Abstract:Objective: The central role of mitochondria is energy metabolism and ATP production, and recent work has revealed the importance of mitochondrial dysfunction in a number of cellular processes, including cell death. There is little information about the role of mitochondria in calcium regulation and signaling in gastrointestinal tissues. To understand the role of mitochondria in intracellular calcium signaling in the enteric nervous system, we studied the relationship of the cytosolic calcium concentration ([Ca2+] c) and mitochondrial matrix calcium concentration ([Ca2+] m), and the effect of mitochondrial dysfunction on the calcium signal in guinea pig enteric neurons. Methods: Primary cultures of enteric neurons from guinea pig taenia coli were double loaded with the cytoplasmic Ca2+ fluorescent indicator fura 2- acetoxymethyl ester (AM) and the mitochondrial Ca2+ fluorescent dye Rhod-2, and intracellular and mitochondrial calcium concentrations were measured using digital imaging microscopy. The cells were confirmed to be viable neurons by activating voltage-dependent currents with potassium chloride. Single neurons were then superfused with agonists in separate experiments, including ATP and glutamate. Neurons were then exposed to the mitochondrial inhibitors FCCP (p-trifluoromethoxy carbonyl cyanide phenyl hydrazone) and Antimycin A, and ([Ca2+] c) and ([Ca2+] m) calcium signals were again measured and analyzed using fluorescent microscopy Results: MitoTracker staining revealed a dense mitochondrial network in the enteric neurons, particularly surrounding the nucleus. Exposure of the cultured neurons to ATP (10−4) and glutamate (10−4) induced increases in ([Ca2+] c), followed by increases in ([Ca2+] m) in all cells analyzed. The response consisted of an initial rapid cytoplasmic response, with an average maximal change in fluorescence (ΔF) of 300 +/− 25 units, followed by a mitochondrial response (ΔF = 40 +/− 10 units) within seconds. The cytoplasmic response lasted an average of 200 +/− 50 milliseconds (ms). Depolarization and inactivation of the mitochondria prior to agonist stimulation (both ATP and Glutamate) with both FCCP (10μM) and Antimycin A (10μM), resulted in ([Ca2+] c) responses of increased amplitude (325 +/− 50 units) and longer duration (300 +/− 20 ms), indicating buffering of the cytoplasmic calcium by mitochondria. Conclusions: Our study demonstrates that mitochondria are important in the modulation of cellular calcium and contribute to calcium homeostasis. Understanding the role of mitochondria in calcium signaling is important, given the implication of mitochondrial calcium and dysfunction in a number of neuronal processes, including necrotic cell death and apoptosis.
