Abstract:Diabetes mellitus-related chronic non-healing wounds have greatly lowered the quality of life for people afflicted and placed a heavy financial burden on the healthcare system. In order to hasten the healing of diabetic wounds, a wearable hydrogel-based triboelectric nanogenerator (BP-TENG) was created as a skin patch, which combines the beneficial effects of electrical and photothermal stimulation. The triboelectric output performance of the BPTENG was adjusted and photothermal conversion capacity was brought about by the black phosphorus nanosheets (BPNS) doped in PVA hydrogel. In just 60 seconds, BP-TENG managed to bring the temperature of the mouse wound region up to 48.6 degrees C under the irradiation of 808 nm near-infrared light. BP-TENG also exhibited spectral antibacterial activity against E. coli, S. aureus, and P. aeruginosa. Furthermore, BP-TENG had the ability to capture the energy from mechanical movements of mice and transform it into electrical stimulation. Under 808 nm near-infrared light irradiation, BP-TENG was shown to significantly speed the healing of skin wounds in the incision mouse model of type I diabetes. According to in vitro research, these benefits may be ascribed to photothermal and electric stimulation, which can both repair the healthy microenvironment of wound area and speed up the deposition and remodeling of collagen during the healing process of diabetic wounds. As far as we are aware, this is the first report on the creation and use of hydrogel-based TENG doped with BP for wound healing. We believe that this study improves the quality of life for patients with persistent non-healing wounds and opens up new possibilities for 2D materials and TENG in the application of wearable healthcare devices.
Symbiotic Algae–Bacteria Dressing for Producing Hydrogen to Accelerate Diabetic Wound Healing

Abstract:Oxidative stress induced by hyperglycemia or chronic inflammation can limit diabetic wound healing, resulting in diabetic foot ulcers. Hydrogen has the potential to act as an antioxidant and scavenge reactive oxygen species, thereby attenuating inflammation in these chronic wounds. However, most of the reported H2 delivery systems for wound healing, including hydrogen gas, hydrogen-rich water, and hydrogen-rich saline, are very short-lived for the low solubility of hydrogen gas. Here, we introduce a hydrogen-producing hydrogel made of living Chlorella and bacteria within a cell-impermeable casing that can continuously produce hydrogen for 60 h. This microbe-hydrogel system can selectively reduce highly toxic •OH and ONOO- species and reduce inflammation. Additional experiments indicated that the microbe-hydrogel dressing could promote cell proliferation and diabetic wound healing by almost 50% at day 3. The symbiotic algae-bacteria hydrogel has excellent biocompatibility and reactive oxygen species scavenging features, indicating it has great promise for clinical use.

Abstract:Abstract Chronic diabetic wounds are at lifelong risk of developing diabetic foot ulcers owing to severe hypoxia, excessive reactive oxygen species (ROS), a complex inflammatory microenvironment, and the potential for bacterial infection. Here we develop a programmed treatment strategy employing live Haematococcus (HEA). By modulating light intensity, HEA can be programmed to perform a variety of functions, such as antibacterial activity, oxygen supply, ROS scavenging, and immune regulation, suggesting its potential for use in programmed therapy. Under high light intensity (658 nm, 0.5 W/cm 2 ), green HEA (GHEA) with efficient photothermal conversion mediate wound surface disinfection. By decreasing the light intensity (658 nm, 0.1 W/cm 2 ), the photosynthetic system of GHEA can continuously produce oxygen, effectively resolving the problems of hypoxia and promoting vascular regeneration. Continuous light irradiation induces astaxanthin (AST) accumulation in HEA cells, resulting in a gradual transformation from a green to red hue (RHEA). RHEA effectively scavenges excess ROS, enhances the expression of intracellular antioxidant enzymes, and directs polarization to M2 macrophages by secreting AST vesicles via exosomes. The living HEA hydrogel can sterilize and enhance cell proliferation and migration and promote neoangiogenesis, which could improve infected diabetic wound healing in female mice.

