Abstract:Abstract Objectives Episodic memory is age-sensitive but can be strengthened by targeted training interventions. The method of loci (MoL) is a classic mnemonic which if successfully implemented greatly improves memory performance. We developed and investigated the effects of a MoL training program implemented in a smart phone application (app) with the aim of studying usage of the application, training effect and its modifiability by age, predictors for MoL proficiency, transfer effects to a face-name memory task, and perceived benefit in everyday memory. Method A total of 359 adults participated. Instruction and training of the MoL, transfer test (face-name paired associates cued recall task), and surveys were performed in an in-house developed app. Results The app interested people across the adult life span. Older adults practiced the most, whereas younger and young-old participants showed the highest level of MoL proficiency. Level of proficiency was modulated by amount of practice, but in the oldest participants this effect was less pronounced. Greater self-rated health was associated with higher level of proficiency. No transfer effect was observed. Among those who answered the survey, about half expressed that MoL training had benefitted memory in their everyday life. Discussion App-based memory training in the MoL can be delivered successfully via an app across the adult life span. Level of performance reached in training is variable but generally high, and mainly influenced by amount of training and age of the participants. Our data suggest plasticity across the life span, but to a lesser degree for adults between 70 and 90 years.

Abstract:The “loci method” is a popular mnemonic device that involves visualising and recalling items at specific points along a familiar route. The loci method has been used for thousands of years, and by many successful memory athletes; yet there have been relatively few educational and clinical applications, possibly owing to empirical uncertainty. The current meta-analysis of 13 randomised controlled trials (RCTs) mostly based in university settings demonstrated the effectiveness of the loci method as a mnemonic device, with a medium effect size ( g = 0.65, 95% confidence interval [CI] = [0.45, 0.85]; I 2 = 45.5%). The effect size remained at similar levels in further analyses adjusting for publication bias, the impact of removing each study, setting, control conditions, outliers, and number of loci method sessions. High risk of experimental bias was indicated, however, as the vast majority of studies did not report procedures to minimise biases relating to random sequence generation and allocation concealment. Overall, this meta-analysis of predominantly university-based RCTs has provided good initial support for the loci method as a mnemonic device and this may encourage future investigations and applications, particularly in educational settings, where it has the potential to improve recall of information relevant to academic success.

Abstract:It is said that our species use mnemonics - that "magic of memorization" - to engrave an enormous amount of information in the brain. Yet, it is unclear how mnemonics affect memory and what the neural underpinnings are. In this electroencephalography study, we examined the hypotheses whether mnemonic training improved processing-efficiency and/or altered encoding-pattern to support memory enhancement. By 22-day training of a digit-image mnemonic (a custom memory technique used by world-class mnemonists), a group of children showed increased short-term memory after training, but with limited gain generalization. This training resulted in regular odd-even neural patterns (i.e., enhanced P200 and theta power during the encoding of digits at even- versus odd- positions in a sequence). Critically, the P200 and theta power effects predicted the training-induced memory improvement. These findings provide evidence of how mnemonics alter encoding pattern, as reflected in functional brain organization, to support memory enhancement.
Increased recognition memory precision with decreased neural discrimination

Abstract:Mnemonic discrimination (MD), the ability to distinguish between similar experiences in memory, is essential for memory precision. We hypothesized that training mnemonic discrimination should improve memory precision by increasing the neural activity differences between a new experience and a similar familiar experience (repeat). Participants performed a 2-week, web-based training program. For group 1, memory similarity was adapted to performance. Group 2 trained non-adaptively and group 3 served as active control. Adaptivity improved training gain and led to transfer to other memory tasks. Surprisingly, training reduced neural activity differences between new (lure) and similar familiar (repeat) experiences. Thus, instead of enhancing neural activity to lures and decreasing neural activity to repeats, training led to increased activity to repeats. In several brain regions this pattern was associated with improved MD. These findings highlight an unexpected neural mechanism underpinning improved memory precision in recognition memory.

