Abstract:The effect of salts on activity and stability and enantioselectivity of Candida rugosa lipase in isooctane was studied. The activity of the lipase lyophilized with LiCl, NaCl or KCl and native lipase for the esterification of lauric acid with 1-propanol in isooctane at aw=0.33 was 0.057, 0.044, 0.033 and 0.027gg−1min−1, respectively. The water content played a main role on the effect of lipase activity and lyophilization process did not change the conformation of the lipase. Salt incorporation could keep the conformation of the lipase and prevented the large change of optimum pH condition happening. The stability of the lyophilized lipase was 3.4 times as much as that of native lipase and the lipase thermostability was also improved. The enantioselectivity of lyophilized lipase (20.3) was about 1.6 times as much as that of the native lipase (13.0).

Abstract:Lipases are useful as catalysts, particularly for the kinetic resolution of racemic alcohols and esters. However, their industrial applications are limited by their poor activities in organic media. We recently found that a lipoprotein lipase from Burkholderia species displays dramatically enhanced activity in organic solvent if the protein is coated with glucose-headed surfactant (GHS). Here we investigate the molecular basis of this enhanced enzymatic activity in organic solvent by performing molecular dynamics simulations on Burkholderia cepacia lipase as a model enzyme. Our simulation results indicate that the enhanced activity of lipase stems from a dual function of GHSs different in water and organic solvent. GHS molecules maintain the open conformation of lipase by providing lipid-like microenvironment surrounding the active site in water and stabilize its native active conformation by providing water-like microenvironment around the surface of the lipase in the organic solvent. Our data also suggest the role of organic cosolvent that can facilitate closed-to-open conformational changes during the freeze-drying process. The computational approach in this study lays its potential for guiding the design of more effective surfactant molecules to improve the activity of lipases in organic solvent.

Abstract:Lipase is one of hydrolase enzyme that catalyzed hydrolysis reaction of triacylglycerols into free fatty acids and monoglycerides or glycerol. These biocatalysts are widely used in several industries, namely food and pharmaceutical industry. The activity of lipase can increase significantly if the substrate forms an emulsion. Since biosurfactant has been known to have emulsification characteristic, the effect of biosurfactant addition into lipase is necessary to be investigated. It is the first report that evaluate the effect of microbial surfactant on lipase activity. The purpose of this research is to analyze the stability of biosurfactant emulsion under various conditions, such as salinity and pH as well as their potential to enhance lipase activity. Biosurfactant used was Halomonas elongata BK-AG18 from the collection of Biochemistry Research Group ITB, Bandung, Indonesia. It was found that after the addition of NaCl, there was no significant decrease in the emulsification activity of the biosurfactant. The emulsification index (IE24) of biosurfactant with several NaCl concentrations (2, 5, 10, 15, 20, and 25%) was obtained around 50%. Stability test of biosurfactant at pH range 4-10 showed the highest IE24 of biosurfactant was obtained at pH 6. The effect of biosurfactants on lipase hydrolysis activity is also discussed in this article. Lipase hydrolysis activity was tested using p-nitrophenyl palmitate substrate. The highest lipase activity was obtained after the addition of 70% biosurfactant (v/v) at 0.026 units. This study shows that biosurfactant from H. elongata BK-AG18 has the potential to increase lipase activity.

Abstract:The primary objective of the present article is to understand how the geometric constraints at the surfactant head affect the lipase activity in the reverse micellar interface. To resolve this issue, surfactants were designed and synthesized, and activity was measured in /water/isooctane/n-hexanol reverse micellar systems at z ([alcohol]/[surfactant])=5.6, pH 6.0 (20 mM phosphate), 25 degrees C across a varying range of W0 ([water]/[surfactant]) using p-nitrophenylalkanoates as the substrate. It was observed that lipase activity increases from surfactants to with the increment in surface area per molecule (Amin) because of the substitution by the bulky tert-butyl group at the polar head. However, the activity was found to be similar for despite an enhancement in the hydrophilic moieties at the interface. This unchanged lipase activity is presumably due to the comparable surface area of to originating from the rigidity at the surfactant head. Noticeably, the enzyme activity improved from with the simultaneous increment of both the hydroxyl group and the flexibility of the headgroup whereas that for increased exclusively with the flexibility of the headgroup. The common parameter in both groups of surfactants and is the flexibility of the headgroup, which possibly enhance Amin and consequently the lipase activity. Thus, the geometric constraints at the surfactant headgroup play a crucial role in modulating the lipase activity profile probably because of the variation in interfacial area.