Microenvironmental pH modulating oxygen self-boosting microalgal prodrug carboxymethyl chitosan/hyaluronic acid/puerarin hydrogel for accelerating wound healing in diabetic rats

Abstract:Chronic diabetic wounds are characterized by a range of detrimental features, including hypoxia, elevated levels of reactive oxygen species, impaired angiogenesis, chronic inflammation, and an increased susceptibility to bacterial infections. We have developed an innovative multifunctional hydrogel system based on carboxymethyl chitosan, which incorporates embedded microalgae PCC7942 along with hyaluronic acid and puerarin, termed PCC7942@carboxymethyl chitosan/hyaluronic acid/puerarin hydrogel. It demonstrated outstanding capabilities in exudate absorption, mechanical flexibility, hemostatic action, and antibacterial efficacy. Furthermore, it effectively modulated the pH of wound microenvironment through the hydrolysis of amide bonds, thereby establishing a favorable low-pH microenvironment. Microalgae in hydrogel covered in the wound exhibited stable and continuous oxygen production within 24 h, with more efficiency in dissolved oxygen penetration through skin. Furthermore, prodrugs such as hyaluronic acid and puerarin from hydrogel displayed the controlled release behavior and facilitated the fast and enhanced accumulation of drugs at wound site, thereby accelerating the process of wound healing via enhanced angiogenesis and anti-inflammation effects. In summary, the healing-promoting effect of PCC7942@carboxymethyl chitosan/hyaluronic acid/puerarin hydrogel in type 1 diabetic rats can be attributed to the synergistic effects of microalgae, hyaluronic acid, and puerarin, which collectively accelerated wound healing rate and improved the quality of wound recovery.

Abstract:Chronic wounds caused by hyperglycaemia, hypoxia and bacterial infections are common complications in diabetic patients and chronic wound repair is extremely challenging in clinical practice. A series of hydrogels QPMP with good antioxidant and antimicrobial functions were prepared based on quaternized chitosan (QCS), oxidized pullulan polysaccharide (OP), dopamine-coated polypyrrole (PPY@PDA), and Chlorella vulgaris . The Schiff base cross-linking between the quaternized chitosan (QCS) and oxidized pullulan polysaccharide (OP) constitutes the basic skeleton of the hydrogel, and imparts a certain antimicrobial ability to the hydrogel. Chlorella vulgaris continuously produced oxygen under light conditions to relieve wound hypoxia and promote wound healing. The incorporation of PPY@PDA gave the hydrogel near-infrared (NIR) irradiation-assisted bactericidal activity and antioxidant activity, and as a conductive hydrogel, the hydrogel can be used to sense wound exudate and temperature changes, which can help to achieve the integration of diagnosis and treatment of wound healing. Most importantly, in a chronic wound model, the QPMP hydrogel was more effective in controlling the level of wound inflammation and promoting collagen deposition, angiogenesis, and early wound closure compared to the HeraDerm dressing. Therefore, this conductive oxygen-producing hydrogel is extremely beneficial for chronic wound healing in diabetes.

Abstract:Impaired wound healing is a major healthcare problem in patients with diabetes often resulting in gangrene, microbial infection and amputation of affected limb. The delay or absence in healing process arises from several abnormalities, among them chronic hypoxia is a major concern due to its associated issues such as lack of collagen deposition, epithelization, fibroplasia, angiogenesis, and resistance to infections at the wound site. To address hypoxia, delivery of oxygen at the wound site through oxygen releasing agents have been proven to be effective therapeutics. Several oxygen releasing nanoparticles such as Sodium Percarbonate (SPC), Calcium Peroxide (CPO), Hydrogen Peroxide, Magnesium Peroxide (MPO) have been investigated in wound healing application. However, the uncontrolled/burst release of these nanotherapeutic agents and its accompanied cytotoxicity pose a barrier in expediting the healing process. In this study, a Chitosan-Polyvinyl alcohol (CS-PVA) based hydrogel containing oxygen releasing nanoparticle, calcium peroxide (CPO) was constructed to provide a slow and sustained delivery of oxygen for at least 5 days. In-vitro cell culture studies with this material using fibroblast and endothelial cell line exhibited improved biocompatibility, cell viability and enhanced proliferation in comparison with the control group. Additionally, cell migration study using scratch assay method showed superior cell migration ability of our proposed materials. Furthermore, In vivo study using diabetic rat model showed accelerated wound closure rate compared to untreated control wounds.

