Abstract:The production of nanoparticles enhances the bioactivity of biological molecules for drug delivery to diseased sites. This study explains how silver nanoparticle (AgNP) coating enhanced the protection effects of vanillic acid in male diabetic rats with streptozotocin- (STZ-) induced diabetes. Twenty-four rats were divided into four groups ( n=6 ) for this investigation. The first group (G1) is untreated, whereas diabetes was induced in the other three groups through STZ injection. Diabetic rats that were not getting therapy were included in the second group (G2, STZ-positive), whereas the other diabetic rats were divided into the third group (G3, vanillic acid-treated) and the fourth group (G4, vanillic acid-coated AgNPs treated). The treatment lasted four weeks. In G2, the induction of diabetes significantly (at P=0.05 ) increased in serum glucose, glycated proteins, renal indices, interleukin-6 (IL-6), K+, immunoglobulins, and lipid peroxidation, while decreased Ca++, Na+, and other antioxidants in the kidney tissue homogenate. In addition, pathological altered signs were present in the pancreas and kidneys of diabetic rats. The renal and pancreatic tissues were effectively enhanced by vanillic acid or vanillic acid-coated AgNPs, bringing them very close to their prediabetic conditions. Vanillic acid-coated AgNPs offered a stronger defense against STZ-induced diabetes and lessened the effects of hyperglycemia compared to ordinary vanillic acid. Additionally, using vanillic acid coated with silver nanoparticles greatly increased the antioxidant and antidiabetic activity and reduced inflammation when compared to using vanillic acid alone.

Abstract:A schematic overview of the possible mechanisms involved in the neuroprotective effect of vanillic acid against diabetes‐associated cognitive decline in rats. Hippocampal synaptic dysfunction, oxidative stress, neuroinflammation, and neuronal loss play critical roles in the pathophysiology of diabetes‐associated cognitive decline (DACD). The study aimed to investigate the effects of vanillic acid (VA), a phenolic compound, against DACD and explore the potential underlying mechanisms. Following confirmation of diabetes, rats were treated with VA (50 mg/kg/day; P.O.) or insulin (6 IU/rat/day; S.C.) for 8 consecutive weeks. The cognitive performance of the rats was evaluated using passive‐avoidance and water‐maze tasks. Long‐term potentiation (LTP) was induced at hippocampal dentate gyrus (DG) synapses in response to high‐frequency stimulation (HFS) applied to the perforant pathway (PP) to evaluate synaptic plasticity. Oxidative stress factors, inflammatory markers, and histological changes were evaluated in the rat hippocampus. This study showed that streptozotocin (STZ)‐induced diabetes caused cognitive decline that was associated with inhibition of LTP induction, suppression of enzymatic antioxidant activities, enhanced lipid peroxidation, elevated levels of inflammatory proteins, and neuronal loss. Interestingly, chronic treatment with VA alleviated blood glucose levels, improved cognitive decline, ameliorated LTP impairment, modulated oxidative‐antioxidative status, inhibited inflammatory response, and prevented neuronal loss in diabetic rats at a level comparable to insulin therapy. The results suggest that the antihyperglycemic, antioxidative, anti‐inflammatory, and neuroplastic properties of VA may be the mechanisms behind its neuroprotective effect against DACD.

Renal Protective Effect of Boeravinone B against Diabetic Nephropathy Rats via Inhibition of The Inflammatory and JAK2/STAT3 Signalling Pathway

