Abstract:The paper presents a novel strategy to identify analytical markers of traditional Chinese medicine preparation (TCMP) rapidly via direct analysis in real time mass spectrometry (DART-MS). A commonly used TCMP, Danshen injection, was employed as a model. The optimal analysis conditions were achieved by measuring the contribution of various experimental parameters to the mass spectra. Salvianolic acids and saccharides were simultaneously determined within a single 1-min DART-MS run. Furthermore, spectra of Danshen injections supplied by five manufacturers were processed with principal component analysis (PCA). Obvious clustering was observed in the PCA score plot, and candidate markers were recognized from the contribution plots of PCA. The suitability of potential markers was then confirmed by contrasting with the results of traditional analysis methods. Using this strategy, fructose, glucose, sucrose, protocatechuic aldehyde and salvianolic acid A were rapidly identified as the markers of Danshen injections. The combination of DART-MS with PCA provides a reliable approach to the identification of analytical markers for quality control of TCMP.
Online Analysis of Dynamic Trend Regression and EndpointDetermination for Chinese Traditional Medicine Extraction Process Based on Ultraviolet Spectroscopy

Abstract:With the transition of Chinese traditional medicine manufacture industry, modernization has become the inexorable tendency in its future development. However, during the current Chinese traditional medicine producing process, the lack of online monitoring leads to the lagging of quality detection, as well as quality differences between products. In this paper, aiming at realizing online monitoring and end point automatic determination for Chinese traditional medicine (CTM) extraction unit, which is one of the most important units in CTM producing, ultraviolet (UV) spectroscopy is applied to build UV absorbance dynamic model based on the Lambert-Beer law and the CTM extraction kinetic model which presents a new method of UV absorbance dynamic analysis and endpoint determination, including curve regression, robustness analysis and endpoint calculation. In the experiment for online monitoring homalomena occulta extraction, the online UV spectral collection system for CTM extraction, developed by our laboratory, was applied for spectral collection; meanwhile, solid component contents in the offline samples were measured as reference. During the analysis, first of all, we pretreated the spectrum collected in the current period with interpolation and smoothing, and calculated the mean value within a UV region of 230.2～400 nm to form an absorbance mean value sequence with data obtained in the early measurements; then we verified the linear correlation between the sequence of absorbance and the concentrations of effective component in the solution, the linear correlation coefficient equals 0.982 8, showing a high linearity between UV spectra and solid component contents; finally, we regressed the absorbance mean value sequence with the dynamic model, analyzing its robustness and the extraction endpoint. Experimental results demonstrate that, the robustness analysis could recognize the bad points of measurement during the regression process, and improve the consistency between the regression and the original curves, raising its squared correlation coefficient to more than 0.99; meanwhile, with endpoint determination, we shortened the homalomena occulta extraction process from the original manually set 180 to 122 min effectively. The experiment above proves that this method with UV spectroscopy realizes online monitoring and automatic endpoint determination for the CTM extraction process, and is of significant importance for stabilizing production as well as improving economic benefit.

Abstract:Carbon quantum dots (CQDs) are fluorescent carbon nanomaterials that have been applied to biology, medicine, and optoelectronics, owing to their significant advantages such as simple synthesis methods, low cost, and widely available sources of raw synthesis materials. This review summarizes CQD preparation methods, which include hydrothermal and microwave-assisted synthesis methods, as well as separation methods such as centrifugation, dialysis, and filtration. Additionally, we review the application of CQDs in the detection of active ingredients, primarily phenolic compounds, in traditional Chinese medicine. We also discuss the quenching mechanism of CQD fluorescence using the active ingredients of traditional Chinese medicine. Limitations such as insufficient test selectivity, weak fluorescence intensity, and an unclear quantitative relationship between preparation methods and properties should be resolved for the efficient use of CQDs to detect active ingredients in Chinese medicine.

Abstract:Recent years, research on the detection of heavy metals in Traditional Chinese Medicine (TCM) by laser induced breakdown spectroscopy (LIBS) have gradually increased. Current main methods of establishing calibration curve are based on grounding and pelleting of the tested samples. Although compared to digested samples, grounding and pelleting of the sample is already quite simple, it cannot fully reflect the advantages of LIBS: rapid analysis, and, also, the uneven distribution of heavy metals in the TCM is ignored. In order to avoid grinding and pelleting sample to be tested, and to achieve microdamage quantitative analysis by LIBS, this article presents a new method for establishing calibration curve. The experiment in this paper based on a study with Cd and Pb in leaves of laurel. The preparation of calibration samples and the establishment of calibration methods for microdamage quantitative analysis were presented, which proved the feasibility of microdamage quantitative analysis by LIBS. The square of the linear relationship coefficient R of Pb was higher than 0.82. This method provides a guiding method for the rapid quantitative analysis of heavy metals in TCM by LIBS.

