Abstract:In the present contribution the fabrication and characterization of functionalized gold nanospheres of uniform shape and controlled size is reported. These nano-objects are intended to be used as Surface Enhanced Raman Spectroscopy (SERS) sensors for in-vitro cellular uptake and localization. Thiophenol was used as molecular reporter and was bound to the Au surface by a chemisorption process in aqueous solution. The obtained colloidal solution was highly stable and no aggregation of the single nanospheres into larger clusters was observed. The nanoparticles were incubated in human prostatic cells with the aim of developing a robust, SERS-based method to differentiate normal and tumor cell lines. SERS imaging experiments showed that tumor cells uptake considerably larger amounts of nanoparticles in comparison to normal cells (up to 950% more); significant differences were also observed in the uptake kinetics. This largely different behaviour might be exploited in diagnostic and therapeutic applications.
Multiparametric Assessment of Gold Nanoparticle Cytotoxicity in Cancerous and Healthy Cells: The Role of Size, Shape, and Surface Chemistry

Abstract:In recent years, we and others have become interested in evaluating the use of surface-enhanced Raman scattering (SERS) tags for early cancer detection and in designing new approaches to demonstrate the applicability of this spectroscopic technique in the clinic. SERS-based imaging in particular offers ultra sensitivity up to the single molecule, multiplexing capability, and increased photostability and has been shown to outperform fluorescence. However, to employ SERS tags for early cancer detection, it is important to understand their interaction with cells and determine their cytotoxicity. We have been particularly interested for quite some time in determining if and how gold nanostars, which have been demonstrated as outstanding SERS enhancing substrates, can be safely employed in living systems and translated to the clinic. In this study, we carried out a multiparametric in vitro study to look at the cytotoxicity and cellular uptake of gold nanoparticles on human glioblastoma and human dermal fibroblast cell lines. Cytotoxicity was evaluated by incubating cells with three different morphologies of AuNPs, namely nanospheres, nanorods, and nanostars, each having three different surface chemistries (cetyltrimethylammonium bromide (CTAB), poly(ethylene glycol) (PEG), and human serum albumin (HSA)). Our results showed that the surface chemistry of the nanoparticles had predominant effects on cytotoxicity, and the morphology and size of the nanoparticles only slightly affected cell viability. CTAB-coated particles were found to be the most toxic to cells, and PEGylated nanostars were determined to be the least toxic. Caspase-3 assay and LDH assay revealed that cell death occurs via apoptosis for cancerous cells and via necrosis for healthy ones. Cellular uptake studies carried out via TEM showed that the particles retain their shape even at long incubation times, which may be beneficial for in vivo SERS-based disease detection. Overall, this study provides valuable information on gold-nanoparticle-induced cytotoxicity that can be leveraged for the development of safe and effective nanoparticle-based therapeutic and diagnostic systems.
Bioorthogonal SERS Nanoprobes for Mulitplex Spectroscopic Detection, Tumor Cell Targeting, and Tissue Imaging.

Abstract:A surface-enhanced Raman scattering (SERS) technique shows extraordinary features for a range of biological and biomedical applications. Herein, a series of novel bioorthogonal SERS nanoprobes were constructed with Gold nanoflower (AuNF) and Raman reporters, the signals of which were located in a Raman-silent region of biological samples. AS1411 aptamer was also co-conjugated with AuNF through a self-assembled monolayer coverage strategy. Multiplex SERS imaging using these nanoprobes with three different bioorthogonal small-molecule Raman reporters is successfully achieved with high multiplexing capacity in a biologically Raman-silent region. These Raman nanoprobes co-conjugated with AS1411 showed high affinity for tumor cells with overexpressed nucleolin and can be used for selective tumor cell screening and tissue imaging.

