Abstract:Human hair is naturally colored by melanin pigments, which afford myriad colors from black, to brown, to red depending on the chemical structures and specific blends. In recent decades, synthetic efforts have centered on dopamine oxidation to polydopamine, an effective eumelanin similar to the one found in humans. To date, only a few attempts at polydopamine deposition on human hair have been reported, and their translation to widespread usage and potential commercialization is still hampered by the harsh conditions employed. We reasoned that novel, mild, biocompatible approaches could be developed to establish a metal-free route to tunable, nature-inspired, long-lasting coloration of human hair. Herein, we describe synthetic and formulation routes to achieving this goal and show efficacy on a variety of human hair samples via multiple spectroscopic and imaging techniques. Owing to the mild and inexpensive conditions employed, this novel approach has the potential to replace classical harsh hair dyeing conditions that have raised concerns for several decades due to their potential toxicity.Synthetic melanin nanoparticle coatings on hair surfaces successfully reverse the appearance of melanin loss and hair whitening and provides a novel biomimetic and nontoxic approach to hair dyeing.

Abstract:Melanin exists widely in nature and can afford a variety of colors from black to brown and red according to chemical structure differences and specific mixtures. Inspired by nature, this work reports that tyrosine derivatives with different protecting groups at its N- or C-terminal can be enzymatically oxidized into melanin-like pigments with a wide range of colors. The emergence of colorful pigments can be attributed to the incomplete enzymatic oxidation and polymerization caused by the chemical premodification of the tyrosine molecule. The pigments can be deposited on the surface of the hair to obtain a series of colorful and saturated hair dye effects. Moreover, after the pigments were coated on the hair, we can further deposit silver nanoparticles through in situ reduction, making these coatings have anti-inflammatory and antibacterial potential, thereby expanding their potential use for people with low immunity or those who work in hospitals. Abstract:The doped GA and TA produce a more complex structure of MPN through the synergistic effect of chelating with metal ions to finally obtain more colorful MPN pigments. MPN pigments can be deposited on the hair surface through covalent and non‐covalent interactions, producing a range of highly saturated and stable coloring effects. In this study, we developed a complex polyphenol‐metal ion system, in which two polyphenols represented by gallic acid (GA) and tannic acid (TA) chelated with metal ions leading to the formation of pigments composed of metal‐phenol networks (MPN). The doping of GA and TA creates a mutual synergistic effect, which makes it chelate with metal ions to produce a more complex structure of MPN, and finally obtain more colorful MPN pigments. The MPN pigments can be easily deposited on the hair surface through covalent and non‐covalent interactions, resulting in a series of hair dyeing effects with high color saturation and excellent stability. With many advantages such as low raw material cost, short hair dyeing time, simple process, good biocompatibility, less fading, and antioxidant, this composite polyphenol‐metal ion material offers the possibility to replace the classical irritant hair dyes commonly used in existing cosmetics.

Abstract:Hair dyeing has become a prevalent lifestyle trend, especially within the fashion industry. However, it possesses disadvantages, such as containing carcinogenic and toxic materials. In this study, we developed a biocompatible hair-dyeing technology using a shampoo with a dark polyphenol complex (DPC), referred to as S-DPC. The DPC was formed from a mixture of gallic acid and [1,1′-biphenyl]-2,2′,4,4′,5,5′-hexol and used to enhance both the stability of the hair coating and its ability to scavenge reactive oxygen species (ROS). Colloidal DPC particles play a pivotal role in the coating process of various hair dyes, ensuring the uniform coloring of human hair through intermolecular interactions such as hydrogen bonding. Owing to the effect of a polyphenol complex on hair coating, we observed improved antistatic performance and enhanced mechanical strength, resulting in a substantial increase in elongation at the breaking point from 33.74% to 48.85%. The multicolor S-DPC exhibited antioxidant properties, as indicated by its ROS-scavenging ability, including 2,2-diphenyl-1-picrylhydrazyl inhibition (87–89%), superoxide radical scavenging (84–87%), and hydroxyl radical scavenging (95–98%). Moreover, the in vitro analysis of the DPC revealed nearly 100% cell viability in live and dead assays, highlighting the remarkable biocompatibility of the DPC. Therefore, considering its effectiveness and safety, this biomaterial has considerable potential for applications in hair dyeing.
Metal-Chelation-Assisted Deposition of Polydopamine on Human Hair: A Ready-to-Use Eumelanin-Based Hair Dyeing Methodology

