Abstract:Abstract Water disinfection is a critical step in water and wastewater treatment. The most widely used chlorination suffers from the formation of carcinogenic disinfection by-products (DBPs) while alternative methods (e.g., UV, O 3 , and membrane filtration) are limited by microbial regrowth, no residual disinfectant, and high operation cost. Here, a nanowire-enabled disinfection method, locally enhanced electric field treatment (LEEFT), is introduced with advantages of no chemical addition, no DBP formation, low energy consumption, and efficient microbial inactivation. Attributed to the lightning rod effect, the electric field near the tip area of the nanowires on the electrode is significantly enhanced to inactivate microbes, even though a small external voltage (usually < 5 V) is applied. In this review, after emphasizing the significance of water disinfection, the theory of the LEEFT is explained. Subsequently, the recent development of the LEEFT technology on electrode materials and device configurations are summarized. The disinfection performance is analyzed, with respect to the operating parameters, universality against different microorganisms, electrode durability, and energy consumption. The studies on the inactivation mechanisms during the LEEFT are also reviewed. Lastly, the challenges and future research of LEEFT disinfection are discussed.

Abstract:The promoting effect of electric fields on the growth and lipid production of microalga provides a new research direction for biorefining. However, researchers have rarely applied this technology to the treatment of actual wastewater. In this study, the dry weight and lipid content of microalgae increased by 47.45% and 28.28%, respectively, when optimized for light, growth phase and energization time. By optimizing the initial microalgal concentration and the ratio of nitrogen and phosphorus, the removal of total nitrogen (TN) and ammonia nitrogen (NH4+) increased by 18.5% and 10.9%, respectively. Compared to peroxidase (POD), significant changes in the relative activities of catalase (CAT) and total superoxide dismutase (T-SOD) were observed after the application of an electric field to the microalgae. Along with the cultivation, the relative activities of T-SOD and CAT showed a tendency of an increase and then a decrease compared with the control. This was mainly due to the increased internal secretion of T-SOD and CAT by the microalgae in response to the oxidative stress of the electric field on the microalgae, while the lack of nutrients at the later stages of cultivation also led to the increase of the content of the two enzymes in the control.

Abstract:Efficient drinking water disinfection methods are critical for public health. Locally enhanced electric field treatment (LEEFT) is an antimicrobial method that uses sharp structures, like metallic nanowires, to enhance the electric field at tips and cause bacteria inactivation. Electroporation is the originally designed mechanism of LEEFT. Although oxidation is typically undesired due to byproduct generation and electrode corrosion, it can enhance the overall disinfection efficiency. In this work, we conduct an operando investigation of LEEFT, in which we change the electrical parameters to tune the mechanisms between electrophysical electroporation and electrochemical oxidation. Pure electroporation (i.e., without detectable oxidation) could be achieved under a duty cycle of ≤0.1% and a pulse width of ≤2 μs. Applying 2 μs pulses at 7-8 kV/cm and 0.1% duty cycle results in 80-100% bacteria inactivation with pure electroporation. A higher chance of oxidation is found with a higher duty cycle and a longer pulse width, where the antimicrobial efficiency could also be enhanced. For water with a higher conductivity, a higher antimicrobial efficiency can be achieved under the same treatment conditions, and electrochemical reactions could be induced more easily. The findings shown in this work improve the fundamental understanding of LEEFT and help optimize the performance of LEEFT in real applications.

Abstract:Microalgae hold promise as a renewable energy source for the production of biofuel, as they can convert light energy into chemical energy through photosynthesis. However, cost-efficient harvest of microalgae remains a major challenge to commercial-scale algal biofuel production. We first investigated the potential of electrolytic water as a flocculant for harvesting Tetraselmis sp. Alkaline electrolyzed water (AEW) is produced at the cathode through water electrolysis. It contains mineral ions such as Na+, K+, Ca2+, and Mg2+ that can act as flocculants. The flocculation activity with AEW was evaluated via culture density, AEW concentration, medium pH, settling time, and ionic strength analyses. The flocculation efficiency was 88.7% at 20% AEW (pH 8, 10 min) with a biomass concentration of 2 g/l. The initial biomass concentration and medium pH had significant influences on the flocculation activity of AEW. A viability test of flocculated microalgal cells was conducted using Evans blue stain, and the cells appeared intact. Furthermore, the growth rate of Tetraselmis sp. in recycled flocculation medium was similar to the growth rate in fresh F/2 medium. Our results suggested that AEW flocculation could be a very useful and affordable methodology for fresh biomass harvesting with environmentally friendly easy operation in part of the algal biofuel production process.

