Abstract:<p>Technological advancements in the past few decades have made it possible to manufacture nanomaterials at large scale and engineered nanoparticles (ENPs) are increasingly found in consumer products such as cosmetics, sports products and LED displays. A large amount of these ENPs end up in wastewater and potentially impact the performance of wastewater treatment plants (WWTPs). One important function of the WWTP is nitrification, which is carried out by the actions of two groups of bacteria, ammonia‐oxidizing bacteria (AOB) and nitrite‐oxidizing bacteria (NOB). Since most ENPs are found to have or are designed to have antimicrobial activities, it is a legitimate concern that ENPs entering WWTPs may have negative impacts on nitrification. In this paper, the effects of ENPs on nitrification is discussed, focusing mainly on autotrophic nitrification by AOBs and NOBs. This review also covers ENPs effects on anaerobic ammonium oxidation (ANAMMOX). Generally, nitrifiers in pure and mixed culture can be inhibited by a variety of ENPs, but stress response mechanisms may attenuate toxicity. Long‐term studies demonstrated that a wide range of NPs can cause severe deterioration of AOBs and/or NOBs when the influent concentration exceeded an inhibition threshold. Proposed mechanisms include the generation of reactive oxygen species, dissolved metals, physical disruption of cell membranes, bacterial engulfment, and intracellular accumulation of ENPs. Transient Disturbance of Engineered ZnO Nanoparticles Enhances the Resistance and Resilience of Anammox Process in Wastewater Treatment

Abstract:The increasing use of engineered nanoparticles (NPs) in consumer and industrial products raises concerns about their environmental impacts, but their potential influence on anaerobic ammonium oxidation (anammox) process in wastewater treatment remains unknown. In this study, the response of granule-based anammox reactor to different loads of ZnONPs was investigated. The introduction of 1-5mgL-1 ZnONPs did not affect reactor performance, but 90% of the nitrogen removal capacity was deprived by a shock of 10mgL-1 ZnONPs within 3days. Anammox activity was significantly inhibited, but no significant stimulation of intracellular reactive oxygen species (ROS) production or extracellular lactate dehydrogenase (LDH) activity was observed. The inhibition was thus mainly due to the accumulation of toxic Zn(II) ions in anammox biomass. However, the resistance and resilience of this anammox reactor to ZnONPs were enhanced by intermittent perturbations in the mode of "shock-recovery". The up-regulated abundance of Zn(II)-exporter ZntA might contribute to the enhanced resistance. In addition, these repeated transient disturbances improved the functional specificity of the anammox community despite the reduction of its diversity. Overall, these results may provide useful references for evaluating and controlling the risk of NPs to anammox process.

Abstract:The pervasive use of engineered nanoparticles (NPs) in a wide range of fields raises concerns about their potential environmental impacts. Previous studies confirmed that some NPs had already entered wastewater treatment plants (WWTPs). Wastewater nutrient removal depends on the metabolisms of activated sludge bacteria and their related key enzymes. Therefore, this study compared the possible influences of Al2O3, SiO2, TiO2, and ZnO NPs on the key enzymes activities and microbial community structures involved in wastewater treatment facilities. It was found that long-term exposure to these NPs significantly affected the microbial communities and changed the relative abundances of key functional bacteria, such as ammonia-oxidizing bacteria. Also, the gene expressions and catalytic activities of essential enzymes, such as ammonia monooxygenase, nitrite oxidoreductase, nitrate reductase, and nitrite reductase, were decreased, which finally resulted in a lower efficiency of biological nitrogen removal.

Abstract:Persistence of ammonium nitrogen and heavy metals in wastewater still remains a challenge, and many wastewater treatment plants face the challenge of removing nitrogen under heavy metal stresses. There is no preferred method for the biological treatment of wastewater containing nitrogen and heavy metals with the possible exception of the anaerobic ammonium oxidation (anammox), since it has shown promise for removing nitrogen under heavy metal stresses. This article reviews the recent research results of the nitrogen-removal performance and mechanism by the anammox process under heavy metal stresses, mainly discussing the enhancing and inhibition effects of heavy metals on the performance of the Anammox reactor. The influencing mechanism of heavy metals on the microbial community and extracellular polymeric substances is also presented, and examples are given for explanation. The main problems of the present research are pointed out, and it is proposed that unifying the metal ion concentrations of inhibiting or promoting anammox activity is necessary for the development and industrial application of the anammox process. The information of this review can offer a great possibility for achieving desired nitrogen removal in wastewater treatment under heavy metal stresses and with significant energy savings.

