Abstract:The development and application of nano-drug carriers might provide an excellent opportunity for cancer therapy. However, it is still an important challenge to realize the regulation and control of drug loading by analyzing the assembly process of carrier-loaded drugs. Herein, we show a "self-contained bioactive nanocarrier" system, which is prepared from ursolic acid, one of the very promising biologically active natural products with self-assembly properties. The study decrypts the assembly process of drug-carrier interaction and achieves the regulation of drug loading by controlling the interaction force. This nanocarrier highlights the unique advantages of active natural products in therapeutic efficacy and health benefits. In antitumor experiments, the carrier and drug demonstrated synergistic therapeutic efficacy. Furthermore, the nanocarrier is biosafe and capable of reducing the risk of liver damage induced by chemotherapeutics through the upregulation of key antioxidant pathways. Taken together, this "self-contained bioactive nanocarrier" system makes up for the drawback that conventional nanocarriers have no therapeutic efficacy and health benefits and eliminates the trouble of the toxic side effects associated with chemotherapy agents and the additional toxicity caused by long-term use of nanocarriers.

Abstract:Active ingredients from Chinese medicine (AIFCM) are emerging as a class of anticancer agents for treating hepatocellular carcinoma (HCC). AIFCM have unique advantages over traditional chemotherapeutic drugs, such as their multi-target, multi-level, and coordinated intervention effects against HCC. Recent researches demonstrated their potentials in inhibiting HCC initiation, cell proliferation, angiogenesis, and metastasis. With the advantages of nano-carriers including better bioavailability, significant features of passive or active targeting and stimulus-responsive drug release. Formulation of AIFCM and nano-carriers shows great potential in promoting anti-HCC therapeutic development. Here, we critically assess nano-carriers for delivery and targeting of AIFCM for HCC therapy. First, we offer an overview of major pathogenic factors to induce HCC, including virus infection, alcoholic liver disease, mycotoxin, obesity, and type 2 diabetes. Second, the mechanisms of AIFCM for HCC therapy comprising gene regulation and transduction of different molecular pathways are discussed. Third, significant achievements of AIFCM-encapsulated nano-carriers and AIFCM-modified nano-carriers against HCC are presented. Finally, we provide concluding remarks and future perspectives of AIFCM for HCC therapy, hoping to offer some insight on how to improve AIFCM-based HCC therapy.

Abstract:Nanomedicine of synergistic drug combinations has shown increasing significance in cancer therapy due to its promise in providing superior therapeutic benefits to the current drug combination therapy used in clinical practice. In this article, we will examine the rationale, principles, and advantages of applying nanocarriers to improve anticancer drug combination therapy, review the use of nanocarriers for delivery of a variety of combinations of different classes of anticancer agents including small molecule drugs and biologics, and discuss the challenges and future perspectives of the nanocarrier-based combination therapy. The goal of this review is to provide better understanding of this increasingly important new paradigm of cancer treatment and key considerations for rational design of nanomedicine of synergistic drug combinations for cancer therapy.
Nanomedicine-cum-Carrier by Co-assembly of Natural Small Products for Synergistic Enhanced Antitumor with Tissues Protective Actions.

Abstract:The application of natural small products with self-assembly characteristics in drug-delivery system is attractive for biomedical applications, because of its inherent biological safety and pharmacological activity, and there is no complex structural modification process. However, drug carriers with pharmacological effects have not been developed enough. Here, we report a pure natural Nanomedicine-cum-carrier (NMC) drug delivery system. The NMC is formed by direct co-assembly of two small molecular natural compounds through non-covalent interaction, and a molecular dynamics model for predicting the co-assembly of two small molecular compounds was established. Not only the system shows excellent stability, high drug loading and sustained release characteristics, but also the co-assembled NMC formed by two small molecular compounds has synergistic antitumor effect (CI < 0.7). After drug loading, the antitumor effect is further improved. In addition, this NMC highlights the unique advantages of active natural products in biosafety and health benefits. Compared with free drugs, it can reduce the liver damage caused by chemotherapy drugs through up-regulating key antioxidant pathways. Compared to non-pharmacologically active drug delivery systems, it can reduce the risk of nanotoxicity. Taken together, this co-assembly drug-carrier system breaks the shortcomings that pharmacological active compounds cannot be directly applied, enhances the pharmacological activity of bioactive drug carriers, improves the antitumor efficacy, and slows down the side effects induced by chemotherapy drugs and the additional toxicity caused by long-term use of non-bioactive nanocarriers.

