Abstract:Using the commercially available and economical 6-hydroxycoumarin (6-HC) as the substrate, a dual-emission ratiometric fluorescence sensor was developed to detect tyrosinase (TYR) activity based on 3-aminophenyl boronic acid functionalized quantum dots (APBA-QDs). TYR can catalyze 6-HC, a monohydroxy compound, to form a fluorescence-enhancing o-hydroxy compound, 6,7-dihydroxycoumarin. Owing to the special covalent binding between the o-hydroxyl and boric acid groups, APBA-QDs react with 6,7-dihydroxycoumarin to form a five-membered ring ester dual-emission fluorescence probe for TYR. With an increase in TYR activity, the fluorescence at 675 nm originating from the QDs is gradually quenched, whereas that at 465 nm owing to 6,7-dihydroxycoumarin increases. Referencing the decreasing signal of the dual-emission probe at 675 nm to measure the increasing signal at 465 nm, a ratiometric fluorescence method was established to detect the TYR activity with high sensitivity and selectivity. Under the conditions optimized via response surface methodology, a linear range of 0-0.05 U/mL was obtained for the TYR activity. The detection limit was as low as 0.003 U/mL. This sensing strategy can also be adopted for the rapid screening of the TYR inhibitors.

Abstract:As the biomarker of dermatoses, tyrosinase (TYR) has various biological functions by regulating the production of melanin. Here, we proposed lanthanide luminescent sensors for colorimetric and ratiometric fluorescence dual-readout detection of TYR activity by introducing a characteristic blue fluorescence reaction. With the increase of TYR, the fluorescence signals of lanthanide luminescent probes (dipicolinic acid (DPA)-Eu, DPA-Tb, and DPA-Eu/Tb) are decreased, while the blue signal is turned "on", which improves the sensitivity and selectivity of ratiometric fluorescence sensing, with good linear range and low detection limit (0.05 U mL −1 for the DPA-Eu system or DPA-Tb system, 0.01 U mL −1 for the DPA-Eu/Tb system). In addition, colorimetric analysis of TYR is achieved by recording the absorbance value at 420 nm (The linear range of three system is 0.5–40 U mL −1 ). Moreover, the color changes under natural light and chromaticity changes under UV light facilitate the visual and qualitative analysis of TYR. Furthermore, the lanthanide sensors enable precise quantification of TYR in real samples. Subsequently, arbutin as an inhibitor sufficiently inhibited TYR activity, and arbutin was detected by "turn-off" fluorescence mode. More importantly, this study not only realizes dual-mode detection and inhibition of TYR activity, but also broadens the application of TYR detection in medical research and cosmetics industry, as well as provides a new idea for designing optical sensors.

A fluorescence signal amplification strategy for modification-free ratiometric determination of tyrosinase in situ based on the use of dual-templated copper nanoclusters

Abstract:A fluorescence resonance energy transfer (FRET)-based in situ fluorescence signal amplification strategy is described for the determination of tyrosinase (TYR). In this assay, a dual-templated copper nanocluster (CuNCs) stabilized by bovine serum albumin (BSA) and glycylglycine (Gly-Gly) was used as an energy donor. Metyrosine was employed as a TYR substrate because its enzyme catalytic product (methyldopa) was able to function as a monomer molecule to form fluorescent polymethyldopa (PMeDP) with the assistance of BSA/Gly-Gly CuNCs. In this process, PMeDP can combine with BSA/Gly-Gly CuNCs without extra modification and then acts as an energy receptor, which leads to a remarkable FRET from BSA/Gly-Gly CuNCs to PMeDP. Interestingly, the fluorescence intensity of PMeDP was strengthened greatly in the FRET-based sensor compared to the separate excitation, which provided good sensitivity for TYR sensing. Illuminated under a UV light source, the fluorescence signal change is observed from dark violet to bright green. Therefore, the present sensing system affords a reliable ratiometric assay for TYR determination. Also, the ratio of fluorescence intensity between PMeDP (λem at 505 nm, F505) and BSA/Gly-Gly CuNCs (λem at 415 nm, F415) was used for quantitative determination of TYR. The sensing system was easily operated in aqueous media with an exciting detection limit of 44.0 U L-1. This sensing strategy has been applied to the screening of inhibitors. A Kit for Colorimetric VC Detection Combining with Bionic E-eye

