Abstract:The influences of xylose, glucose, arabinose, fructose and the inhibitors from corn cob hemicellulosic hydrolysate on xylitol production by Candida sp. were investigated. The optimum initial xylose concentration in the culture media was about 100g/L. The inhibitions of acetic acid and furfural in the hydrolysate increased gradually when their concentrations exceeded 1g/L. An increase in the initial pH value from 4.0 to 6.0 effectively reduced the inhibition of acetic acid. A moderate concentration of K~+, Mg~(2+)(especially), Na~+ and H_2PO_4~- enhanced xylitol fermentation. When the salt concentration of 0~4g/L, both the cell growth and the fermentation were strongly suppressed. Synthetic media composed of hemicelluosic sugars were used for xylitol fermentation. The time course of xylitol fermentation on synthetic media was in good agreement with that on the hydrolysate. The results can greatly facilitate further study on the kinetics of xylitol fermentation from hemicellulosic hydrolysate.
Ethanol Production from Hemicellulosic Hydrolysate by a Recombinant Yeast

Abstract:Hemicellulose in corn stover contains mostly pentose (primarily xylose), which can not be utilized by the yeast Sacchromyces cerevisiae. The ethanol production by fermentation of corn stover hemicellulosic hydrolysate with a recombinant yeast Sacchromyces cerevisiae ZU-10 was investigated. The effects of three major fermentation inhibitors in hemicellulosic hydrolysate, such as sulfate ion, acetic acid and furfural, on the growth of yeast cells were examined; and it was found that, for the above three inhibitors, the endurable concentration limits of the yeast Sacchromyces cerevisiae ZU-10 cell are 4 g·L·1, 0.25 g·L·1 and 0.08 g·L·1, respectively. With different operation conditions, the hydrolysis processes of the hemicellulose of corn stover were carried out, and the results show that, using 1% sulphuric acid (solid-liquid ratio of 1:10), 95℃ and 12 h, the hydrolysis yield of hemicellulose can reach 93% with relatively low concentration of inhibitors. The hemicellulosic hydrolysate is available for ethanol production after treating with lime-neutralization, rotor-evaporation and ion-exchange. The sugar concentration of the hemicellulosic hydrolysate correlates with the concentrated degree of the hydrolysate and the concentration of inhibitors in the hydrolysate; and the found suitable sugar concentration of hemicellulosic hydrolysate used for fermentation by the recombinant yeast Sacchromyces cerevisiae ZU-10 is 80 g·L·1. Anaerobic fermentation of the above hemicellulosic hydrolysate was carried out at 30℃ with the inoculums of 1.2 g·L·1 (calculated on dry weight of cells); after 96 h, the ethanol concentration reaches 31.05 g·L·1., and 95.85% xylose is utilized. The results of this study have great significance for the utilization of hemicellulose resource and accelerating the industrialization progress of producing the bio-ethanol from corn stover.

Abstract:We studied the effects of components in lignocellulosic ethanol fermentation broth on ethanol transfer through polydimethylsiloxane?silicalite?1 pervaporation membrane to tackle product inhibition by integrated with ethanol fermentation. Yeasts,corn stover residues and salts could increase the ethanol flux and selectivity through the membrane, whereas glucose and glycerol had negative effects on membrane performance?Lignocellulosic hydrolysates, such as furfural and hydroxyacetone, slightly influenced separation performance?In batch fermentation, ethanol productivity reduced from 12?95 to 10?22 g/( L·h) after 20 h, and became 0 after 60 h?The glucose consumption curve also confirms the same tendency?However, the ethanol fermentation integrated with pervaporation gave a stable ethanol productivity about 13?30 g/( L·h)?Experiments further proved that integrated process of lignocellulosic ethanol fermentation and ethanol?selected pervaporation through polydimethylsiloxane?silicalite?1 membrane could eliminate product inhibition, improve ethanol productivity and glucose conversion rate very effectively.
The Effects of Dilute Acid Hydrolysate By-products of Corn Stover on Ethanol Fermentation of Xylose-utilising Saccharomyces Cerevisiae 6508-127

