Abstract:Objective The transformation of BZR1, a BR signal transduction component to tomato and its effect on characters of fruit of tomato was investigated in present study. Method The bzr1 gene was introduced into tomato cultivar Zhongshusihao via tumefaciens-mediated transformation and confirmed by molecular test and genetic analysis. The content of main quality components and pigments, as well as ethylene evolution rate was analyzed in fruits of transgenic tomato line and non-transgenic line (control) at four typical developmental stages. The expression of BR biosynthetic and regulated genes, as well as ethylene biosynthetic, response, and receptor genes was also analyzed. Result Significantly higher contents of soluble solids, organic acid, soluble proteins, vitamin C, lycopene and total carotenold, as well as the ethylene evolution rate in fruit of turning stage were observed in transgenic tomato line compared with the control. The expression of BR regulated gene LeBR1 was higher in transgenic line than in the control at the stage of seedling and early three developmental stages of fruit. The expression of LeACS4 gene encoding ACC synthase, which catalyzes the rate-limiting step of ethylene biosynthesis, was higher in the fruit of transgenic line than that of control from the stage of mature green to turning. The expression of ethylene response gene ERF1 and receptor gene LeETR1 and LeETR3 in fruit of breaker stage was significantly higher in transgenic line than in the control. Conclusion It was concluded from the study that the transformation of bzr1gene to tomato increased the content of various nutrients in fruit of turning stage, which is the edible part of tomato, and therefore improved the nutrient quality of tomato.

Abstract:Anthocyanins, natural pigments with high antioxidant activities, are widely distributed in the plant kingdom and play important roles in various physiological processes. Much effort has been committed to enhancing the anthocyanin content of these health-promoting pigments in vegetables and grains, for their eye-catching colors and special nutrients. Previously, we reported that the SmMYB1 gene encoding a R2R3 MYB transcription factor participated in the regulation of anthocyanin biosynthesis in the peel of eggplant. In this work, we introduced SmMYB1 into a non-anthocyanin-accumulating eggplant cultivar (Solanum aethiopicum group Gilo) via Agrobacterium-mediated transformation. Genetically engineered plants exhibited high concentrations of anthocyanin in leaves, petals, stamens, and fruit peels under normal growth conditions, especially in fruit flesh. Furthermore, highly methylated anthocyanins, malvidin 3-(p-coumaroyl)rhamnoside(glucoside)-5-glucoside and malvidin 3-(feruloyl)rhamnoside(glucoside)-5-glucoside, were separated from the purple fruit flesh and identified by HPLC–ESI–MS/MS. qRT-PCR analysis revealed that most anthocyanin structural genes were dramatically up-regulated in the tissues of transgenic lines compared with non-transformed plants. In addition, the transgenic seedlings had greater tolerance to freezing stress and better recovery under rewarming conditions. These results provide a good foundation for the breeding of new eggplant cultivars with more healthy agronomic traits in future studies.

Abstract:Carotenoids have a range of diverse biological functions and actions, especially playing an important role in human health with provitamin A activity, anti-cancer activity, enhancing immune ability and so on. Human body can't synthesis carotenoids by itself and must absorb them from outside. However, carotenoid contents in many plant are very low, and many kinds of carotenoid are difficult to produce by chemical ways. With the elucidation of carotenoid biosynthetic pathway and cloning genes of relative enzymes from microorganisms and higher plants, it is possible to regulate carotenoid biosynthesis via genetic engineering. This article reviews gene cloning of carotenoid biosynthetic enzymes in microorganisms and higher plants, and advances in the studies of carotenoid production in heterologous microorganisms and crop plants using gene-manipulated carotenoid biosynthesis.

