Abstract:Engineered nanoparticles released into surface water may accumulate in sediments, potentially threatening benthic organisms. This study determined the toxicokinetics in Chironomus riparius of Ag from pristine silver nanoparticles (Ag NPs), a simulating aged Ag NP form (Ag2S NPs), and AgNO3 as an ionic control. Chironomid larvae were exposed to these Ag forms through water, sediment, or food. The potential transfer of Ag from larvae to adult midges was also evaluated. Results revealed higher Ag uptake by C. riparius upon exposure to Ag2S NPs, while larvae exposed to pristine Ag NPs and AgNO3 generally presented similar uptake kinetics. Uptake patterns of the different Ag forms were generally similar in the tests with water or sediment exposures, suggesting that uptake from water was the most important route of Ag uptake in both experiments. For the sediment bioaccumulation test, uptake was likely a combination of water uptake and sediment particles ingestion. Ag uptake via food exposure was only significant for Ag2S NPs. Ag transfer to the terrestrial compartment was low. In our environmentally relevant exposure scenario, chironomid larvae accumulated relatively high Ag concentrations and elimination was extremely low in some cases. These results suggest that bioaccumulation of Ag in its nanoparticulate and/or ionic form may occur in the environment, raising concerns regarding chronic exposure and trophic transfer. This is the first study determining the toxicokinetics of NPs in Chironomus, providing important information for understanding chironomid exposure to NPs and their potential interactions in the environment.

Abstract:Generalized studies of the world scientific literature on the fate and risk assessment of exposure to silver nanoparticles (NPAg) both at the ecosystem level and at the organism level, as well as in the laboratory. It is emphasized that the toxic effect of silver nanoparticles, mechanisms and methods of action of NPAg on the body of aquatic organisms have been sufficiently studied in laboratory practice. However, there are some gaps and discrepancies between the results of laboratory tests and the study of real environmental consequences, and such inconsistencies hinder the development of appropriate effective measures to achieve environmental well-being. To bridge such gaps, this review summarizes how environmental conditions and the physicochemical properties of NPAg influence conflicting conclusions between laboratory and real-world environmental studies. It is emphasized that modern research on the pathways of entry, transformation and bioaccumulation of silver nanoparticles in natural aquatic ecosystems emphasizes the ability of such nanoparticles to penetrate intact physiological barriers, which is extremely dangerous. It is proved that silver nanoparticles have a toxic effect on microorganisms, macrophytes and aquatic organisms. The toxic effects of NPAg cover almost entire aquatic ecosystems. A study by a number of authors on the factors influencing the mobility, bioavailability, toxicity and environmental fate of Ag nanoparticles was analyzed to assess the environmental risk. In addition, this review systematically examines the various toxic effects of silver nanoparticles in the environment and compares these effects with the results obtained in laboratory practice, which is useful for assessing the environmental effects of such compounds. The dangerous chronic effects of low-concentration NPAg (μg/l) on natural aquatic ecosystems over a long period of time (months to several years) have been described in detail. In addition, the prospects for future studies of NPAg toxicity in natural freshwater environments are emphasized. Abstract:Silver nanoparticles (AgNPs) present unique properties, such as the induced localized surface plasmon resonance (LSPR) provoked under illumination with a proper wavelength, allowing these nanomaterials to be applied in fields such as catalysis and biomedicine. The study of AgNPs is also highly relevant from the environmental pollution viewpoint due to their high production and application in commercial products. Consequently, AgNPs reach aquatic environments and can be plasmonically stimulated under natural light conditions. This study investigates the toxic effects promoted by AgNPs under plasmonic excitation on the survival and physiology of the crustacean Daphnia similis. Two AgNP shapes (spherical and triangular) with plasmon bands absorbing in different spectral regions in the visible range were studied. The organisms were exposed to different AgNP concentrations under five different light conditions. Survival and changes in enzymatic biomarkers of oxidative stress and lipid storage were evaluated. Under LSPR conditions, we observed increased lethality for both AgNP shapes. LSPR effects of AgNPs showed mortality 2.6 and 1.7 times higher than the treatment under dark conditions for spherical and triangular morphologies respectively. The enzymatic assays demonstrated that plasmonic treatments triggered physiological responses. Significantly decreased activities were observed exclusively under LSPR conditions for both AgNP shapes. Considering all treatments, spherical AgNPs showed lower LC50 values than triangular ones, indicating their higher toxic potential. Our results demonstrate that LSPR AgNPs can induce biological responses associated with oxidative stress and survival. Therefore, this study highlights the potential risks of environmental contamination by plasmonically active metallic nanomaterials. These materials can enhance their toxicity when light-excited, yet the results also indicate promising opportunities for light-based therapies.

