Abstract:In this study, a method for detection of an ssRNA viral pathogen that causes viral flacherie in the silkworm, Bombyx mori (L.) (Lepidoptera: Bombycidae), was used for the detection of B. mori infectious flacherie virus (BmIFV). A combination of nested and reverse transcriptase polymerase chain reaction was used for detection. Although BmIFV has been reported in almost all the sericultural regions of the world, there had been no reports of BmIFV incidence in India. Therefore, the confirmation of the presence of BmIFV in Karnataka, India, is of great significance. The present method is advantageous because it can be used to detect the virus by using samples from infected midgut tissues, thus simplifying and avoiding laborious genome isolation procedures. This method could help in early detection of BmIFV disease pathogens and help reduce crop losses.

The paper introduced the history of the survey and described the virus genome organization that encodes corresponding putative proteome. We gave out an overview of BmIFV on replication mechanism, polyprotein cleavage, protein processing, sequence alignment, and epidemiology, Meanwhile. Comparing BmIFV with other picorna(-like) virus, the studies on cell factor involving in virus infection, virus gene function and epidemiological investigation are important.
Biochemical Effects of Fluoride on Haemolymph of the Silkworm, Bombyx Mori L.

Abstract:Changes in the Ca2+ and Mg2+ ion concentrations, catalase activity, and protein metabolism in the haemolymph of four different strains of silkworms fed a special diet supplemented with fluoride were investigated. Fluoride exposure caused decreases in the concentrations of the two ions, with significant differences between strains in their tolerance to fluoride. Catalase activity in the haemolymph of fluoride-exposed silkworm larvae of the Hang 8 strain was over 24 times higher than in the controls at its peak in the middle of the 4(th) instar. In contrast, glutamate dehydrogenase (GDH) activity and the levels of protein and free amino acids in the haemolymph were significantly lowered by fluoride treatment in all strains. These results suggest that biochemical changes in the haemolymph can be used as biomarkers to evaluate the adverse health effects and the economic impact of fluoride exposure on the silkworm, Bombyx mori L.

Abstract:The differential response of silkworm, Bombyx mori L. to phytoecdysteroid when administered at different ages of 5th instar was studied on a popular Chinese silkworm hybrid, Xinhang x Keming reared on the ‘Hu Sang 197' variety of mulberry leaves. Phytoecdysteroid, a chlolest -7 ene- 6- one- carbon skeleton (C27) from a plant source, Radyx achyranthes was administered to 5th instar silkworm per os at a rate of 2 µm per larva to different batches of silkworm at 48 h, 72 h and at the onset of cocoon spinning when a few larvae were ripe. The larval duration, larval mounting duration, cocoon characters and reeling traits were influenced by phytoecdysteroid. The intensity of influence was dependent on the time of application. The larvae treated at 48 h improved almost all economic traits significantly with a marginal increase in larval duration. In the larvae treated at 72 h, the economic traits were adversely affected but the larval duration was shortened by about 24 hours. In the larvae treated at the onset of cocoon spinning, the mounting duration was substantially reduced without much affect on the cocoon traits which would be a big benefit in commercial sericulture. Abstract:Effect of prostaglandin F2α (PGF2α) on carbohydrate accumulation in gonads of the multivoltine silkworm (hybrid: Xinhang×Keming) has been studied by means of topical application to larvae of the silkworm. Increased weights of larvae and reproduction organs, as well as carbohydrate metabolism in gonads of the silkworm, Bombyx mori L. was found after treatment with prostaglandin F2α. The increase in weight (larvae 21.9%, testis 28.9%, and ovary 33.3%) was associated with increases in the biomolecules (20–30%) and LDH and aldolase activity (18–25%). The results suggest that the accumulation of carbohydrates denotes a higher extent of utility of the energy sources in function of the testes and ovaries, and the routine application of prostaglandin F2α would be helpful in improving the reproductivity and egg quality of the silkworm.

Abstract:Changes in the activities of alkaline phosphatase (ALKP), adenosine triphosphatase (ATPase), and phosphorylase in the midgut of silkworm (Bombyx mori L.) larvae exposed to NaF-treated mulberry leaves were investigated. The activity of ALKP was markedly reduced by fluoride compared to the control. In the presence of Ca(OH)(2) or MgSO4, the ALKP activity was only slightly affected. The activities of ATPase and phosphorylase in the midgut were also reduced by IF treatment. These results suggest that certain midgut enzyme activities of silkworm larvae that are adversely affected by fluoride pollution can be used to monitor the health of silkworms and, in the case of ALKP, can also be countered by Ca or Mg salts.

