Abstract:In the hexagonal columnar phase of chiral polymers a bias towards cholesteric twist competes with braiding along an average direction. When the chirality is strong, topological defects proliferate, leading to either a tilt grain boundary phase or a new ``moire state'' with twisted bond order. This moire phase can melt leading to a new phase: the chiral hexatic. I will discuss some recent experimental results from the NIH on DNA liquid crystals in the context of these theories.

Abstract:<p>The very chemical structure of DNA that enables biological heredity and evolution has non‐trivial implications for the self‐organization of DNA molecules into larger assemblies and provides limitless opportunities for building functional nanostructures. This progress report discusses the natural organization of DNA into chiral structures and recent advances in creating synthetic chiral systems using DNA as a building material. How nucleic acid chirality naturally comes into play in a diverse array of situations is considered first, at length scales ranging from an individual nucleotide to entire chromosomes. Thereafter, chiral liquid crystal phases formed by dense DNA mixtures are discussed, including the ongoing efforts to understand their origins. The report then summarizes recent efforts directed toward building chiral structures, and other structures of complex topology, using the principle of DNA self‐assembly. Discussed last are existing and proposed functional man‐made nanostructures designed to either probe or harness DNA's chirality, from plasmonics and spintronics to biosensing.</p>

Molecular engineering of chiral colloidal liquid crystals using DNA origami

Abstract:Establishing precise control over the shape and the interactions of the microscopic building blocks is essential for design of macroscopic soft materials with novel structural, optical and mechanical properties. Here, we demonstrate robust assembly of DNA origami filaments into cholesteric liquid crystals, 1D supramolecular twisted ribbons and 2D colloidal membranes. The exquisite control afforded by the DNA origami technology establishes a quantitative relationship between the microscopic filament structure and the macroscopic cholesteric pitch. Furthermore, it also enables robust assembly of 1D twisted ribbons, which behave as effective supramolecular polymers whose structure and elastic properties can be precisely tuned by controlling the geometry of the elemental building blocks. Our results demonstrate the potential synergy between DNA origami technology and colloidal science, in which the former allows for rapid and robust synthesis of complex particles, and the latter can be used to assemble such particles into bulk materials.

On the origin of chirality in plasmonic meta-molecules

Abstract:Chirality is a fundamental feature in all domains of nature, ranging from particle physics over electromagnetism to chemistry and biology. Chiral objects lack a mirror plane and inversion symmetry and therefore cannot be spatially aligned with their mirrored counterpart, their enantiomer. Both natural molecules and artificial chiral nanostructures can be characterized by their light-matter interaction, which is reflected in circular dichroism (CD). Using DNA origami, we assemble model meta-molecules from multiple plasmonic nanoparticles, representing meta-atoms accurately positioned in space. This allows us to reconstruct piece by piece the impact of varying macromolecular geometries on their surrounding optical near fields. Next to the emergence of CD signatures in the instance that we architect a third dimension, we design and implement sign flipping signals through addition or removal of single particles in the artificial molecules. Our data and theoretical modelling reveal the hitherto unrecognized phenomenon of chiral plasmonic-dielectric coupling, explaining the intricate electromagnetic interactions within hybrid DNA-based plasmonic nanostructures.

Abstract:Artificially designed structures with chirality are able to possess novel mechanical properties, for example, chiral honeycombs with negative Poisson's ratio and chiral acoustic metamaterials. Inspired by the chiral nature of double-helical structure of DNA molecule, in this paper, we present a design of helical structure with chirality and study its mechanical behaviors through experimental characterization, theoretical analysis and numerical simulation. A compression-twisting coupling deformation mode is observed for different types of DNA-inspired helical structures due to the competition among compression, bending and twisting energy of the helical structures and tension energy of the interlinks. We find that interlinks between two intertwined helices are decisive to the compression-twisting coupling deformation mode and both chiral and non-chiral behaviors of the structures can be quantitively captured by our established theoretical and numerical models. The influences of various geometric parameters on the stiffness and deformation mode are discussed. Finally, we present two types of superposed structures that behave differently, showing a high designability of the DNA-inspired structures. This study exploits the chirality of bio-inspired structures and uses it in structural design with novel mechanical properties, which may shed light on the development of bio-inspired mechanical metamaterials, impact energy absorbers, multi-functional composite materials and among others.

