Abstract:A novel method for speciation analysis of inorganic arsenic was developed by on-line hyphenating microchip capillary electrophoresis (chip-CE) with hydride generation atomic fluorescence spectrometry (HG-AFS). Baseline separation of As(III) and As(V) was achieved within 54s by the chip-CE in a 90mm long channel at 2500V using a mixture of 25mmoll−1 H3BO3 and 0.4mmoll−1 CTAB (pH 8.9) as electrolyte buffer. The precisions (RSD, n=5) ranged from 1.9 to 1.4% for migration time, 2.1 to 2.7% for peak area, and 1.8 to 2.3% for peak height for the two arsenic species at 3.0mgl−1 (as As) level. The detection limits (3σ) for As(III) and As(V) based on peak height measurement were 76 and 112μgl−1 (as As), respectively. The recoveries of the spikes (1mgl−1 (as As) of As(III) and As(V)) in four locally collected water samples ranged from 93.7 to 106%.

Abstract:It has been challenging to analyze inorganic arsenic (iAs) with anion exchange HPLC-Electrospray Ionization-Mass spectrometry (HPLC-ESI-MS), because arsenite (As(III)) is difficult to retain on column and the salts in mobile phase causes ionization suppression of iAs. To address these issues, a method has been developed involving the determination of arsenate (As(V)) with mixed mode HPLC-ESI-MS and the conversion of As(III) to As(V) for total iAs. As(V) was separated from other chemicals on Newcrom B, a bi-modal HPLC column involving anion exchange and reverse phase interaction. The elution employed a two-dimensional gradient, including a formic acid gradient to elute As(V) and a concurrent alcohol gradient to elute organic anions used in sample preparations. As(V) was detected by Selected Ion Recording (SIR) in negative mode at m/z = 141 with a QDa (single quad) detector. As(III) was quantitatively converted to As(V) by mCPBA oxidation and measured for total iAs. By replacing salt with formic acid in elution, the ionization efficiency for As(V) was greatly enhanced in ESI interface. The limit of detection (LOD) for As(V) and As(III) were 0.0263 μM (1.97 ppb) and 0.0398 μM (2.99 ppb), respectively. The linear range was 0.05-1 μM. The method has been used to characterize iAs speciation change in the solution and precipitation in a simulated iron-rich groundwater caused by air exposure.

Abstract:Nanoliter high-performance liquid chromatography shows low consumption of solvents and samples, offering one of the best choices for arsenic speciation in precious samples in combination with inuctively coupled plasma mass spectrometry. A systematic investigation on coupling nanoliter high-performance liquid chromatography to inductively coupled plasma mass spectrometry from instrument design to injected sample volume and mobile phase was performed in this study. Nanoflow mobile phase was delivered by flow splitting using a conventional high-pressure pump with reuse of mobile phase waste. Dead volume was minimized to 60 nL for the sheathless interface based on the previously developed nanonebulizer. Capillary columns for nanoliter high-performance liquid chromatography were found to be sensitive to sample loading volume. An apparent difference was also found between the mobile phases for nanoliter and conventional high-performance liquid chromatography. Baseline separation of arsenite, arsenate, monomethylarsenic, and dimethylarsenic was achieved within 11 min on a 15 cm C-18 capillary column and within 12 min on a 25 cm strong anion exchange column. Detection limits of 0.9-1.8 mu g/L were obtained with precisions variable in the range of 1.6-4.2%. A good agreement between determined and certified values of a certified reference material of human urine (GBW 09115) validated its accuracy along with good recoveries (87-102%).

Abstract:Multi-elemental speciation analysis in a single run reduces costs, waste and time, and also sample consumption, which is favorable to a large set of instable samples. This work demonstrates mercury and arsenic speciation in a single run using high performance liquid chromatography with inductively coupled plasma mass spectrometric detection (HPLC/ICP-MS). Thiol compounds in mobile phases interfered with the elution of arsenite because of coordination action, which was overcome by gradient elution (elution of As(III) by thiol-free mobile phases at first). Four arsenic forms (As(III), DMA, MMA and As(V)) and three mercury species (Hg(II), MeHg and EtHg) were separated and detected by HPLC/ICP-MS using 4 mM tetrabutylammonium hydroxide (TBAH) (pH 6.0) in the first 1.0 min and 4 mM TBAH + 20 mM L-cysteine (pH 6.0) in the next 9.0 min. The detection limits were obtained at 0.078 for As(III), 0.081 for DMA, 0.078 for MMA and 0.15 mu g L-1 for As(V), and 0.016 for Hg(II), 0.027 for MeHg and 0.032 mu g L-1 for EtHg, and the reproducibilities (50 mu g L-1 for each As-species and 10 mu g L-1 for each Hg-species) were all lower than 3.7%. The proposed method was successfully applied for speciation analyses of arsenic and mercury in five lotus roots, which indicated DMA (3.5-7.9 mu g kg(-1)) as the only arsenic species and MeHg (0.6-1.0 mu g kg(-1)) as the only mercury species. Analyses of two certified reference materials demonstrated good agreement between determined values and certified values. All results prove its advantages including high sensitivity and high analytical efficiency, which are beneficial for routine speciation analyses of arsenic and mercury in a single run in environmental and food fields.

