Abstract:Vitamin C (VC) maintains the activity of sulfhydrylase and the reduced state of glutathione, and acts as a detoxifying agent. It is widely found in plant tissues as well as in fresh fruits and vegetables. Accurate determination of vitamin C content is of great significance for diet and health care. In this work, MnO2 nanoflakes as the biomimetic oxidase was used to react with VC and TMB was used as a chromogenic agent. The color of the final solution reflects the concentration of VC. And the bionic E-eye as a portable in-situ detection platform was designed to analyzed the color change in 10 s by photographing. By optimizing experimental conditions, the linear range of this method for VC detection is from 7.8 μM to 62.5 μM and the detection limit 7.65 μM which is much lower than other methods. Besides, this methods shows good selectivity, short reaction time (10 min) and also was used for VC detection in artificial urine samples, which shows great potential in disease monitoring.

Abstract:Adenosine can be a possible biomarker for monitoring the progress of lung cancer. Here, a sensitive and easily operated colorimetric methods based on aptasensor for the detection of adenosine has been developed. Gold nanoparticles (AuNPs) will aggregate in high-salt solutions, which causes color change, and single-stranded DNA will prevent this from happening. Based on changes in solution color, adenosine can be quantitatively determined. Besides a home-made bionic electronic-eye (E-eye) system was developed as a portable in-situ detection platform. Two different aptamers were used for comparison for adenosine detection. The linear correlation existed between 5.0-60.0μM with a detection limit of 0.17μM in water with modified aptamer. Moreover, the performance of the method was validated by testing the artificial urine samples with the linear range from 5.0 to 50.0μM and a detection limit of 0.48μM in artificial urine. This simple and cost-effective approach indicating its great potential for point-of-care applications.

Abstract:Due to their unique optical properties, narrow size distributions, and good biological affinity, gold nanoparticles have been widely applied in sensing analysis, catalytic, environmental monitoring, and disease therapy. The color of a gold nanoparticle solution and its maximum characteristic absorption wavelength will change with the particle size and inter-particle spacing. These properties are often used in the detection of hazardous chemicals, such as pesticide residues, heavy metals, banned additives, and biotoxins, in food. Because the gold nanoparticles-colorimetric sensing strategy is simple, quick, and sensitive, this method has extensive applications in real-time on-site monitoring and rapid testing of food quality and safety. Herein, we review the preparation methods, functional modification, photochemical properties, and applications of gold nanoparticle sensors in rapid testing. In addition, we elaborate on the colorimetric sensing mechanisms. Finally, we discuss the advantages and disadvantages of colorimetric sensors based on gold nanoparticles, and directions for future development.

A colorimetric sensor based on multiple elements doped carbon dot nanozyme for rapid detection of 1-naphthol in human urine samples

Abstract:The residual carbaryl in crops can cause serious damage to the human kidney and nervous system after entering the human body, which may be metabolized to 1-naphthol (1-NAP) and excreted through urine. 1-NAP is often used as the biomarker for carbaryl exposure, so the intake or leakage of carbaryl can be monitored by detecting the concentration of 1-NAP. Herein, Co, N, P ternary co-doped carbon dots (CoNP-CDs) derived from vitamin B12 were synthesized by a facile hydrothermal method. CoNP-CDs exhibited oxidase-like activity and excellent peroxidase-like activity, which was attributed to the Fenton-like reaction of Co2+/Co3+ and the presence of pyrrole N and P elements, which together provided multiple active sites for chromogenic substrates. Due to the dual enzyme-like activity of CoNP-CDs, hydroxyl radicals (OH) and superoxide radicals (O2-) were generated during the catalytic process, which could rapidly oxidize colorless 3,3',5,5'-tetramethyl benzidine (TMB) to blue oxidation products (oxTMB). The α-carbon in 1-NAP can be attacked by OH, and the catalytic oxidation process of TMB can be inhibited by the consumption of OH, so that the blue color of the solution became lighter. Based on this principle, a smartphone-assisted colorimetric sensing platform was constructed for the detection of 1-NAP, and which resulted in a linear range of 1.07-37.3 μM and a visual detection limit of 0.68 μM. Moreover, the colorimetric sensing system showed satisfactory recoveries in the detection of human urine samples. The colorimetric sensing system owned the advantages of fast response, strong selectivity and simple operation, and provided a potential strategy for the on-site detection of 1-NAP.

