Abstract:A regeneration protocol for cultured endosperm of Actinidia chinensis were set up via organogenesis from callus tissues derived from endosperm explants of Actinidia chinensis ＇ Jintao＇ in vitro. The effects of different media, plant growth regulators, sucrose concentrations and dark treatment on callus induction and shoot formation were studied. The addition of 2,4-D significantly increased percentage of calli formation form the endosperm explants. Addition of 20g/L of sucrose resulted in better organogenesis form, endosperm callus. Dark culture could promote the growth of endosperm callus and organ formation, and the best efficiency was obtained after 7d of culture in the dark.

Abstract:Ophiocordyceps sinensis (Berk.) is a complex is formed by Hepialidae larvae and Hirsutella sinensis. Infestation by H. sinensis, interaction with host larvae, and fruiting body development are three crucial processes affecting the formation of O. sinensis. However, research on the molecular mechanism of O. sinensis formation has been hindered by the lack of effective genetic transformation protocols. Therefore, Agrobacterium tumefaciens-mediated transformation (ATMT) was adopted to genetically transform two H. sinensis strains and optimize the transformation conditions. The results revealed that the most suitable Agrobacterium strain for H. sinensis transformation was AGL1, and that the surfactant Triton X-100 could also induce ATMT, although less effectively than acetosyringone (AS). In addition, the endogenous promoters of H. sinensis genes had a stronger ability to drive the expression of the target gene than did the exogenous promoter. The optimal transformation conditions were as follows: AS and hygromycin B concentrations of 100 μM and 50 μg/mL, respectively; A. tumefaciens OD600 of 0.4; cocultivation at 18 °C for 24 h; and H. sinensis used within three passages. The results lay a foundation for the functional study of key regulatory genes involved in the formation of O. sinensis.

Optimization of Protoplast Preparation and Regeneration of a Medicinal Fungus Antrodia cinnamomea

Abstract:Antrodia cinnamomea is a unique medicinal fungus in Taiwan. It has been found rich in some pharmacologically active compounds for anti-cancer, hangover, and immune regulation etc. With the in-depth study of these components, it would be interesting and important to establish a molecular system for basic studies of A. cinnamomea. Thus, we would like to set up a foundation for this purpose by studying the A. cinnamomea protoplast preparation and regeneration. Firstly, we studied the optimization method of protoplast preparation of A. cinnamomea, and found various factors that may affect the yield during protoplast preparation, such as mycelial ages, pH values, and osmotic stabilizers. Secondly, in the regeneration of protoplasts, we explored the effects of various conditions on the regeneration of protoplasts, including different media and osmotic pressure. In addition, we found that citrate buffer with pH value around 3 dramatically increased the regeneration of protoplasts of A. cinnamomea, and provided a set of regeneration methodology for A. cinnamomea.

Abstract:A 3 factor, 3 level Box-Behnken factorial design combined with response surface methodology was used to optimize the fermentation process for production of extracellular polysaccharide (EPS) from Hirsutella sinensis . The 3 independent variables were temperature (X 1 ), initial pH (X 2 ), and the glucose concentration (X 3 ). Experimental data were fitted to a 2 nd order polynomial equation using multiple regression. The optimal fermentation conditions of the production of EPS were a temperature of 18.21°C, an initial pH of 5.81, and a glucose concentration of 7.39 g/L. The maximum predicted EPS production of 2.41 g/L was close to the actual experimental EPS yield (2.42±0.038%), demonstrating the model validity. EPS from H. sinensis had a direct in vitro immuno-stimulating activity using murine macrophage RAW264.7 cells, and stimulated release of several major cytokines (IL-1β, NF-κB, TNF-α, and iNOS) in a dose-dependent manner.

