Abstract:Melatonin, a tryptophan-derived molecule, is endogenously generated in animal, plant, fungal and prokaryotic cells. Given its antioxidant properties, it is involved in a myriad of signaling functions associated with various aspects of plant growth and development. In higher plants, melatonin (Mel) interacts with plant regulators such as phytohormones, as well as reactive oxygen and nitrogen species including hydrogen peroxide (H2O2), nitric oxide (NO) and hydrogen sulfide (H2S). It shows great potential as a biotechnological tool to alleviate biotic and abiotic stress, to delay senescence and to conserve the sensory and nutritional quality of postharvest horticultural products which are of considerable economic importance worldwide. This review provides a comprehensive overview of the biochemistry of Mel, whose endogenous induction and exogenous application can play an important biotechnological role in enhancing the marketability and hence earnings from postharvest horticultural crops.

Abstract:Melatonin (MEL), which is also known as N-acetyl-5-methoxytryptamine, is an indoleamine that is widely distributed and synthesized from tryptophan. It serves as a fundamental regulator of various physiological processes in plants themselves. These processes include the modulation od cardiac rhythms, immunity, and antioxidation. Additionally, it is a versatile molecule in plants that has been extensively researched for its involvement in stress tolerance, biosynthesis, degradation, and physiological functions. This article summarizes the latest scientific discoveries regarding the effects of melatonin on flowering, fruit maturation, root development, seed germination, and senescence in plants. The article also highlights melatonin multiple pathways of action in alleviating the adverse effects of biotic stresses, including the regulation of the redox state, gene expression and hormone networks. Furthermore, melatonin has been found to alter the contents of secondary metabolites, such as alkaloids, terpenoids, and polyphenols, and to modulate post-harvest fruit ripening by controlling the expression of genes associated with ethylene. Recent finding suggests that melatonin is a crucial player in the plant's reaction to abiotic stress, working in conjunction with various phytohormones, nitric oxide (NO) and reactive oxygen species (ROS). There has been extensive research on the capacity of melatonin to alleviate the impact of different abiotic stresses, such as drought, extreme temperature, salt, and heavy metal stress. Considering its multifaceted functions in in controlling and regulating plant growth and development, we suggest that melatonin is a master regulator in fruit plants.

Abstract:Precise floral transition from vegetative growth phase to reproductive growth phase is very important in plant life cycle. In flowering genetic pathways, DELLA proteins are master transcriptional regulators of gibberelic acid (GA) pathway, and FLOWERING LOCUS C (FLC) is a core repressor of vernalization pathway as well as downstream of DELLAs. As a crucial messenger in plants, the possible involvement of melatonin (N-acetyl-5-methoxytryptamine) in flowering and underlying molecular mechanism are unknown in Arabidopsis. In this study, we found that exogenous melatonin treatment delayed floral transition in Arabidopsis. Exogenous melatonin treatment conferred protein stabilizations of DELLAs [REPRESSOR of ga1-3 (RGA) and RGA-LIKE3 (RGL3)], without regulating the transcripts of DELLAs and endogenous GA level. Notably, exogenous melatonin delayed plant flowering and DELLA-activated transcripts of FLC were alleviated in della mutants, and those were exacerbated in DELLA overexpressing plants. Taken together, this study provides direct link between melatonin and floral transition, and indicates the novel involvement of DELLAs-activated FLC in melatonin-mediated flowering in Arabidopsis.

