Abstract:Gram-positive and Gram-negative pathogens are able to evade the host immune system, persist within the human host, and lead to severe disease and even death. At present, bacterial infections are one of the leading causes of morbidity and mortality across the world. The development of novel antibacterial agents still represents a challenging endeavor, which is mainly attributed to the continuous emergence of more virulent and drug-resistant pathogens. Benzofuran constitutes the core of diverse pharmacologically active compounds, and there are a variety of approved benzofuranbased drugs in the market or currently going through different clinical phases or registration statuses. Benzofurans could exert antibacterial activity through various mechanisms and possess pronounced activity; therefore, benzofuran is a useful skeleton for the discovery of novel antibacterial agents. This review focuses on the recent advancement of naturally occurring benzofuran derivatives and hybrids of benzofurans with other pharmacophores as antibacterial agents, covering articles published between January 2015 and July 2021. The chemical structures and structure-activity relationships are also discussed.

Abstract:Benzofuran is a heterocyclic compound found naturally in plants and it can also be obtained through synthetic reactions. Multiple physicochemical characteristics and versatile features distinguish benzofuran, and its chemical structure is composed of fused benzene and furan rings. Benzofuran derivatives are essential compounds that hold vital biological activities to design novel therapies with enhanced efficacy compared to conventional treatments. Therefore, medicinal chemists used its core to synthesize new derivatives that can be applied to a variety of disorders. Benzofuran exhibited potential effectiveness in chronic diseases such as hypertension, neurodegenerative and oxidative conditions, and dyslipidemia. In acute infections, benzofuran revealed anti-infective properties against microorganisms like viruses, bacteria, and parasites. In recent years, the complex nature and the number of acquired or resistant cancer cases have been largely increasing. Benzofuran derivatives revealed potential anticancer activity with lower incidence or severity of adverse events normally encountered during chemotherapeutic treatments. This review discusses the structure–activity relationship (SAR) of several benzofuran derivatives in order to elucidate the possible substitution alternatives and structural requirements for a highly potent and selective anticancer activity.

Abstract:The benzofuran moiety constitutes a main component of enormous biologically active natural and synthetic heterocycles. Such heterocycles have distinctive therapeutic potential and are employed in various clinical drugs. A number of publications have dealt with the synthesis and extraction of benzofuran-based heterocycles to investigate their antimicrobial potential. This review describes the antimicrobial activity of various natural and synthetic benzofuran scaffolds. The antimicrobial activity of benzofurans is thoroughly investigated against several bacterial (Gram-positive and Gram-negative) and fungal microorganisms compared with several reference antibiotic drugs. The effects of the electronic nature of substituents on the activity of benzofurans through SAR study were reported. This article also highlights the recent natural and synthetic benzofuran-based organic molecules between 2019–2022 that have had success in terms of their antimicrobial activity. Many of the described benzofurans are promising candidates as antimicrobial agents based on their activity. Most used antibiotics target infections caused by the gram-positive pathogen S. aureus . Interestingly, most of the described benzofurans are promising inhibitors against S. aureus with either equipotent or more potent activity than the reference antibiotic drugs. These findings will encourage medicinal chemists to explore these new avenues for human health promotion to reduce suffering.

Abstract:Abstract: 2,3-Dihydrobenzofuran and coumaranone are readily available in numerous naturally occurring compounds. They mostly exist in plenty of food plants and medicinal plants. Such compounds constitute a series of flavor components and bioactive molecules. Their preparation has been an attractive field of research. In the past few decades, great efforts have been made in the preparation of the 2,3-dihydrobenzofuran structure through both metal-catalyzed and organocatalyzed ways. Visible light-promoted reactions sprang up in the early 21st century and represent a green manner of transformations. Under the irradiation of visible light, radicals could be generated under milder conditions. Thus, visible light-promoted reactions spread widely in the field of chemical synthesis. In recent years, visible light-promoted preparation of 2,3-dihydrobenzofuran and coumaranone has been developed by different groups, including both intramolecular and intermolecular reactions. The benign reaction conditions allow better functional group-tolerance and lead to diverse structures. Several reviews on the synthesis of 2,3-dihydrobenzofuran have been reported. However, visible light-promoted approaches to such structures have not been well reviewed. Our review will cover the literature that has been reported on the discovery of 2,3-dihydrobenzofuran in food and visible light-promoted preparation of 2,3- dihydrobenzofuran, attempting to summarize the existing methods and provide guidance to the chemists on the present challenges.

Abstract:A naturally occurring benzofuran, namely 5-(3-hydroxypropyl)-7-methoxy-2-(3'-methoxy-4'-hydroxyphenyl)3-benzo[b]furancarbaldehyde was isolated from the roots of S. miltiorrhiza Bunge ''Danshen''. Its structure was established by means of spectroscopic methods as well as by a total synthesis. Both the natural and the synthetic compounds showed a high potency (IC50 = 17 nM and 10 nM, respectively) in the bovine adenosine A1 radioligand binding.

Abstract:Abstract A new benzofuran compound, named 2-[5-hydroxy-4-methoxy-2-(3-p-henyl-trans-allyloxy)benzyl]-5-hydroxy-6-methoxy-3-phenylbenzofuran (1), together with (+)-obtusafuran (2) and isoparvifuran (3), was isolated from the heartwood of Dalbergia latifolia. Their structures were elucidated by a combination of spectroscopic methods and comparison with the literature. Compounds 2 and 3 were obtained from this plant for the first time. Compound 1 exhibited moderated antioxidant effect for scavenging 1,1-diphenyl-2-picryhydrazyl (DPPH) free radical (IC50 = 96.7 ± 8.9 μM).

