Abstract:Suzuki-Miyaura cross-coupling reaction is an efficient and utilized method for the direct formation of carbon-carbon bonds. The effectiveness and efficiency of Suzuki-Miyaura cross-coupling reaction and its applications have been the topic of interest for synthetic chemists for the last few decades. Green chemistry is the area where we use eco-friendly products. Suzuki coupling includes palladium or nickel catalyzed coupling reaction, which involves ester of boric acids or simply boric acids with the organic halides or pseudohalide. In recent years, these catalytic systems have been developed in a green environment for Suzuki reaction (Suzuki-Miyaura cross-coupling reaction). This review epitomizes the Suzuki-Miyaura cross-coupling reaction using efficient catalysts in various green media.

Abstract:Abstract In the history of catalysis and organic synthesis, boric chemistry has been developed into one of the most varied and practical disciplines. Several useful Suzuki–Miyaura cross-coupling (SMC) reactions as well as hydroborylation reactions are regarded the essential tools inside the chemical synthesis toolkit accompanied by researchers of the industry or the academia. Novel synthesis of the less electrically and sterically ongoing carbon–boron synthetic reagents is carried out to ensure a unique coupling reaction utilizing metals along with these reagents which draws considerable interest in accelerating the discovery of creative uses for otherwise difficult organic adducts in many disciplines. This article details the noteworthy advancements in the use of traditional metal-catalyzed carbon–carbon coupling processes with cutting-edge coupling partners such as carbon–boron reagents often the beta-alkyl Suzuki–Miyaura coupling since 2001. The current review covers alkylboranes, organotrifluoroborates, 9-BBN, alkylboronic acids and boronic esters as useful reagents in SMCs that will help synthetic chemists in developing new compounds. Abstract:During the past several decades, Suzuki-Miyaura cross-coupling reaction has emerged and flourished as one of the most remarkable and practical methods to construct C-C bond.The substrates, organoboronic compounds, are usually more stable, less toxic and easier to obtain compared with other metal or metalloid alternatives.Suzuki-Miyaura cross-coupling reaction possesses extraordinary merits of demonstrated efficiency, broad functional group compatibility and mild reaction conditions.It has been the premier method to construct C-C bond for organic synthetic chemists and has been widely applied in the industrial syntheses.The rapid development of Suzuki-Miyaura cross-coupling reaction is accompanied with the progress of mechanism studies.It is well accepted that the catalytic cycle of Suzuki-Miyaura cross-coupling reaction contains three main steps: oxidative addition of organic halides to Pd catalysts, transmetalation between oxidative addition intermediates and organoboronic compounds, and reductive elimination from transmetalation intermediates to form C-C bond along with regenerating Pd catalysts.However, these three main steps can be influenced by diverse reaction conditions, and different reaction pathways may occur within each single step.Furthermore, beyond the oxidative addition and transmetalation intermediates, other transient but crucial intermediates may exist.To figure out these issues and discover more insights into Suzuki-Miyaura cross-coupling reaction, numerous researchers have designed and conducted systematic investigations.Herein, the recent progress of mechanism studies is summarized and discussed.The steps of oxidative addition, transmetalation and reductive elimination are discussed in detail.Moreover, a brief introduction of transition-metal-free and base-free Suzuki-Miyaura cross-coupling reactions which show more possibilities of the reaction mechanism is given.Besides summary of precedent research works, our opinions about future development of the mechanism studies are also given.Although breakthroughs have been made in the mechanism studies, further investigations are needed to bring comprehensive knowledge of this reaction and help us further optimize the reaction conditions.

Abstract:Synthetic organic chemistry experienced a significant advance in the last quarter of the 20th century with the advent of the transition-metal catalyzed cross-coupling reactions. The utility of these reactions was universally acknowledged and expressed in the Chemistry Nobel prize awarded to professors Heck, Suzuki and Negishi in 2010. In this scenario, the discovery of the Suzuki-Miyaura reaction was a landmark that occupies a privileged position because of its versatility, compatibility and pivotal contribution to diverse disciplines, including materials science and drug discovery. Despite the substantial progress attained for this reaction until 2010, there is still room for improvement, particularly, if we are concerned about the health of the Earth and aware of the paramount importance to produce substances under the guidelines of sustainable and Green Chemistry. In the present review, we summarize the efforts devoted by a wide and specialized scientific community to upgrade the Suzuki-Miyaura reaction during the post-Nobel prize period. An overview covering new catalytic systems, methods and conditions is provided, together with some applications and updated mechanistic viewpoints.

