Abstract:AbstractThe outbreak of coronavirus disease 2019 (COVID-19) has caused more than 80 813 confirmed cases in all provinces of China, and 21 110 cases reported in 93 countries of six continents as of 7 March 2020 since middle December 2019. Due to biological nature of the novel coronavirus, named severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) with faster spreading and unknown transmission pattern, it makes us in a difficulty position to contain the disease transmission globally. To date, we have found it is one of the greatest challenges to human beings in fighting against COVID-19 in the history, because SARS-CoV-2 is different from SARS-CoV and MERS-CoV in terms of biological features and transmissibility, and also found the containment strategies including the non-pharmaceutical public health measures implemented in China are effective and successful. In order to prevent a potential pandemic-level outbreak of COVID-19, we, as a community of shared future for mankind, recommend for all international leaders to support preparedness in low and middle income countries especially, take strong global interventions by using old approaches or new tools, mobilize global resources to equip hospital facilities and supplies to protect noisome infections and to provide personal protective tools such as facemask to general population, and quickly initiate research projects on drug and vaccine development. We also recommend for the international community to develop better coordination, cooperation, and strong solidarity in the joint efforts of fighting against COVID-19 spreading recommended by the joint mission report of the WHO-China experts, against violating the International Health Regulation (WHO, 2005), and against stigmatization, in order to eventually win the battle against our common enemy — COVID-19.

Abstract:Recently emerged human‐to‐human transmission of COVID‐19, a novel lethal strain (Chan et al., 2020) of coronavirus, caused a global pandemic burden, affecting hundreds of thousands of individuals, having life‐threatening outcomes, not only in medically compromised persons but also in perfectly healthy young individuals with immunocompetent system. Apparently, this specific coronavirus must possess special abilities to spread and compromise the immune mechanisms in humans. Unfortunately, there are limited evidence‐based data available, as, which is understandable clinicians and scientists must focus on life‐saving aspects (Prompetchara et al., 2020). At the same time, the question arises, whether this strain possesses unique abilities to penetrate via oropharyngeal epithelial barriers and, which is even more intriguing, why infection of SARS‐CoV‐2 triggers such abnormal 'cytokine storm' and immunosuppression? For unknown reasons, SARS‐CoV‐2 does not seem to be effectively defeated by human first line protective mechanisms, neither biomolecular, nor cellular, as number of death cases in middle aged, healthy persons were reported, regardless their immune status (Mehta et al., 2020).

Abstract:COVID-19 caused by SARS-CoV-2 has created formidable damage to public health and market economy. Currently, SARS-CoV-2 variants has exacerbated the transmission from person-to-person. Even after a great deal of investigation on COVID-19, SARS-CoV-2 is still rampaging globally, emphasizing the urgent need to reformulate effective prevention and treatment strategies. Here, we review the latest research progress of COVID-19 and provide distinct perspectives on the mechanism and management of COVID-19. Specially, we highlight the significance of Human Identical Sequences (HIS), hyaluronan, and hymecromone (“Three-H”) for the understanding and intervention of COVID-19. Firstly, HIS activate inflammation-related genes to influence COVID-19 progress through NamiRNA-Enhancer network. Accumulation of hyaluronan induced by HIS-mediated HAS2 upregulation is a substantial basis for clinical manifestations of COVID-19, especially in lymphocytopenia and pulmonary ground-glass opacity. Secondly, detection of plasma hyaluronan can be effective for evaluating the progression and severity of COVID-19. Thirdly, spike glycoprotein of SARS-CoV-2 may bind to hyaluronan and further serve as an allergen to stimulate allergic reaction, causing sudden adverse effects after vaccination or the aggravation of COVID-19. Finally, antisense oligonucleotides of HIS or inhibitors of hyaluronan synthesis (hymecromone) or antiallergic agents could be promising therapeutic agents for COVID-19. Collectively, Three-H could hold the key to understand the pathogenic mechanism and create effective therapeutic strategies for COVID-19.