Abstract:Effect of indole-3-acetic acid (IAA) on economic characters of bivoltine pure races, Baiyu and Qinfeng were investigated. Fortification of mulberry leaf with indole-3-acetic acid (IAA) has proved to be successful in improving the crop yield. The increase in economic characters suggests that IAA has stimulatory effect on cocoon yield, silk quality and fecundity rate. Hence IAA can be utilized in sericulture for the improvement of yield and has practical advantage in grainages and reeling industry.

Abstract:The effects of UV-light on biochemical parameters of posterior silk gland of mulberry silkworm, Bombyx mori L. were investigated in two popular bivoltine hybrids, Jufang (Chinese) × Chenxin (Japanese) and Chenxin × Jufang fed an artificial diet. Larvae of the first day of fifth instar were used for the present investigation. The larvae were irradiated with 20 W of UV-light at the intensity of 40 cm above the larvae for one of three exposure times (30, 60 or 120 min) just before feeding. UV treatment produced no significant effects at 30 min. However, at 60 or 120 min there was a significant decrease in silk gland weight and the levels of total protein, free amino acids, RNA and carbohydrates. UV-light thus affects the biosynthetic activities of silk glands.

Abstract:The gut microbiota has a crucial role in the growth, development and environmental adaptation in the host insect. The objective of our work was to investigate the microbiota of the healthy silkworm Bombyx mori gut and changes after the infection of B. mori cypovirus (BmCPV). Intestinal contents of the infected and healthy larvae of B. mori of fifth instar were collected at 24, 72 and 144 h post infection with BmCPV. The gut bacteria were analyzed by pyrosequencing of the 16S rRNA gene. 147(135) and 113(103) genera were found in the gut content of the healthy control female (male) larvae and BmCPV-infected female (male) larvae, respectively. In general, the microbial communities in the gut content of healthy larvae were dominated by Enterococcus, Delftia, Pelomonas, Ralstonia and Staphylococcus, however the abundance change of each genus was depended on the developmental stage and gender. Microbial diversity reached minimum at 144 h of fifth instar larvae. The abundance of Enterococcus in the females was substantially lower and the abundance of Delftia, Aurantimonas and Staphylococcus was substantially higher compared to the males. Bacterial diversity in the intestinal contents decreased after post infection with BmCPV, whereas the abundance of both Enterococcus and Staphylococcus which belongs to Gram-positive were increased. Therefore, our findings suggested that observed changes in relative abundance was related to the immune response of silkworm to BmCPV infection. Relevance analysis of plenty of the predominant genera showed the abundance of the Enterococcus genus was in negative correlation with the abundance of the most predominant genera. These results provided insight into the relationship between the gut microbiota and development of the BmCPV-infected silkworm.

Abstract:Two strains of mulberry silkworm, namely Pure Mysore and NB4D2, at fifth instar first day were inoculated with 40l of BmCPV suspension from the stock of 1.215 X 106 PIB/ml in 0.75% of NaCl solution by ‘oral injection’. Three batches of such inoculated worms were allowed to continue larval development at 20±1°C, 24±1°C and 30±1°C in separate BOD incubators. The crude extract of midgut tissue was prepared from the survivals and used to count the total number of polyhedra per larva in each batch. Further, during virus multiplication at different rearing temperatures, the total amount of glucose, proteins and amylase as well as succinate dehydrogenase activity levels in haemolymph were studied. The experimental results clearly indicated that the atmospheric temperature has a direct influence on a number of polyhedral formations and biomolecules.

Abstract:The low efficiency of the oral infectivity of recombinant polyhedrin-negative baculovirus is a major bottleneck in the application of the baculovirus expression system in the silkworm (Bombyx mori L). In this study, the effects of a fluorescent whitening agent on improving the oral infection for the recombinant Bombyx mori nuclear polyhedrosis virus in silkworm larva and their possible mechanism were investigated. The results showed that the peroral infection can be remarkably enhanced by adding VBL into the larval artificial diet. The maximum infection rate reached as high as 90% with the concentration of VBL (1%), which was then considered as optimal. The total protease activity and pH value of the larval intestinal juice were found to be lower when compared to the control, indicating an abnormal physiological change of the larval digestive system by VBL, which, in turn, resulted in improved peroral infection of recombinant virus.