Abstract:During the dilute acid pretreatment of lignocellulosic materials such as corn stover, hemicellulose is hydrolyzed into monosaccharides, and meanwhile, toxic by-products are simultaneously generated, which may influence ethanol fermentation thereafter. Studies on the inhibitory effects of the by-products on ethanol fermentation are of practical use for further improvement of ethanol yield from lignocellulosic materials. Five by-products, including acetic acid, formic acid, vanillin, furfural and 5-hydroxymethylfurfural, were identified to be the main components in the hydrolysate of dilute acid pretreatment of local corn stover, which were added into the medium at different concentrations to study their impacts on the growth and ethanol fermentation of a recombinant xylose-utilizing yeast strain, S. cerevisiae 6508-127. The ethanol production was inhibited by formic acid and acetic acid to a lesser extent than that to the growth, and formic acid was shown to be much more toxic than acetic acid, showing severe inhibitory effects at the concentration of 1g/L, half of the concentration for acetic acid which showed remarkably negative effects on ethanol fermentation. Vanillin caused a much longer lag-phase in growth when the concentration was 2g/L, and the lag-phase was not obvious at lower concentrations. At the concentration of 6g/L, vanillin completely inhibited the fermentation as well as the cell growth. 5-Hydroxymethylfurfural was showed to remarkably inhibit ethanol production, but the biomass yield was higher by exogenous addition of 5-Hydroxymethylfurfural than control. Furfural at 0.5~1.5g/L inhibited the cell growth, but the ethanol yield was higher than that of the control experiment. It was also found that vanillin, furfural and 5-hydroxymethylfurfural could be assimilated and metabolized by S. cerevisiae 6508-127 under the experimental conditions.

Abstract:Compounds inhibitory to enzymatic hydrolysis and fermentation are generated from neutral steam exploded corn stover in the process of producing bio-ethanol. In this study, weak acids were identified as main yeast inhibitors, while phenols and aldehyde contribute to the inhibition to a lower degree. Main weak acids in hydrolysates are acetic acid and formic acid, for which critical levels for yeast inhibition are 6 and 4g/L, respectively. The inhibitory effect of these compounds can be greatly overcome by increasing pH of hydrolysates to 6.0–9.0, but there is a risk of bacterial contamination when fermenting at high pH. The relationship of pH, total solids of hydrolysates, fermentation and contamination was studied in detail. Results indicate that the contamination by bacteria when fermenting at high pH can be prevented effectively using hydrolysates with total solids of more than 20%. Meanwhile, ethanol yield is improved significantly.

Abstract:The pretreatment of lignocellulosic biomass is important for efficient bioethanol conversion, but causes undesirable by-products that inhibit microbial growth, conversely affecting the bioconversion efficiency. In this study, the main inhibitors derived from dilute phosphoric acid plus steam-exploded poplar wood were identified as 0.22 g/L furfural, 3.63 g/L acetic acid, 0.08 g/L syringaldehyde, etc., indicating the green nature and low toxicity of the pretreatment process. The effects of the three typical inhibitors (furfural, acetic acid, and syringaldehyde) on Saccharomyces cerevisiae 1517RM growth were analyzed and shown to prolong the lag phase of microbial growth to different degrees. In all the inhibitor groups, the ergosterol secretion was boosted, indicating low cell membrane fluidity and robustness of the strain to an adverse environment. The cell electronegativity and morphology of S. cerevisiae 1517RM also changed under different growth conditions, which was helpful for monitoring the physicochemical properties of cells. Furfural, acetic acid, and syringaldehyde had a synergistic effect on each other, providing an important reference to improving the subsequent ethanol fermentation process.

Abstract:The present study investigated the effects of formic acid, acetic acid, furfural, 5-HMF, and ethanol on activity of two commercial cellulases from Novozyme and Youtell. The carboxylic acid (formic acid and acetic acid) showed little impact on cellulose hydrolysis, but furan derivate (furfural, 5-HMF) performed higher inhibitory effects. The significant decrease of enzyme activity (Novozyme 84%, Youtell 75.8%) happened as addition of 6 g/L furfural. The synthetic solution containing four inhibitors with similar concentration as the acid-pretreated corn stover hydrolysate decreased enzyme activity by ~10%. But the real pretreatment liquid significantly decreased the enzyme activity by ~50% (Novozyme) and ~53% (Youtell). Ethanol (12%) also cut the enzyme activity down by 45%. These results suggested that the cellulase activity may be hindered by many potential inhibitors, which would determine the proper fermentation types between simultaneous saccharification and fermentation (SSF) and separate hydrolysis and fermentation (SHF).

