Abstract:Selective and economic conversion of lignocellulosic biomass components to bio-based fuels and chemicals is the major goal of biorefineries, but low yields and selectivity for fuel precursors such as sugars, furanics, and lignin-derived monomers pose significant disadvantages in process economics. In this Minireview we summarize the existing protection strategies used in biomass chemocatalytic conversion processes and focus the discussions on the mechanisms, challenges, and opportunities of each strategy. We introduce a concept of using analogous methods to manipulate biomass catalytic conversion pathways during the upgrading of carbohydrates to fuels and chemicals. This Minireview may provide new insights into the development of selective biorefining processes from a different perspective, expanding the options for selective conversion of biomass to fuels and chemicals.

Abstract:The grand challenge facing the chemical and allied industries in the twenty-first century is the transition to greener, more sustainable manufacturing processes that efficiently use raw materials, eliminate waste and avoid the use of toxic and hazardous materials. It requires a paradigm shift from traditional concepts of process efficiency, focusing on chemical yield, to one that assigns economic value to replacing fossil resources with renewable raw materials, eliminating waste and avoiding the use of toxic and/or hazardous substances. The need for a greening of chemicals manufacture is readily apparent from a consideration of the amounts of waste generated per kilogram of product (the E factors) in various segments of the chemical industry. A primary source of this waste is the use of antiquated 'stoichiometric' technologies and a major challenge is to develop green, catalytic alternatives. Another grand challenge for the twenty-first century, driven by the pressing need for climate change mitigation, is the transition from an unsustainable economy based on fossil resources--oil, coal and natural gas--to a sustainable one based on renewable biomass. In this context, the valorization of waste biomass, which is currently incinerated or goes to landfill, is particularly attractive. The bio-based economy involves cross-disciplinary research at the interface of biotechnology and chemical engineering, focusing on the development of green, chemo- and biocatalytic technologies for waste biomass conversion to biofuels, chemicals and bio-based materials. Biocatalysis has many benefits to offer in this respect. The catalyst is derived from renewable biomass and is biodegradable. Processes are performed under mild conditions and generally produce less waste and are more energy efficient than conventional ones. Thanks to modern advances in biotechnology 'tailor-made' enzymes can be economically produced on a large scale. However, for economic viability it is generally necessary to recover and re-use the enzyme and this can be achieved by immobilization, e.g. as solid cross-linked enzyme aggregates (CLEAs), enabling separation by filtration or centrifugation. A recent advance is the use of 'smart', magnetic CLEAs, which can be separated magnetically from reaction mixtures containing suspensions of solids; truly an example of cross-disciplinary research at the interface of physical and life sciences, which is particularly relevant to biomass conversion processes.

Abstract:This critical review provides a survey illustrated by recent references of different strategies to achieve a sustainable conversion of biomass to bioproducts. Because of the huge number of chemical products that can be potentially manufactured, a selection of starting materials and targeted chemicals has been done. Also, thermochemical conversion processes such as biomass pyrolysis or gasification as well as the synthesis of biofuels were not considered. The synthesis of chemicals by conversion of platform molecules obtained by depolymerisation and fermentation of biopolymers is presently the most widely envisioned approach. Successful catalytic conversion of these building blocks into intermediates, specialties and fine chemicals will be examined. However, the platform molecule value chain is in competition with well-optimised, cost-effective synthesis routes from fossil resources to produce chemicals that have already a market. The literature covering alternative value chains whereby biopolymers are converted in one or few steps to functional materials will be analysed. This approach which does not require the use of isolated, pure chemicals is well adapted to produce high tonnage products, such as paper additives, paints, resins, foams, surfactants, lubricants, and plasticisers. Another objective of the review was to examine critically the green character of conversion processes because using renewables as raw materials does not exempt from abiding by green chemistry principles (368 references).

Abstract:The increasing role of biocatalysis in the green and sustainable manufacture of chemicals is discussed. In the last two decades the breadth and scope of biocatalysis has increased enormously as a result of remarkable advances in metagenomics, protein engineering and bioinformatics. Moreover, the use of enzymes has become more cost-effective through advances in immobilization technologies and the application of the immobilized enzymes in continuous flow operation in packed bed reactors. Consequently, biocatalysis is already the method of choice for the synthesis of enantiopure chiral products for the pharmaceutical and fine chemical industries. Further applications in commodity chemicals manufacture are currently being stimulated by the increasing maturity of biocatalysis and the ongoing transition to a bio-based circular economy based on the valorization of organic waste on the road to net zero manufacturing.