Abstract:Nanosilver, due to its small particle size and enormous specific surface area, facilitates more rapid dissolution of ions than the equivalent bulk material; potentially leading to increased toxicity of nanosilver. This, coupled with their capacity to adsorb biomolecules and interact with biological receptors can mean that nanoparticles can reach sub-cellular locations leading to potentially higher localized concentrations of ions once those particles start to dissolve or degrade in situ. Further complicating the story is the capacity for nanoparticles to generate reactive oxygen species, and to interact with, and potentially disturb the functioning of biomolecules such as proteins, enzymes and DNA. The fact that the nanoparticle size, shape, surface coating and a host of other factors contribute to these interactions, and that the particles themselves are evolving or ageing leads to further complications in terms of elucidating mechanisms of interaction and modes of action for silver nanoparticles, in contrast to dissolved silver species. This review aims to provide a critical assessment of the current understanding of silver nanoparticle toxicity, as well as to provide a set of pointers and guidelines for experimental design of future studies to assess the environmental and biological impacts of silver nanoparticles. In particular; in future we require a detailed description of the nanoparticles; their synthesis route and stabilisation mechanisms; their coating; and evolution and ageing under the exposure conditions of the assay. This would allow for comparison of data from different particles; different environmental or biological systems; and structure-activity or structure-property relationships to emerge as the basis for predictive toxicology. On the basis of currently available data; such comparisons or predictions are difficult; as the characterisation and time-resolved data is not available; and a full understanding of silver nanoparticle dissolution and ageing under different conditions is observed. Clear concerns are emerging regarding the overuse of nanosilver and the potential for bacterial resistance to develop. A significant conclusion includes the need for a risk-benefit analysis for all applications and eventually restrictions of the uses where a clear benefit cannot be demonstrated.

Ageing Alters the Physicochemical Properties of Silver Nanoparticles and Consequently Compromises Their Acute Toxicity in Mammals.

Abstract:Despite numerous investigations into AgNP-induced toxicity, little has been taken into consideration the potential health impacts of aged AgNPs in comparison to fresh AgNPs. In the current study, we scrutinized the potential effects of aged AgNPs in animals. We first found that AgNPs underwent morphological transformations after natural ageing in aqueous solution upon exposure to air and sunlight for 9 days, as characterized by significant aggregation with increase of particle size approximately by 2 fold. Meanwhile, dissolved Ag ions from aged AgNPs increased by 33% compared to fresh AgNPs. Strikingly, the acute exposure results showed that aged AgNPs induced lower toxicity in mice relative to fresh AgNPs. Aged AgNPs caused milder local inflammation in the peritoneal cavity of mice, as evidenced by 63% reduction of tumor necrosis factor α (TNF-α) than that induced by fresh AgNPs. The deposition mass of aged AgNPs in the liver, spleen, lung and kidney was diminished by 69%, 39%, 83% and 40%, respectively, compared to the distribution profiles in response to fresh AgNPs. Whereby, milder splenic hyperemia was observed, and no significant hepatoxicity was found. Additionally, aged AgNPs provoked milder increase of periphery leukocytes and malondialdehyde (MDA) in mice in comparison to fresh AgNPs. Taken together, this study unraveled that the ageing process elicited remarkable alterations to physicochemical properties and toxic effects as well. This study would provide new insights into the potential health impacts of AgNPs under transformation-determined exposure scenarios.
In Vivo toxicological assessment of biologically synthesized silver nanoparticles in adult Zebrafish (Danio rerio)