Abstract:Cross coupling reactions have changed the way complex molecules are synthesized. In particular, Suzuki-Miyaura and Buchwald-Hartwig amination reactions have given opportunities to elegantly make pharmaceutical ingredients. Indeed, these reactions are forefront at both the stages of drug development, medicinal chemistry, and process chemistry. On one hand, these reactions have given medicinal chemists a tool to derivatize the core molecule to arrive at scaffold rapidly. On the other hand, these cross couplings have offered the process chemists a smart tool to synthesize the development candidates safely, quickly, and efficiently. Generally, the application of cross coupling reactions is broad, and this review will specifically focus on their real (pharma) world applications in large scale synthesis those appeared in last two years.

Abstract:Palladium-catalyzed Suzuki-Miyaura cross-coupling or aryl halides is widely employed in the synthesis of many important molecules in synthetic chemistry, including pharmaceuticals, polymers and functional materials. Herein, we disclose the first palladium-catalyzed decarbonylative Suzuki-Miyaura cross-coupling of amides for the synthesis of biaryls through the selective activation of the N-C(O) bond of amides. This new method relies on the precise sequence engineering of the catalytic cycle, wherein decarbonylation occurs prior to the transmetallation step. The reaction is compatible with a wide range of boronic acids and amides, providing valuable biaryls in high yields (>60 examples). DFT studies support a mechanism involving oxidative addition, decarbonylation and transmetallation and provide insight into high N-C(O) bond activation selectivity. Most crucially, the reaction establishes the use of palladium catalysis in the biaryl Suzuki-Miyaura cross-coupling of the amide bond and should enable the design of a wide variety of cross-coupling methods in which palladium rivals the traditional biaryl synthesis from aryl halides and pseudohalides.

Abstract:The story of C-C bond formation includes several reactions, and surely Suzuki-Miyaura is among the most outstanding ones. Herein, a brief historical overview of insights regarding the reaction mechanism is provided. In particular, the formation of the catalytically active species is probably the main concern, thus the preactivation is in competition with, or even assumes the role of the rate determining step (rds) of the overall reaction. Computational chemistry is key in identifying the rds and thus leading to milder conditions on an experimental level by means of predictive catalysis.

Abstract:Acyl Suzuki cross-coupling involves the coupling of an organoboron reagent with an acyl electrophile (acyl halide, anhydride, ester, amide). This review provides a timely overview of the very important advances that have recently taken place in the acylative Suzuki cross-coupling. Particular emphasis is directed toward the type of acyl electrophiles, catalyst systems and new cross-coupling partners. This review will be of value to synthetic chemists involved in this rapidly developing field of Suzuki cross-coupling as well as those interested in using acylative Suzuki cross-coupling for the synthesis of ketones as a catalytic alternative to stoichiometric nucleophilic additions or Friedel-Crafts reactions.

Abstract:The Suzuki-Miyaura coupling is among the most important C-C bond-forming reactions available due to its reliability, chemoselectivity, and diversity. Aryl halides and pseudohalides such as iodides, bromides, and triflates are traditionally used as the electrophilic coupling partner. The expansion of the reaction scope to nontraditional electrophiles is an ongoing challenge to enable an even greater number of useful products to be made from simple starting materials. Herein, we present how an NHC-based Pd catalyst can enable Suzuki-Miyaura coupling where the C(acyl)-O bond of aryl esters takes on the role of electrophile, allowing the synthesis of various ketone-containing products. This contrasts known reactions of similar esters that provide biaryls via nickel catalysis. The underlying cause of this mechanistic divergence is investigated by DFT calculations, and the robustness of esters compared to more electrophilic acylative coupling partners is analyzed.

Abstract:A cobalt-catalyzed method for the C(sp2)-C(sp3) Suzuki-Miyaura cross coupling of aryl boronic esters and alkyl bromides is described. Cobalt-ligand combinations were assayed with high-throughput experimentation, and cobalt(II) sources with trans-N,N'-dimethylcyclohexane-1,2-diamine (DMCyDA, L1) produced optimal yield and selectivity. The scope of this transformation encompassed steric and electronic diversity on the aryl boronate nucleophile as well as various levels of branching and synthetically valuable functionality on the electrophile. Radical trap experiments support the formation of electrophile-derived radicals during catalysis.