Abstract:Objective: Chronic inflammation is a common feature in diabetes, especially when blood sugar levels are poorly controlled. This chronic low-grade inflammation can affect various organs, including the kidneys. Podocyte damage play a key role in the development of diabetic nephropathy (DN). The aim of the study was to evaluate the nephroprotective effect of Boeravinone B (BB) against streptozotocin (STZ) induced DN in rats and explore the underlying mechanism. Materials and methods: In this experimental study, the rats received intraperitoneal injections of STZ (60 mg/kg) to induce DN. Various doses of BB (2.5, 5, and 7.5 mg/kg) were administered orally. Glucose levels, body weights, and organ weights (hepatic and renal) were assessed. Renal, histomorphological, antioxidant, hepatic, and cytokine levels were determined, as were the mRNA expression levels of JAK2 and STAT3. At end of the experimental study, the rats were sacrificed and their renal tissues were removed for histopathological assessment. Results: BB treatment decreased glucose levels and increased body weights. This treatment suppressed hepatic weights, increased renal tissue weights, and also decreased renal parameters like uric acid, urea, bilirubin, creatinine (Cr) and, albumin. There was a decrease (P<0.001) in histomorphological parameters such as kidney hypertrophy index (KHI), mean glomerular volume (MGV), foot process fusion ratio (FPFR), and glomerular basement membrane thickness (GBMT) after treatment with BB. In addition, this treatment improved the levels of renal podocin, renal CD2- associated protein (CD2AP) and suppressed hepatic parameter levels. BB treatment (P<0.001) altered antioxidant parameters and cytokine levels, and suppressed mRNA expressions of JAK2, STAT3, RAGE, KIM-1, NAGL, and S100A8. Conclusion: Administration of BB showed renal protective effects against STZ-induced DN in rats via the reduction of oxidative stress and inflammatory reactions.

Abstract:Vanadium compounds have been well recognized for hypoglycemic effects, but questions remain on gastrointestinal disturbance and possible tissue vanadium accumulation thus slowing the acceptance of vanadium compounds as diabetic therapeutic agents. Our intestinal permeability and toxicity studies of vanadium compounds have suggested that the co-administration of vanadate with Salvia miltiorrhiza Bunge decoction could benefit the therapeutic use of hypoglycemic vanadium compounds. In the present paper, we tested the hypoglycemic effects of vanadate ingested in an aqueous extract of S. Bunge using a streptozocin (STZ)-induced diabetic rat model. Oral administration of vanadate in S. Bunge herbal decoction produced a stable (free of hypoglycemic shock) and long-lasting (∼70day) control of blood glucose status. Effective protection of animal organs from hyperglycemic damage was also observed. As expected, the herbal extract significantly alleviated vanadium toxicity, i.e. GI stress and metal accumulation. In addition, the result suggesting that vanadium-induced amelioration of the diabetic state appears to be secondary to the preservation of a functional portion of the pancreatic β-cells which initially survived STZ-toxicity. These studies provide new insight into the therapeutic treatment of diabetics with vanadium compounds.
Effects of Nicotinamide on Prevention and Treatment of Streptozotocin-Induced Diabetes Mellitus in Rats.

Abstract:To study the effect of nicotinamide (NAA) on prevention and treatment of STZ-induced diabetes mellitus in rats.30 Sprague-Dawley rats were randomly divided inot three groups: 20 rats were given NAA 1 g/kg/day intramuscularly (10 rats) or orally (10 rats) as experimental groups and the other 10 rats by normal saline orally as control group. Three days later, all 30 rats received streptozotocin (STZ) 55 mg/kg by intraperitioneal injection and took NAA or normal saline continuously as described above for 14 days. During the study, fasting blood glucose (FBG) was measured before and 72 hours after STZ injection and every 2-3 days later on with Glucometer, 24 hours urinary glucose excretion (UGE) was determined once every 4 days with glucose oxidase, body weight was determined every week, the volume of 24-hour water intake and urine excretion were determined twice and once a week respectively.Two weeks after STZ injection, FBG and 24 hours UGE of all rats taking NAA were normal or nearly normal, islet function and morphology revealed no significant differences compared with those of the normal, whereas all rats of control group remained hyperglycemic (> 16.6 mmol/L) and markedly higher 24 hours UGE as compared with the normal islets appeared atrophic with decrement of B cell population.The results showed that NAA may effectively prevent the occurrence of STZ-induced diabetes mellitus in rats, suggesting that NAA may be an effective agent for prevention and treatment of insulin-dependent diabetes mellitus (IDDM) in prediabetic and early stage of IDDM.
Stevia extract safeguards rats from diabetic renal injury by mitigating markers of inflammation, oxidative stress, and apoptosis

