Abstract:The organic electronic ion pump (OEIP) is an on-demand electrophoretic drug delivery device, that via electronic to ionic signal conversion enables drug delivery without additional pressure or volume changes. The fundamental component of OEIPs is their polyelectrolyte membranes which are shaped into ionic channels that conduct and deliver ionic drugs, with high spatiotemporal resolution. The patterning of these membranes is essential in OEIP devices and is typically achieved using laborious microprocessing techniques. Here, the development of an inkjet printable formulation of polyelectrolyte is reported, based on a custom anionically functionalized hyperbranched polyglycerol (i-AHPG). This polyelectrolyte ink greatly simplifies the fabrication process and is used in the production of free-standing OEIPs on flexible polyimide (PI) substrates. Both i-AHPG and the OEIP devices are characterized, exhibiting favorable iontronic characteristics of charge selectivity and the ability to transport aromatic compounds. Further, the applicability of these technologies is demonstrated by the transport and delivery of the pharmaceutical compound bupivacaine to dorsal root ganglion cells with high spatial precision and effective nerve blocking, highlighting the applicability of these technologies for biomedical scenarios.

Abstract:Unintended inhibition of the cardiac potassium channel human ether-a-go-go-related gene (hERG) is considered the main culprit in drug-induced arrhythmias known as torsades de pointes. Electrophysiology is the most reliable in vitro screening method for identifying potential cardiac hERG liabilities, but only the recent advent of planar electrode-based voltage clamp electrophysiology promises sufficient throughput to support the drug testing needs of most drug discovery programs. We have assessed the reliability of this new format of the voltage clamp technology in measuring the activity of small molecules on the hERG channel. Based on the results herein of a screening against a panel of well-characterized hERG-active and -inactive molecules, we demonstrate that planar electrode electrophysiology, utilizing the Sealchip and PatchXpress technology platform (AVIVA Biosciences Corp., San Diego, CA), is comparable to traditional electrophysiology based on glass micropipettes in its reliability and data content. The new technology will allow significantly higher throughput and more thorough testing of pharmaceutical compounds.

Abstract:Implantable devices offer an alternative to systemic delivery of drugs for the treatment of neurological disorders. A microfluidic ion pump (µFIP), capable of delivering a drug without the solvent through electrophoresis, is developed. The device is characterized in vitro by delivering γ-amino butyric acid to a target solution, and demonstrates low-voltage operation, high drug-delivery capacity, and high ON/OFF ratio. It is also demonstrated that the device is suitable for cortical delivery in vivo by manipulating the local ion concentration in an animal model and altering neural behavior. These results show that µFIPs represent a significant step forward toward the development of implantable drug-delivery systems.

Abstract:The challenge of treating neurological disorders has motivated the development of implantable devices that can deliver treatment when and where it is needed. This study presents a novel brain implant capable of electrophoretically delivering drugs and recording local neural activity on the surface of the brain. The drug delivery is made possible by the integration of a microfluidic ion pump (µFIP) into a conformable electrocorticography (ECoG) device with recording cites embedded next to the drug delivery outlets. The µFIP ECoG device can deliver a high capacity of several biologically important cationic species on demand. The therapeutic potential of the device is demonstrated by using it to deliver neurotransmitters in a rodent model while simultaneously recording local neural activity. These developments represent a significant step forward for cortical drug-delivery systems.