Abstract:The effect of NaOH concentration in pretreatment on fermentation inhibitor components in rice straw hydrolysis was evaluated using GC-MS.The analysis provided theoretical support to eliminate the fermentation inhibition of hydrolysis and enhance ethanol productivity.The results showed that inhibitors were more evenly distributed in control group, alkanes (31.31％) , acids (21.06％) , ketones (18.81％) and phenols (11.09％) were the main components.After alkali-pretreatment, inhibitors were intensively distributed in hydrolysis, in which phenols, acids and ketones were the main components;phenols were absolutely the most dominated components which could achieve 84.69％ maximum.Moreover, the concentration of phenols in hydrolysis increased as alkali concentration increased.However, the effect of alkali concentration on total inhibitors contents in hydrolysis was not obvious.Taken toxicity and concentration of inhibitors into consideration, phenols were the strongest inhibitors in alkali pretreatment hydrolysis.The hydrolysis was less influenced by inhibitors with 2％ NaOH.At the above pretreatment condition, Ferulic acid, n-Hexadecanoic acid and Megastigmatrienone need to be selectively removed from the hydrolysis.

Abstract:In the process of producing H2 from lignocellulosic materials, inhibitory compounds could be potentially formed during pre-treatment. This work experimentally investigated the effect of lignocellulose-derived inhibitors on growth and hydrogen production by Thermoanaerobacterium thermosaccharolyticum W16. Representative compounds presented in corn stover acid hydrolysate were added in various concentrations, individually or in various combinations and subsequently inhibitions on growth and H2 production were quantified. Acetate sodium was not inhibitory to T. thermosaccharolyticum W16, rather than it was stimulatory to the growth and H2 production. Alternatively, furfural, hydroxymethylfurfural (HMF), vanillin and syringaldehyde were potent inhibitors of growth and hydrogen production even though these compounds showed inhibitory effect depending on their concentrations. Synergistic inhibitory effects were exhibited in the introduction of combinations of inhibitors to the medium and in hydrolysate with concentrated inhibitors. Fermentation results from hydrolysates revealed that to increase the efficiency of this bioprocess from corn stover hydrolysate, the inhibitory compounds concentration must be reduced to the levels present in the raw hydrolysate.
Effect of lignocellulose-derived inhibitors on the growth and D-lactic acid production of Sporolactobacillus inulinus YBS1-5

Abstract:The impact of lignocellulose-derived inhibitors on the cell growth and D-lactic production of Sporolactobacillus inulinus YBS1-5 was investigated. At high concentrations, both furans and phenolics, such as furfural, HMF, syringaldehyde and vanillin, affected cell growth and D-lactic acid production and syringaldehyde exhibited the highest. Further experiments showed that only vanillin caused cellular membrane damage. Based on the Biolog approach, in vivo studies on intact S. inulinus cells indicated that phenolics had a stronger inhibitory effect than furan derivatives on the metabolic activity of the concerned substrates related with the key enzymes of D-lactic acid fermentation. The direct in vitro inhibitory effect of the model compounds on the four key enzymes displayed similar patterns. Syringaldehyde was the strongest inhibitor. In general, comparison with published results for other microorganisms indicated that strain YBS1-5 was a robust microorganism against inhibitors of lignocellulose hydrolysate. Notably, in concentrated corn stover hydrolysate, S. inulinus YBS1-5 produced 70.7 g/L D-lactic acid, which was 87.7 % of the yield from the control experiment. However, the fermentation time was prolonged 36 h. In order to improve fermentation rate, a detoxification technology or more robust mutant to phenolics especially syringaldehyde should be developed.
Fermentable Sugars and Microbial Inhibitors Formation from Two-Stage Pretreatment of Corn Stalk with Variation in Particle Size and Severity Factor