Abstract:As we enter 2022, we are faced with the fact that coronavirus disease 2019 (COVID-19) has been devastating the world for more than 2 full years. Since the original outbreak of COVID-19 in December 2019, the disease has proved to be a major threat to global health and, perhaps more importantly, one that is not likely to be resolved soon. Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the virus that causes COVID-19, is easily transmissible and highly contagious. [1,2] When the virus first entered the human body and subsequently spread among people, an important biological phenomenon occurred—adaptation to a new host. SARS-CoV-2 has undergone frequent mutations, producing multiple variants of consequence to human health. [3] Rapidly emerging variants such as Omicron have shown strong immune evasiveness. Coupled with the substantial amount of global travel nowadays, ongoing variant emergence makes COVID-19 extremely difficult to contain and even more difficult to eliminate or eradicate. Furthermore, COVID-19 poses a major threat to the global economy. [4] Needless to say, there is a high degree of uncertainty associated with COVID-19 and the future. [5,6]

Abstract:Coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), poses an imminent threat to our lives. Although animal models and monolayer cell cultures are utilized for pathogenesis studies and the development of COVID-19 therapeutics, models that can more accurately reflect human-relevant responses to this novel virus are still lacking. Stem cell organoids and bioengineered organs-on-chips have emerged as two cutting-edge technologies used to construct biomimetic in vitro three-dimensional (3D) tissue or organ models. In this review, the key features of these two model systems that allow them to recapitulate organ physiology and function are introduced. The recent progress of these technologies for virology research is summarized and their utility in meeting the COVID-19 pandemic is highlighted. Future opportunities and challenges in the development of advanced human organ models and their potential to accelerate translational applications to provide vaccines and therapies for COVID-19 and other emerging epidemics are also discussed.

Abstract:Emerging infectious diseases have accompanied the development of human society while causing great harm to humans, and SARS-CoV-2 was only one in the long list of microbial threats. Many viruses have existed in their natural reservoirs for a very long time, and the spillover of viruses from natural hosts to humans via interspecies transmission serves as the main source of emerging infectious diseases. Widely existing viruses capable of utilizing human receptors to infect human cells in animals signal the possible outbreak of another viral infection in the near future. Extensive and close collaborative surveillance across nations, more effective wildlife trade legislation, and robust investment into applied and basic research will help to combat the possible pandemics of new emerging infectious diseases in the future.

