Abstract:A new resorufin-based spectroscopic probe, 7-(p-acetoxyphenylmethyloxy)-3H-phenoxazin-3-one (1), has been developed for detecting carboxylesterase activity. The probe is designed by introducing p-acetoxyphenylmethyloxy as a bifunctional moiety to resorufin. The p-acetoxyphenylmethyloxy moiety not only quenches the spectroscopic signal of resorufin, but also serves as a recognition unit for carboxylesterase. As a result, the prepared latent spectroscopic probe 1 shows very low background signal, which is rather desired for achieving sensitive detection. The specific cleavage of the carboxylic ester bond by carboxylesterase induces the hydrolysis of the probe, resulting in the release of resorufin and thereby the recovery of both color and fluorescence signal. This behaviour leads to the development of a simple and sensitive fluorescent method for assaying carboxylesterase activity, with a detection limit of 8.6 × 10(-5) U mL(-1), which is much more sensitive than the existing fluorescence approaches. Moreover, experimental results showed that the probe 1 is cell membrane permeable, and its applicability has been demonstrated for monitoring carboxylesterase activity in HeLa cells.

Abstract:In this study, a new red lysosome-targeted fluorescence off-on probe has been rationally designed, synthesized and applied to highly selective and sensitive fluorescence sensing of carboxylesterase in live cells, sera and tissues. The probe was readily synthesized by coupling N-methylated benzoindole with 4-(acetyloxy)benzaldehyd, in which the acetyl group acted as a quenching and recognizing role and hemicyanine skeleton worked as the fluorescent signal unit. As a consequence, the prepared spectroscopic probe displays quite low background signal. Upon addition of carboxylesterase, the latent probe could be easily hydrolyzed to liberate the fluorophore hydroxy hemicyanine and show the off-on spectroscopic signal, which is desirable for obtaining sensitive detection with high specificity. The newly developed probe has been successfully used to monitor and image carboxylesterase activity in complex biological samples such as serum, live cells and tissues. Moreover, this probe shows great promise for biomedical studies, including obesity disease, hepatocellular carcinoma and drug screening researches.

Abstract:In vivo selective fluorescence imaging of carboxylesterase 2 (CES2) remains a great challenge because existing fluorescence probes can potentially suffer from interference by other hydrolases. In addition, some fluorescent probes that have been separately reported for measuring CES2 activity in vitro are affected by autofluorescence and absorption of the biological matrix due to their limited emission wavelength or short Stokes shift. Herein, based on the substrate preference and catalytic performance of CES2, a novel and NIR fluorescent probe was developed, in which a hemi-cyanine dye ester derivative was used as the basic fluorescent group. In the presence of CES2, the probe was hydrolyzed to expose the fluorophore CZX-OH (λabs ∼ 675 nm, λem ∼ 850 nm), which led to a notable red-shift in the fluorescence (∼175 nm) spectrum. Confocal imaging of cells and live mice demonstrated that the fluorescent signal of this probe was related to the real activities of CES2 in cancer cells. All these results will powerfully promote the screening of CES2 regulators and the analysis of CES2-related physiological and pathological processes.
Preparation of a Highly Selective "Off-On" Rhodamine-Based Fluorescent Probe for the Specific Determination of Carboxylesterase 2 and Cell Imaging

Abstract:Abstract Given the important role of carboxylesterase 2 (CES2) in the metabolism of various esters, it is of significance to develop a tool to determine endogenous CES2 activity in a rapid and highly selective manner. In this work, an "off-on" rhodamine-based fluorescent probe is reported as an effective tool for CES2 activity. Based on the substrate recognition preference of CES2 (large alcohol and small acyl groups), the probe with dimethyl carbamate as the recognition group performs well in terms of selectivity and sensitivity to CES2. The fluorescence switching control of the probe for CES2 was achieved by using the spirolactone structure of rhodamine. The probe shows the fast generation of a fluorescence signal at 634 nm upon hydrolysis of CES2 with excitation at 578 nm. The results showed a linear relationship between the fluorescence intensity at 634 nm and the CES2 activity from 0 to 4 μg/mL. The probe reacts rapidly with CES2, and the reaction is stable within 40 minutes. The probe is highly selective for CES2, with a detection limit as low as 0.303 μg/mL. In addition, the probe has currently been successfully utilized to evaluate CES2 activity in living cells. Hence, this probe is anticipated to be significant in identifying endogenous CES2 activity in intricate biological settings.