Abstract:Living algaes (mainly Euglena gracilis, Spirulina platensis, S. elongatus PCC7942, Chlamydomonas reinhardtii, Chlorella) seeding wound dressings provide a brand‐new treatment strategy for the effective care of diabetic chronic wounds and gradually form a very flexible and efficient concept of "living material" application based on living algae to replace traditional nonliving drugs. Diabetic chronic wounds (DCWs) are a severe complication of diabetes, and current treatments present several deficiencies, including unsatisfactory efficacy and inability to continuously supply oxygen. This review highlights the recent advances in a novel therapeutic strategy that addresses these challenges: considerable biological powerful inclusions, oxygen or hydrogen and ROS production, DCW glucose consumption, chemotaxis at the wound site, highly editable, great biological safety, low cost, and readily available. Over the past decade, this strategy has shown encouraging results in improving the healing delay induced by hyperglycemia, forming a new concept of "living material" application based on living algae. This review discusses the unique advantages of algae as a treatment strategy, the applications and characteristics of different types of living algae in wound dressings, and the challenges and improvement strategies for this innovative approach.

Abstract:Chronic hypoxia and ischemia make diabetic wounds non‐healing. Cellular functions of diabetic chronic wounds are inhibited under a pathological environment. Therefore, we developed a composite hydrogel system to promote diabetic wound healing. The composite hydrogel system consists of ε‐poly‐lysine, calcium peroxide, and borosilicate glass (BG). The hydrogel supplies continuous dissolved oxygen molecules to the wound that can penetrate the skin tissue to restore normal cellular function and promote vascular regeneration. Biofunctional ions released from BGs can recruit more macrophages through neovascularization and modulate macrophage phenotypic transformation. Combining oxygen‐mediated vascular regeneration and ion‐mediated inflammatory regulation significantly accelerated diabetic wound healing. Abstract:Oxygen plays an important role in diabetic chronic wound healing by regulating various life activities such as cell proliferation, migration, and angiogenesis. Therefore, oxygen-delivering systems have drawn much attention and evolved continuously. Here, we propose that an active Chlorella vulgaris (Cv)-loaded separable microneedle (MN) can be used to control oxygen delivery, which then promotes wound healing. The Cv-loaded microneedles (CvMN) consist of a polyvinyl acetate (PVA) substrate and gelatin methacryloyl (GelMA) tips with encapsulated Cv. Once CvMN is applied to diabetic wound, the PVA basal layer is rapidly dissolved in a short time, while the noncytotoxic and biocompatible GelMA tips remain in the skin. By taking advantage of the photosynthesis of Cv, oxygen would be continuously produced in a green way and released from CvMN in a controlled manner. Both in vitro and in vivo results showed that CvMN could promote cell proliferation, migration, and angiogenesis and enhance wound healing in diabetic mice effectively. The remarkable therapeutic effect is mainly attributed to the continuous generation of dissolved oxygen in CvMN and the presence of antioxidant vitamins, gamma-linolenic acid, and linoleic acid in Cv. Thus, CvMN provides a promising strategy for diabetic wound healing with more possibility of clinical transformations.

Abstract:Diabetic wound treatment remains a major challenge due to the difficulties of eliminating bacterial biofilm and relieving wound hypoxia. To address these issues simultaneously, a multifunctional Dex-SA-AEMA/MnO2/PDA (DSAMP) hydrogel platform was developed with excellent biocompatibility and porous structure. The hydrogel could absorb the exudate, maintain humidity and permeate oxygen, which was prepared by encapsulating polydopamine (PDA) and manganese dioxide (MnO2) into Dex-SA-AEMA (DSA) hydrogel by UV irradiation. With the addition of PDA, the DSAMP hydrogel was proved to eliminate the biofilm after NIR photodynamic therapy (PTT, 808 nm) irradiation at 54 °C. Furthermore, in order to mitigate hypoxia wound microenvironment, MnO2 nanoparticles were added to convert the endogenous hydrogen peroxide (H2O2) into oxygen (O2, 16 mg L−1). The diabetic wound in vivo treated by DSAMP hydrogel was completely healed on 14 days. It was revealed that the DSAMP hydrogel possessed a great potential as dressing for diabetic chronic wound healing.

