Abstract:Eurotium cristatum is the dominant fungi species in Fuzhuan brick tea (FBT), which may be responsible for the color, taste, and associated health benefits. In this study, eight E. cristatum strains were first isolated from various brands of FBT in China. Six benzaldehyde derivatives 1 - 6 were then isolated and characterized from these FBT-derived E. cristatum, four of which were also present in the spores of E. cristatum. The anti-inflammatory activities of these benzaldehyde derivatives were evaluated in lipopolysaccharide-induced RAW264.7 cells. These compounds could inhibit the expression both of inducible nitroxide synthase and cyclooxygenase-2. Our results suggest that the benzaldehyde derivatives from E. cristatum may be at least partially responsible for the observed health benefits of FBT.

Abstract:Fu-brick tea (FBT) is predominately fermented by Eurotium Cristatum, FBT polysaccharides (FTPs) and Eurotium Cristatum extracellular polysaccharides (ECPs) are the main active substances in FBT and Eurotium Cristatum, respectively. FTPs was shown to exhibit higher levels of uronic acids, proteins, and polyphenols as compared to ECPs (p < 0.05), contributing to the superior antioxidant activity observed in FTPs. Additionally, FTPs had better water solubility and thermal stability than ECPs. Interestingly, in vitro digestive simulation revealed that FTPs and ECPs resist digestion in the stomach and small intestine. Excitingly, utilizing in vitro fermentation with feces from healthy individuals and type 2 diabetes mellitus (T2DM) patients demonstrated that FTPs and ECPs promote the production of SCFAs. Still, FTPs resulted in greater SCFAs contents than ECPs (p < 0.05). Moreover, FTPs and ECPs fermentation by T2DM patients' fecal microbiota affected different metabolomic pathways. Our findings suggested that FTPs holds great promise for application in functional foods.

Abstract:Fuzhuan brick tea is a dark tea produced using tea plant leaves, and its quality is strongly affected by Eurotium cristatum. The superior Eurotium cristatum strain could enhance functional components in Fuzhuan brick tea, improving its quality and economic value. To research the lipid-lowering substance of Eurotium cristatum in Fuzhuan brick tea from Anhua country tea production area, several wild-type Eurotium cristatum strains were screened for lovastatin production. Of all 20 wild-type strains, 6 Eurotium cristatum strains could produce lovastatin, and the H20 test strain could produce the highest lovastatin. The organ of lovastatin production was studied, and its yield in sexual propagule (ascospore and ascocarp) was the highest. Some fermentation parameters for lovastatin produced by the H20 strain were researched, and the optimum condition, such as PDA medium containing additional 8% soluble starch, 32 °C, and 120 r/min, are reported. These results are promising for developing one Fuzhuan brick tea with a high lipid-lowering function and expanding the scope of tea plant forest resource application.

Abstract:Bioassay-guided fractionation, based on inhibition of -glucosidase, of Euonymus alatus led to the isolation of a new compound, catechin lactone A (7), and seven known compounds (1-6 and 8). Their structures were established on the basis of extensive 1D- and 2D-NMR experiments. The absolute configurations of compound 7 have been determined using vibrational circular dichroism spectroscopy. All these compounds were evaluated for their inhibition activities on -glucosidase and differentiation of 3T3-L1 cells. Compounds 1, 3-5, 7 and 8 showed potent yeast -glucosidase inhibition with IC50 values of 119.1, 39.6, 31.6, 23.0, 71.8 and 102.5 mu M, respectively; compounds 3-5, 7 and 8 can remarkably inhibit the differentiation of 3T3-L1 pre-adipocytes. This is the first report about -glucosidase and 3T3-L1 inhibitory activities of compounds 3, 7 and 8.
Dynamical Changes of Volatile Metabolites and Identification of Core Fungi Associated with Aroma Formation in Fu Brick Tea During the Fungal Fermentation

