Abstract:Phospholipids are ubiquitous components of biomembranes and common biomaterials used in many bioengineering applications. Understanding adsorption of phospholipids at the air-water surface plays an important role in the study of pulmonary surfactants and cell membranes. To date, however, the biophysical mechanisms of phospholipid adsorption are still unknown. It is challenging to reveal the molecular structure of adsorbed phospholipid films. Using combined experiments with constrained drop surfactometry and molecular dynamics simulations, here, we studied the biophysical mechanisms of dipalmitoylphosphatidylcholine (DPPC) adsorption at the air-water surface. It was found that the DPPC film adsorbed from vesicles showed distinct equilibrium surface tensions from the DPPC monolayer spread via organic solvents. Our simulations revealed that only the outer leaflet of the DPPC vesicle is capable of unzipping and spreading at the air-water surface, whereas the inner leaflet remains intact and forms an inverted micelle to the interfacial monolayer. This inverted micelle increases the local curvature of the monolayer, thus leading to a loosely packed monolayer at the air-water surface and hence a higher equilibrium surface tension. These findings provide novel insights, to our knowledge, into the mechanism of the phospholipid and pulmonary surfactant adsorption and may help understand the structure-function correlation in biomembranes.

Abstract:When inhaled nanoparticles (NPs) come into the deep lung, they develop a biomolecular corona by interacting with the pulmonary surfactant. The adsorption of the phospholipids and proteins gives a new biological identity to the NPs, which may alter their subsequent interactions with cells and other biological entities. Investigations of the interaction between the cell membrane and NPs coated with such a biomolecular corona are important in understanding the role of the biofluids on cellular uptake and estimating the dosing capacity and the nanotoxicology of NPs. In this paper, using dissipative particle dynamics, we investigate how the physicochemical properties of the coating pulmonary surfactant lipids and proteins affect the membrane response for inhaled NPs. We pinpoint several key factors in the endocytosis of lipid NPs, including the deformation of the coating lipids, coating lipid density, and ligand-receptor binding strength. Further studies reveal that the deformation of the coating lipids consumes energy but on the other hand promotes the coating ligands to bind with receptors more tightly. The coating lipid density controls the amount of the ligands as well as the hydrophobicity of the lipid NPs, thus affecting the endocytosis kinetics through the specific and nonspecific interactions. It is also found that the hydrophobic surfactant proteins associated with lipids can accelerate the endocytosis process of the NPs, but the endocytosis efficiency mainly depends on the density of the coating surfactant lipids. These findings can help understand how the pulmonary surfactant alters the biocompatibility of the inhaled NPs and provide some guidelines in designing an NP complex for efficient pulmonary drug delivery.
Study Interactions Between Inhaled Nanoparticles and Pulmonary Surfactant Using Molecular Dynamics Simulations

Abstract:Respiratory system is the most important way for the nanoparticles in the environment getting into the human body. Interaction with the pulmonary surfactant film, being the first line of host defense, represents the initial nano-bio interaction in the lungs. Such interaction determines the fate of the inhaled nanoparticles and their potential therapeutic or toxicological effect. Despite considerable progress in optimizing physicochemical properties of nanoparticles for improved delivery and targeting, the mechanisms by which inhaled nanoparticles interact with the pulmonary surfactant are still largely unknown. As it is hard to experimentally observe the molecular behavior in complex biological environment, we investigated the interaction between inhaled nanoparticles and pulmonary surfactant using molecular dynamics simulation. We developed a coarse-grained model for the pulmonary surfactant that contains different lipids and protein fractions, simulated the translocation of nanoparticles through the pulmonary surfactant film, investigated the mechanisms of toxicity caused by the nanoparticles, and unveiled the structure and molecular conformation of the pulmonary surfactant corona formed on nanoparticles of representative surface properties. Our simulation results shed light on understanding the molecular mechanism of nano-bio interactions in the respiratory system.
Influence of Composition and Temperature on Dynamic Properties of Mixed Monolayers of Pulmonary Lipids