Abstract:Laser-induced breakdown spectroscopy (LIBS) technique is widely used to analyze the composition and the concentration of elements in materials,which is based on the atomic excitation and emission spectrum.It has many advantages,such as rapid,multi-element simultaneous detection and without sample pretreatment.In this paper,research progress on the application of metal element detection in such fields as alloy,soil,water,and food is reviewed.It is considered that there are some problems in this technology at present,such as low detection accuracy,poor repeatability,unstable detection signal.It is also found that the research and development of LIBS based metal element detection instrument should be developed towards the direction of conventional,portable and high precision.
Quantitative aluminum alloys analysis using laser-induced breakdown spectroscopy

Abstract:An intense laser radiation was focused on the surface of the sample to generate a plasma plume which vaporized a small amount of alloy material. The emission spectrum emitted as the plasma cooling off, which contained the information of elemental species and concentration in alloy, was collected and analyzed by high-resolution echelle grating spectrometer coupled to intensified charge coupled device (ICCD) camera. A series of standard aluminum alloy reference samples were analyzed for their alloying elements, including Fe, Si, and Cu. Results for Fe, Si, and Cu indicated limits of detection that ranged from 6 × 10-5 - 10 × 10-5 and almost all measurement relative errors less than 5% when mass concentration are over 0.01%. The absolute measurement errors for element with mass concentration under 0.01% were less than 0.001%.

Abstract:Chinese medicinal injection, made of active components extracted from Chinese medicine or Chinese medicinal compound, is a novel dosage form of Chinese patent medicine in China and is pivotal in the traditional Chinese medicine(TCM) industry. The quality control standard of Chinese medicinal injection determines its safety and efficacy. The quantitative nuclear magnetic resonance(qNMR) spectroscopy is a non-targeted, non-invasive, and non-destructive technique with high reproducibility, short measurement time, convenient sample preparation, a broad range of linearity, and no requirement on the reference substance of tested components, which is advantageous as compared with traditional chromatographic methods, and it can provide information about the molecular composition of the tested samples. Therefore, in light of multiple challenges in the quality control of Chinese medicinal injection, such as complex composition, difficulties in quantitative analysis, and the shortage of reference substances, the application of qNMR spectroscopy combined with chemometrics techniques was proposed for the quality evaluation of Chinese medicine reference substances, Chinese medicinal injection, and intermediates in the production process, as well as for the stability analysis of Chinese medicinal injection. This study is expected to provide references for the application of qNMR spectroscopy in the quality control of Chinese medicinal injection.

Abstract:Heavy metal pollution is more and more serious in agriculture products.In the experiment,the rice polluted by cadmium is used as an example.We adopt coaxial dual pulse laser induced breakdown spectroscopy and the experimental parameters are optimized.The application of dual pulse-laser induced breakdown spectroscopy (DP-LIBS) in the area of monitoring the quality and safety of the farm products are explored.The feasibility of improving the sensitivity and accuracy for detecting the heavy metal in agricultural products is disscussed.The intensity of LIBS spectra and the real concentration of Cd of the nine concentration gradients samples are obtained by LIBS and anodic stripping voltammetry.By using univariate linear scaling regression model,three characteristic spectrum of Cd is analyzed quantitatively.The results indicate that the lowest mass fraction of Cd is detected by DP-LIBS with 5.03 μg/g and all the spectra lines have good correlation coefficients.The experiment shows that DP-LIBS can improve the detecting sensitivity and accuracy of Cd,its concentration information of Cd is got according to the linear regression model.
[Rapid multi-elemental analysis on four precious Tibetan medicines based on LIBS technique]

