Abstract:Durability problem of dentin bonding originated from defective bonding interfaces caused by inadequate infil-tration of adhesives into demineralized dentin matrix (DDM) is one of the biggest chanllenges in clinical dentistry, while the scientific mechanism remains unclear. We propose that due to poor physical compatibility, the DDM hydrogel-like interface formed by phosphorylated non-collagenous-proteins can be the primary cause of inadequate infiltration of adhesive containing high-content hydrophobic marcromonomers and the formation of numerous defects in the DDM-cured adhesive interface phase. To test this assumption, we develop a novel bonding strategy based on the hydrogel-like interface dehydrogelation refactoring via actively inducing in situ formation of numerous amorphous calcium phosphate nanoparticles within 30 s. The easy-to-collapse DDM is remolded into a robust porous hybrid structure of low polarity. Irrespective of using a wet or dry DDM, the improved physical compatibility between the DDM interface and adhesives greatly promotes infiltration of various adhesives and the integration of the DDM-cured adhesive interface phase. The resulting defect-low bonding interface greatly improves bonding strength and stability indicating the central role of this long -neglected hydrogel-like DDM interface in the problems associated with dentin bonding, which enriches the theory of adhesive dentistry. Free from hard-to-get advanced adhesive materials and sophisticated operations, accessibility and homogeneity of high-quality dentin bonding may benefit clinical dentistry.

Abstract:Numerous approaches have been developed to improve the resin-dentin bond performance, among which the bio-application of mussel-derived compounds have drawn great attention recently. To assess the performance of N-(3,4-dihydroxyphenethyl)methacrylamide (DMA), a mussel-derived compound, as a functional monomer in dental adhesive, its potential property to cross-link with dentin collagen and polymerize with adhesive will first be evaluated by transmission electron microscopy (TEM), attenuated total reflectance technique of Fourier transform infrared (ATR-FTIR), and atomic force microscopy (AFM) via Peakforce QNM mode. After validating the influence of DMA on collagen and adhesive separately, the overall performance of DMA/ethanol solution as a primer in dentin bonding was examined using micro-tensile bond strength (μTBS) testing, fracture pattern observation, and nanoleakage evaluation both immediately and after 10,000 times thermocycling aging. The inhibitory effect of DMA on endogenous metalloproteinases (MMPs) was evaluated by in situ zymography using confocal laser scanning microscopy (CLSM) and the cytotoxicity of DMA was evaluated using cell counting kit-8. Results demonstrated that DMA successfully cross-linked with dentin collagen via non-covalent bonds and had no influence on the polymerization and mechanical properties of the adhesive. Furthermore, even after 10,000 times thermocycling aging, the μTBS and nanoleakage expression of the DMA-treated groups showed no significant change compared with their immediate values. In situ zymography revealed reduced endogenous proteolytic activities after the application of DMA, and no cytotoxicity effect was observed for DMA concentration up to 25 μmol/L. Thus, DMA could be used as a novel, biocompatible functional monomer in dentin bonding.

The application of mussel-inspired molecule in dentin bonding

Abstract:Objectives: To evaluate the effect of dopamine methacrylamide (DMA) pretreatment on resin-dentin bond durability. Methods: Forty acid-etched dentin surfaces were randomly allocated into five groups, and pretreated with one of the primers: 0.1, 1.0, and 10 mmol/L DMA/ dimethyl sulfoxide (DMSO) solutions, pure DMSO, and deionized water (control) before bonding. The bonded teeth of each group were randomly distributed into an immediate subgroup (water storage at 37 °C for 24 h) and an aged subgroup (thermocycling, 5-55 °C, 10,000 times). The teeth were sectioned into 0.9 mm slabs; two middle slabs were used for nanoleakage evaluation, while the remaining slabs were sectioned into beams for micro-tensile bond strength (MTBS) test. Fractured mode and nanoleakage were observed under SEM. FTIR was used to assess the effect of primers on the degree of conversion (DC) of adhesive. Results: The MTBS was significantly affected by primers (p < 0.001), aging conditions (p < 0.001) and their interactions (p < 0.001) as revealed by two-way ANOVA. Significant increase in nanoleakage was observed in both the aged control and DMSO groups when compared to their immediate values (p < 0.05); while no differences were observed in the aged DMA-treated groups (p > 0.05). Among the three DMA-treated groups, 1.0 mM DMA/DMSO primer manifested the highest MTBS with the lowest nanoleakage after thermocycling aging. Application of 10 mM DMA/DMSO significantly decreased the DC of adhesive (p < 0.05). Conclusions: The 1.0 mM DMA/DMSO solution is a potential functional primer to enhance the durability of resin-dentin bond. Clinical significance: Sixty seconds pretreatment of 1.0 mM DMA/DMSO solution on dentin surfaces provides dentists an alternative choice to preserve dentin bond strength. Dopamine methacrylamide might strengthen the integrity of hybrid layer by serving as a bridge connecting the demineralized collagen fibrils and adhesive polymer network.