Abstract:Permanent dyeing of gray hair has become an increasingly active area in the cosmetics industry because of the increasingly aging population in developed countries. So far, <i>p</i>-phenylenediamine (PPD) and related diamine-based monomeric compounds have been widely used for the dyeing processes, but toxicological studies have revealed such compounds to be carcinogenic and allergenic. Here, we for the first time demonstrated that polydopamine, a mimic of human eumelanin, gives rise within a commercially acceptable period of time (i.e., 1 h) to deep black colors (i.e., natural Asian hair colors) in human keratin hairs in the presence of ferrous ions. The dyed hairs showed excellent resistance to conventional detergents, and the detailed color was readily varied by changing the kind of metal ion used. SEM images and FT-IR-ATR spectra suggested that the extent of polydopamine aggregation was crucial for the dyeing efficiency. High-resolution (15 T) FT-ICR mass spectrometry performed on the products detached from hairs with either 0.1 N HCl or NaOH indicated that similar polydopamine products were recruited into the hair matrices whether in the presence or absence of metal-based chelating. Polydopamine chains were determined using EPR and ICP-OES to use chelation of ferrous ions to self-assemble as well as to bind keratin surfaces in the dyeing conditions. Also, mice subjected to skin toxicity tests showed much greater viability and much less hair loss with our dyeing agents than with PPD. In conclusion, this study showed that a safe eumelanin mimic may be used to permanently dye gray hair, and showed three kinds of deposition mechanisms (i.e., innate binding ability of polydopamine, metal-assisted self-assembly of polydopamine, and metal-related bridging between keratin surface and polydopamine) to be involved.

Abstract:Fast and facile coating strategies play a key role in surface engineering and functionalization of materials for various applications. Herein, we report a rapid and eco-friendly hair dyeing process for natural gray hair via the formation of metal-phenolic networks (MPNs). MPNs composed of gallic acid display high performance, and the coloration is tunable by varying the metal ion types. MPN-based hair dyeing is tolerant to repeated washing (at least 50 times) with detergent solution without color fading and can be discolored in acidic solution (pH < 2). The mechanism of self-assembled MPNs for hair dyeing is investigated by Raman and UV-vis absorption spectroscopy. Cell studies in vitro and skin toxicity tests in vivo demonstrate the advantages (i.e., biocompatibility and hair regrowth) of MPNs for hair dyeing compared to p-phenylenediamine. The reported strategy for hair dyeing avoids the use of toxic substances present in common hair dyes and has negligible damage to the hair structures and tensile strength.

Abstract:There is an on-going demand in recent years for safer and "greener" hair coloring agents with the global consumer awareness of the adverse effects of synthetic hair dyes. The belief in sustainability and health benefits has focused the attention of the scientific community towards natural colorants that serve to replace their synthetic toxic counterparts. This review article encompasses the historical applications of a vast array of natural plant hair dyes and summarizes the possible coloration mechanisms (direct dyeing and mordant dyeing). Current information on phytochemicals (quinones, tannins, flavonoids, indigo, curcuminoids and carotenoids) used for hair dyeing are summarized, including their botanical sources, color chemistry and biological/toxicological activities. A particular focus is given on research into new natural hair dye sources along with eco-friendly, robust and cost-effective technologies for their processing and applications, such as the synthetic biology approach for colorant production, encapsulation techniques for stabilization and the development of inorganic nanocarriers. In addition, innovative in vitro approaches for the toxicological assessments of natural hair dye cosmetics are highlighted.

