Abstract:In this paper the chitosanase-producing microbes from the marine environment were studied.The result showed that marine medium can significantly adapt to strain isolation,and the surface of shell was best source of chitonsanase-producing bacterial strain.KJT-A and KJT-D,which were isolated from the crab shell surface of xuwang marine culture zone in zhoushan island,were classified into Pseudomonas and Penicillium based on morphology,analysis of physiology and biochemistry and molecular biology methods.The growth of two strains in different temperature and salinity also discussed.

Abstract:Chitosan is a biopolymer material extracted from marine biomass waste such as shrimp and crab shells, which has good biocompatibility and degradability with great potential for application in the field of wastewater treatment and soil remediation. The higher the degree of deacetylation (DD), the better the adsorption performance of chitosan. Chitin deacetylase (CDA) can specifically catalyze the deacetylate of chitin in a green reaction that is environmentally friendly. However, the scarcity of high yielding chitin deacetylase strains has been regarded as the technical bottleneck of chitosan green production. Here, we screened a natural chitin degrading bacterium from coastal mud and identified it as Bacillus cereus ZWT-08 by re-screening the chitin deacetylase activity and degree of deacetylation values. By optimizing the medium conditions and enzyme production process, ZWT-08 was cultured in fermentation medium with 1% (m/V) glucose and yeast extract at pH 6.0, 37°C, and a stirring speed of 180 r/min. After fermenting in 5 L fermenter for 48 h, the deacetylation activity of the supernatant reached 613.25 U/mL. Electron microscopic examination of the chitin substrate in the fermentation medium revealed a marshmallow-like fluffy texture on its structural surface. Meanwhile, 89.29% of the acetyl groups in this chitin substrate were removed by enzymatic digestion of chitin deacetylase produced by ZWT-08, resulting in the preparation of chitosan a degree of deacetylation higher than 90%. As an effective strain for chitosan production, Bacillus cereus ZWT-08 plays a positive role in the bioconversion of chitin and the upgrading of the chitosan industry.

Abstract:Alternative methods were evaluated for chitin isolation from Acheta domesticus. Chemical demineralization was compared to fermentation with Lactococcus lactis, citric acid treatment, and microwave treatment, leading to a degree of demineralization of 91.1 ± 0.3, 97.3 ± 0.8, 70.5 ± 3.5, and 85.8 ± 1.3%, respectively. Fermentation with Bacillus subtilis, a deep eutectic solvent, and enzymatic digestion were tested for chitin isolation, generating materials with less than half the chitin content when compared to alkaline deproteinization. Chitosan was produced on a large scale by deacetylation of the chitinous material obtained from two selected processes: the chemical treatment and an alternative process combining L. lactis fermentation with bromelain deproteinization. The chemical and alternative processes resulted in similar chitosan content (81.9 and 88.0%), antioxidant activity (59 and 49%), and degree of deacetylation (66.6 and 62.9%), respectively. The chitosan products had comparable physical properties. Therefore, the alternative process is appropriate to replace the chemical process of chitin isolation for industrial applications.

Optimization of conditions for isolation of high quality chitin from shrimp processing raw byproducts using response surface methodology and its characterization

Abstract:Chitin is one of the most abundant bioactive biopolymer on earth. It is commercially extracted from seafood processing crustacean shell byproducts by harsh thermochemical treatments. The extraction conditions, the source and pretreatment of raw material significantly affect its quality and bioactivity. In this investigation response surface methodology (RSM) has been applied to optimize and evaluate the interaction of variables for extraction of high quality chitin from shrimp processing raw byproducts. Variables such as, concentration of HCl (%, v/v) 4.5 (for wet) and 4.9 (for dry), reaction time 3 h, solid liquid ratio of HCl (w/v) 1:5.5 (for wet) and 1:7.9 (for dry) with two treatments achieved >98 % demineralization of shrimp byproduct. Variables such as, concentration of NaOH 3.6 % (w/v), reaction time 2.5 h, temperature 69.0 ± 1 °C, solid liquid ratio of NaOH 7.4 (w/v) and two treatments accomplished >98 % deproteinization of demineralized byproducts. Significant (p ≤ 0.05–0.001) interactive effects were observed between different variables. Chitin obtained in these conditions had residual content (%, w/w) of ash <0.4 and protein <0.8 and the degree of N-acetylation was >93 % with purity of >98 %. In conclusion, the optimized conditions by RSM can be applied for large scale preparation of high quality chitin from raw shrimp byproduct.