Abstract:Vitamin C (VC) maintains the activity of sulfhydrylase and the reduced state of glutathione, and acts as a detoxifying agent. It is widely found in plant tissues as well as in fresh fruits and vegetables. Accurate determination of vitamin C content is of great significance for diet and health care. In this work, MnO2 nanoflakes as the biomimetic oxidase was used to react with VC and TMB was used as a chromogenic agent. The color of the final solution reflects the concentration of VC. And the bionic E-eye as a portable in-situ detection platform was designed to analyzed the color change in 10 s by photographing. By optimizing experimental conditions, the linear range of this method for VC detection is from 7.8 μM to 62.5 μM and the detection limit 7.65 μM which is much lower than other methods. Besides, this methods shows good selectivity, short reaction time (10 min) and also was used for VC detection in artificial urine samples, which shows great potential in disease monitoring.

Abstract:The abnormal expression or activation of tyrosinase is the pathogenesis of several diseases such as albinism, vitiligo, and melanoma.Detecting and inhibiting tyrosinase activity is of great value in the diagnosis and treatment of these diseases.The detection and inhibition of tyrosinase and its application in whitening products are reviewed, with special emphasis on development of its probes and inhibitors.Tyrosinase is an important enzyme in controlling the formation of melanin in melanosome, and plays key a role in the pigmentation of hair and skin. The abnormal expression or activation of tyrosinase is associated with several diseases such as albinism, vitiligo, melanoma and Parkinson. Excessive deposition of melanin could cause diseases such as freckles and brown spots in human body, and it is also closely related to browning of fruits and vegetables and insect molting. Detecting and inhibiting the activity of tyrosinase is of extraordinary value in the progress of diagnosis and treatment of these diseases. Therefore, many selective optical detection probes and small molecular inhibitors have been developed, and have made significant contributions to the basic and clinical research of these diseases. In this paper, the detection and inhibition of tyrosinase and their application in whitening products are reviewed, with special emphasis on development of fluorescent probes and inhibitors. Abstract:Evolution of fluorometric sensing methods has brought an enormous opportunity and exhibited novel potentials for enzymatic activities determination. Herein, we have established an accurate and straightforward fluorescent sensing approach for tyrosinase (TYR) activity based on the formation of fluorescent silicon nanoparticles (Si NPs). Influenced by TYR-induced hydroxylation of phenol into catechol, and an easy reaction between catechol and 3(2-aminoethylamino) propyl (dimethoxymethylsilane) (AEAPDMMS) to generate Si NPs, we have designed a label-free fluorescence turn-on probe for TYR activity measurement. After optimization of all reaction parameters, the proposed approach could selectively determine TYR activity in the linearity range of 0.08–9 U mL −1 with the limit of detection (LOD) of 0.03 U mL −1 . The practicability of the method in real sample was assessed by measuring TYR activity in human serum sample with successful results. In addition, Si NPs were wonderfully acceptable for Hela and SiHa cells imaging by virtue of their tolerable cytotoxicity and impressive biocompatibility. Therefore, the designed fluorescent biosensor exhibited a potential and widespread implementation in clinical diagnoses assays.

Abstract:It is challenging to develop a selective and sensitive approach for monitoring tyrosinase in cell environment. Herein, a sensing strategy based on ion current rectification induced by nanochannels was established to electrochemically detect tyrosinase. Functionalized alumina nanochannels were decorated by tyramine to selectively determine tyrosinase via specific reaction between phenolic hydroxyl groups of tyramine and tyrosinase. Furthermore, Fe2+ was bonded with dihydroxyl groups generated by tyrosinase in nanochannels to amplify charge changes, enhancing detection sensitivity. The developed electrochemical sensor showed a good linearity towards tyrosinase from 2 to 50 U/mL and was successfully employed to determine tyrosinase in cell lysates.