Abstract:Due to the damage of scalp because of chemical dyes, increasing attention had been paid to the ecological dyeing of human hair. To address the need for ecological restoration and dyeing of human hair, herein, a novel approach has been proposed via laccase-mediated in situ oxidation of dihydroxy phenylalanine (dopamine). As laccase can catalyze dopamine oxidation and then polymerization to produce strongly colored polymers, different biological coloration processes of human hair based on this process were investigated: (i) Human hair was treated with laccase alone; (ii) The absorption of human hair with dopamine, followed by further catalysis of air; (iii) The absorption of human hair with dopamine, followed by further catalysis of laccase; (iv) Simultaneous laccase-mediated polymerization and dyeing of human hair; and (v) Polymerization of Dopamine with laccase and then dyeing human hair. The surface properties of dyed hair were evaluated in terms of ultraviolet (UV)-protection, color fastness, Fourier transforming infrared spectrum (FT-IR), optical microscope, and scanning electron microscopy (SEM). The results show that best hair dyeing effect was achieved via simultaneous laccase-mediated polymerization and dyeing. The dyed hair obtained K/S values of 28, light fastness of level 5, and UPF of 100+. The optical microscope and SEM observations show that the surface of colored hair fibers was covered with thin burrs, which can be attributed to polydopamine attached with physical adhesive force and covalent reaction. FT-IR analysis shows that functional groups were present on the surface of the dyed hair which can be considered as introduction of hydroxyl radical groups. This biological stain technique may be widely used in dyeing of hair with black color.

Abstract:Nature provides a great source of inspiration for the development of sustainable materials with excellent properties, among which melanin with optical, electronic, and radiation protection properties are considered to be promising coloring materials. However, compared to chemical pigments, the single color, complex oxidation process, and poor solubility of natural melanin strongly limit their further applications. Here, we introduce a series of melanin-like polymeric pigments with amino acid-encoded physicochemical properties by a simple three-component reaction system. Our protocol enables artificial control of the chromophore structures through the rational design of the substrates and dopants, thereby combining the safety and functionality of biopigments with the color richness of chemical dyes. Similar to the photoprotective effect of natural melanin, the polymeric pigments showed excellent antioxidant activity in reducing free radicals and have the advantages of iridescent color, strong tinting strength, stability, and affordability. Furthermore, due to their ability to dye substrates, these biomimetic are expected to become new low-cost bioactive chromophores and find various biochemical applications such as in clothing and hair dyeing, food addition, and anticounterfeiting detection.

Abstract:There is a growing interest in using naturally-occurring plant colorants as alternative to the toxic synthetic colors for hair coloration. However, poor color reproducibility and color fastness on the human hair remain a constant problem for natural hair dyeing. This study presents a systematic investigation into the dyeing performances of dye extracts of four hair dye plants (henna, walnut husks, sappanwood and Chinese gallnuts) for hair coloration with the focus on their photostability on the dyed human hair and the underlying mechanisms. First, the optimal dyeing conditions for each of the four plant colorants were obtained on yak hairs by assessing the effects of different dyeing variables ( i.e. metallic mordants, mordant concentrations, mordanting methods and pHs) on color strength and CIE coordinates. Then, the plant colorants with ferrous mordant as the most influential metallic mordant to produce dark colors were investigated in terms of dyeability and photostability on human bleached hairs and natural gray hairs. The results demonstrated that the photosensitivity of naturally dyed human hairs is related to the nature of both hair dye colorant and the dyeing substrate. ESR measurement and fluoresce probing of hydroxyl radicals in hair fibers revealed that dyeing human hairs with natural plant colorants can inhibit, to varying extents, the photogeneration of oxidative radicals in human hairs, which may in turn provide the dyed human hairs with certain photo-protecting effects.

Abstract:Recently, developments in the field of cosmetics have led to a renewed interest in hair dyeing. However, damage to the hair during the dyeing process has increased hesitation in attempting hair dyeing. As a result, hair dyes with minimal side effects have been in constant demand, and are being developed. In this study, natural-extract polyphenols, pyrogallol, and gallic acid are coordinated by CuCl2 in a NaCl aqueous solution to form an oligomer, which creates an ion-channel coating on the hair surface to protect it. This work attempts to develop fast, simple, and damage-free hair-dye ingredients based on pyrogallol and gallic acid. The morphology and elements of polyphenols coated on hair are characterized. The results reveal that the hair is dyed with the polyphenol-based dye reagent successfully. Moreover, the thickness of the dyed hair continuously rises ten times after dyeing. The tensile strength of the dyed hair is also measured, showing an upward and downward trend. These results reflect the fact that pyrogallol and gallic acid are considered to be the essential and functional polyphenols, and can build ion blocks on hair, which can create new multifunctional coating materials.