Abstract:The ability to form durable memory is critical for human survival and development, but its underlying cognitive and neural mechanisms have not been well understood. In particular, existing studies have not clearly dissociated the neural processes supporting short- and long-duration memories. The present study addressed this issue with functional MRI and a modified subsequent memory paradigm. Participants were asked to make semantic judgment on a list of 320 words in the scanner. Half of the words were tested after a short delay (i.e., 1day, T1) and again after a long delay (i.e., 1week, T12), whereas the other half were tested only once after the long delay (T2). Materials forgotten during T1 were categorized as forgotten trials, and those remembered during T2 were categorized as long-duration trials. In contrast, trials remembered during T1 but not during T12 were categorized as short-duration trials. We found that compared to forgotten trials, short-duration trials showed decreased activation in the posterior cingulate cortex (PCC) and precuneus, which is consistent with many previous observations. Importantly, long-duration trials showed stronger activity in the left inferior frontal gyrus (LIFG) but less deactivation in the PCC relative to short-duration trials. Psychophysiological interactions (PPI) analysis revealed stronger functional connectivity between LIFG and PCC for long-duration trials than for forgotten trials. Our results suggest that strong PCC activity, in combination with strong LIFG activity, supports long-lasting memory.

Abstract:Memory brain states may influence how we experience an event. Memory encoding and retrieval constitute neurally dissociable brain states that individuals can selectively engage based on top-down goals. To the extent that memory states linger in time – as suggested by prior behavioral work – memory states may influence not only the current experience, but also subsequent stimuli and judgments. Thus lingering memory states may have broad influences on cognition, yet this account has not been directly tested utilizing neural measures of memory states. Here we address this gap by testing the hypothesis that memory brain states are modulated by memory judgments, and that these brain states persist for several hundred milliseconds. We recorded scalp electroencephalography (EEG) while participants completed a recognition memory task. We used an independently validated multivariate mnemonic state classifier to assess memory state engagement. We replicate prior behavioral findings; however, our neural findings run counter to the predictions made on the basis of the behavioral data. Surprisingly, we find that prior responses modulate current memory state engagement on the basis of response congruency. That is, we find strong engagement of the retrieval state on incongruent trials – when a target is preceded by a correct rejection of a lure and when a lure is preceded by successful recognition of a target. These findings indicate that cortical brain states are influenced by prior judgments and suggest that a non-mnemonic, internal attention state may be recruited to in the face of changing demands in a dynamic environment.

Abstract:What we remember reflects both what we encounter, such as the intrinsic memorability of a stimulus, and our internal attentional state when we encounter that stimulus. Our memories are better for memorable images and images encountered in an engaged attentional state. Here, in an effort to modulate long-term memory performance, we manipulated these factors in combination by selecting the memorability of presented images contingent on individuals' natural fluctuations in sustained attention. Can image memorability and attentional state be strategically combined to improve memory? Are memorable images still well remembered during lapses in sustained attention, and conversely, can attentive states rescue memory performance for forgettable images? We designed a procedure to monitor participants' sustained attention dynamics on the fly via their response time fluctuations during a continuous performance task with trial-unique scene images. When high- or low-attentional states were detected, our algorithm triggered the presentation of high- or low-memorability images. Afterwards, participants completed a surprise recognition memory test for the attention-triggered images. Results demonstrated that memory performance for memorable items is not only resistant to lapses in sustained attention but also that memory cannot be further improved by encoding memorable items in engaged attentional states. On the other hand, memory performance for low-memorability images can be rescued by attentive encoding states. In sum, we show that both memorability and sustained attention can be leveraged in real time to maximize memory performance. This approach suggests that adaptive cognitive interfaces can tailor what information appears when to best support overall memory.

Abstract:Reactivation renders consolidated memory labile again, and the ensuing temporary reconsolidation process is highly susceptible to mnemonic modification. Here, we show that memories in such an unstable state could be influenced by sheer behavioral means, bypassing the need for pharmacological intervention. Across several experiments using a "face-location association" paradigm in which participants experienced a "Learning - New learning - Final-test" procedure, we demonstrate that reactivated memory traces were hampered when the new learning was strategically administered at between 0-min and 20-min delay. Using fMRI, we further advance our theoretical understanding that this lability can be mechanistically explained by the differential activation in the hippocampal-amygdala memory system implicated by the post-activation new-learning whereas the mnemonic intrusion caused by newly learned memories is efficaciously reconciled by the left inferior frontal gyrus.