Abstract:Abstract An on-line method capable of the separation of arsenic species was developed for the speciation of arsenite As(III), arsenate As(V), monomethylarsenic (MMA) and dimethylarsenic acid (DMA) in biological samples. The method is based on the combination of high-performance liquid chromatograph (HPLC) for separation, UV photo oxidation for sample digestion and hydride generation atomic fluorescence spectrometry (HGAFS) for sensitive detection. The best separation results were obtained with an anion-exchange AS11 column protected by an AG11 guard column, and gradient elution with NaH2PO4 and water as mobile phase. The on-line UV photo oxidation with 1.5% K2S2O8 in 0.2 mol L–1 NaOH in an 8 m PTFE coil for 40 s ensures the digestion of organoarsenic compounds. Detection limits for the four species were in the range of 0.11–0.15 ng (20 μL injected). Procedures were validated by analysis of the certified reference materials GBW09103 freeze-dried human urine and the results were in good agreement with the certified values of total arsenic concentration. The method has been successfully applied to speciation studies of blood arsenic species with no need of sample pretreatment. Speciation of arsenic in blood samples collected from two patients after the ingestion of realgar-containing drug reveals slight increase of arsenite and DMA, resulting from the digestion of realgar.

Abstract:For the past few years many studies have been performed to determine arsenic (As) speciation in drinking water, food chain and other environmental samples due to its well-recognized carcinogenic and toxic effects relating to its chemical forms and oxidation states. This review provides an overview of analytical methods, preconcentration and separation techniques, developed up to now, using HGAAS and ETAAS for the determination of inorganic As and organoarsenic species in environmental samples. Specific advantages, disadvantages, selectivity, sensitivity, efficiency, rapidity, detection limit (DL), and some aspects of recent improvements and modifications for different analytical and separation techniques, that can define their application for a particular sample analysis, are highlighted. HG-AAS has high sensitivity, selectivity and low DL using suitable separation techniques; and it is a more suitable, affordable and much less expensive technique than other detectors. The concentrations of HCl and NaBH(4) have a critical effect on the HG response of As species. Use of l-cysteine as pre-reductant is advantageous over KI to obtain the same signal response for different As species under the same, optimum and mild acid concentration, and to reduce the interference of transition metals on the arsine generation. Use of different pretreatment, digestion, separation techniques and surfactants can determine As species with DL from ngL(-1) to μgL(-1). Out of all the chromatographic techniques coupled with HGAAS/ETAAS, ion-pair reversed-phase chromatography (IP-RP) is the most popular due to its higher separation efficiency, resolution selectivity, simplicity, and ability to separate up to seven As species for both non-ionic and ionic compounds in a signal run using the same column and short time. However, a combination of anion- and cation-exchange chromatography seems the most promising for complete resolution up to eight As species. The ETAAS method using different separation techniques and chemical modifiers can determine As species in seawater with DL of 0.02 μgL(-1), eliminate interferences of transition metals, improve the selectivity of the measurement, and enhance the sensitivity.

Abstract:It is known that arsenic has different toxicological properties dependent upon both its oxidation state for inorganic compounds, as well as the different toxicity levels exhibited for organic arsenic compounds. The field of arsenic speciation analysis has grown rapidly in the recent years, especially with the utilization of chromatographic analysis. Extraction method as sequential extraction combined to microwave assisted digestion were applied to water and sediments samples. This paper describes the essential background and toxicity of arsenic in environment, and more importantly, some currently used chromatographic applications and sample handling procedures necessary to accurately detect arsenic in its various forms. Experiments and work using ultra high-performance liquid chromatography and inductively coupled plasma optical emission spectrometry for arsenic speciation of environmental samples are presented in this research.