Abstract:A highly selective method is presented for the colorimetric determination of dopamine (DA) using gold nanoparticles (AuNPs). DA caps on the surface of AuNPs (DA-AuNPs) induces the aggregation of AuNPs in alkaline solution. The DA-AuNPs are modified by the hydrolysate of thioglycolic acid (TGA(2)(-)) through Au-S bonds. The aggregation of AuNPs is accelerated by TGA(2-), due to the strong hydrogen-bonds (NH⋯OC and OH⋯OC) formed between TGA(2)(-) and DA. Upon the addition of DA, the solution shows a color change from red to purple (or yellow), which is also monitored to detect DA in human urine and fetal bovine serum samples. Here, the limits of colorimetric detection are as low as 10(-7)M observed in Milli-Q water, urine and serum. Based on UV-vis absorption spectra, the limits of detection have been calculated to be 3.3×10(-8)M, 1.0×10(-7)M and 9.4×10(-8)M in Milli-Q water, urine, and serum, respectively. All the limits of detection are lower than the lowest abnormal concentrations of DA in urine (5.7×10(-7)M) and blood (1.6×10(-5)M). The good linear ranges from 0 to 10(-6)M are used for the quantitative assay of DA in urine and serum samples. The applicability of our detection system is also verified by analysis of DA in urine and serum samples. The developed approach is without using complex financial instruments.

Abstract:Creatinine level is a crucial indicator in the clinical assessment and diagnosis of renal diseases, and achieving simple and accurate detection of urinary creatinine levels in resource-limited point-of-care settings is of great significance in the timely prevention and diagnosis of kidney diseases. As a popular zero-dimensional material, gold nanoparticles (AuNPs) exhibit intriguing optical properties and thus have become a promising material for many sensing detection applications. Here, we proposed a simple, efficient, and sensitive quantitative detection of creatinine by studying the relative absorbance (Δ A ) of AuNPs in absence and presence of creatinine. The method relies on the aggregation of AuNPs via ligand-exchanged of citrate ions and creatinine on the surface of AuNPs to achieve colorimetric detection. With this assay, the limit of detection for creatinine was as low as 0.16 mM, and the dynamic detection range was 0.5 to 20 mM under optimized conditions. In our experiments, the specificity of the proposed method was investigated and successfully applied to detect creatinine in urine sample. It reveals that the proposed colorimetric protocol has demonstrated a high sensitivity and selectivity for creatinine, and has a potential practicability in clinical diagnostics.

Abstract:Ensuring the vitamin C requirements are met through dietary intake or supplements is essential for health. In this study, a simple and rapid visual detection method for semi-quantitative analysis of vitamin C was proposed based on sodium citrate-stabilized silver nanoparticles (SC-AgNPs). As a validation of SC-AgNPs as a colorimetric sensor for vitamin C, color changes were observed gradually, shifting from transparent yellow to deep brown after the addition of vitamin C at various concentrations. That is also supported by an increase in absorption intensity at the peak wavelength of 417 nm, which was analyzed using a UV-Vis spectrophotometer. Based on the spectral approach method, the linear relationship between absorbance values and vitamin C is in the range of 0-4.2 mM, with a coefficient correlation of 0.99 and sensitivity of 0.95/mM. Furthermore, the feasibility of the SC-AgNPs for vitamin C detection in healthy beverages and tablet pharmaceuticals has been investigated.