Abstract:The enhancement effect of 3-mercaptopropionic acid capped gold nanoporticles (NPs) in drug delivery and as biomarkers of drug-resistant cancer cells has been demonstrated through fluorescence microscopy and electrochemical studies. The results of cell viability experiments and confocal fluorescence microscopy studies illustrate that these functionalized Au NPs could play on important role in efficient drug delivery and biomarking of drug-resistant leukemia K562/ADM cells. This could be explore as a; novel strategy to inhibit multidrug resistance in targeted tumor cells and as a sensitive method for the early diagnosis of certain cancers. Our observations also indicate that the interaction between the functionalized Au NPs and biologically active molecules on the surface of leukemia cells may contribute the observed enhancement in cellular drug uptake.
Robust Encapsulation of Biocompatible Gold Nanosphere Assemblies for Bioimaging via Surface Enhanced Raman Scattering

Abstract:The use of easily fabricated spherical gold nanoparticles as surface‐enhanced Raman scattering (SERS) contrast agents for bioimaging is limited by various stability, biocompatibility, and optical issues. A strategy for the preparation of gold nanoparticle assemblies encapsulated with Raman‐active molecules within a biocompatible polymer shell is presented, as well as their detection in cells by 3D SERS mapping.Controlled assembly of gold nanoparticles (AuNPs) into clusters is a promising avenue for the development of sensitive bioimaging and diagnostic tools based on surface‐enhanced Raman scattering (SERS). However, several challenges, such as biocompatibility or colloidal and structural stability in biological environments, remain before AuNPs can be used as a tool for in vivo bioimaging. A versatile strategy for the preparation of colloidally stable and biocompatible AuNP clusters (AuNPCs) is introduced with high SERS signals, which are used as SERS contrast bioimaging agents (SERS tags). By tuning the ligand ratio of Raman reporter molecules to stabilizing polymer on the surface of each AuNP, aggregation can be carefully controlled. The resulting AuNPCs exhibit redshifted surface plasmon resonances in the near‐infrared (NIR) region, as well as distinct electromagnetic hotspots that give rise to SERS analytical enhancement factors above 104, compared to non‐clustered spherical AuNPs. Thanks to the protective polymer shell, high levels of cellular uptake with low cytotoxicity are observed, allowing 3D SERS mapping of cells with sufficiently high spatial resolution to detect AuNPCs within intracellular organelles.

Abstract:In this study, we describe a 2D-SERS sensor obtained by deposition of spherical gold nanoparticles (AuNPs) onto a suitably functionalized metal surface. Morphological analysis of the SERS surface by SEM and AFM demonstrated a uniform and stable distribution of the active nanoparticles. Following p-mercaptoaniline (pMA) functionalization, the sensor was characterized by co-localized Raman measurements, demonstrating a significant enhancement in Raman signals with homogeneous SERS activity across the entire sampled area. The as-prepared SERS sensor was demonstrated to be suitable for Therapeutic Drug Monitoring (TDM) of 6-mercaptopurine (6-MP), exhibiting a linear correlation between analyte concentration and SERS intensity in the range 5 – 20 μM. This work highlights the potential of 2D-SERS sensors for hypersensitive and accurate analytical measurements, particularly in the biomedical field.

Abstract:Gold Fluorescence Nanoparticles have revealed the great promise in transforming the drug discovery, photodynamic treatment, and cancer detection, among other biological treatments. The integration of these gold nanoparticles into SERS techniques has unlocked a new scope of possibilities, offering enhanced sensitivity, efficient recognition, and specificity in detecting large/small molecular structures. Through their unique properties, gold fluorescence nanoparticles hold tremendous potential to address the crucial challenges in the biomedical field, that promising the significant advancements in diagnosis and treatment methodologies. Nanotechnology has emerged as a pivotal tool in biomedical research, particularly in developing advanced sensing platforms for disease diagnosis and therapeutic monitoring. Since gold nanoparticles are biocompatible and have special optical characteristics, they are excellent choices for surface‐enhanced Raman scattering (SERS) sensing devices. Integrating fluorescence characteristics further enhances their utility in real‐time imaging and tracking within biological systems. The synergistic combination of SERS and fluorescence enables sensitive and selective detection of biomolecules at trace levels, providing a versatile platform for early cancer diagnosis and drug monitoring. In cancer detection, AuNPs facilitate the specific targeting of cancer biomarkers, allowing for early‐stage diagnosis and personalized treatment strategies. The enhanced sensitivity of SERS, coupled with the tunable fluorescence properties of AuNPs, offers a powerful tool for the identification of cancer cells and their microenvironment. This dual‐mode detection not only improves diagnostic accuracy but also enables the monitoring of treatment response and disease progression. In drug detection, integrating AuNPs with SERS provides a robust platform for identifying and quantifying pharmaceutical compounds. The unique spectral fingerprints obtained through SERS enable the discrimination of drug molecules even in complex biological matrices. Furthermore, the fluorescence property of AuNPs makes it easier to track medication distribution in real‐time, maximizing therapeutic effectiveness and reducing adverse effects. Furthermore, the review explores the role of gold fluorescence nanoparticles in photodynamic therapy (PDT). By using the complementary effects of targeted drug release and light‐induced cytotoxicity, SERS‐guided drug delivery and photodynamic therapy (PDT) can increase the effectiveness of treatment against cancer cells. In conclusion, the utilization of gold fluorescence nanoparticles in conjunction with SERS holds tremendous potential for revolutionizing cancer detection, drug analysis, and photodynamic therapy. The dual‐mode capabilities of these nanomaterials provide a multifaceted approach to address the challenges in early diagnosis, treatment monitoring, and personalized medicine, thereby advancing the landscape of biomedical applications.