Abstract:The long road from emerging biotechnologies to commercial “green” biosynthetic routes for chemical production relies in part on efficient microbial use of sustainable and renewable waste biomass feedstocks. One solution is to apply the consolidated bioprocessing approach, whereby microorganisms convert lignocellulose waste into advanced fuels and other chemicals. As lignocellulose is a highly complex network of polymers, enzymatic degradation or “saccharification” requires a range of cellulolytic enzymes acting synergistically to release the abundant sugars contained within. Complications arise from the need for extracellular localisation of cellulolytic enzymes, whether they be free or cell-associated. This review highlights the current progress in the consolidated bioprocessing approach, whereby microbial chassis are engineered to grow on lignocellulose as sole carbon sources whilst generating commercially useful chemicals. Future perspectives in the emerging biofoundry approach with bacterial hosts are discussed, where solutions to existing bottlenecks could potentially be overcome though the application of high throughput and iterative Design-Build-Test-Learn methodologies. These rapid automated pathway building infrastructures could be adapted for addressing the challenges of increasing cellulolytic capabilities of microorganisms to commercially viable levels.

Abstract:The chemical industry is currently reliant on a historically inexpensive, petroleum-based carbon feedstock that generates a small collection of platform chemicals from which highly efficient chemical conversions lead to the manufacture of a large variety of chemical products. Recently, a number of factors have coalesced to provide the impetus to explore alternative renewable sources of carbon. Here we discuss the potential impact on the chemical industry of shifting from non-renewable carbon sources to renewable carbon sources. This change to the manufacture of chemicals from biological carbon sources will provide an opportunity for the biological research community to contribute fundamental knowledge concerning carbon metabolism and its regulation. We discuss whether fundamental biological research into metabolic processes at a holistic level, made possible by completed genome sequences and integrated with detailed structural understanding of biocatalysts, can change the chemical industry from being dependent on fossil-carbon feedstocks to using biorenewable feedstocks. We illustrate this potential by discussing the prospect of building a platform technology based upon a concept of combinatorial biosynthesis, which would explore the enzymological flexibilities of polyketide biosynthesis.

Abstract:To meet decarbonisation goals and implement a more sustainable circular economy model, the chemical industry needs to transition from fossil to renewable sources of carbon. Current chemical production is dominated by petroleum, where this broadly uniform feedstock is separated using a single, simple process to give a small range of heteroatom-free molecules that are the platform to a myriad of products. In a biorefinery, however, many feedstocks of widely varying composition using markedly different technologies are processed to give one or two of a wide range of bioderived platform molecules. Here, recent publications are used to highlight selection of the most suitable second or third generation feedstocks, converted using integrated, complementary processes to generate multiple products. This approach generates a range of chemicals, more fully using the carbon source of choice in a sustainable manner, generating more value, which together makes the realisation of the biorefinery concept draw ever closer.

Abstract:Biocatalysis has the potential to address the need for more sustainable organic synthesis routes. Protein engineering can tune enzymes to perform in cascade reactions and for efficient synthesis of enantiomerically enriched compounds, using both natural and new-to-nature reaction pathways. This review highlights recent achievements in biocatalysis, especially the development of novel enzymatic syntheses to access versatile small molecule intermediates and complex biomolecules. Biocatalytic strategies for the degradation of persistent pollutants and approaches for biomass valorization are also discussed. The transition of chemical synthesis to a greener future will be accelerated by implementing enzymes and engineering them for high performance and new activities.

Abstract:Advancements in cell-free synthetic biology are enabling innovations in sustainable biomanufacturing, that may ultimately shift the global manufacturing paradigm toward localized and ecologically harmonized production processes. Cell-free synthetic biology strategies have been developed for the bioproduction of fine chemicals, biofuels and biological materials. Cell-free workflows typically utilize combinations of purified enzymes, cell extracts for biotransformation or cell-free protein synthesis reactions, to assemble and characterize biosynthetic pathways. Importantly, cell-free reactions can combine the advantages of chemical engineering with metabolic engineering, through the direct addition of co-factors, substrates and chemicals –including those that are cytotoxic. Cell-free synthetic biology is also amenable to automatable design cycles through which an array of biological materials and their underpinning biosynthetic pathways can be tested and optimized in parallel. Whilst challenges still remain, recent convergences between the materials sciences and these advancements in cell-free synthetic biology enable new frontiers for materials research.

