Abstract:A method was developed for the preparation of temperature-responsive multifunctional nanoreactor by coating the poly (N-isopropylacrylamide) (PNIPAM) on the external surface of silicallanthanide-POMs/Au spheres. The nanoreactor showed the repeated on/off catalytic activity and switchable luminescence by temperature stimuli. For example, the nanocomplexes showed strong red luminescence in solution and excellent catalytic activity in the reduction of 4-nitrophenol at 25 degrees C. However, when the temperature was 40 degrees C, the red luminescence almost disappeared in solution and catalytic activity was sharply decreased. Moreover, in solutions of K4Fe(CN)(6) containing Na2SO4 at 25 degrees C, it demonstrated the well-defined cyclic voltammograms (CV) peaks with large peak currents, showing the "on" state; at 40 degrees C, the CV response was significantly suppressed, showing the "off" state. The smart nanoreactors have great potential for applications in many fields, such as bio-materials, controlled labeling catalysis, switches and so on. (C) 2017 Elsevier B.V. All rights reserved.
Bidirectional Regulation of Intracellular Enzyme Activity Using Light‐Driven Nano‐Inhibitors

Abstract:Photochemical regulation provides precise control over enzyme activities with high spatiotemporal resolution. A promising approach involves anchoring "photoswitches" at enzyme active sites to modulate substrate recognition. However, current methods often require genetic mutations and irreversible enzyme modifications for the site-specific anchoring of "photoswitches", potentially compromising the enzyme activities. Herein, we present a pioneering reversible nano-inhibitor based on molecular imprinting technique for bidirectional regulation of intracellular enzyme activity. The nano-inhibitor employs a molecularly imprinted polymer nanoparticle as its body and azobenzene-modified inhibitors ("photoswitches") as the arms. By using a target enzyme as the molecular template, the nano-inhibitor acquires oriented binding sites on its surface, resulting in a high affinity for the target enzyme and non-covalently firm anchoring of the azobenzene-modified inhibitor to the enzyme active site. Harnessing the reversible isomerization of azobenzene units upon exposure to ultraviolet and visible light, the nano-inhibitor achieves bidirectional enzyme activity regulation by precisely docking and undocking inhibitor at the active site. Notably, this innovative approach enables the facile in situ regulation of intracellular endogenous enzymes, such as carbonic anhydrase. Our results represent a practical and versatile tool for precise enzyme activity regulation in complex intracellular environments.

Abstract:Empowering biocatalyst-modified electrodes with the ability to both enforce and perceive will enable the development of intrinsically switchable bioelectrode systems, which exhibit autonomous and heteronomous actions specific to living organisms. However, the electrocatalytic activity of switchable bioelectrodes reported so far has been controlled by changes in the rate of substrate transport to biocatalysts. Here, we prepared a cup-stacked carbon nanofiber (CSCNF) electrode modified with a thermoresponsive N-isopropylacrylamide-based polymer containing peroxidase model compounds (HP). As CSCNFs worked as a converter from near-infrared (NIR) light to heat, bioelectrocatalytic activity of the electrode to H2O2 reduction was reversibly controlled by changes in the amount of electroactive HP, based on expanded and contracted states of the polymers induced by not only environmental temperature changes but also external NIR light irradiation. This intrinsically switchable bioelectrode technique would hold promise for adding new performances in electrochemical biosensors and biofuel cells, for example, autonomous and heteronomous tunable sensitivity and capacity.
Photo‐Switchable Peroxidase/Catalase‐Like Activity of Carbon Quantum Dots