Abstract:Hepatocellular carcinoma (HCC) is one of the most common malignant tumors worldwide, with high rates of recurrence and death. Surgical resection and ablation therapy have limited efficacy for patients with advanced HCC and poor liver function, so pharmacotherapy is the first-line option for those patients. Traditional antitumor drugs have the disadvantages of poor biological distribution and pharmacokinetics, poor target selectivity, high resistance, and high toxicity to nontargeted tissues. Recently, the development of nanotechnology has significantly improved drug delivery to tumor sites by changing the physical and biological characteristics of drugs and nanocarriers to improve their pharmacokinetics and biological distribution and to selectively accumulate cytotoxic agents at tumor sites. Abstract:Cancer, once considered as an incurable disease, today, becomes not that difficult to be treated due to the newly emerging nanotechnology and the rapid development of “smart” drug delivery systems. New cancer therapeutics ought to overcome the defects of conventional drug delivery systems, such as nonspecific bio-distribution or targeting, short circulation time, etc. Tumor targeted drug delivery systems basically includes two strategies: passive and active targeting. Passive targeting is often referred to the enhanced permeability and retention effect (EPR effect) due to the enhanced vascular penetration of tumor capillaries and limited lymphatic drainage of lymphatic capillaries. Active targeting systems are based on local stimuli characteristic of the target pathological zone (such as, increased temperature or lowered pH values, redox potential changes, characteristic of inflamed, the receptor mediated internalization by target cells, etc.). Intriguingly, the combination of the two strategies above is promising and exciting. Herein, we give a brief review of tumor targeted drug delivery systems based on tumor cell itself and its external milieu. Abstract:Chemotherapy is one of the important means of tumor therapy. However, most of the anti-tumor drugs that currently used in clinic are hydrophobic non-specific drugs, which seriously affect the efficacy of drugs. With the development of nanotechnology, drug efficacy can be improved by selecting appropriate biodegradable nanocarriers for achieving the controlled release, targeting and higher bioavailability of drugs. This paper reviewed the research progress of anti-tumor drug nanoparticle carriers, which mainly summarized the materials used for anti-tumor drug nanoparticle carriers and their effects in anti-tumor drugs, as well as the targeted drug delivery methods of anti-tumor drugs based on nanocarriers.</p>

Abstract:Hepatocellular carcinoma (HCC), due to the lack of efficient diagnostic methods and short of available treatments, becomes the third main cause of cancer deaths. Novel treatments for HCCs are thus in great need. The fast-growing area of drug delivery provides intriguing possibility to design nanocarriers with unique properties. The nanocarriers performanced as drug deliver vehicles enable the design of diverse drug delivery systems, which could serve multiple purposes, including improved bioavailability, controlled or triggered release and targeted delivery, leading to enhanced drug efficacy and lowered drug toxicity. This paper provides an overview on the types of delivery vehicles, functions of drug nanocarriers and types of ligand-based targeting systems and highlights the advances made towards better HCC treatments.

Abstract:Nanoformulations with advantages in drug delivery, safety and pharmacodynamics have been booming as a promising strategy for cancer therapy. However, the traditional nanocarrier still suffers from the low drug loading capacity, potential systematic toxicity, unclear metabolism, and other uncertainties. To overcome these issues, carrier-free nanodrugs with desirable bioactivity were developed rapidly and drawn considerable attention. Meanwhile, the multifunctional self-delivery nanoarcheticture fabricated by a simple and "green" method, has significant advantages in synergistic cancer therapy and inhibition of multidrug resistant (MDR). Till now, carrier-free nanoparticles for tumor theranostics, phototherapy, chemotherapy, diagnose and synergistic therapy, have made outstanding progress. In this review, we make an integrated and exhaustive overview of lately reports on carrier-free nanodrug delivery systems formed by several active agents. We summarize the self-assembly and modified strategies, with emphasis on application superiority of carrier-free nanocrystal, and give new insight into the establishment of ideal nanosystems for cancer treatment.

Abstract:Anticancer nanomedicines must overcome a series of physiological and pathological barriers to reach the site of action, and this often leads to low overall therapeutic efficiency. Herein, we present tumor acidity regulated rapid aggregation of nanocarriers around tumor blood vessels to construct drug reservoirs for on-demand drug delivery. We designed two nanocarriers, one that protonates extremely quickly to expose click reactive groups on its surface and that subsequently reacted with the other noncarrier via click chemistry to rapidly form ag-glomerates as drug reservoirs. In proof-of-concept experiments, vascular disrupting agent combretastatin A4 (CA4) and hypoxia-activated prodrug PR-104A were loaded into the two nanocarriers, respectively. CA4 was enriched near blood vessels while PR-104A diffused deep into tumors and was activated by profound hypoxia levels induced by CA4. This combination produced effective adaptive therapy for tumors with different hypoxia levels.