Abstract:Increasing the provitamin A content in staple crops via carotenoid metabolic engineering is one way to address vitamin A deficiency. In this work a combination of methods was applied to specifically increase β-carotene content in wheat by metabolic engineering. Endosperm-specific silencing of the carotenoid hydroxylase gene (TaHYD) increased β-carotene content 10.5-fold to 1.76 μg g(-1) in wheat endosperm. Overexpression of CrtB introduced an additional flux into wheat, accompanied by a β-carotene increase of 14.6-fold to 2.45 μg g(-1). When the "push strategy" (overexpressing CrtB) and "block strategy" (silencing TaHYD) were combined in wheat metabolic engineering, significant levels of β-carotene accumulation were obtained, corresponding to an increase of up to 31-fold to 5.06 μg g(-1). This is the first example of successful metabolic engineering to specifically improve β-carotene content in wheat endosperm through a combination of methods and demonstrates the potential of genetic engineering for specific nutritional enhancement of wheat.

Abstract:Transgenic approach is an excellent way for the clarification of gene function, but it is generally difficult to create transgenic plants for most of the fruit trees including peach. Even if transgenic lines are successfully obtained, it will be extremely difficult to evaluate fruit traits due to the long juvenile phase of the plants. To overcome this problem, usage of virus vector is one of the excellent approaches. In this study, we evaluated gene function related to fruit traits in peaches via virus-induced gene silencing (VIGS). Carotenoid cleavage dioxygenase 4 (CCD4) is proposed to be the key factor responsible for carotenoid degradation in white flesh peaches. Then, we knocked down the CCD4 gene in the white flesh peaches (“Akatsuki” and “Manami”) via VIGS system. Resultantly, yellow pigmentation and increased contents of carotenoids including lutein, ß-carotene, ß-cryptoxanthin, zeaxanthin, and violaxanthin were observed in the agroinfiltration portions of the white flesh peaches, which is a direct evidence that CCD4 is a determinant for yellow flesh trait of peach. Our results suggested the possible application of VIGS system for functional studies of the genes related to fruit traits.

Abstract:Fruit color is a genetic trait and a key factor for consumer acceptability and is therefore receiving increasing importance in several breeding programs. Plant pigments offer plants with a variety of colored organs that attract animals for pollination, favoring seed dispersers and conservation of species. The pigments inside plant cells not only play a light-harvesting role but also provide protection against light damage and exhibit nutritional and ecological value for health and visual pleasure in humans. Tomato (Solanum lycopersicum) is a leading vegetable crop; its fruit color formation is associated with the accumulation of several natural pigments, which include carotenoids in the pericarp, flavonoids in the peel, as well as the breakdown of chlorophyll during fruit ripening. To improve tomato fruit quality, several techniques, such as genetic engineering and genome editing, have been used to alter fruit color and regulate the accumulation of secondary metabolites in related pathways. Recently, clustered regularly interspaced short palindromic repeat (CRISPR)-based systems have been extensively used for genome editing in many crops, including tomatoes, and promising results have been achieved using modified CRISPR systems, including CAS9 (CRISPR/CRISPR-associated-protein) and CRISPR/Cas12a systems. These advanced tools in biotechnology and whole genome sequencing of various tomato species will certainly advance the breeding of tomato fruit color with a high degree of precision. Here, we attempt to summarize the current advancement and effective application of genetic engineering techniques that provide further flexibility for fruit color formation. Furthermore, we have also discussed the challenges and opportunities of genetic engineering and genome editing to improve tomato fruit color.

Abstract:Carotenoid content is a primary determinant of wheat nutritional value and affects its end-use quality. Wheat grains contain very low carotenoid levels and trace amounts of provitamin A content. In order to enrich the carotenoid content in wheat grains, the bacterial phytoene synthase gene (CrtB) and carotene desaturase gene (CrtI) were transformed into the common wheat cultivar Bobwhite. Expression of CrtB or CrtI alone slightly increased the carotenoid content in the grains of transgenic wheat, while co-expression of both genes resulted in a darker red/yellow grain phenotype, accompanied by a total carotenoid content increase of approximately 8-fold achieving 4.76 mu g g(-1) of seed dry weight, a beta-carotene increase of 65-fold to 3.21 mu g g(-1) of seed dry weight, and a provitamin A content (sum of alpha-carotene, beta-carotene, and beta-cryptoxanthin) increase of 76-fold to 3.82 mu g g(-1) of seed dry weight. The high provitamin A content in the transgenic wheat was stably inherited over four generations. Quantitative PCR analysis revealed that enhancement of provitamin A content in transgenic wheat was also a result of the highly coordinated regulation of endogenous carotenoid biosynthetic genes, suggesting a metabolic feedback regulation in the wheat carotenoid biosynthetic pathway. These transgenic wheat lines are not only valuable for breeding wheat varieties with nutritional benefits for human health but also for understanding the mechanism regulating carotenoid biosynthesis in wheat endosperm.

