Abstract:Engineered nanomaterials (ENMs) have been widely exploited in several industrial domains as well as our daily life, raising concern over their potential adverse effects. While in general ENMs do not seem to have detrimental effects on immunity or induce severe inflammation, their indirect effects on immunity are less known. In particular, since the gut microbiota has been tightly associated with human health and immunity, it is possible that ingested ENMs could affect intestinal immunity indirectly by modulating the microbial community composition and functions. In this perspective, we provide a few pieces of evidence and discuss a possible link connecting ENM exposure, gut microbiota and host immune response. Some experimental works suggest that excessive exposure to ENMs could reshape the gut microbiota, thereby modulating the epithelium integrity and the inflammatory state in the intestine. Within such microenvironment, numerous microbiota-derived components, including but not limited to SCFAs and LPS, may serve as important effectors responsible of the ENM effect on intestinal immunity. Therefore, the gut microbiota is implicated as a crucial regulator of the intestinal immunity upon ENM exposure. This calls for including gut microbiota analysis within future work to assess ENM biocompatibility and immunosafety. This also calls for refinement of future studies that should be designed more elaborately and realistically to mimic the human exposure situation.

Abstract:The human body harbors a myriad of microbial communities with the highest concentration and diversity found within the intestine whose disturbance is associated with many diseases. The potential of gut microbiota modulation to confer health benefits has resulted in tremendous efforts of testing extrinsic materials, e.g., probiotics, prebiotics, diet modulation and fecal microbiota transplantation. Meanwhile, the assembly of gut microbiota follows its intrinsic rules, including host selective pressures, bacteria-bacteria interactions, fungi-bacteria interactions, and virus-bacteria interactions. Understanding how these intrinsic factors and mechanisms shape the indigenous gut microbiota can provide crucial knowledge to guide evidence-based microbiota modulation therapies. Hence, this review focuses on our current understanding of the gut microbiota assemblage shaped by host, resident bacteria, fungi as well as virus.

Abstract:In natural and engineered environments, microorganisms often exist as complex communities, which are key to the health of ecosystems and the success of bioprocesses in various engineering applications. With the rapid development of nanotechnology in recent years, engineered nanomaterials (ENMs) have been considered one type of emerging contaminants that pose great potential risks to the proper function of microbial communities in natural and engineered ecosystems. The impacts of ENMs on microorganisms have attracted increasing research attentions; however, most studies focused on the antimicrobial activities of ENMs at single cell and population level. Elucidating the influence of ENMs on microbial communities represents a critical step toward a comprehensive understanding of the ecotoxicity of ENMs. In this mini-review, we summarize and discuss recent research work on the impacts of ENMs on microbial communities in natural and engineered ecosystems, with an emphasis on their influences on the community structure and function. We also highlight several important research topics which may be of great interest to the research community.

Abstract:Nanomaterials are widely employed in everyday life, including food and engineering. Food additives on a nanoscale can enter the body via the digestive tract. The human gut microbiota is a dynamically balanced ecosystem composed of a multitude of microorganisms that play a crucial role in maintaining the proper physiological function of the digestive tract and the body's endocrine coordination. While the antibacterial capabilities of nanomaterials have received much interest in recent years, their impacts on gut microbiota ought to be cautioned about and explored. Nanomaterials exhibit good antibacterial capabilities in vitro. Animal studies have revealed that oral exposure to nanomaterials inhibits probiotic reproduction, stimulates the inflammatory response of the gut immune system, increases opportunistic infections, and changes the composition and structure of the gut microbiota. This article provides an overview of the impacts of nanomaterials, particularly titanium dioxide nanoparticles (TiO2 NPs), on the gut microbiota. It advances nanomaterial safety research and offers a scientific foundation for the prevention, control, and treatment of illnesses associated with gut microbiota abnormalities.

Abstract:Nanotechnology is employed across a wide range of antibacterial applications in clinical settings, food, pharmaceutical and textile industries, water treatment and consumer goods. Depending on type and concentration, engineered nanomaterials (ENMs) can also benefit bacteria in myriad contexts including within the human body, in biotechnology, environmental bioremediation, wastewater treatment, and agriculture. However, to realize the full potential of nanotechnology across broad applications, it is necessary to understand conditions and mechanisms of detrimental or beneficial effects of ENMs to bacteria. To study ENM effects, bacterial population growth or viability are commonly assessed. However, such endpoints alone may be insufficiently sensitive to fully probe ENM effects on bacterial physiology. To reveal more thoroughly how bacteria respond to ENMs, molecular-level omics methods such as transcriptomics, proteomics, and metabolomics are required. Because omics methods are increasingly utilized, a body of literature exists from which to synthesize state-of-the-art knowledge. Here we review relevant literature regarding ENM impacts on bacterial cellular pathways obtained by transcriptomic, proteomic, and metabolomic analyses across three growth and viability effect levels: inhibitory, sub-inhibitory or stimulatory. As indicated by our analysis, a wider range of pathways are affected in bacteria at sub-inhibitory vs. inhibitory ENM effect levels, underscoring the importance of ENM exposure concentration in elucidating ENM mechanisms of action and interpreting omics results. In addition, challenges and future research directions of applying omics approaches in studying bacterial-ENM interactions are discussed.