Abstract:The laser-induced breakdown spectroscopy (LIBS) was applied to perform a qualitative elementary analysis on four precious Tibetan medicines, i. e. Renqing Mangjue, Renqing Changjue, 25-herb coral pills and 25-herb pearl pills. The specific spectra of the four Tibetan medicines were established. In the experiment, Nd: YAG and 1 064 nm-baseband pulse laser were adopted to collect the spectra. A laser beam focused on the surface of the samples to generate plasma. Its spectral signal was detected by using spectrograph. Based on the National Institute of Standard and Technology (NIST) database, LIBS spectral lines were indentified. The four Tibetan medicines mainly included Ca, Na, K, Mg and other elements and C-N molecular band. Specifically, Fe was detected in Renqing Changjue and 25-herb pearl pills; heavy mental elements Hg and Cu were shown in Renqing Mangjue and Renqing Changjue; Ag was found in Renqing Changjue. The results demonstrated that LIBS is a reliable and rapid multi-element analysis on the four Tibetan medicines. With Real-time, rapid and nondestructive advantages, LIBS has a wide application prospect in the element analysis on ethnic medicines.
[Quantitative determination of trace heavy metals in polluted soil by laser induced breakdown spectroscopy]

Abstract:Based on laser induced breakdown spectroscopy data analysis, a quantitative determination method of elements was introduced. This method takes the advantage of Fourier analysis to study different part of the spectrum-white noise, thermal radiation and emission spectral lines. Through band-pass filter, emission spectral lines and most noise were separated, only a little white noise was missed. By calculated similarity between detected line and its corresponding unit characteristic line, convolution intensity was introduced to measure the detected line intensity. With Cu 324.75 nm as the analysis line and Ti 337.28 nm as the base line, the way of separating spectrum mentioned before was carried out to detect trace Cu pollution in soil. Results contained two bright spots. First, The correlation coefficient between Cu concentration and the spectral intensity was up to 0.997 9. Last but not the least, the detectability of Cu in soil was 44 mg x kg(-1), which is below the trace element thresholds for Class 2 soil used in the environmental quality standard for soil in China. At the same time, the relative error is always below 10%. Therefore the feasibility of this method for portable and instant detection of heavy metals in polluted soil was verified.

Abstract:<p>In the micro-laser-induced breakdown spectroscopy field, when the laser ablated crater on the surface of the sample is less than 10 μm, it is difficult to detect the plasma emission line because of short lifetime of excited plasma, and simultaneous quantitative analysis of multiple elements is rarely reported. In this paper, a picosecond LIBS micro-area analysis scheme is proposed. Compared with the femtosecond laser micro-area analysis system, this scheme is more mature and reliable, and is more beneficial to engineering applications. In addition, in this study, the effect of the number of laser pulses on the crater morphology was studied. It was observed that a certain crater depth would have an enhancement effect on the signal, and by controlling the laser energy, a crater less than the diffraction limit was obtained. Moreover, a multielement simultaneous analysis on a set of molybdenum-copper stainless steel samples containing 40%-70% iron was performed when the ablated crater had an outer diameter of approximately 5 μm and an inner diameter of approximately 3 μm. By accumulating spectral intensity, background removal, and abnormal point processing, the calibration results for the eight elements of Mn, Cr, Ni, Cu, Ti, V, Nb and Al were not less than 0.95 using the basic calibration method. Among them, the calibration results for Ni, Cu, V, Nb, Al elements are all higher than 0.99 and the detection limits of the obtained elements range from 0.04% to 1.8%.</p>

Abstract:In the present work, spatial character of emission spectrum was analyzed, the effect of laser energy and sample, attribute on the best detection position for the highest signal to noise ratio was studied, and sonic experimental investigations with LIBS technique to detect trate Cu in polluted soil were carried out in our laboratory. A Q-switched Nd : YAG laser operating at 1 064 nm with pulse width of 10 ns and repetition frequency of 1 Hz was utilized. The laser pulse was focused by lens with focal length of 10 cm to generate, microplasmas on the surface of printed circuit board and soil samples. The sample was adjustable by vernier construction to detect the emission spectrum of the microplasmas from different position. Experiments showed that the intensity of thermal radiation and atomic radiations evolved differently while the detection position changed. It was verified that thermal radiation reduced rapidly with the distance from the center of spark increasing, while the intensity of atomic radiations increased firstly and decreased after intensity maximum was reached. The method of separating thermal radiation and atomic radiations in space brought on high signal-to-noise ratio. It was found that the best detection position was 0.75 mm off the center of the spark for soil sample while the laser energy was 10 mJ, and the distance increased with the growth of laser energy. With Cu 324.75 nm and Cu 327.39 nm as the analysis lines, the best detection position was selected to detect trace Cu pollution in soil. Internal standard method was used to determine the relation between Cu concentration and Its intensity. It was concluded that the detection limit of Cu in soil was 67 mg . kg(-1), which is below the trace element thresholds for Class 2 soil defined in the Environmental Quality Standard for Soil in China. It was proven an effective way to achieve higher signal-to-noise ratio by adjusting the location of spectral measurements. This method was viable for trace Cu detection in polluted soil.

