Abstract:Arsenic is an element widely present in nature. Additionally, it may be found as different species in several matrices and therefore it is one of the target elements in chemical speciation. Although the number of studies in terrestrial plants is low, compared to matrices such as fish or urine, this number is raising due to the fact that this type of matrix are closely related to the human food chain. In speciation analysis, sample preparation is a critical step and several extraction procedures present drawbacks. In this review, papers dealing with extraction procedures, analytical methods, and studies of species conservation in plants cultivated in terrestrial environment are critically discussed. Analytical procedures based on extractions using water or diluted acid solutions associated with HPLC-ICP-MS are good alternatives, owing to their versatility and sensitivity, even though less expensive strategies are shown as feasible choices.

Abstract:It is known that arsenic has different toxicological properties dependent upon both its oxidation state for inorganic compounds, as well as the different toxicity levels exhibited for organic arsenic compounds. The field of arsenic speciation analysis has grown rapidly in the recent years, especially with the utilization of chromatographic analysis. Extraction method as sequential extraction combined to microwave assisted digestion were applied to water and sediments samples. This paper describes the essential background and toxicity of arsenic in environment, and more importantly, some currently used chromatographic applications and sample handling procedures necessary to accurately detect arsenic in its various forms. Experiments and work using ultra high-performance liquid chromatography and inductively coupled plasma optical emission spectrometry for arsenic speciation of environmental samples are presented in this research.

Abstract:Long term exposure to arsenic through consumption of contaminated groundwater has been a global issue since the last five decades; while from an alternate standpoint, arsenic compounds have emerged as unparallel chemotherapeutic drugs. This review highlights the contribution from arsenic speciation studies that have played a pivotal role in the progression of our understanding of the biological behaviour of arsenic in humans. We also discuss the limitations of the speciation studies and their association with the interpretation of arsenic metabolism. Chromatographic separation followed by spectroscopic detection as well as the utilization of biotinylated pull-down assays, protein microarray and radiolabelled arsenic have been instrumental in identifying hundreds of metabolic arsenic conjugates, while, computational modelling has predicted thousands of them. However, these species exhibit a variegated pattern, which supports more than one hypothesis for the metabolic pathway of arsenic. Thus, the arsenic species are yet to be integrated into a coherent mechanistic pathway depicting its chemicobiological fate. Novel biorelevant arsenic species have been identified due to significant evolution in experimental methodologies. However, these methods are specific for the identification of only a group of arsenicals sharing similar physiochemical properties; and may not be applicable to other constituents of the vast spectrum of arsenic species. Consequently, the identity of arsenic binding partners in vivo and the sequence of events in arsenic metabolism are still elusive. This resonates the need for additional focus on the extraction and characterization of both low and high molecular weight arsenicals in a combinative manner.
Arsenic speciation based on ion exchange high-performance liquid chromatography hyphenated with hydride generation atomic fluorescence and on-line UV photo oxidation

Abstract:Abstract An on-line method capable of the separation of arsenic species was developed for the speciation of arsenite As(III), arsenate As(V), monomethylarsenic (MMA) and dimethylarsenic acid (DMA) in biological samples. The method is based on the combination of high-performance liquid chromatograph (HPLC) for separation, UV photo oxidation for sample digestion and hydride generation atomic fluorescence spectrometry (HGAFS) for sensitive detection. The best separation results were obtained with an anion-exchange AS11 column protected by an AG11 guard column, and gradient elution with NaH2PO4 and water as mobile phase. The on-line UV photo oxidation with 1.5% K2S2O8 in 0.2 mol L–1 NaOH in an 8 m PTFE coil for 40 s ensures the digestion of organoarsenic compounds. Detection limits for the four species were in the range of 0.11–0.15 ng (20 μL injected). Procedures were validated by analysis of the certified reference materials GBW09103 freeze-dried human urine and the results were in good agreement with the certified values of total arsenic concentration. The method has been successfully applied to speciation studies of blood arsenic species with no need of sample pretreatment. Speciation of arsenic in blood samples collected from two patients after the ingestion of realgar-containing drug reveals slight increase of arsenite and DMA, resulting from the digestion of realgar.