Abstract:OBJECTIVES: The humid oral environment adversely affects the interaction between a functionalised primer and dentine collagen after acid-etching. Robust adhesion of marine mussels to their wet substrates instigates the quest for a strategy that improves the longevity of resin-dentine bonds. In the present study, an etching strategy based on the incorporation of biomimetic dopamine methacrylamide (DMA) as a functionalised primer into phosphoric acid etchant was developed. The mechanism and effect of this DMA-containing acid-etching strategy on bond durability were examined.METHODS: Etchants with different concentrations of DMA (1, 3 or 5 mM) were formulated and tested for their demineralisation efficacy. The interaction between DMA and dentine collagen, the effect of DMA on collagen stability and the collagenase inhibition capacity of the DMA-containing etchants were evaluated. The effectiveness of this new etching strategy on resin-dentine bond durability was investigated.RESULTS: All etchants were capable of demineralising dentine and exposing the collagen matrix. The latter strongly integrated with DMA via covalent bond, hydrogen bond and Van der Waals' forces. These interactions significantly improve collagen stability and inhibited collagenase activity. Application of the etchant containing 5 mM DMA achieved the most durable bonding interface.CONCLUSION: Dopamine methacrylamide interacts with dentine collagen in a humid environment and improves collagen stability. The monomer effectively inactivates collagenase activity. Acid-etching with 5 mM DMA-containing phosphoric acid has the potential to prolong the longevity of bonded dental restorations without compromising clinical operation time.CLINICAL SIGNIFICANCE: The use of 5 mM dopamine methacrylamide-containing phosphoric acid for etching dentine does not require an additional clinical step and has potential to improve the adhesive performance of bonded dental restorations.

Abstract:Mussels can form tough and long-lasting adhesions to organic and inorganic surfaces in saline and impactive severe aquatic environments. Similar to mussel adhesion, dentin bonding occurs in a wet environment. However, unlike mussels, it is difficult to achieve long-lasting bonds with dentin. Moreover, water is considered a major hindrance in dentin bonding. Inspired by the synergistic effect of cationic lysine (Lys) and catechol on the elimination of the hydration layer during mussel adhesion, a catechol-and Lys-functionalized polymerizable polymer (catechol Lys methacrylate [CLM]) was synthesized to replicate the complex synergy between amino acids and catechol. The bond-promoting potential of 5 mg/mL CLM primer was confirmed using an in vitro wet dentin-bonding model, which was characterized by an improvement in bond strength and durability. CLM can adhere to wet demineralized dentin, with Lys acting as a molecular vanguard to expel water. Subsequently, a myriad of interfacial interactions can be obtained by introducing the catechol group into the interface. Addi-tionally, tough and long-lasting adhesion, similar to that formed by mussels, can be achieved by grafting CLM onto type I collagen via covalent bonds, hydrogen bonds, Van der Waals interactions, and cation-pi interactions, which can enhance the mechanical and chemical stability of collagen, increase the enzymatic resistance of collagen, and provide additional physical/chemical adhesion to dentin bonds. Catechol-and cationic Lys-functionalized poly-mers can improve the stability of the resin-dentin interface under wet conditions.