Abstract:Chitosan is a versatile biopolymer due to its biocompatibility, biodegradability, antimicrobial, non-toxic, mucoadhesive, and highly adsorptive properties. Chitosan and its derivatives have been used for many biomedical applications. Currently, crustacean shells and other marine organisms are the significant sources of chitin/chitosan production worldwide. However, extraction from marine sources presents several challenges, including an unstable supply of raw materials. Large-scale chitosan extraction from crustacean sources harms the environment by involving harsh processing steps such as alkali deproteinization. Recently many studies have been carried out focusing on alternative sources or eco-friendlier routes for production of chitosan. This paper briefly overviews recent studies on fungi and insect cuticles as alternative chitosan sources. Milder extraction processes for fungal chitosan and the superior quality of the resultant polymer make it highly desirable for biological applications. Biological techniques involving fermentation and enzymatic processing of the raw materials are looked at in detail. In the concluding remarks, the paper highlights the potential of using a combination of "green" technologies and briefly looks at potential biological/biomedical applications of extracted chitinous materials.

Deciphering the Role of Burkholderia Cenocepacia Membrane Proteins in Antimicrobial Properties of Chitosan

Abstract:Chitosan, a versatile derivative of chitin, is widely used as an antimicrobial agent either alone or mixed with other natural polymers. Burkholderia cenocepacia is a multidrug-resistant bacteria and difficult to eradicate. Our previous studies shown that chitosan had strong antibacterial activity against B. cenocepacia. In the current study, we have investigated the molecular aspects for the susceptibility of B. cenocepacia in response to chitosan antibacterial activity. We have conducted RNA expression analysis of drug efflux system by RT-PCR, membrane protein profiling by SDS–PAGE, and by LC-MS/MS analysis following the validation of selected membrane proteins by real-time PCR analysis. By RT-PCR analysis, it was found that orf3, orf9, and orf13 were expressed at detectable levels, which were similar to control, while rest of the orf did not express. Moreover, shotgun proteomics analysis revealed 21 proteins in chitosan-treated cells and 16 proteins in control. Among them 4 proteins were detected as shared proteins under control and chitosan-treated cells and 17 proteins as uniquely identified proteins under chitosan-treated cells. Among the catalog of uniquely identified proteins, there were proteins involved in electron transport chain and ATP synthase, metabolism of carbohydrates and adaptation to atypical conditions proteins which indicate that utilization and pattern of chitosan is diverse which might be responsible for its antibacterial effects on bacteria. Moreover, our results showed that RND drug efflux system, which display the ability to transport a variety of structurally unrelated drugs from a cell and consequently are capable of conferring resistance to a diverse range of chemotherapeutic agents, was not determined to play its role in response to chitosan. It might be lipopolysaccharides interaction with chitosan resulted in the destabilization of membrane protein to membrane lyses to cell death. Membrane proteome analysis were also validated by RT-qPCR analysis, which corroborated our results that of membrane proteins.
Production and isolation of chitosan from Mucor rouxii

Abstract:A method for the lab-scale production and isolation of chitosan (polyglucosamine) from hyphal walls of Mucor rouxii was developed. Hyphal wall yields were generally 16 to 22% on a dry cell weight basis, of which 35 to 40% was glucosamine. Chitosan was readily extracted from purified, mycelial walls with acetic, formic, and hydrochloric acids; the last named was the most efficient. The yield of chitosan isolated ranged from 4 to 8% of the dry weight of the cell wall material.

Abstract:Chitosan was isolated and purified from the mycelia of Rhizomucor miehei and Mucor racemosus. To characterize the extracted materials, their FTIR spectra were compared with that of shrimp chitosan. Final degree of deacetylation which determined by (1)H NMR was obtained as 98.6% for chitosan from R. miehei (named as C1) and 97.1% for chitosan from M. racemosus (named as C2), respectively. To investigate the antimicrobial properties of the isolated fungal chitosans, minimum inhibitory concentration (MIC) values were performed against twelve strains of bacteria and fungi. Data obtained generally showed that the antibacterial and antifungal activities of the purified fungal chitosans were more effective against Escherichia coli, Pseudomonas aeroginosa, Candida albicans and Candida glabrata in comparison to the shrimp chitosan. Taken together, the results suggest that the use of the fungal chitosan could be of interest as a suitable alternative source to shrimp chitosan.