Abstract:Arsenic is a hazardous element that causes environmental pollution. Due to its toxicological effects, it is crucial to quantify and minimize the hazardous impact on the ecology. Despite the significant advances in analytical techniques, sample preparation is still crucial for determining target analytes in complex matrices. Several factors affect the direct analysis, such as trace-level analysis, advanced regulatory requirements, complexity of sample matrices, and incompatible with analytical instrumentation. Along with the development in the sample preparation process, microextraction methods play an essential role in the sample preparation process. Microextraction techniques (METs) are the newest green approach that replaces traditional sample preparation and preconcentration methods. METs have minimized the limitation of conventional sample preparation methods while keeping all their benefits. METs improve extraction efficacy, are fast, automated, use less amount of solvents, and are suitable for the environment. Microextraction techniques with less solvent consumption, such as solid phase microextraction (SPME) solvent-free methods, and liquid phase microextraction (LPME), are widely used in modern analytical procedures. SPME development focuses on synthesizing new sorbents and applying online sample preparation, whereas LPME research investigates the utilization of new solvents.

Abstract:The toxic and carcinogenic properties of inorganic and organic arsenic species make their determination in natural water vitally important. Determination of individual inorganic and organic arsenic species is critical because the toxicology, mobility, and adsorptivity vary substantially. Several methods for the speciation of arsenic in groundwater, surface-water, and acid mine drainage sample matrices using field and laboratory techniques are presented. The methods provide quantitative determination of arsenite [As(III)], arsenate [As(V)], monomethylarsonate (MMA), dimethylarsinate (DMA), and roxarsone in 2-8 min at detection limits of less than 1 microg arsenic per liter (microg As L(-1)). All the methods use anion exchange chromatography to separate the arsenic species and inductively coupled plasma-mass spectrometry as an arsenic-specific detector. Different methods were needed because some sample matrices did not have all arsenic species present or were incompatible with particular high-performance liquid chromatography (HPLC) mobile phases. The bias and variability of the methods were evaluated using total arsenic, As(III), As(V), DMA, and MMA results from more than 100 surface-water, groundwater, and acid mine drainage samples, and reference materials. Concentrations in test samples were as much as 13,000 microg As L(-1) for As(III) and 3700 microg As L(-1) for As(V). Methylated arsenic species were less than 100 microg As L(-1) and were found only in certain surface-water samples, and roxarsone was not detected in any of the water samples tested. The distribution of inorganic arsenic species in the test samples ranged from 0% to 90% As(III). Laboratory-speciation method variability for As(III), As(V), MMA, and DMA in reagent water at 0.5 microg As L(-1) was 8-13% (n=7). Field-speciation method variability for As(III) and As(V) at 1 microg As L(-1) in reagent water was 3-4% (n=3).

Abstract:Abstract Food and water are main sources of human exposure to arsenic. It is important to determine arsenic species in food because toxicities of arsenic vary greatly with its chemical speciation. Extensive research has focused on high concentrations of arsenic species in marine organisms. The concentrations of arsenic species in freshwater fish are much lower, and their determination presents analytical challenges. We summarize in this review the current state of knowledge on arsenic speciation in freshwater fish and discuss challenges and research needs. Fish samples are typically homogenized, and arsenic species extracted using water/methanol with the assistance of sonication and enzyme treatment. Arsenic species in the extracts are commonly separated using high performance liquid chromatography (HPLC) and detected using inductively coupled plasma mass spectrometry (ICPMS). Electrospray ionization tandem mass spectrometry (ESI MS/MS), used in combination with HPLC and ICPMS, provides complementary information for the identification and characterization of arsenic species. The methods and perspectives discussed in this review, covering sample preparation, chromatography separation, and mass spectrometry detection, are directed to arsenic speciation in freshwater fish and also applicable to studies of other food items. Despite progresses made in arsenic speciation analysis, a large fraction of total arsenic in freshwater fish remains unidentified. It is challenging to identify and quantify arsenic species present in complex sample matrices at very low concentrations. Further research is needed to improve the extraction efficiency, chromatographic resolution, detection sensitivity, and characterization capability.