Gold-Etched Silver Nanowire Endoscopy: Toward a Widely Accessible Platform for Surface-Enhanced Raman Scattering-Based Analysis in Living Cells

Abstract:Recently, our group introduced the use of silver nanowires (AgNWs) as novel non-invasive endoscopic probes for detecting intracellular Raman signals. This method, although innovative and promising, relies exclusively on the plasmonic waveguiding effect for signal enhancement. It, therefore, requires sophisticated operational tools and protocols, drastically limiting its applicability. Herein, an advanced strategy is offered to significantly enhance the performance of these endoscopic probes, making this approach widely accessible and versatile for cellular studies. By uniformly forming gold structures on the smooth AgNW surface <i>via</i> a galvanic replacement reaction, the density of the light coupling points along the whole probe surface is drastically increased, enabling high surface-enhanced Raman scattering (SERS) efficiency upon solely focusing the excitation light on the gold-etched AgNW. The applicability of these gold-etched AgNW probes for molecular sensing in cells is demonstrated by detecting site-specific and high-resolved SERS spectra of cell compartment-labeling dyes, namely, 4′,6-diamidino-2-phenylindole in the nucleus and 3,3′-dioctadecyloxacarbocyanine on the membrane. Abstract:Precise detection and effective treatment are crucial to prolong cancer patients' lives. Surface-enhanced Raman scattering (SERS) imaging coupled with photothermal therapy has been considered a precise and effective strategy for cancer theranostics. Nevertheless, Raman reporters employed in the literature usually possessed multiple shift peaks in the fingerprint region, which are overlapped with background signals from endogenous biological molecules. Herein, we fabricated a new kind of bioorthogonal Raman reporter and aptamer functionalized SERS nanotags. The SERS nanotags demonstrated a strong Raman signal at 2205 cm(-1) in the biologically Raman-silent region and recognized MCF-7 breast cancer cells for Raman imaging with high specificity. Laser irradiation induced serious toxicity of MCF-7 cells due to the excellent photothermal capability of the SERS nanotags. After intravenous administration of the SERS nanotags, tumor Raman spectral detection and mapping in living mice were successfully achieved. Further in vivo antitumor experiments manifested that the aptamer-modified SERS nanotags significantly restrained tumor growth after laser irradiation with 99% inhibition rate and good biocompatibility. These results clearly revealed that the SERS nanotags could serve as a novel and precise theranostic platform for in vivo cancer diagnosis and photothermal therapy.
Raman reporter-coated gold nanorods and their applications in multimodal optical imaging of cancer cells