Abstract:The charge balance mechanism for two amino acid inhibitory neurotransmitter ions, glycine (Gly), and gamma‐aminobutyric acid (GABA) is clarified as zwitterion protonation and deprotonation near polarized electrodes. The concentration changes of glycine and GABA are realized via electrosorption in nanoporous carbon, which enables switchable capacitance in carbon‐based ionic electrolyte double‐layer transistors. Neurotransmitters carrying specific chemical information play a key role in the nervous system, which inspires ion‐based devices to mimic biological functions of nervous system, including sensing, transduction, and computing. In this work, the interaction mechanism of zwitterionic amino acids, glycine (Gly), and gamma‐aminobutyric acid (GABA) (two common inhibitory neurotransmitters) are analyzed with porous carbon electrodes. The charge balance of these zwitterionic neurotransmitters relies on zwitterion protonation in the vicinity of the negative electrodes (produced cations adsorbed by the negative electrode) and deprotonation near the positive electrode (produced anions to balance the positive charges) as demonstrated based on the theoretical dissociation analyses and local pH measurements. Additionally, electrosorption‐induced concentration changes of Gly and GABA in aqueous solutions and phosphate‐buffered saline (PBS) solutions are realized by electrosorption via nanoporous carbon electrodes. Based on this mechanistic understanding, the ionologic functions in printed ionic electrolyte double‐layer transistors are demonstrated. These neurotransmitter‐based iontronic devices hold potential for further organism‐machine interfacing and neuromorphic computing applications.

Abstract:Bioelectronic devices must be fast and sensitive to interact with the rapid, low-amplitude signals generated by neural tissue. They should also be biocompatible and soft, and should exhibit long-term stability in physiologic environments. Here, we develop an enhancement-mode, internal ion-gated organic electrochemical transistor (e-IGT) based on a reversible redox reaction and hydrated ion reservoirs within the conducting polymer channel, which enable long-term stable operation and shortened ion transit time. E-IGT transient responses depend on hole rather than ion mobility, and combine with high transconductance to result in a gain–bandwidth product that is several orders of magnitude above that of other ion-based transistors. We used these transistors to acquire a wide range of electrophysiological signals, including in vivo recording of neural action potentials, and to create soft, biocompatible, long-term implantable neural processing units for the real-time detection of epileptic discharges. E-IGTs offer a safe, reliable and high-performance building block for chronically implanted bioelectronics, with a spatiotemporal resolution at the scale of individual neurons.

Abstract:A new concept in iontophoresis is presented, where a redox mediator solution is used to control electrode reactions and sustain continuous drug delivery. It allows the solvent‐free delivery of various drugs over a theoretically unlimited period, offering the potential to improve treatment options for various diseases, including hard‐to‐treat cancers. Drug delivery into the brain and spinal cord is fundamentally limited by the blood‐brain barrier which impedes the use of the vast majority of drugs. Implants based on iontophoresis use an applied voltage to deliver charged drug molecules, allowing solvent‐free delivery directly into the site of interest and overcoming issues associated with systemic exposure to the drug. However, during continuous delivery over long periods, electrochemical reactions occur at the electrodes leading to corrosive gas formation. Here, the concept of redox flow iontophoresis is presented, where a redox mediator solution is used to control electrode reactions and sustain continuous delivery for theoretically unlimited duration. As a proof‐of‐concept, a redox flow iontophoresis‐based brain implant that can continuously deliver the cancer drug doxorubicin at stable rates exceeding 2 nmol min−1 is demonstrated. This new concept enables the continuous delivery of various potent drugs into the brain and spinal cord and therefore has the potential to improve treatment options for various diseases.

Abstract:Ion coupling has provided an additional method to modulate electric properties for solid-state materials. Here, phosphorosilicate glass (PSG)-based electrolyte gated protonic/electronic coupled indium-tin-oxide electric-double-layer (EDL) transistors are fabricated. The oxide transistor exhibits good electrical performances due to an extremely strong proton gating behavior for the electrolyte. With interfacial electrochemical doping, channel conductances of the oxide EDL transistor can be regulated to different levels, corresponding to different initial synaptic weights. Thus, activity dependent synaptic responses, such as excitatory post-synaptic current, paired-pulse facilitation and high-pass filtering, are discussed in detail. The proposed proton conductor gated oxide EDL synaptic transistors with activity dependent synaptic plasticities may act as fundamental building blocks for neuromorphic system applications.
High-resolution iontophoresis for single-synapse stimulation

