Abstract:Rhus typhina L. (staghorn sumac) represents a valuable species in terms of its medicinal properties and its wide variety of uses, which deserves more attention in the scientific world. Its fruits contain important amounts of anthocyanins, vitamins, minerals and essential amino acids, while gallic acid and several hydrolyzed tannins are produced by the leaves. Although staghorn sumac has a milky sap that can produce dermatitis and its pollen causes severe allergies, the species has been shown to be useful in treating other disorders such as stomach aches, conjunctivitis and improving the condition of people with diabetes. In the last 10 years the most studies have focused on the ecological impact of this plant and its resistance to different environmental characteristics and less on its bioactivity and chemical composition. As any other invasive species, R. typhina modifies the soil properties and the structure of local plant communities, but is sensitive to light and ozone pollution. In terms of its utility, this species of sumac finds its place in the field of gastronomy, in the production of fuels, cosmetics, pesticides, but also in paper manufacturing.

Abstract:Rhus typhina, commonly called staghorn sumac, is native to North America and is widely planted as an ornamental deciduous shrub all over the temperate region. In this study, the complete chloroplast genome (plastome) of R. typhina was determined using genome-skimming method. The complete plastome of staghorn sumac was 160,204 bp in length with a canonical quadripartite structure, including a large single-copy region of 87,789 bp, a small single-copy region of 19,319 bp, and a pair of inverted repeats regions of 26,548 bp each. A total of 113 unique genes were annotated in this plastome, including 79 protein-coding genes, 30 tRNA genes, and 4 rRNA genes. The overall guanine-cytosine (GC) content was 37.8%. As shown in the ML phylogenetic tree, R. typhina was closely related to R. chinensis, and Rhus was sister to Pistacia within Anacardiaceae famliy.

Abstract:Taxus is a genus of 12 species mainly distributed in the Northern Hemisphere, with four species in North America, one in Europe, and the remaining native to Southeast Asia. Taxus contains numerous compounds of taxane derivatives, polysaccharides, and flavanoids. The compound with the greatest significance for humans is an oxygen-rich diterpene called taxol, which can be used as an inhibitor of cancer cell growth. Many new biological activities of these chemical compounds have been discovered to fight against various cancers (e.g., ovarian, breast, esophagus, gastric, lung, and prostate cancers), anti-oxidation, cure inflammatory and HIV effects, and eliminate free radicals. This review revises the information on the morphological characteristics and geographical distribution, generalizes the relationship of special secondary metabolites among species, summarizes the main pharmacological activities of the compounds contained in plant, and also puts forward some effective strategies for the exploitation and protection of the Taxus species. These works may be helpful for the researcher and utilizer.

Abstract:Rhus typhina is a valuable plant used in the pharmaceutical, cosmetic, and food industries due to the presence of biologically active substances accumulated in its organs, especially in secretory structures, i.e. trichomes and secretory ducts. Light microscopy, scanning electron microscopy, and transmission electron microscopy were used to examine the structure of glandular and non-glandular trichomes, as well as secretory ducts present in inflorescence peduncles of R. typhina. The chemical composition of the secretion produced by trichomes and ducts was assessed using histochemical techniques, including observations under brightfield and fluorescence microscopes. Two types of capitate glandular trichomes producing secretions with a similar composition and non-glandular trichomes exhibiting secretory activity were identified. The secretion of glandular trichomes was dominated by acidic and neutral lipids, essential oil, sesquiterpenes, and steroid-containing terpenes. The schizogenic secretory ducts located in the phloem produced a viscous milky substance with acidic polysaccharides, acidic lipids, phenolic compounds, and proteins. The secretion was released into the duct lumen through notches in the walls of the secretory epithelial cell facing the duct lumen. The location, type, and traits of the non-glandular trichomes and secretory structures, as well as the composition of the secreted products are considered important taxonomic features in the family Anacardiaceae and the Rhus genus. Additionally, these characters are important diagnostic markers for the pharmacobotanical identification of the species in medicinal and cosmetic raw materials. The various compounds present in the secretory structures of R. typhina may contribute to plant protection against pathogens or herbivory and probably play a role as attractants for pollinators and seed dispersers.
De Novo Transcriptome Characterization of Rhodomyrtus Tomentosa Leaves and Identification of Genes Involved in Α/β-Pinene and Β-Caryophyllene Biosynthesis