Abstract:We report the preparation of a kind of surface-enhanced Raman scattering (SERS) tags and explore their applications in multifunctional optical imaging of cancer cells. The proposed nanoparticles (SERS tags) are prepared by connecting dye molecules directly onto the surfaces of gold nanorods through Au–S or Au–N interactions. The dye molecules are used as Raman reporters, while gold nanorods are used as enhanced materials due to their localized surface plasmon resonance effect. Multilayered polymers are further coated onto the surfaces of the nanoparticles to reach better stability and biocompatibility. Gold nanorods with different aspect ratios and different dye molecules conjugated are compared in order to achieve the diversity of SERS tags and find out the optimized condition of SERS tags with the highest signal intensity. Our experiments show that the resulting nanoparticles, which are uptaken by cancer cells, can provide not only dark field cells images but also multiplexing SERS images. Abstract:A novel, high-sensitivity, biocompatible SERS tag with core-shell structure based on gold nanoparticles containing alkynyl molecule core -silver nanoparticle satellites shell was fabricated for the first time to be used for live cancer cells Surface enhanced Raman scattering (SERS) imaging. (E)-2-((4-(phenylethynyl)benzylidene) amino) ethanethiol (PBAT) synthesized facilely in our lab is the Raman-silence region reporter which is advantage for bioorthogonal SERS cell imaging. In order to enhance the intensity of the Raman tags for live cancer cell imaging, a series of news measures have been adopted. Firstly, reporter molecules of the PBAT were added twice, which is embedded in the gold core with the reduction of tetrachloroaurate and then PBAT is conjugated again on disperse gold nanoparticles (PBAT-Au). Furthermore, numerous Ag nanoparticles self-assembly were densely arranged around PBAT-Au core surface (PBAT- Au@Ag), just like a circle of satellites cluster, which produce obvious "hot spots" effects enhancing the signal of the Raman tags enormously. Finally, Bovine serum albumin (BSA) and polydopamine (PDA) coated on the PBAT- Au@Ag successively, defined as (PBAT-Au@Ag@BSA@PDA), which make as-synthesized nanocomposites own features of bio-compatibility and facilitates antibody modification. Compared with Au@PBAT@PDA, PBAT-Au@Ag@BSA@PDA with core-shell satellites structure showed 10-fold increase in the Raman signals intensity. Moreover, PBAT-Au@Ag@BSA@PDA nanocomposites were successfully applied in the Raman imaging of human glioma cells (U251) by the recognition of the anti-epidermal growth factor receptor (EGFR). All experimental results demonstrated that the nanocomposites have high value and huge potential application in the live cancer cells imaging and biomedical diagnostics in the near future.
SERS Nanoprobes in Biologically Raman Silent Region for Tumor Cell Imaging and in Vivo Tumor Spectral Detection in Mice

Abstract:Surface enhanced Raman scattering (SERS) is widely used in biological applications due to its high sensitivity and deep tissue penetration. Most SERS nanoprobes with fluorophore attachments have Raman signals between 1400 and 1700 cm −1 . Here, a series of SERS nanoprobes anchored with alkyne moieties in the biologically Raman‐silent region through CAu bond formation is prepared. To achieve target‐specific recognition, two nucleic acid aptamers (AS1411 and MUC1) and two control oligonucleotides (C‐con and T‐con) with the same alkyne moiety are also attached on the Au surface, respectively. Both aptamer‐charged SERS nanoprobes successfully target MCF‐7 cells, which could possibly achieve cross‐check of target cells and overcome the possible false positive results. And the limit of detection reaches up to five cancer cells, which has great potential in circulating tumor cells detection. Further in vivo studies indicate that both SERS nanoprobes are successfully employed to target tumors in living mice with an obvious signal (≈2205 cm −1 ) at the Raman silent region after tail intravenous injection for the first time.
Stimuli Responsive Molecular Exchange of Structure Directing Agents on Gold Nanobipyramids: Cancer Cell Detection and Synergistic Therapeutics