Abstract:The present study examines the deleterious effect of biologically synthesized silver nanoparticles in adult zebrafish. Silver nanoparticles (AgNPs) used in the study were synthesized by treating AgNO3 with aqueous leaves extract of Malva crispa Linn., a medicinal herb as source of reductants. LC50 concentration of AgNPs at 96 h was observed as 142.2 μg/l. In order to explore the underlying toxicity mechanisms of AgNPs, half of the LC50 concentration (71.1 μg/l) was exposed to adult zebrafish for 14 days. Cytological changes and intrahepatic localization of AgNPs were observed in gills and liver tissues respectively, and the results concluded a possible sign for oxidative stress. In addition to oxidative stress the genotoxic effect was observed in peripheral blood cells like presence of micronuclei, nuclear abnormalities and also loss in cell contact with irregular shape was observed in liver parenchyma cells. Hence to confirm the oxidative stress and genotoxic effects the mRNA expression of stress related (MTF-1, HSP70) and immune response related (TLR4, NFKB, IL1B, CEBP, TRF, TLR22) genes were analyzed in liver tissues and the results clearly concluded that the plant extract mediated synthesis of AgNPs leads to oxidative stress and immunotoxicity in adult zebrafish.

Abstract:BackgroundWhen suspended in cell culture medium, nano-objects composed of soluble metals such as silver can dissolve resulting in ion formation, altered particle properties (e.g. mass, morphology, etc.), and modulated cellular dose. Cultured cells are exposed not just to nanoparticles but to a complex, dynamic mixture of altered nanoparticles, unbound ions, and ion-ligand complexes. Here, three different cell types (RAW 264.7 macrophages and bone marrow derived macrophages from wild-type C57BL/6 J mice and Scavenger Receptor A deficient (SR-A(−/−)) mice) were exposed to 20 and 110 nm silver nanoparticles, and RAW 264.7 cells were exposed to freshly mixed silver ions, aged silver ions (ions incubated in cell culture medium), and ions formed from nanoparticle dissolution. The In Vitro Sedimentation, Diffusion, Dissolution, and Dosimetry Model (ISD3) was used to predict dose metrics for each exposure scenario.ResultsSilver nanoparticles, freshly mixed ions, and ions from nanoparticle dissolution were toxic, while aged ions were not toxic. Macrophages from SR-A(−/−) mice did not take up 20 nm silver nanoparticles as well as wild-types but demonstrated no differences in silver levels after exposure to 110 nm nanoparticles. Dose response modeling with ISD3 predicted dose metrics suggest that amount of ions in cells and area under the curve (AUC) of ion amount in cells are the most predictive of cell viability after nanoparticle and combined nanoparticle/dissolution-formed-ions exposures, respectively.ConclusionsResults of this study suggest that the unbound silver cation is the ultimate toxicant, and ions formed extracellularly drive toxicity after exposure to nanoparticles. Applying computational modeling (ISD3) to better understand dose metrics for soluble nanoparticles allows for better interpretation of in vitro hazard assessments.

An insight into silver nanoparticles bioavailability in rats.

Abstract:A comprehensive study of the bioavailability of orally administered silver nanoparticles (AgNPs) was carried out using a rat model. The silver uptake was monitored in liver and kidney tissues, as well as in urine and in feces. Significant accumulation of silver was found in both organs, the liver being the principal target of AgNPs. A significant (similar to 50%) fraction of silver was found in feces whereas the fraction excreted via urine was negligible (<0.01%). Intact silver nanoparticles were found in feces by asymmetric flow field-flow fractionation (AsFlFFF) coupled with UV-Vis analysis. Laser ablation-ICP MS imaging showed that AgNPs were able to penetrate into the liver, in contrast to kidneys where they were retained in the cortex. Silver speciation analysis in cytosols from kidneys showed the metallothionein complex as the major species whereas in the liver the majority of silver was bound to high-molecular (70-25 kDa) proteins. These findings demonstrate the presence of Ag(I), released by the oxidation of AgNPs in the biological environment.
Toxicity of Synthesized Silver Nanoparticles in a Widespread Fish: A Comparison Between Green and Chemical