Abstract:In natural and engineered environments, microorganisms often exist as complex communities, which are key to the health of ecosystems and the success of bioprocesses in various engineering applications. With the rapid development of nanotechnology in recent years, engineered nanomaterials (ENMs) have been considered one type of emerging contaminants that pose great potential risks to the proper function of microbial communities in natural and engineered ecosystems. The impacts of ENMs on microorganisms have attracted increasing research attentions; however, most studies focused on the antimicrobial activities of ENMs at single cell and population level. Elucidating the influence of ENMs on microbial communities represents a critical step toward a comprehensive understanding of the ecotoxicity of ENMs. In this mini-review, we summarize and discuss recent research work on the impacts of ENMs on microbial communities in natural and engineered ecosystems, with an emphasis on their influences on the community structure and function. We also highlight several important research topics which may be of great interest to the research community.

Abstract:Engineered nanoparticle materials (ENMs) are widely and increasingly produced and employed in many sectors. The use of diverse ENMs potentially leads to the release of multiple ENMs into the environment. These ENMs after discharge will be end in wastewater treatment plant and present in sludge. This work investigated the effect of multi-ENMs systems of ZnO and TiO2 on sludge anaerobic fermentation and the related toxicity mechanism. Results revealed that the toxicity of ZnO ENMs on anaerobic fermentation was reduced in the presence of TiO2 ENMs. Investigation on the change of free Zn2+ and reactive oxygen species (OH and H2O2) suggested that both of free Zn2+ and ROS contributed to the toxicity mechanism. Zn2+ decrease was the main reason for the reduced toxicity in multi-ENMs systems. ROS mainly led to the reduction of cell viability in anaerobic fermentation systems. The presence of TiO2 in the multi-ENMs systems promoted the recovery of enzyme activity, cell viability and bacteria abundance.

Abstract:A Although the toxicity effects of engineering nanoparticles (NPs) in biological wastewater nitrogen removal (BNR) system have been extensively attracted, the impacts of co-existing contaminants from wastewater on NP toxicity have been less addressed. In this study, the effects of ammonia on ZnO NP toxicity to typical ammonia oxidizing bacteria-Nitrosomonas europaea were investigated, as indicated by the cell density, membrane integrity, ammonia oxidation rate, and AMO activity. After 6-h's exposure to 10 mg/L ZnO NPs, the cell density, membrane integrity, ammonia oxidation rate, and AMO activity was dramatically suppressed despite of the increasing ammonia loading. Ammonia at varying concentrations did not obviously affect ZnO NPs toxicity to cell density. The presence of ammonia at 100 or 200 mg/L significantly alleviated the antibacterial effects of ZnO NPs on cells. The reduction of the concentration of released Zn2+ might be responsible for the compromised ZnO NPs toxicity. However, the presence of extremely dosed ammonia at 200 mg/L imposed restrictions on further alleviation of ZnO NPs toxicities probably due to the production of free ammonia and acclamation of nitrite. All these findings would provide new insights for risk assessment of the combined effects of NPs with other co-existing contaminants in the BNR system.

Abstract:Numerous researches have been carried out to study the effects of emerging contaminants in wastewater, such as antibiotics, nanomaterials, heavy metals, and microplastics, on the anammox process. However, they are fragmented and difficult to provide a comprehensive understanding of their effects on reactor performance and the metabolic mechanisms in anammox bacteria. Therefore, this paper overviews the effects on anammox processes by the introduced emerging contaminants in the past years to fulfill such knowledge gaps that affect our perception of the inhibitory mechanisms and limit the optimization of the anammox process. In detail, their effects on anammox processes from the aspects of reactor performance, microbial community, antibiotic resistance genes (ARGs), and functional genes related to anammox and nitrogen transformation in anammox consortia are summarized. Furthermore, the metabolic mechanisms causing the cell death of anammox bacteria, such as induction of reactive oxygen species, limitation of substrates diffusion, and membrane binding are proposed. By offering this review, the remaining research gaps are identified, and the potential metabolic mechanisms in anammox consortia are highlighted.