Abstract:Fu Brick Tea (FBT), a distinguished dark tea, derives its unique sensory identity from a specialized fungal fermentation process. However, the effect of core functional fungi on sensory attributes during FBT fungal fermentation still needs to be systematically explored. Employing a synergistic blend of electronic sensory evaluation, microbiomics, and volatile metabolomics, this study delves into the dynamic evolution of fungi and volatile metabolites (VMs) during fungal fermentation. Utilizing fungal sequencing and Gas Chromatography-Mass Spectrometry (GC-MS) techniques, we unveiled 234 OTUs, with g_Eurotium emerging as a dominant fungus. Additionally, we identified 288 contributing VMs, 35 of which exhibited notable fluctuations during fermentation. These associated fungi influenced crucial metabolic pathways, enhancing the synthesis of sweet-scented components like acetophenone, while repressing woody and fecal aromas, exemplified by nerolidol, skatole, and indole. This study reveals the evolution of fungi and VMs during FBT fungal fermentation, and suggests 4 possible pathways of substance synthesis in which fungi may be involved, delivering valuable insights into the nuanced processes governing the quality formation of FBT.

Abstract:Fuzhuan brick tea (FBT), a distinctive Chinese dark tea with the predominant fungus of Eurotium cristatum, offered significant health benefits to Chinese people. In the current study, the in vivo bioactivities of E. cristatum (SXHBTBU1934) fermented green tea and spores of E. cristatum fermented on wheat were investigated, respectively. The methanol extract of fermented green tea and spore of E. cristatum both showed potent lipid-lowering activity in the blood of a high-fat diet induced hyperlipidemia model in golden hamsters and significantly reduced the accumulation of fat granules in the liver. These results indicated that the key active components were produced by E. cristatum. Chemical investigations suggested similar components in the two extracts and led to the identification of a new alkaloid, namely variecolorin P (1), along with four known structurally related compounds, (-)-neoechinulin A (2), neoechinulin D (3), variecolorin G (4), and echinulin (5). The structure of the new alkaloid was elucidated by HRESIMS, 1H, 13C, and 2D NMR analysis. The lipid-lowering activity of these compounds was evaluated using an oleic acid-induced HepG2 cell line model. Compound 1 significantly reduced the lipid accumulation in the HepG2 cell line with an IC50 value of 0.127 μM.

Abstract:Eurotium cristatum as the dominant fungi species of Fuzhuan brick tea in China, can produce multitudinous secondary metabolites (SMs) with various bioactivities. Polyketides are a very important class of SMs found in E. cristatum and have gained extensive attention in recent years due to their remarkable diversity of structures and multiple functions. Therefore, it is necessary to explore the polyketides produced by E. cristatum at the genomic level to enhance its application value. In this paper, 12 polyketide synthase (PKS) genes were found in the whole genome of E. cristatum E1 isolated from Fuzhuan brick tea. In addition, the qRT-PCR results further demonstrated that these genes were expressed. Moreover, metabolic analysis demonstrated E. cristatum E1 can produce a variety of polyketides, including citreorosein, emodin, physcion, isoaspergin, dihydroauroglaucin, iso-dihydroauroglaucin, aspergin, flavoglaucin and auroglaucin. Furthermore, based on genomic analysis, the putative secondary metabolites clusters for emodin and flavoglaucin were proposed. The results reported here will lay a good basis for systematically mining SMs resources of E. cristatum and broadening its application fields.

Abstract:Eurotium cristatum isolated from natural "Fu" brick tea was inoculated into pan-fired green tea.After fermentation of 30 days,the taste and chemical components of the tea were measured.The antioxidant activity of the fermented tea was also evaluated by using 1,1-diphenyl-2-picrylhydrazyl radical 2,2-diphenyl-1-(2,4,6-trinitrophenyl)hydrazyl(DPPH) and 2,2'-azinobis-(3-ethylbenzthiazoline-6-sulphonate)(ABTS).The results showed that the quality of the fermented tea was improved,and the treatment reduced the bitter taste and increased the content of theaflavin and thearubin.The scavenging abilities of the fermented tea reached 75% and 56% respectively in determination by DPPH and ABTS radical methods,and were similar to those of "Fu" brick tea produced naturally from Hunan Province,or reached 64.7% and 80.6%,and were higher than those of "Fu" brick tea produced naturally from Zhejiang Province.