Abstract:Nanozymes possess multi-enzyme activities over the natural enzymes, which produce multi-pathway synergistic effects for varies of biomedical applications. Unfortunately, their multi-enzyme activities are in fighting, significantly reducing the synergistic effects. Dynamic regulation of their multi-enzyme activities is the bottleneck for intelligent therapies. Herein, we construct a novel oxygen-nitrogen functionalized carbon quantum dots (O/N-CQDs) with peroxidase-like (Reactive oxygen species (ROS) producer) activity. Interestingly, the peroxidase-like activity can be reversibly converted to catalase-like (ROS scavenger) activity under visible light irradiation. It is found that both the peroxidase/catalase-like activity of O/N-CQDs can be precisely manipulated by the light intensity. The mechanism of switchable enzyme activities is attributed to the polarization of quinoid nitrogen in polyaniline (PANI) precursor retained on O/N-CQDs under visible light, which consumes the ROS to produce O2 and H2O. As a proof-of-concept demonstration, we are able to non-intrusively up and down regulate the ROS level in cells successfully by simply switching off and on the light respectively, potentially facilitating the precise medicine based on the development of the disease. Indeed, the photo-switchable peroxidase/catalase-like activity of O/N-CQDs opens a non-invasive strategy for better manipulations of the multi-activity of nanozymes, promising their wider and more intelligent biomedical applications.

Abstract:Smart supramolecular nanoenzymes with temperature-driven switching property have been successfully constructed by the self-assembly of supra-amphiphiles formed by the cyclodextrin-based host-guest chemistry. The self-assembled nanostructures were catalyst-functionalized and thermosensitively-functionalized through conveniently linking the catalytic center of glutathione peroxidase and thermosensitive polymer to the host cyclodextriri molecules. The ON-OFF switches for the peroxidase activity by reversible transformation of nanostructures from tube to sphere have been achieved through changing the temperature. We anticipate that such intelligent enzyme mimics could be developed to use in an antioxidant medicine with controlled catalytic efficiency according to the needs of the human body in the future.

Abstract:Gated photochromism is of interest for the operation and control of modern high-tech optofunctional materials. For further advancing this topic towards the achievement of multifunctional molecular switching, however, it remains a great challenge to incorporate multiple fluorescence regulation into gated photochromism in one unimolecular system. Herein, it is reported that a dithienylethene derivative DTEN with a Schiff base connection can be facilely synthesized by one-step coupling, and it enabled distinct color and spectral changes upon different stimuli, including ultraviolet, visible light, Ni2+, and Al3+. Relying on hydrazine and hydroxy units in this molecule, compound DTEN exhibited novel Ni2+-locked photochromic characteristics originating from complexation of the compound with Ni2+ in a 2:1 stoichiometry. On the other hand, a 1:1 complexation between compound DTEN and Al3+ could allow both of the initial and photostationary states of DTEN to display fluorescent enhancement and a redshift, realizing a dual-fluorescence "turn-on" sensing of Al3+ by light. On this basis, it is argued that the switching of the coordination mode between DTEN and Ni2+ or Al3+ brings up the possibility of tunable photoswitching by multiple stimuli, which offers a novel way for future development of multifunctional switching materials with different input and output signals, as exemplified by the construction of a delicate molecular circuit.

Abstract:Plasmonic oligomers are near-field-coupled assemblies of metallic nanoparticles. Both their scattering/absorption spectra and the spatial distribution of the electromagnetic field can be tailored through the hybridization of plasmonic modes hosted by individual particles. Such a control on the field distribution opens new routes to deliver light at a deep subwavelength scale in targeted locations (“hot spots”). However, active control of hot spots in plasmonic oligomers and their observation in the near field are highly challenging. ON-OFF nanopores for optical control of transmembrane ionic communication

Abstract:Nanoscale photoswitchable proteins could facilitate precise spatiotemporal control of transmembrane communication and support studies in synthetic biology, neuroscience, and bioelectronics. Through covalent modification of the alpha-hemolysin protein pore with arylazopyrazole photoswitches, we have produced "photopores" that transition between iontronic resistor and diode modes in response to irradiation at orthogonal wavelengths. In the diode mode, a low-leak OFF-state nanopore exhibits a reversible increase in unitary conductance of more than 20-fold upon irradiation at 365 nm. A rectification ratio of >5 was achieved with photopores in the diode state by either direct or alternating voltage input. Unlike conventional electronic phototransistors with intensity-dependent photoelectric responses, the photopores regulated current output solely based on the wavelength(s) of monochromatic or dual-wavelength irradiation. Dual-wavelength irradiation at various relative intensities allowed graded adjustment of photopore conductance. By using these properties, photonic signals were converted into ionic signals, highlighting the potential applications of photopores as components of smart devices in synthetic biology.