Abstract:The increasing use of engineered metal oxide nanoparticles (MONPs) in consumer products raises great concerns about their environmental impacts, but their potential impacts on anaerobic ammonium oxidation (anammox) process in wastewater treatment remain unclear. In this study, the presence of MONPs (1, 50, 200 mg L-1) exhibited no visible effects on the nitrogen removal performance of anammox reactors, but high levels (200 mg L-1) of SiO(2)NPs, Al(2)O(3)NPs and CeO(2)NPs had a distinct effect on shaping the anammox community. Long-term exposure of MONPs caused different responses in the relative abundance of Ca. Kuenenia, the level of functional gene HzsA and the activities of three key enzymes involved in anammox metabolism, but no significant inhibition effects on specific anammox activity were detected. Overall, the effects of MONPs on anammox community structure and sludge properties depended on their types and levels and followed the order SiO2 > GeO2 > A1(2)O(3) > TiO2.

Abstract:Engineered nano-ZnO is extensively utilized in both production and daily life, leading to its inevitable entry into the wastewater treatment system through various pathways. Nitrogen removal microorganisms in wastewater treatment systems are highly susceptible to environmental impacts. The antibacterial properties of nano-ZnO can impede the biological nitrogen removal (BNR) process and adversely affect the nitrogen removal performance. A comprehensive understanding of the inhibitory effect and mechanism of nano-ZnO on the BNR process is crucial in devising appropriate countermeasures to ensure optimal nitrogen removal performance. This review provides an overview of the sources of nano-ZnO in the environment, its impact on the BNR process, and the inhibition mechanism, and proposes potential methods that can mitigate the inhibitory effect of nano-ZnO. Additionally, future prospects are also discussed. This review serves as a foundation for a deeper understanding of the inhibition of engineered nano-ZnO on the BNR process and aids in guiding efforts to maintain the nitrogen removal performance in the presence of engineered nano-ZnO.
Underlying Promotion Mechanism of High Concentration of Silver Nanoparticles on Anammox Process.

Abstract:Silver nanoparticles (AgNPs) are largely discharged into sewers and mostly accumulate in the sediments and sludge. The toxicity of silver nanoparticles to environmental microorganisms has attracted great attention. However, the effect of AgNPs on anaerobic ammonium-oxidizing (anammox) granules remains unknown. Here we present the underlying promotion mechanism of AgNPs on anammox granules from a morphological and molecular biology perspective. Our results demonstrate a positive effect of AgNPs on the proliferation of anammox bacteria. AgNPs resulted in a change in the three-dimensional structure of anammox granules and led to larger pore size and higher porosity. In addition, the diffusion capacity of the substrate and metal ions was enhanced. In addition, the expression of anammox-related enzymes, such as nitrite oxidoreductase (NirS), hydrazine dehydrogenase (Hdh), and hydrazine synthase (HZS) was upregulated. Therefore, the growth rate and the nitrogen removal performance of the anammox granules were improved. Our findings clarify the underlying mechanism of AgNPs on anammox granules and provide a promising method for the treatment of AgNPs-rich wastewater.
Short-term Impacts of Cu, CuO, ZnO and Ag Nanoparticles (nps) on Anammox Sludge: CuNPs Make a Difference.

Abstract:The increasing application of engineered nanoparticles (NPs) has posed an emerging challenge to wastewater treatment processes. The short-term impacts of CuNPs, CuONPs, ZnONPs and AgNPs on anaerobic ammonium oxidation (anammox) process were investigated firstly in this study. CuONPs, ZnONPs and AgNPs up to 50 mg g(-1) suspended solid (SS) did not affect anammox activity, reactive oxygen species (ROS) production or cell membrane integrity. However, 1.25 mg g(-1) SS CuNPs significantly inhibited the anammox activity and the loads that caused 50% inhibition were 4.64 +/- 1.24 and 3.27 +/- 0.79 mg g(-1) SS for anammox granules and flocs, respectively. 5 mg g(-1) SS CuNPs caused serious accumulation of the toxic intermediate N2H4. Furthermore, CuNPs interacted with extracellular polymeric substances by specifically bonding to tyrosine or tryptophan-containing groups, C-O-C in polysaccharides and -OH in polymeric compounds. Abstract:Nano materials have been widely used in various industrial production field and wastewater treatment process due to their unique physical and chemical properties. When nano materials enter into water treatment facilities, they may affect the physicochemical properties of sludge and microbial communities of the therein sludge, thus affect the performance and stability of the biological treatment. The influence of the nano metal and its metallic oxide on characteristics of activated sludge, aerobic granular sludge and anaerobic granular sludge were summarized from aspects of removal of organic matters, ammonia nitrogen and total nitrogen, and methane production of anaerobic granular sludge. The future development direction of nano metal and its metallic oxide was proposed, which could provide a reference for the intensive study of the potential impact of the said kind of material on biological sewage treatment.