Abstract:By the orthogonalty design, the factors which influenced the regeneration of Brassica oleracea L. var. alboglabra were studied, the results showed that the major factor is the explant, and the next are NAA, BAP, sucrose and AgNO_3, the best recipe of the culture medium for the regeneration is: MS+NAA 0.03 mg/L+BAP 2mg/L+1% sucrose+AgNO_37.0 mg/L+0.8% agar. The optimum explant is the hypocotyls with the regeneraion frequency 97.5%.
Shoot regeneration process and optimization of Agrobacterium-mediated transformation in Sinningia speciosa

Abstract:The florist’s Gloxinia, Sinningia speciosa, which bears considerable flower trait variations, is an emerging model plants to study floral traits development. However, the investigation of the genetic information linking these floral traits is limited due to a lack of a reliable and efficient transformation system for functional studies. This study aims to optimize a stable genetic transformation system for S. speciosa. Detailed regeneration process and tissue culture parameters are also elucidated. The results show that the plant regeneration, initiated from a single perivascular parenchyma cell, can be induced from leaf and petiole explants in the presence of 1 mg/mL 6-benzylaminopurine (BA) and 0.1 mg/mL naphthalene-acetic acid (NAA) through embryogenesis. In the presence of 0.1 mg/mL NAA only, the adventitious roots form prior to the re-differentiation of shoot tissues in leaf explants. When the proximal end of the petiole is orientated upright with the distal end to the medium, it results in higher success of regeneration, suggesting that hormone supplies must follow endogenous basipetal auxin polarity. Using a glucuronidase (GUS) reporter gene construct, maximum transformation (3.13%) was obtained after a 3 day pre-culture and 5 day co-culture from cotyledons and leaves of 3-week-old seedlings inoculating Agrobacterium strain EHA105. The putative transgenic lines were validated by RT-PCR, Southern blotting and GUS activity. Our result demonstrates that young seedlings are the best material for transformation, probably because young leaves are only a few cell layers thick allowing inner perivascular cell (the origin of regeneration) to be more accessible for Agrobacterium infiltration.

Design and Optimization of a Greener Sinomenine Hydrochloride Preparation Process Considering Variations among Different Batches of the Medicinal Herb

Abstract:The current methods used to industrially produce sinomenine hydrochloride involve several issues, including high solvent toxicity, long process flow, and low atomic utilization efficiency, and the greenness scores of the processes are below 65 points. To solve these problems, a new process using anisole as the extractant was proposed. Anisole exhibits high selectivity for sinomenine and can be connected to the subsequent water-washing steps. After alkalization of the medicinal material, heating extraction, water washing, and acidification crystallization were carried out. The process was modeled and optimized. The design space was constructed. The recommended operating ranges for the critical process parameters were 3.0-4.0 h for alkalization time, 60.0 degrees C-80.0 degrees C for extraction temperature, 2.0-3.0 (volume ratio) for washing solution amount, and 2.0-2.4 mol.L-1 for hydrochloric acid concentration. The new process shows good robustness because different batches of medicinal materials did not greatly impact crystal purity or sinomenine transfer rate. The sinomenine transfer rate was about 20% higher than that of industrial processes. The greenness score increased to 90 points since the novel process proposed in this research solves the problems of long process flow, high solvent toxicity, and poor atomic economy, better aligning with the concept of green chemistry. Enhanced Synthesis of Coenzyme Q10 by Reducing the Competitive Production of Carotenoids in Rhodobacter Sphaeroides