Abstract:A significant challenge exists in obtaining chiral nanostructures that are amenable to both solution-phase self-assembly and solid-phase preservation, which enable the observation of unveiled optical responses impacted by the dynamic or static conformation and the incident excitations. Here, to meet this demand, we employed DNA origami technology to create quasi-planar chiral satellite-core nanoparticle superstructures with an intermediate geometry between the monolayer and the double layer. We disentangled the complex chiral mechanisms, which include planar chirality, 3D chirality, and induced chirality transfer, through combined theoretical studies and thorough experimental measurements of both solution- and solid-phase samples. Two distinct states of optical responses were demonstrated by the dynamic and static conformations, involving a split or nonsplit circular dichroism (CD) line shape. More importantly, our study on chiral nanoparticle superstructures on a substrate featuring both a dominant 2D geometry and a defined 3D represents a great leap toward the realization of colloidal chiral metasurfaces.

Abstract:Tertiary chirality describes the handedness of supramolecular assemblies and relies not only on the primary and secondary structures of the building blocks but also on topological driving forces that have been sparsely characterized. Helical biopolymers, especially DNA, have been extensively investigated as they possess intrinsic chirality that determines the optical, mechanical, and physical properties of the ensuing material. Here, we employ the DNA tensegrity triangle as a model system to locate the tipping points in chirality inversion at the tertiary level by X-ray diffraction. We engineer tensegrity triangle crystals with incremental rotational steps between immobile junctions from 3 to 28 base pairs (bp). We construct a mathematical model that accurately predicts and explains the molecular configurations in both this work and previous studies. Our design framework is extendable to other supramolecular assemblies of helical biopolymers and can be used in the design of chiral nanomaterials, optically active molecules, and mesoporous frameworks, all of which are of interest to physical, biological, and chemical nanoscience.

Abstract:Confinement and hydrodynamic interactions often play an important role in the fluctuation dynamics of soft matter systems, which can typically be studied using light scattering techniques. With experimental and theoretical methodologies, I demonstrate here that chirality is an additional critical parameter that leads to diverging decay times and correlation lengths in chiral liquid crystal cells with a fully unwound cholesteric helix. This study combines light scattering measurements made in a tailored microscope geometry and theoretical calculations of the decay dynamics of chiral orientational fluctuations-including hydrodynamics-to establish the existence of two soft chiral modes of fluctuations driving the destabilization of the unwound cholesteric. Despite the achirality of the equilibrium state of unwound cholesterics, this study indicates that chirality hides itself in the orientational fluctuation modes and plays a major role in their dynamics, which can be exploited to locally measure the strength of chirality in frustrated chiral liquid crystal cells.

Abstract:Chiral growth exists prevalently in natural materials. The mechanism underlying the formation of chiral morphologies in biological and man-made materials has been an important issue of both theoretical and technological interest. In this paper, an elastic rod model taking into account chiral microstructures is developed to investigate the formation of chiral morphologies of biological materials. The curvature and twist of chiral shapes are investigated with this model using the variational method of energy. The result shows the misfit of chirality of two-layer structured biological materials may induce various chiral morphologies, such as helices and twisting belts. Furthermore, it was found that cooperative or competitive interactions between anisotropic elasticity and chirality can also lead to the formation of chiral morphologies, and the fibre orientation angles and chiral parameters are the determining factors to the shape, size and handedness of chiral morphologies. This work is expected to shed new light on the physical mechanisms of the formation of various chiral morphologies in the biological world and provide useful guidance for the design of deformation driving and shape control of soft robots and machines.

Abstract:The shape of the building blocks plays a crucial role in directing self-assembly towards desired architectures. Out of the many different shapes, helix has a unique position. Helical structures are ubiquitous in nature and a helical shape is exhibited by the most important biopolymers like polynucleotides, polypeptides and polysaccharides as well as by cellular organelles like flagella. Helical particles can self-assemble into chiral superstructures, which may have a variety of applications, e.g. as photonic (meta)materials. However,a clear and definite understanding of these structures has not been entirely achieved yet. We have recently undertaken an extensive investigation on the phase behaviour of hard helical particles, using numerical simulations and classical density functional theory. Here we present a detailed study of the phase diagram of hard helices as a function of their morphology. This includes a variety of liquid-crystal phases, with different degrees of orientational and positional ordering. We show how, by tuning the helix parameters, it is possible to control the organization of the system. Starting from slender helices, whose phase behaviour is similar to that of rodlike particles, the increase in curliness leads to the onset of azimuthal correlations between the particles and the formation of phases specific to helices. These phases feature a new kind of screw order, of which there is experimental evidence in colloidal suspensions of helical flagella.

