Abstract:This work investigated the roles of sludge micropowder addition in microbial structure and partial nitrification and denitrification (PND) in an extended filamentous aerobic granular sludge-sequencing batch reactor (AGS-SBR) using high-ammonia wastewater. Type 1683 Acinetobacter with a high percentage became the dominant extended filaments, remarkably shifted and remained at a low level, acting as a framework for AGS recovery after micropowder addition. The sludge volume index (SVI5) decreased from 114 to 41.7 mL/g, mixed liquid suspended solids (MLSS) and extracellular polymers (EPS) both increased and balanced at 6836 mg/L and 113.4 mg/g•MLVSS, respectively. COD and NH4+-N were degraded to certain degrees in the end. However, the effluent NO2−-N accumulated to the peak value of 97.6 mg/L on day 100 (aeration stage), then decreased and remained at 45.3 mg/L with development of the stirring and micropowder supplemented in the SBR on day 160 (anoxic stage), while the influent NO2−-N always remained at zero. Interestingly, the influent/effluent NO3−-N both remained at zero throughout the whole experiment. These results demonstrated that PND was successfully obtained in this work. Sludge micropowder addition not only restrained the extended filaments’ overgrowth, but also contributed to PND realization with carbon released. Citrobacter and Thauera played an essential role in the PND process for high-ammonia wastewater treatment. Running condition, wastewater characteristic, and sludge structure played an important role in microbial composition.

Abstract:Extracellular polymeric substances (EPS) play a key role in anaerobic digestion of excess sludge . The influence of EPS removal and re-addition on sludge anaerobic digestion were investigated by batch experiment. The results showed that both EPS removal and re-addition improved sludge anaerobic digestion, the cumulative methane production increased by 14.9 % and 50.4 %. EPS removal promoted sludge hydrolysis and acidification, the soluble chemical oxygen demand (SCOD) and volatile fatty acids (VFAs) concentration increased by 31 % and 29 %. The re-addition of EPS increased the substrate of sludge fermentation, resulting in 1.64 and 1.77 times higher SCOD and VFAs concentration than that of control group. In addition, EPS contained lots of electrochemically active substances, and re-adding EPS increased the electron transfer system activity in anaerobic digestion by 164 %, which enhanced significantly methane production. Both removal and re-addition of EPS altered the microbial community structure in anaerobic digestion, the bacteria with hydrolysis and acidification functions were enriched, such as Chloroflexi and Desulfobacterota . Methanobacterium and Methanosaeta were the dominant methanogens in anaerobic digestion systems, and their relative abundance were increased by removing and re-adding EPS.

Abstract:This study investigated the effect of N-acyl-L-homoserine lactones (AHLs) and EPS on anammox activity, and identified the relationship between AHLs and EPS. The effects of AHLs and different EPS fractions (loosely bound (LB) and tightly bound (TB) EPS) from denitrification sludge (DS) and perchlorate reducing sludge (PRS) on anammox activity were conducted by batch experiments. Compared with the control experiment, the nitrogen removal rate was increased by 82.0%, 73.9% and 73.0%, respectively, when adding DS-EPS (integral DS EPS), PRS-EPS (integral PRS EPS) and DS-LB in anammox biomass. EPS fractions of DS and PRS contained the same kinds of AHLs, including C4-HSL, C6-HSL and C8-HSL, as identified by HPLC-MS. C4-HSL and C6-HSL at 0.1 mg L−1 concentration effectively improved the nitrogen removal rate by induceing the expression of anammox active genes during the anammox process. This study provides a new idea for efficient nitrogen removal from anammox bacteria.

Abstract:In this manuscript,the influence of the aeration intensity on the pollutant removal efficiency,the activity of dehydrogenases,components of extracellular polymeric substances（EPS）,Zeta potential,average size of sludge flocs and membrane fouling was investigated in MBR treatment of synthetic municipal wastewater.The results showed that,with the decrease in aeration intensity,COD removal revealed little variation（〉94%）.The activity of dehydrogenases and VSS/SS ratio dropped.In addition,sludge LB-EPS content increased,but both Zeta potential and the size of sludge flocs declined.The membrane fouling tended to be heavy.With 800—400 L · m-2 · h-1 aeration intensity examined,hydraulic shearing force was not a dominant contribution to the floc size,but Zeta potential played a key role.However,it helped in alleviating membrane fouling.