Abstract:In Rhodobacter sphaeroides, synthesis of coenzyme Qio (CoQ(10)) shares the same precursor geranylgeranyl diphosphate (GGPP) with carotenoids. Therefore, suppression of carotenoids synthesis is supposed to pose positive effects on accumulation of CoQ(10). In this paper, the carotenogenesis pathway was repressed using two different strategies, deletion of carotenogenic genes and overexpression of ppsR, a transcriptional regulator for photosynthesis genes, respectively. Knockout of carotenoids synthesis (crt) genes resulted in undetectable carotenoids as expected. However, the production of CoQ(10), and biomass were both decreased to half as compared to the wild-type strain. In contrast, upon overexpression of ppsR, the production of carotenoids was decreased from 15.7 mg/L to 2.2 mg/L, and the CoQ(10) production and content were enhanced by 28% and 34.2%, respectively. To further enhance the production of CoQ(10), crtE was constitutively co-overexpressed with ppsR to improve the supply of GGPP as a key precursor for the isoprenoid side chain of CoQ(10). The CoQ(10) production and content of the resulting strain RspPE were increased to 73.2 mg/L and 5.67 mg/g, respectively, representing 47% and 55% improvement as compared to the wild type. This result demonstrated that appropriate reduction of carotenoids rather than complete blocking could enhance the CoQ(10) production in Rhodobacter. sphaeroides. (C) 2017 Elsevier B.V. [Reclamation of the Plant Regeneration Efficiency of Brassica Campestris Subsp. Chinensis Var.parachinensis].

Abstract:This investigation has developed an efficient and fast method for plant regeneration from petiole of cotyledon explants of Brassica campestris L. subsp. chinensis Makino var. parachinensis Tsen et Lee. A medium was designed for B. campestris subsp. chinensis var. parachinensis to obtain the high frequency of shoot regeneration, which contained BAP 2 mg/L, NAA 0.75-1.0 mgL and 7.5 mg/L AgNO3 solution to the half of NH4+ concentration's MS basic medium. 60 mL/L coconut milk were added to all of media. In this method, frequency of shoot regeneration of "youqing caixin" reached as high as 91.2% and the number of shoots per explant reached as high as 4.7 plants. The result showed that there was a positive correlation between frequency of shoot regeneration and number of shoots per explant. The little shoots could be observed five days after inoculation and were formed directly. The inducing rate of roots of the shoots reached as high as 100% and the rate of viability of transferred mature plant reached higher than 95%. The regeneration period from petiole with cotyledon to a seedling was shorten to about 49 days. Factors influencing in vitro explant regeneration were studied.
Stain improvement of docosahexenoic acid producing by protoplast mutagenesis

Abstract:This article offered an experimental analysis of the protoplastization and regeneration of Schizochytrium sp.,aiming to get a high yield production strain of DHA.The investigation showed that the ideal result would be got when 0.3 g/L snailase reacted 20 min with cells at 30 ℃,the protoplastization rate could reach 77.78%,and the regeneration rate reached 0.57%.The mutant,called Schizochytrium sp.HX-204M,was obtained by protoplast mutagenesis,combining with metabolic pathway analysis.Its cell dry weight and fatty acid was increased by 12.27% and 13.76%,respectively.In addition,the DHA content was higher than the control.

Abstract:Salvia miltiorrhiza is a traditional Chinese medicinal plant of Labiatae, which has been widely utilized to treat a variety of cardiovascular and cerebrovascular diseases. However, due to the long growth cycle, low content of active ingredients, and serious quality deterioration of S. miltiorrhiza , the use of biotechnology to improve S. miltiorrhiza to meet the growing demand for clinical applications has become a research hotspot. In this study, a novel one-step hairy root regeneration method was developed, which could rapidly obtain hairy roots and regenerated plants with high tanshinone content. By optimizing the parameters of Agrobacterium rhizogenes transformation in S. miltiorrhiza , it was finally established that the explants were infected in Ar.qual (OD 600 = 0.6) for 10 min, co-cultured for 3 days, and then screened on the screening medium containing 7.5 mg/l hygromycin, the maximum transformation frequency can reach 73.85%. GFP and PCR detection yielded a total of 9 positive transgenic hairy root lines and 11 positive transgenic regenerated plants. SmGGPPS1 was successfully overexpressed in positive transgenic regenerated plants, according to the results of qRT-PCR. The content of tanshinone IIA and cryptotanshinone were dramatically enhanced in transgenic regenerated plants and hairy roots by Ultra Performance Liquid Chromatography analysis. Based on the Agrobacterium-mediated transformation of S. miltiorrhiza , this study developed a new method for regenerating plants with transgenic hairy roots. This method provides a foundation for the breeding of S. miltiorrhiza and the sustainable development of medicinal plant resources, as well as provides a useful reference for the application of other species.