Abstract:Metallic nanoparticles are gaining importance in various fields of life due to their large surface area to volume ratio. However, metallic nanoparticles pose different toxic effects on fish when they appear with different shapes and compositions in water. Herein the present study was designed to evaluate the median (LC50) and sub-lethal (1/10th of LC50) concentrations of Ag-Green NPs, 700 μg/L for Ag-Chem NPs, and 50 μg/L for Ag2O-Chem NPs were confirmed in Hypophthalmichthys molitrix. Furthermore, exposure of H. molitrix fingerlings to 10 % of LC50 concentration of these particles induced significantly higher (p < 0.05) activities of serum alanine transaminase, aspartate aminotransferase, lactate dehydrogenase, white blood cells, acetylcholinesterase and catalase, superoxide dismutase, peroxidase, relative gene expressions of antioxidant enzymes, heat shock protein (Hsp70), hypoxia- inducible factor 1-alpha (HIF-1α) and lipid peroxidase level than the control, but decreased hematological parameters with less effects of Ag-Green NPs than chemically synthesized AgNPs. Moreover, the histopathological study also indicated morphological changes in the liver and gills of treated fish groups. The comparative toxicity evaluation revealed the maximum negative effect of Ag2O-Chem NPs followed by Ag-Chem NPs while Ag-Green NPs showed the least toxic effects. Based on our results, replacement of chemically synthesized NPs to green synthesized AgNPs can be recommended in large scale application to reduce the noxious effects to aquatic environment.
Antagonistic in vivo interaction of polystyrene nanoplastics and silver compounds. A study using Drosophila

Abstract:Since heavy metals and micro-/nanoplastics (MNPLs) can share common environmental niches, their potential interactions could modulate their hazard impacts. The current study was planned to evaluate the potential interactions between silver compounds (silver nanoparticles or silver nitrate) and two different sizes of polystyrene nanoplastics (PSNPLs) (PS-50 and PS-500 nm), administered via ingestion to Drosophila larvae. While egg-to-adult survival was not affected by the exposure to silver compounds, PSNPLs, or their coexposures, the combined treatments succeeded to restore the delay of fly emergence induced by silver compounds. Transmission electron microscopy (TEM) and inductively coupled plasma mass spectrometry (ICP-MS) showed the ability of PSNPLs to transport silver compounds (regardless of their form) across the intestinal barrier, delivering them into the hemolymph of Drosophila larvae in a concentration exceeding that mediated by the exposure to silver compounds alone. The molecular response (gene expression) of Drosophila larvae greatly fluctuated, accordingly if exposures were administered alone or in combination. Although PSNPLs produced some oxidative stress in the hemocytes of Drosophila, especially at the highest dose (1 mM), higher levels were observed after silver exposure, regardless of its form. Interestingly, the oxidative stress of silver, especially that produced by nano‑silver, drastically decreased when coexposed with PSNPLs. Similar effects were observed regarding the DNA damage induced in Drosophila hemocytes, where cotreatment decreased the genotoxicity induced by silver compounds. This antagonistic interaction could be attributed to the ability of tiny plastic specks to confine silver, avoiding its bioavailability, and diminishing their potential impacts.

Abstract:The exploitation of silver nanoparticles (AgNPs) in biomedicine represents more than one third of their overall application. Despite their wide use and significant amount of scientific data on their effects on biological systems, detailed insight into their in vivo fate is still lacking. This study aimed to elucidate the biotransformation patterns of AgNPs following oral administration. Colloidal stability, biochemical transformation, dissolution, and degradation behaviour of different types of AgNPs were evaluated in systems modelled to represent biological environments relevant for oral administration, as well as in cell culture media and tissue compartments obtained from animal models. A multimethod approach was employed by implementing light scattering (dynamic and electrophoretic) techniques, spectroscopy (UV–vis, atomic absorption, nuclear magnetic resonance) and transmission electron microscopy. The obtained results demonstrated that AgNPs may transform very quickly during their journey through different biological conditions. They are able to degrade to an ionic form and again reconstruct to a nanoparticulate form, depending on the biological environment determined by specific body compartments. As suggested for other inorganic nanoparticles by other research groups, AgNPs fail to preserve their specific integrity in in vivo settings.

