisms, with the extract from O. compactum exhibiting better activity, particularly against Gram-negative bacteria, certain yeasts, and the fungus Aspergillus niger. The aqueous extracts of both Origanum species demonstrate significant antioxidant activity. O. compactum has a higher total antioxidant capacity (IC50 of 35.083 μg/mL) compared to O. elongatum (IC50 of 77.080 μg/mL). However, O. elongatum has a higher reducing power (35.697 μg/mL) compared to O. compactum (42.563 μg/mL). In vivo evaluations revealed that the aqueous extracts of O. compactum and O. elongatum possess significant antihyperglycemic and anticoagulant properties. The extracts demonstrated a marked reduction in blood glucose levels during the oral glucose tolerance test (OGTT) in Wistar rats and effectively prolonged both prothrombin time (PT) and activated partial thromboplastin time (aPTT), highlighting their ability to inhibit coagulation pathways. Moreover, their comparable efficacy to standard antihyperglycemic medications and absence of severe toxicity, even at high doses, underscore their therapeutic potential for safe and effective treatment applications. Between the two species, O. compactum exhibited superior efficacy in key biological activities such as antioxidant, antimicrobial, and anticoagulant properties, making it a strong candidate for therapeutic applications. This study underscores the value of Origanum species as a rich source of bioactive compounds, offering significant potential in pharmaceuticals, nutraceuticals, and agri-food industries. The findings pave the way for further exploration of their diverse applications.