Effective removing of rotifer contamination in microalgal lab-scale raceway ponds by light-induced phototaxis coupled with high-voltage pulse electroshock

Abstract:Rotifer reproduction control in open microalgae cultivation systems poses a significant challenge for large-scale industries. Conventional methods, such as electric, meshing, and chemical techniques, are often expensive, ineffective, and may have adverse environmental-health impacts. This study investigated a promising control technique through light-induced phototaxis to concentrate rotifers in a specific spot, where they were electroshocked by local-limited exposure dose. The results showed that the rotifers had the most pronounced positive and negative phototropism with phototaxis rates of 66.7 % and -78.8 %, respectively, at blue-light irradiation of 30 µmol∙m-2∙s-1 and red-light irradiation of 22.5 µmol∙m-2∙s-1 for 20 min. The most effective electroshock configuration employed 1200 V/cm for 15 min with a 1-second cycle time and a 10 % duty cycle, resulting in a 75.0 % rotifer removal rate without impacting microalgae growth. The combination of the two light beams could effectively lead rotifers to designated areas where they were electrocuted successfully.
Electro-Fenton Based Technique to Enhance Cell Harvest and Lipid Extraction from Microalgae

Abstract:Currently, lipid extraction remains a major bottleneck in microalgae technology for biofuel production. In this study, an effective and easily controlled cell wall disruption method based on electro-Fenton reaction was used to enhance lipid extraction from the wet biomass of Nannochloropsis oceanica IMET1. The results showed that 1.27 mM of hydroxide radical (HO•) was generated under the optimal conditions with 9.1 mM FeSO4 in a 16.4 mA·cm−2 current density for 37.0 min. After the electro-Fenton treatment, the neutral lipid extraction yield of microalgae (~155 mg) increased from 40% to 87.5%, equal to from 12.2% to 26.7% dry cell weight (DCW). In particular, the fatty acid composition remained stable. The cell wall disruption and lipid extraction processes were displayed by the transmission electron microscope (TEM) and fluorescence microscopy (FM) observations, respectively. Meanwhile, the removal efficiency of algal cells reached 85.2% within 2 h after the reaction was terminated. Furthermore, the biomass of the microalgae cultured in the electrolysis wastewater treated with fresh nutrients reached 3 g/L, which is 12-fold higher than that of the initial after 24 days. These finds provided an economic and efficient method for lipid extraction from wet microalgae, which could be easily controlled by current magnitude regulation.

Abstract:Microalgae show great potential for nutrient removal from piggery digestate. However, full-strength piggery digestate have been found to severely inhibit microalgal growth. In this study, microalgae were coupled into the electric field (EF)system to form an electric field-microalgae system (EFMS). The effects of EF characteristics and environmental conditions on the growth of Desmodesmus sp. CHX1 and the removal of nitrogen and phosphorus in EFMS were explored. The results indicated that the optimal EF parameters for forming a fine EFMS were electrode of Zn (anode)/graphite (cathode), electric frequency of three times per day (10 min/time) and voltage of 12 V. The suitable light intensity and microalgae inoculation concentration for the EFMS were 180 μmol photons/(m2·s) and 0.2 g/L, respectively. Ammonium nitrogen and total phosphorus removal efficiencies were 65.38% and 96.16% in the piggery digestate by EFMS under optimal conditions. These results indicate that EFMS is a promising technology for nutrient removal from piggery digestate.