Speciation analysis of arsenic in landfill leachate

Abstract:As environmental impacts of landfill last from beginning of cell filling to many years after, there is an increasing interest in monitoring landfill leachate composition especially with regards to metals and metalloids. High-performance liquid chromatography (HPLC) coupled with inductively coupled plasma mass spectrometry (ICP-MS) has been applied to the speciation of arsenic in landfill leachates. The difficulty is related to the complexity and heterogeneity of leachate matrices. A soft sample preparation protocol with water dilution and filtration of leachates has proved to be sufficient for the achievement of identification and quantification of arsenic species without matrix effect. The cationic-exchange separation method developed has enabled the detection of six arsenic species (AsIII, MMA, AsV, DMA, AsB, TMAO) in different landfill leachates. The wide range of concentrations of arsenic species (from 0.2 to 250 microg As L(-1)) and their repartition illustrate the high variability of these effluents depending on the nature of the wastes, the landfill management, the climatic conditions and the degradation phase, to list a few. These results provide new information about the chemical composition of these effluents which is useful to better adapt their treatment and to achieve the risk assessment of landfill management.

Abstract:For the past few years many studies have been performed to determine arsenic (As) speciation in drinking water, food chain and other environmental samples due to its well-recognized carcinogenic and toxic effects relating to its chemical forms and oxidation states. This review provides an overview of analytical methods, preconcentration and separation techniques, developed up to now, using HGAAS and ETAAS for the determination of inorganic As and organoarsenic species in environmental samples. Specific advantages, disadvantages, selectivity, sensitivity, efficiency, rapidity, detection limit (DL), and some aspects of recent improvements and modifications for different analytical and separation techniques, that can define their application for a particular sample analysis, are highlighted. HG-AAS has high sensitivity, selectivity and low DL using suitable separation techniques; and it is a more suitable, affordable and much less expensive technique than other detectors. The concentrations of HCl and NaBH(4) have a critical effect on the HG response of As species. Use of l-cysteine as pre-reductant is advantageous over KI to obtain the same signal response for different As species under the same, optimum and mild acid concentration, and to reduce the interference of transition metals on the arsine generation. Use of different pretreatment, digestion, separation techniques and surfactants can determine As species with DL from ngL(-1) to μgL(-1). Out of all the chromatographic techniques coupled with HGAAS/ETAAS, ion-pair reversed-phase chromatography (IP-RP) is the most popular due to its higher separation efficiency, resolution selectivity, simplicity, and ability to separate up to seven As species for both non-ionic and ionic compounds in a signal run using the same column and short time. However, a combination of anion- and cation-exchange chromatography seems the most promising for complete resolution up to eight As species. The ETAAS method using different separation techniques and chemical modifiers can determine As species in seawater with DL of 0.02 μgL(-1), eliminate interferences of transition metals, improve the selectivity of the measurement, and enhance the sensitivity.

Abstract:Nowadays, reliable and robust detectors can be considered standard laboratory instrumentation, which, for most of the elements provide quantitation limits in the lower ng/g range. Despite these advances in detector technology, sample preparation is by far the most important error source in modern analytical method development and can be judged as the "Achilles' heel" of any analytical process regarding reliability of the obtained results and time consumption. The aim of the present review is to highlight modern trends for tin and arsenic speciation, as these analytes can be considered as models for challenges in modern method development in this field. First background information, legislative aspects and current needs are elucidated. Then the role of sample treatment within the process of method development in speciation is discussed, followed by a presentation of modern extraction techniques, matching the requirements for arsenic and tin speciation analysis: to provide mild conditions in order to ensure species preservation, to improve species recovery, to enhance sample throughput and to be suitable for hyphenation with chromatographic separation systems. The review includes applications on tin and arsenic speciation, covering the period of 2001-2006.

Abstract:Arsenic (As) speciation is an interesting topic because it is well recognized that the toxicity of this metalloid ultimately depends on its chemical form. More than 300 arsenicals exist naturally. However, As can be present in four oxidation states: As −III , As 0 , As III and As V . Long-term exposure to As from different sources, such as anthropogenic processes, or water, fauna and flora contaminated with As, has put human health at risk for decades. There are many side-effects correlated with exposure to InAs species, such as skin problems, respiratory diseases, kidney problems, cardiovascular diseases and even cancer. There are different levels and types of As in foods, particularly in vegetables. Furthermore, different chemical methods and techniques have been developed. Therefore, this review focuses on the general properties of various approaches used to identify As species in vegetation samples published worldwide. This includes various approaches (different solvents and techniques) used to extract As species from the matrix. Then, versatile chromatographic and non-chromatographic systems to separate different forms of As are reviewed. Finally, the general properties of the most common instruments used to detect As species from samples of interest are listed.