Abstract:Background: Mnemonic strategy training (MST) has been shown to improve cognitive performance in amnestic mild cognitive impairment (a-MCI), however, several questions remain unresolved. The goal of the present study was to replicate earlier pilot study findings using a randomized controlled design and to evaluate transfer effects and changes in brain activation. Methods: Thirty patients with a-MCI were randomized into MST or education program. At baseline, participants completed clinical and neuropsychological assessments as well as structural and functional magnetic resonance imaging (fMRI). Interventions were administered individually and comprised four sessions, over 2 weeks. MST taught patients to use a three-step process to learn and recall face-name associations. Post-treatment assessment included fMRI, a separate face-name association task, neuropsychological tests, and measures of metamemory. Behavioral (i.e., non-fMRI) measures were repeated after one and 3-months. Results: Participants in the MST condition showed greater improvement on measures of face-name memory, and increased associative strategy use; effects that were accompanied by increased fMRI activation in the left anterior temporal lobe. While all participants reported greater contentment with their everyday memory following intervention, only the MST group reported significant improvements in their memory abilities. There was no clear indication of far-transfer effects to other neuropsychological tests. Conclusion: Results demonstrate that patients with a-MCI not only show stimulus specific benefits of MST, but that they appear capable of transferring training to at least some other cognitive tasks. MST also facilitated the use of brain regions that are involved in face processing, episodic and semantic memory, and social cognition, which are consonant with the cognitive processes engaged by training.

Abstract:Schematic prior knowledge can provide a powerful scaffold for episodic memories, yet the neural mechanisms underlying this scaffolding process are still poorly understood. A crucial step of the scaffolding process is the way in which details of a new episode are connected to an existing schema, forming a robust memory representation that can be easily accessed in the future. A unique testbed for studying this binding process is a mnemonic technique called the Method of Loci (MoL), in which people meaningfully connect items to be remembered with a well-learned list of imagined loci. We collected fMRI data from participants in 3 longitudinal sessions while they were enrolled in a month-long MoL training course, all of whom showed dramatic improvements in their ability to remember lists of 20 or 40 words. We obtained neural patterns when the loci and objects are presented by themselves, when they are combined into an integrated representation at encoding, and when the integrated representation was subsequently retrieved, as well as verbal descriptions from the participants about the way they associated each item to each locus. We found that in default mode network regions, including medial prefrontal cortex (mPFC), the combined representations of loci and items are highly conjunctive: the unified locus-item representation was substantially different from a linear combination of the isolated locus and item representation, reflecting the addition of new integrative details specific to each combined pair. The conjunctive component of the representation reflected the particular creative details generated by individual participants and increased over time as participants gained expertise in MoL. Our findings reveal a critical role for the default mode network in creating meaningful connections between new information and well-learned schematic contexts.

Abstract:Recent experiments in rats and humans have indicated that the effects of non-invasive electrical stimulation are primarily due to transcutaneous stimulation of peripheral nerves, specifically the greater occipital nerve. This stimulation pathway activates communication gateways from the periphery to the brain, impacting memory consolidation. In this invited commentary, I delve into and offer additional insights concerning the enhancement of episodic memory through transcutaneous electrical stimulation of the greater occipital nerve, building upon the findings published by my laboratory in both Science Advances and Elife. Our research on non-invasive transcutaneous electrical stimulation of the greater occipital nerve (NITESGON) has shown to enhance episodic memory consolidation and promote communication between the locus coeruleus (LC) pathway and the hippocampus based on resting connectivity functional MRI. The LC, primarily responsible for releasing noradrenaline and dopamine, plays a crucial role in post-encoding memory stabilization. This suggests that NITESGON can improve memory but does not affect immediate learning. The concept of behavioural tagging, where weak memories can be stabilized through strong or novel events, and how NITESGON activates a memory consolidation through this mechanism are discussed. The role of NITESGON in enhancing memory stabilization is highlighted, providing a non-pharmaceutical solution with minimal side effects. The potential application of NITESGON in neurological conditions, including Alzheimer's disease, attention deficit hyperactivity disorder and post-traumatic stress disorder, is also discussed, emphasizing its promising therapeutic prospects.

Abstract:Persistent, memorandum-specific neuronal spiking activity has long been hypothesized to underlie working memory 1,2 . However, emerging evidence suggests a potential role for 'activity-silent' synaptic mechanisms 3,4,5 . This issue remains controversial because evidence for either view has largely relied either on datasets that fail to capture single-trial population dynamics or on indirect measures of neuronal spiking. We addressed this controversy by examining the dynamics of mnemonic information on single trials obtained from large, local populations of lateral prefrontal neurons recorded simultaneously in monkeys performing a working memory task. Here we show that mnemonic information does not persist in the spiking activity of neuronal populations during memory delays, but instead alternates between coordinated 'On' and 'Off' states. At the level of single neurons, Off states are driven by both a loss of selectivity for memoranda and a return of firing rates to spontaneous levels. Further exploiting the large-scale recordings used here, we show that mnemonic information is available in the patterns of functional connections among neuronal ensembles during Off states. Our results suggest that intermittent periods of memorandum-specific spiking coexist with synaptic mechanisms to support working memory.