Abstract:Plant-derived terpenes are effective in treating chronic dysentery, rheumatism, hepatitis, and hyperlipemia. Thus, understanding the molecular basis of terpene biosynthesis in some terpene-abundant Chinese medicinal plants is of great importance. Abundant in mono- and sesqui-terpenes, Rhodomyrtus tomentosa (Ait.) Hassk, an evergreen shrub belonging to the family Myrtaceae, is widely used as a traditional Chinese medicine. In this study, (+)-α-pinene and β-caryophyllene were detected to be the two major components in the leaves of R. tomentosa, in which (+)-α-pinene is higher in the young leaves than in the mature leaves, whereas the distribution of β-caryophyllene is opposite. Genome-wide transcriptome analysis of leaves identified 138 unigenes potentially involved in terpenoid biosynthesis. By integrating known biosynthetic pathways for terpenoids, 7 candidate genes encoding terpene synthase (RtTPS1-7) that potentially catalyze the last step in pinene and caryophyllene biosynthesis were further characterized. Sequence alignment analysis showed that RtTPS1, RtTPS3 and RtTPS4 do not contain typical N-terminal transit peptides (62-64aa), thus probably producing multiple isomers and enantiomers by terpenoid isomerization. Further enzyme activity in vitro confirmed that RtTPS1-4 mainly produce (+)-α-pinene and (+)-β-pinene, as well as small amounts of (-)-α-pinene and (-)-β-pinene with GPP, while RtTPS1 and RtTPS3 are also active with FPP, producing β-caryophyllene, along with a smaller amount of α-humulene. Our results deepen the understanding of molecular mechanisms of terpenes biosynthesis in Myrtaceae.

Abstract:Abstract Dried and powdered fruits of sicilian sumac (Rhus coriaria L., family Anacardiaceae R.Br.) have been used in Asian countries and some others for a very long time as seasoning (spices «sumac», «zatar»). The research is aimed at an assessment of prospects for use of fruits of staghorn sumac (Rhus typhina L.), growing in the middle zone of the Russian Federation, as a spice, having medicinal properties. The morphological and anatomical, dimensional and weight characteristics of Rh. typhina’s fruits, collected in the arboretum of Tsytsin Main Botanical Garden of the Russian Academy of Sciences (MBG RAS), having medicinal properties, were studied. The content of absolutely dry matter and vitamin C in them was determined. For comparison of biochemical parameters, seasoning «sumac» (Rh. coriaria), purchased on open market, was evaluated. It was established that surface of Rh. typhina’s fruit is covered with multicellular trichomes of two types (covering long single-row and glandular capitate on two-celled pedicle); pericarp is differentiated, its structure as a whole is similar to that, described in the literature; the content of absolutely dry matter in air-dry matter (90.5%) and vitamin C (62.0 mg%) in Rh. typhina’s fruits is slightly less than those for Rh. coriaria’s fruits – 96.5% and 64.6 mg%, respectively. Mass of 1000 fruits Rh. typhina (about 12 g) in MBG RAS arboretum is 1,5 times exceed the according to the literature. Thus, Rh. typhina’s fruits may be recommended for further study as an alternative to Rh. coriaria’s fruits, which are traditionally used for making spices.