Lipase Catalysis in Presence of Nonionic Surfactants

Abstract:Lipase can catalyze varieties of reactions at the interface of aqueous and organic phase. Among various alternatives to modify catalytic performance of lipase, the addition of surfactants, particularly nonionic surfactants, has been widely studied. Low concentrations of nonionic surfactants augment lipase catalysis; on increasing surfactant concentration, often the catalytic performance decreases. Mole ratio of water to (nonionic) surfactant also has a profound effect on lipase activity. Catalytic abilities of some lipases are either enhanced or reduced in the presence of all nonionic surfactants of the same type, whereas for some other lipases, nonionic surfactants of the same type have mixed effect. Nonionic surfactant even changes substrate specificity of lipase. Water-in-ionic liquid microemulsion involving nonionic surfactant often performs better than other systems in improving catalytic ability of lipase. Tween and Triton surfactants often enhance enantiomeric separation catalyzed by lipase. Nonionic surfactants significantly affect activities of immobilized lipase, being present either as a component during immobilization or as a component in reaction medium. Lipases coated with nonionic surfactants act better than reverse micelles and microemulsions containing lipase. Thus, nonionic surfactants help lipase catalyzed processes in various media to enhance production of useful compounds like flavor ester, structured lipids, optically pure compounds, and noncrystalline polymers.

Abstract:The use of enzymes in laundry and dish detergent products is growing. Such tendency implies dedicated studies to understand surfactant-enzyme interactions. The interactions between surfactants and enzymes and their impact on the catalytic efficiency represent a central problem and were here evaluated using circular dichroism, dynamic light scattering, and enzyme activity determinations. This work focuses on this key issue by evaluating the role of the ethyleneoxide (EO) groups of anionic surfactants on the structure and activity of a commercial lipase, and by focusing on the protein/surfactant interactions at a molecular level. The conformational changes and enzymatic activity of the protein were evaluated in the presence of sodium dodecyl sulfate (SDS also denoted as SLE0 S) and of sodium lauryl ether sulfate with two EO units (SLE2 S). The results strongly suggest that the presence of EO units in the surfactant polar headgroup determines the stability and the activity of the enzyme. While SDS promotes enzyme denaturation and consequent loss of activity, SLE2 S preserves the enzyme structure and activity. The data further highlights that the electrostatic interactions among the protein groups are changed by the presence of the adsorbed anionic surfactants being such absorption mainly driven by hydrophobic interactions. © 2016 American Institute of Chemical Engineers Biotechnol. Abstract:Non-ionic surfactants were shown to stabilize the active conformation of thermoalkalophilic lipases by mimicking the lipid substrate while the catalytic interactions formed by anionic surfactants have not been well characterized. In this study, we combined μs-scale molecular dynamics (MD) simulations and lipase activity assays to analyze the effect of ionic surfactant, sodium dodecyl sulfate (SDS), on the structure and activity of thermoalkalophilic lipases. Both the open and closed lipase conformations that differ in geometry were recruited to the MD analysis to provide a broader understanding of the molecular effect of SDS on the lipase structure. Simulations at 298 K showed the potential of SDS for maintaining the active lipase through binding to the sn-1 acyl-chain binding pocket in the open conformation or transforming the closed conformation to an open-like state. Consistent with MD findings, experimental analysis showed increased lipase activity upon SDS incubation at ambient temperature. Notably, the lipase cores stayed intact throughout 2 μs regardless of an increase in the simulation temperature or SDS concentration. However, the surface structures were unfolded in the presence of SDS and at elevated temperature for both conformations. Simulations of the dimeric lipase were also carried out and showed reduced flexibility of the surface structures which were unfolded in the monomer, indicating the insulating role of dimer interactions against SDS. Taken together, this study provides insights into the possible substrate mimicry by the ionic surfactant SDS for the thermoalkalophilic lipases without temperature elevation, underscoring SDS's potential for interfacial activation at ambient temperatures.

Abstract:Interfacially active lipases can be immobilized at a biphasic interface to enhance turnover recyclability and to facilitate product separation. Extensive coarse-grained molecular dynamics simulations of lipase A (LipA) from Bacillus subtilis show a bimodal orientational distribution of the enzyme at an oil-water interface, arising from its ellipsoidal Janus particle-like character. The relative orientational preference can be tuned by pH. The simulations rationalize a rare experimental observation of an order of magnitude increase in the turnover rate of this lipase upon its noncovalent tagging by a single surfactant molecule at the interface, compared to its rate in bulk water. The adsorption free energy, the interfacial activation, a decrease in the number of orientational fluctuations, and an increased rate of translational diffusion, to all of which the Janus character of LipA contributes, are the factors responsible for this enhancement. This study can spur further investigations of the Janus behavior of enzymes to enhance their activity as well as to stabilize the biphasic emulsion needed for interfacial catalysis.