Abstract:Silver nanoparticles (AgNPs) have extensive applications in nanomedicine and parasitology, particularly as antifungal, bactericidal, antiviral, larvicidal, mosquitocidal, and tick-killing agents. Plant-based AgNPs (PB-AgNPs) have been studied as a safer and biocompatible strategy to collaborate in disease control. However, knowledge concerning the toxicity of PB-AgNPs in non-target organisms is still limited. A scientometric and systematic review was conducted to comprehensively understand the potential toxicity associated with these nanoparticles. In brief, the assessment of PB-AgNPs toxicological and ecotoxicity aspects needs to be aligned with their development for target organisms. Our review demonstrates that different PB-AgNPs can cause lethal and sublethal effects like increased oxidative stress, cardiotoxicity, neurotoxicity, hepatotoxicity, hematotoxicity, and, DNA damage, alongside others, particularly in aquatic organisms. Besides, the toxicity of PB-AgNPs for terrestrial and some aquatic organisms remains poorly understood. Additionally, the similar LC 50 range between non-target aquatic organisms and target organisms highlights the potential ecological impact of PB-AgNPs. Comprehensive toxicological assessments and further research are crucial to ensure the safe and sustainable use of PB-AgNPs in a One Health context.

Abstract:The past decades have witnessed a boom in nanotechnology that has led to increasing production and application of silver nanoparticles (AgNPs) in the textile industry due to their antimicrobial properties. Increase in the manufacture and use of NPs inevitably has resulted in their increased release into aquatic environments resulting in the exposure of organisms living in these environments. Recently, the risk of exposure to NPs and the potential interaction with biological systems has received increasing attention. The present study investigated the potential effects of predator cues on the toxicity of environmentally relevant concentrations of AgNPs in Daphnia carinata at organismal and biochemical levels. The results of this study show that exposure to environmentally relevant concentrations of AgNPs can result in adverse effects on daphnids with 24- and 48-h LC 50 values of 3.56 and 1.75 μg/L, respectively. Furthermore, significant inhibition of reproduction was observed at concentrations as low as 0.5 μg/L. Exposure to predator cues alone resulted in an increase in reproduction and inhibition of superoxide dismutase activity in daphnids. However, coexposure to predator cues interacted in an antagonistic manner with AgNPs with a 24-h LC 50 value of 10.81 μg/L compared with 3.56 μg/L for AgNPs alone. In summary, AgNPs could pose risks to aquatic invertebrates at environmentally relevant concentrations. Interestingly, the presence of other factors, such as predator cues, moderated the effects of exposure to AgNPs. Therefore, there is a need to further investigate the potential interactions between NPs and biological factors that can modulate toxicity of NPs for application to the risk assessment of aquatic invertebrates.

Abstract:Concerns have arisen about the health and environmental impacts of the increasing commercial use of silver nanoparticles (AgNPs). However, the toxic mechanisms and target tissues of AgNPs have not been fully defined. In this paper, we investigated the tissue toxicity of mice after intravenous administration of AgNPs at a single-dose of 0.2, 2 or 5 mg per kg (body weight), respectively. Biodistribution, endoplasmic reticulum stress, and oxidative stress were examined in mouse organs at eight hours after exposure. Stress markers, e.g. HSP70, BIP, p-IRE1, p-PERK, chop and xbp-1s proteins/genes, were significantly upregulated in a dose-dependent manner. In the liver, spleen, lung and kidney, high stress accompanied by apoptosis occurred. Low stress levels were observed in the heart and brain. Thus, it is proposed that the liver, spleen, lung and kidney are dominant target tissues of AgNP exposure. The lower stress and toxicity in the heart and brain were in agreement with lower AgNP accumulation. The present results demonstrated that AgNP exposure eventually resulted in permanent toxic damage by gradually imposing stress impacts on target organs. These findings highlight the potent applications of stress markers in future risk evaluation of silver nanoparticle toxicity.