Abstract:Zinc‐containing polydopamine‐based Zn–PDA@SiO2 dental adhesives exhibit a highly efficient and durable dentin bonding. Antibacterial and anti‐enzymatic activities and superior biosafety are remarkably produced by the synergistic action of Zn and PDA in vitro/vivo. As verified by theoretical simulations, enhanced dentin bonding is achieved through the formation of covalent bonds, Zn‐assisted coordinative bonds, and hydrogen bonds between the PDA and collagen. Dental adhesives are widely used in daily practice for minimally invasive restorative dentistry but suffer from bond degradation and biofilm attack. Bio‐inspired by marine mussels having excellent surface‐adhesion capability and high chemical affinity of polydopamine (PDA) to metal ions, herein, experimental zinc (Zn)‐containing polydopamine‐based adhesive formulation, further being referred to as "Zn–PDA@SiO2"‐incorporated adhesive is proposed as a novel dental adhesive. Different Zn contents (5 and 10 mm) of Zn–PDA@SiO2 are prepared. Considering the synergistic effect of Zn and PDA, Zn–PDA@SiO2 not only presents excellent antibacterial potential and notably inhibits enzymatic activity (soluble and matrix‐bound proteases), but also exhibits superior biocompatibility and biosafety in vitro/vivo. The long‐term bond stability is substantially improved by adding 5 wt% 5 mm Zn–PDA@SiO2 to the primer. The aged bond strength of the experimentally formulated dental adhesives applied in self‐etch (SE) bonding mode is 1.9 times higher than that of the SE gold‐standard adhesive. Molecular dynamics calculations indicate the stable formation of covalent bonds, Zn‐assisted coordinative bonds, and hydrogen bonds between PDA and collagen. Overall, this bioinspired dental adhesive provides an avenue technology for innovative biomedical applications and has already revealed promising perspectives for dental restorative dentistry.

Abstract:The durability of the resin–dentin bonding interface is a key issue in clinical esthetic dentistry. Inspired by the extraordinary bioadhesive properties of marine mussels in a wet environment, we designed and synthetized N-2-(3,4-dihydroxylphenyl) acrylamide (DAA) according to the functional domain of mussel adhesive proteins. DAA’s properties of collagen cross-linking, collagenase inhibition, inducing collagen mineralization in vitro, and as a novel prime monomer for clinical dentin adhesion use, its optimal parameters, and effect on the adhesive longevity and the bonding interface’s integrity and mineralization, were evaluated in vitro and in vivo. The results showed that oxide DAA can inhibit the activity of collagenase and cross collagen fibers to improve the anti-enzymatic hydrolysis of collagen fibers and induce intrafibrillar and interfibrillar collagen mineralization. As a primer used in the etch-rinse tooth adhesive system, oxide DAA can improve the durability and integrity of the bonding interface by anti-degradation and mineralization of the exposed collagen matrix. Oxidized DAA (OX-DAA) is a promising primer for improving dentin durability; using 5% OX-DAA ethanol solution and treating the etched dentin surface for 30 s is the optimal choice when used as a primer in the etch-rinse tooth adhesive system.

Abstract:Objectives To evaluate the benefits of a novel dentin-bonding primer, namely, isocyanate-terminated urethane methacrylate precursor (UMP), which can form covalent bonds with demineralized dentin collagen. Methods The synthesized and purified UMP monomer was characterized and tested its effects on the degree of conversion (DC) and wettability of an acetone-based dental adhesive. Then UMP primers of different concentrations were formulated and used to prepare adhesive specimens, which were compared with solvent-treated groups. Primer-treated specimens with and without aging were also compared. To evaluate the bonding interface, microtensile strength tests, nano-indentation tests and nanoleakage- eavaluation were performed using a field-emission scanning electron microscope and nano-indenter. Data were analyzed using SPSS 20.0 software with significance set at α = 0.05 using one-way analysis of variance (ANOVA) and two-way ANOVA to characterize the effects of the primer. Results Treatment with the UMP primer promoted the DC and wettability of the adhesive on the demineralized dentin surface (P < 0.05); it also increased the bond strength of the aged dentin bonding interface (P < 0.05). Nanoleakage was reduced; the bonding interface became more stable, and the continuity and strength of the hybrid layer improved (P < 0.05) following UMP treatment. The application of 5 mM UMP as a primer for dentin bonding could lead to a stable bonding interface and long-lasting bonding effects. Significance The use of 5 mM UMP primer developed in this study could improve dentin bonding durability and has excellent clinical application prospects.