Abstract:In order to investigate the characteristics of extracellular polymeric substances(EPS) of pulse aerated activated sludge and its relationship with denitrification efficiency.A study between loosely bound EPS (LB-EPS) and tightly bound EPS(TB-EPS) was carried out under the condition of the pulse aeration.Aerobic time was set for 5 min and anoxic time for 10 min.The relationship between EPS and denitrification efficiency was also analyzed.The results showed that the content of LB-EPS was lower than that of TB-EPS and the EPS content kept increasing during the whole pulse aeration acclimation process.Three-dimensional excitation-emission matrix (3D-EEM) fluorescence spectroscopy showed that the fluorescence intensity of tryptophan in LB-EPS had a red shift and the fluorescence intensity in TB-EPS increases indicating that the content of tryptophan protein had an increasing trend in the EPS of activated sludge with pulse aeration.The removal rate of ammonia nitrogen(NH4+-N) in pulse aeration reactor was about 80％,which was close to that of continuous aeration reactor.However,the removal rate of total nitrogen(TN) was about 50％,nearly 20％ higher than that of continuous aeration reactor.EPS has a detrimental effect on denitrification.There was a negative correlation between the EPS content and the removal rate of NH4+-N and TN under pulse aeration condition.The negative correlation that were significant between LB-EPS and the removal rate of TN.The correlation between TB-EPS and the removal rate of NH+4-N was also negative.

Abstract:Extracellular polymeric substances (EPS) play an important role in biological wastewater treatment. The influence of extracted EPS in aerobic activated sludge was investigated, with EPS extraction yields represented by total organic carbon. Polysaccharides and proteins in EPS were analyzed to the major component of EPS in activated sludge. Results showed that the difference of treatment technology and culture medium among activated sludge can induce the change of component and content of extracted EPS, especially the ratio between proteins and polysaccharides.

Abstract:In this study, role of extracellular polymeric substances (EPSs) in enhancing nitrogen-removal from ammonium-nitrogen-rich wastewater using aerobic granular sludge (AGS) technology were analyzed. AGS enabled ammonium oxidation and denitrification to occur simultaneously. Air stripping and simultaneous nitrification-denitrification contributed to total-nitrogen removal. Clone-library analysis revealed that close relatives of Nitrosomonas eutropha and heterotrophic denitrifiers were dominant in the AGS, whereas anammox bacteria were not detected. EPSs adsorption of ammonium, nitrite, and nitrate nitrogen results in improved removal of nitrogen in batch experiments.

Abstract:The effects of aeration regimes (intermittent and continuous aeration) on nitritation performance and biofilm EPS composition were evaluated in moving bed biofilm reactors (MBBRs), and a hypothesis that the aeration regimes affect EPS composition by affecting the microbial activity and sludge discharge content was proposed. The effluent NO2-/NH4+ ratio corresponded to that of an anammox reaction (1.07 ± 0.20) for the MBBR with continuous aeration (MBBRcon.), while that in the MBBR with intermittent aeration (20 min on/15 min off) (MBBRint.) was relatively lower (0.75 ± 0.19). Furthermore, the activity of ammonia-oxidizing bacteria in MBBRcon. was 0.4-7.9 mg-N·L-1·h-1 more than that in MBBRint., which was consistent with the lower proportion of dead cells in MBBRcon. compared with MBBRint. (9.4% vs. 31.8%). The higher microbial activity in MBBRcon. led to more sludge discharge than MBBRint., which was reflected in the higher biofilm detachment rate in MBBRcon. compared with MBBRint. (0.15 ± 0.02 vs. 0.11 ± 0.02 g m-2·d-1). The ratio of humic substances to polysaccharides in the EPS was high (0.96 ± 0.08) in the detachment biomass, while the ratios in the nitritation biofilm on carriers from MBBRcon. and MBBRint. were 0.52 ± 0.13 and 0.72 ± 0.16, respectively. We hypothesized that biofilm matrix with high ratios of humic substances to polysaccharides are structurally unstable and prone to fall off. In addition, the higher proportion of dead cells in MBBRint. made the proportion of humic substances in EPS high. Meanwhile, less sludge discharge in MBBRint. than MBBRcon. caused more humic substances to accumulate in the biofilm. These was responsible for the higher ratio of humic substances to polysaccharides in MBBRint. compared with MBBRcon. The findings elucidate the connection between aeration regimes and biofilm EPS composition, and guide the choice of aeration regimes in the design of biofilm reactors for partial nitritation.