Abstract:Objective This study will figure out if stevia extract, a herb with anti-inflammatory and antioxidant characteristics, is effective in treating diabetic nephropathy (DN) and protecting off concerns brought on by diabetic renal impairment. Methods KIM-1, NGAL, and total protein (TP) concentrations in urine are all estimated using biochemical assays in addition to serum glucose and insulin levels. Rat serum were subjected to an ELISA analysis to assess kidney function, glutathione (GSH), and malondialdehyde (MDA). Also, apoptotic mediators alongside inflammatory markers were evaluated using this technique in rat kidney tissue. By using Hematoxylin and eosin stains, under a microscope, the histological alterations in the kidneys were studied. Results Oral administration of Stevia extract (400 mg/kg) for 30 days normalized serum glucose and insulin levels in diabetic rats compared to non-diabetic rats, indicating its antidiabetic efficacy. Stevia administration improved blood creatinine, uric acid, and TP levels in diabetic rats. Stevia's capacity to maintain GSH and lower MDA in diabetic rats' serum showed its antioxidant power. Stevia additionally inhibited the renal tissues of Type 2 diabetes mellitus (T2DM) rats from expressing Bax, caspase-3, and Cyt-c and elevating proinflammatory cytokines like IL-6, IL-1β, and TNF-α. Stevia extract therapy in diabetic rats' kidneys restored anti-antiapoptotic Bcl-2 and P53. In diabetic rats, stevia reduced urine KIM-1, NGAL, and TP levels, pointing to its anti-inflammatory and antiapoptotic characteristics. Comet length showed more DNA damage in renal tissues of diabetics than controls, but stevia extract minimized it. Ultimately, histopathological changes led to confirm these results. Conclusion To sum up, the remarkable anti-inflammatory and antiapoptotic properties of stevia extract, along with its antihyperglycemic and antioxidant actions, are responsible for its protective effects on the DN during the early stage of T2DM. Stevia may therefore be an effective and helpful approach to alleviate DN and other adverse effects.
Effects of enalapril and paricalcitol treatment on diabetic nephropathy and renal expressions of TNF-α, p53, caspase-3 and Bcl-2 in STZ-induced diabetic rats

Abstract:This study aimed to assess the renopreventive effect of enalapril and/or paricalcitol on streptozotocin (STZ) diabetes-induced nephropathy and to elucidate their mechanisms of action through investigation of the effects on renal oxidative stress, antioxidant defense system and expressions of TNF-α, p53, caspase-3, and Bcl-2. Diabetes mellitus was induced in fasting male Wistar rats by single intraperitoneal injection of STZ (45 mg /kg b.w.) dissolved in citrate buffer (pH 4.5). Ten days after STZ injection, the diabetic rats were treated with enalapril (25 mg/l of drinking water) and/or paricalcitol (8 μg/kg b.w. per os) dissolved in 5% DMSO daily for 4 weeks. The obtained data revealed that the treatment of diabetic Wistar rats with enalapril and/or paricalcitol led to significant decreases in the elevated serum urea, uric acid, creatinine, sodium and potassium levels; thereby reflecting the improvement of the impaired kidney function. The deteriorated kidney lipid peroxidation, GSH content and GST and catalase activities in diabetic rats were significantly ameliorated as a result of treatment with enalapril and/or paricalcitol. The elevated fasting and post-prandial serum glucose levels and the lowered serum insulin and C-peptide levels were also improved. The treatment with enalapril and paricalcitol in combination was the most potent in decreasing the elevated serum glucose levels. Moreover, the treatment of diabetic rats successfully prevented the diabetes-induced histopathological deleterious changes of kidney and islets of Langerhans of pancreas. In association, the immunohistochemically detected pro-inflammatory cytokine, TNF-α, and apoptotic mediators, p53 and caspase-3, were remarkably decreased in kidney of diabetic rats as a result of treatment while the expression of anti-apoptotic protein Bcl-2 was increased. Based on these findings, it can be concluded that enalapril and paricalcitol alone or in combination can prevent STZ diabetes-induced nephropathy through amelioration of the glycemic state and antioxidant defense system together with the suppression of oxidative stress, inflammation and apoptosis. However, the treatment of diabetic rats with enalapril and paricalcitol in combination has no further significant improvement effects on renal function and damage when compared with enalapril or paclitaxel treated diabetic groups.

