[Determination of berberine in biological specimen by high performance TLC and fluoro-densitometric method]

Abstract:In this paper, a simple and specific high performance TLC and fluoro-densitometric method for separating and determining berberine in biological specimen was developed. Two hundred microliters of plasma or tissue homogenate mixed with 20 microliters of sodium lauryl sulfate (18%) and quinidine (300 ng, as internal standard) were extracted with 1 ml of chloroform. The developing solvent consisted of ethyl acetate-methyl acetate-methanol-water (5.4:4.6:1.2:1.0). The determination was carried out with a Shimadzu TLC-Scanner CS-910. Berberine spots were measured using an excitation wavelength of 350 nm and an emission wavelength of 550 nm, while using 350 nm and 450 nm for quinidine spots as the excitation and emission wavelength respectively. The average recoveries were 100.3% from plasma and 103.8% from tissue homogenate. The berberine levels in plasma and in tissues were compared in normal mice after oral administration of berberine (100 mg/kg) or the powder of Coptis chinensis (2 g/kg) containing the same amount of berberine. Simultaneously, the hypoglycemic effect of berberine and that of the powder of Coptis chinensis were measured in these mice. Results indicate that the concentrations of berberine in plasma and tissues given the powder of Coptis chinensis orally were higher than those given berberine and that the hypoglycemic effect of the powder was also stronger than that of berberine. Evidently, the changes of the blood glucose level and the level of berberine in plasma showed an opposite relationship.

Abstract:Objective To seek the feasibility of obtaining highly pure berberine hydrochloride. Methods Berberine hydrochloride in Rhizoma coptidis was turned into berberine sulphuric of large dissolubility, and then was turned into crude berberine hydrochloride. Relative highly pure berberine hydrochloride was separated by TLC further. And berberine hydrochloride was separated by column chromatography. Finally, the highly pure berberine hydrochloride was obtained by several crystalli- zations in ethanol, and its content was measured by HPLC.Results The purity of berberine hydrochlo-ride obtained in this method reach over 97%.Conclusion this method is characterized by convenience, practicality,good reproducibility,which can be applied to mass production and supplied for reasonable exploitation of resource.

Abstract:Sample preparation technique based on an organic filter membrane (pH-resolved filter membrane microextraction) (pH-RFMME) was developed, coupled with high-performance liquid chromatography, and used to determine protoberberine alkaloids (jatrorrhizine, epiberberine, coptisine, palmatine, and berberine) in Coptis chinensis at different pH values through a one-step procedure. This green procedure provides a desirable sample pretreatment technology. The main variables affecting the extraction such as filter membrane area (or volumes of extraction solvents), sample pH, eluent pH, ionic strength, extraction stirring rate, extraction time, and sample volume were optimized. Under the optimized conditions, the enrichment factors of the analytes were 40.4–52.0, the linear ranges were 3.2–6250 ng · mL−1 for jatrorrhizine and epiberberine, 6.0–12000 ng · mL−1 for coptisine, 1.8–3600 ng · mL−1 for palmatine, and 18.8–18800 ng · mL−1 for berberine, with r 2 ≥ 0.9945. The limits of detection were less than 0.3 ng · mL−1. Satisfactory recoveries (84.8%–115.5%) and precision (1.8%–10.0%) were also achieved. These results confirmed that pH-RFMME is a simple, rapid, practical, and environmentally friendly method to isolate analytes that exhibit significant differences in acidity or alkalinity from complex samples.

Abstract:Coptidis Rhizoma (Chinese: Huanglian) and Phellodendri Chinensis Cortex (Chinese: Huangbo) are widely used Traditional Chinese Medicine, and often used in combination because of their similar pharmacological effects in clinical practice. However, the quality control methods of the two drugs are different and complicated, which is time consuming and laborious in practical application. In this paper, rapid and simultaneous determination of moisture and berberine in Coptidis Rhizoma (CR) and Phellodendri Chinensis Cortex (PC) was realized by near-infrared spectroscopy (NIRs) combined with global models. Competitive adaptive reweighted sampling (CARS) and successive projection algorithm (SPA) method were applied for variable selection. Principal component analysis (PCA) and partial least squares regression method (PLSR) were applied for qualitative and quantitative analysis, respectively. The characteristic variables of berberine showed similarity and consistency in distribution, providing basis for the global models. For moisture content, the global model had relative standard error of prediction set (RSEP) value of 3.04% and 2.53% for CR and PC, respectively. For berberine content, the global model had RSEP value of 5.41% and 3.97% for CR and PC, respectively. These results indicated the global models based on CARS-PLS method achieved satisfactory prediction for moisture and berberine content, improving the determination efficiency. Furthermore, the greater range and larger number of samples enhanced the reliance of the global model. The NIRs combined with global models could be a powerful tool for quality control of CR and PC.