Callogenesis and Plant Regeneration in Peony (paeonia × Suffruticosa) Using Flower Petal Explants

Abstract:Peony is a traditional Chinese flower with significant ornamental and medicinal value. However, there are still problems, such as serious browning, difficulties in differentiation, and rooting and low regeneration efficiency in the process of the regeneration system established, which have hindered the development of transgenic peony technology. Establishing an efficient regeneration system is considered to be an important goal among peony researchers. Here, we describe a protocol for high-frequency callus induction and establishment of peony plants using flower petals as explants. Murashige and Skoog (MS) medium supplemented with 2.0 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D), 1.5 mg/L N-(phenylmethyl)-9H-purin-6-amine (6-BA), and 0.3 mg/L 1-naphthylacetic acid (NAA) was identified as the best medium for callus induction, achieving an induction rate of up to 98.52%. The highest peony proliferation rate (234%) was achieved on MS supplemented with 0.2 mg/L NAA and 3.0 mg/L 6-BA. The highest callus differentiation rate (34.81%) was achieved on MS supplemented with 2.0 mg/L 6-BA and 0.5 mg/L zeatin (ZT). The highest rooting rate was 23.33% when using 1/2 MS supplemented with 0.1 mg/L NAA and 0.05 mg/L 3-indolebutyric acid (IBA). After acclimation, the plants were transferred to pots, where they showed robust growth. We also observed the surface structures of the calluses using scanning electron microscopy and found that the differentiation characteristics of the calluses were hugely variable and that different surface structures appeared to affect bud differentiation efficiency. The efficient and rapid system for regenerating peonies using petal cultures established here will create new opportunities for the mass reproduction and genetic engineering of peony plants.
Design and optimization of purification process of sinomenine hydrochloride

Abstract:Sinomenine hydrochloride is generally produced from Caulis Sinomenii. At present, the purification process in industrial production suffers from large amount of solid waste, high solvent toxicity, and low sinomenine hydrochloride yield. In this study, a new purification process for sinomenine hydrochloride was proposed by using the extract obtained from acid extraction of Caulis Sinomenii as the starting material. The process included the following steps: alkalization, extraction, water washing, acid–water stripping, drying, and crystallization. 1-Heptanol was used as the extractant. The distribution coefficients of sinomenine and sinomenine hydrochloride in 1-heptanol–water system were 27.4 and 0.0167, respectively. The dissociation constants of sinomenine hydrochloride were 8.27 and 11.24, respectively. Process parameters of the new purification process were optimized with experimental design. The extractant 1-heptanol and sinomenine hydrochloride in the crystallization mother solution can be recycled in the new process. The purity of the obtained sinomenine hydrochloride crystals exceeded 85%, and the yield was about 70%. Compared with current industrial processes, safer extractant, less solid waste, and higher sinomenine hydrochloride yield can be achieved using the new purification process of sinomenine hydrochloride provided in this study.
Enhancement of cordyceps polysaccharide production via biosynthetic pathway analysis in Hirsutella sinensis

Abstract:The addition of various sulfates for enhanced cordyceps polysaccharide (CP) production in submerged cultivation of H. sinensis was investigated, and manganese sulfate was found the most effective. 2mM of manganese sulfate on 0day (d) was investigated as the optimal adding condition, and the CP production reached optimum with 5.33%, increasing by 93.3% compared with the control. Furthermore, the consumption of three main precursors of CP was studied over cultivation under two conditions. Intracellular mannose content decreased by 43.1% throughout 6days cultivation, which corresponded to CP accumulation rate sharply increased from 0 d to 6 d, and mannose was considered as the most preferred precursor for generating CP. Subsequently, mannose biosynthetic pathway was constructed and verified for the first time in H. sinensis, which constituted the important part of CP biosynthesis, and transcriptional levels of the biosynthetic genes were studied. Transcriptional level of gene cpsA was significantly up-regulated 5.35-fold and it was a key gene involved both in mannose and CP biosynthesis. This study demonstrated that manganese sulfate addition is an efficient and simple way to improve CP production. Transcriptional analysis based on biosynthetic pathway was helpful to find key genes and better understand CP biosynthesis.