Abstract:A novel, enzyme-free and aptamer-based colorimetric platform for protein detection has been developed, which takes advantage of aptamer-functionalized magnetic beads (MBs) for target capture, concentration and separation, and aptamer-conjugated gold nanoparticle (AuNP)-catalyzed color bleaching reaction of methyl orange (MO) to generate the colorimetric signals. It was demonstrated that the proposed colorimetric sensing strategy enables simple, cost-effective, sensitive and specific thrombin detection without the use of any enhancing solutions and enzymes. Herein, by naked eye observation, we can detect the human thrombin with a detection limit of approximately 320 pM, which can be further decreased to 30 pM with the help of a UV-vis instrument. In addition, this method also works for targets with two or more binding sites.

Abstract:An easy and reliable method based on a novel electroanalytical nanostructured sensor has been developed to perform quantification of vitamin C in commercial and fortified cow-milk-based formulae and foods for infants and young children. The work is motivated by the need of a reliable analytical tool to be applied in quality control laboratories for the quantitative assessment of vitamin C where its rapid and cost-effective monitoring is essential.The ad hoc designed sensor, based on disposable Screen-Printed Carbon Electrodes modified with Au nanoparticles decorated Reduced Graphene Oxide flakes, exhibits a LOD of 0.088 mg L-1. The low cost, easy sample preparation, fast response and high reproducibility (RSD≈8%) of the proposed method highlight its suitability for usage in quality control laboratories for determining vitamin C in real complex food matrices, envisaging the application of the sensing platform in the determination of other compounds relevant in food chemistry and food manufacturing.

Abstract:Pathogenic bacteria are a major cause of foodborne diseases, which not only seriously threaten human safety but also cause significant losses for the national economy. Therefore, it is very important and urgent to develop a method for the detection of pathogenic bacteria with high accuracy, high sensitivity, and easy interpretation for use in food safety and medical hygiene. Herein, based upon the sensitive color changes induced by the dispersion and aggregation states of gold nanoparticles (AuNPs), a point-of-care (POCT) colorimetric assay was constructed for the rapid and sensitive visual detection of pathogenic bacteria. The POCT visual sensing system is composed of two individual elements: (1) an alkaline phosphatase/graphene oxide (GO@PEI-ALP) nanoconjugate that can release free ALP molecules in the presence of pathogenic bacteria; (2) D-glucose-6-phosphate (pGlu) and 3-aminobenzene boric acid (AMBA)-functionalized AuNPs (pGlu/AMBA-AuNPs) that are cross-linked upon the digestion of pGlu by free ALP molecules, resulting in a significant color change. Under optimized conditions, the detection limit of this sensing system for target bacteria was as low as 24 CFU mL−1 and was successfully applied to complex real samples. This proposed rapid colorimetric assay has high sensitivity, accuracy, and practicability with an intuitive signal and is expected to provide new inspiration for the detection of pathogenic bacteria.

Direct detection of β-agonists by use of gold nanoparticle-based colorimetric assays

Abstract:β-Agonists fed to animals for human consumption pose a serious threat to human health. Fast, broad-spectrum detection methods are needed for on-site screening of various types of β-agonists from animal feeds, meats, and animal body fluids. We developed a colorimetric assay that uses gold nanoparticle (AuNP) plasmon absorption to realize quick detection of β-agonists from liquid samples. β-Agonists showed the capability of directly reducing HAuCl(4) into atomic gold, which involved oxidation of the amine or phenol group on the benzene ring of the β-agonists. The resulting atomic gold formed AuNPs spontaneously, which had strong plasmon absorption at 528 nm. The linear relationship between the concentrations of β-agonists and the AuNPs plasmon absorbance granted quantitative determination of β-agonists in solution. The AuNPs colorimetric assay showed different sensitivities toward β-agonists with different substituent groups on the aromatic ring. β-Agonists with phenol groups had a lower limit of quantitation (LOQ) than those with amine groups. High-resolution transmission electron microscopy (TEM) images revealed the sizes of the AuNPs were in the range 15-25 nm, while X-ray energy-dispersive spectroscopic data suggested the smaller particles observed in TEM with lower contrast may be salt particles from the buffer solution. The developed colorimetric assay can potentially be used for the detection of β-agonists and their analogues from serum, urine, and other liquid samples in the presence of interference from common antibiotics and glucose.