Abstract:The main component of pulmonary surfactant is dipalmitoyl phosphatidylcholine (DPPC), which reduces the surface tension almost to zero when the lung surface is compressed, thus preventing the alveolus from collapse in the course of exhalation. In this work the methods of the surface rheology have been employed to determine the influence of six pulmonary lipids on the dynamic surface properties of a DPPC monolayer at different temperatures and in a wide range of surface tensions. Particular attention has been paid to the region of low surface tensions (lower than 25 mN/m) at temperatures of 25 and 35°С, with these conditions being close to the physiological state on the internal surface of lungs. The addition of lipids with similar molecular structures to DPPC does not affect significantly the dynamic surface properties at a temperature of 25°C. At the same time, the addition of these lipids increases the surface elasticity in the region of low surface tensions at 35°С. However, under these conditions, the presence of lipids with unsaturated hydrocarbon radicals in the surface layer leads to the opposite effect and hinders the achievement of low surface tensions during slow compression. The results obtained have shown the possibility to control the properties of the lipid/DPPC mixed monolayer, which can be considered which can be considered as a model of pulmonary surfactant.

Abstract:Hypothesis Atomistically-detailed models of surfactants provide quantitative information on the molecular interactions and spatial distributions at fluid interfaces. Hence, it should be possible to extract from this information, macroscopical thermophysical properties such as interfacial tension, critical micelle concentrations and the relationship between these properties and the bulk fluid surfactant concentrations. Simulations and Experiments Molecular-scale interfacial of systems containing n-dodecyl β-glucoside (APG12) are simulated using classical molecular dynamics. The bulk phases and the corresponding interfacial regions are all explicitly detailed using an all-atom force field (PCFF+). During the simulation, the behaviour of the interface is analyzed geometrically to obtain an approximated value of the critical micelle concentration (CMC) in terms of the surfactant area number density and the interfacial tension is assessed through the analysis of the forces amongst molecules. New experimental determinations are reported for the surface tension of APG12 at the water/air and at the water/n-decane interfaces. Findings We showcase the application of a thermodynamic framework that inter-relates interfacial tensions, surface densities, CMCs and bulk surfactant concentrations, which allows the in silico quantitative prediction of interfacial tension isotherms.

Abstract:We report on the interaction of pulmonary surfactant composed of phospholipids and proteins with nanometric alumina (Al2O3) in the context of lung exposure and nanotoxicity. We study the bulk properties of phospholipid/nanoparticle dispersions and determine the nature of their interactions. The clinical surfactant Curosurf, both native and extruded, and a protein-free surfactant are investigated. The phase behavior of mixed surfactant/particle dispersions was determined by optical and electron microscopy, light scattering and zeta potential measurements. It exhibits broad similarities with that of strongly interacting nanosystems such as polymers, proteins or particles, and supports the hypothesis of electrostatic complexation. At a critical stoichiometry, micron sized aggregates arising from the association between oppositely charged vesicles and nanoparticles are formed. Contrary to the models of lipoprotein corona or of particle wrapping, our work shows that vesicles maintain their structural integrity and trap the particles at their surfaces. The agglomeration of particles in surfactant phase is a phenomenon of importance since it could change the interactions of the particles with lung cells.

Abstract:Thermophoresis has emerged as a powerful tool for characterizing and manipulating colloids at the nano- and micro-scales due to its sensitivity to colloid-solvent interactions. The use of surfactants enables the tailoring of surface chemistry on colloidal particles and the tuning of interfacial interactions. However, the microscopic mechanisms underlying thermophoresis in surfactant solutions remain poorly understood due to the complexity of multiscale interaction coupling. To achieve a more fundamental understanding of the roles of surfactants, we investigated the thermophoretic behavior of silica beads in both ionic and nonionic surfactant solutions at various background temperatures. We provide a complete mechanistic picture of the effects of surfactants on interfacial interactions through mode-coupling analysis of both electrophoretic and thermophoretic experiments. Our results demonstrate that silica thermophoresis is predominantly governed by the dissociation of silanol functional groups at silica-water interfaces in nonionic surfactant solutions, while in ionic surfactant solutions, the primary mechanism driving silica thermophoresis is the adsorption of ionic surfactants onto the silica surface.