Abstract:Helical structures such as double helical DNA and the α-helical proteins found in biological systems are among the most beautiful natural structures. Chiral nanoarchitectonics, which is used here to describe the hierarchical formation and fabrication of chiral nanoarchitectures that can be observed by atomic force microscopy (AFM), scanning tunneling microscopy (STM), scanning electron microscopy (SEM), or transmission electron microscopy (TEM), is one of the most effective ways to mimic those natural chiral nanostructures. This article focuses on the formation, structure, and function of the most common chiral nanoarchitectures: nanoscale chiral twists and helices. The types of molecules that can be designed and how they can form hierarchical chiral nanoarchitectures are explored. In addition, new and unique functions such as amplified chiral sensing, chiral separation, biological effects, and circularly polarized luminescence associated with the chiral nanoarchitectures are discussed.
The Thermodynamic Origins of Chiral Twist in Monolayer Assemblies of Hard Rod-like Colloids

Abstract:The propagation of chirality across scales is a common but poorly understood phenomenon in soft matter. In this work, we use computer simulations to study chiral monolayer assemblies formed by hard rod-like colloidal particles in the presence of non-adsorbing polymer and characterize the thermodynamic driving forces responsible for the twisting. Simulations show that straight (achiral) rods assemble into monolayers with a spontaneous twist that is either left- or right-handed, while helical (chiral) rods lead to assemblies with preferential chiral features that depend on their handedness and curliness. The onset of chirality in these monolayers can be traced back to small clusters formed at the initial stage of the self-assembly. In these microscopic monolayers, entropy drives twisting in ways that differ from the assumptions on which existing continuum theory is built. Depending on the geometry of the constituent rods, the preferred chiral twist can be driven by entropy gain of the polymers, or of the rods, or both. In addition, the variation of the polymer entropy with twist depends on changes in both the surface area and the volume of the monolayer. Rod fluctuations perpendicular to the monolayer also play an important role in stabilising the twisting.

Defects in Chiral Columnar Phases: Tilt Grain Boundaries and Iterated Moire Maps

Abstract:Biomolecules are often very long with a definite chirality. DNA, xanthan and poly-gamma-benzyl-glutamate (PBLG) can all form columnar crystalline phases. The chirality, however, competes with the tendency for crystalline order. For chiral polymers, there are two sorts of chirality: the first describes the usual cholesteric-like twist of the local director around a pitch axis, while the second favors the rotation of the local bond-orientational order and leads to a braiding of the polymers along an average direction. In the former case chirality can be manifested in a tilt grain boundary phase (TGB) analogous to the Renn-Lubensky phase of smectic-A liquid crystals. In the latter case we are led to a new "moire" state with twisted bond order. In the moire state polymers are simultaneously entangled, crystalline, and aligned, on average, in a common direction. In this case the polymer trajectories in the plane perpendicular to their average direction are described by iterated moire maps of remarkable complexity, reminiscent of dynamical systems.

Abstract:DNA monolayers with inherent chirality play a pivotal role across various domains, including biosensors, DNA chips, and bioelectronics. Nonetheless, conventional DNA chiral monolayers, typically constructed from single-stranded DNA (ssDNA) or double-stranded DNA (dsDNA), often lack structural orderliness and design flexibility at the interface. Structural DNA nanotechnology emerges as a promising solution to tackle these challenges. In this study, we present a strategy for crafting highly adaptable twisted DNA origami-based chiral monolayers. These structures exhibit distinct interfacial assembly characteristics and effectively mitigate the structural disorder of dsDNA monolayers, which is constrained by a limited persistence length of ~50 nm of dsDNA. We highlight the spin-filtering capabilities of four representative DNA origami-based chiral monolayers, demonstrating a maximal one-order-of-magnitude increase in spin-filtering efficiency per unit area compared to conventional dsDNA chiral monolayers. Intriguingly, our findings reveal that the higher-order, tertiary, chiral structure of twisted DNA origami further enhances the spin-filtering efficiency. This work paves the way for the rational design of DNA chiral monolayers.