Abstract:Photoregulation of biomolecules has become crucial tools in chemical biology, because light enables access under mild conditions and with delicate spatiotemporal control. The control of enzyme activity in a reversible way is a challenge. To achieve it, a facile approach is to use photoswitchable inhibitors. This review highlights recent progress in photoswitchable inhibitors based on azobenzenes units. The progress suggests that the incorporation of an azobenzene unit to a known inhibitor is an effective method for preparing a photoswitchable inhibitor, and with these photoswitchable inhibitors, the activity of enzymes can be regulated by optical control, which is valuable in both basic science and therapeutic applications.

Abstract:We demonstrate the use of P3HT-spiropyran blends as an active layer to achieve reversible photoswitching effects in solution-processed organic field-effect transistors (OFETs). Conformational changes of spiropyrans triggered by light with different wavelengths produce two distinct interaction strengths at organic/organic interfaces that can reversibly modulate the channel conductance of OFETs in a noninvasive manner. This concept of organic-organic interfacial modification offers attractive new prospects for the development of organic electronic devices with desired properties.

Abstract:Copper single‐atom nanozyme possessing photothermal‐switched specificity is first reported in this study, where multiple enzyme‐like activities and specific peroxidase‐like activities can be used for sample pretreatment and sensitive sensing, respectively. Taking advantage of these characteristics, a pretreatment‐and‐sensing integration kit is constructed for point‐of‐care testing of various targets without interferences. Various applications lead to the requirement of nanozymes with either specific activity or multiple enzyme‐like activities. To this end, intelligent nanozymes with freely switching specificity abilities hold great promise to adapt to complicated and changeable practical conditions. Herein, a nitrogen‐doped carbon‐supported copper single‐atom nanozyme (named Cu SA/NC) with switchable specificity is reported. Atomically dispersed active sites endow Cu SA/NC with specific peroxidase‐like activity at room temperature. Furthermore, the intrinsic photothermal conversion ability of Cu SA/NC enables the specificity switch by additional laser irradiation, where photothermal‐induced temperature elevation triggers the expression of oxidase‐like and catalase‐like activity of Cu SA/NC. For further applications in practice, a pretreatment‐and‐sensing integration kit (PSIK) is constructed, where Cu SA/NC can successively achieve sample pretreatment and sensitive detection by switching from multi‐activity mode to specific‐activity mode. This study sets the foundation for nanozymes with switchable specificity and broadens the application scope in point‐of‐care testing.

Abstract:The stimulus-responsive regulation of enzyme catalytic activity and selectivity provides a new opportunity to extend the functionality and efficiency of immobilized enzymes. This work aims to design and synthesize a thermo-switchable enzyme@MOF for size-selective biocatalysis and biosensing through the immobilization of Candida rugosa lipase (CRL) within ZIF-8 functionalized with thermally responsive polymer, poly(N-isopropylacrylamide) (PNIPAM) (CRL@ZIF-8-PNIPAM). Unlike free CRL, which does not demonstrate substrate selectivity, we can reversibly tune the pore size of the ZIF-8-PNIPAM nanostructures (open pores or blocked pores) through temperature stimulus and subsequently modulate the substrate selectivity of CRL@ZIF-8-PNIPAM. CRL@ZIF-8-PNIPAM had the highest hydrolytic activity for small molecules (12 mM p-nitrophenol/mg protein/min, 4-nitrophenyl butyrate (p-NP Be)) and the lowest hydrolytic activity for large molecules (0.16 mM p-nitrophenol/mg protein/min, 4-nitrophenyl palmitate (p-NP P)). In addition, CRL@ZIF-8-PNIPAM demonstrated thermo-switchable behavior for large molecules (p-NP P). The p-NP P hydrolytic activity of CRL@ZIF-8-PNIPAM was significantly lower at 40 °C (blocked pores) than at 27 °C (open pores). However, the transition of blocked pores and open pores is a gradual process that resulted in a delay in the "thermo-switchable" catalytic behavior of CRL@ZIF-8-PNIPAM during thermal cycling. CRL@ZIF-8-PNIPAM was also successfully used for the fabrication of electrochemical biosensors for the selective biosensing of pesticides with different molecular sizes.