Abstract:We present a technique for stimulating post-synaptic receptors with neurotransmitter locally at a single synapse and with a concentration profile that is comparable to endogenous stimulation. We modify the technique of iontophoresis to use a 0.1 μm electrode tip for local stimulation, and we combine it with fast capacitance compensation to achieve high-speed application from a high-resistance tip. Ejection of fluorescent dye from the electrode shows that transmitter can be limited to the width of a single synapse and to a time scale similar to an endogenous event. The speed and localization of transmitter is confirmed by iontophoretically stimulating single labeled synapses in cultured hippocampal neurons held under voltage clamp. The amount of transmitter ejected is linear and reproducible over a physiologically relevant range, making this technique useful for examining receptor kinetics and receptor insertion/removal. The system should be capable of delivering any charged neurotransmitter, and we show examples using glutamate and GABA. The technique is also combined with computer-controlled manipulation to study the strength and plasticity of multiple synapses in real-time.

Abstract:Despite optimal therapeutic regimen with currently available antiepileptic drugs (AEDs), approximately a third of epilepsy patients remain drug refractory. Region-specific overexpression of multidrug efflux transporters at the blood brain barrier, such as P-glycoprotein (P-gp), might contribute to multidrug resistance (MDR) by reducing target concentrations of AEDs. Therefore, development of nanomedicine that can modulate P-gp function as well as facilitate targeted AEDs delivery represents a promising strategy for epilepsy intervention. To achieve this, we sought to exploit the possibility of combination of active targeting function of tryptophan by transporter-mediated endocytosis and overcoming MDR by Pluronic block copolymers. Herein, a tryptophan derivate (TD) functionalized Pluronic P123/F127 mixed micelles encapsulating LTG (TD-PF/LTG) was developed to promote AEDs delivery to epileptogenic focus. TD-PF/LTG was about 20 nm in diameter with a spherical shape and high encapsulation efficiency. A rat epilepsy model with pilocarpine was established to evaluate the brain penetration efficiency of the LTG-incorporated polymeric micellar formulation, compared with free LTG formulations. Studies showed that TD-PF/LTG was more efficient than PF/LTG as well as free LTG in delivering the drug to the brain, especially the hippocampus. The enhanced targeted delivery could be ascribed to the increased tryptophan uptake at epileptogenic focus as well as P-gp modulation property of the nanomaterial. Taken together, TD-conjugated Pluronic micelles showed promising potential as a nanoplatform for the delivery of AEDs in refractory epilepsy. (C) 2015 Elsevier Ltd. All rights reserved.

Abstract:Intracellular protein delivery systems have great potential in the fields of therapeutics development and biomedical research. However, targeted delivery, passing through the cell membrane without damaging the cells, and escaping from endosomal entrapment of endocytosed molecular cargos are major challenges of the system. Here, we present a novel intracellular protein delivery system based on modularly engineered botulinum neurotoxin type A (BoNT/A). LH N A domain, consisting of light chain and endosomal escape machinery of BoNT/A, was genetically fused with SpyCatcher (SC) and EGFR targeting affibody (EGFRAfb) to create SC-LH N A-EGFRAfb, a target-specific and protein cargo-switchable BoNT/A-based intracellular protein delivery platform. SC-LH N A-EGFRAfb was purely purified in large quantities, efficiently ligated with multiple ST-fused protein cargos individually, generating a variety of protein cargo-containing intracellular delivery complexes, and successfully delivered ligated protein cargos into the cytosol of target cells via receptor-mediated endocytosis, followed by endosomal escape and subsequent cytosolic delivery. SC-LH N A-EGFRAfb enhanced intracellular delivery efficiency of protein toxin, gelonin, by approximately 100-fold, highlighting the crucial roles of EGFRAfb and LH N A domain as a targeting ligand and an endosomal escape machinery, respectively, in the delivery process. The BoNT-based plug-and-deliverable intracellular protein delivery system has the potential to expand its applications in protein therapeutics and manipulating cellular processes.