Abstract:Based on the response surface methodology, an effective microbial system for diosgenin production from enzymatic pretreated Dioscorea zingiberensis tubers with Trichoderma reesei was studied. The fermentation medium was optimized with central composite design (35) depended on Plackett–Burmann design which identified significant impacts of peptone, K2HPO4 and Tween 80 on diosgenin yield. The effects of different fermentation conditions on diosgenin production were also studied. Four parameters, i.e. incubation period, temperature, initial pH and substrate concentration were optimized using 45 central composite design. The highest diosgenin yield of 90.57% was achieved with 2.67% (w/v) of peptone, 0.29% (w/v) of K2HPO4, 0.73% (w/v) of Tween 80 and 9.77% (w/v) of substrate, under the condition of pH 5.8, temperature 30 °C. The idealized incubation time was 6.5 days. After optimization, the product yield increased by 33.70% as compared to 67.74 ± 1.54% of diosgenin yield in not optimized condition. Scale-up fermentation was carried out in a 5.0 l bioreactor, maximum diosgenin yield of 90.17 ± 3.12% was obtained at an aeration of 0.80 vvm and an agitation rate of 300 rpm. The proposed microbial system is clean and effective for diosgenin production and thus more environmentally acceptable than the traditional acid hydrolysis.

Abstract:Arctic-derived fungus Eutypella sp. D-1 has attracted wide attention due to its huge ability to synthesize secondary metabolites. However, current studies only focus on stimulating its production of new secondary metabolites by OSMAC strategies, and the relationship between secondary metabolites and biosynthetic gene clusters (BGCs) has not been explored. In this study, the preparation and regeneration conditions of Eutypella sp. D-1 protoplasts were explored to lay a foundation for the study of genetic transformation of this fungus. Orthogonal experiment showed that the optimal preparation conditions were 0.75 M NaCl, 20 g/L of lysing enzyme, and 20 g/L of driselase, 28°C for 6 h. The maximum yield of Eutypella sp. D-1 protoplasts could reach 6.15 × 10 6 cells·ml −1 , and the concentration of osmotic stabilizer NaCl was the most important factor for Eutypella sp. D-1 protoplasts. The results of FDA staining showed that the prepared protoplasts had good activity. Besides, the best protoplasts regeneration medium was YEPS, whose maximum regeneration rate is 36%. The mediums with nitrogen sources, such as SR and RM, also had good effects on the Eutypella sp. D-1 protoplast regeneration, indicating that nitrogen sources played an important role on the Eutypella sp. D-1 protoplast regeneration. Subsequent transformation experiments showed that hygromycin resistance genes ( hrg ) could be successfully transferred into the genome of Eutypella sp. D-1, indicating that the prepared protoplasts could meet the needs of subsequent gene manipulation and research. This study lays a foundation for the genetic transformation of Eutypella sp. D-1.