Abstract:Carboxylesterases in human body play pivotal roles for catalyzing the hydrolysis of endogenous and exogenous substances such as lipids, thioesters and amides. Carboxylesterase-2 (CES2) as a major isoform of carboxylesterases is highly expressed in human digestive tract, and usually serves as an important mediator for pharmacokinetics and pharmacodynamics of ester medications or ester prodrugs. Therefore, developing effective strategies to detect and track CES2 in cells or tissues is of great significance for understanding its physiological and pharmacological effects on various diseases and therapeutic outcomes. In this study, we synthesized a fluorescent probe (CHQ-E) with near-infrared (NIR) excitation and emission maxima based on dihydroxanthene fluorophore for highly selective and sensitive detection of CES2. This probe exhibited rapid response (3 min), excellent sensitivity (detection limit = 0.35 ng/mL) and high binding affinity ( K m = 9.4 μM) towards CES2, as well as low cytotoxicity towards living biosamples. Significantly, the maximum fluorescence excitation and emission wavelengths led to excellent optical properties of this probe including less photodamage, few autofluorescence and superior tissue penetration depth (180 μm). Moreover, CHQ-E has been applied to monitoring CES2 activity in drug-induced liver injury and its remediation models. Results of fluorescence imaging in vivo indicate the broad potential of CHQ-E for clinical diagnosis and evaluation of pharmaceuticals.

Abstract:Herein, we report a novel fluorescent light-up probe useful for carboxylesterase assay that is based on a tetraphenylethylene derivative containing carboxylic ester groups. The specific cleavage of the carboxylic ester bonds by carboxylesterase results in the generation of a relatively hydrophobic moiety that self-assembles into supramolecular microfibers, thus giving rise to turn-on fluorescent signals. A high sensitivity towards carboxylesterase was achieved with a detection limit as low as 29pM, which is much lower than the corresponding assays based on other fluorescent approaches.

Abstract:Carboxylesterases (CEs) are involved in biotransformation of numerous drugs, environmental toxicants, and carcinogens containing ester groups. Abnormal CEs levels are associated with various diseases. Herein, we developed a novel water-soluble merocyanine-based probe 1 to detect CEs activity using acetate ester as a recognition site. The notable emission at 510 nm of the probe in neat aqueous solution was partially quenched and red-shifted to 580 nm after adding CEs. The fluorescent ratiometric response of 1 toward CEs activity is sensitive (with the detection limit of 0.98 x 10 (6) U/ml), rapid (within 10 min), and effective in a wide pH range (4.0-10.0). Due to the existence of morpholine as a lysosome-targeting group, the probe was successfully utilized to monitor lysosomal CEs activity. More importantly, the probe was successfully applied to detect and image carboxylesterase activity in zebrafish.

Abstract:Carboxylesterases (CEs) play great role in CEs-related diseases and drug metabolism. Selectively monitoring its activity is important to explore its role in CEs-related diseases and drug combination. Herein, a new "turn-on" near-infrared (NIR) fluorescent probe (CHY-1 ) was reported with large Stokes shift (145 nm) for CEs detection. Dicyanoisophorone-based derivative was chosen as NIR fluorophore and 4-bromobutyrate was the identifying group. What's more, CHY-1 exhibited ultra-sensitivity (LOD ∼ 9.2 × 10 −5 U/mL), high selectivity against Acetylcholinesterase (AChE), Butyrylcholinesterase (BChE) and Chymotrypsin for CEs fluorescence detection under physiological pH and temperature. Furthermore, CHY-1 showed little effect on cell viability at high concentration and featured good optical imaging character for the slight change of CEs activity induced by 5-Fu (5-Fluorouridine, anti-tumor drug) and CEs inhibitor in living cells. Moreover, CHY-1 was also used to detect the activity and distribution of CEs in mice. Taken together, CHY-1 has widely applicable value in the diagnosis of CEs-related diseases and drug combination.