Abstract:Chinese Fuzhuan brick-tea is a unique microbial fermented tea characterized by a period of fungal growth during its manufacturing process. The aim of the present study was to characterize, both physicochemically and microbiologically, traditional industrial production processes of Fuzhuan brick-tea. Fermenting tea samples were collected from the largest manufacturer. Physicochemical analyses showed that the low water content in the tea substrates provided optimal growth conditions for xerophilic fungi. The fungal communities existing in tea materials, fermenting tea, and stored teas were monitored by polymerase chain reaction (PCR)-denaturing gradient gel electrophoresis (DGGE) targeting the D1 region of the 26S rRNA genes, followed by sequencing of the amplicons. Results revealed that the microorganisms were from, or closely related to, the genera Eurotium, Debaryomyces, Aspergillus, Verticillium, Pichia, Pestalotiopsis, Rhizomucor and Beauveria. This is the first report of Debaryomyces participating in the processing of Fuzhuan brick-tea. We concluded that the dominant genera Eurotium, Debaryomyces and Aspergillus are beneficial fungi associated with the fermentation of Fuzhuan brick-tea. The genus Beauveria was present in the stored Fuzhuan brick-tea, which may help protect tea products from insect spoilage. The remaining four genera were of minor importance in the manufacturing of Fuzhuan brick-tea. The predominant Eurotium species, a strain named Eurotium sp. FZ, was phenotypically and genotypically identified as Eurotium cristatum. High performance thin layer chromatography analysis of anthraquinones showed that emodin existed in all the dark tea samples, but physcion was only detectable in the tea fermented by E. cristatum. The PCR-DGGE approach was an effective and convenient means for profiling the fungal communities in Fuzhuan brick-tea. These results may help promote the use of microbial consortia as starter cultures to stabilize and improve the quality of Fuzhuan brick-tea products.

Abstract:The conditions for extracting polysaccharides from tea (Camellia sinensis L.) fruit peel (TFPPs) were studied. Three parameters (temperature, time, and liquid/solid ratio) affecting the extraction of TFPP were optimized using response surface methodology (RSM). Under the optimized conditions, the yield of TFPP was predicted to be 4.98%. The physicochemical properties, in vitro antioxidant activities, and inhibitory effects on α-glucosidase of fractionated TFPPs (TFPP-0, TFPP-20, TFPP-40, and TFPP-60) were investigated. We found that the TFPPs were all acid protein-bound heteropolysaccharides, although with different chemical compositions. They had not only remarkable scavenging activity on 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) and reducing activity, but also excellent inhibitory potential against α-glucosidase in vitro. Our results suggest that tea fruit peel could be treated as a potential bioresource for the development of polysaccharide antioxidants.

Abstract:Introduction: Fuzhuan brick tea (FBT) is a worldwide popular beverage which has the appreciable potential in regulating glycometabolism. However, the reports on the hypoglycemic mechanism of FBT remain limited. Methods: In this study, the hypoglycemic effect of FBT was evaluated in a pharmacological experiment based on Kunming mice. Global metabolomics and network pharmacology were combined to discover the potential target metabolites and genes. In addition, the real-time quantitative polymerase chain reaction (RT-qPCR) analysis was performed for verification. Results: Seven potential target metabolites and six potential target genes were screened using the integrated approach. After RT-qPCR analysis, it was found that the mRNA expression of VEGFA, KDR, MAPK14, and PPARA showed significant differences between normal and diabetes mellitus mice, with a retracement after FBT treatment. Conclusion: These results indicated that the hypoglycemic effect of FBT was associated with its anti-inflammatory activities and regulation of lipid metabolism disorders. The exploration of the hypoglycemic mechanism of FBT would be meaningful for its further application and development.

Abstract:Fuzhuan brick tea is a distinctive Chinese dark tea for its significant health benefits and unique quality characteristics by a unique fungal fermentation stage, in which Eurotium cristatum strains play a crucial role in the post-fermentation process. In this study, the biochemical characteristics and lipid-lowing activity of E. cristatum (MF800948)-fermented loose tea (FT) were investigated. The results showed that total phenolics, flavonoids, catechins and free amino acids were decreased after fermentation, while three purine alkaloids, soluble carbohydrates and theabrownins were significantly increased compared to green tea. In addition, d-limonene (27.71%) and β-myrcene (11.55%) were the predominant volatiles in FT, giving the characteristic fruity and balsamic scent to the fermented loose tea. Correspondingly, the aqueous extract of FT showed potent lipid-lowering activity in a high-fat zebrafish model and the optimum effective concentration was 500 μg/mL, at which the lipid level declined to 51.49% compared to control group. Hence, it was suggested that loose tea fermented by E. cristatum (MF800948) could provide valuable insights for developing a high-quality and hypolipidemic dark tea.