Abstract:Melatonin functions as a negative regulatory signal of seed oil and anthocyanin accumulation during the maturation of A. thaliana seeds. Previous studies have clearly demonstrated that the putative phytohormone melatonin functions directly in many aspects of plant growth and development. In Arabidopsis (Arabidopsis thaliana), the role of melatonin in seed oil and anthocyanin accumulation, and corresponding underlying mechanisms, remain unclear. Here, we found that serotonin N-acetyltransferase1 (SNAT1) and caffeic acid O-methyltransferase (COMT) genes were ubiquitously and highly expressed and essential for melatonin biosynthesis in Arabidopsis developing seeds. We demonstrated that blocking endogenous melatonin biosynthesis by knocking out SNAT1 and/or COMT significantly increased oil and anthocyanin content of mature seeds. In contrast, enhancement of melatonin signaling by exogenous application of melatonin led to a significant decrease in levels of seed oil and anthocyanins. Further gene expression analysis through RNA sequencing and reverse-transcription quantitative PCR demonstrated that the expression of a series of important genes involved in fatty acid and anthocyanin accumulation was significantly altered in snat1-1 comt-1 developing seeds during seed maturation. We also discovered that SNAT1 and COMT significantly regulated the accumulation of both mucilage and proanthocyanidins in mature seeds. These results not only help us understand the function of melatonin and provide valuable insights into the complicated regulatory network controlling oil and anthocyanin accumulation in seeds, but also divulge promising gene targets for improvement of both oil and flavonoids in seeds of oil-producing crops and plants.
Melatonin as an inhibitor of sweet cherries ripening in orchard trees

Abstract:Although melatonin effects on postharvest fruit ripening have been studied in some detail, information is still scarce during pre-harvest. Here, we examined whether or not melatonin may exert a regulatory role during sweet cherries ripening in orchard trees. We evaluated (i) the endogenous variations in melatonin contents, in comparison to those of well-known phytohormones such as ABA, salicylic acid and jsamonic acid, by ultrahigh performance liquid chromatography coupled to tandem mass spectrometry (UHPLC-MS/MS) during fruit ripening over two consecutive years, and (ii) to what extent melatonin treatments at low and high concentrations (at 10-4 M and 10-5 M, respectively) influence fruit ripening on the tree. Endogenous melatonin contents decreased in parallel to those of salicylic acid and jasmonic acid, while ABA contents increased as fruit ripening progressed, thus suggesting an inhibitory role for melatonin in fruit ripening. Furthermore, melatonin treatment at 10-5 M, which transiently increased endogenous melatonin contents at physiological concentrations, delayed anthocyanin accumulation, thus confirming an inhibitory regulatory role for melatonin in fruit ripening. We also found that the endogenous contents of cytokinins, but not those of ABA were transiently affected by melatonin treatment at 10-5 M. It is concluded that melatonin may delay sweet cherries ripening in orchard trees, probably exerting a modulatory role through a hormonal cross-talk. These results have important implications for the use of melatonin in the control of the timing of sweet cherries ripening in orchard trees.

Abstract:Seed germination, an important developmental stage in the life cycle of seed plants, is regulated by complex signals. Melatonin is a signaling molecule associated with seed germination under stressful conditions, although the underlying regulatory mechanisms are largely unknown. In this study, we showed that a low concentration (10 µM or 100 µM) of melatonin had no effect on seed germination, but when the concentration of melatonin increased to 500 µM or 1000 µM, seed germination was significantly inhibited in Arabidopsis. RNA sequencing analysis showed that melatonin regulated seed germination correlated to phytohormones abscisic acid (ABA), gibberellin (GA), and auxin. Further investigation revealed that ABA and melatonin synergistically inhibited seed germination, while GA and auxin antagonized the inhibitory effect of seed germination by melatonin. Disruption of the melatonin biosynthesis enzyme gene serotonin N‐acetyltransferase (SNAT), or N‐acetylserotonin methyltransferase (ASMT) promoted seed germination, while overexpression of ASMT inhibited seed germination. Taken together, our study sheds new light on the function and mechanism of melatonin in modulating seed germination in Arabidopsis.

Abstract:Melatonin is a naturally occurring chemical with pleiotropic effects in various species. In plants, melatonin is associated with a variety of plant physiological processes, including plant growth and development, stress responses, etc. Thus, melatonin may hold promise for improving crop yields and agricultural sustainability. This review describes the biosynthetic mode of melatonin and its properties and summarizes its functions in growth, development, and reproduction. In addition, the role of melatonin in plants facing various stressful environments is elaborated upon, and its relationship with other phytohormones is summarized. Through this review, we recognize the problems and challenges facing melatonin research and propose some feasible solutions.