A Novel Prime-&-rinse Mode Using MDP and MMPs Inhibitors Improves the Dentin Bond Durability of Self-Etch Adhesive.

Abstract:Objectives: The study investigated the effects of novel prime-&-rinse mode using MDP (10-methacryloyloxydecyl dihydmgenphosphate) and matrix metalloproteinase (MMPs) inhibitors on dentin micmtensile bond strengths (MTBS) of self-etch adhesive, resin-dentin interface degradations, and activity of recombinant human (rh) MMP-8, -9. Materials and methods: Eight experimental primers were prepared using 5% and 15% of MDP ethanol-aqueous (1:1) solution in combination with/without MMPs inhibitors (1%benzalkonium chloride (BAC), 1000 mu m/mL polyvinylphosphonic acid (PVPA) and 15%proanthocyanidin (PA)). Ninety human mid-coronal dentin surfaces were applied with the experimental primers, water-sprayed and gently air-dried (prime-&-rinse mode), or not (control, self-etch mode). The specimens were bonded with self-etch adhesive (Clearfil S3 Bond) and composite resin (Clearfil Majesty). The resin-bonded specimens were prepared into multiple micro-beams for MTBS tests after 24 h and 1 yr of water storage. The resin-dentin interfaces were analyzed with SEM/TEM. The inhibitory effects of eight primers on rhMMP-8, 9 were determined. The data were analyzed using two-way ANOVA and LSD multiple comparisons tests. Results: Compared with control, all the primers used in prime-&-rinse mode could significantly improve long-term dentin MTBS (P < 0.05), while 5%MDP-BAC, 15%MDP and 15%MDP+MMPs inhibitors could significantly increase the short-term dentin MTBS (P < 0.05). The SEM/TEM findings revealed that the resin-dentin interfaces were stable over time when the prime-&-rinse mode used. Eight primers possessed the high inhibitory ratio of rh MMP-8, 9. Conclusions: The novel prime-&-rinse mode using 5%MDP-BAC, 15%MDP and 15%MDP+MMPs inhibitors could significantly increase the short- and long-term dentin MTBS of self-etch adhesive. This might be a supplement to contemporary dentin bonding strategies.
Polyphenol-Enriched Extract of Lacquer Sap Used as a Dentine Primer with Benefits of Improving Collagen Cross-Linking and Antibacterial Functions

Abstract:Commercial dentin adhesive systems are applied to restorations due to their resistant bonding properties, but they suffer from the lack of bioactivity and are prone to hydrolysis. Therefore, to overcome these limitations, an eco-friendly natural monomer, urushiol, was adopted to be a primer in dentin bonding due to its interaction with collagen and antibacterial activity, preventing further hydrolysis development. First, urushiol was determined to be capable of improving the biological stability of dentin collagen through cross-linking. Using high-fidelity analytical chemistry techniques, such as Fourier transform infrared spectroscopy, we quantified the effects of urushiol on collagen molecules. It could also effectively decrease weight loss after collagenase ingestion by improving the stability of dentin. Moreover, urushiol inhibited Streptococcus mutans growth as well as its biofilm formation. Finally, we demonstrated that the urushiol primer could improve the bonding strength, particularly after aging. The cross-linking and antibacterial functions of urushiol have provided promising developmental prospects for biomaterials in dentin adhesion.
Effect of a Novel Prime-and-rinse Approach on Short- and Long-Term Dentin Bond Strength of Self-Etch Adhesives.