Abstract:An innovative electro-Fenton enhanced membrane photobioreactor with satisfactory membrane fouling mitigation was constructed for microalgae harvesting. The porous carbon and carbon nanotubes hollow fiber membranes (PC-CHFMs) were used as the separation unit and cathode, simultaneously. H2O2 was generated by cathode reducing O2 in-situ, which would further produce •OH as the main oxidant by coupling H2O2 with Fe2+. The •OH could deeply remove the extracellular organic matter (EOM) deposited on the membrane surface or inside the pores. Experimental results showed that the permeate flux recovery rates of PC-CHFMs by electro-Fenton at the 18th, 29th and 41st day were 100%, 100% and 98.3%, respectively. The corresponding recovery rates by chemical cleaning at the same time were 99.8%, 81.7% and 54.4%. The stable and high permeate flux of PC-CHFMs made a great contribution to the microalgae harvesting efficiency, where the concentration factor could be 4.8 times higher than that of the control group. Filtrating superiority of PC-CHFMs was becoming more prominent with the extension of operating time. In addition, the removal efficiency of NH4+-N and TP in wastewater was approximately 100% at stable culture period.

Abstract:Lipid extraction is a key step of biodiesel production from microalgae, however, the application of traditional extraction methods was limited due to the difficulties of cell disruption as well as solvent toxicity. In this work, pretreatment method using pulsed electric field (PEF), was primarily applied to lipid extraction process from microalgae Chlorella pyrenoidosa grown in wastewater. After the pretreatment with PEF, the yields of fatty acid methyl esters from C.pyrenoidosa was 12.0% higher than traditional pretreatment with ultrasonic. The results indicated that PEF was an effective method for cell disruption. Fluorescence staining and scanning electron microscopy showed that the integrity of the cell membrane of microalgae was damaged under pulsed electric field, which enhanced the penetration of solvents and lipid extraction.

Abstract:Purpose of ReviewThe aim of this review is to summarize the inactivation and removal technologies used for algal-bloom control and to serve the development of theories and technologies for algal bloom prevention and control in the future.Recent FindingsThe efficiency of inactivation and removal of algal blooms largely depends on the density of bloom-forming microalgae faced when the control measures are adopted. Locally enhanced electric field treatment technology has shown the most potential for low-density algal-bloom control, which could effectively inactivate algal cells with low energy consumption. Magnetic capture technology has outstanding advantages for the separation and capture of high-density algae blooms owing to its high efficiency, easy separation, and low cost.SummaryThis article reviews the physical, chemical, physicochemical, biological, and ecological methods of algal-bloom prevention and control. The key to effective prevention of algal blooms is to select the appropriate method based on the difference in microalgae density. For example, under low-density stages, if the inactivation of algae cells can be realized, the occurrence and development of high-density algal blooms may be prevented, and under high-density stages, if the capture of algae cells can be done effectively, the subsequent environmental hazards brought by algal blooms will be reduced or even eliminated. This paper will help in choosing appropriate methods of algal-bloom prevention and control based on the difference in microalgae density and providing suggestions for future research on algal-bloom control.

Abstract:Microalgae harvesting remains a challenging step in microalgae industrialization, thereby provoking the necessity to explore sustainable and economically feasible approaches. This research investigated the use of magnetic flocculated nanoparticles in the harvesting of the common microalgae Chlorella pyrenoidosa and Scenedesmus obliquus. The results showed that magnetic flocculated nanoparticles efficiently adsorbed negatively charged microalgae cells, and a magnetic field could adsorb the magnetic flocculated nanoparticles, thereby harvesting the microalgae cells. Harvesting efficiency was remarkably increased at the optimum magnetic field strength of 0.5 T with the magnetic flocculated nanoparticles at 0.738 g/L, and microalgae broth at pH 9.0, whereas the recovery rates of both C. pyrenoidosa and S. obliquus were around 97% and the sedimentation speed of both was above 2.63 cm/min. This study exemplified the magnetic flocculated nanoparticles-based approach to effectively harvest the microalgae cells.
Low-frequency ultrasound assisted contact-electro-catalysis for efficient inactivation of Microcystis aeruginosa