Abstract:Rapid, sensitive, and reliable bacterial pathogens detection is a chief requirement. The gold nanoparticles (AuNPs) have numerous applications such as in the detection of biomolecules for their high surface to volume ratio and unique optical property facilitating development of highly efficient AuNPs-based bio-sensing tools. Although various molecular detection methods, such as PCR, real-time PCR, and loop-mediated isothermal amplification are sensitive and convenient, these techniques need elaborate work and require special skills to increase their specificity. Smartly fabricated gold nanoparticle (GNPs) play a role as probes for selective detection of pathogens. The AuNPs-based colorimetric methods have become applicable for rapid, simple, reliable and high-efficient, sensitive, inexpensive, and easy detection of the DNA, RNA, and protein biomolecules. Colorimetric detection using AuNPs has been used for rapid and high precision and multiplex detection of a large number and of bacterial pathogens. AuNPs act in functionalized and unfunctionalized ways. AuNPs-based colorimetric methods have incredible advantages compared with many other bacterial detection methods. In spite of many molecular techniques, AuNPs-based colorimetric methods do not require additional devices, fabrication cost, signal processing and interpretation complexities, and costly and complex instruments. This simple and rapid method is suitable, especially in low-income areas and for large number of samples analysis. In this review, applications of AuNPs and AuNPs-based colorimetric methods for bacterial pathogens detection have been overviewed.
Sensitive and highly selective biosensor based on innovative V2O5 nanoparticles for detection of glutathione

Abstract:In this study, V2O5 nanoparticles with flake or prism‐like morphology were synthesized using a two‐step solvothermal synthesis and calcination process for the first time. These nanoparticles exhibited intrinsic oxidase‐like activity, catalyzing the oxidation of 3,3′,5,5′‐tetramethylbenzidine to produce blue oxidized 3,3′,5,5′‐tetramethylbenzidine even in the absence of H2O2, with a characteristic absorption peak at 652 nm. Upon the introduction of glutathione (GSH), the solution color gradually lightened, correlating with a reduction in absorbance. Leveraging these properties, we developed a simple, sensitive, and highly selective colorimetric biosensor utilizing V2O5 nanoparticles for GSH detection in human serum. The developed method demonstrated excellent linearity over a range of 1–30 μM, with a low detection limit of 4.04 nM. Additionally, it exhibited outstanding selectivity against common interfering substances in human serum. Furthermore, this biosensor enabled both naked‐eye detection and spectrophotometric quantitative analysis of GSH. Successful application to spiked serum samples yielded recoveries ranging from 97.1% to 101.7%. Overall, this method offers a promising approach for determining GSH content in human serum, with significant potential for biomedical testing applications. Its rapid and accurate detection capability may contribute to early diagnosis and treatment of various fatal diseases, including cancer and cardiovascular diseases.

Abstract:A simple, rapid, sensitive and field-portable colorimetric technique for the determination of Cr(III) in aqueous solution based on an aggregation-induced color transition of gold nanoparticles (AuNPs) has been developed. AuNPs were first functionalized with a dithiocarbamate-modified N-benzyl-4-(pyridin-4-ylmethyl)aniline ligand (BP-DTC). Chelation of Cr(III) by several of these ligands, bound to different nanoparticles, led to nanoparticle aggregation in solution. This gave rise to a color change from wine-red to blue that was discernible by the naked eye and an easily measurable alteration in the extinction spectrum of the particles. The method could be used to determine Cr(III) with a detection limit of 31ppb. Furthermore, selective detection of trace Cr(III) in aqueous solution in the presence of 12 other transition metal ions has been achieved. Toward the goal of practical applications, the sensor has been further evaluated with a view to monitoring Cr(III) in nutritional supplements and the blood of diabetes patients and also applied in the indirect determination of Cr(VI) in waste water.