Enhancing diabetic wound healing with a pH-responsive nanozyme hydrogel featuring multi-enzyme-like activities and oxygen self-supply

Abstract:Diabetic wound treating remains a challenging due to bacterial infections, oxidative stress, tissue hypoxia, and high glucose levels. Herein, a multi-enzyme-like activities nanocomposite (Mo,Fe/Cu,I-Ag@GOx) was designed and anchored to a multifunctional fluorescence hydrogel. The nanozyme gel, loaded with glucose-oxidase (GOx), exhibits intrinsic GOx, peroxidase (POD)-, oxidase (OXD)-, catalase (CAT)- and superoxide dismutase (SOD)-like activities with pH-switchable glucose-initiated cascade reaction for diabetic wound healing. In the first cascade-reaction, initiated by GOx, the nanozyme gel catalyzes glucose and O2 into gluconic acid and H2O2 to further generate superoxide anion radical (O2·-) and hydroxyl radicals (·OH) to eradicate bacteria. In the second cascade-reaction, as the wound pH changes alkalescent microenvironment, the nanozyme gel simulates SOD to transform O2·- into O2 and H2O2, and then decomposes endogenous and exogenous H2O2 into O2 via CAT-like activity to reduce oxidative stress and alleviate hypoxia. The gel by calcium ion (Ca2+) cross-linked sodium alginate (SA) and chitosan (CS) containing nanozyme was constructed with injectability, adhesion and fluorescence properties, as well as beneficial biocompatible. Importantly, the water/alcohol solubility of the nanozyme gel allows it to be used as a dressing without causing secondary injury to the wound. The multifunctional fluorescence hydrogel exhibits efficiently promote pro-angiogenesis and bacteria-infected wound healing.
Zinc alginate hydrogels with embedded RL-QN15 peptide-loaded hollow polydopamine nanoparticles for diabetic wound healing therapy

Abstract:Chronic wound healing remains a considerable clinical challenge worldwide. We previously identified a pro-regenerative agent, i.e., hollow dopamine nanoparticles (HPDA) loaded with potent pro-healing peptide RL-QN15 (HPDAlR), with significant skin wound healing activity. In the current study, in consideration of clinical application, we successfully prepared a new zinc alginate hydrogel embedded with HPDAlR (HPDAlR&ZA) to treat diabetic wounds. HPDAlR&ZA exhibited no obvious toxicity against keratinocytes, human umbilical vein endothelial cells (HUVECs), human skin fibroblasts (HSFs), and mice. HPDAlR&ZA significantly promoted keratinocyte, HUVEC, and HSF proliferation, as well as HUVEC migration and angiogenesis. HPDAlR&ZA also modulated the expression of cytokines from macrophages. Diabetic mouse full-thickness skin wound and diabetic patient in vitro skin wound models showed that HPDAlR&ZA resisted the inflammatory response by rapidly polarizing M1 macrophages to M2 macrophages, thereby reconstructing blood supply, increasing collagen deposition, and accelerating diabetic wound repair and skin regeneration. In summary, HPDAlR&ZA is a biodegradable hydrogel wound dressing and an efficient treatment strategy for diabetic wound repair, with strong anti-oxidant, anti-inflammatory, and pro-angiogenic features.

Abstract:Accelerating the diabetes-related chronic wounds healing is a long-sought-after goal in diabetes management. However, the therapeutic strategies based on antibiotic or catalysts still face great challenges to break the limitations of antimicrobial resistance, low H2O2 and the blocking effect of bacterial biofilm on antibiotic/catalysts penetration. Herein, we reported a glucose biofuel cell-powered and drug-free antibacterial patch, which consisted of MAF-7 protected glucose oxidase/horseradish peroxidase anode and horseradish peroxidase cathode, for treating the diabetic wounds. This self-powered patch could take the high blood glucose as the fuel to generate electricity and abundant reactive oxygen species (ROS) in situ, synergistically regulating the local hyperglycemia and breaking the limitation of insufficient ROS caused by low H2O2 level. In particular, the electric field created by the GBFC could drive the negatively-charged bacteria to adhere firmly on the electrode surface. As a result, the ROS produced in situ on the electrodes was localized to the bacteria, realizing precise sterilization. In vivo experiments confirmed that this self-powered patch enabled the wounds on diabetic mice to take a mere 10 days to eliminate inflammation and form mature skin with new hair follicles, demonstrating its great potential in treating bacteria-infected diabetic wounds.