Abstract:Silver nanoparticles (AgNPs) are widely used due to its unique physicochemical properties and efficient antimicrobial performance, therefore a large number of AgNPs have entered the aquatic environment and attracted extensive academic attention. This review introduces several commonly used methods for the preparation of AgNPs, including physical synthesis (ball milling, evaporative condensation, and laser ablation), chemical synthesis (chemical reduction, microemulsion, and microwave-assisted synthesis), and green synthesis methods for the preparation of AgNPs using microbial or plant extracts. This is followed by an overview of the applications of materials containing AgNPs and the pathways of AgNPs into the aquatic environment. Then, a comprehensive review of the research progress on the migration and transformation of AgNPs in natural water, sediment, wastewater, and wastewater treatment plants is presented. The transformation behavior of AgNPs in natural freshwater, sewage, and sediments includes aggregation and sedimentation, oxidative dissolution, sulfation, and chlorination, where the AgNPs in sewage and sediments end up in the form of sulfides. AgNPs in wastewater plants affect microbial activity and the removal of organic matter, nitrogen, and phosphorus during biological treatment. Finally, the toxic effects of AgNPs on microorganisms, animals, and plants after discharge into the aquatic environment are summarized. This work presents a full-process, multi-faceted analysis and review of the migration and transformation of AgNPs in aquatic environments, providing a theoretical reference for further research related to AgNPs.

Abstract:Since ancient times, silver has been known for its pronounced bactericidal, antiviral and fungicidal properties. Currently, nanoparticles of this metal are widely used in the food, light and pharmaceutical industries, as well as in medicine. Silver in any form can have a toxic effect not only on pathogens, but also on healthy cells. The biological activity and bioavailability of silver preparations depend on the degree of their solubility in water. In addition, the maximum permissible concentration of soluble forms of silver is an order of magnitude lower than that of insoluble forms. This makes nanoparticles of silver with a hydrophilic coating that form stable colloidal solutions in aqueous media potentially unsafe objects. In this work, we studied the kinetics of the accumulation of silver nanoparticles with an average size of 34 ± 5 nm stabilized with polyvinylpyrrolidone in the organs of laboratory C57Bl/6 mice. The administration of nanoparticles was carried out orally for 30, 60, 120 and 180 days at the dose of 50 μg/day/animal. All the mice developed and gained weight normally during the experiment. No adverse effects were observed. Determination of the silver content in biological tissues of mammals was accomplished by neutron activation analysis. The masses and concentrations of silver in the brain and its different sections (hippocampus, cerebellum, cortex and remnants), as well as in the lungs, testes, liver, blood, kidneys, spleen and heart, were determined. The injection times at which the accumulation curves reached saturation were established. An extremely high accumulation of silver in the testes was shown at 120 days of administration, and a significant accumulation of silver in the lungs and brain was observed. The accumulation of silver in all parts of the brain except the cortex was significant, and its trend was similar to that in the whole brain.

Abstract:The characterization of silver nanoparticles (AgNPs) showed spherical particles of 2.4‐7.2 nm, and aggregates of larger sizes.The crossover evaluation of antimicrobial activity and the toxicological profile of AgNPs showed that low concentrations caused high susceptibility in P. aeruginosa and S. aureus, citotoxicity in 3T3 and HaCat cells, and delayed embryonic development in zebrafish embryos and presented morphological abnormalities, neurotoxicity, and cardiotoxicity in zebrafish larvae. The achievement of complete profiles will lead to project functional and biocompatible nanoparticles. Silver nanoparticles (AgNPs) have a primordial role in the nanotechnology industry since they have great antimicrobial effects against diverse pathogens. AgNP production, use, and discard have been increasing; still, there is debate about the consequences of exposure to AgNPs for animals and humans. The bioaccumulation and toxicity of AgNPs depend on several factors, including AgNP concentration, physicochemical characteristics, and aggregation. So, complete activity and toxicity profiles for each silver nanoparticle synthesized must be performed. The present work aimed to characterize the behavior of specific AgNPs, then crossover of their antimicrobial capacity against Pseudomonas aeruginosa and Staphylococcus aureus with their potential in vitro cytotoxic effects in murine fibroblasts (3T3) and human keratinocytes (HaCat) cell lines, and in vivo general and organ‐specific toxicity in zebrafish embryos and larvae. A high susceptibility of P. aeruginosa and S. aureus to low concentrations of AgNPs was observed, yet a cytotoxic effect was also detected in 3T3 and HaCat cells. Moreover, these concentrations delayed embryonic development in zebrafish embryos and presented morphological abnormalities, neurotoxicity, and cardiotoxicity in zebrafish larvae. The results highlight the importance of jointly evaluating the antimicrobial activity and the toxicological profile of each nanoparticle manufactured; this study presents a suitable platform for such crossover assessment.