Abstract:The presence of arsenic (As) in aquatic environments is often attributable to geogenic processes occurring within aquifers. Arsenic exists in two prevalent oxidation states, with the trivalent arsenite [As(III)] exerting stronger toxicity effects on the aquatic biota than the pentavalent arsenate [As(V)]. The review evaluates the literature available on the main arsenic removal technologies and the application of combined chemical and biological treatments. We provide a synthetic outlook on the potential strategies of biological As(III) oxidation to As(V) by means of cell-detoxifying mechanisms or metabolic processes with the aim to enhance As removal efficiency. Furthermore, the role of microorganisms in the mobility of arsenic in natural systems as well as the distribution of As-resistant bacteria, potentially suitable for arsenic removal, is discussed in the context of a case study carried out in Latium region (Italy), which is known for arsenic contamination of waters.

Abstract:Arsenic is a ubiquitous toxic metalloid causing serious health problems. Speciation analysis of arsenic in human urine provides valuable insights for large-scale epidemiological studies and informs on sources of exposure as well as human metabolism. The Multi-Ethnic Study of Atherosclerosis (MESA) is a valuable cohort for assessing chronic low-moderate arsenic exposure and health effects in an ethnically diverse US population. We present a state-of-the-art arsenic speciation analysis methodology and its application to 7,677 MESA spot urine samples based on high-performance liquid chromatography coupled to inductively coupled plasma mass spectrometry. This method is fast, robust and detects a total of 11 individual As species at method detection limits of 0.02-0.03 μg arsenic/L urine for each individual species. Our analytical approach features excellent method accuracy (98%) and precision (5%) for the main arsenic species in urine (arsenobetaine, methylarsonic acid, dimethylarsinic acid, and total inorganic As); intra- (3-6%) and inter-day coefficients of variability (5-6%); column recovery (96 ± 7%); and spike recovery (97 ± 6%). The main arsenic species were detectable in ≥95% of urine samples due to the implementation of an oxidation step. Each individual minor arsenic species was detectable in ≤25% of all urines, although at least one of them was detectable in almost half the participants. We identified two minor urinary arsenic species as dimethylarsinoylacetic acid and dimethylarsinoylpropionic acid, potential metabolites of seafood-related arsenicals. We observed differences in individual As species excretion by race/ethnicity, with Asian-American participants featuring 3-4 times higher concentrations compared to other participants. We also found differences by site, body mass index, smoking status, rice intake, and water arsenic levels, potentially indicating different exposures or related to individual bio-metabolism. The proposed approach is suitable for epidemiological studies and the collected data will constitute the base for future research on potential health effects of chronic low-level arsenic exposure.

Abstract:Abstract Traditional Chinese medicine contains arsenic (As), which in the natural environment accumulates in plants during the growth of Chinese medicinal materials; there are mineral medicines containing As in Chinese patent medicine such as As 4 S 4 , As 2 S 3 , etc. Due to the toxicity of As-containing compounds and its role in inflammation and treatment of cancers such as leukemia, it is necessary to analyze the chemical form of As. A comprehensive investigation of the compound forms of heavy metals rather than the simple total amount of elements will lay the foundation for the scientific and objective evaluation of the safety of heavy metals. This article summarizes the speciation of As in bulk Chinese medicinal materials and Chinese patent medicines in recent years, and reviews the main research methods of As speciation analysis. The separation and detection combined analysis method focuses on the high-performance liquid chromatography-plasma mass spectrometry and high-performance liquid chromatography-hydride generation-atomic fluorescence spectroscopy, etc. Taking the advanced synchrotron radiation source as the research platform, the use of X-ray near edge absorption fine structure spectrum and micro-area X-ray fluorescence analysis as a microscopic analysis technique supports direct analysis of the As speciation in situ . It is the most promising morphological analysis method.