Abstract:The effects on the absorbance signals obtained using HG-AAS of variations in concentrations of the reaction medium (hydrochloric acid), the reducing agent [sodium tetrahydroborate(III); NaBH(4)], the pre-reducing agent (l-cysteine), and the contact time (between l-cysteine and arsenic-containing solutions) for the arsines generated from solutions of arsenite, arsenate, monomethylarsonic acid (MMA), and dimethylarsenic acid (DMA), have been investigated to find a method for analysis of the four arsenic species in environmental samples. Signals were found to be greatly enhanced in low acid concentration in both the absence (0.03-0.60 M HCl) and the presence of l-cysteine (0.001-0.03 M HCl), however with l-cysteine present, higher signals were obtained. Total arsenic content and speciation of DMA, As(III), MMA, and As(V) in mixtures containing the four arsenic species, as well as some environmental samples have been obtained using the following conditions: (i) total arsenic: 0.01 M acid, 2% NaBH(4), 5% l-cysteine, and contact time<10 min; (ii) DMA: 1.0 M acid, 0.3-0.6% NaBH(4), 4.0% l-cysteine, and contact time <5 min; (iii) As(III): 4-6 M acid and 0.05% NaBH(4) in the absence of l-cysteine; (iv) MMA: 4.0 M acid, 0.03% NaBH(4), 0.4% l-cysteine, and contact time of 30 min; (v) As(V): by difference. Detection limits (ppb) for analysis of total arsenic, DMA, As(III), and MMA were found to be 1.1 (n=7), 0.5 (n=5), 0.6 (n=7), and 1.8 (n=4), respectively. Good percentage recoveries (102-114%) of added spikes were obtained for all analyses.

Abstract:In order to obtain a well understanding of the toxicity and ecological effects of trace elements in the environment, it is necessary to determine not only the total amount, but also their existing species. Speciation analysis has become increasingly important in making risk assessments of toxic elements since the toxicity and bioavailability strongly depend on their chemical forms. Effective separation of different species in combination with highly sensitive detectors to quantify these particular species is indispensable to meet this requirement. In this paper, we present the recent progresses on the speciation analysis of trace arsenic, mercury, selenium and antimony in environmental and biological samples with an emphasis on the separation and detection techniques, especially the recent applications of high performance liquid chromatography (HPLC) hyphenated to atomic spectrometry or mass spectrometry.

Speciation analysis of arsenic in Realgar and in Chinese traditional patent medicine containing Realgar

Abstract:Objective:To develop a method for analysis of arsenic species,i.e.arsenite As(Ⅲ),arsenite As(Ⅴ), monomethylarsonic acid(MMA),and dimethylarsinic acid(DMA)in Realgar and in Chinese traditional patent medicine containing Realgar.Method:Arsenic species in samples were extracted with dilute hydrochloric acid solu- tion(pH2)and the separation were performed on a silica-based strong cation-exchange(SCX)cartridge,and a resin-based strong anion-exchange(SAX)cartridge.Then hydride generation atomic fluorescence spectrometry was used to determine the speciation of arsenic in samples.Results:Calibration curves were linear with regression coeffi- cients of more than 0.999 for all four arsenic species,and the linear ranges of As(Ⅴ)、MMA and DMA were 0-20 ng·mL~(-1)and of As(Ⅲ)was 0-40 ng·mL~(-1).The average recoveries of all four arsenic species were ranged from 81% to 105.4% and precision(RSD)were within 8.7% for DMA,12% for MMA,4.7% for As(Ⅴ)and 3% for As(Ⅲ).Conclution:The proposed method is simple,accurate and is suitable for As speciation analysis in Realgar and in Chinese traditional patent medicine containing Realgar.

Abstract:This work establishes a hyphenated methodology coupling HPLC with ICP-MS for simultaneous speciation analysis of arsenic, mercury and lead for the first time. Four arsenicals (As(III), DMA, MMA and As(V)), four mercurials (Hg(II), MeHg, EtHg and PhHg) and three lead compounds (Pb(II), TML and TEL) were simultaneously analyzed within only 8 min with acceptable resolution (2.0-8.2 for As, 1.6-6.1 for Hg and 2.7-4.0 for Pb). The detection limits were 0.036-0.20 for As-species, 0.023-0.041 for Hg-species, and 0.0076-0.14 mu g L-1 for Pb-species. The developed method was applied for the measurement of five lotus seed samples, indicating the presence of DMA (19.6-28.2 mu g kg(-1)), TML (1.4-2.9 mu g kg(-1)), MeHg (1.2-4.8 mu g kg(-1)) and EtHg (0.8-2.2 mu g kg(-1)). This method provides a promising tool for studying the toxic, metabolic and bioavailable behaviors of arsenic, mercury and lead.