Abstract:Much is known about the effect of surfactants on the heterogeneity of liquid-liquid interfaces, but interfacial properties are sensitive to temperature. The relationship between temperature, surfactant adsorption, and interface structure has not yet been a major research topic. In this work, the effect of temperature on the interfacial structure of the tri-n-butyl phosphate saturated water/octane was investigated through molecular dynamics simulations. We compared the differences in interfacial tension curves of water/organic systems with and without surfactants at different temperatures, focusing on the effects of temperature on surfactant adsorption, interfacial stability, hydrogen bond network, and water extraction. We found the temperature dependence of interfacial protrusion formation and provided a molecular-level understanding of how protrusions promote subsequent water extraction processes.

Constrained drop surfactometry for studying adsorbed pulmonary surfactant at physiologically relevant high concentrations

Abstract:Exogenous surfactant therapy has been used as a standard clinical intervention for treating premature newborns with respiratory distress syndrome. The phospholipid concentrations of exogenous surfactants used in clinical practice are consistently higher than 25 mg/mL; while it was estimated that the phospholipid concentration of endogenous surfactant is approximately in the range between 15 and 50 mg/mL. However, most in vitro biophysical simulations of pulmonary surfactants were only capable of studying surfactant concentrations up to 3 mg/mL, one order of magnitude lower than the physiologically relevant concentration. Using a new in vitro biophysical model, called constrained drop surfactometry, in conjunction with atomic force microscopy and other technological advances, we have investigated the biophysical properties, ultrastructure, and topography of the pulmonary surfactant film adsorbed from the subphase at physiologically relevant high surfactant concentrations of 10-35 mg/mL. It was found that the effect of surfactant concentration on the dynamic surface activity of the surfactant film was only important when the surface area of the surfactant film varied no more than 15%, mimicking normal tidal breathing. The adsorbed surfactant film depicts a multilayer conformation consisting of a layer-by-layer assembly of stacked bilayers with the height of the multilayers proportional to the surfactant concentration. Our experimental data suggest that the biophysical function of these multilayer structures formed after de novo adsorption is to act as a buffer zone to store surface-active materials ejected from the interfacial monolayer under extreme conditions such as deep breathing.NEW & NOTEWORTHY An in vitro biophysical model, called constrained drop surfactometry, was developed to study the biophysical properties, ultrastructure, and topography of the pulmonary surfactant film adsorbed from the subphase at physiologically relevant high surfactant concentrations of 10-35 mg/mL. Abstract:Inhaled nanoparticles traveling through the airways are able to reach the respiratory zone of the lungs. In such event, the incoming particles first enter in contact with the liquid lining the alveolar epithelium, the pulmonary surfactant. The pulmonary surfactant is composed of lipids and proteins that are assembled into large vesicular structures. The question of the nature of the biophysicochemical interaction with the pulmonary surfactant is central to understand how the nanoparticles can cross the air-blood barrier. Here we explore the phase behavior of sub-100 nm particles and surfactant substitutes in controlled conditions. Three types of surfactant mimetics, including the exogenous substitute Curosurf, a drug administred to infants with respiratory distress syndrome are tested together with aluminum oxide (Al2O3), silicon dioxide (SiO2) and polymer (latex) nanoparticles. The main result here is the observation of the spontaneous nanoparticle-vesicle aggregation induced by Coulombic attraction. The role of the surface charges is clearly established. We also evaluate the supported lipid bilayer formation recently predicted and find that in the cases studied these structures do not occur. Pertaining to the aggregate internal structure, fluorescence microscopy ascertains that the vesicles and particles are intermixed at the nano- to microscale. With particles acting as stickers between vesicles, it is anticipated that the presence of inhaled nanomaterials in the alveolar spaces could significantly modify the interfacial and bulk properties of the pulmonary surfactant and interfere with the lung physiology.

