Abstract:Carcass quality and intramuscular fat content of castrates and gilts are compared and a possible dependence of intramuscular fat content on sex and slaughter weight is determined in the pig population of the Slovak Republic. A total of 129 pigs of three different genotypes were included in the experiment. After slaughter, the carcasses were weighed and backfat thickness was measured. On the next day, carcass dissection described by Walstra and Merkus (1995) was determined. Four prime cuts (shoulder, loin, ham and belly) were further dissected to meat, bones and fat with skin. Intramuscular fat content was analysed in a laboratory from the samples (100 g) of musculus longissimus dorsi . The results were statistically analysed using SAS/STAT and a linear model was used to find the dependence of intramuscular fat content. Correlation coefficients between carcass traits and intramuscular fat were also calculated. Sex of pigs, and particularly weight of lean meat and weight of fatty parts had a significant effect on intramuscular fat content ( P < 0.0001 and P = 0.0022) while no effect of the genotype and slaughter weight was observed. Significant differences between castrates and gilts were found in almost all observed traits, e.g. average backfat thickness (29.01 vs. 25.56 mm), percentage of meat (52.77 vs. 57.68%), intramuscular fat content (2.49 vs. 2.00%). Generally, the intramuscular fat in the pig population is not sufficient (2.25%), therefore it would be desirable to include this trait in selection strategies in pig breeding.

Study on the Meat Quality and Fatty Acid of Longissimus Muscle in Different Breeds of Pigs

Abstract:To explore the effect of different breeds on conventional meat quality traits and delicious flavor of longissimus muscle,this study selected the local best breeds of Jinhua pig,hybrid breeds of Jinhua pig,the lean type pig,and foreign lean meat breed large Yorkshire and so on.After slaughtered,the conventional meat quality traits and the contents of fatty acid in the longissimus muscle were measured.The results showed that Jinhua Pigs pH value,meat color,marbling and hydraulic systems were the highest in the six varieties,which were significantly higher than those of foreign lean type breed Yorkshire pigs (P＜ 0.05);Jinhua Pigs Shear force and the rate of water loss were the lowest,which were significantly lower than those of foreign meat type swine (P＜0.05).Among the 13 kinds of fatty acids except the zoomaric acid in longest muscle of the six breeds,the difference was significant (P＜0.05) or extremely significant (P＜0.01).The content of saturated fatty acid,the sum of saturated fatty acids and mono unsaturated fatty acid and the ratio of saturated fatty acids to total unsaturated fatty acids in the longissimus muscle of Jinhua pig were significantly higher than those of Yorkshire (P＜0.01);The content of monounsaturated fatty acid in the longissimus muscle of Jinhua pig was significantly higher than that of Yorkshire pigs (P＜ 0.05);the content of polyunsaturated fatty acid in the longissimus muscle of Jinhua pig was significantly lower than Yorkshire pigs (P＜0.01).The content of saturated fat acid and the sum of saturated fatty acids and mono unsaturated fatty acid in the longissimus muscle of Duroc × Jinhua were higher than those of Landrace×Jinhua and Duroc× Landrace×Jinhua pigs and Yorkshire,but the four breeds had no significant difference (P＞0.05).The content of linoleic acid in Duroc× Landrace×Jinhua pigs among the hybrid varieties of Jinhua pig is the highest,which were significantly higher than that of Duroc×Jinhua pigs and Landrace×Jinhua pigs (P＜0.01).
Effects of H-FABP Gene Polymorphisms and Nutritional Factors on Pork Quality

Abstract:Pork quality is affected by both genetic and nutritional factors. However, few researches focused simultaneously on the effects of these two kinds of factors. In order to study the co-effects of these two kinds of factors simultaneously, we implemented this experiment, 136 PIC hybrid pigs with about 65 kg body weight were randomly divided into 4 groups; each group was fed with a different kind of rations. After 35 days of feeding, the pigs were slaughtered at about 95 kg body weight and the pork quality was evaluated. Then the polymorphism of H-FABP gene was analyzed and an association analysis was conducted. The results are as follows: (1) feed ingredient has very significant effect on meat color (MC), pH24, intramuscular fat (IMF, %), and intramuscular protein (IMP, %); (2) H-FABP gene polymorphism has very significant effect on IMF (%) and IMP (%); (3) the interaction between gene and feed ingredient has significant effect on pH and IMF (%), and pH and IMF (%) were the highest for AA genotype in group 0 and group 1, respectively. These results suggest that both genetic and nutritional factors should be concerned in the improvement of pork quality.