Abstract:Using phosphate as buffer solution, five arsenic species: arsenite(As(II)), monomethylarsonic acid(MMA), arsanilic acid(ANA), sodium dimethylarsinate(DMA) and arsenate (As(V)) have been separated by capillary zone electrophoresis, Experiment conditions were optimized when the on-line UV detection was operated at 191 nm. The reproducibility of this method has been studied using extended light length capillary. Relation between signal peak area and sample concentration has been measured, and using the largest injection volume, the detection limit has been determined. Abstract:This work describes the method of a selective hydride generation-cryotrapping (HG-CT) coupled to an extremely sensitive but simple in-house assembled and designed atomic fluorescence spectrometry (AFS) instrument for determination of toxicologically important As species. Here, an advanced flame-in-gas-shield atomizer (FIGS) was interfaced to HG-CT and its performance was compared to a standard miniature diffusion flame (MDF) atomizer. A significant improvement both in sensitivity and baseline noise was found that was reflected in improved (4 times) limits of detection (LODs). The yielded LODs with the FIGS atomizer were 0.44, 0.74, 0.15, 0.17 and 0.67 ng L(-1) for arsenite, total inorganic, mono-, dimethylated As and trimethylarsine oxide, respectively. Moreover, the sensitivities with FIGS and MDF were equal for all As species, allowing for the possibility of single species standardization with arsenate standard for accurate quantification of all other As species. The accuracy of HG-CT-AFS with FIGS was verified by speciation analysis in two samples of bottled drinking water and certified reference materials, NRC CASS-5 (nearshore seawater) and SLRS-5 (river water) that contain traces of methylated As species. As speciation was in agreement with results previously reported and sums of all quantified species corresponded with the certified total As. The feasibility of HG-CT-AFS with FIGS was also demonstrated by the speciation analysis in microsamples of exfoliated bladder epithelial cells isolated from human urine. The results for the sums of trivalent and pentavalent As species corresponded well with the reference results obtained by HG-CT-ICPMS (inductively coupled plasma mass spectrometry).

Abstract:Based an the reproducibility of flow injection hydride generation in different media and the additivity of absorbance, a now injection hydride generation atomic absorption spectrometry; combined with target factor analysis was used to determine the numbers, identities and concentrations of arsenic species in the synthetic solutions and real samples. The slope values of calibration curves of the 4 pure arsenic species in different HCl concentrations (0.5 similar to 4 mol/L) were used as target test vectors. It shows that the average recovery of 93.9% for speciation arsenic in samples with low concentration (mu g/L) can be achieved.

Abstract:A method for the analysis of arsenic species in aqueous sulfide samples is presented. The method uses an ion chromatography system connected with a Hydride-Generation Atomic Fluorescence Spectrometer (IC-HG-AFS). With this method inorganic As(III) and As(V) species in water samples can be analyzed, including arsenite (HnAs(III)O3(n-3)), thioarsenite (HnAs(III)S3(n-3)), arsenate (HnAs(V)O4(n-3)), monothioarsenate (HnAs(V)SO3(n-3)), dithioarsenate (HnAs(V)S2O2(n-3)), trithioarsenate (HnAs(V)S3O(n-3)) and tetrathioarsenate (HnAs(V)S4(n-3)). The peak identification and retention times were determined based on standard analysis of the various arsenic compounds. The analytical detection limit was ~1-3 µg L(-1) (LOD), depending on the quality of the baseline. This low detection limit makes this method also applicable to discriminate between waters meeting the drinking water standard of max. 10 µg L(-1) As, and waters that do not meet this standard. The new method was successfully applied for on-site determination of arsenic species in natural sulfidic waters, in which seven species were unambiguously identified.

Abstract:Abstract Traditional Chinese medicine contains arsenic (As), which in the natural environment accumulates in plants during the growth of Chinese medicinal materials; there are mineral medicines containing As in Chinese patent medicine such as As 4 S 4 , As 2 S 3 , etc. Due to the toxicity of As-containing compounds and its role in inflammation and treatment of cancers such as leukemia, it is necessary to analyze the chemical form of As. A comprehensive investigation of the compound forms of heavy metals rather than the simple total amount of elements will lay the foundation for the scientific and objective evaluation of the safety of heavy metals. This article summarizes the speciation of As in bulk Chinese medicinal materials and Chinese patent medicines in recent years, and reviews the main research methods of As speciation analysis. The separation and detection combined analysis method focuses on the high-performance liquid chromatography-plasma mass spectrometry and high-performance liquid chromatography-hydride generation-atomic fluorescence spectroscopy, etc. Taking the advanced synchrotron radiation source as the research platform, the use of X-ray near edge absorption fine structure spectrum and micro-area X-ray fluorescence analysis as a microscopic analysis technique supports direct analysis of the As speciation in situ . It is the most promising morphological analysis method.