Working memory processing boosts the neural representation of long-term memories

Abstract:Understanding the interaction between short-term and long-term memory systems is essential for advancing our knowledge of human memory. This study investigates whether working memory processes, specifically attentional prioritization (Experiments 1 and 2) and testing (Experiment 2), can enhance the neural activity associated with recently encoded long-term memories. A total of 86 participants completed a novel three-phase memory task that integrated a traditional long-term memory learning task with a working memory paradigm. In the first phase, participants encoded object-location associations. During the second phase, these associations were reintroduced in a working memory task that manipulated attentional prioritization; participants were required to report the location of the cued object. In the final phase, participants recalled the locations associated with each object. By analyzing both behavioral performance and electroencephalogram (EEG) data collected during this retrieval phase, we found that attentional prioritization in working memory significantly improved long-term memory retrieval, a finding supported by corresponding neural evidence. Additionally, Experiment 2 demonstrated that both prioritization and testing in working memory jointly enhance the neural representation of long-term memories. These findings indicate that working memory processes can dynamically alter the neural patterns underlying long-term memory representations, revealing a more integrated role for working memory in long-term memory consolidation.

Abstract:We investigated memory encoding and retrieval during a quasi-naturalistic spatial-episodic memory task in which subjects delivered items to landmarks in a desktop virtual environment and later recalled the delivered items. Transition probabilities and latencies revealed the spatial and temporal organization of memory. As subjects gained experience with the town, their improved spatial knowledge led to more efficient navigation and increased spatial organization during recall. Subjects who exhibited stronger spatial organization exhibited weaker temporal organization. Scalp-recorded electroencephalographic (EEG) signals revealed spectral correlates of successful encoding and retrieval. Increased theta power ( ) and decreased alpha/beta power ( ) accompanied successful encoding, with the addition of increased gamma ( ) accompanying successful retrieval. Logistic-regression classifiers trained on spectral features reliably predicted mnemonic success in held-out sessions. Univariate and multivariate EEG analyses revealed a similar spectral of successful memory. These findings extend behavioral and neural signatures of successful encoding and retrieval to a naturalistic task in which learning occurs within a spatiotemporal context.

Abstract:Memory is a way for people to acquire information through personal experiences and from media such as books, TV, and the internet. Relying on memory and the ability to gain new ones, the human brain is capable of adapting to survive in the ever-changing environment and can create marvels and build objects out of nature. Therefore, it is memory that creates modern civilization. Memory consists of synapses and neural ensembles in the brain which integrate multisensory information both spatially and temporally. Thus, memory bridges the link between cognitive domain information and physical domain neural connections, making them easier to track and study. Memory is the foundation of the brain's cognitive functions. Consequently, research into how the brain stores and retrieves memories reveals not only how the brain works to generate intelligence, but also suggests possible avenues for creating efficient machine learning algorithms that have evolved over millions of years. Recent breakthroughs in biotechnology have led to discoveries about the mechanisms underlying memory formation and retrieval, providing insight into neural memories at the cellular and synaptic levels as well as understanding neural coding for circuitry integration and operation. Particularly, the discovery of cellular memory engrams in the neocortex has revealed a highly parallel arrangement of memory circuits, which are located in layer ll of widely distributed cortical areas, including sensory cortices, primary visual cortices, and retrosplenial cortices. In this review, we summarize the most recent findings on memory engrams in the hippocampus and neocortex. We highlight the correlation between neural oscillations and cortical engram activities during memory formation. We also explore the evidence that long-range synchrony of theta-coupled gamma oscillations plays a key role in modulating cortical engram activity to create a brain-wide memory network. We explain how memory engrams in the neocortex are integrated and organized by indexing signals from the hippocampus, allowing memories to be registered allocated and retrieved in the brain. Lastly, we shed light on the molecular mechanisms especially epigenetic regulators involved in the physical regulation of memory persistency and flexibility. Deficiencies in the epigenetic regulation of these engrams may lead to memory deficits in various brain diseases. Additionally, the laminar-specific distribution of memory engrams in local cortical columns and the global organization of memory traces in distinct functional cortical regions may inspire the development of novel computing algorithms in machine learning to achieve brain-level intelligence. Taken together, the advances in methodology are paving the way for us to directly investigate the neural mechanisms of memory storage and retrieval in mammalian brain, giving insight into human cognition and inspiring new approaches in machine learning.