Abstract:Rhus coriaria L. (Anacardiaceae), commonly known as sumac, has been used since ancient times for many different applications, and nowadays is used mostly as a spice obtained from its in the Mediterranean and the Middle ground fruits and employed for flavoring and garnishing food, predominantly Eastern regions. Traditionally, sumac has been also used in popular medicine for the treatment of many ailments including hemorrhoids, wound healing, diarrhea, ulcers, and eye inflammation. Sumac drupes are indeed rich in various classes of phytochemicals including organic acids, flavonoids, tannins, and others, which are responsible of their powerful antioxidant capacity, from which treatment of many common diseases such as cardiovascular disease, diabetes, and cancer could benefit. In this work we evaluated the influence of fruit ripeness, conservation, and processing. To this aim, a phytochemical characterization of six different samples of Rhus coriaria L. was carried out. Specifically, headspace solid-phase micro extraction gas chromatography coupled to mass spectrometry and comprehensive two-dimensional liquid chromatography coupled to photodiode array and mass spectrometry detection, were employed. A total of 263 volatile compounds, including terpene hydrocarbons, acids, and aldehydes, as well as 83 polyphenolic compounds, mainly gallic acid derivatives, were positively identified. All samples showed a significant antioxidant activity by means of oxygen radical absorbance capacity, in line with their polyphenolic content and composition. Such findings set a solid ground to support the utilization of this plant as an attractive target for novel nutraceutical approaches and for drug discovery.

Phylogenetic and biogeographic diversification of Rhus (Anacardiaceae) in the Northern Hemisphere

Abstract:Sequences of internal transcribed spacers (ITS) of nuclear ribosomal DNA, the chloroplast ndhF gene, and chloroplast trnL-F regions (trnL intron, and trnL [UAA] 3' exon-trnF [GAA] intergenic spacer) were used for phylogenetic analyses of Rhus, a genus disjunctly distributed in Asia, Europe, Hawaii, North America, and Northern Central America. Both ITS and cpDNA data sets support the monophyly of Rhus. The monophyly of subgenus Rhus was suggested by the combined cpDNA and ITS data, and largely supported in the cpDNA data except that Rhus microphylla of subgenus Lobadium was nested within it. The monophyly of subgenus Lobadium was strongly supported in the ITS data, whereas the cpDNA data revealed two main clades within the subgenus, which formed a trichotomy with the clade of subgenus Rhus plus R. microphylla. The ITS and cpDNA trees differ in the positions of Rhus michauxii, R. microphylla, and Rhus rubifolia, and hybridization may have caused this discordance. Fossil evidence indicates that Rhus dates back to the early Eocene. The penalized likelihood method was used to estimate divergence times, with fossils of Rhus subgenus Lobadium, Pistacia and Toxicodendron used for age constraints. Rhus diverged from its closest relative at 49.1+/-2.1 million years ago (Ma), the split of subgenus Lobadium and subgenus Rhus was at 38.1+/-3.0 Ma. Rhus most likely migrated from North America into Asia via the Bering Land Bridge during the Late Eocene (33.8+/-3.1 Ma). Rhus coriaria from southern Europe and western Asia diverged from its relatives in eastern Asia at 24.4+/-3.2 Ma. The Hawaiian Rhus sandwicensis diverged from the Asian Rhus chinensis at 13.5+/-3.0 Ma. Subgenus Lobadium was inferred to be of North American origin. Taxa of subgenus Lobadium then migrated southward to Central America. Furthermore, we herein make the following three nomenclatural combinations: (1) Searsia leptodictya (Diels) T. S. Yi, A. J. Miller and J. Wen, comb. nov., (2) Searsia pyroides (A. Rich.) T. S. Yi, A. J. Miller and J. Wen, comb. nov., and (3) Searsia undulata (Jacq.) T. S. Yi, A. J. Miller and J. Wen, because our analyses support the segregation of Searsia from Rhus.

Abstract:Rhus coriaria L. (Anacardiaceae), sumac, is a common condiment, appetizer and souring agent in the Mediterranean region that has a long history in traditional medicine. R. coriaria has been prescribed for the treatment of many ailments including diarrhea, ulcer, hemorrhoids, hemorrhage, wound healing, hematemesis, and eye ailments like ophthalmia and conjunctivitis. The plant is also used as diuresis, antimicrobial, abortifacient and as a stomach tonic. Sumac is known to be rich in different classes of phytochemicals including tannins, polyphenols, flavonoids, flavones, organic acids and essential oils and continues to be a hot topic for extensive research work designed for revealing its phytochemical constituents and evaluating its bioactive properties. This review article summarizes the recent phytochemical and diverse bioactivity studies on R. coriaria , especially those concerned with antitumor, antioxidant, hypoglycemic, antimicrobial, and anti‐inflammatory studies.