Abstract:Non‐healing wound is a common complication of diabetic patients associated with high morbidity and mortality. Engineered therapeutic hydrogels have enviable advantages in tissue regeneration, however, they are suboptimal for the healing of diabetic wounds characterized by reactive oxygen species (ROS) accumulation and chronic hypoxia. Here, we present a unique biological metabolism‐inspired hydrogel for ameliorating this hostile diabetic microenvironment. Consisting of natural polymers (hydrazide modified hyaluronic acid and aldehyde modified hyaluronic acid) and a metal‐organic frameworks derived catalase‐mimic nanozyme (ε‐polylysine coated mesoporous manganese cobalt oxide), the engineered nanozyme‐reinforced hydrogels can not only capture the endogenous elevated ROS in diabetic wounds, but also synergistically produce oxygen through the ROS‐driven oxygen production ability. These fascinating properties of hydrogels protected skin cells (e.g., keratinocytes, fibroblasts, and vascular endothelial cells) from ROS and hypoxia‐mediated death and proliferation inhibition. Diabetic wounds treated with the nanozyme‐reinforced hydrogels highlighted the potential of inducing the macrophages polarization from pro‐inflammatory phenotype (M1) to anti‐inflammatory subtype (M2). The hydrogel dressings demonstrated a prominently accelerated healing rate as featured by alleviating the excessive inflammatory, inducing efficiently proliferation, re‐epithelialization, collagen deposition, and neovascularization. This work provides an effective strategy based on nanozyme‐reinforced hydrogel as a ROS‐driven oxygenerator for enhancing diabetic wound healing. This article is protected by copyright. Abstract:The treatment of diabetic wounds remains a major challenge in clinical practice, with chronic wounds characterized by multiple drug-resistant bacterial infections, angiopathy, and oxidative damage to the microenvironment. Herein, a novel in situ injectable HA@MnO2 /FGF-2/Exos hydrogel is introduced for improving diabetic wound healing. Through a simple local injection, this hydrogel is able to form a protective barrier covering the wound, providing rapid hemostasis and long-term antibacterial protection. The MnO2 /ε-PL nanosheet is able to catalyze the excess H2 O2 produced in the wound, converting it to O2 , thus not only eliminating the harmful effects of H2 O2 but also providing O2 for wound healing. Moreover, the release of M2-derived Exosomes (M2 Exos) and FGF-2 growth factor stimulates angiogenesis and epithelization, respectively. These in vivo and in vitro results demonstrate accelerated healing of diabetic wounds with the use of the HA@MnO2 /FGF-2/Exos hydrogel, presenting a viable strategy for chronic diabetic wound repair.

Abstract:Interaction between bacterial species is through quorum sensing (QS), a process that regulates and coordinates genes in virulence activities and biofilm formation. QS contributes to the slowdown of tissue repair during the wound healing process by inducing chronic inflammation of the wound. Here, we developed a multifunctional QS inhibitor based on a bioactive hydrogel system to interrupt the QS of bacteria, relieve hypoxia, and destroy biofilms, and thus accelerate the healing of infected wounds in diabetic mice. We loaded berberine (BBR, a QS inhibitor, and antibacterial agent) into a natural living microalgae Spirulina platensis (SP) to form a bioactive hydrogel (BBR@SP gel) in combination with carboxymethyl chitosan/sodium alginate. Under laser irradiation, the BBR@SP gel could constantly release BBR and produce reactive oxygen species, resulting in a synergistic QS inhibition against methicillin-resistant Staphylococcus aureus (MRSA) combined chemo-photodynamic therapy. The BBR@SP gel also suppresses and destroys biofilm formation and down-regulates the expression of virulence factors. We found that the BBR@SP gel could accelerate MRSA-infected diabetic wound healing by promoting angiogenesis, skin regeneration, and suppressing the inflammatory response. Our work presents an innovative antimicrobial strategy that eliminates drug-resistant bacteria by synergistic chemo-photodynamic killing effect, relieving biofilm hypoxia, and blocking QS simultaneously, which may open up new prospects in solving antimicrobial drug resistance and combating biofilm-related infections.