Abstract:An outbreak of the COVID-19 is currently underway across China. The government has allocated a lot of staff and material resources to control the spread of this epidemic. However, at the meaning time, excessive prevention involving the use of massive disinfectants is happening at places far away from the core outbreak area, and the induced secondary problems must not be ignored. Based on new epidemic data issued on February 14, 2020 from the National Health Commission,[1] the mortality rate of patients with COVID-19 infection is about 2.29% in Wuhan city, the capital of Hubei Province and core outbreak area holding almost 56% of the infected population in China. This rate is lower than the 9.6% observed for severe acute respiratory syndrome (SARS)[2] and 35% for Middle East respiratory syndrome (MERS).[3] Outside Hubei Province, although there are nearly 1000 patients with new coronary pneumonia in several areas, such as Guangdong, Henan and Zhejiang provinces, the mortality rate for these patients is less than 1%, or even close to zero.[4] Chinese health authorities and researchers have identified the disease pathogen and its main route of transmission,[5] but have not yet finalized the intermediate host of COVID-19. Evidence shows that the disease spreads quickly, especially in agglomeration. During the outbreak, cases of family clusters were often reported in China. In addition, cases aboard the Diamond Princess, a cruise ship that was quarantined by the Japanese Health, Labor, and Welfare Ministry for 14 days, have drawn great international attention. As of February 17, 2020, a total of 454 passengers and crew onboard had been diagnosed with COVID-19 infection,[6] reflecting the virus's high spread efficiency in this agglomeration situation. From observations of currently treated cases, the condition is relatively mild, recovery is faster, and the prognosis is better in children compared with adults.[7] Severe cases are rare among confirmed cases in children, possibly because children are less likely to enter crowded areas than adults. Additionally, during the epidemic, parents tend to pay attention to the protection of children and bring them to the hospital during the early stage of symptoms, thus contributing to a low proportion of severe cases among children.[8] To control the disease and treat patients more effectively, mobile cabin hospitals have been built in Wuhan city. However, public concerns about the risk of cross-infection in these hospitals still remain. The good news is that the virus was clinically observed to be self-limiting, which means the new coronavirus may not require intensive antiviral treatment, and some patients can effectively eliminate the virus through their strong immunity. However, self-healing does not mean treatment is not needed.[9] In Chongqing city, approximately 800 km from Wuhan city, everyone must wear a mask even when entering no-man's land, everyday disinfectant is sprayed profusely, and individuals are sprayed with a special disinfection device similar to a carwash when returning a housing estate. The main component used in disinfectants is sodium hypochlorite, which is currently at risk of being abused to control the new virus COVID-19. Sodium hypochlorite is an environmental hazard and corrosive substance. Diverse health effects occur after acute or chronic exposure to sodium hypochlorite. Specifically, dermal/ocular exposure to sodium hypochlorite can cause irritation and damage to skin or eyes, which may become apparent over time; the inhalation of aerosols can cause coughing, sore throat, headache, and nausea.[10] Moreover, sodium hypochlorite is poisonous for aquatic organisms and can persistently pollute the environment by forming organic chlorine compounds, such as absorbable organic halogen.[11] Thus, massive disinfection is not good for the human body or environment, and can even cause long-term damage over time. At present, an effective vaccine for COVID-19 infection is not available yet. As such, the ultimate epidemic prevention relies on individuals’ immunity. Positive moods are often associated with a decreased risk of inflammation through changes of immune response functions.[12] Therefore, in the face of this current epidemic, it is better for those far away from the core outbreak area to improve body immunity by maintaining optimism and using appropriate preventive measures according to different situation. Abstract:The coronavirus disease 2019 (COVID-19) pandemic underscores the critical need to integrate immunomics within the One Health framework to effectively address zoonotic diseases across humans, animals, and environments. Employing advanced high-throughput technologies, this interdisciplinary approach reveals the complex immunological interactions among these systems, enhancing our understanding of immune responses and yielding vital insights into the mechanisms that influence viral spread and host susceptibility. Significant advancements in immunomics have accelerated vaccine development, improved viral mutation tracking, and broadened our comprehension of immune pathways in zoonotic transmissions. This review highlights the role of animals, not merely as carriers or reservoirs, but as essential elements of ecological networks that profoundly influence viral epidemiology. Furthermore, we explore how environmental factors shape immune response patterns across species, influencing viral persistence and spillover risks. Moreover, case studies demonstrating the integration of immunogenomic data within the One Health framework for COVID-19 are discussed, outlining its implications for future research. However, linking humans, animals, and the environment through immunogenomics remains challenging, including the complex management of vast amounts of data and issues of scalability. Despite challenges, integrating immunomics data within the One Health framework significantly enhances our strategies and responses to zoonotic diseases and pandemic threats, marking a crucial direction for future public health breakthroughs.

Abstract:Coronavirus disease 2019 (COVID-19) is currently 2019 novel coronavirus (2019-nCoV), but most of them causing great damage to the health of people worldwide. are of poor quality.” At a press conference on May 31, 2020, Tedros Adhanom Ghebreyesus, the Director-General of the World Health Organization, announced that there have been more than six million cases of COVID-19 worldwide. A study by the United Nations Department of Economic and Social Affairs reported that the global economy could contract by almost 1% in 2020 because of the COVID-19 pandemic. In the face of this unprecedented crisis, everyone, including scientists and the general public, is eager to receive increasingly detailed scientific information about COVID-19. To share the latest research information promptly, both traditional journals and non-peer-reviewed preprint servers have been working tirelessly to release the latest research on this disease and provide support and decision-making information to clinicians and public health officials.