Abstract:This review explores the impact of engineered nanomaterials (ENMs) on soil microbial diversity, function, metabolic pathways, and resilience. With the increasing application of ENMs in agriculture and industrial fields, understanding their interaction with soil microorganisms is crucial. We examine various types of ENMs, their physicochemical properties, and how these influence soil microbial communities. This review highlights the dual role of ENMs, demonstrating both beneficial and detrimental effects on microbial diversity and activity. Fundamental interaction mechanisms, such as altering metabolic pathways and microbial community structure, are discussed. Additionally, we address the implications of ENM-induced changes for soil health and agricultural productivity. Further, the review provides insights into the complex dynamics of ENMs in soil ecosystems and outlines directions for future research to optimize their use while minimizing environmental risks.

Abstract:A staggering number of nanomaterials-based products are being engineered and produced commercially. Many of these engineered nanomaterials (ENMs) are finally disposed into the soil through various routes in enormous quantities. Nanomaterials are also being specially tailored for their use in agriculture as nano-fertilizers, nano-pesticides, and nano-based biosensors, which is leading to their accumulation in the soil. The presence of ENMs considerably affects the soil microbiome, including the abundance and diversity of microbes. In addition, they also influence crucial microbial processes, such as nitrogen fixation, mineralization, and plant growth promoting activities. ENMs conduct in soil is typically dependent on various properties of ENMs and soil. Among nanoparticles, silver and zinc oxide have been extensively prepared and studied owing to their excellent industrial properties and well-known antimicrobial activities. Therefore, at this stage, it is imperative to understand how these ENMs influence the soil microbiome and related processes. These investigations will provide necessary information to regulate the applications of ENMs for sustainable agriculture and may help in increasing agrarian production. Therefore, this review discusses several such issues.

Abstract:The role of the gut microbiome in human health and disease is being increasingly recognized. Gut microbes (including bacteria, fungi and viruses) can be genetically modified to diagnose (as biosensors) and treat (detoxification, controlled biosynthesis and precision targeting) the dysbiosis of the microbiome, which has been linked to several cancers and metabolic, autoimmune and infectious diseases. However, conventional manipulation of single microbial strains is often insufficient, and engineering a mutually supportive and collaborative network of gut microbes — 'a keystone consortium' — could be more effective. In this Review, we summarize gut microbiome engineering strategies against selected diseases and critically discuss their translational potential. We focus mainly on genetic engineering approaches, but we also discuss complementary strategies such as encapsulation, coupling with electronic devices, orthogonal diet engineering and faecal microbiota transplantation.

Abstract:Engineered nanomaterials (ENMs) are widely used in industrial, commercial, and medical fields due to their unique physicochemical properties and diverse functional advantages. Human exposure or direct contact with ENMs via various routes, including dermal, inhalation, and systemic administration, may allow ENMs to interact with immune systems, leading to immunostimulation or immunosuppression outcomes depending on antigen properties. The actions of various immunological responses and how ENMs' intrinsic properties impact immune responses are fascinating but under-investigated. Herein, we systemically review state-of-the-art advances in various molecular mechanisms of ENM-induced immunological responses to outline the critical roles of material properties on immunity. Through the growing awareness of nano quantitative structure-activity relationship (QSAR), we further conceptualize how to utilize ENMs’ immunomodulatory properties to develop safe, precise, and effective immunomodulation strategies based on high throughput and predictive discovery paradigms. We also discuss the challenges and opportunities for clinical translation using nano-enabled immune modulation for disease treatment.

Abstract:Emerging nanoscience and nanotechnology inevitably facilitate discharge of engineered nanomaterials (ENMs) into the environment. Owing to their versatile physicochemical properties, ENMs invariably come across and interact with various pollutants already existing in the environment, leading to considerable uncertainty regarding the risk assessment of pollutants. Nevertheless, the underlying mechanisms of the complicated joint toxicity are still largely unexplored. This review aims to aid in understanding the interaction of ENMs and pollutants from the perspective of ecological and environmental health risk assessment. Based on related research published from 2005 to 2018, this review focuses on summarizing the effect of ENMs on the toxicity of pollutants both in vivo and in vitro. Physicochemical interaction appears as a main factor affecting ENMs-pollutants joint toxicity, with the mechanisms and the resultants for ENM-pollutant adsorption been illustrated. Cellular and molecular mechanisms involved in the joint toxicity of ENMs and pollutants are discussed, including the effect of ENMs on the bioaccumulation, biodistribution, and metabolism of pollutants, as well as the defense responses of organisms against such pollutants. Future in-depth investigation are suggested to focus on further exploring biological mechanisms (especially for the antagonized effect of ENMs against pollutants), using more advanced mammalian models, and paying more attention to the realistic exposure scenarios.