Abstract:This study investigated the effects of the prime‐and‐rinse approach, using a 10‐methacryloyloxydecyl dihydrogen phosphate (MDP)‐containing primer, on the short‐ and long‐term dentin microtensile bond strengths (MTBSs) of mild self‐etch adhesives. Half of sixty human midcoronal dentin surfaces were polished as control (self‐etch approach), and the other half were polished and further treated with a 15% MDP‐containing primer and thoroughly sprayed with water as prime‐and‐rinse approach. The dentin surfaces were treated with a self‐etch adhesive, and a composite resin was placed on the surfaces. The following materials were used: Clearfil S3 Bond+Clearfil Majesty; G‐Bond+Gradia Direct; Adper Easy One+Z250; and i Bond+Charisma. The MTBS was examined after 24 h and 14 months in water storage. The resin–dentin interfaces were analysed using scanning electron microscopy/transmission electron microscopy. Pretreated dentin surfaces were further analysed using scanning electron microscopy and micro‐Raman spectroscopy. Compared with the self‐etch approach, the prime‐and‐rinse approach significantly increased the dentin MTBS, regardless of the duration of storage. The scanning electron microscopy/transmission electron microscopy findings revealed that the prime‐and‐rinse approach removed most of the dentin smear layer. The Raman spectra of the MDP‐treated dentin reveal the characteristic spectra of collagen, hydroxyapatite, and the monomer. Therefore, the prime‐and‐rinse approach using MDP‐containing primers prior to the application of mild self‐etch adhesives significantly increases the short‐ and long‐term MTBS of dentin.

Abstract:OBJECTIVES:The study investigated the solid reactants of amorphous calcium phosphate (ACP) nanoparticles with 10-methacryloyloxydecyl dihydrogen phosphate (MDP) in ethanol-aqueous solution, and the effect of ACP-MDP suspension as a novel primer on dentin micro-tesnisl bond strengths (MTBS) prior to application of self-etch adhesives.MATERIALS AND METHODS:The ACP nanoparticles were synthesized. The solid reactants of ACP nanoparticles with MDP ethanol-aqueous solution were analyzed by TEM, SEM, XRD and AFM. After the ACP-MDP complexes were attacked by alkaline, acidic and self-etch adhesive challenges, they were analyzed with TEM. The polished mid-coronal dentin surfaces of six third molars were treated with ACP-MDP suspension, 5% MDP solution or not (served as control), and thoroughly water-sprayed. The pretreated dentin surfaces were applied with a self-etch adhesive (Clearfil S3 Bond) and placed with a composite resin (Clearfil Majesty). After 24 h, the resin-bonded samples were cut into multiple beams before the MTBS tests were performed. The failure modes were determined by a stereo-microscopy. The pretreated dentin surfaces were analyzed by SEM. The resin-dentin interfaces were analyzed with TEM.RESULTS:The ACP nanoparticles in the MDP ethanol-aqueous solution at certain molar ratio of Ca:MDP (0.12:1) could self-assemble into ACP-MDP complexes consisting of nanolayering structures. The ACP-MDP self-assembly went from ACP nanoparticles, beaded structures, and twig-like structures to networks. The ACP-MDP complexes can be resistant to adhesive challenge, but not to alkaline and acidic challenges. ACP-MDP complexes possessed a similar modulus of elasticity to dentin, and were stable at least for 3 months. Furthermore, the dentin surfaces treated with ACP-MDP suspension could significantly increase the dentin MTBS when compared with control and those treated with 5% MDP solution (P < 0.05).CONCLUSION:The ACP-MDP suspension at certain molar ratio of Ca:MDP (0.12:1) could be self-assembled into ACP-MDP complexes consisting of nanolayering structures, and could be used as a novel primer to greatly improve the dentin bond strengths.

Abstract:The dentin collagen matrix that is not completely enveloped by resin adhesive is vulnerable to degradation by intrinsic collagenases during the etch-and-rinse process, which contributes to the deterioration of the bonding interface. Current commercial adhesives have no functional components that can form covalent bonds to the dentin collagen matrix. In this study, a photocurable aldehyde, 4-formylphenyl acrylate (FA), was synthesized and for the first time applied as a primer in adhesive dentistry to covalently bind to collagen. Experimental groups with different concentrations of FA (1%, 3%, 5%, 7%, 9%) were prepared as primers. The cytotoxicity was evaluated by live/dead-cell staining and thiazolyl blue tetrazolium bromide assay. The interaction of FA with collagen was examined by attenuated total reflection Fourier transform infrared spectroscopy, hydroxyproline release under the degradation of type I collagenase, and thermogravimetric analysis. An optimal group was selected based on the degree of conversion of 2 universal adhesives and further divided depending on the treatment time (20 s, 30 s, 1 min, 2 min). The bonding performances were evaluated by microtensile strength before and after aging. Finally, the bonding interface was observed under confocal laser scanning microscopy and scanning electron microscope. The results indicated that FA demonstrated good biocompatibility, dentin modification capability, and infiltration. It not only effectively cross-linked dentin collagen to improve its stability against enzymatic hydrolysis and modify the adhesive interface but also potentially acted as a diluting monomer to induce deep penetration of adhesive resin monomers into the dentin. The bonding strength after aging was improved without jeopardizing the degree of conversion of 2 commercial adhesives. Such prominent advantages of using FA to improve the bonding performance promotes its further application in adhesive dentistry.