Abstract:Chitin is the second most common polymer after cellulose in earth, existing in the shells of crustaceans like crab and shrimp. Chitosan is a natural amino-polysaccharide derived from chitin, known as one of the most abundant organic materials in nature, it has been widely used in several applications due to its natural origin and exceptional properties such as biocompatibility, biodegradability, non-toxicity, and chelating of metal ions. Chitin and chitosan are characterized by deacetylation degree, one of the most important chemical characteristics that can influence the performance of chitosan in many applications. Chitosan is usually prepared by a thermochemical method, consuming time, energy and reagents. In this review, various methods of chitosan extraction will be approached and compared; the importance of a new method of ecological extraction will be emphasized. Moreover, in order to improve the chitosan functionality, and better control these physicochemical properties, several chemical modifications have been reported. These chemical modifications lead to a broad range of derivatives with a wide range of applications in many fields. Recent examples of the distinct applications, with particular emphasis on environmental applications, have been presented.

Abstract:Background: In the past few years, the production of shrimp shell waste from the seafood processing industries has confronted a significant surge. Furthermore, insignificant dumping of waste has dangerous effects on both nature and human well-being. This marine waste contains a huge quantity of chitin which has several applications in different fields. The chitinase enzyme can achieve degradation of chitin, and the chitin itself can be used as the substrate as well for production of chitinase. In the current study, the chitinase enzyme was produced by Thermomyces lanuginosus. The extracellular chitinase was purified from crude extract using ammonium sulfate precipitation followed by DEAE-cellulose ion-exchange chromatography and Sephadex G-100 gel filtration chromatography. The stability and activity of chitinase with different pH, temperature, different times for a reaction, in the presence of different metal ions, and different concentration of enzyme and substrate were analyzed. Result: The chitinase activity was found to be highest at pH 6.5, 50 °C, and 60 min after the reaction began. and the chitinase showed the highest activity and stability in the presence of β-mercaptoethanol (ME). The SDS-PAGE of denatured purified chitinase showed a protein band of 18 kDa. Conclusion: The characterization study concludes that Cu2+, Hg2+, and EDTA have an inhibitory effect on chitinase activity, whereas β-ME acts as an activator for chitinase activity. The utilization of chitin to produce chitinase and the degradation of chitin using that chitinase enzyme would be an opportunity for bioremediation of shrimp shell waste.

Abstract:Abstract Marine pollution is a significant issue in recent decades, with the increase in industries and their waste harming the environment and ecosystems. Notably, the rise in shellfish industries contributes to tons of shellfish waste composed of up to 58% chitin. Chitin, the second most ample polymer next to cellulose, is insoluble and resistant to degradation. It requires chemical-based treatment or enzymatic hydrolysis to cleave the chitin polymers. The chemical-based treatment can lead to environmental pollution, so to solve this problem, enzymatic hydrolysis is the best option. Moreover, the resulting biopolymer by-products can be used to boost the fish immune system and also as drug delivery agents. Many marine microbial strains have chitinase producing ability. Nevertheless, we still lack an economical and highly stable chitinase enzyme for use in the industrial sector. So we isolate a novel marine bacterial strain Achromobacter xylosoxidans from the shrimp waste disposal site using chitin minimal medium. Placket–Burman and central composite design statistical models for culture condition optimisation predicted a 464.2 U/ml of chitinase production. The culture conditions were optimised for maximum chitinase production recording up to 467 U/ml. This chitinase from the A. xylosoxidans was 100% active at an optimum temperature of 45 °C (withstand up to 55 °C) and pH 8 with 80% stability. The HPLC analysis of chitinase degraded shellfish waste reveals a major amino acid profile composition—arginine, lysine, aspartic acid, alanine, threonine and low levels of isoleucine and methionine. These chitinase degraded products and by-products can be used as supplements in the aquaculture industry.