Abstract:Lipase was immobilized with Cu3(PO4)2⋅3H2O to synthesize lipase-Cu3(PO4)2⋅3H2O hybrid microflowers (Lip-MFs). Surfactants (Tween 80, Triton X-100 and CTAB) were added to study their effect on the morphology and activity of Lip-MFs. SEM images revealed that surfactants can increase the size of Lip-MFs and make the hierarchical structure more compact. Further, compared with other surfactants, the activity of CTAB-Lip-MFs was 1.45 times of that of free lipase, demonstrating CTAB is conducive to improve the activity of lipase.

Abstract:In this work, we present the effects of ionic and zwitterionic surfactants on the hydrolytic activity of Candida rugosa lipase (CRL), one of the most important and widely used microbial lipases. A series of amine N-oxide surfactants was studied to explore the relationship between their molecular structures and their effect on catalytic properties of CRL. These zwitterionic amphiphiles are known for their ability to form aggregates that can increase their size, thanks to a sphere-rod transition, without any additive. Enzyme activity seemed to be improved by morphological changes of micelles from spherical to rod-like, and the structure of the monomers played a crucial role in this transition. In fact, all the amine oxides investigated provoked superactivation, but the CRL activity increased by lengthening the alkyl chain of N-oxide surfactants, whereas it decreased in the presence of bulky head groups. Superactivity was mainly because of an increase in kcat (0.57 s-1 in buffer, 0.80-1.99 s-1 in surfactant solutions) and, in some cases, a decrease in KM (2 × 10-3 M in buffer, 1.08-4.28 × 10-3 M in surfactant solutions). Micelles seemed to play a dual role: superactivity occurred at surfactant concentrations higher than their critical micelle concentration, but, on the other hand, micelles subtracted the substrate from the bulk, making it unavailable for the catalysis.

Abstract:Although polythiocyanogen (SCN)(x), has been known for a long time, its solid state structure continues to be uncertain and controversial. Reported here are density functional calculations for linear polymers comprising repeating 1,2,4-dithiazole or 1,2,4-thiadiazole rings, an S-S bridged isomer, and a variety of cyclic phyrin-like systems resulting from cyclization of these linear polymers. The computed energies and comparison of GIAO-derived relative magnetic shieldings for the (13)C and (15)N nuclei with experimental values reported by Woollins and co-workers confirms their assignment of the polymer as based on the 1,2,4-dithiazole ring. It is proposed here that the cyclic forms of this polymer are a better match with experiment than the purely linear forms; the (33)S spectra are likely to be the best discriminator for these diverse structures. A number of these cyclic forms are modelled on the conformations of analogous phyrins such as the recently reported dodecaphyrin, and have lemniscular figure-eight or helical topologies which are recognized here as being double or higher twisted Möbius pi-conjugated rings. The aromatic character of several forms is dissected using ELF(pi) (electron localization) analyses. Based on the propensity of the phyrin rings to form both mono and dinuclear metal complexes, and a specific analogy to a uranylpentaphyrin, it is proposed that a UO(2) complex of e.g. a (SCN)(2) pentamer might be isolable.
Simulation Study on Molecular Behavior of Rhamnolipids and Biobased Zwitterionic Surfactants at the Oil/Water Interface: Effect of Rhamnose Moiety Structures

Abstract:Molecular behavior of rhamnolipid mixed with a biobased zwitterionic surfactant at an n-hexadecane/water interface has been studied, and the effects of a rhamnose moiety and composition are evaluated. Results showed that rhamnolipid abundantly interacts with biobased surfactant EAB by means of hydrophobic interactions between aliphatic tails and electrostatic interactions between headgroups, including the attractive interaction between COO- of rhamnolipids and N+ of biobased surfactants and the repulsive interaction between COO- of both surfactants. Dirhamnolipid has a larger number of bound Na+ and a more stable bound structure of COO- ∼ Na+, which screens the repulsive interaction between two kinds of surfactants and shows a more homogeneous distribution with biobased surfactants. The interfacial tension between n-hexadecane and water has been synergistically reduced by dirhamnolipids mixed with biobased surfactants at a higher molar ratio of biobased surfactants. Monorhamnolipids show a strengthened interaction with N+ of biobased surfactants and a more stable hydrogen bond with water relative to that of dirhamnolipids, and there is no synergistic effect in lowering the interfacial tension for the mixture of monorhamnolipids and biobased surfactants. The present work provides details of the molecular behavior of biosurfactant rhamnolipids mixed with biobased surfactants and obtains the key factor in affecting the interfacial properties of the binary system.