Abstract:Fruit specific promoter (2A12) from Lycopersicom esculentum and cDNA of isopentenyl-transferase (ipt) from Ti plasmid of Agrobacterium tumerfaciens C58 were cloned by PCR procedure respectively. Two plant expression vectors with 2A12/gus or 2A12/ipt were respectively constructed. These two chimeric genes were transferred into tomato by Agrobacterium mediated procedure. The results of Southern hybridization showed that the fusion genes had been integrated into tomatoes. The result of gus histochemical staining showed that 2A12 had high fruit specific expressive capability in transgenic tomato. The ipt expression resulted in accumulation of high level of cytokinins (CTKs) in fruit lead to developmental changes in fruits and seeds. The fruit of ipt transformed tomato had the hyperplastic placenta with very few seeds or even seedless. The shelf life of transgenic fruits elongated for 1–2 weeks. The ratio of fruit set, the dry weight of fruit and the crude protein content in fruit were increased, while the soluble sugar of fruits decreased.
Fine Mapping of Candidate Gene Controlling Anthocyanin Biosynthesis for Purple Peel in Solanum melongena L.

Abstract:Fruit color is an intuitive quality of horticultural crops that can be used as an evaluation criterion for fruit ripening and is an important factor affecting consumers' purchase choices. In this study, a genetic population from the cross of green peel 'Qidong' and purple peel '8 guo' revealed that the purple to green color of eggplant peel is dominant and controlled by a pair of alleles. Bulked segregant analysis (BSA), SNP haplotyping, and fine genetic mapping delimited candidate genes to a 350 kb region of eggplant chromosome 10 flanked by markers KA2381 and CA8828. One ANS gene (EGP22363) was predicted to be a candidate gene based on gene annotation and sequence alignment of the 350-kb region. Sequence analysis revealed that a single base mutation of 'T' to 'C' on the exon green peel, which caused hydrophobicity to become hydrophilic serine, led to a change in the three-level spatial structure. Additionally, EGP22363 was more highly expressed in purple peels than in green peels. Collectively, EGP22363 is a strong candidate gene for anthocyanin biosynthesis in purple eggplant peels. These results provide important information for molecular marker-assisted selection in eggplants, and a basis for analyzing the regulatory pathways responsible for anthocyanin biosynthesis in eggplants.

Abstract:The accumulation of carotenoids in plants is a key nutritional quality in many horticultural crops. Although the structural genes encoding the biosynthetic enzymes are well-characterized, little is known regarding photoperiod-mediated carotenoid accumulation in the fruits of some horticultural crops. Herein, we performed physiological and transcriptomic analyses using two cucumber genotypes, SWCC8 (XIS-orange-fleshed and photoperiod-sensitive) and CC3 (white-fleshed and photoperiod-non-sensitive), established under two photoperiod conditions (8L/16D vs. 12L/12D) at four fruit developmental stages. Day-neutral treatments significantly increased fruit β-carotene content by 42.1% compared to short day (SD) treatments in SWCC8 at 40 DAP with no significant changes in CC3. Day-neutral condition elevated sugar levels of fruits compared to short-day treatments. According to GO and KEGG analyses, the predominantly expressed genes were related to photosynthesis, carotenoid biosynthesis, plant hormone signaling, circadian rhythms, and carbohydrates. Consistent with β-carotene accumulation in SWCC8, the day-neutral condition elevated the expression of key carotenoid biosynthesis genes such as PSY1, PDS, ZDS1, LYCB, and CHYB1 during later stages between 30 to 40 days of fruit development. Compared to SWCC8, CC3 showed an expression of DEGs related to carotenoid cleavage and oxidative stresses, signifying reduced β-carotene levels in CC3 cucumber. Further, a WGCNA analysis revealed co-expression between carbohydrate-related genes (pentose-phosphatase synthase, β-glucosidase, and trehalose-6-phosphatase), photoperiod-signaling genes (LHY, APRR7/5, FKF1, PIF3, COP1, GIGANTEA, and CK2) and carotenoid-biosynthetic genes, thus suggesting that a cross-talk mechanism between carbohydrates and light-related genes induces β-carotene accumulation. The results highlighted herein provide a framework for future gene functional analyses and molecular breeding towards enhanced carotenoid accumulation in edible plant organs.