Abstract:The dwindling supply of the petroleum product and its carbon footprint has initiated search for a sustainable fuel and alternate feed-stocks. One such underexplored feedstock is chitin, a waste derived from sea food processing. The limitation of insolubility and crystallinity inherent in chitin is addressed with the chitin hydrolysates. In the present study, a chitinases producing marine isolate was isolated from the sediments of Arabian Sea from a depth of 20 m. In order to increase the expression of the chitinases, sequential optimisation using one factor at a time and Taguchi experimental designs were employed which resulted in a yield of 13.46 U/mL which was 2.62 fold higher than the initial bioprocess condition values. In a two-step refinery protocol, Candida albicans was evolved towards chitooligosaccharides using chemically synthesized hydrolysates. In a fed –batch fermentation design the Candida yielded a 12.8 % conversion of these commercial chitin oligosaccharides into bioethanol in a run time of 48 h. This is the first report demonstrating the potential of Candida to utilise chitin oligosaccharides for the production of bioethanol.

Abstract:Shell fish processing industry is very common in coastal areas. While processing, only the meat is taken, the head and shells are discarded as waste. On an average, the sea food industry produces 80,000 tons of waste per year. The sheer amount of waste makes degradation a slow process causing accumulation of waste over a period of time. A very simple and effective solution to this environmental hazard is the recycling of shell waste to commercially viable products like chitin. Chitosan is the N-acetyl derivative of chitin obtained by N-deacetylation. Chitosan is widely used in food and bioengineering industries for encapsulation of active food ingredients, enzyme immobilization, as a carrier for controlled drug delivery, in agriculture as a plant growth promoter. Chitosan is also a defense elicitor and an antimicrobial agent. Chitosan has interesting properties such as biodegradability, biocompatibility, bioactivity, nontoxicity and polycationic nature. This review presents structural characteristics and physicochemical properties of chitosan. The methods of preparation of chitosan nanoparticles are detailed. Applications of chitosan nanoparticles are discussed. Applications include drug delivery, encapsulation, antimicrobial agent, plant growth-promoting agent and plant protector.

Extraction of Fungal Chitosan by Leveraging Pineapple Peel Substrate for Sustainable Biopolymer Production

Abstract:The cost-effective production of commercially important biopolymers, such as chitosan, has gained momentum in recent decades owing to its versatile material properties. The seasonal variability in the availability of crustacean waste and fish waste, routinely used for chitosan extraction, has triggered a focus on fungal chitosan as a sustainable alternative. This study demonstrates a cost-effective strategy for cultivating an endophytic fungus isolated from Pichavaram mangrove soil in a pineapple peel-based medium for harvesting fungal biomass. Chitosan was extracted using alkali and acid treatment methods from various combinations of media. The highest chitosan yield (139 ± 0.25 mg/L) was obtained from the pineapple peel waste-derived medium supplemented with peptone. The extracted polymer was characterized by FTIR, XRD, DSC, and TGA analysis. The antioxidant activity of the fungal chitosan was evaluated using DPPH assay and showed an IC50 value of 0.22 mg/L. Subsequently, a transparent chitosan film was fabricated using the extracted fungal chitosan, and its biodegradability was assessed using a soil burial test for 50 days. Biodegradation tests revealed that, after 50 days, a degradation rate of 28.92 ± 0.75% (w/w) was recorded. Thus, this study emphasizes a cost-effective strategy for the production of biopolymers with significant antioxidant activity, which may have promising applications in food packaging if additional investigations are carried out in the future.

Abstract:Chitosan is an amino-polysaccharide made of glucosamine and N-acetyl-D-glucosamine. Owing to its biocompatible, biodegradable, and nontoxic nature, chitosan is considered as biomaterial, and these unique properties attested that chitosan has greater potential for biological applications. As cationic molecule, chitosan interacts with negatively charged gastrointestinal (GI) mucosal surface and hence is considered as potent mucoadhesive. Therefore chitosan-based encapsulation techniques provide better viability of probiotic microorganisms and protecting the latter in food products and at GI tract. In recent eons, chitosan or chitosan-based nanocomposites are considered as an attractive supportive matrix for enzyme immobilization because of the presence of reactive groups like amino and hydroxyl which supports long-term reusability of the immobilized biocatalyst. The current assignment highlights the recent research and cutting-edge strategies regarding chitosan-based nanomaterials in the field of probiotics encapsulation and enzyme immobilization.