Abstract:Meat quality is a critical aspect of pig breeding. In addition to genetics, meat quality is also influenced by nutritional and environmental factors. In this study, three pig breeds, Shengxianhua, Jiaxing, and Qinglian Black (SXH, JXB and QLB), were used as experimental animals. Transcriptional analysis was performed on the longissimus thoracis (LT) muscle to investigate variations in intramuscular fat (IMF), inosine monophosphate (IMP), amino acids, and muscle fiber morphology across different breeds. Ingenuity canonical pathway analysis (IPA) identified biological processes and key driver genes related to metabolism and muscle development. Additionally, weighted gene co-expression network analysis (WGCNA) revealed gene modules associated with IMP. KEGG and GO analyses identified specific biological processes and signaling pathways related to IMP, including the Oxidative Phosphorylation pathway and rRNA Metabolic Processes. These findings provide novel insights into the molecular regulatory mechanisms underlying meat quality variations among pig breeds.
Effect of different protein sources on the meat quality of Złotnicka Spotted pigs with particular emphasis of slaughter body weight

Abstract:Abstract The aim of the study was to determine the impact of protein feeding from Fabaceae plants and the final slaughter weight on the quality of meat of native Złotnicka Spotted pigs. The experiment was carried out on 96 fattening pigs (48 barrows and 48 gilts) of the Złotnicka Spotted breed. Animals were divided into 4 feeding groups: group A – control cereal-soybean mixture, and 3 experimental mixtures containing different types of domestic protein sources (groups B - lupin, C - faba beans and D - peas). All diets were isoproteic and isoenergetic within the fattening phase, i.e. grower and finisher. In addition, pigs were fattened to 3 different slaughter weights: 120, 130 and 140 kg. The type of protein had no effect on fattening and slaughter traits. These traits (feed intake [kg], gaily gain during fattening [g/day], mean backfat thickness [mm]) were influenced by the body weight of the pigs at slaughter (p<0.01). It was found a statistically significant interaction effect between feeding group and body weight on the fat, the fatty acid profile of meat ( longissimus lumborum ) and on oxidative stability (thiobarbituric acid reactive substances - TBARS) of meat. Increasing the slaughter weight to 140 kg affects increasing the fat content (p<0,05) of meat ( longissimus lumborum ) primarily in groups of pigs fed on cereal-soy mix (A) and faba beans (C). In addition, fat up to 140 kg and feeding with faba beans (C) and peas (D) has a beneficial effect by reducing the proportion of PUFA n-6/n-3 in meat (p<0,05). Finally, the benefit of feeding pigs with faba beans (C) up to a slaughter weight of 140 kg is the reduction of TBARS of meat (p<0,05). In conclusion, increasing the slaughter weight up to 140 kg and the use in the feed mixture of faba beans improves the nutritional value and oxidation durability of pork.

Abstract:The objective of the study was to analyse the fatty acid profile (FA) and amino acid profile (AA) in the musculus longissimus lumborum et thoracis ( MLLT ) in two contrast pig populations. The study was carried out on a total of 39 pigs (19 barrows and 20 gilts) of the indigenous Prestice Black-Pied Pig (PB) and 37 pigs (17 barrows and 20 gilts) of the three-breed commercial hybrid of Large White × Landrace × Large White sireline (CH). The pigs were kept in the same conditions of diet and management. The slaughter was carried out at the average weight of 95 kg. Sex of the evaluated pigs was also taken into account. The CH pigs showed lower contents of saturated FA ( p < 0.01), monounsaturated FA (MUFA; p < 0.05) and a higher content of n -6 polyunsaturated FA (PUFA; p < 0.05), more favourable health indexes, expressed as lower values of atherogenic ( p < 0.001), thrombogenic ( p < 0.001) and saturation (saturated/unsaturated FA) index ( p < 0.001) and positively higher values of hypocholesterolemic/hypercholesterolemic FA ratio ( p < 0.01). The gilts showed lower content of MUFA ( p < 0.05). Significant ( p < 0.05) interaction effects between genotype and sex were recorded for the caprylic acid, capric acid, lauric acid, tridecylic acid, myristic acid, stearic acid, arachidic acid, α-linolenic and γ-linolenic acid ( p < 0.01), with the highest content found in PB barrows. Differences in the AA profiles in the MLLT muscle were only minimal. The PB pigs showed higher proportion ( p < 0.05) of threonine, isoleucine and aspartic acid. Higher proportions of threonine and valine were observed in barrows ( p < 0.05). The results indicate that the evaluated populations of pigs are characterised by significantly different composition of FA. The health indexes suggest more favourable values in the meat of CH pigs. The aim of the study was comparison of fatty acid (FA) and amino acid (AA) composition in meat of two contrast pig genotypes. Regarding the FA profile, the modern hybrid showed more favourable health indexes than the Prestice Black-Pied Pig (PB). These two populations of pigs, kept in the same conditions, had similar composition of AAs in meat.