Abstract:Objective:To investigate the protective effect of novel cannabinoid(CB)1 receptor antago-nist ZH-1 01-S on kidneys in streptozotocin induced diabetic rats.Methods:Diabetic models were estab-lished by intraperitoneal injection of streptozotocin,rats were randomized into diabetes mellitus group(DM group,normal saline,i.p.),low dose group(0.3 mg/kg ZH-1 01-S,i.p.),middle dose group(1 .0 mg/kg ZH-1 01-S,i.p.),and high dose group(3.0 mg/kg ZH-1 01-S,i.p.);and healthy rats were selected as control group(normal saline,i.p.).After 8 weeks,the serum urea nitrogen(BUN),serum creatinine (Scr),urine albumin excretion rate(UAER),and the level of malondialdehyde(MDA)and the activity of superoxide dismutase(SOD)in renal tissues were detected;the pathological changes of renal tissues were observed by HE staining;the expression of voltage-dependent anion channel(VDAC)protein in renal tis-sues were examined by Western blotting.Results:Compared with control group,DM group showed higher levels of UAER,Scr and BUN(P<0.05 ),decreased SOD activity(P<0.01 ),and increased expression of MDA(P<0.05);decreased VDAC expression(P<0.01 );glomerular volume increased,renal tubular epithelial cell edema,mesangial matrix increased.Compared with DM group,high dose group had lower levels of UAER,Scr and BUN,increased SOD activity,lower content of malondialdehyde;increased VDAC expression(P<0.01 ),the pathological morphology of the kidney was improved.Conclusion:ZH-1 01-S showed protective effect on the kidney of diabetic rats,which may be related to the inhibition of oxi-dative stress and the improvement of kidney mitochondrial function.
Protective Effects of Riboflavin on Diabetic Nephropathy in STZ-induced Diabetic Rats

Abstract:AIM:To explore the protective effects of riboflavin on the kidney in streptozotocin (STZ)-induced diabetic rats.METHODS:Male Sprague-Dawley rats were randomly divided into 3 groups:normal control group,diabetic model group and riboflavin-treated group.Diabetes was induced by a single injection of STZ (dissolved in 0.01 mol/L citrate buffer,pH 4.5,65 mg/kg,ip) in rats.The biochemical methods were used to measure the contents of urine protein and malondialdehyde in the kidney,and the activities of superoxide dismutase (SOD) and catalase (CAT) in serum and renal tissues.Furthermore,the protein expression of TGF-β1 and plasminogen activator inhibitor-1(PAI-1) in renal cortex was detected by Western blotting.The morphological changes of renal tissue were observed under microscope.RESULTS:Compared to the diabetic model group,riboflavin significantly increased the activities of SOD and CAT (P0.01) in the serum and renal tissues,and decreased the contents of urine protein and MDA (P0.01) in the renal tissues in riboflavin treated group.The levels of TGF-β1 and PAI-1 in the renal cortex were dramatically decreased in the treated diabetic rats compared to the diabetic model rats (P0.01).CONCLUSION:Riboflavin inhibits the protein expression of TGF-β1 and PAI-1 in renal tissue of STZ-induced diabetic rats.Riboflavin may alleviate the pathologic changes and play an important protective role in diabetic kidneys.
Effect of arctiin on glomerular filtration barrier damage in STZ-induced diabetic nephropathy rats

