Abstract:For several decades, although studies of rat physiology and behavior have abounded, research on rat emotions has been limited in scope to fear, anxiety, and pain. Converging evidence for the capacity of many species to share others' affective states has emerged, sparking interest in the empathic capacities of rats. Recent research has demonstrated that rats are a highly cooperative species and are motivated by others' distress to prosocial actions, such as opening a door or pulling a chain to release trapped conspecifics. Studies of rat affect, cognition, and neural function provide compelling evidence that rats have some capacity to represent others' needs, to instrumentally act to improve their well-being, and are thus capable of forms of targeted helping. Rats' complex abilities raise the importance of integrating new measures of rat well-being into scientific research.

Observational fear behavior in rodents as a model for empathy

Abstract:Empathy enables social mammals to recognize and share emotion with others and is well-documented in non-human primates. During the past few years, systematic observations have showed that a primal form of empathy also exists in rodents, indicating that empathy has an evolutionary continuity. Now, using rodents exhibiting emotional empathy, the molecular and cellular study of empathy in animals has begun in earnest. In this article, we will review recent reports that indicate that rodents can share states of fear with others, and will try to highlight new understandings of the neural circuitry, biochemistry and genetics of empathic fear. We hope that the use of rodent models will enhance understanding of the mechanisms of human empathy and provide insights into how to treat social deficits in neuropsychiatric disorders characterized by empathy impairment.

Abstract:Prosocial behaviors (i.e., actions that benefit others) are central for social interactions in humans and other animals, by fostering social bonding and cohesion. To study prosociality in rodents, scientists have developed behavioral paradigms where animals can display actions that benefit conspecifics in distress or need. These paradigms have provided insights into the role of social interactions and transfer of emotional states in the expression of prosociality, and increased knowledge of its neural bases. However, prosociality levels are variable: not all tested animals are prosocial. Such variation has been linked to differences in animals' ability to process another's state as well as to contextual factors. Moreover, evidence suggests that prosocial behaviors involve the orchestrated activity of multiple brain regions and neuromodulators. This review aims to synthesize findings across paradigms both at the level of behavior and neural mechanisms. Growing evidence confirms that these processes can be studied in rodents, and intense research in the past years is rapidly advancing our knowledge. We discuss a strong bias in the field towards the study of these processes in negative valence contexts (e.g., pain, fear, stress), which should be taken as an opportunity to open new venues for future research.
Neural correlates of ingroup bias for prosociality in rats

Abstract:Prosocial behavior, in particular helping others in need, occurs preferentially in response to distress of one's own group members. In order to explore the neural mechanisms promoting mammalian helping behavior, a discovery-based approach was used here to identify brain-wide activity correlated with helping behavior in rats. Demonstrating social selectivity, rats helped others of their strain ('ingroup'), but not rats of an unfamiliar strain ('outgroup'), by releasing them from a restrainer. Analysis of brain-wide neural activity via quantification of the early-immediate gene c-Fos identified a shared network, including frontal and insular cortices, that was active in the helping test irrespective of group membership. In contrast, the striatum was selectively active for ingroup members, and activity in the nucleus accumbens, a central network hub, correlated with helping. In vivo calcium imaging showed accumbens activity when rats approached a trapped ingroup member, and retrograde tracing identified a subpopulation of accumbens-projecting cells that was correlated with helping. These findings demonstrate that motivation and reward networks are associated with helping an ingroup member and provide the first description of neural correlates of ingroup bias in rodents.

Abstract:Empathy is an essential function for humans as social animals. Emotional contagion, the basic form of afffective empathy, comprises the cognitive process of perceiving and sharing the affective state of others. The observational fear assay, an animal model of emotional contagion, has enabled researchers to undertake molecular, cellular, and circuit mechanism of this behavior. Such studies have revealed that observational fear is mediated through neural circuits involved in processing the affective dimension of direct pain experiences. A mouse can also respond to milder social stimuli induced by either positive or negative emotional changes in another mouse, which seems not dependent on the affective pain circuits. Further studies should explore how different neural circuits contribute to integrating different dimensions of affective empathy.