Abstract:Surface-engineered gold nanoparticles have been considered as versatile systems for theranostics applications. Moreover, surface covering or stabilizing agents on gold nanoparticles especially gold nanobipyramids (AuNBPs) provides an extra space for cargo molecules entrapment. However, it is not well studied yet and also the preparation of AuNBPs still remains dependent largely on cetyltrimethylammonium bromide (CTAB), a cytotoxic surfactant. Therefore, the direct use of CTAB stabilized nanoparticles is not recommended for cancer theranostics applications. Herein, we address an approach of dodecyl ethyl dimethylammonium bromide (DMAB) as biocompatible structure directing agent for AuNBPs, which also accommodate anticancer drug doxorubicin (45%), an additional chemotherapeutics agent. Upon near-infrared light (NIR, 808 nm) exposure, engineered AuNBPs exhibit (i) better phototransduction (51 °C) due to NIR absorption ability (650-900 nm), (ii) photo triggered drug release (more than 80%), and (iii) synergistic chemophototherapy for breast cancer cells. Drug release response has been evaluated in tumor microenvironment conditions (84% in acidic pH and 80% at high GSH) due to protonation and high affinity of thiol binding with AuNBPs followed by DMAB replacement. Intracellular glutathione (GSH, 5-7.5 mM) replaces DMAB from AuNBPs, which cause easy aggregation of nanoparticles as corroborated by colorimetric shifts, suggesting their utilization as a molecular sensing probe of early stage cancer biomarkers. Our optimized recipe yield is monodisperse DMAB-AuNBPs with ∼90% purity even at large scales (500 mL volume per batch). DMAB-AuNBPs show better cell viability (more than 90%) across all concentrations (5-500 ug/mL) when directly compared to CTAB-AuNBPs (less than 10%). Our findings show the potential of DMAB-AuNBPs for early stage cancer detection and theranostics applications.

Surface Plasmon Resonance Scattering and Absorption of Biofunctionalized Gold Nanoparticles for Targeted Cancer Imaging and Laser Therapy

Abstract:The localized surface plasmon resonance of gold nanoparticles enables them to be excellent light scattering sensing reagents and efficient lignt-heat convertors for potential diagnostics and therapeutics. In this work, gold nanoparticles of 15 nm in size were synthesized and conjugated with a kind of arginine-glycine-aspartic acid peptide (RGD) to target the cancer cells. Under a conventional dark field microscope, the scattering images from normal and cancer cells are very different. Only a few of gold nanoparticles bind to the membrane of normal cells due to nonspecific interaction. Yet for cancer cells, the concentration of gold nanoparticles inside cancer cells is much higher than that inside normal cells, showing a strong light scattering signal. By exposing these cells to an external laser irradiation, cancer cells were selectively destructed because of the heat conversion of gold nanoparticles inside them, but normal cells still kept alive due to the absence of gold nanoparticles in them. These results incidated that gold nanoparticles might be potential contrast reagents for targeted cancer imaging and laser therapy.

Abstract:This paper presents the fabrication and characterization of new gold-silver core-shell nanoparticles labeled with para-mercaptobenzoic acid (4MBA) molecules and demonstrates their use as SERS-nanotags with ultra-bright traceability inside cells and ability to convey spectrally-coded information about the intracellular pH by means of Surface-Enhanced Raman Spectroscopy (SERS). Unlike to previous reported studies, our fabrication procedure includes in the first step the synthesis of chitosan-coated gold nanoparticles as a seed material with subsequent growing of a silver shell. The bimetallic core-shell structure is revealed by transmission electron microscopy (TEM), high-angle annular dark field scanning transmission electron microscopy (HAADF-STEM), energy-dispersive X-ray elemental mapping (EDX) and the presence of two interacting localized surface plasmon resonance (LSPR) modes in UV-Vis extinction spectrum. The high SERS activity and sensitivity of as fabricated 4MBA-chit-Au-AgNPs nano-constructs to different pH in solution is investigated under 532 and 633 nm laser lines excitation. Next, in view of future studies in cancer diagnosis, the in vitro antiproliferative effects of SERS-nanotags against human ovarian adenocarcinoma cells (NIH:OVCAR-3) are evaluated. The capacity to operate as bright SERS nanotags with precise localization at a single cell level as well as intracellular pH indicators is clearly demonstrated by performing cell imaging under scanning confocal Raman microscopy.