Abstract:Infection and oxidative stress seriously hinder the healing of diabetic wounds, resulting in various serious health and clinical problems. Herein, a sustainable biological hydrogen (H2)-producing hyaluronic acid-based hydrogel patch (HAP-Chl) was constructed by loading an imidazolium-based poly(ionic liquid) (PIL) flocculated live Chlorella as a diabetic wound dressing. The PIL can flocculate Chlorella through electrostatic interactions between PIL and Chlorella to form Chlorella agglomerates, endowing the Chlorella in the central agglomerates with the ability to continuously produce H2 for 24 h under mild conditions. Combining the membrane disruption-related bactericidal mechanism of PIL and the antioxidant properties of the produced H2, HAP-Chl was determined to be antibacterial and antioxidant. In addition to exhibiting biocompatible and nontoxic activities, subsequent Staphylococcus aureus-infected chronic wound studies revealed that HAP-Chl is capable of promoting the healing of chronic wounds by effectively killing bacteria, reducing extensive ROS, relieving inflammation, and promoting the deposition of mature collagen and angiogenesis. This study provides a new strategy for constructing an in situ sustainable H2-producing hydrogel, enabling the formation of novel antibacterial and antioxidant material platforms with potential for wound dressing applications.

Abstract:Exposed chronic wounds are usually covered by hypoxic tissues accompanied with necrosis, persistent inflammation and anaerobic infections. Since atmospheric oxygen air can only penetrate about 0.3 mm tissues, meanwhile oxygen from the circulation is difficult to reach chronic wounds through the destroyed blood vessels, a solution to deliver oxygen locally and permeably is urgently needed. Herein we report a technique to reform traditional gel-based wound dressings by adding lyophilized oxygen encapsulated nanoparticles, which can deliver dissolved oxygen locally into wound surface. This delivery technique can potentially evaluate the therapeutic effects on hypoxic epithelial, endothelial and fibroblasts in vitro. Further experiments confirm the effects on both open wounds bearing and flap transplanted diabetic mice models. Considering its biocompatibility, effectiveness and practicality, we believe our hydrogel has significant transformation value to care and accelerate the healing of various clinical wounds, especially chronic wound.

Abstract:A novel oxygen generating system HA‐L‐NB/PFE@cp by combining conjugated polymers/chloroplast composite nanoparticles with photo‐triggered hydrogels is constructed. The enhanced photosynthesis of composite nanoparticles effectively relieves hypoxia, promotes cell migration, and favors photodynamic antibacterial. Hydrogels can quickly anchor on tissues. Both in vitro and in vivo assays demonstrate excellent chronic wounds healing effect. Diabetic chronic wounds are characterized by local hypoxia, impaired angiogenesis, and bacterial infection. In situ, self‐supply of dissolved oxygen combined with the elimination of bacteria is urgent and challenging for chronic nonhealing wound treatment. Herein, an oxygen‐generating system named HA‐L‐NB/PFE@cp involving biological photosynthetic chloroplasts (cp)/conjugated polymer composite nanoparticles (PFE‐1‐NPs@cp) and light‐triggered hyaluronic acid‐based (HA‐L‐NB) hydrogel for promoting diabetic wound healing is introduced. Briefly, conjugated polymer nanoparticles (PFE‐1‐NPs) possess unique light harvesting ability, which accelerates the electron transport rates in photosystem II (PS II) by energy transfer, elevating photosynthesis beyond natural chloroplasts. The enhanced release of oxygen can greatly relieve hypoxia, promote cell migration, and favor antibacterial photodynamic therapy. Additionally, the injectable hydrogel precursors are employed as a carrier to deliver PFE‐1‐NPs@cp into the wound. Under light irradiation, they quickly form a gel by S‐nitrosylation coupling reaction and in situ anchor on tissues through amine‐aldehyde condensation. Both in vitro and in vivo assays demonstrate that the oxygen‐generating system can simultaneously relieve wound hypoxia, eliminate bacteria, and promote cell migration, leading to the acceleration of wound healing. This study provides a facile approach to develop an enhanced oxygen self‐sufficient system for promoting hypoxic tissue, especially diabetic wound healing.