Abstract:Microbiota plays a crucial role in human health and disease; therefore, the modulation of this complex and yet widely unexplored ecosystem is a biomedical priority. Numerous antibacterial alternatives have been developed in recent years, imposed by the huge problem of antibioresistance, but also by the people demand for natural therapeutical products without side effects, as dysbiosis, cyto/hepatotoxicity. Current studies are focusing mainly in the development of nanoparticles (NPs) functionalized with herbal and fruit essential oils (EOs) to fight resistant pathogens. This is due to their increased efficiency against susceptible, multidrug resistant and biofilm embedded microorganisms. They are also studied because of their versatile properties, size and possibility to ensure a targeted administration and a controlled release of bioactive substances. Accordingly, an increasing number of studies addressing the effects of functional nanoparticles and plant products on microbial pathogens has been observed. Regardless the beneficial role of EOs and NPs in the treatment of infectious diseases, concerns regarding their potential activity against human microbiota raised constantly in recent years. The main focus of current research is on gut microbiota (GM) due to well documented metabolic and immunological functions of gut microbes. Moreover, GM is constantly exposed to micro- and nano-particles, but also plant products (including EOs). Because of the great diversity of both microbiota and chemical antimicrobial alternatives (i.e., nanomaterials and EOs), here we limit our discussion on the interactions of gut microbiota, inorganic NPs and EOs. Impact of accidental exposure caused by ingestion of day care products, foods, atmospheric particles and drugs containing nanoparticles and/or fruit EOs on gut dysbiosis and associated diseases is also dissected in this paper. Current models developed to investigate mechanisms of dysbiosis after exposure to NPs/EOs and perspectives for identifying factors driving EOs functionalized NPs dysbiosis are reviewed.

Response of soil microbial communities to engineered nanomaterials in presence of maize (Zea mays L.) plants

Abstract:With the intended application of engineered nanomaterials (ENMs) in agriculture, accurate assessment the effect of these ENMs on soil microbial communities is especially necessary. Here, maize plants were cultivated in soil amended by SiO2, TiO2, and Fe3O4 ENMs (100 mg kg-1 soil) for four weeks. The impact of ENMs on bacterial community structure of the rhizosphere soil was investigated by using high-throughput sequencing. In addition, metabolites of maize rhizosphere soil were quantified by gas chromatography-mass spectrometry (GC-MS) based metabolomics. We found that the disturbance of ENMs on soil microbes are in the follow of Fe3O4>TiO2>SiO2. Exposure of Fe3O4 ENMs significantly reduced the abundance of nitrogen-fixation related bacteria Bradyrhizobiaceae (from 2.94% to 2.40%) and iron-redox bacteria Sediminibacterium (from 2.15% to 2.07%). Additionally, Fe3O4 ENMs significantly increased populations of Nocardioides (from 1.63% to 1.77%), Chitinophaga sancti (from 1.12% to 2.08%), Pantoea (from 1.31% to 2.22%), Rhizobiumand (from 1.41% to 1.74%) and Burkholderia-Paraburkholderia (from 1.50% to 2.09%), which are associated with carbon cycling and plant growth promoting. This study provides a perspective on the response of rhizosphere microbial community and low molecular weight metabolites to ENMs exposure, providing a comprehensive understanding of the environmental risk of ENMs.

Abstract:Nano-Al2O3 has been widely used in various consumer products and water treatment processes because of its unique physicochemical properties. The probability of human exposure to nano-Al2O3 increases significantly, of which oral ingestion is an important route. However, effects and underlying mechanisms of nano-Al2O3 on gut microbiota and resistome are still not well delineated. Here, we systematically investigated the effects of nano-Al2O3 on the human gut microbiome by an in vitro simulator of human colon microbial ecosystem. Results indicated that nano-Al2O3 interfered with the gut microbiota, and significantly suppressed the short-chain fatty acids metabolism, which might pose adverse effects on the host. More seriously, high level of nano-Al2O3 (50 mg/L) was more destructive to the gut flora, though the damage might be temporary. In addition, sub-inhibitory low-dose of nano-Al2O3 (0.1 mg/L) significantly enhanced the abundance of antibiotic resistance genes (ARGs) after 7-day exposure. This is attributed to that low concentration of nano-Al2O3 can promote horizontal transfer of ARGs by increasing cell membrane permeability and relative abundance of transposase (e.g. tnpA, IS613, and Tp614). Our findings confirmed the adverse effects of nano-Al2O3 on the human gut resistome and emphasized the necessity to assess potential risks of nanomaterials on the human gut health.