Abstract:Diabetic nephropathy (DN) is the major life-threatening complication of diabetes. Abnormal permeability of glomerular basement membrane plays an important role in DN pathogenesis. This study was performed to assess the effect of arctiin, the lignan constituent from Arctium lappa L., on metabolic profile and aggravation of renal lesions in a rat model of streptozotocin (STZ)-induced DN. STZ-induced diabetic rats were treated with arctiin at the dosage of 60 or 40 mg/kg/day via intraperitoneal injection for 8 weeks. Blood glucose and 24-h urinary albumin content were measured, and kidney histopathological changes were monitored. RT-PCR and immunohistochemistry were used to detect the mRNA and protein levels of nephrin, podocin and heparanase (HPSE) in the kidney cortex of rats, respectively. Treatment with arctiin significantly decreased the levels of 24-h urinary albumin, prevented the sclerosis of glomeruli and effectively restored the glomerular filtration barrier damage by up-regulating the expression of nephrin and podocin and down-regulating HPSE level. Our studies suggest that arctiin might be beneficial for DN. The effects of arctiin on attenuating albuminuria and glomerulosclerosis are possibly mediated by regulating the expression of nephrin and podocin and HPSE in STZ-induced diabetic rats.
Vinpocetine mitigates proteinuria and podocytes injury in a rat model of diabetic nephropathy

Abstract:Podocyte injury and glomerular basement membrane thickening have been considered as essential pathophysiological events in diabetic nephropathy. The aim of this study was to investigate the possible beneficial effects of vinpocetine on diabetes-associated renal damage. Male Wistar rats were made diabetic by injection of streptozotocin (STZ). Diabetic rats were treated with vinpocetine in a dose of 20mg/kg/day for 6 weeks. Treatment with vinpocetine resulted in a marked decrease in the levels of blood glucose, glycosylated haemoglobin, creatinine, blood urea nitrogen, urinary albumin and albumin/creatinine ratio along with an elevation in creatinine clearance rate. The renal contents of advanced glycation end-products, interleukin-10, tissue growth factor-β, nuclear factor (NF)-κB and Ras-related C3 botulinum toxin substrate 1 (Rac 1) were decreased. Renal nephrin and podocin contents were increased and their mRNA expressions were replenished in vinpocetine-treated rats. Moreover, administration of vinpocetine showed improvements in oxidative status as well as renal glomerular and tubular structures. The current investigation revealed that vinpocetine ameliorated the STZ-induced renal damage. This beneficial effect could be attributed to its antioxidant and antihyperglycemic effects parallel to its ability to inhibit NF-κB which eventually modulated cytokines production as well as nephrin and podocin proteins expression.
Cardioprotective effect of vanillic acid against doxorubicin-induced cardiotoxicity in rat

Abstract:BACKGROUND AND PURPOSE: Doxorubicin (DOX) is an effective agent for the treatment of many neoplastic diseases. Cardiotoxicity is the major side effect of this drug and limits its use. Vanillic acid (VA) is a pharmaceutical compound from the phenolic acids family. The present study is an attempt to investigate the possible helpful effects of VA against DOX-induced cardiotoxicity in rats.EXPERIMENTAL APPROACH: For induction of cardiotoxicity, male Wistar rats received total of six doses of DOX (2.5 mg/kg i.p.) three times per week from days 14 to 28. Treatment groups received daily oral doses of VA (10, 20, and 40 mg/kg) two weeks before DOX injection and then plus DOX for 2 weeks. At the end of experiment, systolic blood pressure (SBP) and heart rate (HR) were detected using tail-cuff method. Lactate dehydrogenase (LDH), creatine phosphokinase-MB (CK-MB), serum glutamic oxaloacetic transaminase (SGOT), malondialdehyde (MDA), and ferric reducing antioxidant power (FRAP) were measured in serum samples. Troponin-I and toll-like receptor 4 (TLR4) were measured in cardiac tissue. All the measurements processed spectrophotometrically using commercial ELISA kits. Cardiac tissue was also processed for histopathological examination.FINDINGS / RESULTS: Treatment with VA significantly increased SBP compared to the DOX group and restored HR near to the normal level. Administration of VA at all of doses, decreased serum levels of LDH, SGOT, CK-MB, MDA, cardiac troponin-I, cardiac TLR4 and increased FRAP value.CONCLUSION AND IMPLICATIONS: These results suggest that VA may exert cardioprotective effects against DOX-induced cardiotoxicity by decreasing oxidative stress and biomarkers of cardiotoxicity, suppression of TLR4 signaling and consequently inflammation pathway.
Amorphophallus campanulatus tuber extract protects diabetic nephropathy in streptozotocin-induced diabetic nephropathy rat model by regulating oxidative stress and TNF-α inflammatory pathway

