Abstract:A new type of core-shell upconversion nanoparticles which can be effectively excited at 795 nm has been designed and synthesized through spatially confined doping of neodymium (Nd(3+)) ions. The use of Nd(3+) ions as sensitizers facilitates the energy transfer and photon upconversion of a series of lanthanide activators (Er(3+), Tm(3+), and Ho(3+)) at a biocompatible excitation wavelength (795 nm) and also significantly minimizes the overheating problem associated with conventional 980 nm excitation. Importantly, the core-shell design enabled high-concentration doping of Nd(3+) (~0 mol %) in the shell layer and thus markedly enhanced the upconversion emission from the activators, providing highly attractive luminescent biomarkers for bioimaging without autofluorescence and concern of overheating.

Abstract:Lanthanide-doped upconversion nanoparticles capable of converting near-infrared excitation into UV and visible emissions have attracted considerable research interests in recent years. Low-density near-infrared light needed for excitation of these nanoparticles is nondestructive and highly penetrating, offering significant technological advantages for biological and photonic applications such as remote neuron modulation, super-resolution imaging, and short-wavelength lasing. In this review, we describe several emerging classes of upconversion nanoparticles that have been developed to promote these technological applications by providing boosted emission intensities. We explain how heavy doping, thermal stimulating, and hybridizing can be harnessed to maximize absorption of excitation light and minimize quenching processes in these nanomaterials.

Abstract:Upconversion nanoparticles (UCNs) attract intensive attentions in biomedical applications. They have shown great potential in bioimaging, biomolecule detection, drug delivery, photodynamic therapy and cellular molecules interactions. Due to the anti-Stokes optical property and NIR excitation, UCNs overcome the drawbacks encountered in conventional luminescent biomarkers. High signal to noise ratio, low cytotoxicity and stable high throughput results are obtained using UCNs as luminescent labels or light triggers in biomedical applications. In this review article, the reason for choosing UCNs as biomedical agents, the progress of the UCNs development and case studies of their biomedical applications will be discussed.

Abstract:Activating upconversion nanoparticle-based photoresponsive nanovectors (UCPNVs) by upconversion visible light at low-power near-infrared (NIR) excitation can realize deeper biotissue stimulation with a minimized overheating effect and photodamage. Here, we demonstrate a facile strategy to construct new surface-decorated UCPNVs based on Passerini three-component reaction (P-3CR) in highly convenient and effective manners. Such UCPNVs materials have a tailored deprotecting wavelength that overlaps upconversion blue light. By using 3-perylenecarboxaldehyde, Tm3+/Yb3+ ion-doped UCNP-containing isocyanides, and antitumor agent chlorambucil as the three components, the resulting monodisperse UCPNV exhibits an efficient release of caged chlorambucil under a very low 976 nm power. This approach expands the synthetic toolbox to enable quick development of UCPNVs for UCNP-assisted low-power NIR photochemistry.

Abstract:Near-infrared (NIR) light responsive nanomaterials have attracted considerable attention due to their location in the biological window. Especially, lanthanide-doped nanomaterials exhibit unique upconversion luminescence (UCL) properties with low background interference and long luminescence lifetime, which makes them promising in imaging diagnosis of cancers. Compared with traditional upconversion nanomaterials excited by 980 nm laser, 808 nm excitation can overcome the disadvantages of high heating effect and weak penetration depth induced by excitation source. Therefore, developing 808 nm excited upconversion nanoprobes will be important for the in vivo bio-imaging and visualized theranostics of tumors in deep-tissue. In the review paper, we systematically summarized the synthesis strategy and luminescence modulation of Nd3+-sensitized upconversion nanomaterials under 808 nm excitation, discussed the influence of excitation source on heating effect and penetration depth, and particularly focused on their applications in UCL imaging, multi-modal imaging and imaging-guided therapy of cancers.

Abstract:With the rapid development of nanotechnology, the unique rare-earth lanthanide-doped upconversion nanocrystals (UCNs), which can convert tissue-penetrable near-infrared (NIR) photonic irradiation into ultraviolet, visible, and NIR emissions, have a significant potential in bioimaging, diagnosis, and therapy, as well as in photovoltaic systems and optical data storage. Despite the promising achievements made in the past decade, critical challenges associated with low upconversion efficiencies and the overheating effect induced by NIR laser-irradiation still remain in the biomedical fields. In high demand are more well-defined material design and unique structural modifications that are capable of solving these technical concerns and promoting such promising NIR light-mediated upconversion nanocrystals for their further application in the medical sciences. Recent advances in upconversion nanomaterials have witnessed a tremendous development towards enhancing their photon conversion efficiency, which provides great opportunities in expanding the potential of the UCNs in bioimaging diagnosis and anticancer therapy. Hence, this review is mainly focused on summarizing the fundamental principles and strategies that improve upconversion luminescence and the approaches to reduce the local thermal effect on the basis of a rational design of UCNs. In addition, the future perspectives in the development of UCNs for biomedical applications are also proposed.