Abstract:Journal of Dental Research, Ahead of Print. The water-rich nature of the dentin bonding microenvironment, coupled with the stresses on the bonding interface, contributes to the hydrolytic degradation of the hybrid layer, resulting in a decline in bonding durability and, ultimately, restoration failure. Currently, the 3-step etch-and-rinse technique remains the gold standard for dentin bonding, and the bonding mechanism mainly involves a physical interaction with little chemical bonding. In this study, we have developed a siloxane-modified polyurethane monomer (SPU) with acrylate and siloxane modifications that chemically binds to both collagen and hydroxyapatite in dentin. Formulated as a bisphenol A-glycidyl methacrylate alternative, the SPU monomer-based adhesive was designed to improve dentin bonding quality and durability. Attenuated total reflection Fourier transform infrared spectroscopy, thermogravimetric analysis, X-ray photoelectron spectroscopy, scanning electron microscopy, transmission electron microscope, and hydroxyproline release assays were performed on SPU-treated collagen, hydroxyapatite, and acid-etched dentin slices to dentin. The physicochemical properties of the configured SPU adhesives were profiled for polymerization behavior, water contact angle, and tensile strain and strength. The bonding effectiveness was assessed through micro-tensile strength, nano-leakage tests conducted on the bonded samples before and after thermal cycle aging. Finally, we further conducted in vivo and in vitro experiments to assess the biocompatibility of adhesives. The results showed that the siloxane groups of SPU monomer could covalently bind to dentin collagen and hydroxyapatite. The incorporation of SPU in the adhesive led to a significant increase in adhesive polymerization (P < 0.05) and tensile strain at break up to 134.11%. Furthermore, the SPU adhesive significantly improved dentin bond strength (P < 0.05), reduced interfacial nano-leakage (P < 0.05), and displayed good biocompatibility. In conclusion, the application of SPU, which achieves dual chemical bonding with dentin, can improve the quality of the hybrid layer, buffer the interfacial stresses, enhance the interfacial resistance to hydrolysis, and provide a feasible strategy to extend the service life of adhesive restorations.

Abstract:Objectives The main reason for restoration failure is secondary caries caused by biofilm acids. Replacing the failed restorations accounts for 50–70% of all operative work. The objectives of this study were to incorporate a new quaternary ammonium monomer (dimethylaminododecyl methacrylate, DMADDM) and nanoparticles of silver (NAg) into a primer and an adhesive, and to investigate their effects on antibacterial and dentin bonding properties. Methods Scotchbond Multi-Purpose (SBMP) served as control. DMADDM was synthesized and incorporated with NAg into primer/adhesive. A dental plaque microcosm biofilm model with human saliva was used to investigate metabolic activity, colony-forming units (CFU), and lactic acid. Dentin shear bond strengths were measured. Results Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of the new DMADDM were orders of magnitude lower than those of a previous quaternary ammonium dimethacrylate (QADM). Uncured primer with DMADDM had much larger inhibition zones than QADM (p<0.05). Cured primer/adhesive with DMADDM-NAg greatly reduced biofilm metabolic activity (p<0.05). Combining DMADDM with NAg in primer/adhesive resulted in less CFU than DMADDM alone (p<0.05). Lactic acid production by biofilms was reduced by 20-fold via DMADDM-NAg, compared to control. Incorporation of DMADDM and NAg into primer/adhesive did not adversely affect dentin bond strength. Conclusions A new antibacterial monomer DMADDM was synthesized and incorporated into primer/adhesive for the first time. The bonding agents are promising to combat residual bacteria in tooth cavity and invading bacteria at tooth-restoration margins to inhibit caries. DMADDM and NAg are promising for use into a wide range of dental adhesive systems and restoratives.
Novel biomimetic peptide-loaded chitosan nanoparticles improve dentin bonding via promoting dentin remineralization and inhibiting endogenous matrix metalloproteinases