Abstract:Energy is one of the most complex fields of study and an issue that influences nearly every aspect of modern life. Over the past century, combustion of fossil fuels, particularly in the transportation sector, has been the dominant form of energy release. Refining of petroleum and natural gas into liquid transportation fuels is also the centerpiece of the modern chemical industry used to produce materials, solvents, and other consumer goods. In the face of global climate change, the world is searching for alternative, sustainable means of producing energy carriers and chemical building blocks. The use of biofuels in engines predates modern refinery optimization and today represents a small but significant fraction of liquid transportation fuels burnt each year. Similarly, white biotechnology has been used to produce many natural products through fermentation. The evolution of recombinant DNA technology into modern synthetic biology has expanded the scope of biofuels and bioproducts that can be made by biocatalysts. This opinion examines the current trends in this research space, highlighting the substantial growth in computational tools and the growing influence of renewable electricity in the design of metabolic engineering strategies. In short, advanced biofuel and bioproduct synthesis remains a vibrant and critically important field of study whose focus is shifting away from the conversion of lignocellulosic biomass toward a broader consideration of how to reduce carbon dioxide to fuels and chemical products.

Abstract:Biomass incorporates carbon captured from the atmosphere and can serve as a renewable feedstock for producing valuable chemicals and fuels. Here we look at how electrochemical approaches can impact biomass valorization, focusing on identifying chemical transformations that leverage renewable electricity and feedstocks to produce valorized products via electro-privileged transformations. First, we recommend that the field should explore widening the spectrum of platform chemicals derived from bio-feedstocks, thus offering pathways to molecules that have historically been derived from petroleum. Second, we identify opportunities in electrocatalytic production of energy-dense fuels from biomass that utilize water as the hydrogen source and renewable electricity as the driving force. Finally, we look at the potential in electrochemical depolymerization to preserve key functional groups in raw feedstocks that would otherwise be lost during harsh pre-treatments in traditional depolymerization routes. On the basis of these priorities, we suggest a roadmap for the integration of biomass and electrochemistry and offer milestones required to tap further into the potential of electrochemical biomass valorization.

Abstract:The unabated carbon dioxide (CO 2 ) emission into the atmosphere has exacerbated global climate change, resulting in extreme weather events, biodiversity loss, and an intensified greenhouse effect. To address these challenges and work toward carbon (C) neutrality and reduced CO 2 emissions, the capture and utilization of CO 2 have become imperative in both scientific research and industry. One cutting-edge approach to achieving efficient catalytic performance involves integrating green bioconversion and chemical conversion. This innovative strategy offers several advantages, including environmental friendliness, high efficiency, and multi-selectivity. This study provides a comprehensive review of existing technical routes for carbon sequestration (CS) and introduces two novel CS pathways: the electrochemical-biological hybrid and artificial photosynthesis systems. It also thoroughly examines the synthesis of valuable C n products from the two CS systems employing different catalysts and biocatalysts. As both systems heavily rely on electron transfer, direct and mediated electron transfer has been discussed and summarized in detail. Additionally, this study explores the conditions suitable for different catalysts and assesses the strengths and weaknesses of biocatalysts. We also explored the biocompatibility of the electrode materials and developed novel materials. These materials were specifically engineered to combine with enzymes or microbial cells to solve the biocompatibility problem, while improving the electron transfer efficiency of both. Furthermore, this review summarizes the relevant systems developed in recent years for manufacturing different products, along with their respective production efficiencies, providing a solid database for development in this direction. The novel chemical-biological combination proposed herein holds great promise for the future conversion of CO 2 into advanced organic compounds. Additionally, it offers exciting prospects for utilizing CO 2 in synthesizing a wide range of industrial products. Ultimately, the present study provides a unique perspective for achieving the vital goals of "peak shaving" and C-neutrality, contributing significantly to our collective efforts to combat climate change and its associated challenges.