Abstract:Abstract Background: Fruit skin color intensity is one of the most important economic traits of purple eggplant. A wide diversity for fruit skin color intensity exists in purple eggplant and the accumulation of anthocyanins and chlorophylls of fruit skin mainly affected color intensity. However, limited information is available contributing to the molecular mechanisms underlying fruit skin color intensity variation in eggplant.Results: In the present study, variation of two purple eggplant advanced lines EP26 and EP28, with different fruit skin color intensity was investigated. Comparative transcriptome analysis of EP26 and EP28 identified a total of 2218 differential expressed genes (DEGs) at two different developmental stages. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis showed that these DEGs were mainly involved in flavonoid biosynthesis and photosynthesis. In addition, a total of 131 transcription factors including MYB , bHLH , WRKY , and NAC exhibited dynamic changes, which might be responsible for the variation of fruit pigments accumulation between EP26 and EP28.Conclusions: Taken together, these results expand our knowledge of molecular mechanisms underlying fruit skin color intensity variation in purple eggplant, which allowing for improvement of fruit coloration in eggplant breeding.
Developing carotenoids-enhanced tomato fruit with multi-transgene stacking strategies

Abstract:As natural dominant pigments, carotenoids and their derivatives not only contribute to fruit color and flavor quality but are regarded as phytochemicals beneficial to human health because of various bioactivities. Tomato is one of the most important vegetables as well as a main dietary source of carotenoids. So, it's of great importance to generate carotenoid-biofortified tomatoes. The carotenoid biosynthesis pathway is a network co-regulated by multiple enzymes and regulatory genes. Here, we assembled four binary constructs containing different combinations of four endogenous carotenoids metabolic-related genes, including SlORHis, SlDXS, SlPSY, and SlBHY by using a high efficiency multi-transgene stacking system and a series of fruit-specific promotors. Transgenic lines overexpression SlORHis alone, three genes (SlORHis/SlDXS/SlPSY), two genes (SlORHis/SlBHY), and all these four genes (SlORHis/SlDXS/SlPSY/SlBHY) were enriched with carotenoids to varying degrees. Notably, overexpressing SlORHis alone showed comparable effects with simultaneous overexpression of the key regulatory enzyme coding genes SlDXS, SlPSY, and SlORHis in promoting carotenoid accumulation. Downstream carotenoid derivatives zeaxanthin and violaxanthin were detected only in lines containing SlBHY. In addition, the sugar content and total antioxidant capacity of these carotenoids-enhanced tomatoes was also increased. These data provided useful information for the future developing of biofortified tomatoes with different carotenoid profiles, and confirmed a promising system for generation of nutrients biofortified tomatoes by multiple engineering genes stacking strategy.

Abstract:An increasing interest in carotenoids as nutritional sources of provitamin A and health-promoting compounds has prompted a significant effort in metabolic engineering of carotenoid content and composition in food crops. The strategy commonly used in plants is to increase the biosynthetic capacity by altering the carotenogenic enzyme activities. The recent isolation of the Or gene from a cauliflower orange mutant has brought a new endeavor for carotenoid enhancement by increasing the sink strength to sequester and store the synthesized carotenoids. Potato as one of the major staple crops usually accumulates low levels of carotenoids. In this chapter, we describe a detailed protocol for metabolic engineering of carotenoids in potato plants with the Or gene and the analysis of the Or transformants.

