Abstract:The utilization of plant extract, microbial metabolites, and phytochemicals for the synthesis of green nanoparticles has gained popularity in the fields of biotechnology and biomedical sciences due to its cost-effectiveness, environmentally benign nature, and significant pharmacological effects. The present investigation involved the production of copper oxide (CuO) nanoparticles (NPs) and copper oxide-zinc oxide (CuO-ZnO) nanocomposites (NCs) by a green synthesis approach, employing Parkia timoriana bark extract as a stabilizing and capping agent. The nanoparticles were analyzed using different characteristic methods such as UV–visible spectroscopy (UV–Vis), thermogravimetric analysis (TGA), atomic force microscopy (AFM), high-resolution transmission electron microscopy (HR-TEM), and X-ray photoelectron microscopy (XPS). In UV–visible studies, the 250–400-nm peaks indicate the biosynthesis of CuO NPs and CuO-ZnO NCs. FTIR results indicate stretching vibration of the Cu–O and Zn–O bonds at 775.38 cm −1 and 448.34 cm −1 . XRD analysis displayed the crystalline nature of the biosynthesized nanoparticle, and the size of CuO NPs and CuO-ZnO NCs were 20.2 nm and 23.4 nm, respectively. FE-SEM results indicate the crystalline structure and spherical shape of CuO NPs and CuO-ZnO NCs. Antioxidant and antidiabetic assays were performed to compare the biological activity of mono and nanocomposite. In silico molecular docking studies showed the compounds' therapeutic potential in the presence and absence of CuO, ZnO, and CuO-ZnO nanoparticles. The docking approach placed these drug-like compounds in the α-amylase binding pocket. CuO-ZnO NC at a 100 μg/ml concentration demonstrated higher antioxidant activity (69.07%) than CuO NP (58.94%). In the α-amylase inhibition experiment, CuO NP and CuO-ZnO NC had IC50 values of 62.43 and 57.57 μg/ml, respectively. The IC 50 value for CuO NP and CuO-ZnO NC in the α-glucosidase inhibition assay was 66.78 μg/ml and 57.51 μg/ml, respectively. Thus, CuO-ZnO NC possesses higher antioxidant and antidiabetic properties than other nanoparticles in this study.