Abstract:The supramolecular interaction of beta-cyclodextrin dimer with berberine hydrochloride was studied in aqueous KH2PO4-H3PO4 buffer solution of pH 2.00 at room temperature by spectrofluorimetry. The apparent association constant of the complex was 1.53 x 10(4) L mol(-1). Based on the significant enhancement of fluorescence intensity of supramolecular sandwich complexes, a spectrofluorimetric method with high sensitivity and selectivity was developed for the determination of berberine hydrochloride in aqueous solution in presence of ethylenediamine linked beta-CD dimer. The linear range of the method was 12.8-1.00 x 10(4 )ng mL(-1) with the detection limit 3.6 ng mL(-1). There was no interference from the normally used in tablets and serum constituents. The proposed method was successfully applied to the determination of berberine hydrochloride in tablets and serum. And then it has a promising potential in therapeutic drug monitoring, pharmacokinetics and clinical application.
Research on Method of Microwave Extraction of Berberine in Coptis Chinensis

Abstract:Using microwave method to extract berberine in coptis chinensis,extraction rate as the evaluation index,examines sulfuric acid concentration、the radiation time、radiation power、solid-liquid ratio to the influence of different conditions on berberine extraction rate,and has made the comparison with other methods.The results show the optimum conditions for extraction: radiation time 4min,sulfuric acid concentration 0.06mol / L,the radiation power of fire,and solid-liquid ratio 1: 80.The optimum extraction rate was 45.9%.Compared to the existing traditional method,microwave method is time saving,easy to operate,and the extraction rate of berberine can increase about 35%.

Abstract:This study aims to qualitatively analyze protoberberine alkaloids in crude extract of Coptidis Rhizoma using HPLC with ESI-MS/MS. Possible specific molecular weights of protoberberine alkaloids were firstly deduced according to literatures and were adopted to screen the alkaloids in the HPLC with ESI-MS of crude extract of Coptidis Rhizoma. As a result, 21 protoberberine alkaloids were found, including compounds of very low concentration and compounds coeluted in one peak. Among these, two compounds were positively identified and verified by comparison with standards. Ten of these compounds were first reported in this study for Coptidis Rhizoma. In addition, chromatographic retention parameters a and c of all compounds were obtained using their retention times under five gradient conditions and were applied to confirm the deduction about the structures of protoberberine alkaloids by tandem mass data.
Naturally occurring proteinaceous nanoparticles in Coptidis Rhizoma extract act as concentration-dependent carriers that facilitate berberine absorption

Abstract:Pharmacological activities of some natural products diminish and even disappear after purification. In this study, we explored the mechanisms underlying the decrease of acute oral toxicity of Coptidis Rhizoma extract after purification. The water solubility, in vitro absorption, and plasma exposure of berberine (the major active compound) in the Coptidis Rhizoma extract were much better than those of pure berberine. Scanning electron microscopy, laser scanning confocal microscopy (LSCM), and dynamic light scattering experiments confirmed that nanoparticles attached to very fine precipitates existed in the aqueous extract solution. The LSCM experiment showed that the precipitates were absorbed with the particles by the mouse intestine. High-speed centrifugation of the extract could not remove the nanoparticles and did not influence plasma exposure or acute oral toxicity. However, after extract dilution, the attached precipitates vanished, although the nanoparticles were preserved, and there were no differences in the acute oral toxicity and plasma exposure between the extract and pure berberine. The nanoparticles were then purified and identified as proteinaceous. Furthermore, they could absorb co-dissolved berberine. Our results indicate that naturally occurring proteinaceous nanoparticles in Coptidis Rhizoma extract act as concentration-dependent carriers that facilitate berberine absorption. These findings should inspire related studies in other natural products.
Characterization Of The Transportation Of Berberine In Coptidis Rhizoma Extract Through Rat Primary Cultured Cortical Neurons