Abstract:Microbial inhibitors including weak acids, furan derivatives and phenolic compounds are key problems of cellulosic bio-fuels production by fermentation.Most of these inhibitors are sugars and lignin degradation compounds, which are almost unavoidable during pretreatment processes.While, most of the one stage pretreatment has been conducted at high severity factors of 3.5 or more to get high sugar yield, with increase in severity factor, high concentration of microbial inhibitors were formed and significantly affected downstream biofuel yield.Thus, a two-stage pretreatment of corn stalk, hydrothermal followed by oxalic acid, under low severity factor and its enzymatic degradability was investigated in this study to identify fermentable sugar production and corresponding microbial inhibitors formation.Additionally, effect of equivalent severity factors of 2 to 3.5 and particle sizes of 1 to 35 mm were also studied systematically.Particle size of 15 mm was found as an optimum size at an equivalent severity factor of 2.5.Sugars 61.99 ± 0.03 g and inhibitors 5.12 ± 0.01 g from 100 g of corn stalk were obtained at the optimum particle size and pretreatment condition.The highest glucan conversion and recovery at the optimum conditions were 92.95± 0.08 and 78.42± 0.07%, respectively.Overall, the two-stage pretreatment process with the larger particle size and low equivalent severity factor could be an alternative to reduce microbial inhibitors formation and excessive biomass processing cost.
Inhibitor Tolerance of a Recombinant Flocculating Industrial Saccharomyces Cerevisiae Strain During Glucose and Xylose Co-Fermentation.

Abstract:Lignocellulose-derived inhibitors have negative effects on the ethanol fermentation capacity of Saccharomyces cerevisiae. In this study, the effects of eight typical inhibitors, including weak acids, furans, and phenols, on glucose and xylose co-fermentation of the recombinant xylose-fermenting flocculating industrial S. cerevisiae strain NAPX37 were evaluated by batch fermentation. Inhibition on glucose fermentation, not that on xylose fermentation, correlated with delayed cell growth. The weak acids and the phenols showed additive effects. The effect of inhibitors on glucose fermentation was as follows (from strongest to weakest): vanillin>phenol>syringaldehyde>5-HMF>furfural>levulinic acid>acetic acid>formic acid. The effect of inhibitors on xylose fermentation was as follows (from strongest to weakest): phenol>vanillin>syringaldehyde>furfural>5-HMF>formic acid>levulinic acid>acetic acid. The NAPX37 strain showed substantial tolerance to typical inhibitors and showed good fermentation characteristics, when a medium with inhibitor cocktail or rape straw hydrolysate was used. This research provides important clues for inhibitors tolerance of recombinant industrial xylose-fermenting S. cerevisiae.

Abstract:Abstract Background Presence of inhibitory chemicals in lignocellulose hydrolysates is a major hurdle for production of second-generation bioethanol. Especially cheaper pre-treatment methods that ensure an economical viable production process generate high levels of these inhibitory chemicals. The effect of several of these inhibitors has been extensively studied with non-xylose-fermenting laboratory strains, in synthetic media, and usually as single inhibitors, or with inhibitor concentrations much higher than those found in lignocellulose hydrolysates. However, the relevance of individual inhibitors in inhibitor-rich lignocellulose hydrolysates has remained unclear. Results The relative importance for inhibition of ethanol fermentation by two industrial second-generation yeast strains in five lignocellulose hydrolysates, from bagasse, corn cobs and spruce, has now been investigated by spiking higher concentrations of each compound in a concentration range relevant for industrial hydrolysates. The strongest inhibition was observed with industrially relevant concentrations of furfural causing partial inhibition of both D-glucose and D-xylose consumption. Addition of 3 or 6 g/L furfural strongly reduced the ethanol titer obtained with strain MD4 in all hydrolysates evaluated, in a range of 34 to 51% and of 77 to 86%, respectively. This was followed by 5-hydroxymethylfurfural, acetic acid and formic acid, for which in general, industrially relevant concentrations caused partial inhibition of D-xylose fermentation. On the other hand, spiking with levulinic acid, 4-hydroxybenzaldehyde, 4-hydroxybenzoic acid or vanillin caused little inhibition compared to unspiked hydrolysate. The further evolved MD4 strain generally showed superior performance compared to the previously developed strain GSE16-T18. Conclusion The results highlight the importance of individual inhibitor evaluation in a medium containing a genuine mix of inhibitors as well as the ethanol that is produced by the fermentation. They also highlight the potential of increasing yeast inhibitor tolerance for improving industrial process economics.