Abstract:Surfactants and polymers are two common chemicals used to stabilize the wet foam, but molecular-level insights into the structures, dynamics and hydrogen bonds of these agents and water at the surface are still lack. Herein a series of all-atom molecular dynamics simulations were conducted to explore the behaviors of surfactants/polymers at liquid films. Simulations confirmed that OPC water model can better reproduce the surface tension, and surface tension decreases while interface thickness increases as the increase of surfactant surface concentration. Our findings provide molecular insights on the morphology transitions of surfactant monolayers, which is dependent on the van der Waals energy among surfactants. Surfactant monolayers greatly retard the mobility of the interfacial water molecules, but such capability only exists nearby interfacial area. By adding polymers inside, water dynamics can be retarded further through the hydrogen bonding effects. In other words, polymers in the bulk solution produce a “bolt-like” effect which constrains the water around the chains.

Abstract:The surface parameter of protonated surfactin molecules and the structural properties of the protonated surfactin monolayer adsorbed at the air/water interface have been studied by molecular dynamics simulation. The simulation was performed at 293 K and the interfacial concentration of surfactin was set in a range of 0.70-2.20 nm(2) molecule(-1). The results show that the interfacial concentration greatly affects the molecular orientation of surfactin, the structure of the peptide ring backbone and the spatial arrangement of the surfactin monolayer. The peptide ring backbone of the surfactin molecule exhibits a structural flexibility, and a more packed structure is adopted at higher interfacial concentration. The hydrophobic contacts between surfactin molecules and the stability of the secondary structures, beta-turn structure in Leu2 -> Asp5 and the beta-sheet domains, are enhanced when the surfactin molecules are in a very packed situation.

Abstract:Inhaled nanoparticles (<100 nm) reaching the deep lung region first interact with the pulmonary surfactant, a thin lipid film lining the alveolar epithelium. To date, most biophysical studies have focused on particle-induced modifications of the film interfacial properties. In comparison, there is less work on the surfactant bulk properties and on their changes upon particle exposure. Here we study the viscoelastic properties of a biomimetic pulmonary surfactant in the presence of various engineered nanoparticles. The microrheology technique used is based on the remote actuation of micron-sized wires via the application of a rotating magnetic field and on time-lapse optical microscopy. It is found that particles strongly interacting with lipid vesicles, such as cationic silica (SiO2, 42 nm) and alumina (Al2O3, 40 nm) induce profound modifications of the surfactant flow properties, even at low concentrations. In particular, we find that silica causes fluidification, while alumina induces a liquid-to-soft solid transition. Both phenomena are described quantitatively and accounted for in the context of colloidal physics models. It is finally suggested that the structure and viscosity changes could impair the fluid reorganization and recirculation occurring during breathing.

Abstract:Within the deep lung, pulmonary surfactant coats the air-liquid interface at the surface of the alveoli. This complex mixture of amphiphilic molecules and proteins modifies the surface tension and mechanical properties of this interface to assist with breathing. In this study, we examine the effects on pulmonary surfactant function by two industrially used compounds composing surfactants and polymers. Using an experimental method previously developed to imitate the in vivo exposure in the alveoli, we quantify the change in the dilational rheology of the pulmonary surfactant due to the introduction of two widely used chemicals; Benzalkonium Chloride (BAC) and Polyhexamethylene Biguanide (PHMB). We observe that these chemicals alter the dilational rheology of the surfactant monolayer. Using a mechanistic theory, we are able to semi-quantitatively model the changes induced by the introduction of these compounds to the pulmonary surfactant.