Abstract:Purpose: To assess the effect of Amorphophallus campanulatus tuber (Ac) extract in the protection of diabetic nephropathy in streptozotocin (STZ) induced diabetic nephropathy (DN) rat model. Methods: Diabetes was induced with STZ (60 mg/kg, i.p.), and DN was confirmed after six weeks of STZ administration with the estimation of kidney function test. Further rats were treated with Ac 250 and 500 mg/kg p.o. for next four week. Oxidative stress and level of inflammatory cytokines were estimated in the kidney tissue of DN rats. Histopathology of kidney tissue was performed using hematoxylin and eosin staining. Results: There was improvement in the body weight of Ac treated groups than DN group of rats. Blood glucose level was observed to be reduced in Ac treated groups than DN group on 42nd and 70th day of protocol. Treatment with Ac ameliorated the altered level of kidney function tests (creatinine and BUN), enzymes of liver function (aspartate aminotransferase and alanine aminotransferase), and lipid profile in the serum of DN rats. Oxidative stress parameters (malondialdehyde and reactive oxygen species enhances and reduction in the level of glutathione and superoxide dismutase) and inflammatory cytokines such as interleukin-6, tumour necrosis factor-α, and monocyte chemoattractant protein-1 reduces in the tissue of Ac treated group than DN group. Treatment with Ac also attenuates the altered histopathological changes in the kidney tissue of DN rats. Conclusions: The report suggests that Ac protects renal injury in DN rats by regulating inflammatory cytokines and oxidative stress.
Oral administration of veratric acid, a constituent of vegetables and fruits, prevents cardiovascular remodelling in hypertensive rats: a functional evaluation

Abstract:In our previous studies, veratric acid (VA) shows beneficial effect on hypertension and its associated dyslipidaemia. In continuation, this study was designed to investigate the effect of VA, one of the major benzoic acid derivatives from vegetables and fruits, on cardiovascular remodelling in hypertensive rats, primarily assessed by functional studies using Langendorff isolated heart system and organ bath system. Hypertension was induced in male albino Wistar rats by oral administration of N ω -nitro-l-arginine methyl ester hydrochloride (l-NAME) (40 mg/kg body weight (b.w.)) in drinking water for 4 weeks. VA was orally administered at a dose of 40 mg/kg b.w. l-NAME-treated rats showed impaired cardiac ventricular and vascular function, evaluated by Langendorff isolated heart system and organ bath studies, respectively; a significant increase in the lipid peroxidation products such as thiobarbituric acid-reactive substances and lipid hydroperoxides in aorta; and a significant decrease in the activities of superoxide dismutase, catalase, glutathione peroxidase and levels of GSH, vitamin C and vitamin E in aorta. Fibrotic remodelling of the aorta and heart were assessed by Masson's Trichrome staining and Van Gieson's staining, respectively. In addition, l-NAME rats showed increased heart fibronectin expression assessed by immunohistochemical analysis. VA supplementation throughout the experimental period significantly normalised cardiovascular function, oxidative stress, antioxidant status and fibrotic remodelling of tissues. These results of the present study conclude that VA acts as a protective agent against hypertension-associated cardiovascular remodelling.