Abstract:Laser induced breakdown spectroscopy is an emerging elemental analytical technique with the advantage of fast response, minimal sample preparation and multi-element analysis capacities. This work studied practical use of LIBS for rapid quantification of 4 (Cu, Ni, Cr, Pb) heavy metal elements in 169 agricultural soil samples. The objective is to conclude an appropriate method to reduce the interference of matrix effect in soils within the scope of data analysis, by comparing several univariate and multivariate methods of LIBS data interpretation (full spectrum and emission lines). The proposed multivariate methods, such as the least absolute shrinkage and selection operator and principal components regression were found to be effective in reducing the matrix interference and the predictive performance was stable in our experiment, approaching normalized root mean squared error of 6.84%, 8.87%, 9,71%, 10.76% for Cu, Ni, Cr and Pb, respectively. Meanwhile, the performance of univariate analysis suffered from such effect. The results showed the importance of utilizing proper multivariate chemometrics for LIBS data interpretation when quantifying the heavy metals in soils.

Abstract:Laser-Induced Breakdown Spectroscopy (LIBS) instruments are increasingly recognized as valuable tools for detecting trace metal elements due to their simplicity, rapid detection, and ability to perform simultaneous multi-element analysis. Traditional LIBS modeling often relies on empirical or machine learning-based feature band selection to establish quantitative models. In this study, we introduce a novel approach-simultaneous multi-element quantitative analysis based on the entire spectrum, which enhances model establishment efficiency and leverages the advantages of LIBS. By logarithmically processing the spectra and quantifying the cognitive uncertainty of the model, we achieved remarkable predictive performance (R2) for trace elements Mn, Mo, Cr, and Cu (0.9876, 0.9879, 0.9891, and 0.9841, respectively) in stainless steel. Our multi-element model shares features and parameters during the learning process, effectively mitigating the impact of matrix effects and self-absorption. Additionally, we introduce a cognitive error term to quantify the cognitive uncertainty of the model. The results suggest that our approach has significant potential in the quantitative analysis of trace elements, providing a reliable data processing method for efficient and accurate multi-task analysis in LIBS. This methodology holds promising applications in the field of LIBS quantitative analysis.
The multi-dimensional characterization of Al-doped CuO thin films by laser-induced breakdown spectroscopy (LIBS) with nanosecond and picosecond lasers

Abstract:Glasses coated with various optical thin film materials are widely used in people's life and scientific research. In this study, a series of aluminum-doped (Al-doped) copper oxide (CuO) thin films with different sputtering pressures and powers were deposited on soda-lime glass substrates by RF-magnetron sputtering to explore the effect of doping element on the optical properties of the thin films. However, laser-induced breakdown spectroscopy (LIBS) with nanosecond and picosecond lasers was employed to achieve the multi-dimensional analysis of the Al element. With ns-LIBS, the rapid quantitative analysis was performed by drawing the calibration curves of LIBS intensity ratio of Al/Cu versus concentration ratio under different sputtering pressures and powers. Furthermore, the 2D chemical mappings were drawn to provide a more complete and detailed relative spatial distribution of the Al element according to the LIBS intensity ratio of Al/Cu. Meanwhile, the thin film thickness was determined by the picosecond laser pulse number since the depth ablated by a single laser pulse was assessed as nearly 46.75 ± 4.25 nm. Moreover, the variation trends of optical band gaps with sputtering parameters were similar to that of LIBS intensity ratios with sputtering parameters, as suggested by analyzing the transmission spectra of thin films. The results demonstrated that the multi-dimensional characterization (quantitative analysis, 2D chemical mapping, thin-film thickness estimation, and optical properties analysis) of Al-doped CuO thin films prepared by RF-magnetron sputtering can be effectively realized by LIBS.