Abstract:Development of simple and reliable sensor for detecting vitamin content is of great significance for guiding human nutrition metabolism, overseeing the quality of food or drugs, and advancing the treatment of related diseases. In this work, a simple electrochemical sensor was conveniently fabricated by modification a carbon electrode with vertically-ordered mesoporous silica film (VMSF), enabling highly sensitive electrochemical detection of vitamin B2 (VB2) based on the dual enrichment of the analyte by the supporting electrode and nanochannels. The widely used glassy carbon electrode (GCE), was preactivated using simple electrochemical polarization, The resulting preactivated GCE (p-GCE) exhibited improved potential resolution ability and enhanced peak current of VB2. Stable modification of VMSF on p-GCE (VMSF/p-GCE) was achieved without introducing another binding layer. The dual enrichment effect of the supporting p-GCE and nanochannels facilitated sensitive detection of VB2, with a linear concentration ranged from 20 nM to 7 μM and from 7 μM to 20 μM. The limit of detection (LOD), determined based on a signal-to-noise ratio of three (S/N = 3), was found to be 11 nM. The modification of ultra-small nanochannels of VMSF endowed VMSF/p-GCE with excellent anti-interference and anti-fouling performance, along with high stability. The constructed sensor demonstrated the capability to achieve direct electrochemical detection of VB2 in turbid samples including milk and leachate of compound vitamin B tablet without the need for complex sample pretreatment. The fabricated electrochemical can be easily regenerated and has high reusability. The advantages of simple preparation, high detection performance, and good regeneration endow the constructed electrochemical sensor with great potential for direct detection of small molecule in complex samples.

On the basis of this property, we constructed a novel sensing platform for the determination of urea, urease, and urease inhibitor. The limit of detection for urea and urease was 5 μM and 1.8 U/L, respectively. The half-maximal inhibition value IC50 of acetohydroxamic acid was found to be 0.05 mM. Urea in human urine and urease in soil were detected with satisfied results.

Abstract:A gold nanoparticle (AuNP)–based sensing strategy based on rapid reduction of Au(I→0) is proposed. As a proof-of-concept study, the proposed sensing principle is designed for simultaneous and colorimetric detection and discrimination of multiple proteins. In the presence of H2O2, the target proteins could reduce Au(I) (i.e. HAuCl2) to AuNPs with different sizes, shapes and dispersion/aggregation states, thus resulting in rapidly colorimetric identification of different proteins. The optical response (i.e. color) of AuNPs is found to be characteristic of a given protein. The color response patterns are characteristic for each protein and can be quantitatively differentiated by statistical techniques. The sensor array is capable of discriminating proteins at concentrations as low as 0.1 μg/mL with high accuracy. A linear relationship was observed between the total Euclidean distances and protein concentration, providing the potential for protein quantification using this sensor array. The limit of detection (LOD) for catalase (Cat) is 0.08 μg/mL. The good linear range (from 0 to 8 μg/mL) has been used for the quantitative assay of Cat. To show a potentially practical application, this method was used to detect and discriminate proteins in human urine and tear samples.

Abstract:A sensitive, simple, and rapid chemiluminescence method has been employed for monitoring of sub-nanogram level of vitamin C in human urine. The procedure was based on strong decrease of chemiluminescence by vitamin C in reaction with luminol and periodate. It has been found that quenching of chemiluminescence was linearly proportional to the logarithm of vitamin C concentration in the range of 0.1-100 ng mL(-1). The analytical pro cedure, including sampling and washing, was completed within 0.5 min, giving an analytical throughput of 120 samples per hour. The limit of detection was 003. ng mL(-1) (3 sigma) and relative standard deviation was lower than 2.5%. The proposed method was successfully applied to the monitoring of excretion of vitamin C with human urine during 24 h after oral administration of tablets of vitamin C. Possible mechanism of chemiluminescence changes has been also discussed.