Abstract:Introduction: Empathic behaviors are driven by the ability to understand the emotional states of others along with the motivation to improve it. Evidence points towards forms of empathy, like targeted helping, in many species including rats. There are several variables that may modulate targeted helping, including sex, sensory modalities, and activity of multiple neural substrates. Methods: Using a model of social contact-independent targeted helping, we first tested whether sex differences exist in helping behavior. Next, we explored sex differences in sensory and affective signaling, including direct visualization and an analysis of ultrasonic vocalizations made between animal pairs. Finally, we examined the neural activity in males and females of multiple regions of interest across time. Here, we aim to examine any behavioral differences in our lab's social contact independent targeted helping task between males and females. Results and Discussion: These findings are the first to intimate that, like other prosocial behaviors, males and females may exhibit similar social-independent targeted helping behavior, but the underlying sensory communication in males and females may differ. In addition, this is the first set of experiments that explore the neural correlates of social-independent targeted helping in both males and females. These results lay the groundwork for future studies to explore the similarities and differences that drive targeted helping in both sexes.

Abstract:In recent years, a growing number of pre-clinical studies have made use of the social abilities of mice, asking how gene variants (e.g., null, transgenic or mutant alleles) give rise to abnormalities in neurodevelopment. Two distinct courses of research provide the foundation for these studies. One course has mostly focused on how we can assess “sociability” using metrics, often automated, to quantitate mouse approach and withdrawal responses to a variety of social stimuli. The other course has focused on psychobiological constructs that underlie the socio-emotional capacities of mice, including motivation, reward and empathy. Critically, we know little about how measures of mouse sociability align with their underlying socio-emotional capacities. In the present work, we compared the expression of sociability in adolescent mice from several strains versus a precisely defined behavioral model of empathy that makes use of a vicarious fear learning paradigm. Despite substantial strain-dependent variation within each behavioral domain, we found little evidence of a relationship between these social phenotypes (i.e., the rank order of strain differences was unique for each test). By contrast, emission of ultrasonic vocalizations was highly associated with sociability, suggesting that these two measures reflect the same underlying construct. Taken together, our results indicate that sociability and vicarious fear learning are not manifestations of a single, overarching social trait. These findings thus underscore the necessity for a robust and diverse set of measures when using laboratory mice to model the social dimensions of neuropsychiatric disorders.

Abstract:Key PointsObservational and experimental studies dating back to the 1950s demonstrate that mammals spontaneously help distressed conspecifics. Research emphasizes the untrained, unrewarded nature of this behaviour, which is also biased towards familiar individuals, thus arguing against explanations that are exclusively based on associative learning or conditioning.The perception–action model extends an existing motor theory on overlapping representations to emotional phenomena; it states that observers who attend to a target's state understand and 'feel into' it through personal distributed representations of the target, the state and the situation. Easily observed manifestations of this mechanism are emotional contagion and motor mimicry, which have been demonstrated in many animals. In cognitive forms of empathy, the same representations are accessed from the top-down.Experiments on two common mammalian expressions of empathy — the consolation of distressed individuals and spontaneous assistance to those in need — support the crucial role of caught distress and arousal because these behaviours are suppressed by anti-anxiety medication and engage the same neuropeptide system that supports social attachment.The Russian-doll model seeks to arrange forms of empathy into layers that are built on top of each other — with the layers ranging from emotional contagion to more cognitive forms of empathy — in a functionally integrated whole based on perception–action processes. Perspective-taking is well developed in some non-human species, as manifested by theory-of-mind and targeted helping.One can segregate emotional and cognitive empathy (as well as felt and observed states) in the brains of observers, but all forms require some initial access to the observer's distributed, shared, personal representations of the target's state. At least in the initial phase of processing, this access helps to decode the target's state and provide subsequent processing with content and meaning, even if the shared state is not experienced, or is incomplete or inaccurate.Empathic pain does not usually include the peripheral sensation of the target's injury, but it can include sensory information when the stimuli and task instructions emphasize the specific nature of the feeling at the location of the injury.

Abstract:While empathic emotion is closely related to prosocial behavior in human and animals, neuronal substrate that accounts for empathy-driven prosocial action remains unclear. We recorded ACC and InC neurons in rats when they observed another rat in pain through the vicarious pain test. We discovered neurons with anti-mirror properties in ACC and InC besides those with mirror properties. ACC neurons show higher coupling between activation of self in pain and of others in pain, while InC has higher ratio of neurons that showed anti-mirror properties. During others in pain, ACC neurons activated more when noise poking toward others and InC neurons activated more when freezing. Motivated by the distinct behavioral correlations and the discovery of affective anti-mirror neurons, we examined neuronal activities in the rescuing task where a rat was trapped in a restrainer to further illustrate prosocial function. Both ACC and InC showed specific activation to rat rescuing, which is specifically contributed by the mirror but not anti-mirror neurons. Our work introduces a neuronal framework that explains empathy-driven prosocial action along with accurate self–other distinction.