Abstract:The intuitive impression of pork is extremely important in terms of whether consumers are enthusiastic about purchasing it. Flesh color and intramuscular fat (IMF) are indispensable indicators in meat quality assessment. In this study, we determined the flesh color and intramuscular fat at 45 min and 12 h after slaughter (45 mFC, 45 mIMF, 12 hFC, and 12 hIMF) of 1518 commercial Duroc × Landrace × Large White (DLY) pigs. We performed a single nucleotide polymorphism (SNP) genome-wide association study (GWAS) analysis with 28,066 SNPs. This experiment found that the correlation between 45 mFC and 12 hFC was 0.343. The correlation between 45 mIMF and 12 hIMF was 0.238. The heritability of the traits 45 mFC, 12 hFC, 45 mIMF, and 12 hIMF was 0.112, 0.217, 0.139, and 0.178, respectively, and we identified seven SNPs for flesh color and three SNPs for IMF. Finally, several candidate genes regulating these four traits were identified. Three candidate genes related to flesh color were provided: SNCAIP and PRR16 on SSC2, ST3GAL4 on SSC5, and GALR1 on SSC1. A total of three candidate genes related to intramuscular fat were found, including ABLIM3 on SSC2, DPH5 on SSC4, and DOCK10 on SSC15. Furthermore, GO and KEGG analysis revealed that these genes are involved in the regulation of apoptosis and are implicated in functions such as pigmentation and skeletal muscle metabolism. This study applied GWAS to analyze the scoring results of flesh color and IMF in different time periods, and it further revealed the genetic structure of flesh color and IMF traits, which may provide important genetic loci for the subsequent improvement of pig meat quality traits.

Effect of an Outdoor Access System on the Growth Performance, Carcass Characteristics, and Longissimus lumborum Muscle Meat Quality of the Prestice Black-Pied Pig Breed

Abstract:The effect of an outdoor-access vs. conventional indoor system on the growth, carcass characteristics, and longissimus lumborum muscle (LL) meat quality was evaluated in 24 Prestice Black-Pied pigs, during the growing-finishing period. Two groups received the same complete diet and were housed separately under conventional indoor conditions, with only one group having full access to pasture (350 m2/pig). The animals showed acceptable growth rates (outdoor vs. indoor, average of 740 g/d vs. 700 g/d), feed intake (average of 2700 g/d), and feed conversion ratios (FCR) (average of 3.3 vs. 3.5). The rearing system significantly affected the fatty acid composition of the LL. Outdoor pigs had lower ratios of n − 6/n − 3 polyunsaturated fatty acids, saturation indexes, atherogenic indexes, and thrombogenic indexes, compared with indoor-raised pigs. No differences were recorded in carcass characteristics, physical meat quality traits (pH45, pH24, drip loss, water holding capacity), or the chemical composition of the meat (crude protein, cholesterol, intramuscular fat, hydroxyproline, and tocopherol). The sensory analysis of grilled LL muscle found that outdoor pigs received lower evaluation scores for tenderness, juiciness, and chewiness, but had a better overall acceptance compared to pigs reared indoors.