Abstract:The ability to discriminate between stimuli relies on a chain of neural operations associated with perception, memory and decision-making. Accumulating studies show learning-dependent plasticity in perception or decision-making, yet whether perceptual learning modifies mnemonic processing remains unclear. Here, we trained human participants of both sexes in an orientation discrimination task, while using functional magnetic resonance imaging (fMRI) and transcranial magnetic stimulation (TMS) to separately examine training-induced changes in working memory (WM) representation. fMRI decoding revealed orientation-specific neural patterns during the delay period in primary visual cortex (V1) before, but not after, training, whereas neurodisruption of V1 during the delay period led to behavioral deficits in both phases. In contrast, both fMRI decoding and disruptive effect of TMS showed that intraparietal sulcus (IPS) represented WM content after, but not before, training. These results suggest that training does not affect the necessity of sensory area in representing WM information, consistent with the sensory recruitment hypothesis in WM, but likely alters the coding format of the stored stimulus in this region. On the other hand, training can render WM content to be maintained in higher-order parietal areas, complementing sensory area to support more robust maintenance of information. SIGNIFICANCE STATEMENT There has been accumulating progresses regarding experience-dependent plasticity in perception or decision-making, yet how perceptual experience moulds mnemonic processing of visual information remains less explored. Here, we provide novel findings that learning-dependent improvement of discriminability accompanies altered WM representation at different cortical levels. Critically, we suggest a role of training in modulating cortical locus of WM representation, providing a plausible explanation to reconcile the discrepant findings between human and animal studies regarding the recruitment of sensory or higher-order areas in WM.

Abstract:Mimicking memory processes, including encoding, storing, and retrieving information, is critical for neuromorphic computing and artificial intelligence. Synaptic behavior simulations through electronic, magnetic, or photonic devices based on metal oxides, 2D materials, molecular complex and phase change materials, represent important strategies for performing computational tasks with enhanced power efficiency. Here, a special class of memristive materials based on persistent luminescent memitters (termed as a portmanteau of "memory" and "emitter") with optical characteristics closely resembling those of biological synapses is reported. The memory process and synaptic plasticity can be successfully emulated using such memitters under precisely controlled excitation frequency, wavelength, pulse number, and power density. The experimental and theoretical data suggest that electron-coupled trap nucleation and propagation through clustering in persistent luminescent memitters can explain experience-dependent plasticity. The use of persistent luminescent memitters for multichannel image memorization that allows direct visualization of subtle changes in luminescence intensity and realization of short-term and long-term memory is also demonstrated. These findings may promote the discovery of new functional materials as artificial synapses and enhance the understanding of memory mechanisms.

Abstract:The long-term fate of a memory is not exclusively determined by the events occurring at the moment of encoding. Research at the cellular, circuit, and behavioral levels is beginning to reveal how neurochemical activations in the moments surrounding an event can retroactively and proactively rescue weak memory for seemingly mundane experiences. We review emerging evidence showing enhancement of weakly formed memories encoded minutes to hours before or after a related motivationally relevant experience. We discuss proposed neurobiological mechanisms for strengthening weak memories formed in temporal proximity to a strong event, and how this knowledge could be leveraged to improve memory for information that is prone to forgetting.

Abstract:Spectral features of human electroencephalographic (EEG) recordings during learning predict subsequent recall variability. New method. Capitalizing on these fluctuating neural features, we develop a non-invasive closed-loop (NICL) system for real-time optimization of human learning. Participants play a virtual navigation and memory game; recording multi-session data across days allowed us to build participant-specific classification models of recall success. In subsequent closed-loop sessions, our platform manipulated the timing of memory encoding, selectively presenting items during periods of predicted good or poor memory function based on EEG features decoded in real time. We observed greater memory modulation (difference between recall rates when presenting items during predicted good vs. poor learning periods) for participants with higher out-of-sample classification accuracy. This study demonstrates greater-than-chance memory decoding from EEG recordings in a naturalistic virtual navigation task with greater real-world validity than basic word-list recall paradigms. Here we modulate memory by timing stimulus presentation based on noninvasive scalp EEG recordings, whereas prior closed-loop studies for memory improvement involved intracranial recordings and direct electrical stimulation. Other noninvasive studies have investigated the use of neurofeedback or remedial study for memory improvement. These findings present a proof-of-concept for using non-invasive closed-loop technology to optimize human learning and memory through principled stimulus timing, but only in those participants for whom classifiers reliably predict out-of-sample memory function.