The Effect of Mixtures and Additives on Dissolving Surfactant Lamellar Phases

Abstract:Understanding the dissolution process of surfactant solutions is important in formulating product design processes. The main goal of this study is to gain further insights into how additives and mixtures affect surfactant dissolution processes. To achieve this goal, dissipative particle dynamic simulations were used. Lamellar phases at 80% volume of surfactant were initially equilibrated with water. After reaching an equilibrium state, the dissolution simulations were carried out for different surfactant mixtures. To track the dissolution process, different metrics were used, including visual analysis, local concentration analysis, diffusion, and cluster size calculations. Results show that by having a mixture of surfactants, the diffusion of the micelles is not affected only by the size of the micelles as in pure surfactant systems, but there is also an effect due to the composition of the micelles. When oil is added to a surfactant system, the system acts like a longer chain surfactant system, but only when the chain of oil becomes sufficiently long.

Abstract:The Brownian dynamics (BD) simulation of a dilute surfactant solution is conducted in a steady shear flow. The rodlike micelle is assumed as a rigid rod composed of lined-up beads. A novel intercluster potential model is introduced for describing the interactions between micelles. In the model, the Lennard-Jones and the soft-sphere potentials are used as inter-bead potentials for end-end and interior-interior beads, respectively. The micelles are combined at their ends to form a network structure at lower shear rates and are disconnected to become more and more parallel to the shear flow direction with increasing shear rate. The change of micellar microstructures with the variation of the shear rate results in shear thinning characteristics of the computed shear viscosities and first normal stress difference coefficients. The effects of surfactant solution concentration on the micellar structures and rheological properties are also investigated. Results show that the shear viscosities and the first normal stress difference coefficients increase with increasing the viscosity of the surfactant solution.

Abstract:Surfactants play an increasingly important role across diverse scientific and industrial domains. Gaining a deeper understanding of their molecular behavior at various interfaces is thus becoming ever more essential. Despite considerable advances in experimental techniques, challenges in capturing the detailed molecular-level behavior of surfactants at interfaces persist. In this work, we discuss the potential of combining various experimental methods with atomistic molecular dynamics (MD) simulations in studies of surfactant interfacial layers. MD simulations have emerged as a powerful tool that provides detailed insights into molecular structures and dynamic properties, some of which are inaccessible through experimental means alone. By re-examining existing MD simulation data and directly comparing them with experiments, we illustrate how MD simulations can be used to validate and support thermodynamic models and interpret spectroscopy and scattering data. While combining scattering experiments on Langmuir layers of insoluble surfactants with simulations seems to be well-established by now, we emphasize the growing capability of scattering techniques to investigate also the more disordered Gibbs layers of soluble surfactants. Here, MD simulations can now connect the pressure and adsorption isotherms with the equation of state. In light of the ongoing parallel developments of computational and experimental methods, their synergistic use can be expected to drive future progress in surfactant research.

Abstract:Thin liquid films are central to everyday life. They are ubiquitous in modern technology (pharmaceuticals, coatings), consumer products (foams, emulsions) and also serve vital biological functions (tear film of the eye, pulmonary surfactants in the lung). A common feature in all these examples is the presence of surface-active molecules at the air-liquid interface. Though they form only molecularly-thin layers, these surfactants produce complex surface stresses on the free surface, which have important consequences for the dynamics and stability of the underlying thin liquid film. Here we conduct simple thinning experiments to explore the fundamental mechanisms that allow the surfactant molecules to slow the gravity-driven drainage of the underlying film. We present a simple model that works for both soluble and insoluble surfactant systems. We show that surfactants with finite surface rheology influence bulk flow through viscoelastic interfacial stresses, while surfactants with inviscid surfaces achieve stability through opposing surface-tension induced Marangoni flows.

Abstract:The water-oil-rock system's surfactant and electrostatic interactions are essential for removing oil droplets from rock substrates.