Abstract:AbstractCoronavirus disease 2019 (COVID‐19) continues to threaten human health, economic development, and national security. Although many vaccines and drugs have been explored to fight against the major pandemic, their efficacy and safety still need to be improved. Cell‐based biomaterials, especially living cells, extracellular vesicles, and cell membranes, offer great potential in preventing and treating COVID‐19 owing to their versatility and unique biological functions. In this review, the characteristics and functions of cell‐based biomaterials and their biological applications in COVID‐19 prevention and therapy are described. First the pathological features of COVID‐19 are summarized, providing enlightenment on how to fight against COVID‐19. Next, the classification, organization structure, characteristics, and functions of cell‐based biomaterials are focused on. Finally, the progress of cell‐based biomaterials in overcoming COVID‐19 in different aspects, including the prevention of viral infection, inhibition of viral proliferation, anti‐inflammation, tissue repair, and alleviation of lymphopenia are comprehensively described. At the end of this review, a look forward to the challenges of this aspect is presented.

Abstract:Since the outbreak of COVID-19, SARS-CoV-2 has continued to evolve. As the fifth variants of concern (VOCs) announced by the World Health Organization, Omicron has significantly changed from previous VOCs in terms of genome, biological and epidemiological characteristics. Although the severe illness and mortality caused by Omicron infection are significantly lower than those caused by the previous VOCs, Omicron's strong transmission ability has continued to break through the epidemic prevention barrier and human immunity barrier established by prior infections and vaccines. Omicron has opened a new chapter in the COVID-19 pandemic. In this paper, we review the changes in genome and biological characteristics of Omicron and its subspecies as well as the differences in epidemiological characteristics between Omicron and influenza viruses in order to provide scientific basis for the optimization of epidemic prevention and control.

Abstract:Since the outbreak of the COVID-19 pandemic in early December 2019, 81 174 confirmed cases and 3242 deaths have been reported in China as of March 19, 2020. The Chinese people and government have contributed huge efforts to combat this disease, resulting in significant improvement of the situation, with 58 new cases (34 were imported cases) and 11 new deaths reported on March 19, 2020. However, as of March 19, 2020, the COVID-19 pandemic continues to develop in 167 countries/territories outside of China, and 128 665 confirmed cases and 5536 deaths have been reported, with 16 498 new cases and 817 new deaths occurring in last 24 hours. Therefore, the world should work together to fight against this pandemic. Here, we review the recent advances in COVID-19, including the insights in the virus, the responses of the host cells, the cytokine release syndrome, and the therapeutic approaches to inhibit the virus and alleviate the cytokine storm. By sharing knowledge and deepening our understanding of the virus and the disease pathogenesis, we believe that the community can efficiently develop effective vaccines and drugs, and the mankind will eventually win this battle against this pandemic.

Abstract:Humanity has experienced four major pandemics since the twentieth century, with the 1918 Spanish flu, the 2002 severe acute respiratory syndrome (SARS), the 2009 swine flu, and the 2019 coronavirus disease (COVID)-19 pandemics having the most important impact in human health. The 1918 Spanish flu caused unprecedented catastrophes in the recorded human history, with an estimated death toll between 50 – 100 million. While the 2002 SARS and 2009 swine flu pandemics caused approximately 780 and 280,000 deaths, respectively, the current COVID-19 pandemic has resulted in > 6 million deaths globally at the time of writing. COVID-19, instigated by the SARS – coronavirus-2 (SARS-CoV-2), causes unprecedented challenges in all facets of our lives, and never before brought scientists of all fields together to focus on this singular topic. While for the past 50 years research have been heavily focused on viruses themselves, we now understand that the host immune responses are just as important in determining the pathogenesis and outcomes of infection. Research in innate immune mechanisms is crucial in understanding all aspects of host antiviral programmes and the mechanisms underpinning virus-host interactions, which can be translated to the development of effective therapeutic avenues. This review summarizes what is known and what remains to be explored in the innate immune responses to influenza viruses and SARS-CoVs, and virus-host interactions in driving disease pathogenesis. This hopefully will encourage discussions and research on the unanswered questions, new paradigms, and antiviral strategies against these emerging infectious pathogens before the next pandemic occurs.