Abstract:Repurposing naturally occurring enzymes with visible light represents one of the most exciting avenues for developing new enzyme reactivity. This Emerging Topic highlights seminal work in reshaping various classes of enzymes, categorized under three mechanistic scenarios: excitation of enzymatic EDA complexes, visible‐light excitation of flavoproteins, and synergistic dual photo‐enzyme catalysis. Enzymes are natural treasure troves that hold multiple superiority. Enzymatic catalysis has become a powerful tool for asymmetric synthesis, though it is typically limited to a relatively narrow range of reaction types. By integrating the advantages of enzymatic catalysis with photocatalysis, photoenzymatic catalysis not only expands the catalytic capabilities of enzymes but also provides an effective strategy for the stereo‐control of photochemical reactions, thereby emerging as a significant research field. Herein, we focus on new‐to‐nature photoenzymatic catalysis by repurposing naturally occurring enzymes with visible light. Bidirectionally Modulating the Dissipative Phase Transition of UCST Polymers in a Programmable Manner Using Visible Light

Abstract:Light is used to modulate the dissipative phase transitions of spiropyran functionalized upper critical solution temperature polymers in a highly programmable and waste‐free manner. The phase transition temperature can be bidirectionally shifted to a lower or higher direction using light, leading to diametrically opposite transmittance changes of the solutions, which are further applied to produce complementary photopatterning for transient information storage and encryption. Dynamic phase transitions are important in living organisms for sustaining their adaptive structures and functions. The design of artificial dynamic systems using light as a noninvasive input fuel has gained significant interest to mimic such adaptive features. Here the development of light‐fueled dissipative phase transitions of upper critical solution temperature polymers by introducing pendant spiropyrans as photoactive units, is reported. The reversible open‐close photoisomerization of spiropyrans is designed to undergo either an enhancement or a decline of their net charge, which is subsequently used to regulate the electrostatic interactions between polymer chains, and ultimately shift their phase transition temperatures to either a lower or a higher direction. The degree and speed of such bidirectional shifting are flexibly modulated by manipulating the external illumination parameters or varying the compositions/concentrations of the polymers. The diametrically opposite transmittance changes driven by light are utilized to produce a series of complementary patterns and shapes for application in transient information storage and encryption with improved accuracy and security. This work offers a bidirectional spatiotemporal modulation of dissipative phase transitions of polymers using visible light that allows highly tunable and adaptable features shared with their natural counterparts.

Abstract:Materials whose luminescence can be switched by optical stimulation drive technologies ranging from superresolution imaging1-4, nanophotonics5, and optical data storage6-8, to targeted pharmacology, optogenetics, and chemical reactivity9. These photoswitchable probes, including organic fluorophores and proteins, are prone to photodegradation, and often require phototoxic doses of ultraviolet (UV) or visible light. Colloidal inorganic nanoparticles have significant stability advantages over existing photoswitchable materials, but the ability to switch emission bidirectionally, particularly with NIR light, has not been reported with nanoparticles. Here, we present 2-way, near-infrared (NIR) photoswitching of avalanching nanoparticles (ANPs), showing full optical control of upconverted emission using phototriggers in the NIR-I and NIR-II spectral regions useful for subsurface imaging. Employing single-step photodarkening10-13 and photobrightening12,14-18, we demonstrate indefinite photoswitching of individual nanoparticles (>1000 cycles over 7 h) in ambient or aqueous conditions without measurable photodegradation. Critical steps of the photoswitching mechanism are elucidated by modeling and by measuring the photon avalanche properties of single ANPs in both bright and dark states. Unlimited, reversible photoswitching of ANPs enables indefinitely rewritable 2D and 3D multi-level optical patterning of ANPs, as well as optical nanoscopy with sub-Å localization superresolution that allows us to distinguish individual ANPs within tightly packed clusters.