Abstract:Bisphenol A (BPA), an endocrine disruptor, has potential threat to human health due to its widespread use as a plasticizer in many industrial products. Laccase (Lac) can degrade BPA well and the intervention of some surfactants can further improve the degradation of BPA by Lac, but the exact details are not known. In this paper, focused on the Lac and BPA, as well as the surfactants rhamnolipid (RL) and polyethylene glycol (PEG) as research objects, we construct nine research systems by molecular docking, and study the effect of these two surfactants on the interaction of Lac-BPA at the molecular level through molecular dynamics (MD) simulation and MM-PBSA prediction. The results showed that these two surfactants aggregated and encapsulated the enzyme in different degrees during the MD simulations, which separated the surface part of the enzyme from the solvent, thereby weakening the environmental influence on Lac activity and changing the local residues conformation of Lac, leading to the reduction of polar solvation free energy and changing the binding state of Lac-BPA. RL promotes the binding of Lac-BPA primarily by influencing the free energy of polar solvents, while PEG weakens the binding free energy of Lac-BPA complex. These results explain the reason for the lower degradation rate of BPA with the addition of PEG than with the addition of RL in the same system under the appropriate conditions. Our work elucidates the mechanism of action by which two surfactants RL and PEG affect the Lac-BPA interaction which will provide a strong basis for improving the degradation of BPA by Lac at the molecular level.

Abstract:Classical extraction methods used for isolation of active substances from plant material are expensive, complicated and often environmentally unfriendly. The ultrasonic assistance micelle-mediated extraction method (UAMME), based on green chemistry principles, seems to be an interesting alternative. This work aimed to find a connection between the chemical structure of non-ionic surfactants and the efficiency of the extraction process. The effect of hydrophobic chain length and number of ethoxy groups on the quality of Bidens tripartite extracts was investigated. Several ethoxylated fatty alcohols were used: Ceteareth-20, Steareth-20, Oleth-20, Oleth-10, Oleth-5, C12-C13 Pareth-12, C12-C15 Pareth-12 and Ceteareth-12. The bioflavonoid compositions with the HPLC method was determined. The hydrophilic lipophilic balance (HLB) of studied surfactants, as well as the surface tension of surfactant solutions, were compared, to determine the explanation for the obtained differences in bioflavonoids concentration. The structural changes influenced by polyphenol extraction were monitored using Dynamic Light Scattering (DLS) measurements. In this work, probably for the first time, the connection between the chemical structure of non-ionic surfactants and the efficiency of the extraction process was found. The experimental and theoretical approach rationalized the choice of an appropriate eluent. We propose some structurally dependent factors, whose optimal value gave a high efficiency to the UAMME.
A Correlation between the Activity of Candida antarctica Lipase B and Differences in Binding Free Energies of Organic Solvent and Substrate

Abstract:The ability of enzymes to operate in organic solvent is now widely accepted and is the basis for extensive research in enzymology. The challenge is to select the solvent media that allows the modulation of enzyme activity. For a rational selection of a solvent, it is necessary to understand the effect of organic solvent molecules on enzyme structure and the enzymatic reaction on a molecular level. To gain such insight, we combined experimental kinetics studies with full atomic molecular dynamics simulations and found a correlation between the activity of Candida antarctica lipase B (CALB) [for the esterification reaction between butyric acid and ethanol at a fixed water activity] and the binding of the solvent/substrate molecules in the active site region of CALB. We have investigated the influence of four organic solventshexane (HEX), methyl tertiary butyl ether (MTBE), acetonitrile (ACN), and tertiary butanol (TBU)on the catalytic activity of CALB for the esterification reaction. The solvents have been chosen on the basis of different polarity/functional groups. Our study shows that these organic solvents do not alter the overall conformation of CALB; rather, the solvent effects on the performance of the enzyme may be ascribed to binding of solvent molecules to the enzyme active site region and the solvation energy of substrate molecules in the different solvents. Polar solvent molecules interact strongly with CALB and compete with the substrate to bind to the active site region, resulting in an inhibitory effect which is also confirmed by the binding free energies for the solvent and substrate molecules estimated from the simulations. Consequently, the catalytic activity of CALB decreases in polar solvents. This effect is significant, and CALB is over 10 orders of magnitude more active in nonpolar solvents (HEX and MTBE) than in the polar solvents (ACN and TBU). TBU molecules show an exceptional behavior because the solvent molecule forms an extensive hydrogen bond network within the CALB active site region suggesting that solvent molecules rich on hydrogen bond acceptors and donors are poor solvents when used for lipase-catalyzed esterification reactions.