Abstract:Diabetic nephropathy is the one of the major complication of diabetes mellitus which is the main cause of renal failure. The treatment of diabetic nephropathy includes different nonpharmacological and pharmacological approach, but poor prognosis. Our study was designed to investigate the combined effect of resveratrol and vitamin D3 on the glucose homeostasis, histological changes, and cell damage biomarkers in streptozotocin-induced diabetic rat kidneys. Oral treatment with resveratrol (10 mg/kg/day), vitamin D3 (0.5 µgm/kg), and the combination for 10 weeks showed an increase in the average body weights compared to the untreated diabetic rats. The combination showed a highly significant decrease in fasting blood glucose levels, serum cholesterol levels compared to the untreated diabetic rats, there is a significant increase in serum HDL cholesterol levels. There is a highly significant decrease in serum creatinine, urea, albumin levels compared to the untreated diabetic rats. However, the combination treatment provides superior efficacy than individual drug treatments. Histopathological examination of the kidney tissue section revealed that the diabetic rats treated with resveratrol, vitamin D3, and the combination showed noticeable improvement in the histopathological parameters compared to untreated diabetic rats. However, a marked reduction of TNF-α and IL-6 expression was observed in combination treated rats compared to individual drug treatments. Combination therapy was significantly more effective in treating diabetic nephropathy than resveratrol and vitamin D3 alone. Resveratrol and vitamin D3 has a protective role of diabetic kidneys.
Vitamin D regulates insulin and ameliorates apoptosis and oxidative stress in pancreatic tissues of rats with streptozotocin-induced diabetes

Abstract:This study was designed to evaluate the potential therapeutic efficacy of vitamin D (Vit D) in averting the harmful effects of type 2 diabetes mellitus (T2D). Forty male Wistar rats were allotted into four groups: (1) the control, (2) Vit D, (3) streptozotocin (STZ), and (4) STZ + Vit D groups. Rats co-treated with Vit D had significantly ( p < 0.05) decreased levels of cortisol; proinflammatory cytokines, including interleukin-6 (IL-6); and malondialdehyde (MDA). Meanwhile, the levels of insulin significantly ( p < 0.05) increased, whereas the activity of the antioxidant system, including glutathione (GSH), superoxide dismutase (SOD), catalase, and total antioxidant capacity (TAC), significantly ( p < 0.05) decreased. Histopathological examination revealed the destruction of beta cells in the islets of Langerhans in rats with diabetes. Meanwhile, immunoexpression revealed an increase in the immunoreactivity of caspase-3 and endothelial nitric oxide synthase and a reduction in the immunoreactivity of insulin in rats with diabetes. In conclusion, Vit D ameliorated the harmful biochemical impact of diabetes mellitus, probably by increasing insulin secretion and sensitivity, ameliorating β-cell function, and decreasing cortisol levels; also, the anti-inflammatory effect of Vit D reduces the number of proinflammatory cytokines (e.g., IL-6) and increases the activity of the antioxidant system, such as GSH, SOD, TAC, and catalase while reducing lipid peroxidation enzymes (e.g., MDA).

