Effects of long-term exposure to sulfamethoxazole on the sediments in sewage pipe: Extracellular polymeric substance, microbial communities, and antibiotic resistance genes

Abstract:Long-term exposure to antibiotics in sewage pipelines might change the microbial community, and affect the spread of antibiotic resistance genes (ARGs). In this study, the effects of sulfamethoxazole (SMX) exposure on the microbial community in a smaller distance downstream, as well as the changes of water quality and ARGs in the sewage pipe were studied, and the propagation of ARGs in pipeline sediments under SMX was revealed. The results showed that under SMX exposure, the relative abundances of Dechloromonas , unclassified__o_PeM15 , and Nakamurell associated with phosphorus transformation, Clostridium_sensu_stricto_1 , Trichococcus associated with nitrogen transformation and Thiobacillus , a sulfur oxidizing bacterium, were all significantly different (P < 0.01), causing changes in the sewage quality of the pipeline. With SMX exposure concentration increasing, the content of TB-EPS in the sediments increased from 7.64 mg/g VSS at 0 d to 24.35 mg/g VSS at 113 d, and the enhancement of tryptophan-like characteristic fluorescence peak indicated that the microorganisms secreted more proteins for self-protection. SMX exposure in the pipeline increased the relative abundance of the ARGs and intl1 . Intl1 was significantly positively correlated with most ARGs (P < 0.05), indicating that SMX promoted the horizontal transfer of ARGs, and ARGs existed independently of their respective antibiotic inducers. Correlation analysis showed that g__Subgroup_10 , g__unclassified_o__RBG-13–54–9 , g__unclassified_o__SBR1031 , g__unclassified_f__Caldilineaceae , g__Rubellimicrobium , g__unclassified_f__JG30-KF-CM45 and g__Bacillus were the main potential hosts of ARGs. This study helps to understand the effect of antibiotic contamination on the ARGs transmission, and assess the transmission risk of ARGs in sewage pipes.