Abstract:The increasing demand for natural and safer alternatives to traditional hair dyes has led to the investigation of nanomaterials as potential candidates for hair coloring applications. MXene nanosheets have emerged as a promising alternative in this context due to their unique optical and electronic properties. In this study, we aimed to evaluate the potential of Ti 3 C 2 T x (T x = –O, –OH, –F, etc. ) MXene nanosheets as a hair dye. MXene nanosheet-based dyes have been demonstrated to exhibit not only coloring capabilities but also additional properties such as antistatic properties, heat dissipation, and electromagnetic wave shielding. Additionally, surface modification of MXene using collagen reduces the surface roughness of hair and upregulates keratinocyte markers KRT5 and KRT14, demonstrating the potential for tuning its physicochemical and biological properties. This conceptual advancement highlights the potential of MXene nanosheets to go beyond simple cosmetic improvements and provide improved comfort and safety by preventing the presence of hazardous ingredients and solvents while providing versatility.

Abstract:Schematic diagram of dopamine synthesis process on wool fiber. The use of dopamine to synthesize melanin as a natural dye on fabrics has the disadvantages of complicated processing, long processing time, and high cost. This study simplifies the processing conditions of the oxidative polymerization of dopamine and develops wool fabric melanin coatings with different finishing effects by adjusting the reaction time. The results of computer color matching, ultraviolet (UV) protection properties, and color fastness indicated that the treated wool fibers were effectively dyed, and they achieved UV resistance properties comparable to those realized in other studies in a relatively short time. The degree of lightness (L) and UV protection factor of the treated fabric reached up to 13.3 and 80+, respectively. Samples W/PDA 60 and W/PDA 120 absorbed more than 95% of the UV light. The color fastness was greater than Grade 4. The Fourier infrared spectrum and X‐ray diffraction studies revealed that the chemical and crystalline structures of the wool fiber did not change significantly after the treatment. The breaking strength of samples W/PDA 60 and W/PDA 120 was about 50% higher than that of the original wool fabric. This efficient and simple method provides a feasible and promising solution for the color deepening and UV finishing of high value‐added wool products. It can be a potential choice for upgrading the printing and dyeing industries from the perspective of ecological protection.

Structural and Functional Tailoring of Melanin-Like Polydopamine Radical Scavengers

Abstract:Melanin is a ubiquitous but poorly understood polymeric, dark brown to black pigment biomaterial found both in nature and by organic synthesis. Its structural heterogeneity and disordered covalent/noncovalent interactions make it a grand challenge to tune and improve the physical and biological properties of both melanin sources. Herein, we report a facile one-pot fabrication approach for synthetic melanins with controlled size and composition via copolymerization of dopamine and basic amino acid such as arginine in an aqueous solution. The resulting arginine-doped polydopamine melanin-like nanoparticles possess less ordered nonplanar microstructures within the particles, whereas the conventional polydopamine-based melanins contain a variety of compact π-stacked microstructures. The assembly of these distinct polymeric microstructures could lead the preceding ones for greater accessibility to free radicals, with resultant stronger free-radical scavenging effect, and thus, exhibit superior antioxidant performance toward cellular oxidative stress. This work has inspired us to continue to fabricate new synthetic melanin pigments with tunable and improved functions by rational structural tailoring.

Abstract:Most natural biomolecules have two enantiomeric forms and can exist in either of them. Despite the L -type homochirality of natural biopolymers composed of amino acids, a great deal of effort has been devoted to the systems in which chirality dictates molecular self-assembly and corresponding properties. Previously, we demonstrated that chemical pre-modification of tyrosine ( L -Tyr) inspired by natural eumelanin can obtain colorful pigments and surface coatings, but their characteristics like stability still have development space. Here, the racemic reaction systems of tyrosine derivatives ( L/D -Tyr, Boc- L/D -Tyr and Cbz- L/D -Tyr) were designed, and we proved that substrate molecular chirality can affect the reaction rate of the oxidation process, change the intermolecular force and arrangement during assembly, thus influencing the morphology, structures, and properties of the biomimetic pigment materials. At the same time, we found that L + D -tyrosine derivatives can be oxidized and deposited on hair surfaces to obtain the color effect of black and red-brown series, and achieve green-safe biological hair dyeing. Compared with previous studies, it has faster coloring speed, higher color saturation, stronger color fixation stability, good mechanical strength and biocompatibility, providing a new idea for the manufacture of chiral-inspired advanced materials with bio-nanotechnology applications.