Abstract:Chitin is a linear polysaccharide of N-acetylglucosamine, which is highly abundant in nature and mainly produced by marine crustaceans. Chitosan is obtained by hydrolytic deacetylation. Both polysaccharides are renewable resources, simply and cost-effectively extracted from waste material of fish industry, mainly crab and shrimp shells. Research over the past five decades has revealed that chitosan, in particular, possesses unique and useful characteristics such as chemical versatility, polyelectrolyte properties, gel- and film-forming ability, high adsorption capacity, antimicrobial and antioxidative properties, low toxicity, and biocompatibility and biodegradability features. A plethora of chemical chitosan derivatives have been synthesized yielding improved materials with suggested or effective applications in water treatment, biosensor engineering, agriculture, food processing and storage, textile additives, cosmetics fabrication, and in veterinary and human medicine. The number of studies in this research field has exploded particularly during the last two decades. Here, we review recent advances in utilizing chitosan and chitosan derivatives in different technical, agricultural, and biomedical fields.
A Novel Chitosanase from Penicillium oxalicum M2 for Chitooligosaccharide Production: Purification, Identification and Characterization

Abstract:This study discovered a novel chitosanase from Penicillium oxalicum M2 based on a new screening strategy. An extracellular chitosanase was isolated and purified from the fermentation broth of Penicillium oxalicum M2. A 19.34-fold purification was achieved on a cation exchange column. Using sodium dodecylsulfate-polyacrylamide gel electrophoresis (SDS-PAGE) analysis, chitosanase was determined at approximately 42 kDa without any subunits. The sequence of peptide in the protein was identified as SALNKNYITNFSTLR by MALTI-TOF/TOF MS. The maximum catalytic activity of the purified enzyme was 60.45 U/mg at the optimum pH and temperature of 5.5 and 60 °C. The enzyme activity held stability in the range of 35–50 °C and pH 3–4.5. Ca2+, Mn2+, non-ionic surfactants (Tween 20/40/60/80 and Trition X-100) and some common reducing agents (DTT and β-ME) could significantly activate chitosanase. The purified enzyme showed rigorous specificity to chitosan as a substrate. The hydrolysate in the final stage of hydrolysis consisted of chitooligosaccharides with a degree of polymerization ranging from 2 to 5 and without glucosamine or acetylglucosamine. The monomeric enzyme obtained by one-step purification reveal applications potential in sugar industry, and expanded our understanding of the GH75 family chitosanases simultaneously.

Abstract:Due to the versatility of its features and capabilities, chitosan generated from marine crustacean waste is gaining importance and appeal in a wide variety of applications. It was initially used in pharmaceutical and medical applications due to its antibacterial, biocompatible, and biodegradable properties. However, as the demand for innovative materials with environmentally benign properties has increased, the application range of chitosan has expanded, and it is now used in a variety of everyday applications. The most exciting aspect of the chitosan is its bactericidal properties against pathogens, which are prevalent in contaminated water and cause a variety of human ailments. Apart from antimicrobial and water filtration applications, chitosan is used in dentistry, in water filtration membranes to remove metal ions and some heavy metals from industrial effluents, in microbial fuel cell membranes, and in agriculture to maintain moisture in fruits and leaves. It is also used in skin care products and cosmetics as a moisturizer, in conjunction with fertilizer to boost plant immunity, and as a bi-adhesive for bonding woods and metals. As it has the capacity to increase the life span of food items and raw meat, it is an unavoidable component in food packing and preservation. The numerous applications of chitosan are reviewed in this brief study, as well as the approaches used to incorporate chitosan alongside traditional materials and its effect on the outputs.
Modification of chitosan derivatives of environmental and biological interest: a green chemistry approach

Abstract:Chitosan is a non-toxic polyaminosaccharide that is available in a variety of useful forms, and its chemical and biological properties make it a very attractive biomaterial that could be used in a wide variety of medicinal applications. This work focuses on the preparation of different chitosan derivatives by treatment with ethyl cellulose, cellulose triacetate and different carbohydrates in both neutral and slightly acidic media. It also addresses modification with glycidyltrimethyl ammonium chloride, phthalic anhydride and succinic acid derivatives. The obtained derivatives were crosslinked with glutaraldehyde. Thermo-gravimetric (TGA) and FT-IR spectroscopic analyses and electron scanning microscopy (SEM) were used to characterize the obtained products and demonstrate the success of the chitosan-modification process. The obtained products were tested for their ability to uptake transition metal ions from aqueous solutions, and their ion-uptake efficiency was determined with the aid of the ICP-AES technique. The bioactivity of some selected products was tested to study the effect of their concentrations on selected microorganisms. Burkholderia cepaci, Aspergillus niger, and Candida albicans were selected as representative examples of bacteria, yeasts and fungi, respectively.