Abstract:The nitrogen pollution has caused serious environmental problems. The main aims of the present study were to remove the nitrogen from wastewater through ANAMMOX (ANaerobic AMMonium OXidation) process, and let the effluent meet the discharge standard which was significant and advance in the field of ANAMMOX application which was also very important to the environmental protection and water recycling. This study reports the performance of ANAMMOX-EGSB (Expanded Granular Sludge Bed) reactor after feeding with synthetic wastewater. The results showed that the effluent ammonium concentration was 11.90mgN/L and the nitrogen remove efficiency (NRE) reached up to 94.68% when the volumetric nitrogen loading rate (NLR) and volumetric nitrogen removal rate (NRR) were as high as 27.31 and 25.86kgN/(m3·d), respectively. The effluent ammonium concentration meets the Chinese integrated wastewater discharge standard (GB8978-1996) in which the ammonium concentration should be lower than 15mgN/L. The maximum ammonium, nitrite and nitrogen conversion rates (qmax) by the ANAMMOX granular sludge in the reactor were 907.13, 841.76, 1810.10mgN/(g VSS·d) and half saturation constants (KS) were 2.69, 0.44, 3.11mgN/L, respectively.
Effect of Extracellular Polymeric Substances Removal and Re-Addition on the Denitrification Performance of Activated Sludge: Carbon Source Metabolism, Electron Transfer and Enzyme Activity

Abstract:Extracellular polymeric substances (EPS) play an important role in the metabolic activities of activated sludge. The effect of EPS removal (pellet group) and re-addition (pellet + EPS group) on the denitrification of activated sludge were analyzed in this study. The results indicated that EPS removal deteriorated the denitrification performance of the activated sludge. The denitrification rates in the pellet group with EPS extraction degrees of 7.77, 12.37, 35.89, and 100% were only 82.6, 75.7, 64.1, and 26.9%, respectively, of the control group. However, the denitrification rate of the pellet increased by 19–34% by re-adding the extracted EPS. Moreover, when the EPS extraction degree was 7.77%, the denitrification rate of the pellet + EPS group was 10.4% higher than that of the control group. The EPS contain many redox mediators. The electron transport system activity (ETSA) and key enzymes in biological denitrification were improved by re-adding EPS. The ETSA in the pellet + EPS group were 1.66-fold that in control group. The pellet + EPS group had a lower reaction activation energy, which was 42.9% lower with that of the control group. In addition, EPS re-addition promoted the activities of enzymes related to glycolysis by 9.60–18.08%, and nicotinamide adenine dinucleotide yield increased by 6.5%, which enhanced the denitrification rate of activated sludge. This study provides a novel method to enhance the denitrification performance of activated sludge by in situ appropriate desorption of EPS as endogenous redox mediator.
Decoding the Role of Extracellular Polymeric Substances in Enhancing Nitrogen Removal from High-Ammonia and Low-C/N Wastewater in a Sequencing Batch Packed-Bed Biofilm Reactor