Abstract:As a new approach to permanent/oxidative hair dyes, we demonstrated that certain monoazo dyes containing substituents that enable diffusion into hair fibers and subsequent chelation/complexation using benign metal ions (Al3+ and Fe3+), rather than a harsh oxidant and strong alkali, merit consideration in this application area. This is important, because billions of individuals worldwide color their hair using permanent hair dye products that can contain aromatic amines and phenols that display genotoxicity and/or skin sensitization (e.g., para-phenylenediamine, PPD). Herein, our work is extended to an examination of the toxicological properties and durability of metallizable monoazo dyes, in comparison to a commercially used permanent hair dye product. Results from the Salmonella/microsome mutagenicity assay and from acute toxicity tests using aquatic test organisms (Daphnia similis, 48-h and Parhyale hawaiensis, 96-h) indicated that the proposed dyes were significantly less toxic than the top-eight permanent hair dye precursors utilized annually. Regarding durability, monoazo dye chelation in situ led to a higher degree of resistance to removal by washing than a commercial permanent hair dye. Taken together, these results further demonstrate the potential of the dyes as sustainable alternatives to conventional permanent hair dyes.

Abstract:Permanent fluorescent hair dyeing is one of the modern coloration modes to meet the increasing demands of personality and fashion statements. However, the limited durability and narrow Stokes shift of small molecule dyes hinder the permanent performance and distinguished use of fluorescent hair dyes from conventional hair coloration. Here, fluorescent metal nanomaterials, gold nanoclusters, have been utilized as hair dyes and embedded inside hair through a fast protein-assisted biomineralization approach under heating and alkaline conditions. Heat and alkalinity not only ensure that the scales of hair fully open and the dye precursor is deeply pervasive but also largely promote the reducibility of hair protein. The introduction of gold nanocluster bestows a broad range of new photophysical, much enhanced antistatic, mechanical, durable, and biocompatible properties to hair, largely improving the comfort experience and cosmetic performance.

The dual nature of biomimetic melanin

Abstract:Melanin-inspired nanomaterials offer unique photophysical, electronic and radical scavenging properties that are widely explored for health and environmental preservation, or energy conversion and storage. The incorporation of functional melanin building blocks in more complex nanostructures or surfaces is typically achieved via a bottom-up approach starting from a molecular precursor, in most cases dopamine. Here we demonstrate that indeed, the oxidative polymerization of dopamine, for the synthesis of melanin-like polydopamine (PDA), leads to the simultaneous formation of more than one nanosized species with different compositions, morphologies and properties. In particular, a low-density polymeric structure and dense nanoparticles (NP) are simultaneously formed. The two populations could be separated and analyzed in real time using a chromatographic technique free of any stationary phase (flow field fractionation, FFF). The results following the synthesis of melanin-like PDA showed that the NP are formed only during the first 6 hours as a result of a supramolecular self-assembly-driven polymerization, while the formation of the polymer continues for about 36 hours. The two populations were also separated and characterized using TEM, UV-vis absorption spectroscopy, fluorescence and light scattering spectroscopy, DLS, FTIR, ζ-potential measurements, gel electrophoresis and pH titrations. Interestingly, very different properties between the two populations were observed: in particular the polymer contains a higher number of catechol units (8 mmol g-1 -OH) with respect to the NP (1 mmol g-1 -OH) and presents a much higher antioxidant activity. The attenuation of light by NP is more efficient than that by the polymer especially in the Vis-NIR region. Moreover, while the NP scatter light with an efficiency up to 27% they are not fluorescent, and the polymer does not scatter light but shows an excitation wavelength-dependent fluorescence typical of multi-fluorophoric uncoupled systems.