Abstract:Intimately connected to the rule of life, chirality remains a long-time fascination in biology, chemistry, physics and materials science. Chiral structures, e.g., nucleic acid and cholesteric phase developed from chiral molecules are common in nature and synthetic soft materials. While it was recently discovered that achiral but bent core mesogens can also form chiral helices, the assembly of chiral microstructures from achiral polymers has rarely been explored. Here, we reveal chiral emergence from achiral conjugated polymers for the first time, in which hierarchical helical structures are developed through a multistep assembly pathway. Upon increasing concentration beyond a threshold volume fraction, pre-aggregated polymer nanofibers form lyotropic liquid crystalline (LC) mesophases with complex, chiral morphologies. Combining imaging, X-ray and spectroscopy techniques with molecular simulations, we demonstrate that this structural evolution arises from torsional polymer molecules which induce multiscale helical assembly, progressing from nano- to micron scale helical structures as the solution concentration increases. This study unveils a previously unknown complex state of matter for conjugated polymers that can pave way to a new field of chiral (opto)electronics. We anticipate that hierarchical chiral helical structures can profoundly impact how conjugated polymers interact with light, transport charges, and transduce signals from biomolecular interactions and even give rise to properties unimagined before.

Abstract:Developing artificial systems to mimic the emergence of chirality is of vital importance in better understanding the mysterious origin of natural homochirality and guiding the design of advanced chiroptical materials. Herein, we present a DNA-inspired control over the emergence of supramolecular nanoscale chirality in multiple hydrogen bonded self-assemblies. N-Terminated aromatic amino acids bearing serine and aromatic domains could self-assemble into lamellar microsheets without nanoscale chirality, ascribed to their pronounced molecular stacking preference on two-dimensional orientations. Significantly, when achiral bipyridine-, melamine-, and imidazole-based molecular binders that could potentially form hydrogen-bonded complexes with these aromatic amino acids were introduced, the induced chiral nanostructures from the resulting coassemblies were observed. Melamine and its derivatives presented an excellent capability to generate ordered supramolecular structures and induce the nanoscale chirality. Assisted by the key duplex hydrogen bonds between the melamine core and serine segments, chiral nanotubes and ribbons were obtained. This study reveals that multiple hydrogen bonds are the prerequisite for inducing the emergence of chiral nanostructures from integrated coassemblies.

Abstract:An unusual overwinding and a subsequently unwinding behavior of helix of filament with twisted microstructures under stretch is analyzed from the nonlinear variation of material chirality (i.e., chirality in material properties), motivated by recent single-molecule experiments for B-DNA. We establish a refined Cosserat rod model, which reproduces the deformation behaviors of filament incorporating its microscopic chirality. The helix of filament with twisted microstructures is found to exhibit a DNA-like nonlinear variation of material chirality under large elongation. The multiscale interactions between microscopic chirality of twisted microstructures and macroscopic chirality of helix determine the nonlinear coupling behaviors of hierarchically chiral structured materials. Abstract:The functions and transformation mechanisms of nucleic acids in their various states are of great importance in physiology and pathology. Based on the Landau model, this study reveals the mechanism of the existence of chirality and the transformation rules between different chiral conformations by studying the Duffing oscillation response of the local gauge potential on the DNA chain. The research results show that normal chiral DNA and DNA configuration transition behaviors exhibit regular nonlinear periodic oscillations. Moreover, the gauge potential value approaches zero at the B-Z junction site, which may add a criterion for detecting Z-forming sites. In addition, external force and damping play a key role in the chiral gauge potential of nucleic acids, and they can influence, regulate and even control the configuration transition of nucleic acids. Finally, we verified the rationality of our theory by combining X-ray crystal diffraction data of multiple configurations of DNA. This study provides new insights into the behavior and function of chiral DNA in organisms, and provides new possibilities for regulating DNA conformational transition in the future.