Abstract:Although the role of extracellular polymeric substances (EPSs) as a viscous high-molecular polymer in biological wastewater treatment has been recognized, in-depth knowledge of how EPSs affect nitrogen removal remains limited in biofilm-based reactors. Herein, we explored EPS characteristics associated with nitrogen removal from high-ammonia (NH4+-N: 300 mg/L) and low carbon-to-nitrogen ratio (C/N: 2–3) wastewater in a sequencing batch packed-bed biofilm reactor (SBPBBR) under four different operating scenarios for a total of 112 cycles. Scanning electron microscopy (SEM), atomic force microscopy (AFM), and Fourier-transform infrared (FTIR) analysis revealed that the distinct physicochemical properties, interface microstructure, and chemical composition of the bio-carrier were conducive to biofilm formation and microbial immobilization and enrichment. Under the optimal conditions (C/N: 3, dissolved oxygen: 1.3 mg/L, and cycle time: 12 h), 88.9% ammonia removal efficiency (ARE) and 81.9% nitrogen removal efficiency (NRE) could be achieved in the SBPBBR. Based on visual and SEM observations of the bio-carriers, biofilm development, biomass concentration, and microbial morphology were closely linked with nitrogen removal performance. Moreover, FTIR and three-dimensional excitation–emission matrix (3D-EEM) spectroscopy demonstrated that tightly bound EPSs (TB-EPSs) play a more important role in maintaining the stability of the biofilm. Significant shifts in the number, intensity, and position of fluorescence peaks of EPSs determined different nitrogen removal. More importantly, the high presence of tryptophan proteins and humic acids might promote advanced nitrogen removal. These findings uncover intrinsic correlations between EPSs and nitrogen removal for better controlling and optimizing biofilm reactors.

Fluorescent Characteristics and Metal Binding Properties of Different Molecular Weight Fractions in Stratified Extracellular Polymeric Substances of Activated Sludge

Abstract:The combination of heavy metals and extracellular polymeric substances (EPSs) affects the distribution of heavy metals in microbial aggregates, soil, and aquatic systems. This paper aimed to explore the binding mechanisms of EPSs of different molecular weights in activated sludge with heavy metals. We extracted the stratification components of activated sludge EPSs and divided the components into five fractions of different molecular weight ranges. In the three-dimensional fluorescence analysis of each fraction, the EPSs of activated sludge had two peaks, peak A (representing low-excitation tryptophan) and peak B (representing high-excitation tryptophan), and static quenching was the main reason for the fluorescence quenching between the compounds attributable to peak A in activated sludge EPSs and Pb2+ and Cu2+. Further exploration suggested that the EPSs of activated sludge interacted with Cd2+, Pb2+, Cu2+, and Zn2+ to form new substances. The quenching effect of the EPSs with the highest molecular weight (100 kDa–0.7 μm) was more significant, and the binding ability was more stable. This study implies that the application of EPSs from activated sludge is promising. While effectively binding heavy metals, it can also reduce the volume of the excess activated sludge.

Abstract:Extensive use of nanoparticles (NPs) in consumer and industrial products has led to concerns about their potential environmental impacts; however, the influences of different NPs (e.g., nZVI (nano zero-valent iron), Ag NPs, Fe2O3 NPs and MgO NPs) on the anaerobic digestion of sludge have not yet been studied in depth. Additionally, a new guideline or the use of different NPs in the anaerobic digestion of sludge should be established to improve the anaerobic digestion of sludge and avoid inhibitory effects. This study investigated the effects of four representative NPs (i.e., nZVI, Ag NPs, Fe2O3 NPs and MgO NPs) on methane production during the anaerobic digestion of waste activated sludge (WAS). The presence of 10 mg/g total suspended solids (TSS) nZVI and 100 mg/g TSS Fe2O3 NPs increased methane production to 120% and 117% of the control, respectively, whereas 500 mg/g TSS Ag NPs and 500 mg/g TSS MgO NPs generated lower levels of methane production (73.52% and 1.08% that of the control, respectively). These results showed that low concentrations of nZVI and Fe2O3 NPs promoted the amount of microbes (Bacteria and Archaea) and activities of key enzymes but that higher concentrations of Ag NPs and MgO NPs inhibited them.