Abstract:The toxicity and inhibition efficiency of lignocellulose-derived compounds on two kinds of bacteria were evaluated based on Quantitative Structure-Activity Relationship method, which could guide us to eliminate fermentation inhibitors existed in hydrolysates specifically. The results revealed that phenols exhibited the most significant toxicity on microbes, in which the functional group attached to benzene ring could dramatically influence toxicity degree. Furans showed moderate toxicity on microbes, however, short chain fatty acids exhibited feeble toxicity. GC-MS analysis identified inhibitors in rice straw hydrolysates after alkali-pretreatment as main constituent of phenols and acids, the concentration of which were 1.965 mM and 2.965 mM, respectively. Moreover, inhibition efficiency of these inhibitors was evaluated based on their toxicity and concentration. The results showed that inhibition efficiency of long chain fatty acids and phenol acids were significantly more enormous than that of other compounds, in which octadecanoic acid and ferulic acid had maximum inhibition efficiency value. Exploring the Inhibitory Characteristics of Acid Hydrolysates Upon Butanol Fermentation: A Toxicological Assessment

Abstract:This study aimed to quantitatively evaluate the inhibitor tolerance of butanol-producing bacterium Clostridium acetobutylicum. The inhibitory effect of the inhibitors generated by acid pretreatment of biomass feedstock on butanol fermentation decreased in the order of formic acid > oxalic acid > furfural > 5-HMF > Na2SO4. C. acetobutylicum has a small tolerance range for furfural (1.06-2.6 g/L) and 5-HMF (1.99-2.3 g/L). However, the inhibitory effect of Na2SO4 appears to have a wide range, with a chronic toxicity for C. acetobutylicum. All the results could explain, in quantitative manner, the instability of butanol fermentation with C. acetobutylicum from various acid-pretreated feedstocks caused by the fermentation inhibitors. Abstract:One of the important problems during cellulosic bioethanol production is that various byproducts generated during the pretreatment process seriously inhibit the growth and fermentation of Saccharomyces cerevisiae.Among these byproducts,weak acids,mainly including formic acid,acetic acid,and levulinic acid,are dominant.Formic acid is considered to have the strongest inhibitory effect.In this study,we investigated the inhibitory mechanism of formic acid action on xylose utilization during mixed sugar fermentation by an industrial S.cerevisiae xylose-fermenting strain,in order to provide the basics for breeding of inhibitor-tolerant strains.By the fermentation with the addition of acetic acid as the control,the fermentation performance of mixed sugars and xylose alone and the differences in the expression of sugar metabolism related-genes as well as the effect of glucose concentration on xylose fermentation and the expression of xylose metabolism-related genes were studied in the presence of formic acid.In addition,the effects of acetic acid and ethanol (both are metabolites of glucose) on xylose fermentation in the presence of formic acid,the changes of formic acid concentration,and the transcription of formic acid dehydrogenase gene FDH1 during the fermentation of mixed sugars and xylose alone were next investigated.The results showed that glucose inhibited the xylose fermentation specifically only in the presence of formic acid,and the decrease of xylose consumption rate was related to the decrease of the activities of xylose reductase (XR) and xylitol dehydrogenase (XDH).During the fermentation of xylose alone,the fermentation was inhibited only when the acetic acid,ethanol,and formic acid coexisted,and the inhibitory effect increased with the increasing ethanol concentration,to which the decrease of XDH activity was related.However,the synergistic inhibition by acetic acid,ethanol,and formic acid was still much weaker than the inhibitory effect of 60 g/L glucose.The much lower transcription level of the FDH1 gene resulted in a much lower formic acid decomposition rate during the fermentation of mixed sugars;this contributed to the specific inhibition of xylose fermentation by glucose in the presence of formic acid;however,the contribution was limited.To sum up,glucose inhibited the decomposition of formic acid,and the synergistic inhibition by glucose metabolites,acetic acid and ethanol,together with formic acid contributed to the specific serious inhibition of xylose fermentation by glucose in the presence of formic acid.However,there are other unknown inhibitory factors,which need further investigation.