Abstract:This paper reports the design of a proof-of-concept drug delivery device that is actuated using the bubbles formed during electrolysis. The device uses a platinum (Pt) coated nickel (Ni) metal foam and a solid drug in reservoir (SDR) approach to improve the device's performance. This electrochemically-driven pump has many features that are unlike conventional drug delivery devices: it is capable of pumping periodically and being refilled automatically; it features drug release control; and it enables targeted delivery. Pt-coated metal foam is used as a catalytic reforming element, which reduces the period of each delivery cycle. Two methods were used for fabricating the Pt-coated metal: sputtering and electroplating. Of these two methods, the sputtered Pt-coated metal foam has a higher pumping rate; it also has a comparable recombination rate when compared to the electroplated Pt-coated metal foam. The only drawback of this catalytic reformer is that it consumes nickel scaffold. Considering long-term applications, the electroplated Pt metal foam was selected for drug delivery, where a controlled drug release rate of 2.2 μg ± 0.3 μg per actuation pulse was achieved using 4 mW of power.

Abstract:AbstractIontronics is a newly emerging interdisciplinary concept that bridges electronics and ionics. It provides new opportunities for biomimic information processing. Iontronic devices can act as building blocks for neuromorphic platforms. Here, a proof‐of‐principle Hodgkin–Huxley artificial synaptic membrane is proposed for the first time based on inorganic proton conductor. Phosphosilicate glass‐based proton conductor electrolyte demonstrates unique short‐term volatile charging behaviors, indicating potential short‐term synaptic plasticity applications. By using protonic/electronic hybrid oxide transistor configuration, dynamic synaptic membrane potential responses are triggered with gate current spikes. Typical resting potential, excitatory/inhibitory postsynaptic potential behaviors, and membrane depolarization/activation behaviors are mimicked on the proposed Hodgkin–Huxley artificial synaptic membrane. Moreover, proton‐related electrostatic coupling enables the device to possess short‐term synaptic plasticities with low power consumption. The proposed Hodgkin–Huxley artificial synaptic membrane provides a new prototype for neuromorphic system applications.

Abstract:Conjugated polymer-based organic electrochemical transistors (OECTs) are being studied for applications ranging from biochemical sensing to neural interfaces. While new conjugated polymers are being developed that can interface digital electronics with the aqueous chemistry of life, the vast majority of high-performance, high-mobility organic transistor materials developed over the past decades are extremely poor at taking up biologically-relevant ions. Here we incorporate an ion exchange gel into an OECT, demonstrating that this structure is capable of taking up biologically-relevant ions from solution and injecting larger, more hydrophobic ions into the underlying polymer semiconductor active layer in multiple hydrophobic conjugated polymers. Using poly[2,5-bis(3-tetradecylthiophen-2-yl) thieno[3,2-b]thiophene] (PBTTT) as a model semiconductor active layer and a blend of the ionic liquid 1-butyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide (BMIM TFSI) and poly(vinylidene fluoride-co-hexafluoropropylene) (PVDF-HFP) as the ion exchange gel, we demonstrate more than a four order of magnitude improvement in OECT device transconductance and a one hundred-fold increase in ion injection kinetics. We demonstrate the ability of the ion exchange gel OECT to record biological signals by measuring the action potentials of a Venus flytrap plant. These results show the possibility of using interface engineering to open up a wider palette of organic semiconductor materials as OECTs that can be gated by aqueous solutions.

Abstract:The human brain is often likened to an incredibly complex and intricate computer, rather than electrical devices, consisting of billions of neuronal cells connected by synapses. Different brain circuits are responsible for coordinating and performing specific functions. The reward pathway of the synaptic plasticity in the brain is strongly related to the features of both drug addiction and relief. In the current study, a synaptic device based on layered hafnium disulfide (HfS2) is developed for the first time, to emulate the behavioral mechanisms of drug dosage modulation for neuroplasticity. A strong gate‐dependent persistent photocurrent is observed, arising from the modulation of substrate‐trapping events. By controlling the polarity of gate voltage, the basic functions of biological synapses are realized under a range of light spiking conditions. Furthermore, under the control of detrapping/trapping events at the HfS2/SiO2 interface, positive/negative correlations of the An/A1 index, which significantly reflected the weight change of synaptic plasticity, are realized under the same stimulation conditions for the emulation of the drug‐related addition/relief behaviors in the brain. The findings provide a new advance for mimicking human brain plasticity.