Abstract:Numerical taxonomic studies were done on 13 species from 3 genera of tribe Areae in Zhejiang, used as 13 operational taxonomic units (OTUS). 40 morphological characters were used for analysis. Euclid distance coefficients used to show quantitative index of similarity among OTUS were computed by standardized data. The dendrograms from 8 systematic cluster methods including single linkage method, complete linkage method, median method (beta = 0 WPGMA) and (beta = -0.25 WPGMA), centroid method, group average method, variable group average method (beta = -0.25) and variable method (beta = 0.25) were constructed respectively. WPGMA was slected as the optimal one by computing the comparative coefficients of every cluster result and the boundary of taxa in its dendrogram determined by a method to treated IBM computer with the program by BASIC language. The Results showed that 13 OTUs were classified as 3 clusters by broken line L2, namely, cluster I: Typhonium; cluster II: Arisaema; cluster III: Pinellia, and cluster II further as 4 groups by L1, which belong to Section Pistillata, Arisaema, Sinarisaema and Tortuosa respectively. Being consistent with those from classical taxonomy, the results in this study may be helpful to the classification and identification of the medicinal plants from tribe Areae in Zhejiang Privince.

Abstract:Based on a literature survey and a field trip with specimen examination, statistical analysis including principal components analysis (PCA) on morphological characters have been carried out on Rhododendron mariae and related taxa. Cluster analysis and PCA showed three independent groups, which corresponded to three species except R. loniceriflorum. As a consequence, nine taxa are reduced to synonyms of R. mariae, and two new combinations, viz R. mariae subsp. microphyton, and R. mariae subsp. flumineum are made. Rhododendron microphyton var. trichanthum is reduced to a synonym of R. mariae subsp. microphyton, and R. malipoense to a synonym of R. mariae subsp. flumineum. Three names are synonymized and a new combination, viz R. chryocalyx subsp. meridionale, is proposed within R. chryocalyx. The third entity, R. naamkwanense, has got two new synonyms. Rhododendron loniceriflorum, with whitish pubescence on the inner corolla surface, is recognized as the fourth distinct species. Each entity is illustrated with line drawings for identification.

Abstract:Rhus potaninii Maxim is an economically and medicinally important tree species in China. It produces galls (induced by aphids) with a high abundance of tannins. Here, we discuss the histology, cellular structures and their distribution, and the macromolecular components of secretive glandular trichomes on the leaves of R. potaninii. A variation in the density of glandular trichomes and tomenta was found between the adaxial and abaxial sides of a leaf in different regions and stages of the leaf. The glandular trichomes on R. potaninii trees comprise a stalk with no cellular structure and a head with 8–15 cells. Based on staining, we found that the secretion of glandular trichomes has many polysaccharides, phenolic compounds, and acidic lipids but very few neutral lipids. The dense glandular trichomes provide mechanical protection for young tissues; additionally, their secretion protects the young tissues from pathogens by a special chemical component. According to transcriptome analysis, we found enhanced biosynthetic and metabolism pathways of glycan, lipids, toxic amino acids, and phenylpropanoids. This shows a similar tendency to the staining. The numbers of differentially expressed genes were large or small; the averaged range of upregulated genes was greater than that of the downregulated genes in most subpathways. Some selectively expressed genes were found in glandular trichomes, responsible for the chitinase activity and pathogenesis-related proteins, which all have antibacterial activity and serve for plant defense. To our knowledge, this is the first study showing the components of the secretion from glandular trichomes on the leaf surface of R. potaninii.