Abstract:The aim of this study was to investigate the transport behavior and efflux of berberine through the primary culture cortical neurons. High-performance liquid chromatography coupled with an UV-vis detector at 347 nm was applied. The mobile phase was 0.05 m potassium dihydrogen phosphate solution (containing 0.5% triethylamine, pH 3.0)-acetonitrile (73:27, v/v). Neurons were incubated with Coptidis rhizoma extract 6.5 mu g/mL (containing 1.91 mu g/mL berberine) and verapamil, KCN or cimetidine for 2 h, and then lysed in methanol to extract intracellular berberine. A 20 mu l aliquot of sample was injected into the HPLC system to determine berberine concentration. The results showed that metabolic inhibitor KCN and P-glycoprotein inhibitor verapamil could increase berberine concentration within the neurons, indicating that efflux of berberine was energy-dependent and P-glycoprotein was likely to be involved. Moreover, the organic cation transporter inhibitor cimetidine could decrease berberine concentration within the neurons, suggesting that the organic cation transporter might be involved in the berberine transport process. Abstract:Objective: To investigate the pharmacokinetics of berberine in Coptidis Rhizoma extract in rat intestinal and bile.Method: A simple and accurate high-performance liquid chromatography method was used to determine the concentration of berberine in the rat intestinal and bile samples.Result: Following the intravenous administration of Coptidis Rhizoma extract,B/P(concentration in the bile/concentration in the plasma) of berberine was determined to be 69.1～497.9 from 0.25 h to 4 h.Bile excretion was one of the main excretion ways for rats to eliminate berberine of Coptidis Rhizoma alkaloids.Most of berberine in the intestinal was from the bile.While bile was discharged from rats,berberine still could be determined in the intestinal.Conclusion: The results proved that berberine could be transported from blood to intestinal in the animal model with a direction from basolateral membrane to apical membrane.P-glycoprotein is involved in the transport mechanism.
Determination of baicalin and berberine in Ganmao Yixiaoshi Capsule by HPLC

Abstract:AIM:To establish methods for determining baicalin and berberine in Ganmao Yixiaoshi Capsule(Rhizoma et Radix Notopterygii,Radix Angelicae Pubescentis,Radix Puerariae thomsonii,Radix Scutellariae,Rhizoma Coptidis,etc.)by HPLC.METHODS:Hypersil C_ 18(4.6 mm×150 mm,5 μm)column was used,the mobile phase was composed of acetonitrile-0.05 mol/L phosphoric acid(25 ∶75)to determine baicalin,acetonitrile-0.05 mol/L potassium dihydrogen phosphate buffer solution(with 0.3% triethylamine,pH 3.0 adjusted with phosphoric acid)(24 ∶76)to determine berberine.The detection wavelengths were at 276 nm and 346 nm,respectively.RESULTS:The calibration curve was linear in the concentration ranges of 0.176 μg-0.880 μg for baicalin and 0.148 μg-0.845 μg for berberine,the correlation coefficient were 0.999 8 and 0.999 5,respectively.The average recovery of baicalin was 99.30%(RSD= 1.56%)and that of berberine was 98.84%(RSD= 2.41%).CONCLUSION:The method is sensitive,simple and accurate,and can be used for the quality control of Ganmao Yixiaoshi Capsule.
Investigation on Intermolecular Interaction Between Berberine and Beta-Cyclodextrin by 2d Uv-Vis Asynchronous Spectra

Abstract:The interaction between berberine chloride and beta-cyclodextrin (beta-CyD) is investigated via 2D asynchronous UV-Vis spectrum. The occurrence of cross peaks around (420 nm, 420 nm) in 2D asynchronous spectrum reveals that specific intermolecular interaction indeed exists between berberine chloride and beta-CyD. In spite of the difficulty caused by overlapping of cross peaks, we manage to confirm that the 420 nm band of berberine undergoes a red shift, and its bandwidth decreases under the interaction with beta-CyD. The red-shift of the 420 nm band that can be assigned to n-pi* transition indicates the environment of berberine becomes more hydrophobic. The above spectral behavior is helpful in understanding why the solubility of berberine is enhanced by beta-CyD. (C) 2017 Published by Elsevier B.V.