Abstract:The objective of the study was to compare carcass and meat quality among Lithuanian White, Lithuanian Indigenous Wattle and conventional hybrids as well as consumer sensory evaluations. The pigs were slaughtered at a live weight of approximately 95-100 kg. After 24 h of slaughter, carcasses were evaluated and sampled for the analysis of meat quality traits of loins and consumer evaluation of cooked loins and baked bellies. The pigs of both Lithuanian breeds had higher backfat thickness and depth of underbelly but lower loin area compared with hybrids. However, more fatty local breeds had higher intramuscular fat content but lower cholesterol content than hybrids. The loins from local pigs displayed higher pH and colour with lower lightness and yellowness but higher redness and also lower cooking loss, shear force and hardness than conventional hybrids. Pork from lean hybrids had a higher proportion of polyunsaturated fatty acids and more favourable lipid quality indices such as atherogenic (AI) and thrombogenic (TI) indices and the hypocholesterolemic/hypercholesterolemic (h/H) ratio; however, the peroxidisability index (PI) and iodine value (IV) were less favourable compared with local pigs. Consumers evaluated cooked loins and baked bellies. The loins from local breeds scored higher in juiciness, taste and overall acceptability compared with conventional hybrids. However, a higher overall acceptability was observed for the lean bellies of hybrids. The results can be used to increase pork consumption choices.
Pork quality and histological properties of longissimus muscle from boars and early and late immunocastrated pigs

Abstract:Immunocastration has been introduced in pig production to reduce boar taint. However, there is not much information on how different schedule of immunocastration affects meat quality, especially muscle histological properties. In this study, carcass and meat quality characteristics, histological properties of the longissimus dorsi muscle, sensory characteristics and fatty acid composition of meat and fat from entire males (EM), late immunocastrated (LIC, first dose 8 weeks before slaughter, second dose 4 weeks before slaughter) and early immunocastrated pigs (EIC, first dose 13 weeks before slaughter, second dose 8 weeks before slaughter) were compared. Fat thickness was greater in EIC than in LIC and EM. Meat quality parameters differed mainly between EIC and EM, as EIC had a higher pH 45, lower electrical conductivity, lighter and more yellowish loins and lower drip loss than EM. EM had a higher protein and moisture content and a lower fat content than EIC and LIC. No major effect on histological properties was observed, however, the percentage of fast-twitch oxidative fibres tended to be lower in both LIC and EIC animals compared to EM. Pork from EM had higher boar taint and lower pork odour and flavour scores and higher pig and boar taint flavour scores. The results of this study show that the immunocastration schedule influences some carcass, meat and fat quality traits and, because of that, it can be modified to obtain the desired final product.
Effects of various pre-slaughter conditions on pig carcasses and meat quality in a low-input slaughter facility

Abstract:The aim of this study was to determine the effects of gender, slaughter weight, stocking density, lairage time and season on carcass and meat quality parameters in a low-input production system. The study was conducted on 400 slaughter pigs (200 barrows and 200 gilts) with live weight of 115.0 ± 4.70 kg. Barrows had higher slaughter and carcass weights, fat thickness and lower meatiness. Heavier pigs had higher hot and cold carcass weights, fat thickness, lower meatiness, and higher incidences of pale, soft and exudative (PSE) and dark, firm and dry (DFD) meat. Low stocking density during transportation increased skin lesion score, pH 45 value and the incidence of DFD meat; while high stocking density decreased pH 45 value and increased T 45 value and the incidence of PSE meat. Long lairaging reduced slaughter weight, carcass weight, dressing percentage and backfat thickness and increased skin lesion score. After overnight lairaging, pH 45 value and the incidence of DFD meat became higher. After short lairaging, the pH 45 value was lower, while the incidence of PSE meat increased. Pigs slaughtered in winter had the highest slaughter weight, backfat thickness, pH 45 value and incidence of DFD, and the lowest meatiness. Pigs slaughtered in summer had the lowest slaughter and carcass weights and pH 45 value, and the highest incidence of PSE meat. In conclusion, gender influenced carcass quality, slaughter weight and lairage time, and season affected carcass and meat quality, while stocking density had an impact on meat quality. Abstract:The problem of boar taint has recently become a frequent topic due to the ban on surgical castration of piglets without anaesthesia. Genetic selection based on markers that affect the molecular synthesis and degradation of the two main compounds of boar taint - androstenone and skatole - appears to be one of the possibilities. Many genes and their expression profiles associated with androstenone and skatole levels are known, and studies usually analyse them in hybrids. Our study focused on pure pig breeds and their comparison in the genotypes of the CYP2E1 , TEAD3 , HSD3B , and CYB5A genes. We examined four common commercial breeds (Large White, Landrace, Pietrain, Duroc) and compared them with the original Czech autochthonous Přeštice Black Pied breed, which is also classified as a gene source, with the breed being highlighted for its meat quality. Our research shows that there are differences between purebred individuals. Due to breeding programs, genotyping of breeding boars and sows is not only possible but also relevant.