Abstract:OBJECTIVE:To explore the renoprotective effect of Compound Xueshuantong Capsule (XST) on diabetic rat model with nephropathy.METHODS:Twenty-eight male Sprague Dawley diabetic rats were induced to hyperglycaemia (3 days later, fasting blood glucose > 16.7 mmol/L) by peritoneal injection with streptozotocin (STZ, 50 mg/kg). And they were divided into four groups: diabetic nephropathy (vehicle treatment), irbesartan (20 mg×kg(-1)×d(-1)), low-dosage XST (900 mg×kg(-1)×d(-1)) and high-dosage XST (1800 mg×kg(-1)×d(-1)). Seven normal rats were used as control. After a 12-week intervention, urine protein was examined. Pathological morphology was observed by hematoxylin-eosin (HE), Masson and (periodic acid Schiff) PAS stains. Blood nitric oxide (NO), malondialdehyde (MDA) and blood superoxide dismutase (SOD) and urine SOD were detected. And the expression of (matrix metalloproteinase-2) MMP-2 was detected by Western blot in each group.RESULTS:The model rats presented with hyperglycemia, polydipsia, hyperphagia, polyuria and hyper microalbuminuria. The intervention groups showed decreased microalbuminuria and there was no effect on blood glucose or body weight. Glomerular sclerosis and extracellular matrix (ECM) increased in model group and improved in irbesartan and XST groups as judged by HE, Masson and PAS stains. Three intervention groups had no effect on the elevated expression of MMP-2 in diabetic rats. Compared with the model group, the irbesartan, low-dosage and high-dosage XST groups had significantly decreased blood levels of NO ((104.9 ± 11.0) µmol/L vs (41.9 ± 9.6) µmol/L and (14.7 ± 1.9) µmol/L, P < 0.05) and MDA ((19.6 ± 1.6) nmol/L vs (6.6 ± 0.9) mol/L and (4.5 ± 1.2) nmol/L, P < 0.05), increased blood and urine activities of SOD (blood: (222 ± 20)×10(3) vs (231 ± 18)×10(3) and (237 ± 24)×10(3) U/L,P < 0.05), urine: (11.8 ± 1.1)×10(3) vs (23.3 ± 2.0)×10(3) and (25.7 ± 1.8)×10(3) U/L).CONCLUSION:Compound Xueshuantong Capsule may decrease proteinuria through its suppression of oxidative stress and not its improvement of ECM metabolism.
Aldosterone Antagonists in Monotherapy Are Protective against Streptozotocin-Induced Diabetic Nephropathy in Rats

Abstract:Angiotensin converting enzyme inhibitors (ACEi) and angiotensin II receptor blockers (ARB) are the standard clinical therapy of diabetic nephropathy (DN), while aldosterone antagonists are only used as adjuncts. Previously in experimental DN we showed that Na/K ATPase (NKA) is mislocated and angiotensin II leads to superimposed renal progression. Here we investigated the monotherapeutic effect of aldosterone blockers on the progression of DN and renal NKA alteration in comparison to ACEi and ARBs. Streptozotocin-diabetic rats developing DN were treated with aldosterone antagonists; ACEi and ARB. Renal function, morphology, protein level and tubular localization of NKA were analyzed. To evaluate the effect of high glucose per se; HK-2 proximal tubular cells were cultured in normal or high concentration of glucose and treated with the same agents. Aldosterone antagonists were the most effective in ameliorating functional and structural kidney damage and they normalized diabetes induced bradycardia and weight loss. Aldosterone blockers also prevented hyperglycemia and diabetes induced increase in NKA protein level and enzyme mislocation. A monotherapy with aldosterone antagonists might be as, or more effective than ACEi or ARBs in the prevention of STZ-induced DN. Furthermore the alteration of the NKA could represent a novel pathophysiological feature of DN and might serve as an additional target of aldosterone blockers.

Abstract:Objective:To explore the renoprotective effect of L-arginine on streptozotocin-induced diabetic rats and its potential mechanism.Methods:Diabetic models were induced by intraperitoneal injection of streptozotocin STZ in r+ats.Models were randomly divided into two groups:DM group(non-treated diabetic model rats)and L-Arg group(diabetic model rats treated with L-arginine after 4 weeks).The normal non-diabetic rats were as the control group.The excretion of urinary albumin excretion rate(UAER),serum creatinine(SCr) and serum urea nitrogen(BUN)were detected after 8 weeks.Morphological changes of kidney were observed by optics microscope.The levels of nitric oxide(NO),the activity of superoxide dismutase(SOD)and malondialdehyde(MDA) in serum and renal cortex were detected;kidney mitochondrial membrane potential and mitochondrial swelling were measured in the different groups. Results:Compared with DM group,the excretion of UAER,SCr and BUN in L-Arg group were significantly decreased(P<0.05),while the abnormal pathological changes were improved;the levels of NO and the activity of SOD in L-Arg group were also significantly increased(P<0.01,P<0.05),the levels of MDA were significantly reduced(P<0.05);the kidney mitochondrial membrane potential was significantly elevated(P<0.05) and mitochondrial swelling was reinforced. Conclusion:L-arginine protects the kidneys during diabetic model rats possibly by increasing the levels of NO and improving the function of mitochondria.