Abstract:To evaluate the inhibition of by-products that exist in pretreated lignocellulose hydrolysate on bioethanol fermentation, quantitative structure-activity relationship (QSAR) models were established in the present study. These models have the potential to predict the fermentation inhibition to minimize the experimental effort. They also provided an innovative methodology for removing fermentation inhibitors purposefully and for optimizing pretreatment parameters and thus could enhance the bioethanol yield. The results indicated that the fermentation inhibition of phenolic aldehyde were stronger than that of phenolic acid followed by phenolic alcohol in which the fermentation inhibition was weakened by the existence of the methoxy group in the benzene ring. Meanwhile, the formation of the intramolecular hydrogen bonds of fermentation inhibitors also played an important role in weakening their fermentation inhibition. The individual fermentation inhibition of fermentation inhibitors was found strongly related to their molecular descriptors. Furthermore, ferulic acid was chosen as the representative inhibitor for the primary investigation on the combined fermentation inhibition of the various complex binary fermentation inhibitor mixtures, which co-existed in the lignocellulose hydrolysate. As indicated by the results, antagonism occurred mainly under the higher ferulic acid concentrations in the binary mixtures, while a simple additive effect was generated.
Improving cellulosic ethanol fermentation efficiency by converting endogenous water-soluble carbohydrates into citric acid before pretreatment

Abstract:Water-soluble carbohydrates in raw lignocellulosic biomass are converted into hydroxymethylfurfural (HMF) in the acid-based pretreatment, thus increasing the detoxification intensity and reducing the fermentation efficiency of cellulosic ethanol. Therefore, reducing water-soluble carbohydrates in raw corn stover is crucially important to reduce the inhibitors’ generation and improve the ethanol fermentation efficiency. In this study, aerobic solid-state fermentation of corn stover by inoculating Aspergillus niger spores converted 83% of the endogenous water-soluble carbohydrates into citric acid, leading to the decrease of 41% of HMF generation and 8% of sulfuric acid usage during the dry acid pretreatment. The reduced inhibitor generation improved the ethanol fermentability by 11% more ethanol than that of the corn stover without water-soluble carbohydrates’ removal. This suggests that the removal of the water-soluble carbohydrates before pretreatment significantly reduced the inhibitors’ generation in pretreatment and improved the fermentation efficiency of cellulosic ethanol.

Abstract:The inhibitory effect of toxic byproducts mainly derived from the pretreatment of lignocellulosic biomass is one of the limiting factors hindering industrial application. In this work, a simple yet practicable aeration strategy was proposed to enhance microbial tolerance to inhibitors, including formic acid, acetic acid, furfural, and 5-hydroxymethyl furfural (5-HMF). Batch fermentation by Kluyveromyces marxianus CICC 1727-5 using xylose and glucose alone and in combination under different aeration conditions coupled with multiple inhibitors was performed. The fermentation time and residual xylose significantly decreased with high xylitol production (0.7 g/g) and high ethanol production (0.42 g/g) at an aeration rate of 0.5 vvm. The postglucose effect was effectively relieved during co-fermentation by the microaeration strategy. Transcriptomics analysis was performed to determine the mechanism of inhibitor resistance. Transcription of genes related to DNA replication, protein processing, carbon metabolism, biosynthesis of the ribosome, amino acids, peroxidase, sterols, and unsaturated fatty acids were significantly upregulated accompanied by downregulated expression of genes related to autophagy under aeration conditions in the presence of multiple inhibitors. These results provide a valuable reference for future studies on biofuel fermentation from lignocellulosic biomass by a facultative fermenting microbe.
Inhibitory Effects of Biomass Degradation Products on Ethanol Fermentation and a Strategy to Overcome Them

Abstract:The influence of buffers, as well as inhibitors such as formic acid, furfural, HMF, guaiacol, and vanillin, on ethanol formation was investigated. Compared to phosphoric buffer, the acetic and citric buffers were less inhibitory on ethanol fermentation. The addition of formic acid (2.5 g/L) to the buffer reduced the ethanol yield by 8%. Guaiacol (3 g/L) and vanillin (2.5 g/L) decreased ethanol production by 50% and 20%, respectively. Furfural and HMF delayed the yeast fermentation without reducing the total yield. The fermentation was seriously inhibited by the mixture of furfural (1 g/L), HMF (1 g/L), formic acid (1 g/L), vanillin (1 g/L), and guaiacol (1 g/L). The ethanol yield of the fermentation based on enzymatic hydrolyzate from treated biomass was 82%. The addition of 1 g/L MgSO4 as a shielding protector rehabilitated nearly 100% of the total yield.