Abstract:Ripening as a physico-chemical change is part of a continuous developmental process and hormones play a major role in this processes. The present study was carried out to investigate the effect of external melatonin (0 and 10 μM) injection at the light green stage on the ripening of strawberry fruit. The fruit was sampled for morphological, biochemical, and gene expression analysis during (0, 5, 10, and 15 days after treatment). The results showed a lower accumulation of anthocyanin content was observed in fruits treated with 10 μM. The injection of 10 μM melatonin caused a lower total soluble solid content and fruit color, and higher titratable acidity and softening. The total phenol content was higher in fruit treated with 10 μM melatonin, accompanied by increased PAL enzyme activity and gene expression, increased DPPH scavenging capacity, and higher content of quercetin, gallic, caffeic, and chlorogenic acids. The delay in fruit ripening was associated with suppression of H 2 O 2 level and endogenous ABA accumulation caused by lower expression of NCEDs genes. In general, it is concluded that activating the melatonin ROS scavenging cascade might be responsible for the delayed ripening and development of strawberry fruit. Therefore, our study demonstrates that the exogenous application of 10 μM melatonin can slow the ripening of strawberry fruit.

Melatonin promotes ripening and improves quality of tomato fruit during postharvest life

Abstract:In this study, the effect of melatonin on the postharvest ripening and quality improvement of tomato fruit was carried out. The tomatoes were immersed in exogenous melatonin for 2h, and then the related physiological indicators and the expression of genes during post-harvest life were evaluated. Compared with control check (CK), the 50 µM melatonin treatment significantly increased lycopene levels by 5.8-fold. Meanwhile, the key genes involved in fruit colour development, including phytoene synthase1 (PSY1) and carotenoid isomerase (CRTISO), showed a 2-fold increase in expression levels. The rate of water loss from tomato fruit also increased 8.3%, and the expression of aquaporin genes, such as SlPIP12Q, SlPIPQ, SlPIP21Q, and SlPIP22, was up-regulated 2- to 3-fold under 50 µM melatonin treatment. In addition, 50 µM melatonin treatment enhanced fruit softening, increased water-soluble pectin by 22.5%, and decreased protopectin by 19.5%. The expression of the cell wall modifying proteins polygalacturonase (PG), pectin esterase1 (PE1), β-galactosidase (TBG4), and expansin1 (Exp1) was up-regulated under 50 µM melatonin treatment. Melatonin increased ethylene production by 27.1%, accelerated the climacteric phase, and influenced the ethylene signalling pathway. Alteration of ethylene production correlated with altered 1-aminocyclopropane-1-carboxylic acid (ACC) synthase (ACS4) expression. The expression of ethylene signal transduction-related genes such as NR, SlETR4, SlEIL1, SlEIL3, and SlERF2, was enhanced by 50 µM melatonin. The effect of melatonin on ethylene biosynthesis, ethylene perception, and ethylene signalling may contribute to fruit ripening and quality improvement in tomato. This research may promote the application of melatonin on postharvest ripening and quality improvement of tomato fruit as well as other horticultural productions in the future.

Abstract:Melatonin, a trace efficient natural plant signaling molecule, has been widely reported in plant growth and development, abiotic stress, and postharvest preservation and other aspects. In recent years, melatonin has been found to play a positive role in inducing disease resistance in fruits. This paper describes the effect of melatonin on pathogens in vitro and in infected fruits, with a focus on the mechanism by which melatonin induces fruit resistance to invasive diseases. Furthermore, this paper reveals that melatonin and plant signal transduction pathways constitute a regulation network to mediate disease resistance responses in fruits. We conclude this paper with an outlook on future research directions in order to provide a theoretical reference for further research on the mechanism by which melatonin induces disease resistance.