The comparison was carried out on the 8 male and the 8 female animals. The animals were reared under the same housing conditions and were fed with grass hay (ad libitum) , feed straw and calves feed concentrate, and had a free access to the fresh water. The fattening period started from about 70 days of age to required final weight. Length of the fattening period was 180 days. The slaughter and carcass composition, carcass yield, qualitative, physical and technological characteristics of Longissimus thoracis et lumborum muscle were studied. We found a significantly higher proportion of bones and a significantly higher proportion of separable fat in the carcass half in males. Higher proportion of meat were found in females. At the heifers was found, although not significantly, a higher proportion of internal fat (rumen, intestinal, kidney, pelvic). Significantly different pH measured 24 hours post mortem was found between males and females (P<0.01). Parameter of meat colour – lightness was higher identified in male calves at 24 hour and 7 days post mortem . Higher redness value was identified in female calves at 24 hour and 7 days post mortem . Sex Effects on Carcass Characteristics, Meat Quality Traits and Meat Amino Acid and Fatty Acid Compositions in a Novel Duroc Line Pig.

Abstract:In-depth studies of carcass characters and meat quality could provide insight both for breeding improvement and food development in pigs. Breed and gender are two main factors affected the carcass and meat altitude, which plays important roles in pork industry. The aim of this study was to evaluate the sex effects on carcass characteristics and meat quality traits in a novel Duroc strain pig crossbred from French line, American line and Canadian line pigs. A total of 30 pigs (15 surgical-castrated males and 15 females) with similar birthweight (1.8 ± 0.13 kg) was used in experiment. During the experiment period, all pigs were fed same commercial diets. Overall, female pigs observed higher (p < 0.05) carcass weight, slaughter backfat, loin muscle area, loin muscle depth, carcass yield, pH on 45 min, meat histidine and essential amino acid (AA) compositions, and eicosenoic, unsaturation and free fatty acids (FA) compositions compared with meat from castrated males. Whereas, castrated males' meat showed better altitude (p < 0.05) on meat lightness, meat moisture content percentage, total umami AA and stearic acid and saturated FA compositions than those from female ones. In conclusion, the results of this study provide evidence on the sex effects on meat quality and carcass parameters in Duroc strain pigs. Furthermore, this study also give a reference on the relationship between sex and carcass and meat characteristics in Durco strain pigs.
Pork Quality of Two Lithuanian Breeds: Effects of Breed, Gender and Feeding Regimen

Abstract:The diversity of breeds is an important factor influencing carcass and meat quality traits that are also associated with other different effects. The objective of this study was to investigate the effects of breed, gender, and feeding level in the finishing phase on the carcass and meat quality of Lithuanian White and Lithuanian Indigenous Wattle pigs reared indoors. After 60 kg weight, half of the animals from both breeds were fed a restricted diet of approximately 82% of average ad libitum feeding intake, and the other half of pigs were further fed ad libitum to the end of the experiment. Feed restriction during the finishing phase decreased daily gain and weight of pigs at slaughter, and backfat thickness at the tenth rib (p ˂ 0.001, p ˂ 0.01, and p ˂ 0.05, respectively). Lithuanian White pigs demonstrated higher (p ˂ 0.001) growth rate, live body weight at slaughter and carcass weight, and had a higher (p ˂ 0.01) length of carcass and loin area and lower (p ˂ 0.05) backfat thickness at the last rib and at two points at the lumbar area compared with Lithuanian Indigenous Wattle pigs. The semimembranosus muscle of Lithuanian White pigs had relatively (8.1%) lower (p ˂ 0.001) proportions of saturated and 41.2% higher (p ˂ 0.001) proportions of polyunsaturated fatty acids compared with the pigs of the Lithuanian Indigenous Wattle breed, whereas in the longissimus muscle and subcutaneous tissue, the breed only affected the n-6/n-3 ratio (p ˂ 0.01 and p ˂ 0.001, respectively). Gender showed an effect on saturated fatty acids in all the studied tissues, however, the effects on polyunsaturated fatty acids were found in the semimembranosus muscle and subcutaneous tissue (p ˂ 0.05 and p ˂ 0.001) and the effects on monounsaturated fatty acids was found only in the semimembranosus muscle (p ˂ 0.01). These differences in the fatty acid composition of the semimembranosus muscle and subcutaneous tissue of females exhibited more favorable lipid quality indices compared with castrated males (p ˂ 0.001). The semimembranosus muscle of Lithuanian White pigs also showed more favorable lipid quality indices in relation to healthy nutrition compared with Lithuanian Indigenous Wattle pigs (p ˂ 0.001). This study is one of the steps toward the development and utilization of endangered breeds. The obtained information can be used to increase choice in pork production and consumption, and provides new insights for research into the conservation of local breeds.