Abstract:Bioprocesses use cost-effective renewable resources as feedstock to produce various products with less energy consumption under mild conditions. The technical progress in the modification of biocatalysts including enzymes and cells has significantly improved their efficiencies. In this article, the strategies involving molecular, cellular and community levels for improving various bioprocesses are reviewed with specific examples presented. To modify of enzyme molecule to better fit biomanufacture, semi-rational design of enzyme molecules and intensification of substrate channelling have been proved as effective methods. Progress in metabolic engineering and synthetic biology has promoted engineering the intelligent microbial cell factories. Through modularization of non-native synthesis pathways, microbial cell factories can be optimized at whole cellular levels, and consequently achieved the efficient production of pharmaceuticals, biofuels and chemical compounds as well as natural products. The use of synergistic interactions in microbial communities would provide an opportunity to engineer more complex and robust functions. In addition, several special technologies including bioprocessing under microgravity conditions and nanobiotechnology provide novel ways for developing new applied areas and opportunity to improve bioprocess efficiency.

Abstract:With crude oil reserves dwindling, the hunt for a sustainable alternative feedstock for fuels and materials for our society continues to expand. The biorefinery concept has enjoyed both a surge in popularity and also vocal opposition to the idea of diverting food-grade land and crops for this purpose. The idea of using the inevitable wastes arising from biomass processing, particularly farming and food production, is, therefore, gaining more attention as the feedstock for the biorefinery. For the three main components of biomass—carbohydrates, lipids, and proteins—there are long-established processes for using some of these by-products. However, the recent advances in chemical technologies are expanding both the feedstocks available for processing and the products that be obtained. Herein, this review presents some of the more recent developments in processing these molecules for green materials, as well as case studies that bring these technologies and materials together into final products for applied usage.

Abstract:The role and power of biocatalysis in sustainable chemistry has been continuously brought forward step by step to its present outstanding position. The problem-solving capabilities of biocatalysis have been realized by numerous substantial achievements in biology, chemistry and engineering. Advances and breakthroughs in the life sciences and interdisciplinary cooperation with chemistry have clearly accelerated the implementation of biocatalytic synthesis in modern chemistry. Resource-efficient biocatalytic manufacturing processes have already provided numerous benefits to sustainable chemistry as well as customer-centric value creation in the pharmaceutical, food, flavor, fragrance, vitamin, agrochemical, polymer, specialty, and fine chemical industries. Biocatalysis can make significant contributions not only to manufacturing processes, but also to the design of completely new value-creation chains. Biocatalysis can now be considered as a key enabling technology to implement sustainable chemistry.

Abstract:The use of biorefineries for the production of chemicals as well as materials and energy products is key to ensuring a sustainable future for the chemical and allied industries. Through the integration of green chemistry into biorefineries, and the use of low environmental impact technologies, we can establish future supply chains for genuinely green and sustainable chemical products. The first step in these future biorefineries should be the benign extraction of surface chemicals; here the use of greener solvents, such as supercritical carbon dioxide and bioethanol, should be considered. The residues will often be rich in lignocellulosics and the effective separation of the cellulose is a major challenge which may, in the future, be assisted by greener solvents, such as ionic liquids. Lignin is nature's major source of aromatics; we need new ways to produce small aromatic building blocks from lignin in order to satisfy the enormous and diverse industrial demand for aromatics. Fermentation can be used to convert biomass into a wide range of bioplatform chemicals in addition to ethanol. Their green chemical conversion to higher value chemicals is as important as their efficient production; here clean technologies such as catalysis – notably biocatalysis and heterogeneous catalysis – the use of benign solvents, and energy efficient reactors are essential. Thermochemical processes for the conversion of biomass, such as the production of pyrolysis oil, will also play an important role in future biorefineries and here again green chemistry methods should be used to go to higher value downstream chemicals. Abstract:Originating from the desire to improve sustainability, producing fuels and chemicals from the conversion of biomass and waste plastic has become an important research topic in the twenty-first century. Although biomass is natural and plastic synthetic, the chemical nature of the two are not as distinct as they first appear. They share substantial structural similarities in terms of their polymeric nature and the types of bonds linking their monomeric units, resulting in close relationships between the two materials and their conversions. Previously, their transformations were mostly studied and reviewed separately in the literature. Here, we summarize the catalytic conversion of biomass and waste plastics, with a focus on bond activation chemistry and catalyst design.