Abstract:Fuzhuan brick-tea, a fungal-fermented tea, is commonly consumed in northwest China; in places such as Sinkiang and Tibet and is thought to be helpful in digestion. To better understand Fuzhuan brick-tea and its function on digestion, the Fuzhuan brick-tea’s chemical compounds were surveyed at pivotal process phases, and its effects on pancreatic enzymes in vitro were studied. Most of the changes in amino acids, proteins, polyphenols, catechins and organic acids were found during fungal fermentation phase. All the infusions of Fuzhuan brick-tea samples had promotional effects on pancreatic amylase and protease and no effect on pancreatic lipase. Correlation analysis and principle component analysis between the main compounds of Fuzhuan brick-tea and the activities of two pancreatic enzymes were performed. The results showed that among ten significantly related compounds, the catechins and organic acids were particularly correlated with these two pancreatic enzymes’ activities. The present work confirmed the importance of microbial fermentation in the compositional changes of Fuzhuan brick-tea and its effects on two pancreatic enzymes in vitro, and suggested the possible application of microbial fermented tea such as Fuzhuan brick-tea in digestive aid.

Abstract:Background: Eurotium cristatum (E. cristatum) is the probiotic fungus in Fu-brick tea, with which fermentation brings a unique flavor and taste and health-promoting effects. Tartary buckwheat leaves are rich in functional active substances such as flavonoids and phenolic compounds, yet are not effectively utilized. Methods: Tartary buckwheat leaves were processed into raw green tea first and subsequently fermented with E. cristatum to develop a novel fermented leaf tea. The tea quality was evaluated by the aspects of the sensory scores by E-tongue, the volatile compounds by HS-SPME-GC-MS, the non-volatile profile by biochemical and UPLC-MS/MS methods and the antioxidant activity by the colorimetric assay. Results: Fermented leaf tea displayed a golden yellow color, a unique “flower” aroma and a dark-tea taste, with an improved sensory acceptability. Fermentation raised the content of volatile heterocyclic and aromatic compounds, alkenes and other aromatic components, which produced a unique floral flavor. The proportion of sour, bitter and astringency accounting non-volatile compounds such as phenolic acids and amino acids decreased, while the proportion of umami and sweet accounting substances such as responsible amino acids increased. Fermented leaf tea displayed a relative stronger total antioxidant activity against ABTS. Conclusion: E. cristatum fermentation exerted positive effects on Tartary buckwheat leaf tea quality.

Abstract:Polysaccharides from Fuzhuan brick tea (FBTPS), one of most important bioactive components in tea, showed various health-promoting functions. Our previous work demonstrated that the crude FBTPS (CFBTPS) could modulate the gut microbiota. However, which purified fraction in CFBTPS contributing to the modulation of gut microbiota remains unclear. Thus, the fermentation characteristics and probiotic activity of a purified fraction (FBTPS-2-1) of CFBTPS were evaluated in this work. The results showed that gut microbiota could utilize FBTPS-2-1 to produce short-chain fatty acids including acetic, propionic, n-butyric and n-valeric acids. FBTPS-2-1 could modulate the structure and metabolic pathways of gut microbiota. FBTPS-2-1 could increase the health-promoting gut microbiota such as Prevotellaceae and Bifidobacteriaceae, and decreased the harmful bacteria such as Enterobacteriaceae and Fusobacteriaceae. The results of metagenomics showed that Prevotella copri and Megamonas funiformis were the dominant bacteria after fermentation of FBTPS-2-1. Furthermore, FBTPS-2-1 could regulate the biosynthesis and metabolism pathways of gut microbiota. Thus, the enrichment of food with FBTPS-2-1 is expected as a potential strategy for promoting human health due to modulation of gut microbiota.