Abstract:Lanthanide-doped upconverting nanoparticles (UCNPs) possess uniqueanti-Stokes optical properties, in which low energy near-infrared (NIR) photons can beconverted into high energy UV, visible, shorter NIR emission via multiphoton upconversionprocesses. Due to the rapid development of synthesis chemistry, lanthanide-doped UCNPscan be fabricated with narrow distribution and tunable multi-color optical properties. Theseunique attributes grant them unique NIR-driven imaging/drug delivery/therapeuticapplications, especially in the cases of deep tissue environments. Abstract:Upconversion nanoparticles (UCNPs) that emit high-energy photons upon excitation by the low-energy near-infrared (NIR) light are emerging as new optical nano-probes useful in biomedicine. Herein, we load Chlorin e6 (Ce6), a photosensitizer, on polymer-coated UCNPs, forming a UCNP-Ce6 supramolecular complex that produces singlet oxygen to kill cancer cells under NIR light. Excellent photodynamic therapy (PDT) efficacy is achieved in tumor-bearing mice upon intratumoral injection of UCNP-Ce6 and the followed NIR light exposure. It is further uncovered that UCNPs after PDT treatment are gradually cleared out from mouse organs, without rendering appreciable toxicity to the treated animals. Moreover, we demonstrate that the NIR-induced PDT based on UCNP-Ce6 exhibits a remarkably increased tissue penetration depth compared to the traditional PDT using visible excitation light, offering significantly improved treatment efficacy for tumors blocked by thick biological tissues. Our work demonstrates NIR light-induced in vivo PDT treatment of cancer in animals, and highlights the promise of UCNPs for multifunctional in vivo cancer treatment and imaging.

Abstract:Photodynamic therapy (PDT) is a non-invasive treatment modality for a variety of diseases including cancer. PDT based on upconversion nanoparticles (UCNPs) has received much attention in recent years. Under near-infrared (NIR) light excitation, UCNPs are able to emit high-energy visible light, which can activate surrounding photosensitizer (PS) molecules to produce singlet oxygen and kill cancer cells. Owing to the high tissue penetration ability of NIR light, NIR-excited UCNPs can be used to activate PS molecules in much deeper tissues compared to traditional PDT induced by visible or ultraviolet (UV) light. In addition to the application of UCNPs as an energy donor in PDT, via similar mechanisms, they could also be used for the NIR light-triggered drug release or activation of 'caged' imaging or therapeutic molecules. In this review, we will summarize the latest progresses regarding the applications of UCNPs for photodynamic therapy, NIR triggered drug and gene delivery, as well as several other UCNP-based cancer therapeutic approaches. The future prospects and challenges in this emerging field will be also discussed.

Abstract:Photodynamic therapy (PDT) has been considered as a promising and noninvasive strategy for clinical cancer treatment. Nonetheless, building a smart "off-on" theranostic PDT platform to spatiotemporally control the generation of reactive oxygen species in the PDT treatment still remains challenging. Here, we have rationally developed photoswitching upconversion nanoparticles (UCNPs) with orthogonal emissive properties in response to two distinct near-infrared (NIR) emissions at 808 and 980 nm, , red emission with 980 nm excitation and green emission with 808 nm excitation. Unlike traditional photoswitching UCNPs, these specially designed core-shell-shell structured UCNPs do not require complicated multilayer doping as their red and green upconversion luminescence both originate from the same activator Er ions in the core structure. As a proof of concept, we have demonstrated the capability of these orthogonal emissive UCNPs for imaging-guided PDT in a real-time manner, where the red emission excited by 980 nm light is used to trigger PDT and the green emission with 808 nm excitation is to diagnose and monitor the therapeutic treatment. Our study suggests that such specially designed UCNPs with orthogonal emissions hold great promise for NIR light-targeted and imaging-guided therapy under precisely spatiotemporal control.

Abstract:Controlled activation or release of biomolecules is very crucial in various biological applications. Controlling the activity of biomolecules have been attempted by various means and controlling the activity by light has gained popularity in the past decade. The major hurdle in this process is that photoactivable compounds mostly respond to UV radiation and not to visible or near-infrared (NIR) light. The use of UV irradiation is limited by its toxicity and very low tissue penetration power. In this study, we report the exploitation of the potential of NIR-to-UV upconversion nanoparticles (UCNs), which act as nanotransducers to absorb NIR light having high tissue penetration power and negligible phototoxicity and emit UV light locally, for photoactivation of caged compounds and, in particular, used for photo-controlled gene expression. Both activation and knockdown of GFP was performed in both solution and cells, and patterned activation of GFP was achieved successfully by using upconverted UV light produced by NIR-to-UV UCNs. In-depth photoactivation through tissue phantoms and in vivo activation of caged nucleic acids were also accomplished. The success of this methodology has defined a unique level in the field of photo-controlled activation and delivery of molecules.