Abstract:In the field of quantitative analysis utilizing laser induced breakdown spectroscopy (LIBS), ensuring the stability and precision of the technique is a crucial consideration. This is particularly pertinent for certain engineering applications, where utilizing electric translation stages for sample scanning is not feasible. In this study, a compact optical scanning LIBS analysis system was developed using a deflection mirror design to replace the translation platform. The system achieved rapid scanning and precise analysis, with a total weight of less than 5 kg. Moreover, to quantitatively analyze Mn, Ni, Cr, Cu, and other elements in various alloy steels, a general method for selecting spectral analysis line combination was proposed. The calibration model was established utilizing 16 alloy steel samples, and its validity was assessed through a verification set of seven samples (with element content ranging from 0.021 to 2.0 wt%). The results indicate a high degree of accuracy. The calibration model determination coefficients ( ) for Mn, Ni, Cr, and Cu elements exceeded 0.99. The root-mean-square error of validation (RMSEV) was less than 0.05, and the relative standard deviation (RSD) was not more than 4%. Additionally, the relative prediction errors for Mn range from 2.98% to 67.44%, for Ni range from 0.57% to 15.93%, for Cr range from 3.91% to 55.56%, and for Cu range from 0.95% to 38.1%.

Abstract:A nanoparticle-coated graphite target (NCGT) is presented to improve the analysis accuracy and stability of laser-induced breakdown spectroscopy (LIBS). A stable, relatively homogeneous, and close to optically thin laser-induced breakdown plasma was obtained by dispersing sample nanoparticles on a high-purity graphite substrate. Spectral structures dominated by the characteristic lines of carbon and the samples can greatly simplify spectral identification and avoid line interference. To maximize the analysis accuracy and stability, a series of experimental conditions were optimized step by step according to the spectral intensity and signal-to-noise ratio of the lines. Based on the final optimized conditions, the relative standard deviation values of Mg, Fe, and Sr elemental content in Chinese medicinal material (CMM) samples were reduced from 17.7, 16.6, 12.1% of the pressed target to 4.8, 9.5, and 4.5% of the NCGT, respectively. Comparisons with the inductively coupled plasma mass spectrometry (ICP-MS) results demonstrated that the present method has great potential for detection of LIBS.

Abstract:In this work, a simple analytical strategy combining high-performance liquid chromatography–diode array detection (HPLC-DAD) and the chemometric method was developed for the simultaneous quantification of 5-hydroxymethyl-2-furfural (HMF), paeoniflorin (PAE), ferulic acid (FER), baicalin (BAI), and berberine (BER) in a Chinese medicine formula Wen-Qing-Yin (WQY). The alternating trilinear decomposition (ATLD) algorithm and alternating trilinear decomposition assisted multivariate curve resolution (ATLD-MCR) algorithm were used to realize the separation and rapid determination of five target analytes under the presence of time shifts, solvent peaks, peak overlaps, and unknown interferences. All analytes were eluted within 10 min and the linear correlation coefficients of calibration sets were between 0.9969 and 0.9996. In addition, the average recoveries of the five active compounds obtained by ATLD and ATLD-MCR analysis were in the range of 91.8–112.5% and 88.6–101.6%, respectively. For investigating the accuracy and reliability of the proposed method, figures of merit including limit of detection (LOD), limit of quantitation (LOQ), sensitivity (SEN), and selectivity (SEL) were calculated. The proposed analytical strategy has the advantages of being fast, simple, and sensitive, and can be used for the qualitative and quantitative analysis of WQY, providing a feasible option for the quality monitoring of the traditional Chinese medicine formula.

Abstract:Chinese herbal medicines are often referred to as Chinese materia medica (CMM). Composite formulae containing mixtures of CMM are prescribed for treatment and prevention of diseases in the practice of traditional Chinese medicine (TCM). Some of the well-known CMM formulae (Fufang in Chinese) are manufactured and marketed as proprietary Chinese medicines (PCM). Quality assessment and assurance of these products are difficult; they are a challenging task. Mid-infrared spectroscopy, a classic molecular structure analysis method, has been innovatively applied in the quality control of TCM, and has gained significant impact and advancement in analytical fields. Infrared fingerprinting features appear particularly suitable for the identification of multicomponent matrices in samples whose chemical integrity has not been altered or destroyed because no extraction procedure is needed. This review summarizes and gives an overall view on the application of mid-infrared and two-dimensional correlation infrared (2D-IR) spectroscopy as well as chemometric techniques in the identification of CMM, investigation of TCM processing procedures, and analysis of herb extracts and preparations.