Abstract:There are more than 100 different arsenic with different characteristics in the soil-water-plant ecosystem. The identification and quantification of individual arsenic species is essential for understanding the distribution, environmental fate and behavior, metabolism and toxicity of arsenic. Due to the hazardous nature of arsenic, people have a high interest in the measurement of arsenic species. The reaction of the formation of arsenic speciation in the soil-water-plant environment is briefly studied. There is little information on methods used to quantify arsenic forms and species in contaminated soil, water and plant. The purpose of this article is to understand the available sample pretreatment, extraction, separation, detection and method validation techniques for arsenic speciation analysis of arsenic species in soil, water and plant. The performances of various sample preparation and extraction processes, as well as effective separation techniques, that contribute greatly to excellent sensitivity and selectivity in arsenic speciation when coupling with suitable detection mode, and method validity are discussed. The outlines of arsenic speciation techniques are discussed in view of the importance to the completeness and accuracy of analytical data in the soil-water-plant samples. To develop cheap, fast, sensitive, and reproducible techniques with low detection limits, still needed to confine research on arsenic speciation present in environmental matrices.

Abstract:It is recognized that aluminium (Al) is a potential environmental hazard. Acidic deposition has been linked to increased Al concentrations in natural waters. Elevated levels of Al might have serious consequences for biological communities. Of particular interest is the speciation of Al in aquatic environments, because Al toxicity depends on its forms and concentrations. In this paper, advances in analytical methodologies for Al speciation in environmental and biological samples during the past five years are reviewed. Concerns about the specific problems of Al speciation and highlights of some important methods are elucidated in sections devoted to hybrid techniques (HPLC or FPLC coupled with ET–AAS, ICP– AES, or ICP–MS), flow-injection analysis (FIA), nuclear magnetic resonance (27Al NMR), electrochemical analysis, and computer simulation. Abstract:In order to obtain a well understanding of the toxicity and ecological effects of trace elements in the environment, it is necessary to determine not only the total amount, but also their existing species. Speciation analysis has become increasingly important in making risk assessments of toxic elements since the toxicity and bioavailability strongly depend on their chemical forms. Effective separation of different species in combination with highly sensitive detectors to quantify these particular species is indispensable to meet this requirement. In this paper, we present the recent progresses on the speciation analysis of trace arsenic, mercury, selenium and antimony in environmental and biological samples with an emphasis on the separation and detection techniques, especially the recent applications of high performance liquid chromatography (HPLC) hyphenated to atomic spectrometry or mass spectrometry.

Quantification of Arsenic in Soil Samples Collected in an Industrial Area of Brindisi (Apulia, Italy): Speciation Analysis and Availability

Abstract:Arsenic (As) is a well-known toxic metalloid, but environmental risks due to excessive As content in soils or sediments depend on the chemical forms present and their relative mobility. Long-term exposure to arsenic may cause several diseases. In order to assess the possible risks in the heavily impacted Consorzio per lo Sviluppo Industriale e di Servizi Reali alle Imprese (Consortium for Industrial Development and Effective Services for Business, S.I.S.R.I.) industrial area of Brindisi (Apulia, southern Italy), 38 soil samples were collected in the area, from 18 sampling points previously determined as outliers. Total As determination, speciation analysis, and a cession test with acetic acid were performed. Speciation analysis was performed by HPLC coupled to hydride generation-atomic absorption spectroscopy (HG-AAS). Total As determination obtained by mineralization showed a concentration range between 51.8 and 169.6 mg kg−1, which is higher than the limit of 50 mg kg−1 established by D.M. (Ministerial Decree) 471/99 for industrial areas. The highest concentrations of extracted As were obtained in the top-soil layers. As(III) and As(V) were detected in all the samples, while the concentrations of the organic species monomethyl arsonic acid (MMAA) and dimethyl arsenic acid (DMAA) were always under the detection limit. The samples releasing the highest As quantities in the acetic acid cession test were in every circumstance collected from the superficial soil levels. The different amounts of As determined in the sampling sites could depend on the distance from the specific sources of pollution, even if it is very difficult to identify them in a very complex industrial zone such as the S.I.S.R.I. area of Brindisi. In this study, As occurs mainly as relatively immobile or slowly exchangeable forms: for this reason, it is more abundant in the top-soil and is little affected by the action of rainwater, which transports only reduced quantities of As into the deeper layers.