Abstract:Melon is an important fruit in the Japanese food market. Accordingly, to be a leading variety in Japan, the fruit must meet demands from growers, markets, and consumers. Melon breeders have made great efforts to incorporate such demands into breeding programs. We have developed methods for improving two important traits, shelf-life and fruit size, by genetic engineering. First, we isolated and characterized genes related to these characteristics and subsequently used the genes to develop methods for altering shelf-life and fruit size. We isolated two ethylene receptor genes, Cm-ERS1 and Cm-ETR1, which we expected to be related to shelf-life, and developed methods for altering the ethylene sensitivity of the plants. We also isolated a gene encoding 3-hydroxy-3-methylglutaryl coenzyme A reductase, Cm-HMGR, which we expected to be related to fruit enlargement, and developed methods for altering fruit size. These two methods will provide basic technologies for the molecular breeding of melon.

Abstract:Cassava (Manihot esculenta Crantz) is a staple crop in tropical and subtropical regions, valued for its high starch content in roots. Effective genetic transformation and genome editing of cassava require efficient screening methods for transgenic and edited plants. In this study, a visual selection marker system using an R2R3-MYB transcription factor anthocyanin 1 gene (HbAN1, LOC110667474) from a rubber tree (Hevea brasiliensis Müll. Arg.) has been developed to facilitate the identification of transgenic cassava plants. Transgenic cassava lines expressing HbAN1 accumulated anthocyanins in their leaves, allowing for easy visual identification without the need for destructive assays or specialized equipment. Importantly, the accumulation of anthocyanins did not affect the regeneration or transformation efficiency of cassava. Additionally, the AR-CRISPR/Cas9-gRNA system with the HbAN1 gene as a marker produced MeCDD4 gene-edited cassava mutants with purple leaves, demonstrating successful editing. This anthocyanin-based visual reporter (AR) system will provide an effective tool for genetic transformation and genome editing in cassava.

Abstract:Carotenoid pigments are valuable components of the human diet. A notable example is β-carotene, or provitamin A, which is converted into the derivatives astaxanthin and capsanthin, via the common intermediate zeaxanthin. To generate rice varieties producing diverse carotenoids beyond β-carotene, we specifically used a Capsicum β-carotene hydroxylase gene, B (CaBch) and a codon optimized version of the same gene, stB (stBch) to increase zeaxanthin synthesis. We also used a recombinant BAK gene (CaBch-2A-HpBkt), consisting of the CaBch sequence and a Haematococcus β-carotene ketolase gene (HpBkt) linked by a bicistronic 2 A sequence, as well as a codon optimized recombinant stBAK gene (stBch-2A-stBkt) to create astaxanthin synthesis. The four cassettes to seed-specifically express the B, stB, BAK and stBAK genes were individually combined with a PAC gene (CaPsy-2A-PaCrtI) cassette to previously impart β-carotene-enriched trait in rice endosperm. The single T-DNA vectors of B-PAC, stB-PAC, BAK-PAC and stBAK-PAC resulted in the accumulation of zeaxanthin and astaxanthin in the endosperm of the transgenic rice seeds. In addition, an extended version on the carotenoid pathway was introduced into rice to allow the production of capsanthin, by intercrossing a B-PAC rice line with a Ccs rice line, which harbors a Capsicum capsanthin-capsorubin synthase gene. Ultimately, we developed three functional rice varieties: B-PAC (0.8 μg/g zeaxanthin, deep yellow), stBAK-PAC (1.4 μg/g ketocarotenoids, including astaxanthin, pinkish red) and B-PAC x Ccs (0.4 μg/g of ketoxanthophylls, including capsanthin, orange-red) with the similar levels of total carotenoids to PAC rice, suggesting the capacity was dependent on β-carotene levels. Collectively, a combination of genetic engineering and conventional breeding is effective for multi-step metabolic engineering and biochemical pathway extension.
Production of Transgenic Rice Containing apl Gene Encoding a Ferredoxin-like Amphipathic Protein from Sweet Pepper