Abstract:Eurotium cristatum is a dominant fungus in the post-fermentation of Fu brick tea, however, its potential role in affecting the volatile profile and non-volatiles metabolites during the liquid-state fermentation of dark tea is not clear. In this study, dark tea was prepared by liquid-state fermentation with E. cristatum . A total of 51 volatile compounds were identified by HS-SPME-GC-MS, which mainly consisted of ketones, aldehydes, esters, and alcohols. There were 12 volatiles (ROAV >1) contributed to the overall aroma, including linalool, citral, methyl salicylate, etc. The overall flavor of tea infusion had been greatly improved after fermentation. The content of ester-type catechins , total phenols , total flavonoids , theaflavins, thearubigins, soluble sugars, and free amino acids decreased, while the content of theabrownins and soluble proteins increased. Meanwhile, the content of nonester type catechins and gallic acid increased and then decreased. The contents of various volatiles and metabolites change under the action of E. cristatum , coordinating to form a fermented dark tea infusion with mint, flower and fruit aroma and mellow and slightly astringent quality. The antioxidant activities showed a positive correlation with substances such as gallic acid and polyphenols. The results showed that the liquid-state fermentation with E. cristatum can effectively influence the flavor characteristics and antioxidant capacity of dark tea infusion.

Identification and Analysis of “Golden-Flower” Fungus from Fu-Brick Tea in Shaanxi Province

Abstract:By dilution plate method,using potato dextrose agar medium,the "Golden-Flower" fungus Fu-brick tea produced in Shaanxi province,the birthplace of Fu-brick tea,was identified and analyzed for the first time.Strain JW0 was obtained through separation and purification.According to colony characteristics,morphology observation and back inoculation in Southern Shaanxi dried green tea,the strain was confirmed as Eurotium cristatum(Raper Fennell) Malloch Cain belonging to Eurotium genus,Eurotiaceae family,Eurotiales order,Ascomycetes.Its anamorph was Aspergillus cristatellus Kozak.,synonym of A.cristatus Raper Fennell,A.spiculosus Blaser.

Abstract:This study investigated the potential fat-thermogenic effects of Eurotium cristatum, and elucidated the underlying mechanisms. The 12-week administration of E. cristatum in HFD-fed obese mice reduced body weight and improved glucolipid metabolism disorders. The administration of E. cristatum also efficiently promoted thermogenesis by increasing the expression of UCP1 and PRDM16 in both interscapular brown adipose tissue (iBAT) and inguinal white adipose tissue (iWAT) of HFD-fed mice. Furthermore, E. cristatum shaped the gut microbiome by increasing the abundance of Parabacteroides and Akkermansia muciniphila, and also elevated the levels of cecal short-chain fatty acids, particularly propionate and acetate. Of note, A. muciniphila was highly negatively correlated with body weight gain (r = -0.801, p < 0.05) and the iWAT index (r = -0.977, p < 0.01), suggesting that A. muciniphila may play an important role in the thermogenic mobilization induced by E. cristatum. Continuous supplementation with A. muciniphila suppressed adipose accumulation, improved glucolipid metabolism, and enhanced the thermogenic activity of iWAT and iBAT. Collectively, our results propose that boosted A. muciniphila acts as a key microbe in tea-derived probiotic E. cristatum-mediated fat-thermogenic and anti-obesity effects.

Abstract:As living standards advance, an escalating emphasis is placed on health, particularly in relation to prevalent chronic metabolic disorders. It is necessary to explore safe and effective functional foods or drugs. Fu brick tea (FBT) is a kind of dark tea fermented by fungi. The extracts are rich in compounds that can effectively relieve metabolic diseases such as hyperglycemia and hyperlipidemia, protect the liver, improve human immunity, enhance antioxidant activity, and regulate intestinal flora. This paper summarizes the biological activities and mechanisms of the extracts, polysaccharides, and small molecular compounds of FBT, which provides a certain theoretical basis for the rational, systematic, comprehensive development and utilization of the FBT resources. It is expected to develop and apply these active substances in health care products and natural medicines and provide more beneficial and diversified FBT products for human beings.