Research on the Preparation and Regeneration of Protoplast from High-concentration Mash Resistant Saccharomyces Cerevisia

Abstract:The preparation and regeneration conditions of protoplast from a high-concentration mash resistant S. cerevisia strain was studied. The results showed that the optimum time for protoplast preparation was the exponential growth of 7 h after the inoculum, and the optimum enzyme for cell wall hydrolysis was snailase. Through L16(5)4 orthogonal experiment, the optimum preparation conditions of S. cerevisia protoplast were identified as follows: 1.0 % of snailase, 0.7mol/L of KCl high osmotic buffer, 1.5 h hydrolysis, 0.1 % of β-mercaptoethanol protectant and hydrolysis temperature at 26 ℃. Under the above conditions, the formation rate and the regeneration rate of protoplast were 80.84 % and 36.03 % respectively. A further study showed that higher regeneration rate (39.78 %) of S. cerevisia protoplast could be obtained when the prepared protoplast was regenerated on the sandwich culture of 7 % sucrose media.
Optimization of Protoplast Preparation and Regeneration Conditions of Valsa mali var.mali

Abstract:Protoplasts with high quality and high regeneration rate are essential in genetic transformation and gene modification of fungi .Using highly pathogenic Valsa mali var.mali strain SDAU11-175 as research object , the protoplast preparation and regeneration conditions were optimized from five aspects of enzyme types, mycelia age, enzymolysis time, osmotic stabilizer and regeneration medium .The results showed that the conditions to obtain the maximum protoplast output were culturing mycelia for 54 hours and treating them with 3%driselase and 3% lysing enzymes for 3 hours at 28℃ using 0 .7 mol/L NaCl as osmotic stabilizer . The protoplasts regenerated best on YPS medium with the regeneration rate as 42 .21%.

Abstract:To establish an efficient in vitro regeneration system and lay a foundation for further research, the effects of shelling, plant hormones, explant position, infection parameters and other factors on the in vitro regeneration of luffa cotyledons were studied. The results showed that the best medium for inducing adventitious buds from luffa cotyledons was Murashige and Skoog (MS) + Thidiazuron (TDZ) 0.3 mg· L −1 + Indole-3-Butyric acid (IBA) 0.02 mg· L −1 + AgNO 3 2 mg· L −1 + sucrose 3 %, and the induction rate reached 95.83 %. However, different cotyledon cutting methods resulted in different regeneration rates of adventitious buds. Using this regeneration system, the ACS7 gene was successfully transferred into luffa by Agrobacterium-mediated transformation.

Efficient Regeneration of Hedychium coronarium through Protocorm-Like Bodies

Abstract:Hedychium coronarium J. Koenig is a multipurpose plant with significant economic value, but it has been overexploited and listed as a vulnerable, near threatened or endangered species. In vitro culture methods have been used for propagating disease-free propagules for its conservation and production. However, explant contamination has been a bottleneck in in vitro propagation due to the use of rhizomes as the explant source. Plants in the family Zingiberaceae have pseudostems that support inflorescences, while rhizomes are considered true stems. The present study, for the first time, reported that the pseudostem bears nodes and vegetative buds and could actually be true stems. The evaluation of different sources of explants showed that mature node explants derived from the stem were the most suitable ones for in vitro culture because of the lowest contamination and the highest bud break rates. Culture of mature node explants on MS medium supplemented with 13.32, 17.76, and 22.20 μM 6-benzylaminopurine (BA), each in combination with 9.08 μM thidiazurin (TDZ) and 0.05 μM α-naphthaleneacetic acid (NAA) induced the conversion of buds to micro-rhizomes in six weeks. More than 96% of the micro-rhizomes cultured on MS medium supplemented with 17.76 μM BA, 6.81 μM TDZ, and 2.46 μM indole-3-butyric acid (IBA) were converted to globular-shaped clumps with protocorm-like bodies (PLBs). Further culture of a piece of the clumps induced more than 15 adventitious shoots. Adventitious roots were produced at the base of adventitious shoots, and plantlets were readily transplanted to a substrate for acclimatization in a shaded greenhouse. The survival rate of the plants in the greenhouse was up to 90%. Plants grew vigorously, and there were no off-types from the regenerated 11,100 plants. Our study also, for the first time, shows that H. coronarium can be regenerated via PLBs, which may represent a new way of the in vitro propagation of H. coronarium. The established protocol could be used for the increased propagation of H. coronarium for conservation or commercial production.