Abstract:The objective of this study was to investigate the effects of single nucleotide polymorphisms (SNPs) in nine candidate genes on meat quality traits in longissimus dorsi muscle of pigs. Each SNP allele's main effects were as follows: ADRB3 c. 1192G allele increased intramuscular fat (IMF; p<0.01) and yellowness (p<0.05). MC4R c. 745G allele increased body weight (p<0.05). LEPR c. 232T allele increased rib eye area and decreased tenderness (p<0.05). HSL c. 873G allele increased rib eye area and decreased IMF and backfat thickness (p<0.05). LPL c. 74T allele increased IMF, tenderness, and yellowness (p<0.05). MYF5 c. 1205A allele improved tenderness (p<0.05). H-FABP c. 1322T allele improved tenderness and yellowness (p<0.05). PPAR c. 175G allele decreased water loss (p<0.05). The identified genetic markers might be useful for evaluation and utilization of meat quality related gene in the effort to improve porcine meat quality.
Production performance, carcass composition, and adipose tissue traits of heavy pigs: influence of breed and production system

Abstract:Both breed and production systems are responsible for production efficiency and quality traits of pork. Effects of breed and production system within breed on growth, body fatness, and adipose tissues traits were assessed in the pure Basque (B, nonselected, local French) and conventional Large White (LW) breeds, reared either in a conventional (C, slatted floor), alternative (A, indoor straw bedding and outdoor area), or extensive (E, free range) system. A total of 100 castrated males were produced in 2 replicates, each involving 50 pigs distributed in 5 treatments based on breed and production system (i.e., BC, BA, BE, LWC, and LWA [10 pigs/group and per replicate]). From 35 kg BW to slaughter at around 145 kg BW, the BC, BA, LWC, and LWA pigs received the same growing and finishing diets, whereas the BE pigs had free access to the natural resources of the E pen and received a standard growing-finishing diet at restricted allowance according to the farming practices of the B pork chain. The B pigs had lower (P < 0.001) ADG and G:F than the LW pigs and were much older (P < 0.001) at slaughter. The LWA pigs had similar ADG but lower (P = 0.03) G:F than the LWC. Within the B breed, the BA had higher (P = 0.04) and the BE lower (P < 0.001) ADG compared with BC pigs. The B pigs had a higher (P < 0.001) carcass dressing an exhibited around 2-fold higher (P < 0.001) back fat proportion, perirenal fat weight and LM lipid content than the LW pigs. Compared with C, the A system decreased (P = 0.04) carcass dressing within LW but did not influence carcass traits within B pigs. The E system decreased (P ≤ 0.05) carcass dressing, back fat proportion, and LM lipid content in BE compared with BC pigs. The B pigs exhibited larger (P < 0.001) adipocytes in both subcutaneous adipose tissue (SCAT) and LM than the LW pigs. Malic enzyme activity was higher in SCAT of B than LW pigs despite their greater fatness, and was higher (P ≤ 0.01) in BA but lower (P < 0.001) in BE than in BC pigs. The B pigs had higher (P < 0.001) MUFA but lower (P ≤ 0.006) SFA and PUFA fatty acid percentages in SCAT than the LW pigs. Compared with C, the A system had scarce influence on FA composition within each breed, whereas the E system led to lower (P = 0.015) SFA and greater (P < 0.001) PUFA in SCAT of the B pigs. Altogether, the E production system can counteract the genetic potential of B pigs for growth rate but also body fatness.
Performance, Carcass Traits, Pork Quality and Expression of Genes Related to Intramuscular Fat Metabolism of Two Diverse Genetic Lines of Pigs