Abstract:A novel method for the synthesis of benzofuran analogues via condensation of 2-hydroxybenzaldehyde or 2-hydroxyphenyl ketone with methyl 4-bromocrotonate was reported. Moderate to good yield was obtained with cesium carbonate as base in DMF at 70 degrees C. The new-generated double bond was (E)-configuration determined by X-ray diffraction. All benzofuran derivatives were determined by H-1 NMR, C-13 NMR and HRMS techniques.

Abstract:Furocoumarins are an important class of heterocyclic compounds with a fused tricyclic structure of coumarin and furan rings. They are commonly found in bioactive natural products and have a diverse range of biological and pharmaceutical properties, including cytotoxicity, photosensitivity, insecticidal, antibacterial, and antifungal activity, among others. The elegant linear/angular tricyclic skeleton and superior pharmacological properties, make them ideal for building and developing advanced biological scaffolds for biomedical applications. As a result, the family of furocoumarins has been the focus of intensive research, and lots of encouraging progress have been achieved in recent years. This review summarizes the most recent methods reported for the synthesis of the furocoumarin derivative family, along with their applications in medicinal chemistry covering from 2018 to 2022.

Abstract:Seven new quinoxaline derivatives 3a-3g were synthesized through two steps convenient approach. Their structures were confirmed by Fourier transform infrared, proton nuclear magnetic resonance, and high-resolution mass spectrometry. The synthesized compounds 3a-3g showed good fluorescent intensity in dimethyl sulfoxide. They were also shown to possess good antifungal activities. The structure-activity relationship was studied by density functional theory calculations.

Abstract:A palladium(0)-catalyzed cascade reaction for the efficient synthesis of 2,3-disubstituted benzofuran derivatives 3 containing a 3-trisubstituted alkene functional group in moderate yields from alkynyl-substituted benzynes 1 and aryl halides 2 has been developed. This method provides an efficient and alternative approach to benzofurans which are very useful heterocyclic compounds with biological and pharmacological potentials. A plausible mechanism involving intramolecular ene reaction, intermolecular insertion, and β-H elimination is proposed.

Abstract:α-Methylene-γ-butyrolactone scaffold and benzofuran pharmacophore demonstrate a number of valuable medicinal properties. Based on bioactivity-guided mixed synthesis principle, we have designed and synthesized a series of new derivatives combining these two substructures. Antifungal activity of the products against some pathogenic fungi and cytotoxicity has been tested. QSAR has been performed. All зкщвгсеы demonstrate high activity against B. cinerea and G. graminis. Compounds with the 4-fluorophenyl group and compound connected with the cinnamic aldehyde structure demonstrate superior in vitro and in vivo activity. Results of SARs and QSAR studies exhibit that the lower electron density around the α-methylene-γ-butyrolactone backbone structure and smaller steric hindrance on the exocyclic carbon-carbon double bond are beneficial for antifungal activity. The results indicate benzbutyrolactone derivatives with α-methylene-γ-butyrolactone and benzofuran pharmacophore as highly active low-toxic compounds with the fungicide potential potential.

The reactions, performed in the presence of K2CO3, EtOH/H2O and Pd(II) complex as catalyst, gave the corresponding products in good to excellent yields. The methodology allows the facile production of heterobiaryl compounds, a unique architectural motif that is ubiquitous in medicinal chemistry.

Abstract:The unsatisfactory effects of conventional bactericides and antimicrobial resistance have increased the challenges in managing plant diseases caused by bacterial pests. Here, we report the successful design and synthesis of benzofuran derivatives using benzofuran as the core skeleton and splicing the disulfide moieties commonly seen in natural substances with antibacterial properties. Most of our developed benzofurans displayed remarkable antibacterial activities to frequently encountered pathogens, including Xanthomonas oryzae pv oryzae (Xoo), Xanthomonas oryzae pv oryzicola (Xoc), and Xanthomonas axonopodis pv citri (Xac). With the assistance of the three-dimensional quantitative constitutive relationship (3D-QSAR) model, the optimal compound V40 was obtained, which has better in vitro antibacterial activity with EC50 values of 0.28, 0.56, and 10.43 μg/mL against Xoo, Xoc, and Xac, respectively, than those of positive control, TC (66.41, 78.49, and 120.36 μg/mL) and allicin (8.40, 28.22, and 88.04 μg/mL). Combining the results of proteomic analysis and enzyme activity assay allows the antibacterial mechanism of V40 to be preliminarily revealed, suggesting its potential as a versatile bactericide in combating bacterial pests in the future.

Abstract:INTRODUCTION: The benzopyran derivatives present a wide variety of biological activity and behaviour. At the same time the benzopyran derivatives support their use as therapeutic agents for multiple diseases. Their structural characteristics correlated to physicochemical properties seem to define the extent of the biological activity. Areas covered: This review summarizes new patents published on new benzopyran derivatives from 2009 to 2016. Expert opinion: Many benzopyran derivatives have vivo/vitro biological responses. Their clinical evaluation will be critical to assess therapeutic utility. The compounds containing benzopyran moiety is well defined as lead compounds for design of new more promising molecules.