Abstract:Silver nanoparticles (AgNPs) are some of the most attractive nanomaterials for commercial applications, but they can also be a very challenging issue from the point of view of cytotoxicity and phytotoxicity as agents that damage genetic information within a cell causing mutations. AgNPs can be easily biosynthesized mainly by fungi which release enzymes in response to stress conditions, reducing silver ions to nanoscale size. Nowadays, mycogenic AgNPs are becoming increasingly relevant for several industrial and environmental applications, therefore, we urgently need to understand in detail how these AgNPs act and their potential effects on the environment to take measures to prevent the occurrence of hazards. The present work had the goal of evaluating the effect of spherical AgNPs synthesized by Penicillium citrinum IBCLP11 (AgNPsIBCLP11), average diameter of 57.5 ± 4 nm, against pathogenic bacteria Pseudomonas aeruginosa IPT322, Staphylococcus aureus IPT246 and Klebsiella pneumoniae IPT412 and their potential toxicity to the freshwater organisms Chlorella vulgaris, Palaemon pandaliformis and Danio rerio. AgNPsIBCLP11 presented the minimum inhibitory concentration for IPT322 strain of 15 μg•mL-1, while for IPT246 and IPT412, it ranged from 20-25 μg•mL-1. For all freshwater organisms studied, the AgNPs toxicity was lower than their metal precursor. Chlorella vulgaris growth and survival rates after 72 h of exposure to AgNPsIBCLP11, were not affected at concentrations ranging from 10 to 100 μg•L-1. AgNPsIBCLP11 and silver nitrate (AgNO3) caused no acute toxicity on adult zebrafish, although they did induce several physiological changes. In view of the results obtained, the AgNP showed promising potential for their use in freshwater systems decontamination, especially against pathogenic microorganisms, making it possible to reduce deleterious impacts on the ecosystem and enabling the conservation and management of this natural resource.

Abstract:For the first time in the world behavioral functions of laboratory mammals exposed to silver nanoparticles were studied with the regard to age. Silver nanoparticles coated with polyvinylpyrrolidone with the size of 8.7 nm were used in the present research as a potential xcenobiotic. Elder mice adapted to the xcenobiotic better than younger animals. Younger animals demonstrated more drastic anxiety than the elder ones. Thus, it is concluded that adaptive homeostasis non-linearly changes with the age. Presumably, it may improve during the prime of life and start to decline just after certain stage.

Abstract:Silver nanoparticles (AgNPs) are widely used in commerce, however, the effect of their physicochemical properties on toxicity remains debatable because of the confounding presence of Ag+ ions. Thus, we designed a series of AgNPs that are stable to surface oxidation and Ag+ ion release. AgNPs were coated with a hybrid lipid membrane comprised of L-phosphatidylcholine (PC), sodium oleate (SOA), and a stoichiometric amount of hexanethiol (HT) to produce oxidant-resistant AgNPs, Ag–SOA–PC–HT. The stability of 7-month aged, 20–100 nm Ag–SOA–PC–HT NPs were assessed using UV–Vis, dynamic light scattering (DLS), and inductively coupled plasma mass spectrometry (ICP-MS), while the toxicity of the nanomaterials was assessed using a well-established, 5-day embryonic zebrafish assay at concentrations ranging from 0–12 mg/L. There was no change in the size of the AgNPs from freshly made samples or 7-month aged samples and minimal Ag+ ion release (<0.2%) in fishwater (FW) up to seven days. Toxicity studies revealed AgNP size- and concentration-dependent effects. Increased mortality and sublethal morphological abnormalities were observed at higher concentrations with smaller nanoparticle sizes. This study, for the first time, determined the effect of AgNP size on toxicity in the absence of Ag+ ions as a confounding variable.