Abstract:Frequent cyanobacterial blooms pose a serious threat to the aquatic ecosystem and human health, so developing an efficient algae removal method is a long-term goal for bloom management. Current technologies for algal bloom control need urgent improvement in terms of algicide recovery, eco-friendliness and cost. Here we propose a contact-electro-catalytic method, using polytetrafluoroethylene (PTFE) film as a reusable catalyst. This contact-electro-catalytic approach involves the generation of reactive oxygen species (e.g., O 2 • – , HO•, 1 O 2 and H 2 O 2 ) through water-PTFE contact electrification under the low-frequency ultrasonic waves, facilitating the inactivation of algae. The removal rate of the cyanobacterium Microcystis aeruginosa ( M. aeruginosa ) exposured to the water-PTFE contact-electro-catalytic system is almost five times greater than that of ultrasound alone after 5 h. A mechanistic investigation revealed that the contact-electro-catalytic system damaged the photosynthetic activity, antioxidant system and membrane integrity of the cells. Additionally, LC-MS metabolomic analysis indicated that this system caused substantial significant disruptions in the TCA cycle, amino acid metabolism, purine metabolism and phospholipid metabolism. Three-dimensional fluorescence spectroscopy suggested contact-electro-catalysis could further availably degrade the organic matter. We anticipate that this method can provide an eco-friendly, highly efficient and economic approach for effective control of harmful algal blooms.

Abstract:Livestock wastewater (LW) is characterized by the significant presence of organic matter, nitrogen (N), and phosphorus (P), as well as heavy metals (HMs), antibiotics, endocrine disruptors (EDCs), microplastics (MPs), and antibiotic resistance genes (ARGs). Microalgae-based technologies present a promising alternative for the bioremediation of LW, as they can efficiently eliminate pollutants while simultaneously utilizing renewable resources. However, microalgae-based LW treatment in remediating multiple contaminants of emerging concern (CECs) has been comprehensively evaluated by few articles. Consequently, the impact of inhibitory components in LW on microalgae and the response of microalgae to such exposure remain unclear. This review provides a critical and updated overview of the potential use of microalgae as LW bioremediation agents for the simultaneous removal of conventional pollutants and CECs. Several models and algorithms have been suggested as viable approaches for improving microalgal growth through the optimization of cultivation parameters. The effects of the inhibitory components in LW on microalgae and the removal mechanisms of hazards were comprehensively illustrated, highlighting the behavior of CECs (e.g., antibiotics, EDCs, MPs, and ARGs). Moreover, conventional and cutting-edge strategies for improving the performance of microalgae-based LW treatment, including pretreatment, optimization of the cultivation mode and light, genetic engineering, random mutagenesis, and microalgae-microbial fuel cells, were further discussed. Furthermore, perspectives for further improvement were proposed. Importantly, research should focus on verifying the specific removal mechanisms of CECs at the cellular level, distinguishing the combined effect of pollutants in LW, optimizing multi-objective microalgae-based LW treatment, and developing more sustainable and user-friendly strategies.

CO2 capture/biofixation and lipid extraction by membrane contactors for enhancing microalgal cultivation

Abstract:Recent soaring oil prices and diminishing world oil reserves, coupled with the increased emission of greenhouse gases and the predicted threat of climate change, have generated renewed interest in using algae as an alternative and renewable feedstock for energy production. Aimed at intensifying microalgal CO2 fixation and lipid production enhancement, membrane contactors have been applied to capture CO2 from air, enhance microalgal cultivation and extract lipid from algae. In order to capture CO2 from air, three kinds of membrane contactors with absorption agents, Carbonic Anhydrase (CA)-hydrogel and ionic liquid, were designed. The photo-bioreactor integrated with a hollow-fiber membrane module to supply CO2 has been designed for Chlorella vulgaris cultivation. Using a membrane contactor, the residence time of CO2 in the photo-bioreactor can be prolonged to enhance CO2 biofixation. The biofixation rate has reached 0.38 g/(L·h), which is more than 2-3 times those in conventional reactors. In order to increase lipid productivity, the effects of cultivation conditions, including KNO3-level, CO2 concentration and irradiance, on the cell growth, chlorophyll content and lipid accumulation of Chlorella vulgaris are systematically investigated. The lipid productivity has reached 40 mg/(L·d) under select cultivated conditions, which is more than 2.5-fold of that reported. Finally, to simplify the algal lipid production process and thus to reduce costs, microalgal lipid extracted in situ and continuously produced in the bioreactor integrated with membrane-based extraction has been investigated in this study.
Enhancing microalgal biomass production in lab-scale raceway ponds through innovative computational fluid dynamics-based electrode deflectors