Abstract:Tyrosinase (TYR) is as a well-known polyphenol oxidase and important biomarker of melanocytic lesions. Thus, developing powerful methods to determine TYR activity is of great value in the early diagnosis of skin disease. Direct surface-enhanced Raman scattering (SERS) detection of biomolecules is usually affected by non-specific interference and complicate structure of the analytes. It is a challenge to develop Raman-active molecules with specific recognition to analytes in complex media. Here, we report a novel colorimetric and surface-enhanced Raman scattering (SERS) dual-readout assay for the determination of TYR using commercially available and economical 4-mercaptophenyl boronic acid (4-MPBA) as a Raman-active and recognition molecule. 4-MPBA provides a unique interactive boronic acid group to the diol group of TYR substrate and exhibits good SERS signal. Also, the introduction of magnetic beads could promptly improve the anti-interference ability of dual-mode sensor. The TYR-incubated tyramine-modified magnetic beads could obviously change the concentration of 4-MPBA-AuNPs in the presence of O2 and ascorbic acid, where the ultraviolet visible (UV-vis) absorption and SERS intensity were directly related to the concentration of TYR added. The dual-mode sensor had a rapid response to TYR within 1 min under optimized conditions and had high selectivity for TYR with a limit of detection at 0.001 U/mL. In addition, the dual-mode strategy showed promising prospects in the determination of TYR activity in serum samples and could be used to screen TYR inhibitors.

Abstract:Tyrosinase (TYR) detection will be crucial in the prevention and early diagnosis of some illnesses, including epilepsy, albinism, and melanoma, due to the close linkage of abnormal expression of TYR with the occurrence of these diseases. Therefore, how to obtain a fluorescent probe that has a high fluorescence quantum yield and high specificity for detecting TYR activity will be of significance. In this work, we improved the rhodamine structure by expanding the π-conjugation system and using asymmetric synthesis to provide a fluorescent probe with good optical properties for the specific detection of TYR. The fluorescence quantum yield of fluorophore released from the probe by the enzymatic reaction with TYR was as high as 0.22, which was significantly greater than that of other previously reported fluorescent probes. We also chose 3-hydroxybenzyl oxygen as the recognition group, which can successfully lessen the interference from reactive oxygen species (ROS). At the same time, the probe demonstrated great sensitivity in detecting TYR activity, and the detection limit of TYR for the probe was calculated to be 0.45 U/mL. In addition, the probe has currently been successfully utilized to evaluate TYR activity in cells. Therefore, this probe is anticipated to be crucial in identifying TYR activity in challenging settings.
Debut of a novel AIE-based fluorescent probe as tyrosinase tracer to image skin pigmentary disorders

Abstract:Tyrosinase (TYR) functions as the critical enzyme in biosynthesis of melanin, and its abnormality is an important hallmark for skin pigmentary disorders. This renders the visualization of tyrosinase dynamics a strategically vital way to facilitate clinical diagnostic and monitoring. However, the use of small molecule fluorescent probes to assess tyrosinase levels and evaluate related therapeutics and drugs has been limited. This is mainly due to the aggregation-induced quenching (ACQ) effects of existing fluorescent probes. In this study, we introduce the first aggregation-induced emission (AIE) small molecule fluorescent probe, BTFTYR, for simple, highly sensitive, and effective detection of tyrosinase activity. The specific fluorescence signal generated by tyrosinase catalytic reaction enables the imaging of pigment cells. We demonstrate its application in visualizing tyrosinase with high contrast in living vertebrates, benefiting from the inherent photobleaching resistance of AIE materials, large Stokes shift, and considerable tissue permeability. We also show the outstanding skin penetration capability of BTFTYR in a skin model. Moreover, we present the first comprehensive evaluation of relevant clinical therapeutic approaches or drugs, including ultraviolet B (UVB) irradiation, kojic acid, and α-MSH, by interpreting the tyrosinase variation with BTFTYR. Finally, our studies confirm that BTFTYR neither causes skin inflammation nor produces visceral toxicity, making it a promising chemical tool for medical diagnosis and drug screening. This tool may further help elucidate the pathomechanisms associated with tyrosinase.

Abstract:Tyrosinase inhibitors have the ability to resist melanin formation and can be used for clinical and cosmetic, so it is becoming extremely crucial to search a rapid and effective method for detecting t the activity of tyrosinase. In this study, a sensing probe based on Nitrogen-doped graphene quantum dots (N-GQDs) were prepared with carbamide and citric acid. Tyrosinase can oxidize dopamine to dopamine quinone, which can quench the fluorescence of N-GQDs based on the principle of fluorescence resonance energy transfer (FRET) process, and then the detection of tyrosinase activity can be achieved. The result demonstrated that the fluorescence intensity of N-GQDs was a linear correlation with the activity of tyrosinase. Wide detection linear ranges between 0.05 and 5 U/mL and high selectivity. The detection range of tyrosinase was 0.05 to 5 U/mL and LOD of 0.005 U/mL. According to the above, the fluorescence method established in this work could be successfully used for the trace analysis of tyrosinase and it was verified that KA is an inhibitor of tyrosinase.