Abstract:Carrier-free nanomedicines without structural modification are attractive for the development of natural small molecules (NSMs) and biomedical applications. Moreover, the combination of NSMs is expected to obtain nanomedicines with high efficacy and low side effects due to their inherent pharmacological activities and health benefits. However, poor water solubility and low bioavailability of NSMs limit their wider biomedical and clinical applications. In this study, we revealed the co-assembly properties of pentacyclic triterpenoids and constructed a series of carrier-free nanodrugs, which are co-assembled nanoparticles (NPs) formed by the combination of two NSMs via a supramolecular assembly strategy. Experimental work and simulation studies were combined to reveal the co-assembly mechanism of non-covalent interactions between NSMs. Not only do co-assembled NPs have rapid cellular uptake ability and passive targeting tumor ability based on the EPR effect, but also their constituent units could arrest the cell cycle at different stages of tumor cells and induce apoptosis, showing synergistic anti-tumor effects (CI < 0.7). Compared with self-assembled NPs and positive control, co-assembled NPs show the strongest therapeutic effect in vivo. Importantly, the co-assembled NPs highlight the unique advantages of NSMs in terms of biosafety and health benefits, and systemic toxicity and histological examination confirm that co-assembled NPs have reliable biosafety, and no side effects and nano toxicity risks were observed.

Abstract:The nanocarrier-based delivery system has emerged as a promising candidate for cancer therapy; nevertheless, their quality problems, variation between batches, and carrier-related toxicity issues have restricted their clinical utilization. Compared with traditional carrier-based nanoparticles, carrier-free nanodrug delivery systems preferred to overcome all these drawbacks and will have a wide range of applications in biomedicine and nanotechnology. Herein, we developed a novel carrier-free nanodrug Asp-UA consisted of the classical drug aspirin and the natural plant drug UA via a green and simple approach. The Asp-UA NPs were investigated for shape, particle size, zeta potential, stability, and UV-vis spectroscopy absorption. Cellular uptake study showed that Asp-UA NPs could be easily internalized by HepG2 cells; cellular study demonstrated that Asp-UA NPs held better inhibitory efficiency on tumor metastasis with low toxicity in vitro and in vivo. Moreover, Asp-UA NPs could obviously suppress the progress of cancer metastasis by H22 cells in vivo. Overall, Asp-UA NPs possess a variety of advantages and hold promise to become an alternative to the treatment of cancer metastasis.

Abstract:The encapsulation of therapeutic agents into nano-based drug delivery system for cancer treatment has received considerable attention in recent years. Advancements in nanotechnology provide an opportunity for efficient delivery of anticancer drugs. The unique properties of nanoparticles not only allow cancer-specific drug delivery by inherent passive targeting phenomena and adopting active targeting strategies, but also improve the pharmacokinetics and bioavailability of the loaded drugs, leading to enhanced therapeutic efficacy and safety compared to conventional treatment modalities. Small molecule drugs are the most widely used anticancer agents at present, while biological macromolecules, such as therapeutic antibodies, peptides and genes, have gained increasing attention. Therefore, this review focuses on the recent achievements of novel nano-encapsulation in targeted drug delivery. A comprehensive introduction of intelligent delivery strategies based on various nanocarriers to encapsulate small molecule chemotherapeutic drugs and biological macromolecule drugs in cancer treatment will also be highlighted.

Abstract:Nanoparticles can be enriched at tumor site and improve the therapeutic efficacy of many chemotherapy drugs with the well-known enhanced permeability and retention (EPR) effect. While conventional preparations of materials for nanoscale drug delivery system mainly focused on chemical synthesis, recently the combination of synthetic carrier and natural biomimetic carrier has gained more and more attention. As a new generation of biomimetic nanoparticles, cell membrane-coated nanoparticles combine the complex biological functions of natural membranes and the physicochemical properties of synthetic nanomaterials for a more effective drug delivery. Herein, we briefly review the recent advances on cell membrane-coated nanoparticles for tumor-targeted drug delivery via the prolonging systemic circulation lifetime and the active targeting effect. Since the preferential accumulation of cell membrane-coated nanoparticles in tumor site, they are able to improve the therapeutic efficacy of conventional chemotherapy drugs in antitumor treatment as well as to reduce the systemic toxicity. We also introduce a systematic targeted strategy for the promising application of this platform on brain tumors.

Critical Reviews in Therapeutic Drug Carrier Systems

Abstract:Nanotechnology has a profound effect on many areas of scientific research. Having grown exponentially, the focus of nanotechnology has been on therapeutic activity, such as cancer treatment. Lipid-based nanocarriers have attracted increasing scientific and commercial attention in the last few years as alternative carriers for the delivery of anticancer drugs. Lipid-based nanocarriers have played significant roles in the formulation of anticancer drugs to improve therapeutics. Shortcomings frequently encountered with anticancer compounds, such as poor solubility, normal tissue toxicity, poor specificity and stability, as well as the high incidence rate of drug resistance, are expected to be overcome through use of lipid-based nanocarriers. In this review, the advantages and methods of using nanocarriers to improve cancer treatment efficiency will be discussed. In addition, types of lipid-based nanocarriers are presented and hotspots in research are highlighted. It is anticipated that, in the near future, lipid-based nanocarriers will be further improved to deliver cytotoxic anticancer compounds in a more efficient, specific and safe manner.