Abstract:Abstract Background Gut microbiome can be readily influenced by external factors, such as diet, antibiotic, bacterial/viral infection and environmental toxicants. Gut microbiota-mediated effects of engineered nanomaterials as such become the new frontiers in nanotoxicology. Methods An integrated approach combining 16S rRNA gene sequencing and liquid chromatography-mass spectrometry (LC-MS) metabolomics was used to determine the potential mechanistic pathway by which disturbed gut microbiota induced by ZnONPs might modulate host physiology and neurobehavior. Results Herein, we showed oral exposure to zinc oxide nanoparticles (ZnONPs), one typical kinds of nanomaterials used widely in the food industry, could cause the neurobehavioral dysfunctions in mice, manifested by spatial learning and memory deficits and locomotor activity inhibition. Our mechanistic results elucidated that ZnONPs exposure led to a marked disturbance of gut microbial composition but did not alter the microbiome α-diversity indexes. We also provided new evidence that neurobehavioral impairments induced by ZnONPs was closely associated with perturbation in the gut microbiota composition that were both specific to changes of neurobehavior-related genes (such as Bdnf and Dlg4 ) and correlated with serum and hippocampal metabolic disorders. Our data further identified a unique metabolite [DG(15:0/0:0/22:4n6)] that linked the relationships among gut microbiota, metabolites and neurobehavior-related genes. Conclusions ZnONPs exposure not only alters the gut microbiome community but also substantially disturbs its metabolic profiles, and therefore leading to neurobehavioral impairments vi gut-brain axis. These findings will provide a novel view for understanding the ZnONPs neurotoxicity through gut-brain axis and may lead to new potential prevention and treatment strategies.

Abstract:With the dramatically increasing use of nanotechnology-enabled consumer products, engineered nanomaterials (ENMs). ENMs are inevitably discharged into environment, posing unknown risks to ecosystem structure and function. Plants play important roles in soil and water resources in the environment. Although these ENMs have been widely used in medicine, agriculture, and consumer products for great economic benefit, their unique properties, may cause adverse effects on plants. Thus, toxicity and disposition of ENMs in terrestrial plants, especially agricultural crops, should be fully characterized. This mini-review summarizes the current literature on: (1) the distribution and biotransformation of ENMs; (2) ENMs induced phytotoxicity at physiological and molecular levels; (3) nutrient displacement or enhancement in ENMs treated plants; (4) trophic transfer of ENMs in terrestrial food chains.

Abstract:Nanoscale matter has unique attributes that potentiate their widespread application from biomedicine, engineering, food, cosmetics, sensing, to energy. Due to the rapid industrialization of engineered nanomaterials (ENMs) as a result of increasing consumer interests, more ENMs will be released into the environment, greatly increasing the probability of their contact with humans. Once these ENMs enter the human body or an ecosystem, they may initiate various biochemical responses in unprecedented ways, thus raising important concerns in regard to their effects on human health and the ecosystem. Here, the shifts in the intricate metabolic pathways instigated by ENMs are highlighted and those altered metabolic changes are extrapolated for the elucidation of environmental health and risk assessment. Furthermore, the toxico-metabolomics knowledge of ENMs provides the opportunity to design novel nanomedicine with high efficacies by capitalizing on their abilities to rewire metabolic networks for the treatment of diseases.

Abstract:The application of engineered nanomaterials (ENMs) is increasing in all walks of life, inevitably resulting in a high risk of ENMs entering the natural environment. Recent studies have demonstrated that phytoaccumulation of ENMs in the environment may be detrimental to plants to varying degrees. However, plants primarily assimilate ENMs through the roots, which are inevitably affected by rhizomicroorganisms. In this review, we focus on a group of common rhizomicroorganisms-arbuscular mycorrhizal fungi (AMF). These fungi contribute to ENMs immobilization and inhibition of phytoaccumulation, improvement of host plant growth and activation of systematic protection in response to excess ENMs stress. In present review, we summarize the biological responses of plants to ENMs and the modulatory mechanisms of AMF on the immobilization of ENMs in substrate-plant interfaces, and indirectly regulatory mechanisms of AMF on the deleterious effects of ENMs on host plants. In addition, the information of feedback of ENMs on mycorrhizal symbiosis and the prospects of future research on the fate and mechanism of phyto-toxicity of ENMs mediated by AMF in the environment are also addressed. In view of above, synergistic reaction of plants and AMF may prove to be a cost-effective and eco-friendly technology to bio-control potential ENMs contamination on a sustainable basis.