Abstract:We aimed to evaluate the performance, carcass and pork quality traits, as well as the mRNA expression of genes related to intramuscular fat deposition in female pigs from different genetic lines. A total of eighteen female pigs (Large White × Landrace × Duroc × Pietrain) × (Large White × Landrace) (Hybrid) averaging 88.96 ± 3.44 kg in body weight and twelve female pigs (Duroc) × (Large White × Landrace) (Duroc) averaging 85.63 ± 1.55 kg in body weight were assigned to a completely randomized design experimental trial that lasted 45 days. Pigs from both genetic lines received the same diet, which was initially adjusted for their nutritional requirements from 0 to 17 days of age and subsequently adjusted for nutritional requirements from 17 to 45 days of age. The performance of pigs did not differ among groups (p > 0.05). Duroc pigs showed a lower backfat thickness (p < 0.03) and greater intramuscular fat content (p < 0.1). A greater mRNA expression of the peroxisome proliferator-activated receptor gamma gene (PPARγ, p = 0.008) and fatty acid protein translocase/cluster differentiation (FAT/CD36, p = 0.002) was observed in the Longissimus dorsi muscle of Duroc pigs. Similarly, a greater expression of PPARγ (p = 0.009) and FAT/CD36 (p = 0.02) was observed in the Soleus muscle of Duroc pigs. Overall, we observed that despite the lack of differences in performance between the genetic groups, Duroc pigs had greater intramuscular fat content than hybrid pigs. The increased intramuscular fat content was associated with an increase in the mRNA expression of key transcriptional factors and genes encoding enzymes involved in adipogenesis and lipogenesis in glycolytic and oxidative skeletal muscle tissues.

Abstract:Shanzhu is a local swine with good meat quality in China, so that it is known as one of the most important female parents for new crossbreds nowadays. For this reason,Shanzhu swine( S) is used to cross with Duroc( D) or Berk-shire(B)swine(Both are famous lean swine in the world). In our research,the aim is to select the best crossbreds through meat quality test. Results show that because of crossbreds, meat productivity can be raised significantly, especially in D×S crossbreds( P<0.05,P<0.01) . S appeares good meat quality such as reasonable meat color and mar-bling. These good meat qualities can be transferred to offsprings stably,they are better in D×S and DS×S crossbreds. An-alyzing data,the share force of M. longissimus dorsi( LD) is at the same lever between 6 groups. As above,D×S crossbreds is the reasonable combining for inheriting good meat quality from S. Analyzing major factors on meat color and building up regression equations,they are Ymeatcolor=0.205Xa?-0.067Xb?-0.128XL+7.738(R=0.67,P=0.000),Ymeatcolor=1.423XpH1-0.625XpH24-2.691(R=0.56,P=0.000)and Ymeatcolor= -0.209Xdriploss-0.011Xwaterloss+4.205(R=0.49, P=0.003) ,respectively. As we know,meat color is influenced by many factors,such as a?,b? as well as L values,pH value,drip loss,water loss rate and so on. Fresh meat also appeared reddish,which is related with a?and L values high-ly. The second relative effects are pH1 value and drip loss.

Abstract:Background: Selection for increasing intramuscular fat content would definitively improve the palatability and juiciness of pig meat as well as the sensorial and organoleptic properties of cured products. However, evidences obtained in human and model organisms suggest that high levels of intramuscular fat might alter muscle lipid and carbohydrate metabolism. We have analysed this issue by determining the transcriptomic profiles of Duroc pigs with divergent phenotypes for 13 fatness traits. The strong aptitude of Duroc pigs to have high levels of intramuscular fat makes them a valuable model to analyse the mechanisms that regulate muscle lipid metabolism, an issue with evident implications in the elucidation of the genetic basis of human metabolic diseases such as obesity and insulin resistance. Results: Muscle gene expression profiles of 68 Duroc pigs belonging to two groups (HIGH and LOW) with extreme phenotypes for lipid deposition and composition traits have been analysed. Microarray and quantitative PCR analysis showed that genes related to fatty acid uptake, lipogenesis and triacylglycerol synthesis were upregulated in the muscle tissue of HIGH pigs, which are fatter and have higher amounts of intramuscular fat than their LOW counterparts. Paradoxically, lipolytic genes also showed increased mRNA levels in the HIGH group suggesting the existence of a cycle where triacylglycerols are continuously synthesized and degraded. Several genes related to the insulin-signalling pathway, that is usually impaired in obese humans, were also upregulated. Finally, genes related to antigen-processing and presentation were downregulated in the HIGH group. Conclusion: Our data suggest that selection for increasing intramuscular fat content in pigs would lead to a shift but not a disruption of the metabolic homeostasis of muscle cells. Future studies on the post-translational changes affecting protein activity or expression as well as information about protein location within the cell would be needed to to elucidate the effects of lipid deposition on muscle metabolism in pigs.