Abstract:Clinical applications of many anti-cancer drugs are restricted due to their hydrophobic nature, requiring use of harmful organic solvents for administration, and poor selectivity and pharmacokinetics resulting in off-target toxicity and inefficient therapies. A wide variety of carrier-based nanoparticles have been developed to tackle these issues, but such strategies often fail to encapsulate drug efficiently and require significant amounts of inorganic and/or organic nanocarriers which may cause toxicity problems in the long term. Preparation of nano-formulations for the delivery of water insoluble drugs without using carriers is thus desired, requiring elegantly designed strategies for products with high quality, stability and performance. These strategies include simple self-assembly or involving chemical modifications via coupling drugs together or conjugating them with various functional molecules such as lipids, carbohydrates and photosensitizers. During nanodrugs synthesis, insertion of redox-responsive linkers and tumor targeting ligands endows them with additional characteristics like on-target delivery, and conjugation with immunotherapeutic reagents enhances immune response alongside therapeutic efficacy. This review aims to summarize the methods of making carrier-free nanodrugs from hydrophobic drug molecules, evaluating their performance, and discussing the advantages, challenges, and future development of these strategies.

Abstract:There are numerous possible implications of nanotechnology in cancer therapy. To improve the therapeutic benefits of anticancer drugs nano-delivery systems (NDS) have been established with significant solubility, degradability and cytocompatibility. These NDS are the result of several advantages that high-performance nanoparticles hydroxyapatite (HA) advance rapidly in specific cancer treatments. Salinomycin is a pharmaceutical drug that serves to suppress tumour cells and the growth of many cancers as a chemical inhibitor. Nevertheless, Salinomycin's low bioavailability is the main challenge to be used. In this work, HA chitosan-coated nanoparticles were utilised to enhance the efficiency and availability of Salinomycin as a pH-sensitive biopolymer. HA nanoparticles were initially produced using the sol-gel technique for this purpose. Then a coating of chitosan was coated with the nanocarrier under regulated circumstances, and the salinomycin medicine was encapsulated. For the characterization of nanocarriers were employed techniques such as scanning electron microscopy, X-ray diffraction, and Fourier transformations infrared spectroscopy. Different bioassays have tested nano-drugs developed in the second part. In this connection, we examined the cytotoxicity rate of the hepatocellular carcinoma HepG2 and Caco-2 by the MTT assay and the apoptosis induction rate by ethidium bromide and acridine orange staining method. It has been established that histological analysis is very suitable for nanomedicine, which does not cause apparent stress like swelling and inflammation organs of the mouse. The findings could offer solid evidence for nanoparticles' apoptotic cell death mechanism and a novel cancer therapy approach.Graphic

Carrier-free nanodrugs are emerging safe drug delivery systems, which are composed of multiple components, such as drugs, bioactive molecules and functional ingredients, avoiding the usage of inert carrier materials and offering advantages that include high drug loading, low toxicity, synergistic therapy, versatile design, and easy surface functionalization. Therefore, how to design multicomponent carrier-free nanodrugs is becoming a priority. In this review, the common strategies of rapid construction of multicomponent carrier-free nanodrugs are briefly explored from the perspective of methodology. The properties of organic-organic, organic-inorganic and inorganic-inorganic multiple carrier-free nanosystems are analyzed according to wettability and in-depth understanding is provided. Further advances in the applications of multiple carrier-free nanodrugs are outlined in anticipation of grasping the intrinsic nature for the design and development of carrier-free nanodrugs.

Abstract:With the advancements in nanotechnology and biomaterials science, the development of nanodrug delivery systems (Nano-DDSs) has provided opportunities for the realization of precise targeted treatment of malignant tumors. Liposomes have become a type of DDS with early clinical application and mature development due to their excellent tissue-targeting capacity and outstanding biocompatibility. However, several obstacles remain, such as recognition and clearance by the immune system, a short half-life, and poor tumor targeting. To address these problems, we propose a new method to transform liposomes, using fusion to reassemble the extracted natural cell membranes and artificial phospholipids to form a composite nanolipid carrier (recombined lipid nanocarriers (RLNs)). We evaluated the different types of cell membrane composite lipid nanocarriers based on parameters such as particle size, stability, drug loading and release capabilities, in vitro and in vivo tumor-targeting efficacy, and safety. The results indicated that these novel tumor cell-derived membrane fusion lipid nanocarriers exhibited promising antitumor effects and safety profiles, offering insights for precision cancer treatment.