Abstract:<p>An aggregation-induced emission (AIE) fluorescent protein chromophore-based probe (CBZ-FP) for detection of human carboxylesterases (CESs) was designed and synthesized. CBZ-FP exhibited good cell permeability with a large stokes shift (116 nm) and can be applied to reveal the actual activities of CES1 in living cells associated with pesticides detoxification process. CBZ-FP can also serve as a fluorescence indicator of pesticide exposure in the way of hydrolyzing the carboxylic acid ester group in CBZ-FP. Therefore, CBZ-FP has high selectivity for CESs and can detect real-time activity of CES1 in biological samples. Molecular docking study was used to explore the binding of CESs and CBZ-FP. Finding that only one specific activity site of CESs can bind with probe. In view of the fact that, the biotransformation of drugs and poisons containing ester groups can carry out normally depending on CESs, Carboxylesterase probes are expected to contribute to the characterization of relevant disease.</p>

Abstract:Pesticide residues pose a significant threat to food safety and human health, necessitating the development of novel detection tools. Pesticides can inhibit the activity of certain biological enzymes, so enzyme inhibition is one of the methods of pesticide detection. In this study, we developed a novel near-infrared fluorescent probe named TCFCl-CES based on the tricyanofuran structure, for ultrasensitive detection of carboxylesterase (CES). TCFCl-CES exhibits strong and stable fluorescence, excellent specificity. Notably, the fluorescence intensity of TCFCl-CES shows a linear relationship with CES concentration, achieving an exceptionally low detection limit of 4.41 × 10-5 u/mL. This ultrasensitive probe can also effectively detect pesticide residues in vegetables and monitor CES activity in cells and liver tissues. TCFCl-CES stands out for its rapid and accurate detection capabilities, making it an essential tool for accurately monitoring pesticide residue. It also has great potential for tracking CES activity in biological systems. Additionally, it offers a robust solution for food safety and improving pesticide residue analysis.

Abstract:A benzothiazole fluorescent probe (probe 1) was developed for the determination of carboxylesterase and carbamate pesticides. Probe 1 can be used as a tool for the rapid detection of carbamate pesticides using smartphones. A benzothiazole fluorescent probe (probe 1) was developed for the determination of carboxylesterase and carbamate pesticides. The probe used benzothiazole as the fluorescent group and chlorophenyl acetate as the recognition group. The recognition mechanism of probe 1 for carboxylesterase was based on the enzymatic hydrolysis of chlorophenyl acetate by carboxylesterase. The solution of probe 1 gradually changed from light blue to dark blue as the concentration of carbamate pesticide increased. Probe 1 could detect the concentration of carbamate pesticide based on the linear functional relationship between the R/B*G (red, blue, green) value of the probe 1 solution photo and the concentration of carbaryl without professional personnel and equipment. Additionally, probe 1 was successfully used to detect the concentration of carbaryl in actual samples based on the curve function relationship between the fluorescence intensity of probe 1 at 434 nm and the concentration of carbaryl.

Abstract:Pesticide residues significantly affect food safety and harm human health. In this work, a series of near-infrared fluorescent probes were designed and developed by acylating the hydroxyl group of the hemicyanine skeleton with a quenching moiety for monitoring the presence of organophosphorus pesticides in food and live cells. The carboxylic ester bond on the probe was hydrolyzed catalytically in the presence of carboxylesterase and thereby the fluorophore was released with near-infrared emission. Notably, the proposed probe 1 exhibited excellent sensitivity against organophosphorus based on the carboxylesterase inhibition mechanism and the detection limit for isocarbophos achieved 0.1734 μg/L in the fresh vegetable sample. More importantly, probe 1 allowed for situ visualization of organophosphorus in live cells and bacteria, meaning great potential for tracking the organophosphorus in biological systems. Consequently, this study presents a promising strategy for tracking pesticide residues in food and biological systems.