Abstract:Tyrosinase (Tyr) is known as an important cancer-related biomarker, which shows significant over-expression in malignant tumor cells, such as melanoma. Developing highly sensitive and selective fluorescent probe for monitoring tyrosinase in living system is still challenging. In this project, we report a novel fluorescein-based highly chemoselective fluorescent probe (FT-3) capable of tracking tyrosinase in vitro and in vivo in turn-on model. FT-3 contains a 3-hydroxybenzyloxy unit as the tyrosinase recognition moiety, which precluded from interference of other competitive omnipresent ROS in living system and realized the highly chemoselective detection of tyrosinase. Furthermore, the probe worked in aqueous solution and showed more than 24-fold fluorescence enhancement in the presence of tyrosinase, which endows FT-3, an excellent signal to noisy ratio. In addition, we have successfully employed FT-3 to monitoring tyrosinase activity in different living cells and zebrafish models. We believe that our newly designed fluorescent probe will provide an alternative for tracking tyrosinase in complex living system.
Detection of Tyrosinase in Real Food Samples and Living Cells by a Novel Near-Infrared Fluorescence Probe

Abstract:A new near-infrared fluorescence probe was developed and applied to the fluorescence detection of tyrosinase in real food samples and living cells. The probe (E)-2-(2-(6-((3-hydroxybenzyloxy)carbonylamino)-2,3-dihydro-1H-xanthen-4-yl)vinyl)-3,3-dimethyl-1-propyl-3H-indolium (1) was designed and synthesized by coupling 3-hydroxybenzyl alcohol via carbamate bond with an amino hemicyanine skeleton, based on the high anti-interference ability of 3-hydroxybenzyl alcohol to reactive oxygen species and its binding affinity to tyrosinase. Compared with the existing tyrosinase probes, the proposed probe exhibits superior analytical performance, such as high selectivity, high sensitivity, superior spatiotemporal sampling ability, fluorescence signal switching at 706 nm, and low detection limit of 0.36 U mL-1. More importantly, the probe has been successfully used to monitor tyrosinase in the browning of apple slices for the first time, and the results indicated that the strongest fluorescence intensity could be achieved at 2.5 h to realize precise visual recognition of tyrosinase. Notably, the probe determined tyrosinase in real food samples (apple, banana, cheese, and red wine) with a stable average recovery range of 95.7-108.3% and has been successfully used to monitor tyrosinase in the living B16 cells. The superior properties of the probe make it of great potential use in food nutritional value evaluation and clinical diagnosis of melanin-related diseases.

Abstract:Tyrosinase (TYR) is a multifunctional copper-containing enzyme that plays a critical role in the biosynthetic pathway of melanin. Thus, the detection of TYR activity possesses vast importance from clinical diagnosis to the food industry. However, most TYR detection methods are expensive, complicated, and time-consuming. Herein, a functional nanofluidic heterochannel composed of an ultrathin tyramine-modified mesoporous silica layer (Tyr-MS) and alumina oxide (AAO) arrays is constructed by an interfacial super-assembly method. The heterochannel with plenty of enzyme catalytic sites for TYR provides the response of the ion current signal against TYR concentrations. Introducing enzymatic reaction paves the way for the heterochannel to achieve label-free, selective, specific detection of TYR. Notably, a highly sensitive detection of TYR with a limit of 2 U mL-1 was obtained by optimizing the modified conditions. Detailed investigations and theoretical calculations further reveal the mechanism for the detection performance. This work provides a simple, low-cost, quick response, and label-free platform based on functional nanofluidic devices for enzyme-sensing technologies.

Abstract:Rapid-response in situ fluorogenic reactions in aqueous solution are important for designing sensitive and stable sensing platforms. Herein, a wavelength-changeable and rapid-response (within 5 s) fluorescence sensing platform for monitoring tyrosinase (TYR) activity is constructed. The developed assay is based on TYR catalyzing the hydroxylation of mono-phenol to o-diphenol and MnO4--triggered fluorogenic between dopamine (DA) and phenol derivatives in aqueous solution. The fluorescence wavelength can be changeable from 470 to 550 nm with strong fluorescence according to different phenol derivatives. Our proposed sensor not only exhibits a good recovery for TYR in high serum concentration (20 %), but also has been successfully applied to the screening of TYR inhibitors modeled on kojic acid. Furthermore, a paper-based wavelength-changeable fluorescence sensor was developed for on-site detection of TYR activity in potatoes with high recovery, which is consistent with our previously reported method. Consequently, the proposed sensing system has broad prospects in the practical application of TYR-associated food monitoring and clinical diagnosis.