Probing Conformational Changes and Interfacial Recognition Site of Lipases With Surfactants and Inhibitors

Abstract:Structural studies on lipases by X-ray crystallography have revealed conformational changes occurring in the presence of surfactants/inhibitors and the pivotal role played by a molecular "lid" of variable size and structure depending on the enzyme. Besides controlling the access to the enzyme active site, the lid is involved in lipase activation, formation of the interfacial recognition site (IRS), and substrate docking within the active site. The combined use of surfactants and inhibitors has been critical for a better understanding of lipase structure-function relationships. An overview of crystal structures of lipases in complex with surfactants and inhibitors reveals common structural features and shows how surfactants monomers interact with the lid in its open conformation. The location of surfactants, inhibitors, and hydrophobic residues exposed upon lid opening provides insights into the IRS of lipases. The mechanism by which surfactants promote the lid opening can be further investigated in solution by site-directed spin labeling of lipase coupled to electron paramagnetic resonance spectroscopy. These experimental approaches are illustrated here by results obtained with mammalian digestive lipases, fungal lipases, and cutinases.

Abstract:A series of 100 ns-long molecular dynamics calculations for binary bilayers of 1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC) and 33 mol% ethylene-glycol-ethers () were performed under the isothermal–isobaric condition ( K and atm) to investigate systematically changes in physico-chemical properties of the binary bilayer depending on the length of the hydrophilic head group and the hydrophobic tail. Remarkable differences were observed in membrane properties between the bilayers mixed with surfactants with a large hydrophilic head group and a small hydrophilic head group. In the former, the binary bilayers extended laterally with more disorder in lateral arrangement of the lipid molecules and more disordered acyl tail conformation of the DMPC molecules in the binary bilayers than those in pure DMPC bilayer. In the latter, the bilayers shrunk laterally with more order in lateral arrangement of the lipids and more ordered DMPC acyl tail conformation. The molecular mechanism of these changes in membrane properties is discussed on the basis of two-dimensional radial distribution functions of the lipid mass centres, radius of gyration of lipids and void distribution in membranes. The umbrella effect of phosphocholine head groups was investigated.

Effect of Surfactant on Phenanthrene Metabolic Kinetics by Citrobacter Sp. SA01

Abstract:To attain a better understanding of the effects of surfactants on the metabolic kinetics of hydrophobic organic compounds, the biodegradation of phenanthrene by Citrobacter sp. SA01 was investigated in a batch experiment containing Tween 80, sodium dodecyl benzene sulfonate and liquid mineral salt medium. The Monod model was modified to effectively describe the partition, phenanthrene biodegradation and biopolymer production. The results showed that Tween 80 and sodium dodecyl benzene sulfonate (each at 50mg/L) enhanced phenanthrene metabolism and poly-β-hydroxybutyrate production as indicated by the increasing amounts of intermediates (by 17.2% to 47.9%), and percentages of poly-β-hydroxybutyrate (by 107.3% and 33.1%) within the cell dry weight when compared to their absence. The modified Monod model was capable of predicting microbial growth, phenanthrene depletion and biopolymer production. Furthermore, the Monod kinetic coefficients were largely determined by the surfactant-enhanced partition, suggesting that partitioning is a critical process in surfactant-enhanced bioremediation of hydrophobic organic compounds.

Abstract:Organic solvent-stable lipases have pronounced impact on industrial economy as they are involved in synthesis by esterification, interesterification, and transesterification. However, very few of such natural lipases have been isolated till date. A study of the recent past provided few pillars to rely on for this work. The three-dimensional structure, inclusive of the surface and active site, of 29 organic solvent-stable lipases was analyzed by subfamily classification and protein solvent molecular docking based on fast Fourier transform correlation approach. The observations revealed that organic solvent stability of lipases is their intrinsic property and unique with respect to each lipase. In this paper, factors like surface distribution of charged, hydrophobic, and neutral residues, interaction of solvents with catalytically immutable residues, and residues interacting with essential water molecules required for lipase activity, synergistically and by mutualism contribute to render a stable lipase organic solvent. The propensity of surface charge in relation to stability in organic solvents by establishing repulsive forces to exclude solvent molecules from interacting with the surface and prohibiting the same from gaining entry to the protein core, thus stabilizing the active conformation, is a new finding. It was also interesting to note that lipases having equivalent surface-exposed positive and negative residues were stable in a wide range of organic solvents, irrespective of their LogP values.