Abstract:Supercritical fluid was used to extract berberine from rhizome of Coptis chinensis Franch. The recovery of berberine was compared with various modifiers, i.e. methanol and 95% ethanol with and without surfactant Tween 80, and 1,2-propanediol. The results show that the yield obtained after 3 h extraction with 1,2-propanediol-modified supercritical carbon dioxide was the highest (from 6.91%, w/w at 200 bar to 7.53%, w/w at 500 bar), while that obtained with 95% ethanol modified-supercritical carbon dioxide was the lowest (from 0.15%, w/w at 300 bar to 0.19%, w/w at 600 bar). The recovery of berberine was not improved by adding 5% Tween 80 in supercritical fluid.

Spectrofluorometric determination of berberine using a novel Au nanocluster with large Stokes shift

Abstract:Berberine hydrochloride (BHC), a natural isoquinoline alkaloid, is widely applied as a an agent in traditional Chinese medicine. Almost all the traditional methods for BHC detection require complicated preprocessing steps or expensive instruments. In this article, we report a simple, rapid, sensitive, and selective method for BHC detection using fluorescent gold nanoclusters (F-AuNCs) as the fluorescent probe with a large Stokes shift of 237 nm. The F-AuNCs prepared with citrate-stabilized stannous chloride and hydrogen tetrachloroaurate(III) as raw materials in an aqueous medium display strong and stable fluorescence at 566 nm. When F-AuNCs are mixed with BHC, the fluorescence of F-AuNCs is effectively quenched. Under optimized conditions, this method allows sensitive and selective measurements of BHC in a concentration ranging from 1.0 × 10-6 to 1.0 × 10-4 mol L-1 with a detection limit of 7.5 × 10-8 mol L-1, which is relatively low among reported spectral methods. This method provides excellent selectivity for the detection of BHC against inorganic anions and natural amino acids. In addition, the BHC content in two different types of berberine tablets was successfully determined by this method and the results showed high accuracy. Graphical Abstract ᅟ.
Determination of Berberine and Phellodendrine in Cortex Phellodendri by HPLC

Abstract:Objective To establish a method for determination of Berberine and Phellodendrine in Cortex Phellodendri(CP).Methods Reverse-phase HPLC method was used for determination of Berberine and Phellodendrine in different batches of CP.The chromatographic conditions were:Merck-lichrospher RP-C18 column(4.6×250 mm,5 μm),flow rate being 0.8 mL/min,column temperature at 25 ℃,detection wavelength at 284nm for Phellodendrine and 245nm for Berberine.Results The content of Berberine was 4 times and that of Phellodendrine in Chuan CP 2～3 times as much as that of Guan CP.Conclusion The method is proved to be feasible for quality assessment of Cortex Phellodendri.Limited contents of Berberine and Phellodendrine should be laid out for the great difference in Chuan CP and Guan CP.
Determination Of The Coptis Extract Alkaloids Using Near-Infrared Diffuse Reflectance Spectroscopy

Abstract:A quick and nondestructive method using near-infrared (NIR) diffuse reflectance spectroscopy was presented to determine the contents of berberine, palmatine, jatrorrhizine and total alkaloid of Coptis extract. Partial least square(PLS) was utilized to build the calibration models using second derivative spectra with high performance liquid chromatography as a reference method for each components. Root mean square error of prediction (RMSEP) for berberine, palmatine, jatrorrhizine and total alkaloid were 0.184, 0.109, 0.054 and 0.325, respectively. The recoveries were determined to be 97.67% similar to 99.59%, 96.63% similar to 100.70%, 95. 15% similar to 101.15% and 97.41% similar to 99.89% for berberine, palmatine, jatrorrhizine and total alkaloid, respectively. The relative standard deviations were 0.3% for berberine, 0.6% for palmatine, 1.8% for jatrorrhizine, and 0.3% for total alkaloid, respectively. The results show that the presented method has a good reproducibility and stability and can be applied to on-line detection in industrial process.