Abstract:BACKGROUND: Plant melatonin appears to be a multi-regulatory molecule, similar to those observed in animals, with many specific functions in plant physiology. In recent years, the number of studies on melatonin in plants has increased significantly. One of the most studied actions of melatonin in plants is its effect on biotic and abiotic stress, such as that produced by drought, extreme temperatures, salinity, chemical pollution and UV radiation, among others.SCOPE: This review looks at studies in which some aspects of the relationship between melatonin and the plant hormones auxin, cytokinin, gibberellins, abscisic acid, ethylene, jasmonic acid and salicylic acid are presented. The effects that some melatonin treatments have on endogenous plant hormone levels, their related genes (biosynthesis, catabolism, receptors and transcription factors) and the physiological actions induced by melatonin, mainly in stress conditions, are discussed.CONCLUSIONS: Melatonin is an important modulator of gene expression related to plant hormones, e.g. in auxin carrier proteins, as well as in metabolism of indole-3-acetic acid (IAA), gibberellins, cytokinins, abscisic acid and ethylene. Most of the studies performed have dealt with the auxin-like activity of melatonin which, in a similar way to IAA, is able to induce growth in shoots and roots and stimulate root generation, giving rise to new lateral and adventitious roots. Melatonin is also able to delay senescence, protecting photosynthetic systems and related sub-cellular structures and processes. Also, its role in fruit ripening and post-harvest processes as a gene regulator of ethylene-related factors is relevant. Another decisive aspect is its role in the pathogen-plant interaction. Melatonin appears to act as a key molecule in the plant immune response, together with other well-known molecules such as nitric oxide and hormones, such as jasmonic acid and salicylic acid. In this sense, the discovery of elevated levels of melatonin in endophytic organisms associated with plants has thrown light on a possible novel form of communication between beneficial endophytes and host plants via melatonin.

Abstract:Abstract Melatonin (N-acetyl-5-methoxytryptamine) is an indole molecule widely found in animals and plants. It is well known that melatonin improves plant resistance to various biotic and abiotic stresses due to its potent free radical scavenging ability while being able to modulate plant signaling and response pathways through mostly unknown mechanisms. In recent years, an increasing number of studies have shown that melatonin plays a crucial role in improving crop quality and yield by participating in the regulation of various aspects of plant growth and development. Here, we review the effects of melatonin on plant vegetative growth and reproductive development, and systematically summarize its molecular regulatory network. Moreover, the effective concentrations of exogenously applied melatonin in different crops or at different growth stages of the same crop are analysed. In addition, we compare endogenous phytomelatonin concentrations in various crops and different organs, and evaluate a potential function of phytomelatonin in plant circadian rhythms. The prospects of different approaches in regulating crop yield and quality through exogenous application of appropriate concentrations of melatonin, endogenous modification of phytomelatonin metabolism-related genes, and the use of nanomaterials and other technologies to improve melatonin utilization efficiency are also discussed.

Abstract:<p>Melatonin is a pleiotropic molecule with multiple functions in plants. Since the discovery of melatonin in plants, numerous studies have provided insight into the biosynthesis, catabolism, and physiological and biochemical functions of this important molecule. Here, we describe the biosynthesis of melatonin from tryptophan, as well as its various degradation pathways in plants. The identification of a putative melatonin receptor in plants has led to the hypothesis that melatonin is a hormone involved in regulating plant growth, aerial organ development, root morphology, and the floral transition. The universal antioxidant activity of melatonin and its role in preserving chlorophyll might explain its anti‐senescence capacity in aging leaves. An impressive amount of research has focused on the role of melatonin in modulating postharvest fruit ripening by regulating the expression of ethylene‐related genes. Recent evidence also indicated that melatonin functions in the plant's response to biotic stress, cooperating with other phytohormones and well‐known molecules such as reactive oxygen species and nitric oxide. Finally, great progress has been made towards understanding how melatonin alleviates the effects of various abiotic stresses, including salt, drought, extreme temperature, and heavy metal stress. Abstract:Melatonin is a pleiotropic molecule with many diverse actions in plants. It is considered primarily an antioxidant with important actions in the control of reactive oxygen and nitrogen species (ROS and RNS), among other free radicals, and harmful oxidative molecules present in plant cells. In addition, plant melatonin is involved in multiple physiological actions, such as growth, rooting, seed germination, photosynthesis, and protection against abiotic and/or biotic stressors. The recent identification of the first plant melatonin receptor opened the door to this regulatory molecule being considered a new plant hormone. However, due to the diversity of its actions, melatonin has also been proposed as a plant master regulator. Here, we discuss the most recent data in respect to both perspectives.