Abstract:Upconversion is an optical process that involves the conversion of lower-energy photons into higher-energy photons. It has been extensively studied since mid-1960s and widely applied in optical devices. Over the past decade, high-quality rare earth-doped upconversion nanoparticles have been successfully synthesized with the rapid development of nanotechnology and are becoming more prominent in biological sciences. The synthesis methods are usually phase-based processes, such as thermal decomposition, hydrothermal reaction, and ionic liquids-based synthesis. The main difference between upconversion nanoparticles and other nanomaterials is that they can emit visible light under near infrared irradiation. The near infrared irradiation leads to low autofluorescence, less scattering and absorption, and deep penetration in biological samples. In this review, the synthesis of upconversion nanoparticles and the mechanisms of upconversion process will be discussed, followed by their applications in different areas, especially in the biological field for biosensing.

Abstract:NO is a key vasodilator in mammalian cardiovascular systems and has been shown to sensitize tissue to γ-radiation. However, photochemical NO precursors that can be activated by these NIR wavelengths are extremely limited. G. D. Stucky, D. Zhao, P. C. Ford, F. Zhang and co-workers have addressed this issue by utilizing the known NIR to visible upconversion properties of lanthanide-doped NaYF4 nanocrystals. This result, described on page 3800, is a potential game changer in multiphoton excitation-based therapeutic delivery of NO and other small molecule bioregulators to physiological targets.

Abstract:<p>Light in the near‐infrared (NIR) spectral region is increasingly utilized in bioapplications, providing deeper penetration in biological tissues owing to the lower absorption and scattering in comparison with light in the visible range. Lanthanide‐doped luminescent nanoparticles with excitation and/or emission in the NIR range have recently attracted tremendous attention as one of the prime candidates for noninvasive biological applications due to their unique optical properties, such as large Stokes shift, spectrally sharp luminescence emissions, long luminescence lifetimes, and excellent photostability. Herein, recent advances of lanthanide‐doped nanoparticles with NIR upconversion or downshifting luminescence and their uses in cutting‐edge biophotonic applications are presented. A set of efficient strategies for overcoming the fundamental limit of low luminescence brightness of lanthanide‐doped nanoparticles is introduced. An in‐depth literature review of their state‐of‐art biophotonics applications is also included, showing their superiority for high‐resolution imaging, single‐nanoparticle‐level detection, and efficacy for tissue‐penetrating diagnostics and therapeutics.</p>

Abstract:Near-infrared (NIR) dye-sensitized upconversion nanoparticles (UCNPs) can broaden the absorption range and boost upconversion efficiency of UCNPs. Here, we achieved significantly enhanced upconversion luminescence in dye-sensitized core/active shell UCNPs via the doping of ytterbium ions (Yb(3+)) in the UCNP shell, which bridged the energy transfer from the dye to the UCNP core. As a result, we synergized the two most practical upconversion booster effectors (dye-sensitizing and core/shell enhancement) to amplify upconversion efficiency. We demonstrated two biomedical applications using these UCNPs. By using dye-sensitized core/active shell UCNP embedded poly(methyl methacrylate) polymer implantable systems, we successfully shifted the optogenetic neuron excitation window to a biocompatible and deep tissue penetrable 800 nm wavelength. Furthermore, UCNPs were water-solubilized with Pluronic F127 with high upconversion efficiency and can be imaged in a mouse model.

Abstract:With multiple emissions ranging from NIR-IIb to visible lights, near-infrared light-excited lanthanide nano-particle (LnNP) is an ideal in-vivo theranostic platform to achieve imaging guided phototherapy. However, current reported LnNPs typically demonstrate simultaneous up and downconversion emissions with fixed single excitation light, which impairs therapeutic efficiency and generates side effect during navigation. Here we develop a lanthanide-based conversion switching nanoparticle (CSNP) with independent activation of 1550 nm NIR-IIb downconversion emission under 808 nm excitation and 345/450 nm upconversion emission under 980 nm excitation. CSNP is modified with Cy-GSH to quench NIR-IIb emission and photosensitizer hypocrellin A. In vivo delivery of CSNP is traced via 808 nm irradiation, and Cy-GSH changes structure in response to glutathione to activate NIR-IIb imaging. This indicates the tumor position and timing to switch for 980 nm irradiation to activate hypocrellin A for photodynamic therapy. Orthogonal activation of CSNP up/down conversion emissions demonstrates high tumor-to-normal tissue ratio in vivo and good therapeutic result, would have promising po-tential as a theranostics platform.