Abstract:A method is presented for the determination of inorganic arsenic species by hydride generation high resolution inductively coupled plasma mass spectrometry. In the presence of 1.5mg/ml of L-cysteine, arsenic(V) is indirectly determined using the difference between As(III) concentrations determined after 2 and 8min of reaction time, whereas the As(III) concentration is obtained by taking the difference between the concentration of arsenic(III) determined after 8min and the concentration of arsenic(III) converted from arsenic(V) after 8min. 25 fold amounts of arsenic(III) and 100 fold amounts of arsenic(V) hardly affected the determination of 1ng/ml arsenic(V) and 100ng/l arsenic(III), respectively. Interferences from transition metal ions were removed by using a Chelex 100 resin. The detection limits for arsenic(III) and arsenic(V) were 11.8 and 39.7ng/l, respectively. The proposed method for the simultaneous determination of inorganic arsenic species was applied to the determination of As(III) and As(V) in spiked and Japanese standard river water samples. Agreement within statistical uncertainty was obtained in all instances.
Speciation Analysis of Arsenic and Their Distribution Characteristic in Sediments of Taihu Lake by HPLC-ICP-MS

Abstract:0.3 mol/L phosphoric acid and 0.1 mol/L ascorbic acid were used to extract the sediment samples following a microwave-assisted extraction procedure.2.0 mmol/L NaH2PO4/0.2 mmol/L EDTA(pH 6.0) were used as the mobile phase,to determine the concentration of As(Ⅲ),As(Ⅴ),MMA and DMA in the extracted liquid by high performance liquid chromatography-inductiuely coupled plasma-mass spectrometry(HPLC-ICP-MS).The chromatographic peaks of 4 arsenic species are completely separated in 10 min.The linear correlations are satisfactory.The recoveries are between 94.2% and 110%.The extraction efficiency of arsenic species were 80.4%—98.7%.The results show that arsenic species in the surface sediments of Taihu Lake are mainly inorganic arsenic.And organic arsenic species are not detected.The content of As(Ⅴ) in the samples are higher than that of As(Ⅲ),which indicate that the surface sediments of Taihu Lake basically are in the oxidizing environment.In the recent 20 years,the arsenic content in the sediment of Taihu Lake had increased evidently.The arsenics were inorganic species in the vertical profiles of sediment,and there were no remarkable differences for the relative ratios of the arsenic species in each section.

Abstract:As environmental impacts of landfill last from beginning of cell filling to many years after, there is an increasing interest in monitoring landfill leachate composition especially with regards to metals and metalloids. High-performance liquid chromatography (HPLC) coupled with inductively coupled plasma mass spectrometry (ICP-MS) has been applied to the speciation of arsenic in landfill leachates. The difficulty is related to the complexity and heterogeneity of leachate matrices. A soft sample preparation protocol with water dilution and filtration of leachates has proved to be sufficient for the achievement of identification and quantification of arsenic species without matrix effect. The cationic-exchange separation method developed has enabled the detection of six arsenic species (AsIII, MMA, AsV, DMA, AsB, TMAO) in different landfill leachates. The wide range of concentrations of arsenic species (from 0.2 to 250 microg As L(-1)) and their repartition illustrate the high variability of these effluents depending on the nature of the wastes, the landfill management, the climatic conditions and the degradation phase, to list a few. These results provide new information about the chemical composition of these effluents which is useful to better adapt their treatment and to achieve the risk assessment of landfill management.

Abstract:Hydride generation inductively coupled plasma-atomic emission spectrometry (HG ICP-AES) was used as a continuous detection system for the determination of arsenic in the eluate from a high-performance liquid chromatographic (HPLC) system. Four arsenic species [arsenite As(III), arsenate As(V), monomethylarsonate (MMA), and dimethylarsinate (DMA)] present in the urine samples of patients treated intravenously with arsenite, were analyzed separately by HPLC-HG-ICP-AES using a non-polar C18 column. This analytical method allowed the sensitive determination of the arsenic species in the submicrogram per liter range. Urine samples collected on different days after arsenite administration were found to contain arsenite predominantly--monomethylarsonate and dimethylarsinate were also detected.