Abstract:Sericulture, often known as silk farming is the practice of rearing silkworms for production of raw silk. One of the major impediments faced by the sericulture community is the silkworm diseases that cause significant crop losses. In the present study, the efficacy of phototrophic bacteria as feed supplements in improving the economic traits as well as disease resistance in mulberry silkworm, Bombyx mori L. was assessed. A total of three phototrophic bacteria were isolated and identified as belonging to the genera Marichromatium, Rhodobacter and Rhodopseudomonas based on 16S rRNA gene sequencing analysis. Their morphological, physiological and biochemical characters were studied, grown under ambient conditions and biomass was harvested. Mulberry fortified with the aforementioned phototrophic bacteria in solitude and combinations at varying concentrations (0.5, 1, 2 and 5%) were found to be innocuous to the mulberry silkworm. The best results were observed when mulberry leaf was supplemented with Rhodopseudomonas sp. at 2% concentration which improved survival, cocoon weight, shell weight, shell%, filament length and non-breakable filament length by 5.18, 0.27, 3.86, 3.5, 1.5 and 1.2%, respectively under normal rearing conditions. Under Staphylococcus sp. infected conditions, the aforesaid diet also enhanced survival in silkworms by 16%. On the other hand, the same diet didn’t exhibit any discernible influence on survival against fungal, viral, or microsporidian infections in silkworm.

Abstract:[Aim]Enhancing the yield of silkworm bioreactor and disclosing the relation between the host nutritive condition and the expression of foreign gene in rBmNPV-Bm system.[Methods]The effects of vitamins and inorganic salt in artificial diet for the host insect of baculovirus.silkworm Bombyx mori,on the activity of foreign gene products was studied using BmNPV as an expression vector and phytase as a reporter gone.[Results]The activity of phytase in silkworm haemolymph varied remarkably with the amount of added vitamin and inorganic salt.The phytase activity in silkworm haemolymph reached the maximum when the amount of vitamin C was 1%of the dry weight of artificial diet,vitamin B mixture 0.25%,inorganic salts mixture 1% and the amount of vitamin B6 is 2 mg per 100 g dry matter weight,respectively in the designed test scope.[Conclusions]The expression of phytase gene in BmNPV-Bm expression system could be enhanced efficiently by altering the levels of micronutrients available in the artificial diet for silkworm larvae.

Abstract:Silkworm Bombyx mori L. is one of the most important domesticated insects, which produces luxuriant silk thread in the form of cocoon by consuming mulberry leaves during larval period. The growth and development of silkworm is greatly influenced by environmental conditions. The seasonal differences in the environmental components considerably affect the genotypic expression in the form of phenotypic output of silkworm crop. Bombyx mori nucleopolyhedrovirus (BmNPV) is a pathogen that causes great economic losses in sericulture. In the present investigation, twelve bivoltine silkworm breeds (WM, ND5, NB4D2, U-4, PO1, ND3, U-6, CSR2, SH6, SPO, U-3 and NSP) were studied for temperature and BmNPV stress. After third and fourth moult silkworm larvae were treated with BmNPV inoculum at 25±10C and 30±10C temperature respectively. Increased mortality was observed at 30±10C when compared to 25±10C. However, with respect to breeds the highest total mortality was recorded in breed PO1 and least in breed U-3. Silkworm breed U-3 may have the ability to tolerate BmNPV incidence and can be exploit for evolving disease-tolerant silkworm hybrids.

Abstract:The peritrophic membrane lines the gut of most insects at one or more stages of their life cycles. It facilitates the digestive processes in the guts and protects from invasion by pathogens or food particles. In the current study, a novel PM protein, designated as BmMtch, was identified from the silkworm, Bombyx mori. The open reading frame of BmMtch is 888 bp in length, encoding 295 amino acid residues consisting of two domains (Mito_carr domains) and three transmembrane regions. They are localized on the 11th chromosome as single copy with one exon only. Quantitative real time PCR analysis (qRT-PCR) revealed that BmMtch was mainly expressed in larval fat bodies, Malpighian tubules, testis and ovaries, and could be detected through all stages of the life cycle of silkworm. Immuno-fluorescence analysis indicated that BmMtch was localized within the goblet cell of larval midgut. Western blotting analysis showed that BmMtch were detected in total proteins of PM and larval midgut. The characteristics of BmMtch indicated that BmMtch represents a novel member of insect PM proteins, without chitin-binding domains.