Abstract:AIM:To develop methods for the fingerprint analysis of Rhizoma Coptidis and the determination of berberine, palmatine and jatrorrhizine in Rhizoma Coptidis, and analyze the contents of these three alkaloids in Rhizoma Coptidis under different cultivation conditions, from different areas and processed with different methods.METHODS:Two methods (HPLC-UV and HPLC-MS) have been developed and used in fingerprint analysis of Rhizoma Coptidis. An HPLC method was used to determine the contents of three alkaloids.RESULTS:With HPLC-MS techniques, seven major chromatographic peaks in the fingerprint analysis of Rhizoma Coptidis were identified by their MS spectra and compared with the reference standards. In different cultivation conditions, shading conditions and growing ages have obvious influence on the contents of three alkaloids in Rhizoma Coptidis, while planting density was not the major factor that influenced the contents of three alkaloids. The contents of three alkaloids of Coptidis samples were almost higher than those of Coptidis reference material. For Coptidis samples from different cultivation area, the contents of these three alkaloids were different greatly. For Coptidis samples processed with different methods, the contents of three alkaloids were not influenced obviously by processing methods.CONCLUSION:The results showed that the ecology cultivation method to replace the traditional shading method was feasible and provided the theoretical foundation for scientifically processing Rhizoma Coptidis.

Abstract:\Quantitation of protoberberine alkaloids is an essential guarantee for efficacy control and medication safety of Coptidis Rhizoma (CR) related medicines. Traditional univariate chromatography faced challenges with co-elution, unknown interferences, and retention time shift when analyzing isomeric analytes in varying sample matrices. We presented a chemometrics-enhanced high-performance liquid chromatography-diode array detection (HPLC-DAD) strategy for simultaneous quantification of six protoberberine alkaloids and processed multi-channels chromatographic-spectral data with four second-order calibration algorithms. Chromatographic conditions were firstly optimized. Four groups of predicted samples were modeled individually with the designed calibration set. Mathematical resolutions were then obtained, and pseudo-univariate regression gave the quantitative concentration of each analyte. Four models were scored on fit, linearity, recovery, and robustness, where alternating trilinear decomposition assisted multivariate curve resolution (ATLD-MCR) exhibited an optimal and stable performance. Besides, the resolved spectra presented high consistency with the actual spectra (r≥0.9993). Limits of quantification (LOQ) fully met the pharmacopoeia stipulation and were 0.17, 0.60, 0.19, 0.74, 0.15, and 0.38 μg mL −1 for columbamine, epiberberine, jatrorrhizine, coptisine, palmatine, and berberine, respectively. The importance of this strategy is to exploit collinearity resolution and additional selectivity that permit accurate quantitation at poor chromatographic resolutions, avoiding individual pretreatment and HPLC optimizations for different samples. This study provides a universal alternative for routine quality assessment of protoberberine alkaloids in CR-related medicines.

[Study on traditional Chinese medicine extracts of rhizoma coptidis by FTIR spectroscopy]

Abstract:In the present paper, we applied Fourier transform infrared spectroscopy (FTIR) and second derivative infrared spectroscopy to establish a rapid and efficient analytical method for a preliminary quality control of medicinal herb extracts. The commonly used Chinese herb medicine, Rhizoma coptidis, was taken as an example. The main component-berberine in rhizoma coptidis was analyzed as target compound. The results indicated that the IR spectra of rhizoma coptidis herb, its water and alcohol extracts, exhibit their macroscopic fingerprint characters with fine repeatability, and the corresponding secondary derivative spectra further confirmed the spectral characteristics of the original IR spectra by enhancing spectral resolution. The typical and strongest absorption band of IR spectrum of berberine is at 1,505 cm(-1), which is assigned to aromatic skeleton vibration band, and its intensities in spectra of rhizoma coptidis herb and its different extracts become the main identification standard of the contents of berberine in those samples. In addition, comparing the IR spectra of the medicinal herb and its extracts, it was shown that the main bioactive components, alkaloids in rhizoma coptidis herb, are enriched by extraction, and the contents of berberine in alcohol extracts of rhizoma coptidis are higher than those in water extracts. The spectroscopic technique described is a simple and rapid analytical technique, which could provide valuable information about chemical constituents of medicinal extracts for guiding further chromatographic analysis and separation improvement.