Abstract:With the development of precision medicine, antigen/antibody-targeted therapy has brought great hope to tumor patients; however, the migration of tumor cells, especially a small number of cells flowing into blood or other tissues, remains a clinical challenge. In particular, it is difficult to use functional gold nanomaterials for targeted clinical tumor diagnosis while simultaneously obtaining stable and highly sensitive Raman signals. Therefore, we developed a detection method for functional Au Nanostars (AuNSs) with dual signal enhancement that can specifically track location and obtain high-intensity surface-enhanced Raman scattering (SERS) signals. First, AuNSs with specific optical properties were synthesized and functionalized. The Raman dye 4-mercapto-hydroxybenzoic acid and polyethylene glycol were coupled with the tumor marker, epidermal growth factor receptor, to obtain the targeted SERS probes. In addition, a detection chip was prepared for Raman detection with physical enhancement, exhibiting a 40-times higher signal intensity than that of quartz glass. This study combines physical enhancement and SERS enhancement technologies to achieve dual enhancement, enabling the detection of a highly sensitive and stable Raman signal; this has potential clinical value for antigen/antibody-targeted tumor diagnosis and treatment.

Abstract:Single cell analysis is essential for understanding the heterogeneity, behaviors of cells, and diversity of target analyte in different subcellular regions. Nucleolin (NCL) is a multifunctional protein that is markedly overexpressed in most of the cancer cells. The variant expression levels of NCL in subcellular regions have a marked influence on cancer proliferation and treatments. However, the specificity of available methods to identify the cancer biomarkers is limited because of the high level of subcellular matrix effect. Herein, we proposed a novel technique to increase both the molecular and spectral specificity of cancer diagnosis by using aptamers affinity based portable nanopipette with distinctive surface-enhanced Raman scattering (SERS) activities. The aptamers-functionalized gold-coated nanopipette was used to capture target, while p-mercaptobenzonitrile (MBN) and complementary DNA modified Ag nanoparticles (AgNPs) worked as Raman reporter to produce SERS signal. The SERS signal of Raman nanotag was lost upon NCL capturing via modified DNA aptamers on nanoprobe, which further helped to verify the specificity of nanoprobe. For proof of concept, NCL protein was specifically extracted from different cell lines by aptamers modified SERS active nanoprobe. The nanoprobes manifested specifically good affinity for NCL with a dissociation constant Kd of 36 nM and provided a 1000-fold higher specificity against other competing proteins. Furthermore, the Raman reporter moiety has a vibrational frequency in the spectroscopically silent region (1800-2300 cm-1) with a negligible matrix effect from cell analysis. The subcellular localization and spatial distribution of NCL were successfully achieved in various types of cells, including MCF-7A, HeLa, and MCF-10A cells. This type of probing technique for single cell analysis could lead to the development of a new perspective in cancer diagnosis and treatment at the cellular level.
High Specific Detection and Near-Infrared Photothermal Therapy of Lung Cancer Cells with High Sers Active Aptamer-Silver-Gold Shell-Core Nanostructures

Abstract:Lung cancer is the leading cause of cancer death worldwide. Its early detection is of paramount importance for diagnosis, classification, treatment, and improvement of survivorship. However, current methods are not sensitive enough to detect lung cancer in its nascent stage. We reported an aptamer-Ag-Au shell-core nanostructure-based surface-enhanced Raman scattering (SERS) assay for sensitive and specific detection, and near-infrared (NIR) photothermal therapy of lung adenocarcinoma cells (A549 cells). The nanostructures target the cells with high affinity and specificity via the specific interaction between the aptamer (a 45-base oligonucleotide) and the cell, and distinguish A549 cells from other types of cancer cells (HeLa and MCF-7 cells) and subtypes of lung cancer cells (NCI-H157, NCI-H520, NCI-H1299, and NCI-H446 cells). The nanostructures have a high capability to absorb NIR irradiation and are able to perform photothermal therapy of the cells at a very low irradiation power density (0.20 W cm(-2)) without destroying the healthy cells and the surrounding normal tissues. In addition, the nanostructures exhibit a high SERS activity. Based on the SERS signal of the labeled Raman reporter (Rh6G molecules), we can specifically detect A549 cells at a very low abundance (similar to 10 cells per mL) and monitor the therapy process of the cancer cells. Therefore, this nanostructure-based SERS assay has great potential in specific recognition, sensitive detection, and effective photothermal therapy of lung cancer.