Abstract:Objective: This study aims to synthesize novel chitosan nanoparticles loaded with an amelogenin-derived peptide QP5 (TMC-QP5/NPs), investigate their remineralization capability and inhibitory effects on endogenous matrix metalloproteinases (MMPs), and evaluate the dentin bonding properties of remineralized dentin regulated by TMC-QP5/NPs. Methods: TMC-QP5/NPs were prepared by ionic crosslinking method and characterized by dynamic light scattering method, scanning electron microscopy, transmission electron microscope, atomic force microscope, Fourier transform infrared spectroscopy, and differential scanning calorimetry. The encapsulation and loading efficiency of TMC-QP5/NPs and the release of QP5 were examined. To evaluate the remineralization capability of TMC-QP5/NPs, the mechanical properties, and the changes in structure and composition of differently conditioned dentin were characterized. The MMPs inhibitory effects of TMC-QP5/NPs were explored by MMP Activity Assay and in-situ zymography. The dentin bonding performance was detected by interfacial microleakage and microshear bond strength (μSBS). Results: TMC-QP5/NPs were successfully synthesized, with uniform size, good stability and biosafety. The encapsulation and loading efficiency of TMC-QP5/NPs was respectively 69.63 ± 2.22% and 13.21 ± 0.73%, with a sustained release of QP5. TMC-QP5/NPs could induce mineral deposits on demineralized collagen fibers and partial occlusion of dentin tubules, and recover the surface microhardness of dentin, showing better remineralization effects than QP5. Besides, TMC-QP5/NPs significantly inhibited the endogenous MMPs activity. The remineralized dentin induced by TMC-QP5/NPs exhibited less interfacial microleakage and higher μSBS, greatly improved dentin bonding. Significance: This novel peptide-loaded chitosan nanoparticles improved resin-dentin bonding by promoting dentin remineralization and inactivating MMPs, suggesting a promising strategy for optimizing dentin adhesive restorations.

Abstract:Decades of research have been conducted on 10-Methacryloyloxydecyl dihydrogen phosphate (MDP) through numerous studies. The mechanisms by which its residual calcium salts benefit dentin bonding remain undetermined. The objective of the research was to investigate the role and process of remaining calcium salts in the priming procedure and their capacity for remineralization. The investigation focused on the variations in topological structure, mechanical properties, and chemical interactions between the main agent and the dentin surface. Two adhesive modes including prime-and-rinse(P&R) and prime-and-nonrinse (P&NR) utilized to evaluate the bonding performance and remineralization ability. The findings indicated that both P&R and P&NR methods could eliminate the smear-layer, uncover dentinal-tubules, and generate a textured/rough surface on the dentin. Collagen fibrils exhibited a greater presence of inorganic minerals in the P&NR mode. Compared to control group, both P&R and P&NR groups improved immediate and aging bond strength significantly (P < 0.05).AFM and 3D-STORM revealed MDP and its residual calcium salts distributed in collagen fibrils and expanded collagen matrix. In the P&NR group, TEM revealed that the dentin collagen matrix experienced some remineralization, and there was also mineralization within the collagen fibrils embedded in the bonding interface. Thus, MDP priming improved dentin bonding stability. Residual calcium salts of P&NR process can enhance topological structure of the collagen matrix and induce intrafibrillar mineralization.