Abstract:The design of novel electrode deflector structures (EDSs) introduced a promising strategy for enhancing raceway ponds performance, increasing carbon fixation, and improving microalgal biomass accumulation. The computational fluid dynamics, based flow field principles, proved that the potency of arc-shaped electrode deflector structures (A-EDS) and spiral electrode deflector structures (S-EDS) were optimal. These configurations yielded superior culture effects, notably reducing dead zones by 9.1% and 11.7%, while elevating biomass increments of 14.7% and 11.5% compared to the control, respectively. In comparison to scenarios without electrostatic field application, the A-EDS group demonstrated pronounced post-stimulation growth, exhibiting an additional biomass increase of 11.2%, coupled with a remarkable 23.6% surge in CO 2 fixation rate and mixing time reduction by 14.7%. A-EDS and S-EDS, combined with strategic electric field integration, provided a theoretical basis for promoting microalgal biomass production and enhancing carbon fixation in a raceway pond environment to similar production practices.

Improving electrocoagulation floatation for harvesting microalgae

Abstract:Electro-coagulation floatation (ECF) is a foam-floatation dewatering method that has been shown to be a highly effective, rapid, and scalable separation methodology. In this manuscript, an in-depth analysis of the gas and flocculant levels observed during the process is provided, with microbubbles observed in the 5-80 μm size range at a concentration of 102-103 bubbles mL-1. Electrolysis of microalgae culture was then observed, demonstrating both effective separation using aluminium electrodes (nine microalgal species tested, 1-40 μm size range, motile and non-motile, marine and freshwater), and sterilisation of culture through bleaching with inert titanium electrodes. Atomic force microscopy was used to visualise floc formation in the presence and absence of algae, showing nanoscale structures on the magnitude of 40-400 nm and entrapped microalgal cells. Improvements to aid industrial biotechnology processing were investigated: protein-doping was found to improve foam stability without inducing cell lysis, and an oxalate buffer wash regime was found to dissolve the flocculant whilst producing no observable difference in the final algal lipid or pigment profiles, leaving the cells viable at the end of the process. ECF separated microalgal culture had an algal biomass loading of 13% and as such was ideal for direct down-stream processing through hydrothermal liquefaction. High bio-crude yields were achieved, though this was reduced slightly on addition of the Al(OH)3 after ECF, with carbon being distributed away to the aqueous and solid residue phases. The amenability and compatibility of ECF to integration with, or replacement of, existing centrifugation and settling processes suggests this process may be of significant interest to the biotechnology industry.

Abstract:Pulsed electric field (PEF) processing has emerged as an alternative to thermal pasteurization for the shelf-life extension of heat-sensitive liquids at industrial scale. It offers the advantage of minimal alteration in physicochemical characteristics and functional properties. In this study, a pilot-scale continuous PEF processing (Toutlet < 55 °C) was applied to microalgae Chlorella vulgaris (Cv) suspensions (pH = 6.5), which was proposed as a functional ingredient for plant-based foods. Cv suspensions were inoculated with three distinct food spoilage microorganisms (Pseudomonas guariconensis, Enterobacter soli and Lactococcus lactis), isolated from the Cv biomass. PEF treatments were applied with varying electric field strength Eel of 16 to 28 kV/cm, pulse repetition rate f of 100 to 140 Hz, with a pulse width τ of 20 μs and an inlet product temperature Tin of 30 °C. The aim was to evaluate the PEF-induced microbial reduction and monitor the microbial outgrowth during a 10-day cold storage period (10 °C). Maximum inactivation of 4.1, 3.7 and 3.6 logs was achieved (28 kV/cm and 120 Hz) for the investigated isolates, respectively. Under these conditions, the critical electric field strengths Ecrit, above which inactivation was observed, ranged from 22.6 to 24.6 kV/cm. Moreover, repeated PEF treatment resulted in similar inactivation efficiency, indicating its potential to enhance shelf-life further.