Abstract:Two to three-week-old mature seed-derived rice callus (Oryza sativa L., japonica cv. Eyi 105) was co-transformed via particle bombardment with two plasmids, pJIMB15 and pBISAPl, containing the selectable marker hygromycin phosp hotransferase gene (hpt), the reporter β-glucuronidase gene (gusA) and the ap1 gene. The ap1 gene encodes an amphipathic protein previousl y shown to delay the hypersensitive response induced in non-host plants by the pathogen Pseudomonas syringae pv. syringae(Pss). After two rounds of sel ection on hygromycin (30 mg/L)-containing medium, resistant callus was transfer red to hygromycin (30 mg/L)-containing regeneration medium for plant regenarati on. Thirty-two independent transgenic plants were regenerated from 186 bombarde d callused, with a transformation frequency of 17.2%. Eighty-four percent of th e transgenic plants carried all three transgenes as shown by PCR/Southern blot a nalysis.
Beyond the limits of photoperception: constitutively active PHYTOCHROME B2 overexpression as a means of improving fruit nutritional quality in tomato

Abstract:Photoreceptor engineering has recently emerged as a means for improving agronomically beneficial traits in crop species. Despite the central role played by the red/far-red photoreceptor phytochromes (PHYs) in controlling fruit physiology, the applicability of PHY engineering for increasing fleshy fruit nutritional content remains poorly exploited. In this study, we demonstrated that the fruit-specific overexpression of a constitutively active GAF domain Tyr252 -to-His PHYB2 mutant version (PHYB2Y252H ) significantly enhances the accumulation of multiple health-promoting antioxidants in tomato fruits, without negative collateral consequences on vegetative development. Compared with the native PHYB2 overexpression, PHYB2Y252H -overexpressing lines exhibited more extensive increments in transcript abundance of genes associated with fruit plastid development, chlorophyll biosynthesis and metabolic pathways responsible for the accumulation of antioxidant compounds. Accordingly, PHYB2Y252H -overexpressing fruits developed more chloroplasts containing voluminous grana at the green stage and overaccumulated carotenoids, tocopherols, flavonoids and ascorbate in ripe fruits compared with both wild-type and PHYB2-overexpressing lines. The impacts of PHYB2 or PHYB2Y252H overexpression on fruit primary metabolism were limited to a slight promotion in lipid biosynthesis and reduction in sugar accumulation. Altogether, these findings indicate that mutation-based adjustments in PHY properties represent a valuable photobiotechnological tool for tomato biofortification, highlighting the potential of photoreceptor engineering for improving quality traits in fleshy fruits.
Overexpression of a BR inactivating enzyme gene GhPAG1 impacts eggplant fruit development and anthocyanin accumulation mainly by altering hormone homeostasis

Abstract:Brassinosteroids (BRs) function importantly in plant growth and development, but the roles in regulating fruit development and anthocyanin pigmentation remain unclear. Eggplant ( Solanum melongena L.) is an important Solanaceae vegetable crop rich in anthocyanins. The fruit size and coloration are important agronomic traits for eggplant breeding. In this study, transgenic eggplant exhibiting endogenous BRs deficiency was created by overexpressing a heterologous BRs-inactivating enzyme gene GhPAG1 driven by CaMV 35 S promoter. 35 S:: GhPAG1 eggplant exhibited severe dwarfism, reduced fruit size, and less anthocyanin accumulation. Microscopic observation showed that the cell size of 35 S:: GhPAG1 eggplant was significantly reduced compared to WT. Furthermore, the levels of IAA, ME-IAA, and active JAs (JA, JA-ILE, and H2JA) all decreased in 35 S:: GhPAG1 eggplant fruit. RNA-Seq analyses showed a decrease in the expression of genes involved in cell elongation, auxin signaling, and JA signaling. Besides, overexpression of GhPAG1 significantly downregulated anthocyanin biosynthetic genes and associated transcription regulators. Altogether, these results strongly suggest that endogenous brassinosteroid deficiency arising from GhPAG1 overexpression impacts eggplant fruit development and anthocyanin coloration mainly by altering hormone homeostasis.