Abstract:Objective. This study investigated the effects of prime-and-rinse approach using 10-methacryloyloxydecyl dihydrogen phosphate (MDP) micellar solutions on extrafibrillar demineralization and dentin bond performance of etch-and-rinse adhesive. Methods. The micellar solutions were prepared by adding 15% MDP in two ethanol aqueous (75:25, 55:45 V/V%) solutions, referring to MDP/EtOH75 and MDP/EtOH55. After mid-coronal dentin surfaces were either etched (control) or conditioned with MDP/EtOH75 and MDP/EtOH55 and rinsed, they were applied with adhesive (Adpter Single Bond 2) in dry-or wet-bonding mode and placed with composite resin (Filtek Z350 XT). They were prepared into multiple micro-beams for micro-tensile bond strengths (MTBS) testing after storage in water for 24 h or subjecting to thermocycling. The other pretreated dentin surfaces were analyzed by TF-XRD, ATR-FTIR, HRTEM, FE-SEM, contact angle measurement and nanoindentation testing. The MTBS data was analyzed with two-way ANOVA followed by LSD post-hoc test. Results. MDP/EtOH75 produced significantly greater MTBS values than MDP/EtOH55 and control after thermocycling aging in dry-or wet-bonding mode (P < 0.05). The ATR-FTIR spectrums shows that ratios of phosphate/monomer (1,034 cm(-1)/1,716 cm(-1)) on MDP/EtOH75-, MDP/EtOH(55-)treated dentin surfaces are 0.51 and 0.23, respectively. This is confirmed by HRTEM images and SAED pattern that intrafibrillar minerals were mostly preserved after treatment with MDP/EtOH75. MDP/EtOH75 produced significantly higher elastic modulus and nanohardness on pretreated dentin surface than MDP/EtOH55 (P < 0.05). TF-XRD pattern shows some MDP-Ca salts remained on the primed dentin surface. Significance. Prime-and-rinse approach using MDP/EtOH75 micellar solution could produce mostly extrafibrillar demineralization, and greatly increase dentin bond durability in dry -or wet-bonding mode. Abstract:Dentin bonding is a dynamic process that involves the penetration of adhesive resin monomers into the extrafibrillar and intrafibrillar demineralized collagen matrix using a wet-bonding technique. However, adhesive resin monomers lack the capacity to infiltrate the intrafibrillar space, and the excess water that is introduced by the wet-bonding technique remains at the bonding interface. This imperfectly bonded interface is inclined to hydrolytic degradation, severely jeopardizing the longevity of bonded clinical restorations. The present study introduces a dentin bonding scheme based on a dry-bonding technique, combined with the use of extrafibrillar demineralization and a collagen-reactive monomer (CRM)-based adhesive (CBA). Selective extrafibrillar demineralization was achieved using 1-wt% high-molecular weight (MW) carboxymethyl chitosan (CMCS) within a clinically acceptable timeframe to create a less aggressive bonding substance for dentin bonding due to its selectively extrafibrillar demineralization capacity. CMCS demineralization decreased the activation of in situ collagenase, improved the shrinking resistance of demineralized collagen, and thus provided stronger and more durable bonding than traditional phosphoric acid etching. The new dentin bonding scheme that contained CMCS and CBA and used a dry-bonding technique achieved an encouraging dentin bonding strength and durability with low technical sensitivity. This bonding scheme can be used to improve the stability of the resin-dentin interface and foster the longevity of bonded clinical restorations.

Abstract:Objectives: This work aims to explore the feasibility of chlorhexidine-encapsulated mesoporous silica (CHX@pMSN) as a modifier of a commercial dental adhesive via the evaluation of physicochemical properties and antibacterial capabilities of adhesive-dentin interface. Methods: Therapeutic adhesives were developed in the present study by incorporating CHX@pMSN into a commercial adhesive at four mass fractions (0, 1, 5 and 10 wt.%). The antibacterial capability on Streptococcus mutans (S. mutans) biofilm, conversion degree, adhesive morphology, microtensile bond strength (MTBS) and nanoleakage expression were evaluated comprehensively. Results: MTT and CLSM evaluation showed that CHX@pMSN-doped adhesive inhibits S. mutans biofilm growth, while CHX is released from the modified adhesive continuously. The incorporation of CHX@pMSN did not affect immediate bond strength at the concentration of 1% and 5% (P > 0.05). Moreover, these bonds were mainly preserved in 5% CHX@pMSN group after one month of collagenase ageing. Meanwhile, CHX@pMSN-doped adhesive groups exhibited similar nanoleakage distribution compared with the control. Conclusion: This study showed that the 5% CHX@pMSN-modified adhesive achieved balance amongst unaffected immediate bonding strength, well-preserved bonds against collagenase ageing and effective inhibition of S. mutans biofilm growth. Clinical significance: CHX@pMSN-modified dentin adhesive can potentially extend the service life of adhesive restoration in clinic.