Abstract:As the technological benefits of nanotechnology begin to rapidly move from laboratory to large-scale industrial application, release of nanomaterials to the environment including water, soil, sediment, and biosolids is inevitable. Concerns have been raised that the special properties of nanostructured materials that make them so attractive could potentially lead to unforeseen health risk or environmental hazards. The progress on the sources of nanoparticles, physicochemical properties and environmental or health risk is critically reviewed in this work. The environmental behavior of nanoparticles is analyzed when they are released into the aqueous environment. Toxicological effects of typical nanoparticles on bacteria, aquatic organisms, soil animals and mammalian cells are summarized. The possible toxicological mechanisms on these model microbes are then discussed. Furthermore, the relationships between toxicological effects of nanoparticles on model microbe and self-physicochemical/environmental factors are critically analyzed. The challenges and existing problems of nanotoxicity research are listed. Moreover, we also highlight the future research fields (such as quantitative structure activity relationship, QSAR) need regarding nanoparticles in the environment. It has been point out that more studies are urgently needed to scrutinize the characteristic methods of nanoscale materials and long-term research on the behavior and toxicity of nanoparticles in microbial community.

Abstract:Previous studies investigating the risk of engineered nanoparticles (NPs) to biological wastewater treatment have primarily tested NPs individually; however, limited data are available on the impact of NPs on the anaerobic ammonium oxidation (anammox) process. In this study, the toxicity of CuNPs on anammox granules was investigated individually and in combination with CuONPs or ZnONPs. Exposure to CuNPs at 5mgg-1 suspended solids (SS) decreased the anammox activity to 47.1±8.5%, increased the lactate dehydrogenase level to 110.5±3.4% and increased the extracellular N2H4 concentration by 16-fold but did not cause oxidative stress. The presence of CuONPs or ZnONPs at 5mgg-1 SS did not significantly aggravate or alleviate the toxicity of the CuNPs; however, the introduction of EDTA or S2- could attenuate the adverse effects of the CuNPs, CuONPs and ZnONPs on the anammox granules. EDTA captured Cu ions, whereas S2- shielded and deactivated Cu ions and passivated CuNPs. Therefore, our results indicated that the toxicity of NPs was dependent on the amount of active metal reaching the anammox cells. Overall, the results of this study have filled knowledge gaps and provided insights into the combined toxicity of NPs on anammox biomass.

Abstract:Metal and metal oxide engineered nano particles (MMO-ENPs) are widely applied in various industries due to their unique properties. Thus, many researches focused on the influence on nitrogen transformation processes by MMO-ENPs. This review focuses on the effect of MMO-ENPs on nitrogen fixation, nitrification, denitrification and Anammox. Firstly, based on most of the researches, it can be concluded MMO-ENPs have negative effect on nitrogen fixation, nitrification and denitrification while the MMO-ENPs have no promotion effect on Anammox. Then, the influence factors are discussed in detail, including MMO-ENPs dosage, MMO-ENPs kind and exposure time. Both the microbial morphology and population structure were altered by MMO-ENPs. Also, the mechanisms of MMO-ENPs affecting the nitrogen transformation are reviewed. The inhibition of key enzymes and functional genes, the promotion of reactive oxygen species (ROS) production, MMO-ENPs themselves and the suppression of electron transfer all contribute to the negative effect. Finally, the key points for future investigation are proposed that more attention should be attached to the effect on Anammox and the further mechanism in the future studies.

Anaerobic digestion (AD) is a major technology treating organic-rich wastes such as sewage sludge, kitchen waste, and animal manure. AD allows to convert waste into energy such as biogas and electricity, and resources such as fertilizers. Nevertheless, biogas production from organic waste is limited by the low biodegradability of resistant organic matter and the short retention time in AD bioreactors. As a consequence, recent researches have focused on the use of additives to improve the performance of AD. For example, nanomaterials are increasingly used as additives due to their properties in the promotion of direct inter-species electron transfer between microorganisms, and the improvement of the microbial community structure. In this study, we review firstly the contribution of nanomaterials to system stability, digestive performance and risk control of AD processes systematically. Then we sorted the enhanced performances by co-digestion of nano-magnetite with sewage sludge, kitchen waste, livestock and poultry manure thoroughly. Finally we discussed mechanisms on how nanomaterials such as iron- and carbon-based nanomaterials addition could enhance production of volatile fatty acids, and inhibit the production of hydrogen sulfide and the transmission of antibiotic resistance genes simultaneously.