Abstract:Electro-Fenton enhanced porous carbon (PC) - carbon nanotubes (CNT) hollow fiber membranes loaded with Fe2+ were firstly used to alleviate the membrane fouling caused by microalgal cells and extracellular organic matter (EOM) in microalgae harvesting. Electrical repulsion effect could reduce the deposition of oppositely charged algal cells and EOM on the membrane surface. The generation of •OH in-situ in electro-Fenton process could degrade EOM on the membrane surface and even inside the pores. Continuous mode results illustrated that the electro-Fenton enhanced microalgae harvesting process could significantly retard transmembrane pressure increasing and strengthen microalgae concentration capacity by 2.5 times at the optimally applied voltage (−1.0 V). The pure water permeance after operation was 16 times higher compared to the control (without electro-Fenton), and recovered to 98% of the initial value after the hydraulic cleaning, while the control group only recovered to 88%. In intermittent mode, the membrane permeance could recover to 100% after operation by electro-Fenton regeneration for 30 min with microalgae concentrating from 1.05 g/L to 7.45 g/L. The electro-Fenton-enhanced membrane could not only mitigate the reversible fouling, but also effectively inhibit the irreversible fouling, and presented prominent separation abilities without obvious damage to membranes and microalgae cells.

Abstract:Algal blooms have become a challenging problem in conventional water treatment processes due to numerous negative effects on human health and ecosystems. Photocatalysis has turned out to be a promising solution with the environmental and economic benefits beheld, but the rapid recombination of photogenerated electron and hole inhibited the application in practice. This study proposed a rotating triboelectric nanogenerator (R-TENG) as a self-powered bias for photocatalysis to enhance the algal removal efficiencies. R-TENG harvest waste water wave energy from coagulation tank to convert it into stable DC voltage (31 V) and DC current (27 μA). TiO 2 nanotubes (TNT) serving as the anode in the photocatalysis with TENG system achieved complete inactivation of algae (>97 %) after 7 h of reaction, which was increased by 13.9 % compared with photocatalysis. This enhanced performance was attributed to the improvement of active species generation (e.g., •O 2 - , •OH and h + ). During algal inactivation, the active species destroyed photosynthetic activity (decline of Chlorophyll a), antioxidant system (decrease of superoxide dismutase, catalase and malondialdehyde) and membrane in algal cells. Given the ubiquitous existence of algal cells in surface water, this work offers a novel method to develop high-performance electro-assisted photocatalysis for algal removal.

Abstract:The outbreak of cyanobacteria bloom in surface water has been a long-term global issue, as the adverse impact on aquatic ecosystem and drinking water safety. Electrocoagulation is an efficient method for cyanobacteria removal, but consumption of energy hampers the application. In this study, an efficient and energy generating electrocoagulation process for cyanobacteria removal by a floating iron-air fuel cell (FIAFC) was successfully applied. At different solution conductivities (2.33 ± 0.25 mS/cm and 4.94 ± 0.55 mS/cm), 100 % removals of algal cells were achieved within 50 min and 30 min, respectively. The maximum power densities of FIAFC were 1.34 W/m 2 and 2.48 W/m 2 at the corresponding current densities of 4.58 A/m 2 and 12.43 A/m 2 . In the FIAFC, algal cells were removed mainly through charge neutralization and sweep flocculation processes with zeta potential, scanning electron microscope (SEM) and atomic force microscopy (AFM) analysis. These results suggested that FIAFC was effective for algae removal and electricity generation, which is a promising method for algae - laden water treatment.