A chromosome-scale genome of Rhus chinensis Mill. provides new insights into plant-insect interaction and gallotannins biosynthesis

Abstract:Rhus chinensis Mill., an economically valuable Anacardiaceae species, is parasitized by the galling aphid Schlechtendalia chinensis, resulting in the formation of the Chinese gallnut (CG). Here, we report a chromosomal-level genome assembly of R. chinensis, with a total size of 389.40 Mb and scaffold N50 of 23.02 Mb. Comparative genomic and transcriptome analysis revealed that the enhanced structure of CG and nutritional metabolism contribute to improving the adaptability of R. chinensis to S. chinensis by supporting CG and galling aphid growth. CG was observed to be abundant in hydrolysable tannins (HT), particularly gallotannin and its isomers. Tandem repeat clusters of dehydroquinate dehydratase/shikimate dehydrogenase (DQD/SDH) and serine carboxypeptidase-like (SCPL) and their homologs involved in HT production were determined as specific to HT-rich species. The functional differentiation of DQD/SDH tandem duplicate genes and the significant contraction in the phenylalanine ammonia-lyase (PAL) gene family contributed to the accumulation of gallic acid and HT while minimizing the production of shikimic acid, flavonoids, and condensed tannins in CG. Furthermore, we identified one UDP glucosyltransferase (UGT84A), three carboxylesterase (CXE), and six SCPL genes from conserved tandem repeat clusters that are involved in gallotannin biosynthesis and hydrolysis in CG. We then constructed a regulatory network of these genes based on co-expression and transcription factor motif analysis. Our findings provide a genomic resource for the exploration of the underlying mechanisms of plant-galling insect interaction and highlight the importance of the functional divergence of tandem duplicate genes in the accumulation of secondary metabolites.

Abstract:Rhus typhina, a plant native to eastern North America, has previously been discovered to exhibit antibacterial activity against various ESKAPE pathogens including Staphylococcus aureus and Acinetobacter baumannii. The purpose of this work is to separate and purify compounds from the crude extracts using column chromatography, identify the pure compounds using NMR, and examine the therapeutic potential of these compounds on A.baumannii and S.aureus. A.baumannii is a non-motile, gram-negative, pleomorphic opportunistic pathogen and S.aureus is a non-motile, gram-positive, coccus-shaped, opportunistic pathogen; Both bacteria are major causes of nosocomial infections due to their ability to resist multiple antibiotics. As multi-drug resistance becomes more prevalent, there is a growing demand for research on new therapeutic agents. Supporting previous unpublished research from our laboratory, the data here suggest that ethanolic extracts of R.typhina contain compounds that exhibit antibacterial capabilities against both S. aureus and A. baumannii. Ongoing studies are focusing on determining the structure of the antimicrobial chemicals and the mode of action of these compounds.
A Revision of the rhododendron Taipaoense Complex (subg. tsutsusi Sect. tsutsusi, Ericaceae), Based on Observations of Morphological Characters and Seed Micromorphology

Abstract:In this paper, we present a revision of the Rhododenron taipaoense complex, including R. taipaoense and eight other named species, based on literature survey, field collections and observations, inspection of herbarium specimens, statistical analysis of morphological characters, and SEM observation of seed shape and testa. We evaluated all characters that have previously been used to diagnose these species: indumentum of young shoots and abaxial surfaces of leaves; filament dimensions; style base; leaf color, size, and apex shape; flower number per inflorescence; corolla length; and length of the stamen and pistil. Cluster analysis of 146 individuals showed that individuals from different populations are interdigitated, irrespective of traditional taxonomic limits. Analysis of morphological data using principal coordinate analysis (PCoA) likewise shows a continual gradation between populations, rather than disjunctures between named species. Consequently, only one species is recognized in the present paper, and R. apricum, R. falcinellum (R. apricum var. falcinellum), R. florulentum, R. hepticum, R. piceum, R. rufulum, R. spadiceum, and R. rufoindumentum (nom. nud.) are all reduced to synonyms of R. taipaoense.