To address these limitations, we introduced a relationally designed nanobiohybrid system that utilized light to modulate whole-cell catalysis for chiral alcohol production. By incorporating platinum nanoparticles onto Rhodotorula sp. cell surfaces, the nanobiohybrid capitalized on the photothermal properties of the nanoparticles to regulate the overall cell activity. When exposed to light, the Pt nanoparticles generate heat through the photothermal effect, consequently leading to an increase in the catalytic activity of the whole cells. This innovative approach facilitates control over whole-cell production and provides an efficient method for regulating biocatalytic processes. The findings of this study demonstrate the significant potential of switchable control strategies in biomanufacturing across a wide range of industries.
Thermoplasmonic Effect Enables Indirect ON–OFF Control over the Z‐E Isomerization of Azobenzene‐Based Photoswitch

Abstract:Gold nanoparticles and azobenzene derivatives can be incorporated within one, processable, water‐functional material by using cellulose nanofibers as a functional scaffold. Presence of gold nanorods enables light‐driven, plasmon‐assisted Z‐E isomerization of a photochrome. Statistical modelling indicates that thermoplasmonic effect, triggered upon red‐NIR irradiation, grants an ON–OFF control over the isomerization kinetics in the hybrid material. Proper formulation of systems containing plasmonic and photochromic units, such as gold nanoparticles and azobenzene derivatives, yields materials and interfaces with synergic functionalities. Moreover, gold nanoparticles are known to accelerate the Z‐E isomerization of azobenzene molecules in the dark. However, very little is known about the light‐driven, plasmon‐assisted Z‐E isomerization of azobenzene compounds. Additionally, most of the azobenzene‐gold hybrids are prepared with nanoparticles of small, isotropic shapes and azobenzene ligands covalently linked to the surface of nanostructures. Herein, a formulation of an innovative system combining azobenzene derivative, gold nanorods, and cellulose nanofibers is proposed. The system's structural integrity relies on electrostatic interactions among components instead of covalent linkage. Cellulose, a robust scaffold, maintains the material's functionality in water and enables monitoring of the material's plasmonic‐photochromic properties upon irradiation and at elevated temperatures without gold nanorods aggregation. Experimental evidence supported by statistical analysis suggests that the optical properties of plasmonic nanometal enable indirect control over the Z‐E isomerization of the photochromic component with near‐infrared irradiation by triggering the thermoplasmonic effect. The proposed hybrid material's dual plasmonic‐photochromic functionality, versatility, and ease of processing render a convenient starting point for further advanced azobenzene‐related research and 3D printing of macroscopic light‐responsive structures.

Abstract:Light offers unique opportunities for controlling the activity of materials and biosystems with high spatiotemporal resolution. Molecular photoswitches are chromophores that undergo reversible isomerization between different states upon irradiation with light, allowing a convenient means to control their influence over the system of interest. However, a significant limitation of classical photoswitches is the requirement to initiate the switching in one or both directions using deleterious UV light with poor tissue penetration. Red-shifted photoswitches are hence in high demand and have attracted keen recent research interest. In this Review, we highlight recent progress towards the development of visible- and NIR-activated photoswitches characterized by distinct photochromic reaction mechanisms. We hope to inspire further endeavors in this field, allowing the full potential of these tools in biotechnology and materials chemistry applications to be realized.