Abstract:Extracellular polymeric substances (EPS) in aerobic granular sludge (AGS) play an important role in the removal of heavy metals. In this study, EPS with high protein content (86.8%) were extracted from AGS using a cation exchange resin extraction method. EPS exhibited large adsorption capacities for Pb(II), Cd(II) and Zn(II) of 1587.3, 1470.6 and 1123.6 mg/g respectively, as calculated by the Langmuir isotherm model. The excellent biosorption performance of EPS was closely related to the large concentrations of carboxyl (0.615 mmol/g) and hydroxyl groups (0.525 mmol/g) located on protein surfaces. FTIR and XPS analysis demonstrated that the dominant adsorption mechanism was complexation by carboxyl and hydroxyl groups. Moreover, extracted EPS, both with hydrophobic and hydrophilic groups, were found to comprise an amphiphilic material, which could also capture metal ions through electrostatic attraction and ion-exchange. Furthermore, the heavy metal ions could be efficiently desorbed from EPS under acidic conditions and reused. The study indicates that protein in EPS filled with carboxyl and hydroxyl groups is key to the heavy-metal adsorption process. The results provide basic guidance for the full utilization of EPS in AGS for bioremediation in wastewater treatment. (C) 2015 Elsevier Ltd. All rights reserved.

Abstract:The amount of sewage sludge generated from wastewater treatment plants globally is unavoidably increasing. In recent years, significant attention has been paid to the biorefinery concept based on the conversion of waste streams to high-value products, material, and energy by microorganisms. However, one of the most significant challenges in the field is the possibility of controlling the microorganisms’ pathways in the anaerobic environment. This study investigated two different anaerobic fermentation tests carried out with real waste activated sludge at high organic loading rate (10 g COD L−1d−1) and short hydraulic retention time (HRT) to comprehensively understand whether this configuration enhances extracellular polymeric substance (EPS) and metal solubilisation. The quantity of EPS recovered increased over time, while the chemical oxygen demand to EPS ratio remained in the range 1.31–1.45. Slightly acidic conditions and sludge floc disintegration promoted EPS matrix disruption and release, combined with the solubilisation of organically bound toxic metals, such as As, Be, Cu, Ni, V, and Zn, thereby increasing the overall metal removal efficiency due to the action of hydrolytic microorganisms. Bacteroidetes, Firmicutes, and Chloroflexi were the most abundant phyla observed, indicating that the short HRT imposed on the systems favoured the hydrolytic and acidogenic activity of these taxa.

Abstract:Extracellular polymeric substances (EPS) extracted from waste activated sludge were used as endogenous redox mediator to enhance denitrification performance. The nitrate reduction rate increased 1.42-fold when EPS were added at 75 mg C/L (C represents total organic carbon). EPS addition decreased the charge transfer resistance and improved the electron transport system activity. The nitrate reductase and nitrite reductase activities improved by 29.7% and 25.4%, respectively. The activation energy of the system with EPS addition (25.82 kJ/mol) was 31.1% lower than that of the control group (37.49 kJ/mol). Besides, EPS could be used as electron carriers to accelerate electron transport; its primary role was similar to that of the quinone loop in the electron transfer chain. More importantly, EPS addition enhanced carbon source metabolism, which increased the available nicotinamide adenine dinucleotide yield to 1.21 times that of the control group, and thus promoted the denitrification performance of activated sludge.

Abstract:Introducing carbon sources to achieve nitrogen removal from mature landfill leachate not only increases the costs and carbon emissions but also inhibits the activity of autotrophic bacteria. Thus, this study constructed a double anammox system that combines partial nitrification-anammox (PNA) and endogenous partial denitrification-anammox (EPDA) within an integrated fixed-film activated sludge (IFAS) reactor. In this system, PNA primarily contributes to nitrogen removal pathways, achieving a nitrite accumulation rate of 98.23%. The production of extracellular polymer substances (EPS) in the IFAS reactor is stimulated by introducing co-fermentation liquid. Through the utilization of EPS, the system effectively achieves EPDA with the nitrite transformation rate of 97.20%. Under the intermittent aeration operation strategy, EPDA combined with PNA and anammox in the oxic and anoxic stages enhanced the nitrogen removal efficiency of the system to 99.70 ± 0.12%. The functional genus Candidatus kuenenia became enriched in biofilm sludge, while Thauera and Nitrosomonas predominated in floc sludge.