Abstract:The COVID-19 pandemic is one of those global challenges that transcends territorial, political, ideological, religious, cultural, and certainly academic boundaries. Public health and healthcare workers are at the frontline, working to contain and to mitigate the spread of this disease. Although intervening biological and immunological responses against viral infection may seem far from the physical sciences and engineering that typically work with inanimate objects, there actually is much that can—and should—be done to help in this global crisis. In this Perspective, we convert the basics of infectious respiratory diseases and viruses into physical sciences and engineering intuitions, and through this exercise, we present examples of questions, hypotheses, and research needs identified based on clinicians' experiences. We hope researchers in the physical sciences and engineering will proactively study these challenges, develop new hypotheses, define new research areas, and work with biological researchers, healthcare, and public health professionals to create user-centered solutions and to inform the general public, so that we can better address the many challenges associated with the transmission and spread of infectious respiratory diseases.

Abstract:The COVID-19 pandemic is an ongoing global health threat, yet our understanding of the dynamics of early cellular responses to this disease remains limited 1 . Here in our SARS-CoV-2 human challenge study, we used single-cell multi-omics profiling of nasopharyngeal swabs and blood to temporally resolve abortive, transient and sustained infections in seronegative individuals challenged with pre-Alpha SARS-CoV-2. Our analyses revealed rapid changes in cell-type proportions and dozens of highly dynamic cellular response states in epithelial and immune cells associated with specific time points and infection status. We observed that the interferon response in blood preceded the nasopharyngeal response. Moreover, nasopharyngeal immune infiltration occurred early in samples from individuals with only transient infection and later in samples from individuals with sustained infection. High expression of HLA-DQA2 before inoculation was associated with preventing sustained infection. Ciliated cells showed multiple immune responses and were most permissive for viral replication, whereas nasopharyngeal T cells and macrophages were infected non-productively. We resolved 54 T cell states, including acutely activated T cells that clonally expanded while carrying convergent SARS-CoV-2 motifs. Our new computational pipeline Cell2TCR identifies activated antigen-responding T cells based on a gene expression signature and clusters these into clonotype groups and motifs. Overall, our detailed time series data can serve as a Rosetta stone for epithelial and immune cell responses and reveals early dynamic responses associated with protection against infection.

Abstract:Abstract: The coronavirus outbreak (COVID-19, SARS-CoV-2) greatly impacts the world. Despite great biomedical efforts, approximately 30-50% global population was at least once infected with COVID. Human life expectancy is reduced by COVID-19 epidemics worldwide. It is indispensable to prevent and treat COVID-19 as effectively as possible. In order to well prevent and treat the coronavirus infection, clinical diagnostic or therapeutic paradigms should be updated. Since the outside condition of COVID-19 prevention is imbalanced among regions and countries, global prevention and treatment action should be aimed. Despite global vaccination for COVID, a significant proportion of humans still constantly exposes and infected with viruses. Advanced viral biological knowledge and vaccine techniques can alleviate viral spread and promote therapeutics. Universal and specific preventive and treatment paradigms should be designed, integrated, and introduced. The origin and hidden nature of COVID-19 biology and pathogenesis are broadly understood now. Viral vaccines, pathogenesis, diagnosis, treatment, and personalized medicine are progressing rapidly. Many viral vaccines and personalized medicine should be especially emphasized. To speed up global efforts against COVID-19, new knowledge and breakthroughs of viral transmissibility, vaccine technique innovation, diagnostic widening, and therapeutic variability worldwide are discussed. Different strategic platforms and landscapes can reach different outcomes. By facilitating global machinery against different variants of COVID, viral-induced socioeconomic burden and imbalance could be reduced.