Abstract:Background: P-glycoprotein (P-gp) mediated drug efflux across the blood-brain barrier (BBB) is an important mechanism underlying poor brain penetration of certain antiepileptic drugs (AEDs). Nanomaterials, as drug carriers, can overcome P-gp activity and improve the targeted delivery of AEDs. However, their applications in the delivery of AEDs have not been adequately investigated. The objective of this study was to develop a nano-scale delivery system to improve the solubility and brain penetration of the antiepileptic drug lamotrigine (LTG).Methods: LTG-loaded Pluronic (R) P123 (P123) polymeric micelles (P123/LTG) were prepared by thin-film hydration, and brain penetration capability of the nanocarrier was evaluated.Results: The mean encapsulating efficiency for the optimized formulation was 98.07%; drug-loading was 5.63%, and particle size was 18.73 nm. The solubility of LTG in P123/LTG can increase to 2.17 mg/ml, making it available as a solution. The in vitro release of LTG from P123/LTG presented a sustained-release property. Compared with free LTG, the LTG-incorporated micelles accumulated more in the brain at 0.5, 1, and 4 hours after intravenous administration in rats. Pretreatment with systemic verapamil increased the rapid brain penetration of free LTG but not P123/LTG. Incorporating another P-gp substrate (Rhodamine 123) into P123 micelles also showed higher efficiency in penetrating the BBB in vitro and in vivo.Conclusion: These results indicated that P123 micelles have the potential to overcome the activity of P-gp expressed on the BBB and therefore show potential for the targeted delivery of AEDs. Future studies are necessary to further evaluate the appropriateness of the nanocarrier to enhance the efficacy of AEDs.

Abstract:Objective. The controlled delivery of potassium is an interesting neuromodulation modality, being potassium ions involved in shaping neuron excitability, synaptic transmission, network synchronization, and playing a key role in pathological conditions like epilepsy and spreading depression. Despite many successful examples of pre-clinical devices able to influence the extracellular potassium concentration, computational frameworks capturing the corresponding impact on neuronal activity are still missing. Approach. We present a finite-element model describing a PEDOT:PSS-coated microelectrode (herein, simply ionic actuator) able to release potassium and thus modulate the activity of a cortical neuron in an in-vitro-like setting. The dynamics of ions in the ionic actuator, the neural membrane, and the cellular fluids are solved self-consistently. Main results. We showcase the capability of the model to describe on a physical basis the modulation of the intrinsic excitability of the cell and the synaptic transmission following the electro-ionic stimulation produced by the actuator. We consider three case studies for the ionic actuator with different levels of selectivity to potassium: ideal selectivity, no selectivity, and selectivity achieved by embedding ionophores in the polymer backbone. Significance. This work is the first step toward a comprehensive computational framework aimed to investigate novel neuromodulation devices targeting specific ionic species, as well as to optimize their design and performance, in terms of the induced modulation of neural activity.

Abstract:An organic electrochemical transistor (OECT) is one of the promising devices for bioelectronics due to its high transconductance, encompassing low operation voltage, and good compatibility with aqueous conditions. Despite these advantages, the challenge of balancing ion penetration and electron transport remains a significant issue in OECTs. Herein, we present an amphiphilic interface modification strategy to successfully prepare OECTs in aqueous conditions based on a high-mobility hydrophobic polypyrrole derivative. An amphiphilic interface mixed with an amphiphilic polymer and the active layer markedly promotes ion penetration and results in a significant improvement in performance, with the switch time reduced from several seconds to nearly 100 ms and the transconductance increased by an order of magnitude. The high-performance OECTs fabricated by this method show promising applications in high-performance neuromorphic devices and ECG recording in advancing the field of electrochemical transistors.