This luminescent nanozyme is composed of luminescent Tb3+, catalytic Fe3O4 nanoparticle and antenna ligand 4,5-imidazoledicarboxylic acid (IDA) through flexible MOF (metal-organic framework) structure, and has dual functions of luminescence-sensing and catalysis like peroxidase. The catalytic reaction-triggered-luminescence of Tb3+ ion is able to sense pM level of tyrosine in human serum. Such a luminescent nanozyme can be used to replace the combination of natural enzymes and chromogenic agents currently widely used in enzyme-based assays, providing a promising prospect for innovating various enzyme-based assays.

Abstract:Dopamine (DA) is an electrochemically active compound. In this work, we reported an electrochemical affinity biosensor for the detection of tyrosinase activity with DA-modified gold nanoparticles (DA-AuNPs) as redox probes. Hydrophilic boronic acids are well known to form tight diol-boronate ester covalent bonds with members of the diol family. In the sensing platform, monophenols of tyrosine residues immobilized onto gold electrode were catalytically oxidized by tyrosinase to o-diphenols. The resultant o-diphenols captured 4-mercaptophenylboronic acid (MBA)-capped AuNPs (MBA-AuNPs) through the formation of tight covalent bonds between o-diphenols and boronic acids of MBA-AuNPs. Electrochemically active DA-AuNPs were then attached by the anchored MBA-AuNPs via the interaction of MBA with DA, which facilities the amplified voltammetric detection. The current increased with the increase of tyrosinase concentration. The well-defined redox waves and high sensitivity resulting from dual-amplification of MBA-AuNPs and DA-AuNPs demonstrated that this method would be valuable for probing the activity of enzymes by generating a diol-containing substrate, and designing new types of electrochemical biosensor for sensitive detection of diol derivatives.

Abstract:Tyrosinase (TYR), an important biomarker for melanoma, offered significant information early detection of melanoma and may decrease the likelihood of mortality. Therefore, this article constructed a highly sensitive and selective green fluorescent functionalized carbon quantum dots (TYR-CQDs) for tyrosinase (TYR) activity detection by one-step hydrothermal protocol utilizing catechol, citric acid and urea as precursors. The prepared TYR-CQDs illustrated excellent linear relationship and broad linear range with a low detection limit, which exhibited high accuracy and recovery in quantitative determination of TYR in human serum samples. Furthermore, the TYR-CQDs had successfully realized intracellular TYR detection owing to excellent biocompatibility, high anti-interference ability and good cellular imaging capability, suggesting the potential biomedical applications in early diagnosis of melanoma and other tyrosinase-related diseases.

Abstract:Tyrosinase (TYR) is an important polyphenolic oxidase enzyme and usually regards as a biomarker of melanoma cancer. Highly effective tracking TYR activity in vivo will help to study the mechanism of TYR in living organisms and forecasts related diseases. In this study, we present a novel TYR-activatable fluorescent probe (CHMC-DOPA) for tracking TYR activity in vitro and in vivo. CHMC-DOPA is constructed by incorporating dopamine (DOPA) moiety into a fluorescent chloro-hydroxyl-merocyanine (CHMC) scaffold. Upon exposure to TYR, the dopamine unit in CHMC-DOPA is oxidized to a dopaquinone derivative, and an intramolecular photo-induced electron transfer (PET) process between CHMC fluorophore and o-dopaquinone will take place, the fluorescence of CHMC-DOPA is quenched rapidly. Therefore, the evaluation of TYR activity is established in terms of the relationship between fluorescence quenching efficiency and TYR activity. In our experiments, CHMC-DOPA shows various advantages, such as fast response (8 min), low concentration of TYR activation (0.5 U/mL), good water-solubility, as well as the lowest detection limit (0.003 U/mL) compared with previously reported works. Furthermore, CHMC-DOPA also exhibits excellent cell membrane permeability and low cytotoxicity, which is successfully used to monitor endogenous TYR activity in living cancer cells and zebrafish models. CHMC-DOPA performs well, and we anticipate that this newly designed novel platform will provide an alternative for high effective monitoring TYR activity in biosystems.