Abstract:The phenomenon of empathy entails the ability to share the affective experiences of others. In recent years social neuroscience made considerable progress in revealing the mechanisms that enable a person to feel what another is feeling. The present review provides an in-depth and critical discussion of these findings. Consistent evidence shows that sharing the emotions of others is associated with activation in neural structures that are also active during the first-hand experience of that emotion. Part of the neural activation shared between self- and other-related experiences seems to be rather automatically activated. However, recent studies also show that empathy is a highly flexible phenomenon, and that vicarious responses are malleable with respect to a number of factors--such as contextual appraisal, the interpersonal relationship between empathizer and other, or the perspective adopted during observation of the other. Future investigations are needed to provide more detailed insights into these factors and their neural underpinnings. Questions such as whether individual differences in empathy can be explained by stable personality traits, whether we can train ourselves to be more empathic, and how empathy relates to prosocial behavior are of utmost relevance for both science and society.

Abstract:In mammals, empathy is crucial for living in social groups and caring for others. In this paper, we consider the structural and functional organization of empathy. We propose that empathy subsumes a variety of neurobiological processes and partially dissociable information processing subsystems, each of which has a unique evolutionary history. Even the most advanced and flexible forms of empathy in humans are built on more basic forms and remain connected to core subcortical and neurohormonal mechanisms associated with affective communication, parental care and social attachment processes. Considering empathy within a framework that recognizes both the continuities and the changes within a phylogenetic perspective provides a richer understanding of empathy and related neurobehavioral processes.

Abstract:Social behaviour plays a key role in mental health and wellbeing, and developing greater understanding of mechanisms underlying social interaction—particularly social motivation—holds substantial transdiagnostic impact. Common rodent behavioural assays used to assess social behaviour are limited in their assessment of social motivation, whereas the social operant conditioning model can provide unique and valuable insights into social motivation. Further characterisation of common experimental parameters that may influence social motivation within the social operant model, as well as complementary methodological and analytical approaches, are warranted.

Abstract:A prosocial response to others in distress is increasingly recognized as a natural behavior for many social species, from humans to rodents. While prosocial behavior is more frequently observed towards familiar conspecifics, even within the same social context some individuals are more prone to help than others. For instance, in a rat helping behavior test, rats can release a distressed conspecific trapped inside a restrainer by opening the restrainer door. Typically, rats are motivated to release a trapped cagemate, and consistently release the trapped rat (‘openers’), yet around 30% do not open the restrainer (‘non-openers’). To characterize the difference between these populations, behavioral and neural activity were compared between opener and non-opener rats tested with a trapped cagemate in the helping test. Behaviorally, openers showed significantly more social affiliative behavior both before and after door-opening compared to non-openers. Analysis of brain-wide neural activity based on the immediate early gene c-Fos revealed increased activity in openers in the previously identified prosocial neural network compared to non-openers. The network includes regions associated with empathy in humans (somatosensory cortex, insula, cingulate cortex and frontal cortex), and motivation and reward regions such as the nucleus accumbens. Oxytocin receptor mRNA expression levels were higher in the accumbens but not the anterior insula. Several transcription control pathways were also significantly upregulated in openers’ accumbens. These findings indicate that prosocial behavior may be predicted by affiliative behavior and activity in the prosocial neural network and provide targets for the investigation of causal mechanisms underlying prosocial behavior.

Abstract:Cooperation is a social behavior crucial for the survival of many species, including humans. Several experimental paradigms have been established to study cooperative behavior and related neural activity in different animal species. Although mice exhibit limited cooperative capacity in some behavioral paradigms, it is still interesting to explore their cooperative behavior and the underlying neural mechanisms. Here, we developed a new paradigm for training and testing cooperative behavior in mice based on coordinated lever-pressing and analyzed social interactions between the animals during cooperation. We observed extensive social contact and waiting behavior in cooperating animals, with the number of such events positively correlated with the success of cooperation. Using c-Fos immunostaining and a high-speed volumetric imaging with synchronized on-the-fly scan and readout (VISoR) system, we further mapped whole-brain neuronal activity trace following cooperation. Significantly higher levels of c-Fos expression were observed in cortical areas including the frontal pole, motor cortex, anterior cingulate area, and prelimbic area. These observations highlight social interaction and coordination in cooperative behavior and provide clues for further study of the underlying neural circuitry mechanisms.
Involvement of Ventral Tegmental Area to Anterior Cingulate Cortex Dopamine Projections in Empathetic Behavior in Male C57 Mice