Abstract:As a kind of new biosorbent,aerobic granular sludge which has the advantages of strong adsorption capacity and easily-desorbing property,was paid great attention in the area of heavy metal waste water treatment.In this research,the adsorption behaviors of Pb2+ and Cd2+ by extracellular polymeric substances(EPS) which extracted from aerobic granular sludge using improved centrifugation methods were studied.The results showed that the adsorption capacity of Pb2+ and Cd2+ on EPS was considerably large.The experimental results were fit well with the Langmuir iostherms model.The maximum adsorption capacity for Pb2+and Cd2+by EPS was up to 534.76 mg/g and 478.47 mg/g,respectively.Furthermore,the competitive adsorption experiments presented that EPS exhibited a selective adsorption for Pb2+.Specially,there was a minor inhibition for Pb2+ adsorption in presences of Cd2+,while the adsorption capacity of Cd2+ was greatly decreased when Pb2+ co-existed in solutions.FT-IR and EEM analysis revealed that EPS was rich in hydrophilic groups and hydrophobic groups,which was benefit to combine heavy metals by complexation,ions exchange and chelation.The adsorption of Pb2+ and Cd2+ by EPS was mainly attributed to the —COOH,—NH2,—CH,—OH and —C=O groups from protein fractions of them.The main adsorption mechanism of Pb2+ by EPS was ion exchange and complexation,while for Cd2+ was mainly took effect by complexation.

Abstract:Anaerobic ammonium oxidation (Anammox) is expected to play a promising role in treating low C/N ratio wastewater. In this study, nitrogen removal performance and the effects of extracellular polymeric substances (EPS) on anammox activity were investigated in two different reactors, followed by analysis of 16S rRNA high-throughput sequencing. A sequencing batch reactor (SBR) and an up-flow anammox sludge blanket reactor (UASB) operated for 133 days, with total nitrogen removal efficiencies of 82.09 ± 4.76% and 87.91 ± 2.59% in the phase 3, respectively. The EPS concentration in UASB was about 1.6 times higher than that in SBR. Also, in the batch tests, especially the anammox-EPS of UASB (AU-EPS) additions contributed to better nitrogen removal capability for anammox bacteria (AnAOB). The relative abundance of AnAOB in UASB was observed to reach 19.68% in the end, accounting for a relatively high degree in the whole community. In SBR, the effluent NH4+-N concentration increased when increasing the substrate concentration, followed by the decrease of the relative abundance of AnAOB. Thus, the different feeding strategies would lead to different granulation styles, further influencing the activity and stability of anammox system. This research aims to get a better understanding of the different mechanisms associated with AnAOB enrichment strategies.

Effects of Waste Activated Sludge Extracellular Polymeric Substances on Biosorption

Abstract:Extracellular polymeric substances (EPS) reportedly make up approximately half of the organic matter in activated sludge (AS), and therefore strongly influence AS properties. This study evaluated the component fractions of EPS normalized to volatile suspended solids (VSS) in waste activated sludge (WAS) from a trickling-filter-solids contact process (TF/SC) and its ability to biosorb organic matter from raw wastewater with 30 min of contact time. Biosorption is the process in which organic matter (carbohydrates, proteins, humic acids, DNA, uronic acids, and lipids) in a sorbate, such as raw wastewater, sorbs onto a sorbent such as WAS. A statistically significant correlation was found between both the total concentration of EPS and the proteins component of the EPS and the biosorption removal of soluble chemical oxygen demand (sCOD) and truly soluble COD (ffCOD). Thus, the biosorption of soluble forms of COD can accurately be predicted by quantifying just the amount of proteins in WAS-associated EPS. No significant correlations were found for the biosorption of colloidal COD (cCOD). WAS biosorbed 45–75 mg L−1 of COD in 30 min. WAS absorbed or stored the proteins fraction of the soluble microbial products (SMP) during the biosorption process. Higher concentrations of humic acids were found in the biosorption process effluent than in the untreated wastewater, which warrants further study. Longer cation exchange resin (CER) extraction times yielded more total EPS from the sludge: 90 ± 9, 158 ± 3, and 316 ± 44 mg g−1 VSS, for 45-min, 4-h, and 24-h extraction times, respectively. Thus, EPS extracted represented only 9%, 15.8%, and 31.6% of the VSS, respectively, raising questions about whether the accurate characterization of EPS can be performed using the typical extraction time of 45 min due to different extraction rates for different components. It was found that the humic acids fraction was extracted much more slowly than the other fractions.