Abstract:A novel near-infrared (NIR) fluorescent probe CHC-CES1 based on a hemi-cyanine skeleton for detecting carboxylesterase 1 (CES1) activity was developed. Herein, CHC-CES1 could be specifically hydrolysed to CHC-COOH along with a significant NIR fluorescence signal enhancement at 670 nm. Systematic evaluation indicated that CHC-CES1 possessed an outstanding selectivity and sensitivity towards CES1, and possessed good chemical stability in complex biosamples. Finally, CHC-CES1 was successfully used for the real-time imaging of endogenous CES1 activity in living cells. Moreover, CHC-CES1 was applied to evaluate the inhibitory effects of various pesticides towards CES1, and visually revealed the inhibitory effect of combined residue pesticides.

Abstract:The discrimination of living and dead cells shows great importance in the development of biology, pathology, medicine, and pharmacology research. Herein, we synthesized a simple benzothiazole-based probe, EP, which was characterized via(1)H NMR (hydrogen nuclear magnetic resonance) spectroscopy, C-13 NMR (carbon nuclear magnetic resonance) spectroscopy and HRMS (high-resolution mass spectroscopy). The fluorescence changes in response to esterase were characterized via fluorescence spectroscopy. EP exhibited a 70-fold fluorescence enhancement in the presence of esterase and possessed a very low limit of detection (4.73 x 10(-5) U mL(-1)). EP also showed high selectivity to esterase compared to other biological species. Bright fluorescence appeared in living cells, which was activated by esterase when incubated with EP. In paraformaldehyde or H2O2 pretreated cells, the fluorescence became very weak since esterase became inactive in these cells. In summary, the EP probe can monitor esterase activity both in vitro and in living cells and can be used to evaluate the health status of cells and discriminate living and dead cells effectively.

Abstract:Carboxylesterases (CESs) are critical for metabolizing ester-containing biomolecules and are specifically important in liver metabolic disorders. The modulation of CESs is also an important issue in pharmacology and clinical applications. Herein, we present a near-infrared (NIR) CES fluorescent probe ( NCES ) based on the protection–deprotection of the hydroxyl group for monitoring CES levels in living systems. The NCES probe has good selectivity and sensitivity for CESs with a limit of detection (LOD) of 5.24 mU mL −1 , which allows for tracing the fluctuation of cellular CES after treatment with anticancer drugs and under inflammation and apoptosis states. Furthermore, NCES can be successfully applied for guiding liver cancer surgery with high-contrast in vivo imaging and detecting clinical serum samples from liver cancer patients. This work showed that the NCES probe has great potential in drug development, imaging applications for medical diagnosis, and early-stage detection for clinical liver diseases.

Abstract:A colorimetric fluorescent probe (BTCNA) was developed for the determination of carboxylesterase and carbamate pesticides. The probe used naphthalene–benzothiazole as the fluorescent group and naphthyl acetate as the recognition group. The recognition mechanism of BTCNA for carboxylesterase was based on the enzymatic hydrolysis of naphthyl acetate by carboxylesterase (CES). The test paper of the BTCNA gradually changed from light blue to bright yellow with the increase of CES activity. The probe solution gradually changed from light blue to earth-yellow as the carbaryl concentration increased. There was a linear functional relationship between the R*G (red, green) value of the photo and the CES activity. And a linear functional relationship between the carbaryl concentration and the R*G value of the photo was found. Additionally, BTCNA was successfully used to detect the concentration of carbaryl in actual samples. BTCNA is a rapid detection tool for CES activity and carbamate pesticides using a smartphone.

Abstract:Esterases are common (biological catalyst) that play an essential role in regulating the physiological processes of organisms, in addition to contributing to disease prevention. Herein, we report the design and synthesis of an esterase-activated chemiluminescent probe (CL-EA) for the selective detection of esterases. Using the 1,2-dioxetane scaffold as the chemiluminescent platform and acetoxymethyl ether as the recognition group, the present probe was successfully lighted in the presence of esterases. As a result, a strong chemiluminescent signal was generated in an aqueous solution, thereby allowing the highly sensitive and specific detection of esterases. Furthermore, the probe is used to screen the activity of three plastic-degrading enzymes and image esterases in live cells and in a mouse model.