Abstract:Stereospecific Csp(3) Suzuki-Miyaura cross-coupling could transform stereochemically complex small molecule synthesis into a simple and broadly accessible process. However, most current methods are not compatible with complex building blocks that represent densely packed, multistereogenic center-containing motifs commonly found in natural products and other complex targets. Here, we report a method that enables the alpha-methyl-beta-hydroxyl motif, which is found in > 18 000 natural products as well as other Csp(3)-rich organic fragments, to be embedded within stereochemically defined secondary alkyl boronic ester building blocks that are readily cross-coupled in a stereospecific manner. The steric effect-mediated decrease in reactivity toward transmetalation and deleterious side reactions associated with the cross-coupling of beta-oxygen-containing Csp(3) boronic esters are concomitantly addressed using beta-aryloxysilyl groups as dual-purpose transmetalation-promoting groups and stable beta-oxygen surrogates. Mechanistic studies including real-time HPLC analysis show that a five-membered pinacol siloxaborolate generated in situ is then hydrolyzed to a dihydroxysiloxaborolate that acts as an activated transmetalation partner in a stereospecific process that proceeds with retention of configuration.

Abstract:Suzuki-Miyaura cross-coupling (SMC) reaction by palladium (Pd) catalysis is widely used in cross-coupling chemistry synthesis for C-C bond formation, yet the biaryl products are generally lacking halo substituents due to highly active palladium species. Suitable catalysts according to this property for the regionselective SMC synthesis of biaryl derivatives with halo substituents did not receive due and sufficient attention. Here, we report the facile synthesis of a covalent organic frameworks (COFs)supported nanoparticles Pd catalyst that can region-selectively catalyze the SMC reaction of aryl boronic acids and aryl halides, especially with halogenated aryl boronic acids. The methodology exhibits mild operating conditions, broad functional group tolerance, and environmental friendliness and can be strategically applied for cross-coupling scenarios governed by sequential transition metal catalysts to build with or without a halo-substituted moiety. The Pd/COF-SMC nanoparticles catalysts are stable and reusable and succeed in synthesizing halo-substituted biaryl products in excellent yield, which avoids the homocoupling of halogenated aryl boronic acid. The most outstanding advantage is the precise regulation of nanoparticles Pd activity with the well-designed COF ligand.

Abstract:Cross-coupling reactions are powerful synthetic tools for the formation of remarkable building blocks of many naturally occurring molecules, polymers and biologically active compounds. These reactions have brought potent transformations in chemical and pharmaceutical disciplines. In this review, we have focused on the use of cross-coupling reactions such as Suzuki, Negishi, Heck, Sonogashira and Stille in the total synthesis of some natural products of recent years (2016–2020). A short introduction of mentioned cross-coupling reactions along with highlighted aspects of natural products has been stated in separate sections. Additionally, few examples of natural products via incorporation of more than one type of cross-coupling reaction have also been added to demonstrate the importance of these reactions in organic synthesis.

Abstract:The Pd‐catalyzed Suzuki‐Miyaura (SM) cross‐coupling of organo halides with organoboron compounds is a widely used tool in organic synthesis for the construction of C‐C bonds. Alternative methods using pseudohalides, such as triflates or nonaflates, are also suitable for this transformation. These substrates offer the advantages of simple access from readily available phenols (or enolizable ketones) and possible use for chemodivergent sequential SM couplings using electronically differentiated leaving groups. This review aims to give an overview about the current strategies of Suzuki‐Miyaura cross‐couplings with triflates or nonaflates as leaving groups. In particular, it aims to give a better understanding about the development of new efficient catalyst systems over the last decade overcoming the limitations of the standard catalyst Pd(PPh3)4. The hydrolysis of triflates as a common side reaction and its prevention will be discussed, as well as the possible transformation of triflates into halides as a strategic alternative. A matrix table is provided to guide the practitioners to find suitable reaction conditions for the various combinations of aryl/enol triflate substrates and arylboronic acids with differing electronic character.