Abstract:The Indian isolate of Bombyx mori bidensovirus (BmBDV) is a bipartite virus that comprises of a segmented, non-homologous, two linear single-strands of DNA molecules (VD1 and VD2). It is one of the causative agents of the fatal silkworm disease ‘Flacherie’ that causes severe crop loss for the sericulture farmers. Genome analyses of the Indian isolate of BmBDV revealed that it consists of 6 putative ORFs similar to the Japanese and Chinese isolates. VD1 consists of 4 ORFs while VD2 has 2 ORFs that code for 4 non- structural (NS) and 2 structural (VP) proteins, in total. In this study, we investigated, in detail, the impact of BmBDV pathogenesis on growth and development of the silkworm Bombyx mori, at different developmental stages. Mortality rate and weight uptake analyses were also performed on newly ecdysed 4th instar larvae. BmBDV infection was not found to be developmental stage specific and it occurred at all stages. Onset of mortality took place 8 days post infection (dpi) and 100% mortality occurred at 11 dpi. The infected larvae showed a significant difference in weight uptake wherein from 7 dpi the larvae stopped gaining weight and from 8th dpi started demonstrating the typical symptoms of flacherie. Further, the expression pattern of the 6 viral ORFs were also investigated in the newly ecdysed 4th instar BmBDV infected silkworms. Among all the six ORFs, VD2 ORF 1 and 2 revealed the highest transcript numbers, which was followed by VD1 ORF 4 that encodes for the viral DNA polymerase enzyme. This was the first ever attempt to understand the pathogenesis and the expression pattern of all the six ORF transcripts of the Indian isolate of BmBDV.

Abstract:The present investigation was carried out to assess the rearing performance and total haemocyte count in silkworm Bombyx mori L. on antibiotic supplemented feed at various concentrations. The study was conducted on the silkworm breed APS-45.The worms were reared upto 3rd moult by feeding on mulberry leaves without any treatment. After 3rd moult, three replications were maintained for each treatment and each treatment received different concentration of antibiotics. For each antibiotic three concentrations (0.05%, 0.10% and 0.15%) were prepared and sprayed on the mulberry leaves, a separate batch was also maintained where only distilled water was sprayed and that served as a control. Among the three antibiotics evaluated ceftiofur sodium showed best results followed by oxytetracycline and enroflaxcin which also showed better results in all the parameters and it was found that improved results were obtained with an increase in concentration. The cocoon yield was statistically significant in all the treatments. However, the highest cocoon yield by weight was obtained in ceftiofur sodium. Higher cocoon weight recorded in these antibiotic treated batches indicated a clear difference in their nutrient utilization efficiency. Present investigation reflected that antibiotics have the potential to be used for enhancing the cocoon and raw silk production.

Abstract:Abstract Bombyx mori L. (Lepidoptera: Bombycidae) nucleopolyhedrovirus (BmNPV) is a serious pathogen causing huge economic losses to sericulture. There is growing evidence that the gut microbiota of silkworms plays a critical role in shaping host responses and interactions with viral infection. However, little is known about the differences in the composition and diversity of intestinal microflora, especially with respect to silkworm strain differences and BmNPV infection-induced changes. Here, we aim to explore the differences between BmNPV-resistant strain A35 and susceptible strain P50 silkworm and the impact of BmNPV infection on intestinal microflora in different strains. The 16S rDNA sequencing analysis revealed that the fecal microbial populations were distinct between A35 and P50 and were significantly changed post BmNPV infection in both strains. Further analysis showed that the BmNPV-resistant strain silkworm possessed higher bacterial diversity than the susceptible strain, and BmNPV infection reduced the diversity of intestinal flora assessed by feces in both silkworm strains. In response to BmNPV infection, the abundance of Muribaculaceae increased in P50 and decreased in A35, while the abundance of Enterobacteriaceae decreased in P50 and increased in A35. These results indicated that BmNPV infection had various effects on the abundance of fecal microflora in different silkworm strains. Our findings not only broadened the understanding of host–pathogen interactions but also provided theoretical help for the breeding of resistant strains and healthy rearing of silkworms based on symbiotic bacteria.