Abstract:Melatonin, a multifunctional molecule that is present in all living organisms studied, is synthesized in plant cells in several intercellular organelles including in the chloroplasts and in mitochondria. In plants, melatonin has a relevant role as a modulatory agent which improves their tolerance response to biotic and abiotic stress. The role of melatonin in stress conditions on the primary metabolism of plant carbohydrates is reviewed in the present work. Thus, the modulatory actions of melatonin on the various biosynthetic and degradation pathways involving simple carbohydrates (mono- and disaccharides), polymers (starch), and derivatives (polyalcohols) in plants are evaluated. The possible applications of the use of melatonin in crop improvement and postharvest products are examined.

Abstract:Melatonin acts as a crucial signaling molecule with multiple physiological functions in plant response to abiotic and biotic stresses. However, the impact and regulatory mechanism of melatonin on attenuating tomato fruit fungal decay are unclear. In this study, we investigated the potential roles of melatonin in modulating fruit resistance to Botrytis cinerea and explored related physiological and molecular mechanisms. The results revealed that disease resistance was strongly enhanced by melatonin treatment, and 50 μM was confirmed as the best concentration. Melatonin treatment increased the activities of defense-related enzymes and decreased hydrogen peroxide (H2O2) content with enhanced antioxidant enzyme activities. Moreover, we found that melatonin treatment increased methyl jasmonate (MeJA) content; up-regulated the expressions of SlLoxD, SlAOC, and SlPI II; and reduced the expressions of SlMYC2 and SlJAZ1. We postulated that melatonin played a positive role in tomato fruit resistance to Botrytis cinerea through regulating H2O2 level and JA signaling pathway.

Abstract:Melatonin is an antioxidant that is widely distributed in plants and animals. It is a conservative molecule. In early studies, scientists often used isolation and identification techniques to observe whether the endogenous melatonin cycle in plants was related to the external photoperiod, plant growth, and development cycles, including seed germination, plant rooting, and floral induction. With the development of isolation and identification technology, there is ample evidence that plants possess a variety of melatonin-synthesis pathways. The comprehensive application of molecular biology, genomics, and computational biology has also led to a comprehensive understanding of the physiological functions of plant melatonin. In this paper, we not only highlight the candidate genes from Arabidopsis thaliana and Oryza sativa that might be contributing to increasing plant endogenous melatonin but also elucidate and characterize the role of melatonin in plant growth and development in response to biotic and abiotic stresses.

Abstract:AbstractPhytomelatonin is a newly identified plant hormone, and its primary functions in plant growth and development remain relatively poorly appraised. Phytomelatonin is a master regulator of reactive oxygen species (ROS) signaling and acts as a darkness signal in circadian stomatal closure. Plants exhibit at least three interrelated patterns of interaction between phytomelatonin and ROS production. Exogenous melatonin can induce flavonoid biosynthesis, which might be required for maintenance of antioxidant capacity under stress, after harvest, and in leaf senescence conditions. However, several genetic studies have provided direct evidence that phytomelatonin plays a negative role in the biosynthesis of flavonoids under non-stress conditions. Phytomelatonin delays flowering time in both dicot and monocot plants, probably via its receptor PMTR1 and interactions with the gibberellin, strigolactone, and ROS signaling pathways. Furthermore, phytomelatonin signaling also functions in hypocotyl and shoot growth in skotomorphogenesis and ultraviolet B (UV-B) exposure; the G protein α-subunit (Arabidopsis GPA1 and rice RGA1) and constitutive photomorphogenic1 (COP1) are important signal components during this process. Taken together, these findings indicate that phytomelatonin acts as a darkness signal with important regulatory roles in circadian stomatal closure, flavonoid biosynthesis, flowering, and hypocotyl and shoot growth.