Abstract:Normal empathetic ability is of great importance to survival and reproduction of group living animals. Although some neural mechanisms underlying empathy has been studied, whether ventral tegmental area (VTA) to the anterior cingulate cortex (ACC) dopamine (DA) projections regulates empathy remains unclear. In this study, we found that dopamine release increased in the ACC upon empathetic behavior occurred. In addition, VTA DA neuron projected to the ACC revealed by injection of cholera toxin subunit B (CTB) as retrograde tracing. Also, activation of VTA-ACC DA neuron projections enhanced empathy while inhibition of VTA-ACC DA neuron projections reduced empathy. Microinjection of dopamine type two receptor (D2R) agonist into the ACC increased the empathetic behavior which was decreased by inhibition of VTA-ACC DA pathway. Meanwhile, microinjection of D2R antagonist into the ACC decreased empathy which was increased by activation of VTA-ACC DA pathway. Therefore, DA system and VTA to ACC DA neuron projections play an important roles in regulating empathetic behavior. Abstract:Positive social interactions during the juvenile and adolescent phases of life are essential for proper social and cognitive development in mammals, including humans. During this developmental period, there is a marked increase in peer–peer interactions, signified by the abundance of social play behaviour. Despite its importance for behavioural development, our knowledge of the neural underpinnings of social play behaviour is limited. Therefore, the purpose of this study was to map the neural circuits involved in social play behaviour in rats. This was achieved by examining cellular activity after social play using the immediate early gene c-Fos as a marker. After a session of social play behaviour, pronounced increases in c-Fos expression were observed in the medial prefrontal cortex, medial and ventral orbitofrontal cortex, dorsal striatum, nucleus accumbens core and shell, lateral amygdala, several thalamic nuclei, dorsal raphe and the pedunculopontine tegmental nucleus. Importantly, the cellular activity patterns after social play were topographically organized in this network, as indicated by play-specific correlations in c-Fos activity between regions with known direct connections. These correlations suggest involvement in social play behaviour of the projections from the medial prefrontal cortex to the striatum, and of amygdala and monoaminergic inputs to frontal cortex and striatum. The analyses presented here outline a topographically organized neural network implicated in processes such as reward, motivation and cognitive control over behaviour, which mediates social play behaviour in rats.

Abstract:Despite the abundance and variety of rodent species worldwide, historically few have been used to study the neurobiology of social signaling. In recent years, the diversity of rodent behaviors has been leveraged to advance our understanding of neural circuits underlying social behaviors, and in particular those related to acoustic communication. Here we highlight recent work investigating vocal behaviors across social contexts in multiple rodent species and discuss how these studies reveal both shared and distinct neural circuits.

Abstract:Deficits in empathy have been proposed to constitute a hallmark of several psychiatric disturbances like conduct disorder, antisocial and narcissistic personality disorders. Limited sensitivity to punishment, shallow or deficient affect and reduced physiological reactivity to environmental stressors have been often reported to co-occur with limited empathy and contribute to the onset of antisocial phenotypes. Empathy in its simplest form (i.e. emotional contagion) is addressed in preclinical models through the evaluation of the social transmission of emotional states: mice exposed to a painful stimulus display a higher response if in the presence of a familiar individual experiencing a higher degree of discomfort, than in isolation. In the present study, we investigated whether a reduction of emotional contagion can be considered a predictor of reduced sociality, sensitivity to punishment and physiological stress reactivity. To this aim, we first evaluated emotional contagion in a group of Balb/cJ mice and then discretised their values in four quartiles. The upper (i.e. Emotional Contagion Prone, ECP) and the lower (i.e. Emotional Contagion Resistant, ECR) quartiles constituted the experimental groups. Our results indicate that mice in the lower quartile are characterized by reduced sociability, impaired memory of negative events and dampened hypothalamic-pituitary-adrenocortical reactivity to external stressors. Furthermore, in the absence of changes in oxytocin receptor density, we show that these mice exhibit elevated concentrations of oxytocin and vasopressin and reduced density of BDNF receptors in behaviourally-relevant brain areas. Thus, not only do present results translate to the preclinical investigation of psychiatric disturbances, but also they can contribute to the study of emotional contagion in terms of its adaptive significance.
